Abstract Background Metabolic syndrome (MetS) is highly prevalent in individuals with schizophrenia (SZ), leading to negative consequences like premature mortality. Gut dysbiosis, which refers to an imbalance of the microbiota, and chronic inflammation are associated with both SZ and MetS. However, the relationship between gut dysbiosis, host immunological dysfunction, and SZ comorbid with MetS (SZ-MetS) remains unclear. This study aims to explore alterations in gut microbiota and their correlation with immune dysfunction in SZ-MetS, offering new insights into its pathogenesis. Methods and results We enrolled 114 Chinese patients with SZ-MetS and 111 age-matched healthy controls from Zhejiang, China, to investigate fecal microbiota using Illumina MiSeq sequencing targeting 16 S rRNA gene V3-V4 hypervariable regions. Host immune responses were assessed using the Bio-Plex Pro Human Cytokine 27-Plex Assay to examine cytokine profiles. In SZ-MetS, we observed decreased bacterial α-diversity and significant differences in β-diversity. LEfSe analysis identified enriched acetate-producing genera (Megamonas and Lactobacillus), and decreased butyrate-producing bacteria (Subdoligranulum, and Faecalibacterium) in SZ-MetS. These altered genera correlated with body mass index, the severity of symptoms (as measured by the Scale for Assessment of Positive Symptoms and Scale for Assessment of Negative Symptoms), and triglyceride levels. Altered bacterial metabolic pathways related to lipopolysaccharide biosynthesis, lipid metabolism, and various amino acid metabolism were also found. Additionally, SZ-MetS exhibited immunological dysfunction with increased pro-inflammatory cytokines, which correlated with the differential genera. Conclusion These findings suggested that gut microbiota dysbiosis and immune dysfunction play a vital role in SZ-MetS development, highlighting potential therapeutic approaches targeting the gut microbiota. While these therapies show promise, further mechanistic studies are needed to fully understand their efficacy and safety before clinical implementation.

Abstract Background There is little evidence to comprehensively summarize the adverse events (AEs) profile of intermittent fasting (IF) despite its widespread use in patients with overweight or obesity. Methods We searched the main electronic databases and registry websites to identify eligible randomized controlled trials (RCTs) comparing IF versus control groups. A direct meta-analysis using a fixed-effect model was conducted to pool the risk differences regarding common AEs and dropouts. Study quality was assessed by using the Jadad scale. Pre-specified subgroup and sensitivity analyses were conducted to explore potential heterogeneity. Results A total of 15 RCTs involving 1,365 adult individuals were included. Findings did not show a significant difference between IF and Control in risk rate of fatigue [0%, 95% confidence interval (CI), -1% to 2%; P = 0.61], headache [0%, 95%CI: -1% to 2%; P = 0.86] and dropout [1%, 95%CI: -2% to 4%; P = 0.51]. However, a numerically higher risk of dizziness was noted among the IF alone subgroup with non-early time restricted eating [3%, 95%CI: -0% to 6%; P = 0.08]. Conclusions This meta-analysis suggested that IF was not associated with a greater risk of AEs in adult patients affected by overweight or obesity. Additional large-scale RCTs stratified by key confounders and designed to evaluate the long-term effects of various IF regimens are needed to ascertain these AEs profile.

BackgroundNowadays, the number of oral cancer survivors is increasing, emphasizing the importance of thoroughly understanding diverse causes of death in oral cancer survivors. Our study aimed to investigate the distribution of causes of death after oral cancer diagnosis.MethodsEligible patients were identified between 2004 and 2015 from the Surveillance, Epidemiology, and End Results (SEER) database. We calculated the number of deaths in different demographic and clinicopathological variables during each follow-up period. Standardized mortality ratios(SMRs) were generated for each cause of death after oral cancer diagnosis.ResultsA total of 30538 patients diagnosed with oral cancer were included, and 17654 deaths were reported during follow-up period. 27.08% of deaths were caused by non-caner reasons. The proportion of non-cancer related deaths increased with the extension of survival time, and non-cancer death accounted for 57.93% of all deaths when followed up more than 10 years. The most common non-cancer cause of death was cardiovascular disease (SMR 4.68#; 95%CI 4.46-4.92).ConclusionsNon-cancer causes of death should not be ignored in oral cancer patients. For oral cancer survivors, multidisciplinary follow-up strategy should be recommended to achieve longer survival time.

Summary: A series of ruthenium iron nitride phases with Ru:Fe ratios of ca. 1:3 were synthesized by ammonolysis. When the ammonolysis temperature was above 500°C, the obtained RuxFe3Ny materials had a ε-Fe3N (P6322) structure, while two similar phases were present when the ammonolysis was lower than 500°C. Powder neutron diffraction identified one phase as relating to the ε-Fe3N structure, while the other had a disordered NiAs-type (P63/mmc) structure. These ternary metal nitrides show ammonia synthesis activity at low temperature (200°C–300°C) and ambient pressure, which can be related to the loss of lattice nitrogen. Steady state catalytic performance at 400°C is associated with ruthenium-iron alloy. Additionally, density functional theory calculations were performed using an approximate model for the disordered hexagonal phase, revealing that this phase is more stable than a cubic anti-perovskite phase which has been previously investigated computationally, and corroborating the experimental findings of the present work.

Abstract How to exploit social networks to make internet content spread rapidly and consistently is an interesting question in marketing management. Although epidemic models have been employed to comprehend the spread dynamics of internet content, such as viral videos, the effects of advertising and individual sharing on information dissemination are difficult to distinguish. This gap forbids us to evaluate the efficiency of marketing strategies. In this paper, we modify a classic mean-field SIR (susceptible–infected–recovered) model, incorporating the influences of sharing and advertising in viral videos. We mathematically analyze the global stability of the system and propose an agent-based modeling approach to evaluate the efficiency of sharing and advertising. We further provide a case study of music videos on YouTube to show the validity of our model.

Abstract Objectives This study aims to elucidate the transcriptomic signatures and dysregulated pathways in patients with Systemic Lupus Erythematosus (SLE), with a particular focus on those persisting during disease remission. Methods We conducted bulk RNA-sequencing of peripheral blood mononuclear cells (PBMCs) from a well-defined cohort comprising 26 remission patients meeting the Low Lupus Disease Activity State (LLDAS) criteria, 76 patients experiencing disease flares, and 15 healthy controls. To elucidate immune signature changes associated with varying disease states, we performed extensive analyses, including the identification of differentially expressed genes and pathways, as well as the construction of protein-protein interaction networks. Results Several transcriptomic features recovered during remission compared to the active disease state, including down-regulation of plasma and cell cycle signatures, as well as up-regulation of lymphocytes. However, specific innate immune response signatures, such as the interferon (IFN) signature, and gene modules involved in chromatin structure modification, persisted across different disease states. Drug repurposing analysis revealed certain drug classes that can target these persistent signatures, potentially preventing disease relapse. Conclusion Our comprehensive transcriptomic study revealed gene expression signatures for SLE in both active and remission states. The discovery of gene expression modules persisting in the remission stage may shed light on the underlying mechanisms of vulnerability to relapse in these patients, providing valuable insights for their treatment.

