Abstract Cyclo (Phe-Pro) (cFP), a cyclic dipeptide with notable antifungal, antibacterial, and antiviral properties, shows great promise for biological control of plant diseases. Produced as a byproduct by non-ribosomal peptide synthetases (NRPS), the regulatory mechanism of cFP biosynthesis remains unclear. In a screening test of 997 Tn5 mutants of Burkholderia seminalis strain R456, we identified eight mutants with enhanced antagonistic effects against Fusarium graminearum (Fg). Among these, mutant 88’s culture filtrate contained cFP, confirmed through HPLC and LC-MS, which actively inhibited Fg. The gene disrupted in mutant 88 is part of the Dct transport system (Dct-A, -B, -D), responsible for C4-dicarboxylate transport. Knockout mutants of Dct genes exhibited higher cFP levels than the wild type, whereas complementary strains showed no significant difference. Additionally, the presence of exogenous C4-dicarboxylates reduced cFP production in wild type R456, indicating that these substrates negatively regulate cFP synthesis. Given that cFP synthesis is related to NRPS, we previously identified an NRPS cluster in R456, horizontally transferred from algae. Specifically, knocking out gene 2061 within this NRPS cluster significantly reduced cFP production. A Fur box binding site was predicted upstream of gene 2061, and yeast one-hybrid assays confirmed Fur protein binding, which increased with additional C4-dicarboxylates. Knockout of the Fur gene led to up-regulation of gene 2061 and increased cFP production, suggesting that C4-dicarboxylates suppress cFP synthesis by enhancing Fur-mediated repression of gene 2061.

Abstract Drought stress is a major abiotic stress affecting the performance of wheat (Triticum aestivum L.). The current study evaluated the effects of drought on wheat phenology, physiology, and biochemistry; and assessed the effectiveness of foliar-applied sulfhydryl thiourea to mitigate drought-induced oxidative stress. The treatments were: wheat varieties; V1 = Punjab-2011, V2 = Galaxy-2013, V3 = Ujala-2016, and V4 = Anaaj-2017, drought stress; D1 = control (80% field capacity [FC]) and D2 = drought stress (40% FC), at the reproductive stage, and sulfhydryl thiourea (S) applications; S0 = control-no thiourea and S1 = foliar thiourea application @ 500 mg L−1. Results of this study indicated that growth parameters, including height, dry weight, leaf area index (LAI), leaf area duration (LAD), crop growth rate (CGR), net assimilation rate (NAR) were decreased under drought stress-40% FC, as compared to control-80% FC. Drought stress reduced the photosynthetic efficiency, water potential, transpiration rates, stomatal conductances, and relative water contents by 18, 17, 26, 29, and 55% in wheat varieties as compared to control. In addition, foliar chlorophyll a, and b contents were also lowered under drought stress in all wheat varieties due to an increase in malondialdehyde and electrolyte leakage. Interestingly, thiourea applications restored wheat growth and yield attributes by improving the production and activities of proline, antioxidants, and osmolytes under normal and drought stress as compared to control. Thiourea applications improved the osmolyte defense in wheat varieties as peroxidase, superoxide dismutase, catalase, proline, glycine betaine, and total phenolic were increased by 13, 20, 12, 17, 23, and 52%; while reducing the electrolyte leakage and malondialdehyde content by 49 and 32% as compared to control. Among the wheat varieties, Anaaj-2017 showed better resilience towards drought stress and also gave better response towards thiourea application based on morpho-physiological, biochemical, and yield attributes as compared to Punjab-2011, Galaxy-2013, and Ujala-2016. Eta-square values showed that thiourea applications, drought stress, and wheat varieties were key contributors to most of the parameters measured. In conclusion, the sulfhydryl thiourea applications improved the morpho-physiology, biochemical, and yield attributes of wheat varieties, thereby mitigating the adverse effects of drought. Moving forward, detailed studies pertaining to the molecular and genetic mechanisms under sulfhydryl thiourea-induced drought stress tolerance are warranted.

Abstract Canola, a vital oilseed crop, is grown globally for food and biodiesel. With the enormous demand for growing various crops, the utilization of agriculturally marginal lands is emerging as an attractive alternative, including brackish-saline transitional lands. Salinity is a major abiotic stress limiting growth and productivity of most crops, and causing food insecurity. Salicylic acid (SA), a small-molecule phenolic compound, is an essential plant defense phytohormone that promotes immunity against pathogens. Recently, several studies have reported that SA was able to improve plant resilience to withstand high salinity. For this purpose, a pot experiment was carried out to ameliorate the negative effects of sodium chloride (NaCl) on canola plants through foliar application of SA. Two canola varieties Faisal (V1) and Super (V2) were assessed for their growth performance during exposure to high salinity i.e. 0 mM NaCl (control) and 200 mM NaCl. Three levels of SA (0, 10, and 20 mM) were applied through foliar spray. The experimental design used for this study was completely randomized design (CRD) with three replicates. The salt stress reduced the shoot and root fresh weights up to 50.3% and 47% respectively. In addition, foliar chlorophyll a and b contents decreased up to 61–65%. Meanwhile, SA treatment diminished the negative effects of salinity and enhanced the shoot fresh weight (49.5%), root dry weight (70%), chl. a (36%) and chl. b (67%). Plants treated with SA showed an increased levels of both enzymatic i.e. (superoxide dismutase (27%), peroxidase (16%) and catalase (34%)) and non-enzymatic antioxidants i.e. total soluble protein (20%), total soluble sugar (17%), total phenolic (22%) flavonoids (19%), anthocyanin (23%), and endogenous ascorbic acid (23%). Application of SA also increased the levels of osmolytes i.e. glycine betaine (31%) and total free proline (24%). Salinity increased the concentration of Na+ ions and concomitantly decreased the K+ and Ca2+ absorption in canola plants. Overall, the foliar treatments of SA were quite effective in reducing the negative effects of salinity. By comparing both varieties of canola, it was observed that variety V2 (Super) grew better than variety V1 (Faisal). Interestingly, 20 mM foliar application of SA proved to be effective in ameliorating the negative effects of high salinity in canola plants.

Background Sequestering carbon dioxide (CO2) in agricultural soils promises climate change mitigation as well as sustainable ecosystem services. In order to stabilize crop residues as soil carbon (C), addition of mineral nutrients in excess to crop needs is suggested as an inevitable practice. However, the effect of two macronutrients i.e., nitrogen (N) & phosphorus (P), on C cycling has been found contradictory. Mineral N usually decreases whereas mineral P increases the soil organic C (SOC) mineralization and microbial biomass. How the addition of these macronutrients in inorganic form to an organic-matter poor soil affect C cycling remains to be investigated. Methods To reconcile this contradiction, we tested the effect of mineral N (120 kg N ha−1) and/or P (60 kg N ha−1) in presence or absence of maize litter (1 g C kg−1 soil) on C cycling in an organic-matter poor soil (0.87% SOC) in a laboratory incubation. Soil respiration was measured periodically during the incubation whereas various soil variables were measured at the end of the incubation. Results Contrary to literature, P addition stimulated soil C mineralization very briefly at start of incubation period and released similar total cumulative CO2-C as in control soil. We attributed this to low organic C content of the soil as P addition could desorb very low amounts of labile C for microbial use. Adding N with litter built up the largest microbial biomass (144% higher) without inducing any further increase in CO2-C release compared to litter only addition. However, adding P with litter did not induce any increase in microbial biomass. Co-application of inorganic N and P significantly increased C mineralization in presence (19% with respect to only litter amended) as well as absence (41% with respect to control soil) of litter. Overall, our study indicates that the combined application of inorganic N and P stabilizes added organic matter while depletes the already unamended soil.

Twig blight is a highly destructive disease primarily caused by Pestalotiopsis versicolor. It affects bayberry (Myrica rubra) cultivation and production. However, the molecular and regulatory defence responses to twig blight remain unclear. We inoculated a susceptible cultivar bayberry variety with P. versicolor XJ27 and carefully profiled the disease development. The bayberry disease index increased sharply following inoculation. We conducted integrative transcriptomic and metabolomic analyses to track key genes and secondary metabolomic changes in susceptible bayberry leaves. Herein, we identified 9588 differentially expressed genes (DEGs) and 748 differentially accumulated metabolites (DAMs). Of these, seven structural genes and seven metabolites (including L-phenylalanine, coniferin, p-coumaric acid, cinnamaldehyde, eleutheroside B, ferulaldehyde, and isoeugenol) related to lignin biosynthesis markedly increased following pathogen infection. These observations suggest that lignin biosynthesis may facilitate bayberry defence responses to twig blight. Quantitative real-time PCR analysis further confirmed the higher expression levels of pivotal lignin synthesis genes in infected leaves. In addition, higher foliar lignin levels and increased xylem formation in infected leaves provided evidence that lignin synthesis was enhanced during bayberry response to twig blight. These findings not only provide effective management strategies for bayberry twig blight but also promote the breeding of high-quality resistant varieties.

Rice is a globally important food crop which is sensitive to the presence of a metalloid, arsenic (As). There is limited research pertaining to identifying relevant As-tolerant rice germplasm in adaptive breeding research initiatives, despite the fact that As contamination in rice has long been known. This study served to identify the growth performance of different rice genotypes under high levels of As. Rice seed germination analysis (germination percentage, GP) was performed to categorize the eight different rice genotypes and growing under varying As levels including As25, 25 μM and As50, 50 μM. The Zhenong 41 was identified as the highly tolerant genotypes with lowest decrease in GP by 87 %, plant height (PH) by 26 %, and dry weight (DW) by 16 %; while 9311 was observed to be the most sensitive genotype with highest reduction in GP by 44 %, PH by 48 % and DW by 54 % under As25 stress conditions, compared to control treatment. The higher As50 stress treatment delivered more adverse growth inhibitory effects than the rice plants cultivated under As25. Specifically, the As-sensitive rice genotype 9311 showed significantly higher decrease in foliar chlorophyll contents relative to the other genotypes, especially Zhenong 41 (As-tolerant). During exposure to high As levels, the rice genotype 9311 significantly modulated and augmented the production of MDA and H2O2 by stimulating the activities of POD, SOD, and CAT. This study revealed interesting insights into the responses of rice genotypes to variable As stresses throughout the various growth stages. Overall, the findings of this study could be harnessed to support any ongoing As-tolerant rice breeding agendas for cultivation in As-polluted environments.

