Abstract Aims This study aims to develop and validate an optimal model for predicting worsening heart failure (WHF). Multiple machine learning (ML) algorithms were compared, and the results were interpreted using SHapley Additive exPlanations (SHAP). A clinical risk calculation tool was subsequently developed based on these findings. Methods and results This nested case–control study included 200 patients with chronic heart failure (CHF) from the China‐Japan Friendship Hospital (September 2019 to December 2022). Sixty‐five variables were collected, including basic information, physical and chemical examinations, and quality of life assessments. WHF occurrence within a 3‐month follow‐up was the outcome event. Variables were screened using LASSO regression, univariate analysis, and comparison of key variables in multiple ML models. Eighty per cent of the data was used for training and 20% for testing. The best models were identified by integrating nine ML algorithms and interpreted using SHAP, and to develop a final risk calculation tool. Among participants, 68 (34.0%) were female, with a mean age (standard deviation, SD) of 68.57 (12.80) years. During the follow‐up, 60 participants (30%) developed WHF. N‐terminal pro‐brain natriuretic peptide (NT‐proBNP), creatinine (Cr), uric acid (UA), haemoglobin (Hb), and emotional area score on the Minnesota Heart Failure Quality of Life Questionnaire were critical predictors of WHF occurrence. The random forest (RF) model was the best model to predict WHF with an area under the curve (AUC) (95% confidence interval, CI) of 0.842 (0.675–1.000), accuracy of 0.775, sensitivity of 0.900, specificity of 0.833, negative predictive value of 0.800, and positive predictive value of 0.600 for the test set. SHAP analysis highlighted NT‐proBNP, UA, and Cr as significant predictors. An online risk predictor based on the RF model was developed for personalized WHF risk assessment. Conclusions This study identifies NT‐proBNP, Cr, UA, Hb, and emotional area scores as crucial predictors of WHF in CHF patients. Among the nine ML algorithms assessed, the RF model showed the highest predictive accuracy. SHAP analysis further emphasized NT‐proBNP, UA, and Cr as the most significant predictors. An online risk prediction tool based on the RF model was subsequently developed to enhance early and personalized WHF risk assessment in clinical settings.

Electrical current can effectively improve the plasticity of metallic materials. The tensile deformation behavior of Al alloys under the pulsed electrical current assisted quasi-static unidirectional tension (EAT) has been investigated. Materials under the EAT exhibits periodic electro-softening and strain-hardening behaviors, i.e., a ratchet shape mechanical response. However, establishing a constitutive model to accurately predict the ratchet shape mechanical behavior, especially during the EAT interval, and accurately predicting the strain-hardening behavior of materials are critical issues that need to be solved urgently. In this study, based on the Taylor polycrystalline model, thermal activation theory and dislocation density evolution theory, a two-parameter dislocation density electroplasticity constitutive model with forward and reverse dislocation density evolution was developed to describe the periodic coupling effect of the electro-thermal-mechanical fields during EAT. The tensile deformation behaviors of AA 6061-T6 and AA 7075-T6 under the effect of a pulsed electrical current were quantitatively predicted using the proposed constitutive model. The results show that the correlation coefficient between the predicted and experimental results of the constitutive model can reach 0.84–0.99, implying that the proposed constitutive model can accurately predict the complex electroplasticity behavior of Al alloys during EAT.

Abstract This study investigated the corrosion and oxidation mechanism of the Cr-free Fe-40Ni-15Co superalloy with exposure to a tropical marine atmosphere for one year and a subsequent high temperature of 650 °C for 300 h. The degradation behavior of the samples was evaluated based on surface characterization and analysis. In the tropical marine atmospheres, the corrosion process of the Fe-40Ni-15Co superalloy presented an initial localized type after one month of exposure and gradually developed into uniform. The corrosion products were mainly comprised of Fe2O3 and NiO, suppressing the pitting process. During the high-temperature oxidation process, all samples presented super-parabolic oxidation kinetics. Except for the initial active oxidation due to salt deposits, the corrosion products after atmospheric exposure could significantly enhance the oxidation resistance at 650 °C. An explanation for the degradation mechanism of the Fe-40Ni-15Co superalloy was developed based on the synergistic effects of atmospheric corrosion and high-temperature oxidation.

Patients with rheumatoid arthritis (RA) have an increased risk of ischemic heart failure (IHF), but the shared mechanisms are unclear. This study analyzed RNA sequencing data from five RA and IHF datasets to identify common biological mechanisms and significant biomarkers. One hundred and seventy-seven common differentially expressed genes (CDEGs) were identified, with enrichment analysis highlighting pathways related to sarcomere organization, ventricular myocardial tissue morphogenesis, chondrocyte differentiation, prolactin signaling, hematopoietic cell lineage, and protein methyltransferases. Five hub genes (CD2, CD3D, CCL5, IL7R, and SPATA18) were identified through protein-protein interaction (PPI) network analysis and machine learning. Co-expression and immune cell infiltration analyses underscored the importance of the inflammatory immune response, with hub genes showing significant correlations with plasma cells, activated CD4+ T memory cells, monocytes, and T regulatory cells. Single-cell RNA sequencing (scRNA-seq) confirmed hub gene expression primarily in T cells, activated T cells, monocytes, and NK cells. The findings underscore the critical roles of sarcomere organization, prolactin signaling, protein methyltransferase activity, and immune responses in the progression of IHF in RA patients. These insights not only identify valuable biomarkers and therapeutic targets but also offer promising directions for early diagnosis, personalized treatments, and preventive strategies for IHF in the context of RA. Moreover, the results highlight opportunities for repurposing existing drugs and developing new therapeutic interventions, which could reduce the risk of IHF in RA patients and improve their overall prognosis.

Unified constitutive equations have been developed to model the behaviour of metallic materials under various processing conditions. These constitutive equations usually take the form of a set of ordinary differential equations (ODEs), which must be solved thousands of times in a finite element (FE) process simulation. Thus, an efficient and reliable numerical integration method for large systems is crucial for solving this problem. However, in many constitutive equations, numerical stiffness is often present. This means that the stability requirements, rather than the accuracy, constrain the step size. Therefore, certain numerical methods become unsuitable when the required step size becomes unacceptably small. In this study, a series of mathematical analyses was performed to investigate the difficulties in the numerical integration of three sets of unified viscoplastic/creep constitutive equations. Based on an analysis of the current stiffness assessment methods, a novel index was introduced, that enables an accurate assessment of the stiffness of the ODE-type unified constitutive equations. A computational study was also conducted to benchmark several promising implicit numerical integration methods for viscoplastic/creep constitutive equations. This study can assist researchers in metal forming and other fields in choosing appropriate numerical methods when dealing with stiff ODEs.