N,N-dimethylformamide (DMF) is an organic solvent with stable chemical properties and high boiling point. Based on its good solubility, DMF is widely used in synthetic textile, leather, electronics, pharmaceutical and pesticide industries. However, the DMF pollutes the environment and does harm to human liver function, kidney function, and nerve function. Herein, an efficient DMF-degrading strain, DM175A1-1, was isolated and identified as Paracoccus sulfuroxidans. This strain can use DMF as the sole source of carbon and nitrogen. Whole-genome sequencing of strain DM175A1-1 revealed that it has a 3.99-Mbp chromosome a 120-kbp plasmid1 and a 40-kbp plasmid2. The chromosome specifically harbors the dmfA1 and dmfA2 essential for the initial steps of DMF degradation. And it also carries the some part of genes facilitating subsequent methylotrophic metabolism and glutathione-dependent pathway. Through further DMF tolerance degradation experiments, DM175A1-1 can tolerate DMF concentrations up to 10,000 mg/L, whereas the majority of Paracoccus strains could only show degradation activity below 1,000 mg/L. And the efficiency of organic nitrogen conversion to NH3-N in DMF can reach 99.0% when the hydraulic retention time (HRT) is controlled at 5 days. Meanwhile, it showed a significant degradation effect at a pharmaceutical enterprise in Zhejiang Province with high concentration of DMF wastewater. This study provides a new strain Paracoccus sulfuroxidans DM175A1-1 which shows a significant influence on DMF degradation, and reveals the characterization on its DMF degradation. It lays the foundation for the application of biological method in the efficient degradation of DMF in industrial wastewater.

PurposeThis study aimed to assess the clinical efficacy and safety of the combined approach involving hepatic arterial infusion chemotherapy (HAIC) and tyrosine kinase inhibitors (TKIs) for the treatment of advanced hepatocellular carcinoma (HCC).Patients and methodsIn this multicenter retrospective study conducted from January 2020 to December 2023, we reviewed advanced HCC patients who were treated either with HAIC alone or with a combination of HAIC and TKIs. To address initial disparities between the two groups, we employed propensity score matching (PSM). Tumor response evaluation was performed following RECIST 1.1 criteria. We compared survival outcomes, including overall survival (OS), progression-free survival (PFS), and objective response rate (ORR), between the two treatment groups. Safety assessments were conducted for all patients.ResultsFollowing the eligibility review, 138 patients underwent combined treatment with HAIC and TKIs (HT group), while 198 patients received HAIC monotherapy (HA group) and met the inclusion criteria for enrollment in this study. After PSM, 107 patients were assigned to each group. The HT group exhibited a longer median OS (18.0 versus 8.8 months; hazard ratio [HR], 0.52, p < 0.001) compared to the HA group. Median PFS was also longer in the HT group, although without statistical significance (6.0 versus 4.7 months; HR, 0.85, p = 0.265). The HT group demonstrated a higher ORR (41.1% versus 25.2%; p = 0.020). No significant differences were observed between the two groups in the incidence of all adverse events (AEs) or grade 3/4 AEs (any grade: 81.2% for HT versus 78.8% for HA, p = 0.68; grade 3/4: 18.1% for HT versus 13.6% for HA, p = 0.29). Importantly, all AEs were manageable and acceptable. Notably, no grade 5 AEs occurred in either group.ConclusionCombination therapy involving HAIC and TKIs effectively prolonged survival in advanced HCC patients. It represented a preferable alternative to HAIC monotherapy, with manageable safety.

IntroductionThe rapid spread of plasmid-mediated tet(X4) conferring high tigecycline resistance poses a significant threat to public health. Escherichia coli as the most common pathogen which carries tet(X4) has been widely disseminated in China. Thus, comprehensive investigations are required to understand the mechanism of transmission of tet(X4)-positive E. coli.MethodsIn this study, a total of 775 nonduplicate samples were collected in Guangdong, China from 2019 to 2020. We screened for tet(X4)-positive E. coli by PCR amplification and species identification. Furthermore, we analyzed the phylogenetics and genetic context of tet(X4)-positive E. coli through whole-genome sequencing and long-reads sequencing.ResultsOverall, 146 (18.84%) tet(X4)-positive E. coli were isolated, comprising 2 isolates from humans and 144 isolates from pigs. The majority of tet(X4)-positive E. coli exhibited resistance to multiple antibiotics but all of them were susceptible to amikacin and colistin. Phylogenetic analysis showed that ST877, ST871, and ST195 emerged as the predominant sequence types in tet(X4)-positive E. coli. Further analysis revealed various genetic environments associated with the horizontal transfer of tet(X4). Notably, a 100-kbp large fragment insertion was discovered downstream of tet(X4), containing a replicon and a 40-kbp gene cluster for the bacterial type IV secretion system.DiscussionThe high colonization rate of tet(X4)-positive E. coli in animals suggests that colonization as a key factor in its dissemination to humans. Diverse genetic context may contribute to the transfer of tet(X4). Our findings underline the urgent need for controlling the spread of plasmid-mediated tigecycline resistance.

Objective: To investigate the effect of Tremella fuciformis polysaccharides (TFP) on oxidative damage induced by exhaustive exercise in mice and analyze its mechanism. Methods: L6 cells were treated with TFP for 48 hours, and the activity of L6 was detected by CCK-8 method. L6 cells were divided into three groups, including the control, H2O2, and H2O2+TFP groups, and incubated for 48 hours. LA levels were measured by a biochemical analyzer, and Nrf2, NQO1, and HO-1 protein levels were detected by Western blot assay. C57BL/6 mice were randomly divided into four groups (n=10 for each group), including model group and model+TFP group (50, 100, 200 mg/kg). The model+TFP groups were continuously gavaged with TFP, while the model group was gavaged with the same dose of distilled water once a day for two consecutive weeks. Exhaustive exercise was performed 30 minutes after the last administration, the duration of exhaustive swimming exercise was recorded, and enzyme-linked immunosorbent assay (ELISA) was used to detect liver glycogen (LG) and skeletal muscle glycogen (MG). ELISA was used to detect blood urea nitrogen (BUN), lactic acid (LA), ROS, malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), TNF-α, IL-6 and NF-κBp65 levels. We aimed to investigate the improvement effect of Beclin-1 on the cognitive function of mice with Alzheimer"s disease (AD). Human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) were isolated from neonatal umbilical cord tissues. The cells stably overexpressing Beclin-1 were constructed, and those of passages 5, 10, and 15 were used. Forty-eight AD mice were randomly divided into AD, P5 hUC-MSCs, P15 hUC-MSCs (P15MSCs) and OE Beclin 1-P15 hUC-MSCs (OE Bec-P15MSCs) groups (n=12). Overexpressing Beclin-1 significantly enhanced the proliferation and migration of MSCs of different passages, and reduced G0/G1 arrest. The OE Bec-P15MSCs group had the longest total distance, the shortest time in forced swimming test, and the largest total food consumption in novelty suppressed feeding test. Transplanting hUC-MSCs and overexpressing Beclin-1 significantly reduced Aβ deposition. The expressions of P-tau (Ser396), P-tau (Ser231) and P-tau (Ser235) were significantly inhibited by hUC-MSCs transplantation and Beclin-1 overexpression, especially in the OE Bec- P15MSCs group. Abstract Schizophrenia (SZ) is a complex psychiatric neurodevelopmental disorder with uncertain etiology and pathogenesis. Increasing evidence has recognized the key role of the gut microbiota in SZ. However, few studies have investigated the potential link between oral microbiota and SZ. We studied the tongue coating microbiota and inflammatory profiles of 118 elderly SZ patients and 97 age-matched healthy controls using Illumina MiSeq sequencing and multiplex immunoassays, respectively. Reduced α-diversity, along with a significant difference in β-diversity, were observed in patients with SZ. We have identified SZ-associated oral dysbiosis, characterized by increased Streptococcus and Fusobacterium, as well as decreased Prevotella and Veillonella. These differential genera could potentially serve as biomarkers for SZ, either alone or in combination. Additionally, an elevated Streptococcus/Prevotella ratio could indicate oral dysbiosis. These differential genera formed two distinct clusters: Streptococcus-dominated and Prevotella-dominated, which exhibited different correlations with the altered immunological profiles. Furthermore, we also observed disruptions in the inferred microbiota functions in SZ-associated microbiota, particularly in lipid and amino acid metabolism. Our study provides novel insights into the characteristics of tongue coating microbiota and its associations with immunological disturbances in elderly SZ patients, which offer new targets for the diagnosis and treatment of SZ in the elderly.