This research reported the utilization of novel adsorbent from the de-oiled waste material of orange seeds for preparing simple charcoal (SC) and iron oxide/activated charcoal (Fe2O3/AC) nanocomposites. Batch adsorption experiments were carried out to evaluate the optimized conditions of the experiment. The results obtained indicated that the pseudo-second-order kinetic model best fitted the adsorption data (R2 > 0.99) and that the Freundlich isotherm model best explained the adsorption of dye on Fe2O3/AC. The adsorption process was spontaneous and exothermic in the temperature range of 293–333°K, as explained by calculated thermodynamic parameters, e.g., ΔG°, ΔH°, and ΔS°. The Fourier transform infrared (FTIR) spectroscopy results publicized that carboxyl and amine functional groups are present on the surface of adsorbents, which are responsible for the attachment of dye. The scanning electron microscopy (SEM) results showed that Fe2O3/AC has a porous surface and textual structure, which can efficiently adsorb dye molecules. A zetasizer was utilized for determining the size of the composites, and the thermal stability was determined by performing a thermogravimetric analysis (TGA). The findings of the comparative experiment indicated that Fe2O3/AC are more promising than raw activated carbon for the adsorption of Congo red (CR). The impregnation of iron oxide nanocomposites on an adsorbent resulted in an increased surface area-to-volume ratio, magnetic properties, and excellent reusable capacity. Overall, it can be reported as an innovative procedure promoting the recycling of waste products, which aids in protecting environmental and human health and in the development of the economy.

Chromium (Cr) toxicity hampers ornamental crops’ growth and post-harvest quality, especially in cut flower plants. Nano-enabled approaches have been developing with phenomenal potential towards improving floricultural crop production under heavy metal-stressed conditions. The current pot experiment aims to explore the ameliorative impact of silicon nanoparticles (Si-NPs; 10 mM) and indole butyric acid (IBA; 20 mM) against Cr stress (0.8 mM) in Freesia refracta. The results showed that Cr stress significantly reduced morphological traits, decreased roots-stems biomass, abridged chlorophyll (14.7%) and carotenoid contents (27.2%), limited gas exchange attributes (intercellular CO2 concentration (Ci) 24.8%, stomatal conductance (gs) 19.3% and photosynthetic rate (A) 28.8%), condensed proline (39.2%) and total protein (40%) contents and reduced vase life (15.3%) of freesia plants by increasing oxidative stress. Contrarily, antioxidant enzyme activities, MDA and H2O2 levels, and Cr concentrations in plant parts were remarkably enhanced in Cr-stressed plants than in the control. However, foliar supplementation of Si-NPs + IBA (combined form) to Cr-stressed plants increased defense mechanism and tolerance as revealed by improved vegetative and reproductive traits, increased biomass, photosynthetic pigments (chlorophyll 30.3%, carotenoid 57.2%) and gaseous exchange attributes (Ci 33.3%, gs 25.6%, A 31.1%), proline (54.5%), total protein (55.1%), and vase life (34.9%) of metal contaminated plants. Similarly, the improvement in the activities of peroxidase, catalase, and superoxide dismutase was recorded by 30.8%, 52.4%, and 60.8%, respectively, compared with Cr-stressed plants. Meanwhile, MDA (54.3%), H2O2 (32.7%) contents, and Cr levels in roots (43.3), in stems (44%), in leaves (52.8%), and in flowers (78.5%), were remarkably reduced due to combine application of Si-NPs + IBA as compared with Cr-stressed nontreated freesia plants. Thus, the hypothesis that the synergistic application of Si-NPs + IBA will be an effective approach in ameliorating Cr stress is authenticated from the results of this experiment. Furthermore, the study will be significant since it will demonstrate how Si-NPs and IBA can work synergistically to combat Cr toxicity, and even when added separately, they can improve growth characteristics both under stressed and un-stressed conditions.

Maize is an economically vital cereal crop. However, water deficiency can severely impact its productivity. Thus, it is necessary to implement an essential approach to increase maize yield while navigating the limitations imposed by scarce water supplies. The present study aimed to investigate whether foliar applications of silicon (Si) and zinc (Zn) could mitigate the adverse effects of water deficiency and improve maize growth and yield. Field experiments were conducted in Egypt during two growing seasons (2021–2022) under three irrigation regimes: full irrigation (ET0), moderate stress (ET1), and severe stress (ET2). The treatments comprised foliar sprays of Si, Zn, Si + Zn, and water control. Phenological, growth, physiological, chemical, and yield-related traits were assessed. Results showed that adequate irrigation (ET0) enhanced most parameters compared to water stress treatments. Under ET0, the combined silicon and zinc treatment resulted in the highest values for plant height, leaf area, chlorophyll content, grains per ear, kernel weight, ear size, and yield compared to other foliar treatments. Under drought stress (ET1, ET2), Si + Zn applications maintained superiority in mitigating yield losses. Proline accumulation was highest under severe stress (ET2) in the absence of foliar sprays, indicating greater drought impacts. Correlation analysis revealed positive associations of grain yield with ear size, leaf area, kernel weight, and biological yield. Cluster analysis separated irrigation regimes and visualized the consistently beneficial effects of Si + Zn across all water levels. Overall, the results demonstrate the synergistic potential of Si and Zn supplementation to sustain maize performance and yields under varying water availability.

Abstract Pea (Pisum sativum L.), a globally cultivated leguminous crop valued for its nutritional and economic significance, faces a critical challenge of soil salinity, which significantly hampers crop growth and production worldwide. A pot experiment was carried out in the Botanical Garden, The Islamia University of Bahawalpur to alleviate the negative impacts of sodium chloride (NaCl) on pea through foliar application of ascorbic acid (AsA). Two pea varieties Meteor (V1) and Sarsabz (V2) were tested against salinity, i.e. 0 mM NaCl (Control) and 100 mM NaCl. Three levels of ascorbic acid 0 (Control), 5 and 10 mM were applied through foliar spray. The experimental design was completely randomized (CRD) with three replicates. Salt stress resulted in the suppression of growth, photosynthetic activity, and yield attributes in pea plants. However, the application of AsA treatments effectively alleviated these inhibitory effects. Under stress conditions, the application of AsA treatment led to a substantial increase in chlorophyll a (41.1%), chl. b (56.1%), total chl. contents (44.6%) and carotenoids (58.4%). Under salt stress, there was an increase in Na+ accumulation, lipid peroxidation, and the generation of reactive oxygen species (ROS). However, the application of AsA increased the contents of proline (26.9%), endogenous AsA (23.1%), total soluble sugars (17.1%), total phenolics (29.7%), and enzymatic antioxidants i.e. SOD (22.3%), POD (34.1%) and CAT (39%) in both varieties under stress. Salinity reduced the yield attributes while foliarly applied AsA increased the pod length (38.7%), number of pods per plant (40%) and 100 seed weight (45.2%). To sum up, the application of AsA alleviated salt-induced damage in pea plants by enhancing photosynthetic pigments, both enzymatic and non-enzymatic activities, maintaining ion homeostasis, and reducing excessive ROS accumulation through the limitation of lipid peroxidation. Overall, V2 (Sarsabz) performed better as compared to the V1 (Meteor).

Abstract Cadmium (Cd), being a heavy metal, tends to accumulate in soils primarily through industrial activities, agricultural practices, and atmospheric deposition. Maize, being a staple crop for many regions, is particularly vulnerable to Cd contamination, leading to compromised growth, reduced yields, and potential health risks for consumers. Biochar (BC), a carbon-rich material derived from the pyrolysis of organic matter has been shown to improve soil structure, nutrient retention and microbial activity. The choice of biochar as an ameliorative agent stems from its well-documented capacity to enhance soil quality and mitigate heavy metal stress. The study aims to contribute to the understanding of the efficacy of biochar in combination with GA3, a plant growth regulator known for its role in promoting various physiological processes, in mitigating the adverse effects of Cd stress. The detailed investigation into morpho-physiological attributes and biochemical responses under controlled laboratory conditions provides valuable insights into the potential benefits of these interventions. The experimental design consisted of three replicates in a complete randomized design (CRD), wherein soil, each containing 10 kg was subjected to varying concentrations of cadmium (0, 8 and 16 mg/kg) and biochar (0.75% w/w base). Twelve different treatment combinations were applied, involving the cultivation of 36 maize plants in soil contaminated with Cd (T1: Control (No Cd stress; T2: Mild Cd stress (8 mg Cd/kg soil); T3: Severe Cd stress (16 mg Cd/kg soil); T4: 10 ppm GA3 (No Cd stress); T5: 10 ppm GA3 + Mild Cd stress; T6: 10 ppm GA3 + Severe Cd stress; T7: 0.75% Biochar (No Cd stress); T8: 0.75% Biochar + Mild Cd stress; T9: 0.75% Biochar + Severe Cd stress; T10: 10 ppm GA3 + 0.75% Biochar (No Cd stress); T11: 10 ppm GA3 + 0.75% Biochar + Mild Cd stress; T12: 10 ppm GA3 + 0.75% Biochar + Severe Cd stress). The combined application of GA3 and BC significantly enhanced multiple parameters including germination (27.83%), root length (59.53%), shoot length (20.49%), leaf protein (121.53%), root protein (99.93%), shoot protein (33.65%), leaf phenolics (47.90%), root phenolics (25.82%), shoot phenolics (25.85%), leaf chlorophyll a (57.03%), leaf chlorophyll b (23.19%), total chlorophyll (43.77%), leaf malondialdehyde (125.07%), root malondialdehyde (78.03%) and shoot malondialdehyde (131.16%) across various Cd levels compared to the control group. The synergistic effect of GA3 and BC manifested in optimal leaf protein and malondialdehyde levels indicating induced tolerance and mitigation of Cd detrimental impact on plant growth. The enriched soils showed resistance to heavy metal toxicity emphasizing the potential of BC and GA3 as viable strategy for enhancing maize growth. The application of biochar and gibberellic acid emerges as an effective means to mitigate cadmium-induced stress in maize, presenting a promising avenue for sustainable agricultural practices.