Digital image correlation (DIC) outperforms other strain measurement technologies due to its versatility, accuracy, non-contact nature, and capability to capture full-field strains. However, for aluminium alloys like AA6082 under high-temperature and large-deformation conditions, the shiny effects observed in the gauge area degrade the quality of the captured deformation images, leading to failures in the DIC analyses. In this study, the source of the shiny effects was investigated by analysing the evolution of AA6082 specimen surfaces painted with speckle patterns during hot stamping deformation. It was found that the shiny effects were attributed to the exposure of the reflective base material due to paint peeling and cracking. To overcome these problems, the DIC method was improved by developing a novel procedure for generating more effective speckle patterns and utilising a cross-polarisation optical system (XPOL) to mitigate the shiny effects. The improved DIC method was applied to AA6082 in both uniaxial and biaxial tensile tests under diverse hot stamping conditions, along with applications to boron steel for comparison. These improvements significantly reduced the shiny effects and increased the time over which full-field strains can be measured for AA6082, reaching over 96% (up from 75%) of the fracture time in the uniaxial tests and even 100% in the biaxial tests. The improved DIC method enables the full-field strain measurement for aluminium alloys under hot stamping conditions, allowing for the accurate determination of their thermomechanical properties.

Abstract Background Ischemic heart disease is one of the leading causes of mortality worldwide, and thus calls for development of more effective therapeutic strategies. This study aimed to identify potential therapeutic targets for coronary heart disease (CHD) and myocardial infarction (MI) by investigating the causal relationship between plasma proteins and these conditions. Methods A two-sample Mendelian randomization (MR) study was performed to evaluate more than 1600 plasma proteins for their causal associations with CHD and MI. The MR findings were further confirmed through Bayesian colocalization, Summary-data-based Mendelian Randomization (SMR), and Transcriptome-Wide Association Studies (TWAS) analyses. Further analyses, including enrichment analysis, single-cell analysis, MR analysis of cardiovascular risk factors, phenome-wide Mendelian Randomization (Phe-MR), and protein-protein interaction (PPI) network construction were conducted to verify the roles of selected causal proteins. Results Thirteen proteins were causally associated with CHD, seven of which were also causal for MI. Among them, FES and PCSK9 were causal proteins for both diseases as determined by several analytical methods. PCSK9 was a risk factor of CHD (OR = 1.25, 95% CI: 1.13–1.38, P = 7.47E-06) and MI (OR = 1.36, 95% CI: 1.21–1.54, P = 2.30E-07), whereas FES was protective against CHD (OR = 0.68, 95% CI: 0.59–0.79, P = 6.40E-07) and MI (OR = 0.65, 95% CI: 0.54–0.77, P = 5.38E-07). Further validation through enrichment and single-cell analysis confirmed the causal effects of these proteins. Moreover, MR analysis of cardiovascular risk factors, Phe-MR, and PPI network provided insights into the potential drug development based on the proteins. Conclusions This study investigated the causal pathways associated with CHD and MI, highlighting the protective and risk roles of FES and PCSK9, respectively. FES. Specifically, the results showed that these proteins are promising therapeutic targets for future drug development.

With the growing concern about climate issues and the urgent need to reduce carbon emissions, hydrogen has attracted increasing attention as a clean and renewable vehicle energy source. However, the storage of flammable hydrogen gas is a major challenge, and it restricts the commercialisation of fuel cell electric vehicles (FCEVs). This paper provides a comprehensive review of common on-board hydrogen storage tanks, possible failure mechanisms and typical manufacturing methods as well as their future development trends. There are generally five types of hydrogen tanks according to different materials used, with only Type III (metallic liner wrapped with composite) and Type IV (polymeric liner wrapped with composite) tanks being used for vehicles. The metallic liner of Type III tank is generally made from aluminium alloys and the associated common manufacturing methods such as roll forming, deep drawing and ironing, and backward extrusion are reviewed and compared. In particular, backward extrusion is a method that can produce near net-shape cylindrical liners without the requirement of welding, and its tool designs and the microstructural evolution of aluminium alloys during the process are analysed. With the improvement and innovation on extrusion tool designs, the extrusion force, which is one of the most demanding issues in the process, can be reduced significantly. As a result, larger liners can be produced using currently available equipment at a lower cost.

As a promising solution to the growing demand for lightweighting, hot stamping has gained considerable applications in the automotive industry. Over the past few decades, the market for hot stamping has experienced explosive growth, with ongoing advancements offering potential for further expansion of its applications. This paper provides a historical overview of hot stamping alongside an in-depth analysis of future trends. Scientific publications, patents and industrial applications of hot stamping are systematically reviewed, with major developments in materials, processes, tools, and other relevant aspects being highlighted. Through data analysis, the current state of hot stamping is comprehensively depicted, and the trends in the development of hot stamping are revealed. Additionally, the future of extending hot stamping technologies to a broader range of materials is discussed, with suggestions furnished from both academic and industrial perspectives.

Aluminium extrusion profiles are widely utilised due to their potential to enhance energy efficiency and reduce costs. In this paper, two advanced tool designs for a novel sideways extrusion technique, the Unified design and the Guided design, were proposed. These designs facilitate the production of three-dimensional curved profiles with marked asymmetry and large-scale cross-sections. Numerical simulations of the initial designs revealed considerable variations in bending shapes compared to the target profile. Employing the Taguchi method combined with grey relational analysis, the study identified that the extrudate's bending shape are influenced by the die orifice position and the speed ratio of two rams with different sensitivities. Following this, optimal designs were derived for both design types. For the particular profile in this study, the simulation results showed that the optimal Unified design permitted better welding quality and lower extrusion force. However, the optimal Guided design achieved a bending shape more aligned with the target shape. It can also provide enhanced stability in the extrusion system and higher production yield compared to the optimal Unified design.

Biodegradable zinc (Zn)-based composites are considered to be the next generation of promising orthopedic implant materials due to their degradability and the multiple functionality of combining metal and ceramic phases. However, its clinical bone-implant application is seriously limited due to their poor interface binding ability and mechanical properties. Herein, we developed the biodegradable in-situ Zn–3Cu–10ZnO composite through powder metallurgy using Zn and copper (Cu) powder combined with hot forging, followed by the high-pressure solidification (HPS) process for bone-implant application. Microstructural characterization showed that the HPS process can densify composite, form a uniform distribution of fine ZnO particle, and increase Cu atom solid solubility in the Zn matrix. Mechanical test revealed that the HPS composite exhibited a high compression performance with a compressive yield strength of 340.8 MPa, an ultimate compressive strength of 514.2 MPa, and a failure strain of ≥70%. The HPS composite exhibited the lowest corrosion rate of 25.8 μm/y and 44.3 μm/y measured by electrochemical corrosion and immersion tests, respectively, among these composites. Moreover, the diluted extracts of HPS composite with a concentration of ent% showed a higher cytocompatibility among these composites and better antimicrobial capability than those of the as-cast pure Zn. Accordingly, the HPS Zn–3Cu–10ZnO composite is expected to be a potential biodegradable material for orthopedic applications.