BackgroundBoth Sophora flavescens (SF) and Astragalus mongholicus (AM) are known for their anti-inflammatory, antifibrotic, and anticancer activities. However, the efficacy, multi-target mechanisms, and therapeutic substances of SF-AM herb pair on the progression of hepatitis-cirrhosis-hepatocellular carcinoma hepatocellular carcinoma (HCC) remain unclear.PurposeTo investigate the efficacy, mechanisms, and potential therapeutic substances of SF-AM herb pair in the progression of hepatitis-cirrhosis-HCC.MethodsFirstly, diethylnitrosamine was used to establish the hepatitis-cirrhosis-HCC model. HE staining and non-targeted metabolomics were used to evaluate the efficacy of SF-AM herb pair. Subsequently, the absorbed components of SF-AM herb pair in the plasma of rats were determined through HPLC-Q-TOF-MS/MS analysis. Flow cytometry, Western blot, and qRT-PCR were then employed to assess CD4+ and CD8+ T lymphocytes, PI3K/Akt signaling pathway-related proteins, and their corresponding mRNAs. Simultaneously, the efficacy and mechanism of SF-AM herb pair on HCC were confirmed by in vitro experiments. Finally, Pearson correlation analysis was performed between pharmacodynamic indicators and in vivo components to identify the potential therapeutic substances of SF-AM herb pair.ResultsSF-AM herb pair can alleviate the pathological damage and reverse metabolic abnormalities in hepatitis, cirrhosis, and HCC rats, particularly during the hepatitis and cirrhosis stages. Pharmacological researches have demonstrated that SF-AM herb pair can increase the proportion of CD8+ T lymphocytes, inhibit the expression of PI3K, Akt, p-Akt, NF-κB p65, NF-κB pp65, and Bcl-2, as well as increase the expression of IκBα, Bax, and cleaved caspase-3. These findings suggest that SF-AM herb pair has the ability to enhance immunity, anti-inflammation and promote apoptosis. Cell experiments have shown that SF-AM herb pair can inhibit the proliferation of HepG2 cell and regulate the PI3K/Akt signaling pathway. Moreover, 23 absorbed prototypical components and 53 metabolites of SF-AM herb pair were identified at different stages of HCC rats. Pearson correlation analysis revealed that matrine, cytisine, wogonoside, and isoastragaloside are potential therapeutic substances in SF-AM herb pair for the prevention and treatment of hepatitis, cirrhosis, and HCC.ConclusionIn summary, this study revealed the efficacy, mechanisms, and potential therapeutic substances of SF-AM herb pair in the hepatitis-cirrhosis-HCC axis and provided a reference for its clinical application.

In response to the problems of the traditional punishment-based coal mine safety management model, the active safety-guided management model, which is based on rewards and a combination of rewards and punishments, was proposed. The model has established a more rigid management system by organizing a wide range of high-frequency advanced safety selection and rewards, establishing a special reward system for production and operation management, and implementing a series of active guidance measures such as safety veto by one vote, public announcement of rewards, exposure of penalties, hearings on unsafe behavior, and rewards for violation investigation and handling, while strictly enforcing the rigidity of the system, strengthening safety management, and cashing in safety penalties. The results show that the implementation of this management model is effective, with a 57.69% reduction in the number of unsafe act outputs in Shangyuquan Mine; after strengthening management, the number of unsafe behavior investigations and punishments in Mindong No.1 Mine increased by about four times, and then rapidly decreased by 72.31%. This management model has greatly reduced all kinds of violations and safety hazards of coal mine units, and greatly improved the enthusiasm of employees and grass-roots managers to take the initiative to safety and act proactively.

Abstract. The intestinal mucus layer is a barrier that separates intestinal contents and epithelial cells, as well as acts as the "mucus layer-soil" for intestinal flora adhesion and colonization. Its structural and functional integrity is crucial to human health. Intestinal mucus is regulated by factors such as diet, living habits, hormones, neurotransmitters, cytokines, and intestinal flora. The mucus layer's thickness, viscosity, porosity, growth rate, and glycosylation status affect the structure of the gut flora colonized on it. The interaction between "mucus layer-soil" and "gut bacteria-seed" is an important factor leading to the pathogenesis of non-alcoholic fatty liver disease (NAFLD). Probiotics, prebiotics, fecal microbiota transplantation (FMT), and wash microbial transplantation are efficient methods for managing NAFLD, but their long-term efficacy is poor. FMT is focused on achieving the goal of treating diseases by enhancing the "gut bacteria-seed". However, a lack of effective repair and management of the "mucus layer-soil" may be a reason why "seeds" cannot be well colonized and grow in the host gut, as the thinning and destruction of the "mucus layer-soil" is an early symptom of NAFLD. This review summarizes the existing correlation between intestinal mucus and gut microbiota, as well as the pathogenesis of NAFLD, and proposes a new perspective that "mucus layer-soil" restoration combined with "gut bacteria-seed" FMT may be one of the most effective future strategies for enhancing the long-term efficacy of NAFLD treatment.

The traditional mode of listening teaching has been unable to fully adapt to the development trend of the times, and it is also difficult to meet the learning needs of students in the information age. In this paper, the multi-objective optimization algorithm is used to improve the teaching-training process to optimize the problems existing in the teaching of listening course, to provide a multi-objective teaching optimization algorithm to imitate the English listening teaching and learning process and to deal with the non-linear constraints in the multi-objective optimization problem utilizing a penalty function. The multi-objective teaching optimization algorithm is used as the basis to establish a diversified teaching model for college English listening courses, and the effectiveness of the teaching is verified by the listening learning performance, students’ multidimensional ability, and learning emotion. The results show that after 16 weeks of diversified teaching, the mean scores of students’ listening test scores in the experimental group increased by 9.52-24.50 points compared with those before the experiment, the mean value of short-term memory scores was the highest at 27.98, and the Cohen’s d value was above 0.861, and the mean scores of the students’ learning emotion test increased by 1.2938 points compared with those before the implementation. Applying the diversified teaching mode to the teaching of college English listening courses can improve students’ English listening performance, eliminate their anxiety and fear, and promote the diversification of college English listening teaching.