Abstract Background This ethnobotanical study in Dunyapur, District Lodhran, Pakistan, focuses on traditional medicinal knowledge, exploring 41 plants across 28 families. The research involves 496 informants from diverse backgrounds, including farmers, herbalists, housewives, teachers, and shopkeepers. The prevalence of herbs (68%) aligns with their accessibility and rapid regrowth, shaping the local medicinal landscape. The study investigates socio-demographic features, emphasizing the importance of considering the community's diverse perspectives. Methods The research employs quantitative ethnobotanical data analysis, introducing various indices like PPV, FUV, FIV, RFC, UV, and RI. The analysis of plant growth habits underscores the dominance of herbs, and the method of preparation evaluation identifies decoction as the most common (23%). Leaves (27%) are the most utilized plant part, and Resedaceae stands out with the highest FUV (0.38). FIV highlights the ecological and cultural significance of Poaceae, Boraginaceae, Fabaceae, and Solanaceae. Results The RFC values range from 0.016 to 0.032, with Cucumis melo having the highest value (0.032), indicating its frequent citation and cultural significance. The study reveals specific plants like Melia azedarach, Peganum harmala and Salvadora oleoides with high PR values for skin issues, reflecting their widespread acceptance and effectiveness. Oligomeris linifolia emerges with the highest UV (0.38), emphasizing its greater significance in local traditional practices. Leptadenia pyrotechnica records the highest RI (9.85), underlining its exceptional importance in the community's traditional pharmacopeia. Conclusion The findings offer a holistic understanding of ethnobotanical knowledge in Dunyapur, emphasizing the role of local contexts and ecological factors in shaping traditional plant uses. The study contributes valuable insights into the diverse practices within the community, laying the foundation for sustainable integration of traditional knowledge into broader healthcare frameworks.

Abstract Heavy metal stress affects crop growth and yields as wheat (Triticum aestivum L.) growth and development are negatively affected under heavy metal stress. The study examined the effect of cobalt chloride (CoCl2) stress on wheat growth and development. To alleviate this problem, a pot experiment was done to analyze the role of sulfur-rich thiourea (STU) in accelerating the defense system of wheat plants against cobalt toxicity. The experimental treatments were, i) Heavy metal stress (a) control and (b) Cobalt stress (300 µM), ii) STU foliar applications; (a) control and (b) 500 µM single dose was applied after seven days of stress, and iii) Wheat varieties (a) FSD-2008 and (b) Zincol-2016. The results revealed that cobalt stress decreased chlorophyll a by 10%, chlorophyll b by 16%, and carotenoids by 5% while foliar application of STU increased these photosynthetic pigments by 16%, 15%, and 15% respectively under stress conditions as in contrast to control. In addition, cobalt stress enhances hydrogen peroxide production by 11% and malondialdehyde (MDA) by 10%. In comparison, STU applications at 500 µM reduced the production of these reactive oxygen species by 5% and by 20% by up-regulating the activities of antioxidants. Results have revealed that the activities of SOD improved by 29%, POD by 25%, and CAT by 28% under Cobalt stress. Furthermore, the foliar application of STU significantly increased the accumulation of osmoprotectants as TSS was increased by 23% and proline was increased by 24% under cobalt stress. Among wheat varieties, FSD-2008 showed better adaptation under Cobalt stress by showing enhanced photosynthetic pigments and antioxidant activities compared to Zincol-2016. In conclusion, the foliar-applied STU can alleviate the negative impacts of Cobalt stress by improving plant physiological attributes and upregulating the antioxidant defense system in wheat. Abstract Salinity poses significant challenges to agricultural productivity, impacting crops’ growth, morphology and biochemical parameters. A pot experiment of three months was conducted between February to April 2023 in the Department of Botany, The Islamia University of Bahawalpur. Four brinjal (eggplant) varieties: ICS-BR-1351, HBR-313-D, HBR-314-E, and HBR-334-D were selected and assessed for the effects of salinity on various growth and biochemical attributes. The experiment was completely randomized in design with three replicates each. This study revealed that increased salinity significantly reduced the shoot length, root length, and leaf number across all varieties, with maximum adverse effects observed at a 300mM NaCl concentration. Among the tested varieties, ICS-BR-1351 demonstrated superior performance in most growth parameters, suggesting potential salt tolerance. Biochemically, salinity decreased chlorophyll content across all varieties, with the sharpest decline observed at the highest salt concentration. V4 (HBR-334-D) showed a 57% decrease in chlorophyll followed by V3 (HBR-314-E) at 56%, V2 (HBR-313-D) at 54%, and V1 (ICS-BR-1351) at 33% decrease at maximum salt levels as compared to control. Conversely, carotenoid content increased up to -42.11% in V3 followed by V2 at -81.48%, V4 at -94.11%, and − 233% in V1 at 300mM NaCl stress as compared to respective controls. V3 (HBR-314-E) has the maximum value for carotenoids while V1 has the lowest value for carotenoids as compared to the other three brinjal varieties. In addition to pigments, the study indicated a salinity-induced decrease in total proteins and total soluble sugar, whereas total amino acids and flavonoids increased. Total proteins showed a decrease in V2 (49.46%) followed by V3 (36.44%), V4 (53.42%), and V1 (53.79%) at maximum salt concentration as compared to plants treated with tap water only. Whereas, total soluble sugars showed a decrease of 52.07% in V3, 41.53% in V2, 19.49% in V1, and 18.99% in V4 at the highest salt level. While discussing total amino acid, plants showed a -9.64% increase in V1 as compared to V4 (-31.10%), V2 (-36.62%), and V3 (-22.61%) with high salt levels in comparison with controls. Plant flavonoid content increased in V3 (-15.61%), V2 (-19.03%), V4 (-18.27%) and V1 (-27.85%) at 300mM salt concentration. Notably, salinity elevated the content of anthocyanin, lycopene, malondialdehyde (MDA), and hydrogen peroxide (H2O2) across all varieties. Antioxidant enzymes like peroxidase, catalase, and superoxide dismutase also increased under salt stress, suggesting an adaptive response to combat oxidative damage. However, V3 (HBR-314-E) has shown an increase in anthocyanin at -80.00%, lycopene at -24.81%, MDA at -168.04%, hydrogen peroxide at -24.22%, POD at -10.71%, CAT as-36.63 and SOD as -99.14% at 300mM NaCl stress as compared to control and other varieties. The enhanced accumulation of antioxidants and other protective compounds suggests an adaptive mechanism in brinjal to combat salt-induced oxidative stress. The salt tolerance of different brinjal varieties was assessed by principal component analysis (PCA), and the order of salt tolerance was V1 (ICS-BR-1351) > V4 (HBR-334-D), > V2 (HBR-313-D) > V3 (HBR-314-E). Among the varieties studied, ICS-BR-1351 demonstrated resilience against saline conditions, potentially offering a promising candidate for saline-prone agricultural areas.

Xanthomonas oryzae pv. oryzae (Xoo) is a significant bacterial pathogen responsible for outbreaks of bacterial leaf blight in rice, posing a major threat to rice cultivation worldwide. Effective management of this pathogen is crucial for ensuring rice yield and food security. In this study, we identified and characterized a novel Xoo phage, ZP3, isolated from diseased rice leaves in Zhejiang, China, which may offer new insights into biocontrol strategies against Xoo and contribute to the development of innovative approaches to combat bacterial leaf blight. Transmission electron microscopy indicated that ZP3 had a short, non-contractile tail. Genome sequencing and bioinformatic analysis showed that ZP3 had a double-stranded DNA genome with a length of 44,713 bp, a G + C content of 52.2%, and 59 predicted genes, which was similar to other OP1-type Xoo phages belonging to the genus Xipdecavirus. ZP3’s endolysin LysZP was further studied for its bacteriolytic action, and the N-terminal transmembrane domain of LysZP is suggested to be a signal–arrest–release sequence that mediates the translocation of LysZP to the periplasm. Our study contributes to the understanding of phage–Xoo interactions and suggests that phage ZP3 and its endolysin LysZP could be developed into biocontrol agents against this phytopathogen.