Abstract Purpose The aim of this retrospective study was to examine the clinical outcomes and complications of proximal femur reconstruction (PFR) combined with total hip arthroplasty (THA) in patients with high hip dislocation secondary to septic arthritis (SA). Methods Between September 2016 to September 2021, we performed a series of 15 consecutive PFR combined with THA on patients with high dislocation of the hip secondary to SA, of these,12 hips were reviewed retrospectively, with a mean follow-up of 2.5 years (range, 1.5-6 years). The mean age of the patients at the time of surgery was 52 years (range, 40–70 years). Results All patients were followed up. At 1-year postoperative follow-up, the median HHS increased from 32.50 preoperatively to 79.50 postoperatively. The median VAS decreased from 7 before surgery to 2 at 1 year after surgery. The median LLD reduced from 45 mm preoperatively to 8 mm at 1 year after surgery. The mean operative time 125 ± 15 min (range 103-195 min). Mean estimated blood loss was500 ± 105ml (range 450–870 ml). Mean hospital days 9.5 days (range 6–15 days). Two patients developed nerve injuries that improved after nutritional nerve treatment. One patient had recurrent postoperative dislocation and underwent reoperation, with no recurrence dislocation during the follow-up. There were no cases of prosthesis loosening during the follow-up period. One patient developed acute postoperative periprosthetic joint infection (PJI) that was treated with Debridement, Antibiotics and Implant Retention (DAIR) plus anti-infective therapy, with no recurrence during 2 years of follow-up. Conclusion This study indicates PFR combined with THA shows promise as a technique to manage high hip dislocation secondary to SA, improving early outcomes related to pain, function, and limb length discrepancy.

Developing reusable and easy-to-operate biocatalysts is of significant interest in biodiesel production. Here, magnetic whole-cell catalysts constructed through immobilizing recombinant Escherichia coli cells (containing MAS1 lipase) into Fe3O4–chitosan magnetic microspheres (termed MWCC@MAS1) were used for fatty acid methyl ester (FAME) production from waste cooking oil (WCO). During the preparation process of immobilized cells, the effects of chitosan concentration and cell concentration on their activity and activity recovery were investigated. Optimal immobilization was achieved with 3% (w/v) chitosan solution and 10 mg wet cell/mL cell suspension. Magnetic immobilization endowed the whole-cell catalysts with superparamagnetism and improved their methanol tolerance, enhancing the recyclability of the biocatalysts. Additionally, we studied the effects of catalyst loading, water content, methanol content, and reaction temperature on FAME yield, optimizing these parameters using response surface methodology and Box–Behnken design. An experimental FAME yield of 89.19% was gained under the optimized conditions (3.9 wt% catalyst loading, 22.3% (v/w) water content, 23.0% (v/w) methanol content, and 32 °C) for 48 h. MWCC@MAS1 demonstrated superior recyclability compared to its whole-cell form, maintaining about 86% of its initial productivity after 10 cycles, whereas the whole-cell form lost nearly half after just five cycles. These results suggest that MWCC@MAS1 has great potential for the industrial production of biodiesel.

To solve the problems of poor mechanical properties and local corrosion caused by the coarse dendrites grain and the second phase of as-cast (AC) Zn–Li alloy, AC Zn-0.5Li alloys were treated by ultra-high pressure (3 GPa) solid solution-aging treatment (UHPS-AT) at 375 °C. The microstructure, mechanical, corrosion behavior, cell compatibility, and antibacterial properties of UHPS-AT Zn-0.5Li alloys were characterized and evaluated. After the UHPS-AT treatment, the TEM images showed the presence of large Zn/β-LiZn4 layered grain inside UHPS-AT alloy, and the dendritic grain and coarse second phase disappeared. The mechanical properties and wear resistance of Zn-0.5Li alloys were improved compared with AC alloys after UHPS-AT treatment. However, the compressive yield strength of UHPS Zn-0.5Li alloy decreased after aging, indicating that the strengthening effect of solid solution is better than that of the second phase precipitation, which was also proved by theoretical calculation. A dense passivation film was formed on the surface of the UHPS-AT sample, hindering further corrosion of the alloy, resulting in the change of the corrosion mechanism from local corrosion in the as-cast state to uniform corrosion in the UHPS-AT state. Cytotoxicity assays with Zn-0.5Li alloys showed that extracts at concentrations below 12.5% were not cytotoxic. Moreover, the Zn-0.5Li alloy exhibited good antibacterial properties against E. coli bacteria.

Abstract Purpose To use 18 F-FDG microPET dynamic imaging to preliminarily identify the changes of myocardial glucose metabolism corresponding to different functional phenotypes of diabetic cardiomyopathy (DCM) in mice and elucidate their relationships. Methods Left ventricular function was measured by echocardiography in C57BL/KsJ-db/db (db/db) mice and their controls at 8, 12, 16, and 20 weeks of age to divide DCM stages and functional phenotypes. Myocardial histopathology was used to verify the staging accuracy and list-mode microPET dynamic imaging was conducted. The myocardial metabolic rate of glucose (MRglu) and the glucose uptake rate constant (Ki) were derived via Patlak graphical analysis, and the differences in myocardial glucose metabolism levels in different DCM stages were compared. The key proteins involved in myocardial glucose metabolism signaling pathway were analyzed by Western blotting to elucidate the underlying mechanism of abnormal glucose metabolism in DCM. Results Compared with the controls, the ratio of early diastolic transmitral flow velocity to early diastolic mitral annular tissue velocity (E/e’) of db/db mice was significantly increased from the age of 12 weeks, while the left ventricular ejection fraction (LVEF) was significantly decreased from the age of 16 weeks (all P 0.05). MRglu and SUV were moderately negatively correlated with the E/e’ ratio (r=-0.539 and − 0.512, P = 0.007 and 0.011), which were not significantly correlated with LVEF (P > 0.05). Meanwhile, Ki was not significantly correlated with LVEF or E/e’ ratio. The decreased expression of glucose transporter (GLUT) -4 in db/db mice preceded GLUT-1 and was accompanied by decreased phosphorylated AMP-activated protein kinase (p-AMPK) expression. Myocardial MRglu, Ki and SUV were significantly positively correlated with the expression of GLUT-4 (MRglu: r = 0.537; Ki: r = 0.818; SUV: r = 0.491; P = 0.000 ~ 0.046), but there was no significant correlation with GLUT-1 expression (P = 0.238 ~ 0.780). Conclusions During the progression of DCM, with the changes of left ventricular functional phenotype, abnormal and dynamic changes of myocardial glucose metabolism can occur in the early stage.

Manipulating molecular self-assembly and disassembly in vivo may permit temporal control of drug delivery and release. Here, the authors report a fluorogenic cisplatin prodrug for cancer theranostics by leveraging stimuli-triggered in situ self-assembly and intracellular disassembly processes.

Lightweight curved profiles are widely utilised in the transportation industry considering the increasing need for improving aerodynamic efficiency, aesthetics and cutting emissions. In this paper, curved AZ31 Mg alloy profiles were manufactured in one operation by a novel process, differential velocity sideways extrusion (DVSE), in which two opposed rams were used. Effects of extrusion temperature and velocity (strain rate) on curvature, microstructure, and mechanical properties of the formed profiles were examined. Profile curvature was found to be more readily controlled by the velocity ratio of the bottom ram v2 to the top ram v1, whereas extrusion temperature (T = 250, 300, 350 °C) and extrusion velocity (v1 = 0.1, 1 mm/s) slightly affect curvature for a given velocity ratio. A homogeneous microstructure with equiaxed grains (∼ 4.5 µm) resulted from dynamic recrystallisation (DRX), was observed after DVSE (v2/v1 = 1/2) at 300 °C and v1 = 0.1 mm/s, where the initial billet had an average grain size of ∼25 um. Increasing extrusion temperature leads to grain growth (∼ 5 µm) at 350 °C and v1 = 0.1 mm/s. DRX is incomplete at the relatively low temperature of 250 °C (v1 = 0.1 mm/s), and higher strain rate with v1 = 1 mm/s (T = 300 °C), resulting in inhomogeneous bi-modal necklace pattern grains ranging in size around 1-25 µm for the former and 2-20 µm for the latter. Grain refinement is attributed to DRX during the severe plastic deformation (SPD) arising in DVSE, and initiates at the prior boundaries of coarse grains in a necklace-like manner. Compared with the billet, micro-hardness and ultimate tensile strength of the profiles have been enhanced, which is compatible with grain refinement. Also, an obvious increase in tensile ductility was found. However, yield strength slightly decreases except for the complete DRXed case (300 °C, v1 = 0.1 mm/s), where a slightly higher value was found, indicating strengthening by grain refinement is greater than softening caused by texture modification. The initial billet had a strong basal texture wherein the {0002} basal plane is oriented parallel to the extrusion direction (‘hard’ orientation), while DVSE results in the profiles having weak basal textures and the {0002} basal plane oriented ∼ 5-10o to the extrusion direction (i.e. towards the orientation for easier slip). This significantly modified texture contributes to the softening of the profiles in the extrusion direction, in which tensile tests were performed, and the related elongation improvement.