Abstract Background The regularity of the menstrual cycle directly affects women’s health. Many studies have focused on menstrual health; however, menstrual cycle regularity-related variations in skin physiological characteristics and skin microbiota have been seldom investigated. Methods To investigate the menstrual cycle regularity-related variations in skin physiological characteristics and skin microbiota of 197 cases of Chinese women aged 18–35 years living in shanghai in 2021. Based on a self-evaluation questionnaire, the volunteers were divided into three groups C1 (those with a regular menstrual cycle), C2 (those with a less regular menstrual cycle) and C3 (those with an irregular menstrual cycle). The physiological parameters of facial skin were measured by non-invasive methods and the skin microbiome was analyzed by 16S rRNA high-throughput sequencing. Results In the C3 group, the hydration content was significantly decreased (p 0.05), indicating that the skin barrier integrity weakened with increased menstrual cycle irregularity. Abstract Background Failure time data frequently occur in many medical studies and often accompany with various types of censoring. In some applications, left truncation may occur and can induce biased sampling, which makes the practical data analysis become more complicated. The existing analysis methods for left-truncated data have some limitations in that they either focus only on a special type of censored data or fail to flexibly utilize the distribution information of the truncation times for inference. Therefore, it is essential to develop a reliable and efficient method for the analysis of left-truncated failure time data with various types of censoring. Method This paper concerns regression analysis of left-truncated failure time data with the proportional hazards model under various types of censoring mechanisms, including right censoring, interval censoring and a mixture of them. The proposed pairwise pseudo-likelihood estimation method is essentially built on a combination of the conditional likelihood and the pairwise likelihood that eliminates the nuisance truncation distribution function or avoids its estimation. To implement the presented method, a flexible EM algorithm is developed by utilizing the idea of self-consistent estimating equation. A main feature of the algorithm is that it involves closed-form estimators of the large-dimensional nuisance parameters and is thus computationally stable and reliable. In addition, an R package LTsurv is developed. Results The numerical results obtained from extensive simulation studies suggest that the proposed pairwise pseudo-likelihood method performs reasonably well in practical situations and is obviously more efficient than the conditional likelihood approach as expected. The analysis results of the MHCPS data with the proposed pairwise pseudo-likelihood method indicate that males have significantly higher risk of losing active life than females. In contrast, the conditional likelihood method recognizes this effect as non-significant, which is because the conditional likelihood method often loses some estimation efficiency compared with the proposed method. Conclusions The proposed method provides a general and helpful tool to conduct the Cox’s regression analysis of left-truncated failure time data under various types of censoring.

Functional electrical stimulation (FES) devices are widely employed for clinical treatment, rehabilitation, and sports training. However, existing FES devices are inadequate in terms of wearability and cannot recognize a user’s intention to move or muscle fatigue. These issues impede the user’s ability to incorporate FES devices into their daily life. In response to these issues, this paper introduces a novel wearable FES system based on customized textile electrodes. The system is driven by surface electromyography (sEMG) movement intention. A parallel structured deep learning model based on a wearable FES device is used, which enables the identification of both the type of motion and muscle fatigue status without being affected by electrical stimulation. Five subjects took part in an experiment to test the proposed system, and the results showed that our method achieved a high level of accuracy for lower limb motion recognition and muscle fatigue status detection. The preliminary results presented here prove the effectiveness of the novel wearable FES system in terms of recognizing lower limb motions and muscle fatigue status.

BackgroundAtaxia-telangiectasia (A-T) is a multisystem genetic disorder involving ataxia, oculocutaneous telangiectasia, and immunodeficiency caused by biallelic pathogenic variants in the ATM gene. To date, most ATM variants have been reported in the Caucasian population, and few studies have focused on the genotype–phenotype correlation of A-T in the Chinese population. We herein present a Chinese patient with A-T who carries compound heterozygous variants in the ATM gene and conducted a literature review for A-T in China.Case presentationA 7-year-old Chinese girl presented with growth retardation, ataxia, medium ocular telangiectasia, cerebellar atrophy, and elevated serum alpha-fetoprotein (AFP) level, which supported the suspicion of A-T. Notably, the serum levels of immunoglobulins were all normal, ruling out immunodeficiency. Exome sequencing and Sanger sequencing revealed two likely pathogenic ATM variants, namely NM_000051.4: c.4195dup (p.Thr1399Asnfs*15) and c.6006 + 1G>T (p.?), which were inherited from her father and mother, respectively. From the Chinese literature review, we found that there was a marked delay in the diagnosis of A-T, and 38.9% (7/18) of A-T patients did not suffer from immunodeficiency in China. No genotype–phenotype correlation was observed in this group of A-T patients.ConclusionThese results extend the genotype spectrum of A-T in the Chinese population and imply that the diagnosis of A-T in China should be improved.

An accumulating body of evidence suggests that the bacterium Akkermansia muciniphila exhibits positive systemic effects on host health, mainly by improving immunological and metabolic functions, and it is therefore regarded as a promising potential probiotic. Recent clinical and preclinical studies have shown that A. muciniphila plays a vital role in a variety of neuropsychiatric disorders by influencing the host brain through the microbiota-gut-brain axis (MGBA). Numerous studies observed that A. muciniphila and its metabolic substances can effectively improve the symptoms of neuropsychiatric disorders by restoring the gut microbiota, reestablishing the integrity of the gut mucosal barrier, regulating host immunity, and modulating gut and neuroinflammation. However, A. muciniphila was also reported to participate in the development of neuropsychiatric disorders by aggravating inflammation and influencing mucus production. Therefore, the exact mechanism of action of A. muciniphila remains much controversial. This review summarizes the proposed roles and mechanisms of A. muciniphila in various neurological and psychiatric disorders such as depression, anxiety, Parkinson’s disease, Alzheimer’s disease, multiple sclerosis, strokes, and autism spectrum disorders, and provides insights into the potential therapeutic application of A. muciniphila for the treatment of these conditions.

Introduction: The China Space Station Telescope (CSST) will enter a low Earth orbit around 2024 and operate for 10 years, with seven of those years devoted to surveying the area of the median-to-high Galactic latitude and median-to-high Ecliptic latitude of the sky. To maximize the scientific output of CSST, it is important to optimize the survey schedule. We aim to evaluate the astrometric capability of CSST for a given survey schedule and to provide independent suggestions for the optimization of the survey strategy.Methods: We first construct the astrometric model and then conduct simulated observations based on the given survey schedule. The astrometric solution is obtained by analyzing the simulated observation data. And then we evaluate the astrometric capability of CSST by analyzing the properties of the astrometric solution.Results: The accuracy of parallax and proper motion of CSST is better than 1 mas (⋅ yr−1) for the sources of 18–22 mag in g band, and about 1∼10 mas (⋅ yr−1) for the sources of 22–26 mag in g band, respectively. The results from real survey could be worse since the assumptions are optimistic and simple.Discussion: Optimizing the survey schedule can improve the astrometric accuracy of CSST. In the future, we will improve the astrometric capability of CSST by continuously iterating and optimizing the survey schedule.