The urban agro-ecosystem in developing countries plays a major role in uplifting the socioeconomic status of their inhabitants by confronting the challenges of improving food security and eradicating hunger. Food security and safety has become a serious global concern owing to accumulation of phytotoxic metals and metalloids in different food commodities subjected to different contaminated irrigation sources. The current study was undertaken to assess the concentrations of heavy metals and metalloid in ridge gourd (Luffa cylindrica L.), its planting soil medium and irrigation water sources used for crop production to determine the health risk. The results showed that the mean concentrations of different metals and metalloids in ground water samples i.e. Ni (0.14 mg L-1), Fe (1.29 mg L-1), Cu (0.10 mg L-1), Pb (0.55 mg L-1), As (0.10 mg L-1), Cr (2.38 mg L-1) and Cd (0.05 mg L-1) were over the permissible limits. Soil samples irrigated with ground water showed that concentrations of Cd (23.67 mg kg-1), Cr (26.66 mg kg-1) and As (45.97 mg kg-1) were above the permissible limits. Similarly, vegetable samples irrigated with ground water had a higher level of Cd (0.33 mg kg-1) than their prescribed safe limits. The bio-concentration factor ranged from 0.00 to 8.21 for all irrigation sources. The target hazard quotients (THQ) for Zn, Fe, and Cr were higher than the threshold value when ridge gourd was consumed, indicating a health concern. Similarly, hazard index values were 5.05, 28, 27, and 1.01, from groundwater, canal water, and sewage water, respectively. Target Cancer Risk (TCR) exposed Ni, Cr, Cd and As in canal water were sources of high cancer risk and Pb showed moderate cancer risk for the local community. These findings suggest that human intake of ridge gourd vegetable from the studied area may pose health hazards. Hence, frequent monitoring of different irrigation streams used in urban and semi-urban agriculture is strongly recommended to manage higher concentrations of phytotoxic metals and metalloids in crop produce and their associated health risks for humans.

Laboratory and field assays of three sets of experiments were conducted to evaluate the impact of different phagostimulants alone and in combination with other phagostimulant lure sources, such as ammonium acetate, DAP, and acetic acid, on the attractancy of both sexes of B. zonata. In the first experiment, the laboratory olfactometer study revealed that out of eleven phagostimulants, banana, mulberry, mango, guava, molasses, and protein hydrolysate exhibited moderate attractancy (15.2–60.2%) to B. zonata. Unexpectedly, banana and protein hydrolysate were demonstrated to be highly attractive phagostimulants for starved female B. zonata (53.6% and 60.2%, respectively). In the field study, none of the tested phagostimulants exhibited high attractancy; however, banana, mulberry, protein hydrolysate, guava, mango, and molasses demonstrated moderate attractancy (5.6–35.6%) to B. zonata. In the second experiment, out of five phagostimulant-mixtures, phagostimulant-mixture-4 proved highly attractive (40.5–68.6% and 45.5–51.2%), followed by phagostimulant-mixture-3, which proved to be moderately attractive (17.0–22.5% and 28.4–36.1%) to B. zonata in olfactometer and field studies, respectively. In the third experiment, out of five phagostimulant-AdMixtures, phagostimulant-AdMixture-4 demonstrated strong attractiveness in the olfactometer (41.6–68.7%) and field studies (52.7–58.7%) for B. zonata, while the rest of the AdMixtures demonstrated moderate to no attractiveness for B. zonata. So, phagostimulant-AdMixture-4 with GF-120 could be used in the development of a phagostimulant bait station which attracts the maximum B. zonata population and ultimately provides pest-free fruits to the farmers

Salinity exerts significant negative impacts on growth and productivity of crop plants and numerous management practices are used to improve crop performance under saline environments. Micronutrients, plant growth promoting bacteria and biochar are known to improve crop productivity under stressful environments. Maize (Zea mays L.) is an important cereal crop and its productivity is adversely impacted by salinity. Although boron (B) application, seed inoculation with boron-tolerant bacteria (BTB) and biochar are known to improve maize growth under stressful environments, there is less information on their combined impact in enhancing maize productivity on saline soils. This study investigated the impact of B seed coating combined with seed inoculation with BTB + biochar on maize productivity under saline soil. Four B seed coating levels [0.0 (no seed coating), 1.0, 1.5, 2.0 g B kg−1 seed], and individual or combined application of 5 % (w/w) maize stalk biochar, and seed inoculation with Bacillus sp. MN-54 BTB were included in the study. Different growth and yield attributes and grain quality were significantly improved by seed coating with 1.5 B kg−1 seed coupled with biochar + BTB. Seed coating with 1.5 B kg−1 seed combined with biochar + BTB improved stomatal conductance by 32 %, photosynthetic rate by 15 %, and transpiration ratio by 52 % compared to seed coating (0 B kg−1 seed) combined with biochar only. Similarly, the highest plant height (189 cm), number of grain rows cob−1 (15.5), grain yield (54.9 g plant−1), biological yield (95.5 g plant−1), and harvest index (57.6 %) were noted for B seed coating (1.5 g B kg−1 seed) combined with biochar + BTB inoculation. The same treatment resulted in the highest grain protein and B contents. It is concluded that B seed coating at 1.5 g B kg−1 seed combined with biochar + BTB inoculation could significantly improve yield and quality of maize crop on saline soils. However, further field experiments investigating the underlying mechanisms are needed to reach concrete conclusions and large-scale recommendations.

Hazardous organic dyes, present in the effluents of industries, are continuously polluting the environment. Photodegradation of these dyes on catalyst surface under sunlight irradiation is economic, safe and suitable strategy to protect environment. Hence, the synthesis and applications of Zn1-xNixO (x = 0.00, 0.02, 0.04, 0.06) NPs are reported here. Effect of pH and dopant concentration was studied to modify the electrical, magnetic, antibacterial and photocatalytic properties of Ni doped ZnO NPs. The samples were characterized by Scanning electron microscopy (SEM), Powder X-ray diffraction (XRD), UV–Visible spectroscopy (UV–Vis.), Energy-dispersive X-ray spectroscopy (EDS) and Fourier-transform infrared spectroscopy (FTIR) to determine the morphology, crystallite structure, optical properties, elemental composition and functional group detection, respectively. LCR meter and VSM were used to evaluate the dielectric properties and magnetic properties of Ni doped ZnO NPs, respectively. XRD pattern confirmed the presence of hexagonal wurtzite geometry of ZnO NPs. The structural and morphological analysis showed the increase in crystallinity with little effect on shape of doped NPs by increasing the dopant concentration and slight increase of pH. It was observed that the Ni doped ZnO NPs possess good photocatalytic potential by 94% degradation of 20 ppm solution of methyl orange dye (MO) in just 80 min under sunlight. Moreover, the enhancement in antibacterial potential was also observed with increase in dopant concentration and decrease in crystallite size of doped ZnO NPs. Smaller size NPs were found more effective against gram negative bacterial strains.

Plant immunity includes enemy recognition, signal transduction, and defensive response against pathogens. We experimented to identify the genes that contribute resistance against dieback disease to Dalbergia sissoo, an economically important timber tree. In this study, we investigated the role of three differentially expressed genes identified in the dieback-induced transcriptome in Dalbergia sissoo. The transcriptome was probed using DOP-rtPCR analysis. The identified RGAs were characterized in silico as the contributors of disease resistance that switch on under dieback stress. Their predicted fingerprints revealed involvement in stress response. Ds-DbRCaG-02-Rga.a, Ds-DbRCaG-04-Rga.b, and Ds-DbRCaG-06-Rga.c showed structural homology with the Transthyretin-52 domain, EAL associated YkuI_C domain, and Src homology-3 domain respectively, which are the attributes of signaling proteins possessing a role in regulating immune responses in plants. Based on in-silico structural and functional characterization, they were predicted to have a role in immune response regulation in D. sissoo.

Bacterial brown stripe disease caused by Acidovorax oryzae is a major threat to crop yields, and the current reliance on pesticides for control is unsustainable due to environmental pollution and resistance. To address this, bacterial-based ligands have been explored as a potential treatment solution. In this study, we developed a protein–protein interaction (PPI) network for A. oryzae by utilizing shared differentially expressed genes (DEGs) and the STRING database. Using a maximal clique centrality (MCC) approach through CytoHubba and Network Analyzer, we identified hub genes within the PPI network. We then analyzed the genomic data of the top 10 proteins, and further narrowed them down to 2 proteins by utilizing betweenness, closeness, degree, and eigenvector studies. Finally, we used molecular docking to screen 100 compounds against the final two proteins (guaA and metG), and Enfumafungin was selected as a potential treatment for bacterial resistance caused by A. oryzae based on their binding affinity and interaction energy. Our approach demonstrates the potential of utilizing bioinformatics and molecular docking to identify novel drug candidates for precision treatment of bacterial brown stripe disease caused by A. oryzae, paving the way for more targeted and sustainable control strategies. The efficacy of Enfumafungin in inhibiting the growth of A. oryzae strain RS-1 was investigated through both computational and wet lab methods. The models of the protein were built using the Swiss model, and their accuracy was confirmed via a Ramachandran plot. Additionally, Enfumafungin demonstrated potent inhibitory action against the bacterial strain, with an MIC of 100 µg/mL, reducing OD600 values by up to 91%. The effectiveness of Enfumafungin was further evidenced through agar well diffusion assays, which exhibited the highest zone of inhibition at 1.42 cm when the concentration of Enfumafungin was at 100 µg/mL. Moreover, Enfumafungin was also able to effectively reduce the biofilm of A. oryzae RS-1 in a concentration-dependent manner. The swarming motility of A. oryzae RS-1 was also found to be significantly inhibited by Enfumafungin. Further validation through TEM observation revealed that bacterial cells exposed to Enfumafungin displayed mostly red fluorescence, indicating destruction of the bacterial cell membrane.