Despite advances in immunotherapy for the treatment of cancers, not all patients can benefit from programmed cell death ligand 1 (PD-L1) immune checkpoint blockade therapy. Anti-PD-L1 therapeutic effects reportedly correlate with the PD-L1 expression level; hence, accurate detection of PD-L1 expression can guide immunotherapy to achieve better therapeutic effects. Therefore, based on the high affinity antibody Nb109, a new site-specifically radiolabeled tracer, 68Ga-NODA-cysteine, aspartic acid, and valine (CDV)-Nb109, was designed and synthesized to accurately monitor PD-L1 expression. The tracer 68Ga-NODA-CDV-Nb109 was obtained using a site-specific conjugation strategy with a radiochemical yield of about 95% and radiochemical purity of 97%. It showed high affinity for PD-L1 with a dissociation constant of 12.34 ± 1.65 nM. Both the cell uptake assay and positron emission tomography (PET) imaging revealed higher tracer uptake in PD-L1-positive A375-hPD-L1 and U87 tumor cells than in PD-L1-negative A375 tumor cells. Meanwhile, dynamic PET imaging of a NCI-H1299 xenograft indicated that doxorubicin could upregulate PD-L1 expression, allowing timely interventional immunotherapy. In conclusion, this tracer could sensitively and dynamically monitor changes in PD-L1 expression levels in different cancers and help screen patients who can benefit from anti-PD-L1 immunotherapy.

Zinc (Zn) alloys have been paid increasing attention in the field of biodegradable implantable materials due to their moderate degradation rate compared to magnesium (Mg) and iron (Fe) alloys. In this study, Zn–4Ag, Zn–4Ag–Cu and Zn–4Ag–Mn were prepared to investigate the effects of Cu and Mn elements on the microstructure and properties of Zn–4Ag alloys, and the addition of Cu and Mn improved the mechanical properties and degradation rate of Zn–4Ag alloys. The tensile strength of Zn–Ag after rolling was increased from 141.9 MPa to 168.4 MPa and 206.3 MPa after the addition of Cu and Mn. The degradation rate of Zn–4Ag increased from 0.17 mm/year to 0.22 mm/year and 0.39 mm/year in the first 5 days after the addition of Cu and Mn. The cytotoxicity testing showed good biocompatibility for human umbilical vein endothelial cells (HUVEC) in as-rolled Zn–4Ag–Mn diluted 4 times, and its cytotoxicity showed grade 0 or 1 toxicity with the cell survival rate of 83.7%. The antibacterial experiment showed the highest antibacterial rate for methicillin-resistant Staphylococcus aureus (MRSA) in as-rolled Zn–4Ag–Cu with the antibacterial rate of 90.3%.

Background: Hypertension, a major cardiovascular risk factor, severely impacts patients’ quality of life. Qiangli Dingxuan tablet (QDT) is a formally approved Chinese patent medicine, which has been widely used as an adjunctive treatment for hypertension. This study aimed to investigate the antihypertensive efficacy and safety of QDT combined with amlodipine besylate in patients with essential hypertension.Methods: In this randomized, double-blind, placebo-controlled, parallel-group, multicenter trial conducted in China, patients diagnosed with grade 1 to 2 essential hypertension were randomly assigned in a 1:1 to the treatment of QDT or placebo for 12 weeks, alongside their ongoing treatment with amlodipine besylate. The primary outcome was the change in office blood pressure (BP) from baseline to 12 weeks. In addition, safety analysis included the assessment of vital signs and laboratory values.Results: At baseline, 269 patients were randomly assigned to the QDT group (n = 133) or the placebo group (n = 136), and there were no significant differences in baseline characteristics between the two groups. The primary outcome based on the full analysis set from baseline to 12 weeks showed that the mean difference in the change of office systolic BP reduction between the two groups was 6.86 mmHg (95%CI, 4.84 to 8.88, p < 0.0001), for office diastolic BP, the mean difference in the change of office diastolic BP reduction between the two groups was 4.64 mmHg (95%CI, 3.10 to 6.18, p < 0.0001). In addition, traditional Chinese medicine symptom scores were significantly decreased in the QDT group compared with the placebo group. No severe adverse events attributable to QDT were reported.Conclusion: The combination of QDT and amlodipine besylate demonstrates superior efficacy compared to amlodipine besylate monotherapy in the management of essential hypertension. QDT shows potential as an adjunctive treatment for essential hypertension. This study investigates the evolution of the structural, volumetric and water retention behaviors of a compacted clay during soaking and desiccation considering the influences of freeze-thaw (FT) cycles and saline intrusion. Compacted specimens were subjected to different FT cycles and then submerged in NaCl solution with different concentrations to facilitate the saline intrusion and measure the swelling behaviors. Shrinkage curve and filter paper tests were thereafter performed to reveal the clay's volumetric and water-retention characteristics during desiccation. Mercury intrusion porosimetry and field emission scanning electron microscopy tests were conducted to observe the evolution of the clay's microstructure. Experimental results show that the clay's micropores decrease and macropores increase after FT cycles, which is associated with the migration of water, growth of ice crystals, and development of FT-induced cracks during FT cycles. Similar observations were obtained from specimens after the saline intrusion, which is attributed to the osmotic and osmotically-induced consolidation. FT-induced cracks significantly reduce the clay's swelling and shrinkage potentials. FT cycles result in the shrinkage of micropores which leads to a reduction in the water retention capacity in the low suction range (capillary regime). The salinization suppresses the swelling of the clay and prolongs its primary and secondary swelling stages. The shrinkage potential initially increases and then decreases with increasing saline concentration. Salinization has significant influences on the osmotic suction and thus alters the clay's water-retention curves in terms of total suction. It demonstrates little impact on the clay's water-retention curves in terms of matric suction.