We update the H i surface density measurements for a subset of 17 THINGS galaxies by dealing with the short-spacing problem of the original Very Large Array (VLA) H i images. It is the same sample that Bigiel et al. used to study the relation between H i surface densities and star formation (SF) rate surface densities in galaxy outer disks, which are beyond the optical radius r _25 . For 10 galaxies, the update is based on combining original THINGS VLA H i images with H i images taken by the single-dish FAST in the FEASTS program. The median increment of H i surface densities in outer disks is 0.15–0.4 dex at a given new H i surface density. Several galaxies change significantly in the shape of radial profiles H i surface densities, and seven galaxies are now more than 1 σ below the H i size–mass relation. We update the H i star formation laws in outer disks. The median relation between the H i surface densities and SF rate surface densities based on pixel-wise measurements shifts downward by around 0.15 dex because the H i surface density values shift rightward, and the scatter increases significantly. The scatter of the relation, indicating the star-forming efficiency, exhibits a much stronger positive correlation with the stellar mass surface density than before. Thus, detecting the previously missed, diffuse H i due to the short-spacing problem of the Very Large Array observations is important in revealing the true condition and variation in SF possibly regulated by stellar feedbacks in the localized environment of outer disks.

We present a statistical study of the properties of diffuse H i in 10 nearby galaxies, comparing the H i detected by the single-dish telescope FAST (FEASTS program) and the interferometer Very Large Array (THINGS program), respectively. The THINGS observation missed H i with a median of 23% due to the short-spacing problem of interferometry and limited sensitivity. We extract the diffuse H i by subtracting the dense H i , which is obtained from the THINGS data with a uniform flux-density threshold, from the total H i detected by FAST. Among the sample, the median diffuse-H i fraction is 34%, and more diffuse H i is found in galaxies exhibiting more prominent tidal-interaction signatures. The diffuse H i we detected seems to be distributed in disk-like layers within a typical thickness of 1 kpc, different from the more halo-like diffuse H i detected around NGC 4631 in a previous study. Most of the diffuse H i is cospatial with the dense H i and has a typical column density of 10 ^17.7 –10 ^20.1 cm ^−2 . The diffuse and dense H i exhibit a similar rotational motion, but the former lags by a median of 25% in at least the inner disks, and its velocity dispersions are typically twice as high. Based on a simplified estimation of circumgalactic medium properties and assuming pressure equilibrium, the volume density of diffuse H i appears to be constant within each individual galaxy, implying its role as a cooling interface. Comparing with existing models, these results are consistent with a possible link between tidal interactions, the formation of diffuse H i , and gas accretion.

Strong gravitational lensing is a powerful tool for investigating dark matter and dark energy properties. With the advent of large-scale sky surveys, we can discover strong-lensing systems on an unprecedented scale, which requires efficient tools to extract them from billions of astronomical objects. The existing mainstream lens-finding tools are based on machine-learning algorithms and applied to cutout-centered galaxies. However, according to the design and survey strategy of optical surveys by the China Space Station Telescope (CSST), preparing cutouts with multiple bands requires considerable efforts. To overcome these challenges, we have developed a framework based on a hierarchical visual transformer with a sliding window technique to search for strong-lensing systems within entire images. Moreover, given that multicolor images of strong-lensing systems can provide insights into their physical characteristics, our framework is specifically crafted to identify strong-lensing systems in images with any number of channels. As evaluated using CSST mock data based on a semianalytic model named CosmoDC2, our framework achieves precision and recall rates of 0.98 and 0.90, respectively. To evaluate the effectiveness of our method in real observations, we have applied it to a subset of images from the DESI Legacy Imaging Surveys and media images from Euclid Early Release Observations. A total of 61 new strong-lensing system candidates are discovered by our method. However, we also identified false positives arising primarily from the simplified galaxy morphology assumptions within the simulation. This underscores the practical limitations of our approach while simultaneously highlighting potential avenues for future improvements.

Germination can improve the level of the free amino acid, and further improve the nutritious value of the foxtail millet. However, the effect of germination temperature and time on them is still unclear. The free amino acid of the germinated foxtail millet has been analyzed at different germination temperatures (20, 25, 30, 35, and 40 ℃) and time (0, 6, 12, 18, 24, and 30 h). Results reveal that 21 components of free amino acids increased. The lower temperature or higher temperature is not suitable for the accumulation of the free amino acid. The free amino acids increase to the maximum at the germination temperature of 30 ℃ for 30 h. The total free amino acids and total essential free amino acids can reach (810.7±69.0) and (367.8± 30.2) mg/100g, respectively. Among the amino acids, the contents of alanine and threonine increase to the highest level. The alanine has increased to (120.0±12.0) and (115.0±14.0) mg/100g at the temperature of 30 and 35 ℃, respectively. The threonine has increased to (106.0±1.0) mg/100g at the temperature of 35 ℃. The γ-aminobutyric acid reaches the maximum level at the germination temperature of 35 ℃ and reaches (60.1±1.8) mg/100g. In conclusion, the germination can promote the increase of the free amino acid and adjust the nutrition of the foxtail millet, and the germinated foxtail millet is a potential source for functional food.

Depression is one of the most common psychiatric conditions, characterized by significant and persistent depressed mood and diminished interest, and often coexists with various comorbidities. The underlying mechanism of depression remain elusive, evidenced by the lack of an appreciate therapy. Recent abundant clinical trials and animal studies support the new notion that the gut microbiota has emerged as a novel actor in the pathophysiology of depression, which partakes in bidirectional communication between the gut and the brain through the neuroendocrine, nervous, and immune signaling pathways, collectively known as the microbiota-gut-brain (MGB) axis. Alterations in the gut microbiota can trigger the changes in neurotransmitters, neuroinflammation, and behaviors. With the transition of human microbiome research from studying associations to investigating mechanistic causality, the MGB axis has emerged as a novel therapeutic target in depression and its comorbidities. These novel insights have fueled idea that targeting on the gut microbiota may open new windows for efficient treatment of depression and its comorbidities. Probiotics, live beneficial microorganisms, can be used to modulate gut dysbiosis into a new eubiosis and modify the occurrence and development of depression and its comorbidities. In present review, we summarize recent findings regarding the MGB axis in depression and discuss the potential therapeutic effects of probiotics on depression and its comorbidities.

BackgroundExploring the human microbiome in multiple body niches is beneficial for clinicians to determine which microbial dysbiosis should be targeted first. We aimed to study whether both the fecal and vaginal microbiomes are disrupted in SLE patients and whether they are correlated, as well as their associations with immunological features.MethodsA group of 30 SLE patients and 30 BMI-age-matched healthy controls were recruited. Fecal and vaginal samples were collected, the 16S rRNA gene was sequenced to profile microbiomes, and immunological features were examined.ResultsDistinct fecal and vaginal bacterial communities and decreased microbial diversity in feces compared with the vagina were found in SLE patients and controls. Altered bacterial communities were found in the feces and vaginas of patients. Compared with the controls, the SLE group had slightly lower gut bacterial diversity, which was accompanied by significantly higher bacterial diversity in their vaginas. The most predominant bacteria differed between feces and the vagina in all groups. Eleven genera differed in patients’ feces; for example, Gardnerella and Lactobacillus increased, whereas Faecalibacterium decreased. Almost all the 13 genera differed in SLE patients’ vaginas, showing higher abundances except for Lactobacillus. Three genera in feces and 11 genera in the vagina were biomarkers for SLE patients. The distinct immunological features were only associated with patients’ vaginal microbiomes; for example, Escherichia−Shigella was negatively associated with serum C4.ConclusionsAlthough SLE patients had fecal and vaginal dysbiosis, dysbiosis in the vagina was more obvious than that in feces. Additionally, only the vaginal microbiome interacted with patients’ immunological features.

