Objective: To demonstrate the antinociceptive and antihypersensitivity mechanisms of Cris-104 (1-{2-[5-(4-fluorophenyl)–1H-pyrazol-4-yl]ethyl}piperidine), a novel selective α4β2* nicotinic acetylcholine receptor (nAChR) agonist, in rodent acute/inflammatory and chronic pain models. Materials and methods: Hot-plate and formalin tests in mice were used to examine Cris-104-induced antinociceptive effects on thermal/inflammatory pain. Cris-104 effects on hypersensitivity, norepinephrine (NE) release in the spinal dorsal horn, and neuronal activity in the locus coeruleus (LC) were examined in rats with lumbar spinal nerve ligation using behavioral, microdialysis, and extracellular recording methods. Cris-104 effects on spontaneous locomotion were examined in an open-field test. Results: Cris-104 induced dose-dependent antinociception effects in hot-plate and formalin tests, and these effects were blocked by the general nAChR antagonist mecamylamine, the selective α4β2* nAChR antagonist dihydro-beta-erythroidine, and the α2-adrenoceptor antagonist yohimbine, but not by the α1-adrenoceptor antagonist prazosin. Systemic and spinally perfused Cris-104 increased NE concentrations in microdialysates from the spinal cord in both normal and SNL rats. Systemic Cris-104 increased neuronal activity in the LC of normal rats. Mecamylamine blocked the effects of Cris-104 on spinal NE release and LC neuronal activity. Systemic Cris-104 did not affect locomotor activity significantly. Conclusion: The α4β2 neuronal nAChR agonist, Cris-104, was effective for treatment of pain via descending noradrenergic inhibition of pain signaling. 35. Extracting bridge frequencies from dynamic responses of two passing vehicles

Abstract

A conventional way to identify bridge frequencies is utilizing vibration data measured directly from the bridge. A drawback with this approach is that the deployment and maintenance of the vibration sensors are generally costly and time-consuming. One of the solutions is in a drive-by approach utilizing vehicle vibrations while the vehicle passes over the bridge. In this approach, however, the vehicle vibration includes the effect of road surface roughness, which makes it difficult to extract the bridge modal properties. This study aims to examine subtracting signals of two trailers towed by a vehicle to reduce the effect of road surface roughness. A simplified vehicle-bridge interaction model is used in the numerical simulation; the vehicle - trailer and bridge system are modeled as a coupled model. In addition, a laboratory experiment is carried out to verify results of the simulation and examine feasibility of the damage detection by the drive-by method.

36. Bridge condition assessment from long-term monitoring by means of Bayesian hypothesis test

Abstract

This study presents an approach to reduce effects of environmental and operational factors on long-term monitoring data of bridges. The Bayesian approach comprising both Bayesian regression and Bayesian hypothesis test is applied to investigate monitoring data of an in-service seven-span plate-Gerber bridge. This study considers time-varying temperature and vehicle weights as environmental and operational factors respectively. Vehicle weights were measured utilizing a bridge weigh-in-motion (BWIM) system installed on the bridge. All data was taken from a healthy bridge, since no damage and deterioration was reported during the monitoring period. Observations through the study demonstrated that considering both temperature and vehicle weight as environmental and operational factors in Bayesian regression led to improved regression results than that considering only temperature. It also showed that monitoring the data observed at a specific time could reduce influence of traffic in long-term monitoring. In the Bayesian hypothesis testing utilizing data from the healthy bridge, the bridge was judged as healthy.

Abstract

This paper investigated dynamic responses of steel monorail bridges incorporating train-bridge interaction under strong earthquakes. Two types of steel monorail bridges were considered in the study: a conventional type with steel track-girder; an advanced type with composite track-girder and simplified lateral bracing system. During strong earthquakes, monorail train was assumed standing on the track-girder of monorail bridges. Observations through the analytical study showed that considering the monorail train as additional mass rather than a dynamic system in numerical modeling overestimated effect of the train load on seismic performance of monorail bridges. Earlier plastic deformations at the end bracing of the girder system absorbed seismic energy and reduced the stress at the pier base.

47. Modal parameter identification of short span bridges under a moving vehicle by means of multivariate AR model

Abstract

This study is intended to investigate the feasibility of health monitoring of short span bridges based on modal parameter identification using the vibration data of the bridge induced by a moving vehicle. The idea of this study using the traffic-induced vibration of short span bridges which is a kind of non-stationary process is that the modal parameters identified repeatedly under a given moving vehicle provide a pattern which may provide useful information to decide current health condition of bridges. The autoregressive (AR) model is adopted for identifying modal parameters such as frequency and damping characteristics of the bridges. Mode shapes of bridges are obtained using multivariate AR model. Feasibility of the modal parameter identification for health monitoring of short span bridges is observed through a moving vehicle laboratory experiment. Pattern changes of identified parameters are observable by comparing the identification results between intact and damaged girders, which encourages the use of the method for long term health monitoring even for short span bridges.

49. Effect of Reveromycin A on experimental tooth movement in OPG-/- mice.

Abstract

In osteoprotegerin-deficient (OPG-/-) mice, osteoclast activity causes bone resorption to outpace bone formation, leading to the development of severe osteoporosis. Such mice are therefore useful for investigating the alveolar bone of patients with osteoporosis. Reveromycin A (RM-A) was recently identified as the unique agent acting on osteoclast activation. This study aimed to analyze the effect of RM-A on the orthodontic treatment of OPG-/- mice (a model of osteoporosis patients with high levels of bone turnover). We examined alveolar bone remodeling in OPG-/- and wild-type (WT) mice during continuous tooth movement. The orthodontic force was induced by means of a Ni-Ti closed-coil spring to move the maxillary first molar for 14 days. RM-A sodium salt (1 mg/kg) was administered intraperitoneally twice daily. In OPG-/- mice, the tooth movement distance was longer, alveolar bone resorption was enhanced, the osteoclast count was greater, and serum alkaline phosphatase and tartrate-resistant acid phosphatase levels were higher relative to those in WT mice. However, the administration of RM-A in OPG-/- mice reduced these parameters. We conclude that RM-A normalizes bone metabolism and loss of alveolar bone during continuous tooth movement in OPG-/- mice. [ABSTRACT FROM AUTHOR]