BackgroundPatients with chronic obstructive pulmonary disease (COPD) often present with atrial fibrillation (AF), but the common pathophysiological mechanisms between the two are unclear. This study aimed to investigate the common biological mechanisms of COPD and AF and to search for important biomarkers through bioinformatic analysis of public RNA sequencing databases.MethodsFour datasets of COPD and AF were downloaded from the Gene Expression Omnibus (GEO) database. The overlapping genes common to both diseases were screened by WGCNA analysis, followed by protein-protein interaction network construction and functional enrichment analysis to elucidate the common mechanisms of COPD and AF. Machine learning algorithms were also used to identify key biomarkers. Co-expression analysis, “transcription factor (TF)-mRNA-microRNA (miRNA)” regulatory networks and drug prediction were performed for key biomarkers. Finally, immune cell infiltration analysis was performed to evaluate further the immune cell changes in the COPD dataset and the correlation between key biomarkers and immune cells.ResultsA total of 133 overlapping genes for COPD and AF were obtained, and the enrichment was mainly focused on pathways associated with the inflammatory immune response. A key biomarker, cyclin dependent kinase 8 (CDK8), was identified through screening by machine learning algorithms and validated in the validation dataset. Twenty potential drugs capable of targeting CDK8 were obtained. Immune cell infiltration analysis revealed the presence of multiple immune cell dysregulation in COPD. Correlation analysis showed that CDK8 expression was significantly associated with CD8+ T cells, resting dendritic cell, macrophage M2, and monocytes.ConclusionsThis study highlights the role of the inflammatory immune response in COPD combined with AF. The prominent link between CDK8 and the inflammatory immune response and its characteristic of not affecting the basal expression level of nuclear factor kappa B (NF-kB) make it a possible promising therapeutic target for COPD combined with AF.

For the first time, a novel dithiomaleimides (DTM) based tetra-antennary GalNAc conjugate was developed, which enable both efficient siRNA delivery and good traceability, without incorporating extra fluorophores. This conjugate can be readily constructed by three click-type reactions, that is, amidations, thiol-dibromomaleimide addition and copper catalyzed azide–alkyne cycloaddition (CuAAC). And it also has comparable siRNA delivery efficiency, with a GalNAc L96 standard to mTTR target. Additionally, due to the internal DTMs, a highly fluorescent emission was observed, which benefited delivery tracking and reduced the cost and side effects of the extra addition of hydrophobic dye molecules. In all, the simple incorporation of DTMs to the GalNAc conjugate structure has potential in gene therapy and tracking applications.

Reduction in vehicle weight can significantly reduce energy use in human transportation. However, to gauge efficiency, energy use and weight for a particular vehicle should be related to the number of people being transported while currently there is no convenient means to assess this. Here we statistically analyse the weight, energy consumption, carrying capacity and occupancy level for automobiles, buses, high-speed trains and aircraft. Based on the analysis and inspired by the medical body mass index (BMI), we have proposed a vehicle mass index (VMI), defined as I=A(WtvWtp)n, for the first time enables energy efficiency assessment of different transportations on a global scale, where n a weight sensitivity parameter and A the energy efficiency constant of a theoretically weightless vehicle. We show the VMI ranges and conclude the significant vehicle weight reduction windows to achieve their index lower limits. The possible limits for the VMI and the associated A and n values are also assessed. The concept of VMI could form the basis of a worldwide standard, useful in the current drive for a greener economy.

Hypertension is a major cardiovascular risk factor, which seriously affects the quality of life of patients. Banxia Baizhu Tianma Decoction (BXD) is a Chinese herbal formula that is widely used to treat hypertension in China. This study aimed to evaluate the efficacy and potential mechanism of BXD for hypertension by meta-analysis and network pharmacology. Meta-analysis was performed to explore the efficacy and safety of BXD combined with conventional treatment for hypertension. Network pharmacology was used to explore the molecular mechanism of BXD in antihypertension. A total of 23 studies involving 2,041 patients were included. Meta-analysis indicated that compared with conventional treatment, combined BXD treatment was beneficial to improve clinical efficacy rate, blood pressure, blood lipids, homocysteine, endothelial function, inflammation, and traditional Chinese medicine symptom score. In addition, meta-analysis indicated that BXD is safe and has no obvious adverse reactions. Network pharmacology showed that the antihypertensive targets of BXD may be AKT1, NOS3, ACE, and PPARG. The antihypertensive active ingredients of BXD may be naringenin, poricoic acid C, eburicoic acid, and licochalcone B. Due to the poor methodological quality of the Chinese studies and the small sample size of most, the analysis of this study may have been affected by bias. Hypertension is the most common chronic disease. A large amount of evidence showed that traditional Chinese medicine (TCM) method of tonifying kidney (TK) combined with routine treatment is more effective and safer in the treatment of hypertension. This study integrated meta-analysis, data mining, and network pharmacology to explore the efficacy and potential mechanisms of TK in the treatment of hypertension. Meta-analysis was performed to explore the efficacy and safety of TK combined with routine treatment in the treatment of hypertension. Data mining was used to screen the core herbs of the TK. Network pharmacology was used to predict the antihypertensive mechanism of TK core herbs. A total of 18 studies with 2,024 patients were included in this study. Meta-analysis showed that TK combined with routine treatment was superior to routine treatment alone in lowering blood pressure (systolic and diastolic blood pressures), lowering blood lipids (total cholesterol, triglyceride, low-density lipoprotein cholesterol), improving vascular endothelial functions (nitric oxide, endothelin) and TCM symptoms (headache dizziness, soreness, and weakness of waist and knees). In addition, TK was safe and has no obvious adverse reactions. Data mining showed that the core herbs of TK were Eucommia ulmoides Oliv. (Duzhong), Vitex negundo L. (Huangjing), Taxillus chinensis (DC.) Danser (Sangjisheng), Ligustrum lucidum W.T.Aiton (Nuzhenzi), Astragalus mongholicus Bunge (Huangqi), Rehmannia glutinosa (Gaertn.) DC. (Shudihuang). Network pharmacology predicted that core herbs antihypertensive components were oleanolic acid, ursolic acid, and civetone, and the antihypertensive targets were NOS3, NOS2, MMP9, TNF, PTGS2, HMOX1. In addition, the antihypertensive targets were enriched in cGMP-PKG signaling pathway, calcium signaling pathway, aldosterone-regulated sodium reabsorption, HIF-1 signaling pathway. In conclusion, TK combined with routine treatment for hypertension is effective and safe. The mechanism of TK may be related to GMP-PKG signaling pathway, calcium signaling pathway, aldosterone-regulated sodium reabsorption. Additive Manufacturing (AM) processes, also known as 3D printing, enable geometrically complex parts to be produced layer by layer on the basis of three-dimensional (3D) data generated either by scanning physical objects or using design software. Compared to conventional manufacturing processes, AM offers the elimination of production steps, allowing rapid and relatively easy prototyping of physical objects from 3D model designs, and reproduction of existing objects. Over the last two decades, AM processes have become widespread for the manufacturing of complex-shaped components in numerous industrial sectors, one of their main areas of application being in the aerospace industry. This sector makes extensive use of high-strength aluminium alloys because of their high strength-to-weight and stiffness-to-weight ratios and excellent machinability. However, the applicability of AM processes to high-strength aluminium alloys is still limited by the presence of several types of non-negligible issues and defects in additively manufactured (AMed) aluminium components. Over the years, significant research efforts have been directed at minimising or eliminating these defects and thereby expanding the range of applications of AM in high-strength aluminium alloys. This paper reviews the state of the art in AM of high-strength aluminium alloys for aerospace. The focus is on defects and issues in AMed 2xxx and 7xxx series alloys and recent developments in novel hybrid AM processes to minimise or eliminate the defects.