Introduction and Objectives: The optimal blood pressure (BP) range for patients with metabolic dysfunction-associated fatty liver disease (MAFLD) is currently unknown. This study aimed to explore the relationship between stratified BP levels and MAFLD progression. Patients and Methods: The data of adults who underwent yearly health check-ups were screened to establish both a cross-sectional and a 6-year longitudinal cohort of individuals with MAFLD. BP was classified into the following categories optimal, normal, high-normal, and hypertension. Liver fibrosis was diagnosed with fibrosis-4 (FIB-4) score, nonalcoholic fatty liver disease fibrosis score (NFS), and aspartate aminotransferase-to-platelet ratio index (APRI). Results: A total of 10,232 individuals were included in the cross-sectional cohort. Abstract Background Pulmonary fibrosis is a progressive and usually lethal pulmonary disease. Despite considerable research efforts, no effective therapeutic strategy for pulmonary fibrosis has been developed. NecroX-5 has been reported to possess anti-inflammatory, anti-oxidative and anti-tumor activities. In the present study, we aimed to determine whether NecroX-5 exhibits antifibrotic property in bleomycin (BLM)-induced pulmonary fibrosis. Results We found that pre-treatment with NecroX-5 alleviated inflammatory response, reduced oxidative stress, inhibited epithelial–mesenchymal transition (EMT), and ameliorated pulmonary fibrosis in vivo and in vitro. Our data further indicated that NecroX-5 substantially reduced activation of NLRP3 inflammasome and TGF-β1/Smad2/3 signaling in vivo and in vitro. Additionally, NLRP3 overexpression significantly reversed the protective effects of NecroX-5 in lung epithelial cells exposed to BLM. Conclusions Overall, our results demonstrate the potent antifibrotic properties of NecroX-5 and its therapeutic potential for pulmonary fibrosis.

Distracted driving is one of the main contributors to traffic accidents. In this work, we propose a novel multi-scale local feature fusion network for image-based distracted driver detection. Since the driver is the most important part to infer the distracted driver actions in a single image, our proposed method first detects the driver’s body using person detection. Then capture abundant local body features after a repeated multi-scale feature fusion module. In addition to the features extracted from the whole image, our network also include the important feature of local body feature. The global feature and local feature are finally fused by an OAWS(optimally-weighted strategy). The experimental result shows that our methods achieve comparative performance on our own HY Large Vehicle Driver Dataset and the public AUC Driver Distracted Dataset.

Introduction. Thumb fingertip injuries, often combined with phalanx fracture or torn ligament, are very common. The focus of treatment is on how to rebuild the function and shape of the thumb. Single thumb fingertip pulp injury is more common. However, due to the diverse type of injury, no uniform treatment guideline is described in the literature. Case report. A patient with extensive thumb fingertip pulp incisions, which looked like multiple parallel wounds, was admitted to our Emergency Department. We used the method of bundling to maintain the shape of the fingertip. A single suture to pass through all the incisions was performed and then the suture was tied outside the skin. After that, we bandaged them with sterile bandages. Finally, good contour and fingertip function were restored. Conclusion. We successfully used the bundling method to treat patient with extensive thumb fingertip pulp incisions.

Cordyceps cicadae (Miq.) is an edible fungus with unique and valuable medicinal properties that is commonly used in traditional Chinese medicine, but its anti-aging effects on the skin fibroblast are not well studied. The aim of the present study was to analyze the active components of aqueous C. cicadae extract (CCE), determine the effects of CCE on hyaluronan synthesis in human skin fibroblasts, and explore the underlying mechanisms. The results of this study indicate that CCE was rich in polysaccharides, five alditols (mainly mannitol), eight nucleosides, protein, and polyphenols, which were present at concentrations of 62.7, 110, 8.26, 35.7, and 3.8 mg/g, respectively. The concentration of extract required to inhibit 50% of 2,2-azino-bis (3-ethylbenzothiazo-line-6-sulphonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazil (DPPH) radical scavenging capacities were 0.36 ± 0.03 and 4.54 ± 0.10 mg/mL, respectively, indicating that CCE exhibits excellent antioxidant activities. CCE showed no cytotoxicity to skin fibroblasts at concentrations ≤ 100 μg/mL, and promoted HA synthesis in fibroblasts. Treatment of fibroblast cells with 100 μg/mL CCE enhances the HA content to 1293 ± 142 ng/mL, which is significantly more than that in the non-treatment (NT) group (p = 0.0067). Further, RNA sequencing detected 1,192 differentially expressed genes (DEGs) in CCE-treated fibroblasts, among which 417 were upregulated and 775 were downregulated. Kyoto Encyclopedia of Genes (KEGG) and Genomes pathway (GO) analysis based on RNA sequencing revealed that CCE mainly affected cytokine-cytokine receptor interaction regulated by HA synthesis-related genes. CCE upregulated HA synthase 2 (HAS2), epidermal growth factor (EGF)-related genes, heparin-binding EGF-like growth factor, C-C motif chemokine ligand 2, interleukin 1 receptor-associated kinase 2, and other genes related to fibroblast differentiation and proliferation. CCE downregulated the gene of matrix metallopeptidase 12 (MMP12), which leads to cell matrix loss. RT-qPCR further verified CCE significantly upregulated HAS2 expression and significantly downregulated MMP12 expression, thus promoting hyaluronan synthesis. CCE shows potential as a moisturizer and anti-aging agent in functional foods and cosmetics.

With the rapid advancement of urbanization, the green space well-being (GSWB) of developing countries faces drastic changes and is increasingly threatened. Green and residential spaces are the core elements of GSWB; however, we know very little about the interaction and combination of the two in terms of their effect on GSWB. This study identified the spatiotemporal features of GSWB and critically examined how patterns of residential-green combinations affect GSWB. Based on land-use data for Jinan from 2006 to 2018, and using the spatial measurement tool GeoDa, we found that both green and residential space have increased significantly in central Jinan. At the macro level, the spatial correlation between the two decreased significantly; meanwhile, at the micro level, there are obvious differences in time and geography. This led to differences in the distribution of GSWB between regions with high value and those with low value. We revealed that the development, preservation, and demolition of residential and green spaces influence changes in GSWB. The positive effects on GSWB come from (1) mountain park development policy in built-up areas, (2) theme park development policy in new urban areas, and (3) urban renewal and demolition policy. The negative effects on GSWB come from (1) issues remaining from prior extensive urban development, (2) the replacement of central areas driven by urban branding, and (3) the lack of supervision of nearby facilities for new housing development. To better understand changes in GSWB, it is necessary to consider its internal residential-green spatial collaboration and propose targeted response strategies. This can help to better safeguard the quality of human settlements in the process of urbanization in developing countries.

