2. Çinkonun Laktat Dehidrogenaz Aktivitesi Üzerine İn Vitro Etkisinin İncelenmesi

Çinko, vücuda dışarıdan alınaninorganik bir maddedir ve enzimlerin aktivitelerini düzenlemesi, proteinlerinyapılarına katılıp stabilizasyonlarını sağlaması ve gen ifadesini kontroletmesi nedeniyle organizmada önemli rollere sahiptir. Bu çalışmada çinkosülfatın, glikolizin son basamağında yer alan laktat dehidrogenaz enzimininaktivitesine in vitro etkisinin incelenmesi amaçlanmıştır. Çinko’ nun laktatdehidrogenaz aktivitesi üzerine etkisini incelemek amacıyla; 1mg/ml, 0.5mg/mlve 0.25mg/ml derişimine sahip 3 farklı derişimdeki çinko sülfat varlığındalaktat dehidrogenaz ortama ilave edilmiş ve 5 farklı substrat(piruvat)derişiminde aktivite değerleri spektrofotometrik olarak belirlenmiştir. Km veVmax değerleri saptanmıştır. Laktat dehidrogenazın Vmax değeri 1.43 µmolpiruvat/mg protein/dakika, Km değeri 32.57 mM olarak hesaplanmıştır. Tepkimeortamına 1, 0.5 ve 0.25 mg/ml çinko sülfat eklendiğinde laktat dehidrogenazenzimin sırasıyla %81.2, 77.7 ve 70.7 oranında inhibe olduğu tespit edilmiştir.Sonuç olarak, çinko sülfat derişimindeki artışın, enzimin hem Km hem de Vmaxdeğerlerinde önemli bir azalmaya neden olduğu ve substrat derişimininarttırılsa bile enzim inhibisyonun devam ettiği tespit edilmiştir. Bu durum dainhibisyon tipinin unkompetetif inhibisyon olduğunu göstermektedir.

Bu çalışmada poliamin yolağına ait enzim düzeyleri ve oksidatif stres indeksi 36 akciğer kanseri hastası ve 36 sağlıklı kontrol üzerinde incelenmiştir. Çalışmanın amacı akciğeri kanserinde bu parametrelerin düzeylerinin belirlenerek kontrol grubuna göre farklılıkların belirlenmesidir. Ornitin Dekarboksilaz(ODC) düzeyi (P=0,001, P0,05) ve Ornitin düzeyinde (t=0,17, P>0,05) istatistiksel olarak anlamlı bir fark gözlenmemiştir.Çalışmada akciğer kanserli hastalarda ornitin dekarboksilaz düzeyi, agmatinaz düzeyi, arjinaz enzim aktivitesi, toplam oksidan kapasitesi ve oksidatif stres indeksinde artış gözlendi. Toplam oksidatif kapasite artarken, toplam antioksidan kapasitesinde ise azalma gözlenmiştir. Bu da doğal olarak oksidatif stres indeksinide yükseltmiştir. Çalışmamızın bulgularına dayanarak poliamin düzeyinde oluşabilecek muhtemel artış, poliamin bağımlı olan kanserin seyrini kötüleştiriyor olabilir. Ayrıca oksidatif stresdeki artışın, çalışmamıza dahil edilen hastalarda akciğer kanserinin ortaya çıkışında etkili olup olmadığı bilmesede hastalığın seyri esnasındaki bu artış hastalığın prognozunu kötü yönde etkiliyor olabilir. In this study, enzyme levels of polyamine pathway and oxidative stress index were observed among 36 lung cancer patients and 36 healthy people who are involved to control group. Objective of this study is to determine these parameter levels on lung cancer and then compare differences to control group. Ornithine Decarboxylase(ODC) level (P=0,001, P0,05) and Ornithine level (t=0,17, P>0,05) differences have been found statistically pointless between the patients and control group, In this study, it was observed that ornithine decarboxylase, agmatinase levels, arginase enzyme activity, total oxidant capacity and oxidative stress index among lung cancer patients were decreasing. It was also found that total oxidative capacity was increasing while total antioxidant capacity was decreasing. And this situation naturally led oxidative stress index to increase. According to our study's results, probable increment on polyamine level may aggravate polyamine relation cancer's process. Furthermore, it isn't known wheather increment of oxidative stress can effect occuring lung cancer or not still if this increment happens during the progress of disease this could effect cancer's prognosis in a bad way. 73