Rheological behavior and dynamic softening mechanism of high-strength AA7075 sheet were investigated by performing isothermal uniaxial tensile tests at deformation temperatures of 573−723 K and strain rates of 0.01–10 s−1. On the basis of the obtained flow stress, response surface models for the ultimate tensile stress (UTS) and fracture strain were developed as a function of temperature and strain rate, respectively. Error analysis certified the reliability of the models. Besides, a constitutive model coupling with dislocation evolution was built to predict the deformation behavior of the alloy, and the dislocation evolution law was revealed based on the established constitutive model. In addition, the dynamic softening mechanism of the alloy during hot deformation was exposed according to the Zener-Hollomon parameter map, microstructural observation and apparent activation energy. Owing to precipitates pinning effect on the motion of dislocation and migration of subgrain boundaries, the dominant softening mechanism is concluded to be dynamic recovery (DRV), relating to cross-slip of screw dislocation.

Mitochondria play a key role in cellular metabolism. Mitochondrial dynamics (fusion and fission) and mitophagy, are critical to mitochondrial function. Fusion allows organelles to share metabolites, proteins, and mitochondrial DNA, promoting complementarity between damaged mitochondria. Fission increases the number of mitochondria to ensure that they are passed on to their offspring during mitosis. Mitophagy is a process of selective removal of excess or damaged mitochondria that helps improve energy metabolism. Cardiometabolic disease is characterized by mitochondrial dysfunction, high production of reactive oxygen species, increased inflammatory response, and low levels of ATP. Cardiometabolic disease is closely related to mitochondrial dynamics and mitophagy. This paper reviewed the mechanisms of mitochondrial dynamics and mitophagy (focus on MFN1, MFN2, OPA1, DRP1, and PINK1 proteins) and their roles in diabetic cardiomyopathy, myocardial infarction, cardiac hypertrophy, heart failure, atherosclerosis, and obesity.

Immunotherapy blocking programmed cell death protein 1/programmed death ligand 1 (PD-1/PD-L1) pathway has achieved great therapeutic effect in the clinic, but the overall response rate is not satisfactory. Early studies showed that response to treatment and overall survival could be positively related to PD-L1 expression in tumors. Therefore, accurate measurement of PD-L1 expression will help to screen cancer patients and improve the overall response rate. A small molecular positron emission tomography (PET) probe [18F]LP-F containing a biphenyl moiety was designed and synthesized for measurement of PD-L1 expression in tumors. The PET probe [18F]LP-F was obtained with a radiochemical yield of 12.72 ± 1.98%, a radiochemical purity of above 98% and molar activity of 18.8 GBq/μmol. [18F]LP-F had good stability in phosphate buffer saline (PBS) and mouse serum. In vitro assay indicated that [18F]LP-F showed moderate affinity to PD-L1. Micro-PET results showed that the tumor accumulation of [18F]LP-F in A375 tumor was inferior to that in A375-hPD-L1 tumor. All the results demonstrated that [18F]LP-F could specifically bind to PD-L1 and had a potential application in non-invasive evaluation of PD-L1 expression in tumors.

Solitary wave propagating over a bottom-mounted barrier is simulated using the Incompressible Smoothed Particle Hydrodynamics (ISPH) method in order to study the generation and transport of turbulence associated with flow separation around submerged structures. For an accurate capture of turbulence characteristics during the wave propagation, rather than employing the standard sub-particle scale (SPS) model, the k-ε turbulence model is coupled with the numerical scheme. The results of the numerical model are compared with experimental data, and good agreement is observed in terms of mean velocity, free surface elevation, vorticity fields and turbulent kinetic energy. The numerical model is then employed to investigate the effects of wave non-linearity and geometrical size of the submerged barrier on the flow separation; and calculate the reflection, dissipation and transmission coefficients to evaluate the importance of energy dissipation due to the generation of vortices. The results of this study show that the developed ISPH method with the k-ε turbulence closure model is capable of reproducing the velocity fields and the turbulence characteristics accurately, and thus can be used to perform predictions of comprehensive hydrodynamics of flow-structure interactions in the urban hydro-environment systems.

For industrial applications of innovative metal forming techniques, advanced experimental methods have been developed to characterize the mechanical properties of materials under the corresponding forming conditions. This paper focuses on uniaxial tensile tests for the characterization of the thermomechanical behavior of materials and biaxial tensile tests for the evaluation of the material formability under hot-stamping conditions; these test methods have been improved in recent years. Applications of both the uniaxial and biaxial tensile tests to a boron steel sheet are presented, and the associated experimental results are analyzed. Importantly, new challenges encountered in the development of these experimental methods are discussed and summarized. This paper concludes that more efforts are needed for the standardization of these experimental methods in future.

Biaxial tensile testing methods using cruciform specimens have been developed in the last few decades for the determination of forming limit diagrams (FLDs) and fracture forming limit diagrams (FFLDs) for sheet metals. One of the difficulties associated with this test geometry is the lack of a widely accepted method to determine the necking and fracture strains which are necessary to construct these diagrams. In this study, a novel spatio-temporal method has been proposed for the determination of necking and fracture strains. In the method, two rectangular zones: the base zone (BZ) and the reference zone (RZ) are selected at the location where fracture initiates. The zone RZ includes the zone BZ and both zones have the same side length in the direction parallel to the necking band but different side length in the perpendicular direction. By plotting the thickness reduction within RZ against that in BZ, the onset of localised necking can be determined by finding the intersection of the two straight lines fitted separately using the data in the initial and final stages of deformation. The corresponding limit strains are then determined using the strains within the zone BZ. The method has been successfully applied to uniaxial tensile tests on AA6082 and boron steel dog-bone specimens, and to equi-biaxial tensile tests on AA5754 cruciform specimens.• Compared to widely used existing methods, the novel spatio-temporal method has greater simplicity, stability and accuracy with regard to the determination of localised necking strains.• The spatio-temporal method has good potential to become a standard method for the determination of limit strains for sheet metals.

Reasonable design and development of a low-cost and high-efficiency bifunctional electrocatalyst for oxygen evolution reaction (OER) and oxygen reduction reaction (ORR) is essential for promoting the development of Zinc-air battery technology. Herein, we obtained an integrated catalytic electrode, NiFe nanoparticles supported on nitrogen-doped carbon (NC) directly grown on the carbon cloth (designated as Ni3Fe2@NC/CC), by pyrolysis of bimetallic NiFe metal-organic framework (MOF) precursor. There is a synergistic effect between nickel and iron component, which enhances the bifunctional catalytic activity. In addition, the underlying carbon cloth is conducive to the efficient electron transfer and also benefits the uniform loading of catalytically active materials. Thus, the integrated electrode shows good OER/ORR dual-functional catalytic performance, and the OER overpotential is much lower than that of the traditional drop-coating electrode and precious metal catalyst (IrO2). Moreover, the Ni3Fe2@NC/CC integrated electrode used in zinc-air batteries shows good flexibility and cycle stability. Our findings provide a new avenue for the development of efficient and stable bifunctional oxygen electrocatalysts.