The present study evaluates the variations in the composition of targeted high-value components (phenolics, fatty acids, organic acids and natural sugars) and biological activities of olive fruits, collected at different maturity stages, from two locally developed cultivars BARI Zaitoon-1 (BZ1) and BARI Zaitoon-2 (BZ2). Ultrasound-assisted extraction (UAE) was employed to extract phenolic compounds. The oil yield was significantly increased during the maturation, while a decrease in bioactives extract yield was noted. The maximum oil yield (33.8 %) was recovered from BZ1 olives at the fully ripen stage, while the maximum extract yield (34.5 %) was given by BZ2 at the unripen stage. According to HPLC analysis, total phenolic acids, including sinapic, caffeic, gallic, chlorogenic and benzoic acids in BZ1 and BZ2 olives, were higher at semi-ripen and unripen stages, respectively. Myricetin was detected as the major flavonoid in both cultivars, but its concentration gradually decreased during fruit maturation. A good correlation of total phenolic contents (TPC) with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and total flavonoid contents (TFC) with ferric reducing antioxidant potential (FRAP) was observed. Similarly, an increase in the contents of total sugars and organic acids was noticed corresponding to fruit maturity. Furthermore, the tested olives fruit extracts inhibited the growth of Gram-positive bacterial strains (B. subtilis and S. aureus) more effectively than Gram-negative strains (K. oxytoca, S. typhi, and E. coli). The GC–MS analysis of extracted olive oil revealed that oleic acid was the major fatty acid found at three maturity stages for both cultivars. The current research revealed considerable variations in the selected components profile and biological activities of the tested olive cultivars as a function of fruit maturity. Based on these findings, the uses of olives can be recommended for specific applications to maximize the functional food and nutraceutical benefits of this valuable fruit.

Under the changing climate due to global warming, various abiotic stresses including drought (D) and salinity (S) are expected to further trigger their devastating effects on the already vulnerable crop production systems. This experiment was designed to unravel and quantify the potential role of exogenous application of salicylic acid (SA) in mitigating both D and S stresses and their combination (D+S), with three replications using CRD (Completely Randomized Design). The obtained results of the current study demonstrated significant effects of all three types of stresses (D, S, and D+S) on various parameters in Brassica napus plants. Quantifying these parameters provides a more informative and precise understanding of the findings. Current results revealed that all three stress types (D, S, and D+S) resulted in a reduction in leaf area (13.65 to 21.87%), chlorophyll levels (30 to 50%), gaseous exchange rate (30 to 54%) and the concentration of mineral ions compared to non-stressed plants. However, application of SA helped in mitigating these stresses by ameliorating the negative effects of these stresses. Moreover, Malondialdehyde (MDA) contents, an indicator of lipid per-oxidation and oxidative stress, the levels of antioxidants, proline content, an osmolyte associated with stress tolerance, and sugar content in the leaves were elevated in response to all stress conditions. In addition, the ultra-structures within the leaves were negatively affected by the stresses, while an application of SA considerably minimized the deterioration of these structures thus providing protection to the brassica plants against the stresses. In a nutshell, the findings of this study suggest that SA application in S, D and S+ D stresses provides evasion to the plants by improving different physiological and growth indices. The application of Salicylic Acid (SA) mitigated the negative effects of the stresses on all the above parameters, reducing MDA contents (47%), antioxidants (11 to 20%), proline (28%), sugar contents (20.50%), and minimizing the deterioration of ultra-structures. The findings emphasize the potential mitigatory role of SA in mitigating D and S stresses and highlight the need for further research to understand the underlying mechanisms in detail and explore its practical application in farming practices.

The moist temperate forests extend along the whole length of outer ranges of Himalaya between the subtropical pine forests and sub-alpine timberline formation with a rainfall from 400 mm to 800 mm. The altitudinal range is from 1500 m to 3000 m. The floristic variation in these Himalayan forests is poorly understood especially in the study area. Species composition may be either unpredictable or it may correspond to environmental heterogeneity. Vegetation from 144 stands in between 2000 – 2700 m altitude was sampled. Soil samples were collected to document edaphic conditions. Soils were physically and chemically analyzed. Ordination (DECORANA) and classificatory techniques were used to analyze the vegetation data. No clear disjunct vegetation patterns emerged from these analyses. The major axes brought out by the ordination were related to altitude and although it is possible to relate the units of classification to broad soil types. Soil chemical properties, litter cover and rockiness were significantly associated with the vegetation variation along an altitudinal gradient. The application of classification to the ordination allowed the interpretations of the vegetation variation in terms of topography and predictable climatic factors such as rainfall, wind speed and extent of snow accumulation. The vegetation patterns revealed have been discussed in relation to the general problem of plant community definition in continuous forest type. Our results demonstrate the overlapping rather than clearly discrete boundaries between the vegetation types and species distributions. This overlapping nature of the vegetation types are discussed in terms of overlapping environmental preferences of the species. Our results are consistent with notion that species separate edaphically and landscape scale within the uniform looking forest. This view maintains that differences in soil within the forest are distinct enough to favor different species, and thus create numerous floristically differentiated forest patches along the altitudinal gradients. The findings that non obvious but distinct floristic and edaphic variation exist within the Himalayan moist temperate forests at the landscape scale have important practical implications for forest management. In biodiversity conservation a high degree of habitat heterogeneity implies an increased need for wide-scale information on species distribution and endemism patterns to better assess where the different habitats are, which species they harbor, and where conservation efforts should be concentrated. The overlapping nature of vegetation types and hazy boundaries of the plant communities implies that plant ecologists must continue to attempt the difficult definition of hazy boundaries.

IntroductionClimate change, pest infestation, and soil degradation are significantly reducing wheat (Triticum aestivum L.) yield. Wheat is cultivated in rice-wheat and cotton-wheat cropping systems and escalating global population is exerting substantial pressure on the efficiency of these systems. Conservation tillage and crop rotation could help in lowering soil degradation and pest infestation, and improving wheat yield.MethodsThis three-year study evaluated soil properties, weed infestation and wheat yield under various tillage and cropping systems. Six different cropping systems, i.e., cotton-wheat, sorghum-wheat, mungbean-wheat, rice-wheat, sunflower-wheat, and fallow-wheat (control) and three tillage systems, i.e., conventional tillage (CT), zero-tillage (ZT) and minimum tillage (MT) were included in the study.ResultsThe individual and interactive effects of tillage and cropping systems significantly affected soil properties, weed infestation and yield of wheat crop. Overall, CT resulted in lower soil bulk density and higher porosity, while ZT behaved oppositely at all locations in this regard. Similarly, mungbean-wheat cropping system resulted in lower bulk density and higher porosity and nitrogen (N) contents, while fallow-wheat cropping system resulted in higher bulk density, and lower soil porosity and N contents. Similarly, ZT and CT resulted in higher and lower weed infestation, respectively. Likewise, lower and higher weed density and biomass were recorded in wheat-sorghum and wheat-fallow cropping systems, respectively at all locations. In the same way higher number of productive tillers, number of grains per spike, 1000-grain weight, grain yield, and economic returns of wheat crop were recorded for CT, whereas ZT resulted in lower values of these traits. Regarding interactions, wheat-mungbean cropping system with CT resulted in lower bulk density and higher porosity and N contents, whereas wheat-fallow system with ZT behaved oppositely at all locations in this regard. Similarly, higher and lower values for yield-related traits and economic returns of wheat crop were noted for mungbean-wheat cropping system under CT and fallow-wheat and sorghum-wheat cropping systems under ZT, respectively. It is concluded that the mungbean-wheat cropping system improved wheat productivity and soil health and sorghum-wheat cropping system could lower weed infestation. Therefore, these cropping systems can be practiced to lower weed infestation and improve wheat yield and economic returns.

Recently, the application of cobalt oxide nanoparticles (Co3O4NPs) has gained popularity owing to its magnetic, catalytic, optical, antimicrobial, and biomedical properties. However, studies on its use as a crop protection agent and its effect on photosynthetic apparatus are yet to be reported. Here, Co3O4NPs were first green synthesized using Hibiscus rosa-sinensis flower extract and were characterized using UV–Vis spectroscopy, Fourier transform infrared spectroscopy, X-ray diffraction (XRD), transmission/scanning electron microscopy methods. Formation of the Co3O4NPs was attested based on surface plasmon resonance at 210 nm. XRD assay showed that the samples were crystalline having a mean size of 34.9 nm. The Co3O4NPs at 200 µg/ml inhibited the growth (OD600 = 1.28) and biofilm formation (OD570 = 1.37) of Xanthomonas oryzae pv. oryzae (Xoo) respectively, by 72.87% and 79.65%. Rice plants inoculated with Xoo had disease leaf area percentage (DLA %) of 57.25% which was significantly reduced to 11.09% on infected plants treated with 200 µg/ml Co3O4NPs. Also, plants treated with 200 µg/ml Co3O4NPs only had significant increment in shoot length, root length, fresh weight, and dry weight in comparison to plants treated with double distilled water. The application of 200 µg/ml Co3O4NPs on the Arabidopsis plant significantly increased the photochemical efficacy of PSII (ΦPSII) and photochemical quenching (qP) respectively, by 149.10% and 125.00% compared to the control while the non-photochemical energy dissipation (ΦNPQ) was significantly lowered in comparison to control. In summary, it can be inferred that Co3O4NPs can be a useful agent in the management of bacterial phytopathogen diseases.

Verticillium dahliae is a soilborne fungal pathogen that causes vascular wilt diseases in a wide range of economically important crops, including eggplant. Trichoderma spp. are effective biological control agents that suppress a wide range of plant pathogens through a variety of mechanisms, including mycoparasitism. However, the molecular mechanisms of mycoparasitism of Trichoderma spp. in the degradation of microsclerotia of V. dahliae are not yet fully understood. In this study, the ability of 15 isolates of Trichoderma to degrade microsclerotia of V. dahliae was evaluated using a dual culture method. After 15 days, isolate HZA14 showed the greatest potential for microsclerotial degradation. The culture filtrate of isolate HZA14 also significantly inhibited the mycelial growth and conidia germination of V. dahliae at different dilutions. Moreover, this study showed that T. virens produced siderophores and indole-3-acetic acid (IAA). In disease control tests, T. virens HZA14 reduced disease severity in eggplant seedlings by up to 2.77%, resulting in a control efficacy of 96.59% at 30 days after inoculation. Additionally, inoculation with an HZA14 isolate increased stem and root length and fresh and dry weight, demonstrating plant growth promotion efficacy. To further investigate the mycoparasitism mechanism of T. virens HZA14, transcriptomics sequencing and real-time fluorescence quantitative PCR (RT-qPCR) were used to identify the differentially expressed genes (DEGs) of T. virens HZA14 at 3, 6, 9, 12, and 15 days of the interaction with microsclerotia of V. dahliae. In contrast to the control group, the mycoparasitic process of T. virens HZA14 exhibited differential gene expression, with 1197, 1758, 1936, and 1914 genes being up-regulated and 1191, 1963, 2050, and 2114 genes being down-regulated, respectively. Among these genes, enzymes associated with the degradation of microsclerotia, such as endochitinase A1, endochitinase 3, endo-1,3-beta-glucanase, alpha-N-acetylglucosaminidase, laccase-1, and peroxidase were predicted based on bioinformatics analysis. The RT-qPCR results confirmed the RNA-sequencing data, showing that the expression trend of the genes was consistent. These results provide important information for understanding molecular mechanisms of microsclerotial degradation and integrated management of Verticillium wilt in eggplant and other crops.

