10. Activism, refusal, expertise : responses to digital ubiquity

Abstract

In this thesis I explore if and how digital technologies capture human potential in terms of economic value, and how this shapes imaginaries, subjectivities, and forms of relationality. I investigate ‘digital ubiquity' as both an epistemological and material condition, and how this is manifest in the discourses that theorise, narrativise, advocate and oppose digital technologies in their many facets. I relay these considerations to Mark Fisher's notion of capitalist realism, and address how and where digital technologies - as extensions of capital - materialise capitalist realism as a ubiquitous force in everyday life (2009, p. 2). I consider how these dynamics influence imaginaries for the future, and whose politics and futures are privileged via the epistemic and material conditions of contemporary digital environments. The thesis is comprised, chiefly, of three critical case studies, each offering a response to various dynamics associated with ‘digitally ubiquity'. The first case study explores an ethnography I undertook at a ‘digital detox' camp, which was a space that invited its participants to relinquish use of their digital devices and discussion of their professions for a four-day period. Here, I examine the pertinence of why a ‘detox' from digital technology and work were encompassed with one another, and how the participants of the ‘digital detox' understood digital technologies - or the lack thereof - to influence their behaviours, interpersonal relationships, and senses of self. The second case study explores prospective ‘blockchain for good' initiatives, which are often offered as forms of techno-solutionism to issues surrounding financial inclusion and digital identity. I focus primarily upon how relationships between bodies and digitality are articulated within ‘blockchain for good' discourses, and how these relationships speak to specific structures of power: not simply in terms of an intensification of big data by Western apparatuses, but also in terms of the epistemological and ideological ‘erasures' that emerge when populations are rendered digitally legible in this way (Vazquez, 2011). The final case study explores the discourses employed by a left-wing think-tank entitled Autonomy, who critique the role of work within the UK and pose suggestions for prospective ‘post-work' futures. Here, I continue my considerations around interrelationships between work and identity formation. I further explore if and where neoliberal discourse is invoked within Autonomy's work, reflecting upon how this relates to articulations of ‘radical' politics within a cultural context of ‘capitalist realism'.

11. Exploring student victimisation and wellbeing in the UK higher education context

Abstract

Research into student bullying in Higher Education (HE) has been limited and most is based on childhood bullying research. Bullying in HE could disrupt student mental health and wellbeing at university - a topic that has recently gained traction. Additionally, marginalised student groups may be more at risk (e.g. LGB+). Focus groups were used to explore students’ conceptualisations of bullying and identify differences between childhood and emerging adulthood (EA) bullying behaviour. Themes identified were power imbalances; tactics of HE bullying; bullying for personal or social gain, and; justifications and minimisations for not intervening (bystander intervention). Examples of bullying behaviour from this first study supplemented the childhood and adult bullying literature to create a new HE bullying scale. The scale was tested on two samples to identify the factor structure (N = 243, N =304). The third survey study (N = 441) adopted a correlational design using the developed scale alongside measures of wellbeing, childhood victimisation, and potential mediator variables, such as university belongingness, social connectedness, and Internal Working Models (IWMs). Group differences were found on victimisation, IWMs, social connectedness, and university belonging, especially for SES and sexual orientation, evidencing the disadvantages that minority groups may suffer within HE. Regression-based path-analyses found that IWMs, university bullying, social connectedness, and university belonging mediated the links between childhood victimisation and current wellbeing. Finally, UK university anti-bullying policies were examined. Policy length and quality varied between universities and a review of content is recommended based on the earlier study findings. This research has produced a new university bullying scale and has also explored mechanisms through which childhood victimisation may have negative effects on current HE student wellbeing. The importance of self- and other- beliefs as well as the social context (e.g. belonging) is stressed.