WOS: 000461427300003, Objective: SCUBE1 [signal peptide-CUB (complement C1r/C1 s)-EGF (epidermal growth factor)-like domain-containing protein 1] is a novel biochemical marker. SCUBE1 is thought to play roles both in platelet activation and inflammation, which are important stages for the development of acute ischemic stroke (AIS). The purpose of our study was to determine the diagnostic and prognostic values and temporal change of plasma SCUBE1 levels in AIS patients. Materials and methods: Thirty-five patients diagnosed with AIS at the Cumhuriyet University Faculty of Medicine Neurology Department, between June and December 2017, and a control group of 35 healthy volunteers were included. Results: Median first day SCUBE1 value in the patient group was 97.51 ng/mL, and the median 7th day SCUBE1 value was 32.72 ng/mL. Median control group SCUBE1 value was 27.51 ng/mL. The first day SCUBE1 levels were significantly higher than the 7th day and the control group SCUBE1 levels (p = 0.001, p < 0.001, respectively). The ROC analysis showed that SCUBE1 levels above 68.8 ng/mL can be used as an indicator with high sensitivity and specificity for AIS diagnosis. Multivariate analysis revealed that the first day SCUBE1 had significant independent effects on development of AIS. In correlation analysis, plasma SCUBE1 levels showed a significantly positive correlation with lesion volume, NIHSS and MRS values (p < 0.01). Conclusion: First day plasma SCUBE1 values in AIS patients rised at significant levels compared to the control group. SCUBE1 could use both in the early diagnosis and prediction of prognosis and lesion volume of AIS patients.

WOS: 000418497500006, PubMed ID: 28870974, Background: In this study, it was aimed to investigate the relationship between diabetes and breast cancer and the detection of enzymes and ornithine levels in polyamine synthesis pathway in diabetes, breast cancer and diabetic breast cancer patients. Methods: Ornithine, arginine decarboxylase, ornithine decarboxylase and agmatinase levels have been measured in serum of all groups. Ornithine levels were measured spectrophotometrically. Arginine decarboxylase, ornithine decarboxylase and agmatinase levels were determined by ELISA kits. Results: Except for the diabetic group, the levels of enzymes in the polyamine synthesis pathway were increased in all and statistically significant (P < 0.05). The increase in the levels of agmatinase was very important among the enzymes (P < 0.001). Conclusions: Decreased levels of polyamine synthase enzymes in diabetes mellitus were found to be increased patients with breast cancer. Whether and how diabetes-based breast cancer development relates to increase activity of enzymes responsible for polyamine synthesis requires further mechanistic and prospective monitoring studies in larger patient cohorts.

WOS: 000388274800006, The polyamines are essential for rapidly growing cells. Polyamines is synthesized by two reactions which begin from arginine. The aim of this study is to examine the serum levels of the enzymes located in the putrescine synthesis pathway and their relationship with lung cancer. In addition, the study also aim determine the oxidant-antioxidant status and the oxidative stress index. This study was performed on 36 lung cancer patients and 36 healthy controls. The arginase activity and the ornithine level were determined spectrophotometrically while the arginine decarboxylase, ornithine decarboxylase and agmatinase levels were determined by using Enzyme Linked Immunosorbent Assay (Elisa kit). The total antioxidant (TAS) and total oxidant status (TOS) were determined using a commercial kit auto analyser, oxidative stress index (OSI) was subsequently estimated. We observed an increase in ornithine decarboxylase, agmatinase and arginase levels in cancer patients (p< 0.05). however, no significant difference was observed between the two groups with regards to orithine and arginase decarboxylase levels (p> 0.05). In the patient group, the TAS values decreased (p< 0.05), while the TOS values increased (p< 0.05). In addition, the OSI values increased (p< 0.05). Reduced agmatine level can cause an increase in polyamine synthesis and cell proliferation.