Abstract In present studies we have evaluated mulching impact of Jatropha curcas leaves on soil health and yield of two wheat (Triticum aestivum L.) varieties Wadan-2017 (rainfed) and Pirsabak-2013 (irrigated) under imposed water stress. Mulch of Jatropha leaves was spread on the soil surface at the rate of 0, 1, 3 and 5 Mg ha−1 after seed germination of wheat. Water stress was imposed by skipping irrigations for one month at anthesis stage of wheat maintaining 40% soil field capacity. We found a significant decline in soil microbial biomass carbon (30.27%), total nitrogen (22.28%) and organic matter content (21.73%) due to imposed water stress in non-mulch plots. However, mulch application at 5 Mg ha−1 significantly improved soil organic matter (38.18%), total nitrogen (37.75%), phenolics content (16.95 mg gallic acid equivalents/g) and soil microbial biomass carbon (26.66%) as compared to non-mulch control. Soil health indicators like soil carbonates, bicarbonates, electrical conductivity, chloride ions and total dissolved salts were decreased by 5 Mg ha−1 mulch application. We noted a decline in yield indicators like spike weight (14.74%), grain spike−1 (7.02%), grain length (3.79%), grain width (3.16%), 1000 grains weight (6.10%), Awn length (9.21%), straw weight (23.53%) and total grain yield (5.98%) of wheat due to imposed water stress. Reduction in yield traits of wheat due to water stress was higher in Pirsabak-2013 than Wadan-2017. Jatropha leaves mulch application at 5 Mg ha−1 significantly minimized the loss in yield traits of wheat crop caused by water stress. Jatropha curcas leaves mulch application at 5 Mg ha−1 is recommended for the successful establishment of wheat crop under water deficit conditions.

Abstract The sustainability of the rice–wheat system is threatened due to the deterioration of soil health and emergence of new challenges of climate change caused by low nutrient use efficiency and large scale burning of crop residues. The conservation agriculture based on tillage intensity, crop residue retention and raising green manuring (GM) crops during the intervening period between wheat harvest and rice establishment offers opportunities for restoration of phosphorus (P) dynamics and stimulate phosphatase activities within the macro-and micro-aggregates. Phosphorus and phosphatase activities in the soil aggregates affected by different residue management practices remain poorly understood. Thus, soil samples were obtained after a five-year field experiment to identify the effect of tillage, green manure and residue management on aggregate-associated phosphorus fractions. Four main plot treatments in rice included combination of wheat straw and GM were conventional till puddled transplanted rice (PTR) with no wheat straw (PTRW0), PTR with 25% wheat stubbles retained (PTRW25), PTR without wheat straw and GM (PTRW0 + GM), and PTR with wheat stubbles and GM (PTRW25 + GM). Three sub-plots treatments in the successive wheat crop were conventional tillage (CT) with rice straw removed (CTWR0), zero tillage (ZT) with rice straw removed (ZTWR0) and ZT with rice straw retained as surface mulch (ZTWR100). Results of the present study revealed significantly higher phosphorus fractions (HCl-P, NaHCO3-Pi and NaOH-Po) in treatment PTRW25 + GM and ZTWR100 compared with PTRW0/CTWR0 within both macro- and micro-aggregates. The total phosphorus (P), available P, alkaline phosphatase and phytin-P were significantly higher under ZTWR100 than CTWR0. The principal component analysis identified NaOH-Po, NaHCO3-Pi and HCl-P as the dominant and reliable indicators for evaluating P transformation within aggregates under conservation agriculture-based practices.

Potato cyst nematodes caused by Globodera rostochiensis, are quarantine-restricted pests causing significant yield losses to potato growers. The phytohormone ethylene play significant roles in various plant-pathogen interactions, however, the molecular knowledge of how ethylene influences potato–nematode interaction is still lacking. Precise detection of potato-induced genes is essential for recognizing plant-induced systemic resistance (ISR). Candidate genes or PR- proteins with putative functions in modulating the response to potato cyst nematode stress were selected and functionally characterized. Using real-time polymerase chain reaction (RT-PCR), we measured the quantified expression of four pathogenesis-related (PR) genes, PR2, PR3, peroxidase, and polyphenol oxidase. The activation of these genes is intermediate during the ISR signaling in the root tissues. Using different ethylene concentrations could detect and induce defense genes in infected potato roots compared to the control treatment. The observed differences in the gene expression of treated infected plants are because of different concentrations of ethylene treatment and pathogenicity. Besides, the overexpressed or suppressed of defense- related genes during developmental stages and pathogen infection. We concluded that ethylene treatments positively affected potato defensive genes expression levels against cyst nematode infection. The results emphasize the necessity of studying molecular signaling pathways controlling biotic stress responses. Understanding such mechanisms will be critical for the development of broad-spectrum and stress-tolerant crops in the future.

Rice bacterial leaf blight (BLB) is the most destructive bacterial diseases caused by Xanthomonas oryzae pv. oryzae (Xoo). Phages have been proposed as a green and efficient strategy to kill bacterial pathogens in crops, however, the mechanism of action of phages in the control of phyllosphere bacterial diseases remain unclear. Here, the glasshouse pot experiment results showed that phage combination could reduce the disease index by up to 64.3%. High-throughput sequencing technology was used to analyze the characteristics of phyllosphere microbiome changes and the results showed that phage combinations restored the impact of pathogen invasion on phyllosphere communities to a certain extent, and increased the diversity of bacterial communities. In addition, the phage combination reduced the relative abundance of epiphytic and endophytic Xoo by 58.9% and 33.9%, respectively. In particular, Sphingomonas and Stenotrophomonas were more abundant. According to structural equation modeling, phage combination directly and indirectly affected the disease index by affecting pathogen Xoo biomass and phage resistance. In summary, phage combination could better decrease the disease index. These findings provide new insights into phage biological control of phyllosphere bacterial diseases, theoretical data support, and new ideas for agricultural green prevention and control of phyllosphere diseases.

IntroductionOsmoprotectant supplementation can be used as a useful approach to enhance plant stress tolerance. However, the effect of silymarin and clove fruit extract (CFE) on wheat plants grown under cadmium (Cd) stress has not been studied.MethodsWheat seeds were planted in plastic pots filled with ions-free sand. A ½-strength Hoagland’s nutrient solution was used for irrigation. Pots were treated with eight treatments thirteen days after sowing: 1) Control, 2) 0.5 mM silymarin foliar application [silymarin], 3) 2% CFE foliar application [CFE], 4) CFE enriched with silymarin (0.24 g silymarin L-1 of CFE) [CFE-silymarin], 5) Watering wheat seedlings with a nutritious solution of 2 mM Cd [Cd]. 6) Cadmium + silymarin, 7) Cadmium + CFE, and 8) Cadmium + CFE-silymarin. The experimental design was a completely randomized design with nine replicates. Results and discussionThe Cd stress decreased grain yield, shoot dry weight, leaf area, carotenoids, chlorophylls, stomatal conductance, net photosynthetic rate, transpiration rate, membrane stability index, nitrogen, phosphorus, and potassium content by 66.9, 60.6, 56.7, 23.8, 33.5, 48.1, 41.2, 48.7, 42.5, 24.1, 39.9, and 24.1%, respectively. On the other hand, Cd has an Application of CFE, silymarin, or CEF-silymarin for wheat plants grown under Cd stress, significantly improved all investigated biochemical, morphological, and physiological variables and enhanced the antioxidant enzyme activities. Applying CFE and/or silymarin enhanced plant tolerance to Cd stress more efficiently. Our findings suggest using CFE-silymarin as a meaningful biostimulator for wheat plants to increase wheat plants’ tolerance to Cd stress via enhancing various metabolic and physiological processes.

The evaluation of alien invasive plants (AIPs) environmental and ecological distribution patterns can provide important data as well as a theoretical approach for management methods such as invasion prevention, eradication, and control of established AIPs, and recognition of ecosystems at high threat of invasion. Twenty 100 m2 transects were randomly established at each of the ten sample sites to collect data on the impact of edaphic properties on the spread of alien species in the semi-arid region. After computing the Important Value Index for each recorded species, ordination and indicator species analyses were carried out. In the study area, 104 alien plants from 30 families and 72 genera were found. Poaceae was the largest family, with 18 taxa (17%), followed by Fabaceae (14 taxa; 13%) and Asteraceae (11 taxa; 10%). Herbs made up 51% of these species, grasses 17%, and shrubs 12%. In terms of life form, Therophytes (56%) were dominant, followed by Chamaephytes (12%). The leaf size spectrum of alien plants was dominated by microphylls (48%), followed by mesophylls (23%), and nanophylls (20%). The CCA ordination analysis clearly showed that environmental variables like pH, moisture and soil texture (silt, sand, and clay) significantly (p ≤ 0.002) influenced how species are distributed in the region. In total, twenty indicator alien species were detected across five different semi-arid ecosystems. Parthenium hysterophorus recorded from all ecotypes except wetland with highest IVI from agroecosystem. Prosopis juliflora was observed in urban ecosystems, while Stellaria media was in both agroecosystems and forests, and Lantana camara was only observed in forests. Our findings reveal patterns and processes of alien invasive plant in different ecotypes, but only remediation and continuous monitoring might affect the situation in the near future. In this regard, the baseline data gathered during the current study would be significant in guiding future scientific monitoring of such ecological changes, as well as in landscape ecology management and planning.