16. Functionalized gold nanoparticles for cellular uptake

Abstract

Nanoparticles attract vast international research interest for medical applications due to their ability to be manipulated via the incorporation of various ligands and ligands. These ligands enable the regulation of a diverse range of biomedical processes which take place at a nanometer scale. Two approaches are commonly used to co-functionalize AuNPs, these methods include synthesizing either a mixed monolayer or by using a bifunctional Polyethylene glycol (PEG) linker. This Thesis directly compares the two approaches by incorporating the use of an internalization and endosomal escape peptide to compare the two arrangements for stability, cell viability, internalization and radio sensitization. An extensive research effort employed to confirm peptide arrangement has revealed that difficulties arise due to the higher binding affinity of thiolated ligands to gold in comparison to amine terminated ligands which are commonly used to synthesize a linker surface arrangement. It is shown that the successful synthesis using a bifunctional linker is heavily dependent on the method of attachment coupled with the solvents used. Inductively coupled plasma mass spectrometry (ICP-MS) has indicated a statistical difference for internalization, demonstrating that the linker has elicited a higher internalization response for both cell lines with an overall F value of 181.8 and a P value of 0.0095 (P < 0.05 statistically significant) for the DU145 cell lines resulting in over 3 times the amount of gold within the cells. Stability studies and cell viability studies have also indicated enhanced stability for the linker samples and preliminary data obtained from a clonogenic survival assay has indicated enhanced radiosensitivity, with an SER 1.36 for the linker, which represents a 36% increase in radiosensitivity for the samples in comparison to the untreated control. Concurrent attachment studies have identified that cysteine binding efficiency can be optimized by altering the pH of an AuNP suspension to ̴ 8, while extensive collaboration efforts have established that the solvents used to attach the peptide and/or ligands is paramount to a successful, stable covalent attachment, and an essential criteria for successful synthesis using a linker using a pre-synthesized NHS ester.

19. Student Anti-Bullying and Harassment Policies at UK Universities

Abstract

This article provides the first analysis of all available UK university anti-bullying policies, summarising, comparing, and contrasting the content of policies acquired from university websites. The importance of anti-bullying policies is known from policy research in schools and workplaces but has previously not been investigated in Higher Education. A new coding framework and guidelines were developed to enable our analysis, and universities were given a score. Scores indicated variation between policies, suggesting some students may have inadequate support when consulting their university policy. The findings indicate that all universities should create and implement an anti-bullying policy. Students could be involved in the development of interventions or policies, as co-created initiatives may be more influential. University policy must be up-to-date, inclusive, comprehensive yet concise, and it must be publicised. We propose that our novel coding framework could be of use to policy makers and researchers investigating bullying policy within higher education.

20. Slow on the Draw: The Representation of Turtles, Terrapins and Tortoises in Children's Literature

Abstract

Children's picture books, both fiction and non-fiction, play a vital role in introducing the reader to the natural world. Here we examine the representation of turtles, terrapins and tortoises (Testudines) in 204 English language picture books and find a mean of 3.9 (SD 9.1) basic biological errors per book. Only 83 (40.7%) of the examined books were found to be error-free in the representation of Testudines, with no significant improvement in biological accuracy being observed over time (book publication date range 1974-2017). Suggestions are made as to how biological accuracy of children's literature could be improved to help foster children's understanding and wonder of the natural world. Fantasy and imagination have an important role within children's literature, but here it is argued that the books children read should support future generations having sufficient understanding of the natural world to imagine the solutions to current environmental problems. A role of children's picture books should not be to reinforce biological illiteracy.