Both schizophrenia (SZ) and multiple sclerosis (MS) affect millions of people worldwide and impose a great burden on society. Recent studies indicated that MS elevated the risk of SZ and vice versa, whereas the underlying pathological mechanisms are still obscure. Considering that fecal microbiota played a vital role in regulating brain functions, the fecal microbiota and serum cytokines from 90 SZ patients and 71 age-, gender-, and BMI-matched cognitively normal subjects (referred as SZC), 22 MS patients and 33 age-, gender-, and BMI-matched healthy subjects (referred as MSC) were analyzed. We found that both diseases demonstrated similar microbial diversity and shared three differential genera, including the down-regulated Faecalibacterium, Roseburia, and the up-regulated Streptococcus. Functional analysis indicated that the three genera were involved in pathways such as “carbohydrate metabolism” and “amino acid metabolism.” Moreover, the variation patterns of serum cytokines associated with MS and SZ patients were a bit different. Among the six cytokines perturbed in both diseases, TNF-α increased, while IL-8 and MIP-1α decreased in both diseases. IL-1ra, PDGF-bb, and RANTES were downregulated in MS patients but upregulated in SZ patients. Association analyses showed that Faecalibacterium demonstrated extensive correlations with cytokines in both diseases. Most notably, Faecalibacterium correlated negatively with TNF-α. In other words, fecal microbiota such as Faecalibacterium may contribute to the coexistence of MS and SZ by regulating serum cytokines. Our study revealed the potential roles of fecal microbiota in linking MS and SZ, which paves the way for developing gut microbiota-targeted therapies that can manage two diseases with a single treat.

Objective To investigate the effects of miR-493-5p on proliferation, invasion and migration of ovarian cancer cells and underlying mechanism. Methods Culture human normal ovarian epithelial cell lines IOSE80 and ovarian cancer cell lines A2780, SKOV3, OVCAR3, transfected the miR-493-5p over-expression vectors, miR-493-5p suppressed expression vectors and G protein coupled receptor kinase interacting factor 1 (GIT1) over-expression vectors into SKOV3 cells; RT-qPCR was used to detect miR-493-5p and GIT1 mRNA expression; Western blot was used to detect protein expression; MTT assay was used to detect cell viability; Transwell was used to detect cell migration and invasion; dual luciferase report experiment was applied for detection of the targeting relationship between miR-493-5p and GIT1. Results In ovarian cancer cells A2780, SKOV3, OVCAR3, miR-493-5p were weakly expressed, and GIT1 was highly expressed (P＜0.05); After over-expression of miR-493-5p, the proliferation activity, migration and invasion number of ovarian cancer cell SKOV3 were decreased (P＜0.05). miR-493-5p targeted at the regulation of GIT1 expression, and over-expression of GIT1 reduced the inhibitory effect of miR- 493-5p on proliferation, migration and invasion of ovarian cancer cell SKOV3 and expression of p-MEK1/2, p-ERK1/2, pc-Jun and pc-Fos. Conclusions miR-493-5p can inhibit the proliferation, migration and invasion of ovarian cancer cells SKOV3. The mechanism may be related to the targeted regulation of GIT1 and MEK1/2-ERK1/2 pathway, which thus provides some new targets for diagnosis, prevention and treatment for ovarian cancer.

Columnar jointed rock mass with unique joint network structure is a special type of jointed rock mass, which is a binary structure composed of high strength ‘basalt block’ and specific ‘dominant joint’. Relaxation, opening and slippage of columnar jointed surfaces and disintegration of columns occur easily during excavation under high ground stress, which eventually lead to disastrous collapses in columnar jointed rock mass. The construction safety of underground engineering under high stress is severed restricted by it. By combining acoustic wave, borehole camera and other integrated in-situ testing technology and numerical simulation, the mechanism of stress-structural collapse in columnar jointed rock mass under high stress is studied based on multiple columnar jointed rock mass collapses at left bank tailwater connection pipe of Baihetan hydropower station. The proposed controlling measures of excavation and support are also provided. The key of columnar joined rock mass collapse is the redistribution of stress in surrounding rock mass after the excavation and the strong unloading relaxation of columnar joined basalt, which causes the opening of its internal joint surfaces and structural deterioration, result in the disintegration of basalt columns. Thus, the chain catastrophic process of continuous unloading relaxation and progressive collapse of columnar jointed rock mass is induced. The research can provide reference for the prediction and control of deformation and failure of jointed rock masses in underground engineering under high geo-stress.

Abstract Purpose To investigate the frailty status in Chinese cancer patients through establishing a novel prediction algorithm. Methods The percentage of frailty in various age groups, locations, and tumor types in Chinese cancer patients was investigated. The prediction capacity of frailty on mortality of Chinese cancer patients was analysed by the frailty index composing of routine laboratory data (FI‐LAB) accessible from a blood test and calculated as the ratio of abnormal factors to 22 total variables. The establishment of a novel algorithm, MCP (mortality of cancer patients), to predict the 5‐year mortality in Chinese cancer patients was accomplished and the algorithm's prediction capacity was tested in the training and validation sets using receiver operating characteristic (ROC) analysis. Results We found that the risk of death in cancer patients can be successfully identified through FI‐LAB. The univariable and multivariable Cox regression were used to evaluate the effect of frailty on death. In the 5‐year follow‐up, 20.6% of the 2959 participants (age = 55.8 ± 11.7 years; 43.5% female) died, while the mean FI‐LAB score in baseline was 0.23 (standard deviation = 0.13; range = 0–0.73). Frailty (after adjusting for gender, age, and other confounders) directly correlated with an increased risk of death, hazard ratio of 12.67 (95% confidence interval [CI]: 7.19, 22.31), compared to those without frailty. In addition, the MCP algorithm (MCP) = 3.678 × FI‐LAB + 1.575 × sex + 1.779 × first tumor node metastasis staging, presented an area under the ROC (AUC) of 0.691 (95% CI: 0.656–0.726) and 0.648 (95% CI: 0.613–0.684) in the training and validation sets, respectively. Conclusion Frailty as defined by FI‐LAB was common and indicated a significant death risk in cancer patients. Our novel developed algorithm MCP had a passable prediction capacity on 5‐year MCP.

Abstract Background Microbiome big data from population-scale cohorts holds the key to unleash the power of microbiomes to overcome critical challenges in disease control, treatment and precision medicine. However, variations introduced during data generation and processing limit the comparisons among independent studies in respect of interpretability. Although multiple databases have been constructed as platforms for data reuse, they are of limited value since only raw sequencing files are considered. Description Here, we present MetaGeneBank, a standardized database that provides details on sample collection and sequencing, and abundances of genes, microbiota and molecular functions for 4470 raw sequencing files (over 12 TB) collected from 16 studies covering over 10 types of diseases and 14 countries using a unified data-processing pipeline. The incorporation of tools that enable browsing and searching with descriptive attributes, gene sequences, microbiota and functions makes the database user-friendly. We found that the source of specimen contributes more than sequencing centers or platforms to the variations of microbiota. Special attention should be paid when re-analyzing sequencing files from different countries. Conclusions Collectively, MetaGeneBank provides a gateway to utilize the untapped potential of gut metagenomic data in helping fighting against human diseases. With the continuous updating of the database in terms of data volume, data types and sample types, MetaGeneBank would undoubtedly be the benchmarking database in the future in respect of data reuse, and would be valuable in translational science.