Peptoids mimic the functions of peptides which have a side chain appended to amidic nitrogen instead of α carbon. This structural change in their backbone gives them increased resistance from proteolysis, improved biostability, greater immunogenicity, and better bioavailability. Therefore, they are specifically designed for various biological activities, including antibacterial, antifungal, antioxidant, antifouling, and anticancer properties. The aim of the research is the one-pot synthesis of quinoline-based peptoids 5(a-b) via Ugi-4CR by the reaction of 1R-(-)-myrtenal 1, benzylamine 2, quinoline-based carboxylic acids 3(a-b), and cyclohexyl isocyanide 4. These peptoids were characterized by FT-IR, 1H NMR, 13C NMR, and HR ESI-MS. In computational studies, the spectral results of 5(a-b) were compared with the calculated spectral values computed at B3LYP/6-311G (d,p) level. TD-DFT method was used to predict electron excitation of 5(a-b) and the contribution of orbitals. The electronic transition of peptoids from charge distribution was computed using natural bond order (NBO) analysis. NPA and MEP analysis was calculated to predict charge distribution in 5(a-b). The FMOs analysis was executed to calculate the global reactivity descriptor to predict the reactivity and stability of peptoids. DFT analysis showed that 5b was slightly more reactive than 5a due to extended conjugation. The biological activities were also predicted using an in silico approach that involved molecular docking and molecular dynamic (MD) simulations. The antiulcer, antibacterial, and antifungal activities were predicted based on ligand–protein binding interactions, binding energy calculations, and dissociation constants. 5(a-b) were evaluated in-vitro for anticholinesterase activity, and they showed 71% inhibition. The umbrella sampling was performed to probe ligand–protein binding.

Date palm is an economically important fruit crop in the arid and semi-arid regions of the world. The knowledge of genetic variation, genotype-by-traits comparison, as well as the relationship among several phenological traits is effective for developing breeding populations by choosing the best allelic combinations and employing selection approaches. Information regarding the genetic variability and inter-relationships among fruit characteristics is facilitating the plant breeders to efficiently exploit the date palm germplasm for the introduction of elite genotypes. The present study was conducted to examine genetic variation among different date palm cultivars -collected from two different research stations in Pakistan as well as the relationship among their phenological and biochemical characteristics. Biplot analysis revealed that the cultivars ‘Dhakki’, ‘Chohara’, and ‘Kupra’ possessed the highest fruit and pulp weights. Moreover, the fruits of the cultivars ‘Champa Kali’ and ‘Shakri’ had the maximum TSS (total soluble solids) and total sugar content. Similar variations were observed in the mean values of the studied phenological and biochemical characteristics as in the biplot analysis. Cluster analysis on the basis of phenological and biochemical characteristics divided 50 cultivars into seven clusters, showing differences in the studied characters. A correlation matrix revealed that pulp weight had a strong relationship with fruit weight, length, and diameter. A strong correlation also existed among antioxidant capacity, antioxidant activity, and total phenolic content. These results demonstrated a huge diversity in phenological and biochemical characteristics of date palm cultivars as well as a correlation among several characteristics of the studied germplasm, which can be exploited in future breeding programs.

We investigated the in silico characterization of short-length nucleotide sequences that were differentially expressed in dieback stress-induced transcriptomic analysis. They displayed homology with C-terminal flanking peptides and defensins-like proteins, revealing their antimicrobial activity. Their predicted fingerprints displayed protein signatures related to antimicrobial peptides. These short-length RGAs have been shown to possess structural motifs such as APLT P-type ATPase, casein kinase II (CK2), protein kinase 3, protein kinase C (PKC), and N-glycosylation site that are the attributes of disease resistance genes. The prediction of arginine and lysine residues in active binding sites in ligand docking analysis prophesied them as antimicrobial peptides due to their strong relation with antimicrobial activity. The in silico structural–functional characterization has predicted their role in resistance against microbial pathogens. Moreover, the predicted antimicrobial peptide regions showed their homology with the signature domain of PR-5-like protein and AMP family Thaumatin

Dalbergia sissoo is one of the most economically important trees in forestry, agroforestry, and horticulture. This tree species is severely threatened by dieback. Widespread dieback outbreaks and infestations have drastically destroyed billions of D. sissoo trees. Hence, we attempted to resolve the dieback etiology through phylogenomics associated with D. sissoo mortality. The Ceratocystis species was evaluated using morphologically investigated fungal isolates collected from dieback-affected tissue plants. Based on the symptomatology, we have differentiated dieback from Fusarium wilt and concluded that the Ceratocystis fimbriata sensu lato complex is causing shisham dieback in Pakistan. As the Ceratocystis species complex is a cryptic species complex, we used genomics and phylogenetic analysis for deciphering its evolutionary hierarchical order. The pathogen’s operational taxonomy was unlocked with the help of phylogenomics, and it was discovered that isolates from D. sissoo represent a species distinct from the other species in the C. fimbriata sensu lato species complex. The name Ceratocystis dalbergicans sp. nov. has been given to the fungus causing dieback disease in D. sissoo.

Abstract The current study was performed on 8 years old "Succary" pomegranate cultivar (Punica granatum L.) during the 2019 and 2020 seasons. One hundred pomegranate trees were chosen and sprayed three times at the beginning of flowering, full bloom, and 1 month later with the following treatments: water as control, 0.025, 0.05 and 0.1 mg/L Se; 5 mL/L, 7.5 and 10 mL/L Ag NPs, and 0.5, 1 and 2 mg/L K2Si2O5. The results showed that spraying of SE, Ag NPs, and K2Si2O5 ameliorated the shoot length, diameter, leaf chlorophyll content, set of fruiting percentage, and fruit yield per tree and hectare compared to control through studying seasons. Moreover, they improved the fruit weight, length, and diameter, as well as total soluble solids, total, reduced, and non-reduced sugars percent, while they lessened the juice acidity percentage compared to control. The most obvious results were noticed with Se at 0.1 mg/L, Ag NPs at 10 mL/L, and K2Si2O5 and K2Si2O5 in both experimental seasons over the other applied treatments. By HPLC analysis, peel extracts showed the presence of several bioactive compounds of catechol, syringic acid, p-coumaric acid, benzoic acid, caffeic acid, pyrogallol, gallic acid, ferulic acid, salicylic acid, cinnamic acid, and ellagic acid. The extracts applied to Melia azedarach wood showed promising antifungal activity against Rhizoctonia solani and were considered wood-biofingicides.

Nanoparticles have a positive impact in several subjects especially in agriculture, while their safety is still being debated. Numerous commercial nano pesticide, insecticides, and fertilizers products are found in the local markets without any intensely studies on the side effect of these products on plant, human as well as environmental effects. The present study aimed to evaluate the genotoxicity of commercial amino zinc nanoparticles (AZ NPs) on Triticum aestivum L. during seeds germination and root elongation using concentration ranges (50, 100, and 150 ppm) at different exposure times (8, 16 and 24 hrs). Long term exposure to AZ NPs, exhibited only slight variation in germination rates and the elongation of roots was affected by AZ NPs treatment ranged from 97.66 to 100%. Significant reduction in the mitotic index was 35.33% after 24 hrs and 150 ppm of AZ NPs, was also observed comparing with control which was 88.0%. Genotoxicity was evaluated at a cytological level in root meristems that revealed sever variations in mitotic activity, chromosomal aberrations, and micronuclei release. Results exhibited that nano amino zinc could enter effortlessly into the cells and inhibit the normal cellular function. The decrease in the emergence of chromosomal aberrations resulting from AZ NPs exposure in a dose-dependent manner was clearly indicated that AZ NPs has induced genotoxic effect on wheat root tips.

Minerals play a dynamic role in plant growth and development. However, most of these mineral nutrients are unavailable to plants due to their presence in fixed forms, which causes significant losses in crop production. An effective strategy to overcome this challenge is using mineral solubilizing bacteria, which can convert insoluble forms of minerals into soluble ones that plants can quickly assimilate, thus enhancing their availability in nutrient-depleted soils. The main objective of the present study was to isolate and characterize mineral solubilizing rhizobacteria and to assess their plant growth-promoting potential for Rhodes grass. Twenty-five rhizobacterial strains were isolated on a nutrient agar medium. They were characterized for solubilization of insoluble minerals (phosphate, potassium, zinc, and manganese), indole acetic acid production, enzymatic activities, and various morphological traits. The selected strains were also evaluated for their potential to promote the growth of Rhodes grass seedlings. Among tested strains, eight strains demonstrated strong qualitative and quantitative solubilization of insoluble phosphate. Strain MS2 reported the highest phosphate solubilization index, phosphate solubilization efficiency, available phosphorus concentration, and reduction in medium pH. Among tested strains, 75% were positive for zinc and manganese solubilization, and 37.5% were positive for potassium solubilization. Strain MS2 demonstrated the highest quantitative manganese solubilization, while strains MS7 and SM4 reported the highest solubilization of zinc and potassium through acidifying their respective media. The strain SM4 demonstrated the most increased IAA production in the presence and absence of L-tryptophan. The majority of strains were positive for various enzymes, including urease, catalase protease, and amylase activities. However, these strains were negative for coagulase activity except strains SM7 and MS7. Based on 16S rRNA gene sequencing, six strains, namely, SM2, SM4, SM5, MS1, MS2, and MS4, were identified as Bacillus cereus, while strains SM7 and MS7 were identified as Staphylococcus saprophyticus and Staphylococcus haemolyticus. These strains significantly improved growth attributes of Rhodes grass, such as root length, shoot length, and root and shoot fresh and dry biomasses compared to the uninoculated control group. The present study highlights the significance of mineral solubilizing and enzyme-producing rhizobacterial strains as potential bioinoculants to enhance Rhodes grass growth under mineral-deficient conditions sustainably.