Abstract

Fibroblast growth factor receptor 4 (FGFR4) has a role in many biological processes, including lipid metabolism, tissue repair, and vertebrate development. In recent years, FGFR4 overexpression and activating mutations have been associated with numerous adult and pediatric cancers. As such, FGFR4 presents an opportunity for therapeutic targeting which is being pursued in clinical trials. To understand the role of FGFR4 signaling in disease and development, we generated and characterized three alleles of fgfr4 knockout zebrafish strains using CRISPR/Cas9. To generate fgfr4 knockout crispants, we injected single-cell wildtype zebrafish embryos with fgfr4 targeting guide RNA and Cas9 proteins, identified adult founders, and outcrossed to wildtype zebrafish to create an F1 generation. The generated mutations introduce a stop codon within the second Ig-like domain of Fgfr4, resulting in a truncated 215, 223, or 228 amino acid Fgfr4 protein compared to 922 amino acids in the full-length protein. All mutant strains exhibited significantly decreased fgfr4 mRNA expression during development, providing evidence for successful knockout of fgfr4 in mutant zebrafish. We found that, consistent with other Fgfr4 knockout animal models, the fgfr4 mutant fish developed normally; however, homozygous fgfr4 mutant zebrafish were significantly smaller than wildtype fish at three months post fertilization. These fgfr4 knockout zebrafish lines are a valuable tool to study the role of FGFR4 in vertebrate development and its viability as a potential therapeutic target in pediatric and adult cancers, as well as other diseases. Abstract

ABSTRACT:Background and purpose:To assess cost-effectiveness of late time-window endovascular treatment (EVT) in a clinical trial setting and a “real-world” setting.Methods:Data are from the randomized ESCAPE trial and a prospective cohort study (ESCAPE-LATE). Anterior circulation large vessel occlusion patients presenting > 6 hours from last-known-well were included, whereby collateral status was an inclusion criterion for ESCAPE but not ESCAPE-LATE. A Markov state transition model was built to estimate lifetime costs and quality-adjusted life-years (QALYs) for EVT in addition to best medical care vs. best medical care only in a clinical trial setting (comparing ESCAPE-EVT to ESCAPE control arm patients) and a “real-world” setting (comparing ESCAPE-LATE to ESCAPE control arm patients). We performed an unadjusted analysis, using 90-day modified Rankin Scale(mRS) scores as model input and analysis adjusted for baseline factors. Acceptability of EVT was calculated using upper/lower willingness-to-pay thresholds of 100,000 USD/50,000 USD/QALY.Results:Two-hundred and forty-nine patients were included (ESCAPE-LATE:n= 200, ESCAPE EVT-arm:n= 29, ESCAPE control-arm:n= 20). Late EVT in addition to best medical care was cost effective in the unadjusted analysis both in the clinical trial and real-world setting, with acceptability 96.6%–99.0%. After adjusting for differences in baseline variables between the groups, late EVT was marginally cost effective in the clinical trial setting (acceptability:49.9%–61.6%), but not the “real-world” setting (acceptability:32.9%–42.6%).Conclusion:EVT for LVO-patients presenting beyond 6 hours was cost effective in the clinical trial setting and “real-world” setting, although this was largely related to baseline patient differences favoring the “real-world” EVT group. After adjusting for these, EVT benefit was reduced in the trial setting, and absent in the real-world setting.

Abstract

The processes that shape Earth’s surface are constantly in flux. Continuous movements of soil and water and growth of ecosystems are what create the perfect stillness in Ansel Adam’s landscapes. In this dissertation, we investigate how a landscape responds to changes in the world around it. The tectonic uplift of Puerto Rico continues to shape the critical zone of the Luquillo Mountains today. We find that it has modified patterns of groundwater emergence, such that the river network is entrenching into ridgelines, in part, via the flux of groundwater. We demonstrate this with measurements of groundwater discharge and the grain size of the sediment it carries, compared to long-term channel incision rates derived with the cosmogenic radionuclide 10Be. Rock uplift enhances erosion rates, and a long-held belief in Earth Science is that soil production processes are linked and will respond to an enhanced erosive flux. By applying 10Be dating to measure soil production rates in the evolving landscape of the Luquillo Mountains, we determined that there is no such functional relationship. We re-evaluated the methods and assumptions that have been applied to this question elsewhere, and determined that in fact, soil production is independent of soil depth across the globe. This implies that no safety net will save us from the massive loss of soil due to anthropogenically accelerated erosion. Even outside of managed landscapes, accelerated erosion under climate change is an imminent possibility. We demonstrate this with a record of physical erosion over 26 ky recorded by 10Be concentrations in the layers of floodplain sediments. Erosion rates are greatly accelerated by the predominance of landsliding that occurs due to the onset of intense, frequent tropical storms in the Caribbean. The floodplain sediments contain a contemporaneous record of ecosystem dynamics in the δ13C isotope ratios of buried organic material. Similar to physical erosion in the watershed, ecological dynamics shift to a disturbance-dominated state due to coupling with a more volatile climate.