An internal wave is a typical dynamic process. As an internal wave, an internal solitary wave usually occurs between two layers of fluids with different densities. Compared with general internal waves, internal solitary waves have large amplitudes, fast propagation speeds, short-wave periods, and often have tremendous energy. The propagation causes strong convergence and divergence of seawater and generates a sudden strong current. Due to its various characteristics, the propagation of internal solitary waves can cause serious harm to offshore engineering structures. Therefore, studying the effects of internal solitary waves on risers is vital in preventing environmental pollution caused by riser damage. Although the research on internal solitary waves has achieved very fruitful results, the research on structures is mostly focused on a single condition, and the occurrence of internal solitary wave, as a complex ocean phenomenon, is often accompanied by many situations. Therefore, this paper constructs a numerical simulation of the interaction between two columns of internal solitary waves and risers. This study explores the force and flow field changes of the riser under the condition of multiple internal solitary waves using the Star-CCM+ software in the simulation. The improved K-epsilon turbulence model was adopted to close the three-dimensional incompressible Navier–Stokes equation, and the solitary wave solution of the eKdV equation was used as the initial and boundary conditions. The interaction between single and double internal solitary waves and a riser was calculated, compared, and analyzed using numerical analysis. The experiment results indicate that the conditions of two internal solitary waves differ from those of a single internal solitary wave. After colliding at the riser, the waves gradually merge into a single wave, and the flow field reaches its minimum velocity. Under the two-wave condition, the horizontal force on the riser as a whole is less than the single-wave condition. As the amplitude difference between the two internal solitary waves gradually decreases, the horizontal opposing force received by the riser first increases and then decreases, while the horizontal positive force gradually decreases.

Due to their advantages of good flexibility, low cost, simple operations, and small equipment size, electrochemical sensors have been commonly employed in food safety. However, when they are applied to detect various food or drug samples, their stability and specificity can be greatly influenced by the complex matrix. By combining electrochemical sensors with molecular imprinting techniques (MIT), they will be endowed with new functions of specific recognition and separation, which make them powerful tools in analytical fields. MIT-based electrochemical sensors (MIECs) require preparing or modifying molecularly imprinted polymers (MIPs) on the electrode surface. In this review, we explored different MIECs regarding the design, working principle and functions. Additionally, the applications of MIECs in food and drug safety were discussed, as well as the challenges and prospects for developing new electrochemical methods. The strengths and weaknesses of MIECs including low stability and electrode fouling are discussed to indicate the research direction for future electrochemical sensors.

Legumain has been found overexpressed in several cancers, which serves as an important biomarker for cancer diagnosis. In this research, a novel fluorine-18 labeled radioactive tracer [18F]SF-AAN targeting legumain was designed and synthesized for positron emission tomography (PET) imaging. Nonradioactive probe [19F]SF-AAN was obtained through chemical and solid phase peptide synthesis. After a simple one-step 18F labeling, the radiotracer [18F]SF-AAN was obtained with a high radiochemical conversion rate (>85%) and radiochemical purity (99%) as well as high molar activity (12.77 ± 0.50 MBq/nmol). The targeting specificity of [18F]SF-AAN for detecting legumain activity was investigated systematically in vitro and in vivo. In vitro cellular uptake assay showed that the uptake of [18F]SF-AAN in legumain-positive MDA-MB-468 cells was twice as much as that in legumain-negative PC-3 cells at 4 h. In vivo PET imaging revealed that the tumor uptake of [18F]SF-AAN in MDA-MB-468 tumor-bearing mice was about 2.7 times of that in PC-3 tumor-bearing mice at 10 min post injection. The experimental results indicated that [18F]SF-AAN could serve as a promising PET tracer for detecting the legumain expression sensitively and specifically, which would be beneficial for the diagnosis of legumain-related diseases.

Cs-corrected high-angle-annular-dark-field scanning-transmission-electron microscopy (Cs-corrected HAADF-STEM) was employed to examine the phases in AlCuLi alloy (AA2050), including GP(T1), GP(θ″) and GPB zones with their subsequent nanometer-sized products, T1 (Al2CuLi), θ′ (Al2Cu), and S (Al2CuMg) precipitates, respectively. Under the peak-aging condition, some solute-atom enriched clusters could still be found, and the newly-formed nucleus of GP(θ″) with a mono-layer {100} plane of Cu atoms occurred at the adjacent area of the joint between θ′ and S precipitates or the edge of an individual S precipitate. The transition of a single Cu-layer GP(θ″) → θ′ was presumed to be transformation via in-situ nucleation. The developing GP(θ″) zones and θ′ precipitates were easily subjected to soft impingement. However, hard impingement between two variants of θ′ presumably occurred, wherein one θ′ variant precipitate was blocked out by the other θ′ variant. As for the creep-ageing forming (CAF) treated sample, some precipitates of T1 and θ′ were found to have the cutting characteristic on specific ledges. With an increasing demand for lightweight design of vehicles in automotive and aircraft industries, sheet metals with low density and high strength have been widely and intensively used in forming lightweight structural panel components. Formability is a critical material property in describing deformation ability of sheet metals, and it is usually evaluated by a forming limit diagram (FLD) determined at various forming conditions. FLDs for metallic material are usually obtained experimentally, which is time-consuming and costly. Numbers of theoretical and numerical models have been developed and used to predict the formability of sheet metals. These modelling techniques are primarily developed based on bifurcation theory, geometrical imperfection theory and continuum damage mechanics. This paper covers a comprehensive review of modelling methods used for the formability prediction of lightweight materials for sheet metal forming applications. Aluminium alloys are being increasingly utilised in the automotive and aerospace industries to reduce the weight of vehicles. Extensive research has been conducted to overcome the poor ductility of aluminium alloys at room temperature and improve formability of the materials, to enable complex-shaped panel components to be manufactured. To this end, this paper contains a comprehensive review of widely used forming processes for aluminium alloys, under cold, warm and hot forming conditions, and the material characteristics and equipment used for each process. Based on a detailed analysis from the view of industrial requirements, recent progress in experimentation techniques are reviewed addressing the limitations and improvements of specific forming processes. Furthermore, material modelling methods at both cold and elevated temperature forming conditions have been presented. In addition, finite element (FE) simulations with the implementation of material models are discussed. This review article intends to provide a systematic guide for process designers to choose the most appropriate sheet forming technique for specific industrial applications. An alkaline lipase strain of Pseudomonas aeruginosa HFE733 was isolated from soil samples of domestic waste. The alkaline lipase from P. aeruginosa HFE733 was purified and characterized. The enzyme was purified 9.97-fold by means of ammonium sulphate precipitation, Sephadex G-25 and diethylaminoethyl (DEAE) cellulose chromatography. Purified alkaline lipase protein was analyzed by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE), the molecular mass was 51.0 kDa. The enzyme exhibited maximum activity at 40 °C and pH 8.5. The enzyme is stable at pH 7.0–8.5. It was remarkably activated by some metal ions and chemical reagents, such as Fe3+, Al3+, β-mercaptoethanol, cysteine, DL-dithiothreitol (DTT), Tween 80 and Triton X-100, but suppressed by sodium dodecyl sulfate (SDS) at 10 mmol/L. The P. aeruginosa HFE733 strain and lipase exhibited remarkable potential for biodegradation of oil and organics (measured as chemical oxygen demand (COD)). We demonstrated that it can be used for biodegradation of food wastewater from restaurants.