Microbial compost plays a crucial role in improving soil health, soil fertility, and plant biomass. These biofertilizers, based on microorganisms, offer numerous benefits such as enhanced nutrient acquisition (N, P, and K), production of hydrogen cyanide (HCN), and control of pathogens through induced systematic resistance. Additionally, they promote the production of phytohormones, siderophore, vitamins, protective enzymes, and antibiotics, further contributing to soil sustainability and optimal agricultural productivity. The escalating generation of organic waste from farm operations poses significant threats to the environment and soil fertility. Simultaneously, the excessive utilization of chemical fertilizers to achieve high crop yields results in detrimental impacts on soil structure and fertility. To address these challenges, a sustainable agriculture system that ensures enhanced soil fertility and minimal ecological impact is imperative. Microbial composts, developed by incorporating characterized plant-growth-promoting bacteria or fungal strains into compost derived from agricultural waste, offer a promising solution. These biofertilizers, with selected microbial strains capable of thriving in compost, offer an eco-friendly, cost-effective, and sustainable alternative for agricultural practices. In this review article, we explore the potential of microbial composts as a viable strategy for improving plant growth and environmental safety. By harnessing the benefits of microorganisms in compost, we can pave the way for sustainable agriculture and foster a healthier relationship between soil, plants, and the environment.

Abstract Recent investigations of a Greco-Roman site at Sais have provided well-preserved archaeobotanical remains within a pile of metal fragments. The remains are compared with comparable modern taxa. The morphology and anatomy are studied using Light microscope (LM), Environmental scanning electron microscope (ESEM) and X-ray computed tomography (CT). To investigate the preservation mode, Energy dispersive spectroscopy (EDS) analysis and elemental mapping are conducted. Results revealed that the archaeobotanical remains are exhibiting close affinity with modern juniper cones. Although, the studied archaeobotanical remains are buried for more than 2 millenniums, they underwent early stages of silicification and copper mineralization. These results are discussed in relation to other excavated objects in the find and to our knowledge and understanding of daily life in the Greco-Roman period.

Background Azo dyes are recalcitrant organic pollutants present in textile industry effluents. Conventional treatment methods to remove them come with a range of disadvantages. Nanoparticles and their nanocomposites offer more efficient, less expensive and easy to handle wastewater treatment alternative. Methods In this study, nanoparticles of nickel oxide (NiO-NPs), copper oxide (CuO-NPs) and their nanocomposite (NiO/CuO-NC) were synthesized using co-precipitation method. The functional groups present on the surface of synthesized nanomaterials were verified using Fourier-transform infrared spectroscopy (FTIR). Surface morphology was assessed using scanning electron microscopy (SEM) whereas purity, shape and size of the crystallite were determined using X-ray diffraction (XRD) technique. The potential of these nanomaterials to degrade three dyes i.e., Reactive Red-2 (RR-2), Reactive Black-5 (RB-5) and Orange II sodium salt (OII) azo dyes, was determined in an aqueous medium under visible light (photocatalysis). The photodegradation effectiveness of all nanomaterials was evaluated under different factors like nanomaterial dose (0.02–0.1 g 10 mL−1), concentration of dyes (20–100 mg L−1), and irradiation time (60–120 min). They were also assessed for their potential to adsorb RR-2 and OII dyes. Results Results revealed that at optimum concentration (60 mgL−1) of RR-2, RB-5, and OII dyes, NiO-NPs degraded 90, 82 and 83%, CuO-NPs degraded 49, 34, and 44%, whereas the nanocomposite NiO/CuO-NC degraded 92, 93, and 96% of the said dyes respectively. The nanomaterials were categorized as the efficient degraders of the dyes in the order: NiO/CuO-NC > NiO-NPs > CuO-NPs. The highest degradation potential shown by the nanocomposite was attributed to its large surface area, small particles size, and quick reactions which were proved by advance analytical techniques. The equilibrium and kinetic adsorption of RR-2 and OII on NiO-NPs, CuO-NPs, and NiO/CuO-NC were well explained with Langmuir and Pseudo second order model, respectively (R2 ≥ 0.96). The maximum RR-2 adsorption (103 mg/g) was obtained with NiO/CuO-NC. It is concluded that nanocomposites are more efficient and promising for the dyes degradation from industrial wastewater as compared with dyes adsorption onto individual NPs. Thus, the nanocomposite NiO/CuO-NC can be an excellent candidate for photodegradation as well as the adsorption of the dyes in aqueous media.

Environmental stresses, including heavy metals accumulation, have posed an immense threat to the agricultural ecosystem, leading to a reduction in the yield of crucial crops. In this study, we evaluated the role of quercetin (Qu) in the alleviation of chromium (Cr) stress in Fenugreek (Trigonella corniculata L.). Different levels of Qu were prepared during the experiment, i.e., 15, 25, and 40 μM. For Cr toxification in potted soil, potassium chromate (K2Cr2O7) was used. Cr toxification reduced growth of T. corniculata seedlings. Cr stress also reduced fiber, ash, moisture, carbohydrate, protein, fats, and flavonoid contents. However, seed priming with Qu improved growth and physiochemical characteristics of T. corniculata seedlings grown in normal and Cr-contaminated soil. Seed priming with Qu escalated intercellular CO2 concentration, stomatal conductance, transpiration rate, and photosynthetic rate in T. corniculata seedlings. Application of Qu also increased the activity of antioxidative enzymes, i.e., superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and peroxidase (POD) in T. corniculata seedlings exposed to normal and Cr-contaminated soil. Application of Qu incremented the activity of SOD, POD, CAT, and APX, which were increased by 28, 22, 29, and 33%, respectively, in T. corniculata grown in Cr-toxic soil as compared to control treatment. Chromium stress alleviation was credited to the enhanced activity of the antioxidative defensive system in T. corniculata seedlings. It is proposed that Qu supplementation can be used to mitigate other abiotic stresses in plants.

Abstract In the present study, and for the waste valorization, Moringa oleifera seeds-removed ripened pods (SRRP) were used for papersheet production and for the extraction of bioactive compounds. Fibers were characterized by SEM–EDX patterns, while the phytoconstituents in ethanol extract was analyzed by HPLC. The inhibition percentage of fungal mycelial growth (IFMG) of the treated Melia azedarach wood with M. oleifera SRRP extract at the concentrations of 10,000, 20,000, and 30,000 µg/mL against the growth of Rhizoctonia solani and Fusarium culmorum was calculated and compared with fluconazole (25 µg). The produced papersheet was treated with the ethanol extract (4000, 2000, and 1000 µg/mL) and assayed for its antibacterial activity against Agrobacterium tumefaciens, Erwinia amylovora, and Pectobacterium atrosepticum by measuring the inhibition zones and minimum inhibitory concentrations (MICs). According to chemical analysis of M. oleifera SRRP, benzene:alcohol extractives, holocellulose, lignin, and ash contents were 7.56, 64.94, 25.66 and 1.53%, respectively, while for the produced unbleached pulp, the screen pulp yield and the Kappa number were 39% and 25, respectively. The produced papersheet showed tensile index, tear index, burst index, and double fold number values of 58.8 N m/g, 3.38 mN m2/g, 3.86 kPa m2/g, and 10.66, respectively. SEM examination showed that the average fiber diameter was 16.39 µm, and the mass average of for elemental composition of C and O by EDX were, 44.21%, and 55.79%, respectively. The main phytoconstituents in the extract (mg/100 g extract) by HPLC were vanillic acid (5053.49), benzoic acid (262.98), naringenin (133.02), chlorogenic acid (66.16), and myricetin (56.27). After 14 days of incubation, M. oleifera SRRP extract-wood treated showed good IFMG against R. solani (36.88%) and F. culmorum (51.66%) compared to fluconazole, where it observed 42.96% and 53.70%, respectively. Moderate to significant antibacterial activity was found, where the minimum inhibitory concentration (MIC) values were 500, 650, and 250 µg/mL against the growth of A. tumefaciens, E. amylovora, and P. atrosepticum respectively, which were lower than the positive control used (Tobramycin 10 µg/disc). In conclusion, M. oleifera SRRP showed promising properties as a raw material for pulp and paper production as well as for the extraction of bioactive compounds.

Dalbergia sissoo is an important timber tree, and dieback disease poses a dire threat to it toward extinction. The genomic record of D. sissoo is not available yet on any database; that is why it is challenging to probe the genetic elements involved in stress resistance. Hence, we attempted to unlock the genetics involved in dieback resistance through probing the NBS-LRR family, linked with mostly disease resistance in plants. We analyzed the transcriptome of D. sissoo under dieback challenge through DOP-rtPCR analysis using degenerate primers from conserved regions of NBS domain-encoded gene sequences. The differentially expressed gene sequences were sequenced and in silico characterized for predicting the expressome that contributes resistance to D. sissoo against dieback. The molecular and bioinformatic analyses predicted the presence of motifs including ATP/GTP-binding site motif A (P-loop NTPase domain), GLPL domain, casein kinase II phosphorylation site, and N-myristoylation site that are the attributes of proteins encoded by disease resistance genes. The physicochemical characteristics of identified resistance gene analogs, subcellular localization, predicted protein fingerprints, in silico functional annotation, and predicted protein structure proved their role in disease and stress resistance.