Cadmium (Cd) contamination of agricultural soils poses a potential public health issue for humans. Phytoremediation-based accumulating plants are an effective and sustainable technology for Cadmium remediation of contaminated agricultural soil. The rhizosphere microbiome can promote the growth and Cadmium accumulation in hyperaccumulators, but its taxonomic and functional traits remain elusive. The present study used two ecotypes of Sedum alfredii, an accumulating ecotype (AE) and a non-accumulating ecotype (NAE), as model plants to investigate the rhizosphere microbiome assemblages and influence on plant growth under high cadmium conditions. Our results showed that distinct root microbiomes assembled in association with both ecotypes of S. alfredii and that the assemblages were based largely on the lifestyles of the two ecotypes. In addition, we demonstrated that the functions of the microbes inhabiting the rhizosphere soils were closely associated with root-microbe interactions in both ecotypes of S. alfredii. Importantly, our results also demonstrated that the rhizosphere microbiome assembled in the AE rhizosphere soils contributed to plant growth and cadmium uptake under high cadmium conditions through functions such as nitrogen fixation, phosphorus solubilization, indole acetic acid (IAA) synthesis, and siderophore metabolism. However, this phenomenon was not clearly observed in the NAE. Our results suggest that the rhizosphere microbiome plays important roles in biogeochemical nutrient and metal cycling that can contribute to host plant fitness.

Abstract Non‐alcoholic fatty liver disease (NAFLD) with an increasing incidence is one of the leading causes for liver transplantation and cancer, whereas understandings on the onset and development of this disease are quite limited. Nowadays, gut microbial dysregulation has been proposed to play a critical role in the immune‐metabolic disorders in NAFLD patients via gut‐liver axis. Although definitive crosstalk between gut and liver has not been thoroughly depicted, the nanosized bacterial extracellular vesicles (bEVs) capable of transporting multiple kinds of molecules to distant organs of host in intact form via system circulation act as major mediators in host‐bacterial interactions. A recent review has confirmed the associations between bEVs and NAFLD features such as insulin resistance and liver inflammation, whereas the mechanisms involved have not been elucidated. Considering that gut‐liver crosstalk is the main pathway through which bEVs take part in NAFLD pathogenesis, we summarize the recent findings of bEVs in gut‐liver crosstalk by modulating the reshape of gut microbial system, host intestinal barrier integrity, host immune responses and metabolic homeostasis, as well as the potential roles of bEVs in regulating NAFLD progression.

The organic matter in soft clay tends to affect the properties of cement-stabilized soil. The influence degree of different organic matter varies. In this paper, the influence weights and mechanism of the main organic matter components fulvic acid and humic acid on the mechanical properties of cemented soil were investigated. Impacts of FA/HA (fulvic acid/humic acid) values and curing time on the unconfined compressive strength, deformation characteristics, and microstructure of cemented soil were explored through the unconfined compressive strength test and electrical resistivity test. The results show that with the increase of FA/HA, the unconfined compressive strength of cemented soil gradually decreased and the plastic properties enhanced. The increase in curing time changed the stress-strain relationship of cemented soil, and some specimens showed brittle damage. The initial resistivity and structural property parameters of cemented soil gradually decreased with the increasing FA/HA value and increased with the increase of curing time. It revealed the influence law of FA/HA and curing time change on the microstructure of cemented soil. Based on the experimental results, the quantitative relationship equations between FA/HA and curing time and unconfined compressive strength, failure strain, deformation modulus, and resistivity were established.

Fingerprinting,which is an important part of reconnaissance,the first stage of network attack killing chain,is the prerequisite of successful implementation of network attack.The promotion of the concept of active defense,especially deception defense,encourages the defenders to confuse the attackers by means of fingerprint information hiding and obfuscation,thus reducing the effectiveness of their network reconnaissance.Therefore,the defenders can obtain a certain first-mover advantage in the confrontation,and the confrontation of both sides is also advanced to the stage of reconnaissance.Deception is the strategic confrontation between the rational agents of both sides,game theory is a quantitative science to study the conflict and cooperation between rational decision players.It can model the players and actions of various defensive deception,and guide the defenders to make better use of deception technology.In this paper,the dynamic game model with incomplete information is used to analyze the interactive process from reconnaissance to attack.The possible perfect Bayesian Nash equilibrium are analyzed and calculated,and the equilibrium are discussed based on different scenarios.Suggestions are put forward for the defenders to optimize the deceptive strategy to achieve better anti-fingerprinting effect.

Highlights Panax notoginseng saponins could be biotransformed to generate five main metabolites, including GF1, GRh2, GC-K, PPT and PPD, by human gut microbiota. Gut microbiota profiles were significantly different in high protein, high fat and low fat, plant fiber-rich diet-pattern groups. Correlation analysis revealed potential relationships between metabolites and gut microbial species. Bifidobacterium adolescentis and Lactobacillus rhamnosus were selected as a representative species to metabolize PNS for the concerned metabolites.

Depression in childhood negatively affects the growth and development, school performance, and peer or family relationships of affected children, and may even lead to suicide. Despite this, its etiology and pathophysiology remain largely unknown. Increasing evidence supports that gut microbiota plays a vital role in the development of childhood depression. However, little is known about the underlying mechanisms, as most clinical studies investigating the link between gut microbiota and depression have been undertaken in adult cohorts. In present study, a total of 140 school-aged children (6–12 years) were enrolled, including 92 with depression (male/female: 42/50) and 48 healthy controls (male/female: 22/26) from Lishui, Zhejiang, China. Illumina sequencing of the V3–V4 region of the 16S rRNA gene was used to investigate gut microbiota profiles while Bio-Plex Pro Human Cytokine 27-plex Panel was employed to explore host immune response. We found that, compared with healthy controls, children with depression had greater bacterial richness and altered β-diversity. Pro-inflammatory genera such as Streptococcus were enriched in the depression group, whereas anti-inflammatory genera such as Faecalibacterium were reduced, as determined by linear discriminant analysis effect size. These changes corresponded to altered bacterial functions, especially the production of immunomodulatory metabolites. We also identified the presence of a complex inflammatory condition in children with depression, characterized by increased levels of pro-inflammatory cytokines such as IL-17 and decreased levels of anti-inflammatory cytokines such as IFN-γ. Correlation analysis demonstrated that the differential cytokine abundance was closely linked to changes in gut microbiota of children with depression. In summary, key functional genera, such as Streptococcus and Faecalibacterium, alone or in combination, could serve as novel and powerful non-invasive biomarkers to distinguish between children with depression from healthy ones. This study was the first to demonstrate that, in Chinese children with depression, gut microbiota homeostasis is disrupted, concomitant with the activation of a complex pro-inflammatory response. These findings suggest that gut microbiota might play an important role in the pathogenesis of depression in school-aged children, while key functional bacteria in gut may serve as novel targets for non-invasive diagnosis and patient-tailored early precise intervention in children with depression.