Hepatic stellate cell (HSC) activation occurs along with decreased Perilipin5 (Plin5) and liver fatty acid-binding protein (L-Fabp) expression and coincident lipid droplet (LD) depletion. Conversely, the activated phenotype is reversible in WT HSCs upon forced expression of Plin5. Here, we asked if L-Fabp expression is required for Plin5-mediated rescue of the quiescent phenotype. Lentiviral Plin5 transduction of passaged L-Fabp−/− HSCs failed to reverse activation markers or restore lipogenic gene expression and LD formation. However, adenoviral L-Fabp infection of lentiviral Plin5 transduced L-Fabp−/− HSCs restored both the quiescent phenotype and LD formation, an effect also mediated by adenoviral intestine-Fabp or adipocyte-Fabp. Expression of exogenous Plin5 in activated WT HSCs induced a transcriptional program of lipogenic gene expression including endogenous L-Fabp, but none of the other FABPs. We further demonstrated that selective, small molecule inhibition of endogenous L-Fabp also eliminated the ability of exogenous Plin5 to rescue LD formation and reverse activation of WT HSCs. This functional coordination of L-Fabp with Plin5 was 5′-AMP-activated protein kinase (AMPK)-dependent and was eliminated by AMPK inhibition. Taken together, our results indicate that L-Fabp is required for Plin5 to activate a transcriptional program that restores LD formation and reverses HSC activation.

An analytical model for predicting the shapes of rectangular bars with variable curvatures along their lengths through a novel forming method, differential velocity sideways extrusion (DVSE), previously proposed by the authors, has been developed on the basis of the upper bound method. A new flow line function was presented to describe its deformation field. The plastic deformation zone (PDZ) was assumed to be fan-shaped, where the trajectory of the material flow within the PDZ had an elliptic shape. The proposed continuous flow line function was validated using finite element simulations. The flow patterns, extrusion pressure, curvature, and effective strain predicted by the analytical solutions agreed well with modelling results. Compared to the classical discontinuous simple shear model of channel angular extrusion (CAE) with a 90° die, the new approach was shown to predict the effective strain more closely. The yield strength and work hardening properties of an Al-Cu-Li alloy AA2050 after artificial ageing have been experimentally investigated and modelled in this study. Uniaxial tensile stress-strain curves of the alloy artificially aged for up to 500 h have been acquired and evolutions of main precipitates during ageing have been summarised to elucidate the underlying mechanisms of the observed mechanical properties, such as yield strength and work hardening behaviour. Work hardening analysis with Kocks-Mecking plots has been performed to analyse the shearing-to-bypassing transition progress of the aged alloy and it has been found that the transition does not occur at the peak-ageing state. A new mechanism-based unified constitutive model, comprising three sub-models, has been developed to simultaneously predict the evolutions of microstructures, yield strength and work hardening properties of the artificially aged AA2050. It is the first unified model covering a wide range of artificial ageing conditions from under-ageing to over-ageing, providing an effective tool for performance prediction of the aged alloys for industrial applications. The model has the generic feature and could be applied to artificial ageing of other 2xxx series aluminium alloys with dominant T1 precipitates. A novel manufacturing process, enabling the production of high quality (i.e. with low and controllable residual stress (RS) distributions and good mechanical properties) T-section 7xxx panels, has been established. This process provides a solution to residual stress induced distortion problems, which greatly concerns a range of industries, especially the aircraft industry. This process consists of three sequential steps — water quenching (WQ), cold rolling (CR) and constrained ageing (CA). The effectiveness of this process was experimentally verified through applying this process to laboratory sized 7050 T-section panels. The RS was measured by neutron diffraction and X-ray techniques, in addition to deflections and hardness at each processing stage. An integrated Finite Element (FE) model, including all three steps, was developed to simulate this manufacturing process and predict both the RS and the final strength distributions. It has been concluded that this novel process can effectively reduce the residual stresses from ±300 MPa to within ±100 MPa and produce T-section panels with required mechanical properties (i.e. hardness: ~159 HV10). A cold rolling level of 1.5% was found most appropriate. The residual stress and yield strength distributions were accurately predicted by FE, providing a valuable prediction tool to process optimization for industrial applications. Microstructural evolution during hot forming of aluminium alloys plays a critical role in both the material flow behaviour during the deformation and the post-form mechanical properties in service. This paper presents a comprehensive review on the recrystallisation mechanisms, the interrelations between microstructures and macroscopic responses, and the associated modelling methods for aluminium alloys under hot forming conditions. Particular attention is focused on dynamic recrystallisation (DRX), which occurs during hot forming. The mechanisms, key features, and conditions of occurrence (forming temperature, strain rates, etc.) during hot forming for each type of DRX type are classified. The relationships between microstructures and macroscopic responses, including the flow behaviour, the post-form strength and ductility, are summarised based on existing experimental results. Most importantly, the associated modelling work, describing the recrystallisation and the viscoplastic behaviour under hot forming conditions, is grouped into four types, to enable a clear and concise understanding of the existing quantitative micro–macro interactions, which are particularly valuable for the future development of advanced physically based multi-scale modelling work for hot-forming processes in aluminium alloys.

This paper presents a three-dimensional numerical simulation model of an oil spill for application in emergency treatment methods under icy water conditions. The combined effects of wind, wave, current and ice implemented in our model correspond to Arctic Ocean conditions. A discrete element method combined with an overset grid was adopted to track the trajectory movements of oil film with medium-density ice floes and simulate the flow field of moving ice of large displacement in six degrees of freedom (6DOF). The probability of oil spill area extensions were estimated by a response surface method (RSM). Results showed reduced risk of pollution in icy water conditions and greater drift action of oil film. Accordingly, the spraying location and quantity of oil-dispersant could be rapidly specified.

To mitigate mechanical and chemical degradation of active materials, hollow core–shell structures have been applied in lithium ion batteries. Without embedding of lithium ions, the rigid coating shell can constrain the inward volume deformation. In this paper, optimal conditions for the full use of inner hollow space are identified in terms of the critical ratio of shell thickness and inner size and the state of charge. It is shown that the critical ratios are 0.10 and 0.15 for Si particle and tube (0.12 and 0.18 for Sn particle and tube), and above which there is lack of space for further lithiation.

A series of novel zoledronic acid (ZL) derivatives 1-hydroxy-3-(2-methyl-1H-imidazol-1-yl)propane-1,1-diyldiphosphonic acid (MIPrDP), 1-hydroxy-4-(2-methyl-1H-imidazol-1-yl)butane-1,1-diyldiphosphonic acid (MIBDP), and 1-hydroxy-5-(2-methyl-1H-imidazol-1-yl)pentane-1,1-diyldiphosphonic acid (MIPeDP) were prepared and successfully labeled with 99mTc in high labeling yields. The in vitro stability and in vivo biodistribution of 99mTc-MIPrDP, 99mTc-MIBDP and 99mTc-MIPeDP were investigated and compared. The biodistribution studies indicate that the radiotracer 99mTc-MIPrDP has highly selective uptake in the skeletal system and rapid clearance from soft tissues. The present findings indicate that 99mTc-MIPrDP holds great potential for use in bone imaging.