Abstract

Satellite transmitters were deployed on three green turtles, Chelonia mydas, and two hawksbill turtles, Eretmochelys imbricata, nesting in the Lesser Antilles islands, Caribbean, between 2005 and 2007 to obtain preliminary information about the inter-nesting, migratory and foraging habitats in the region. Despite the extremely small dataset, both year-round residents and migrants were identified; specifically, (1) two green turtles used local shallow coastal sites within 50 km of the nesting beach during all of their inter-nesting periods and then settled at these sites on completion of their breeding seasons, (2) one hawksbill turtle travelled 200 km westward before reversing direction and settling within 50 km of the original nesting beach and (3) one green and one hawksbill turtle initially nested at the proximate site, before permanently relocating to an alternative nesting site over 190 km distant. A lack of nesting beach fidelity was supported by flipper tag datasets for the region. Tagging datasets from 2002 to 2012 supported that some green and hawksbill individuals exhibit low fidelity to nesting beaches, whereas other females exhibited a high degree of fidelity (26 turtles tagged, 40.0 km maximum distance recorded from original nesting beach). Individual turtles nesting on St Eustatius and St Maarten appear to exhibit behavioural plasticity in their inter-nesting behaviour and post-nesting migration routes in the eastern Caribbean. The tracking and tagging data combined indicate that some of the green and hawksbill females that nest in the Lesser Antilles islands are year-round residents, whilst others may nest and forage at alternative sites. 48. A Thematic Analysis of Students’ perceptions and experiences of bullying in Higher Education

Abstract

Bullying in higher education (HE) has been relatively under-researched; despite its likely prevalence and impact on student wellbeing there is scant understanding of students’ lived experiences of bullying. We conducted online and physical focus groups with UK HE students (40 undergraduates from 17 UK universities, mean age: 22), exploring their perceptions and experiences of bullying at university. Thematic analysis was used to identify key issues, specifically 1) the importance of a power imbalance and perpetuation of existing systemic inequality in a HE context; 2) bullying in HE is motivated by attainment of social and personal gains; 3) the tactics used to bully in HE resemble those seen in other contexts, but may be more nuanced; 4) bullying can be minimised and justified within HE, leading to its continued prevalence. We conclude that HE bullying shares features in common with school and workplace bullying, and with sexual harassment. However, further research is needed to accurately define and conceptualise bullying in this unique context. HE providers should consider attending to issues of power and inequality within their bullying and harassment policies. They should also ensure there is clear information and guidance to prevent and reduce bullying in universities.

Abstract

School bullying has been researched extensively, yet research on student bullying at university is still in the early stages and lacks valid measurement instruments. This paper outlines three studies conducted to develop a new scale to measure victimisation and perpetration at university (ultimately focusing on victimisation). Wider bullying literature from the school context and the workplace was consulted alongside an initial qualitative study exploring students’ perceptions of university bullying. For Study One, an Exploratory Factor Analysis on data from a sample of UK university students (N = 243) resulted in a reliable scale with four factors: (1) Psychological Victimisation, (2) Physical Act/Trace Victimisation, (3) Social Victimisation, and (4) Direct Verbal Victimisation. After modification, Study Two tested the altered structure of the scale on a new sample of UK university students (N = 304), finding two alternative two- and three-factor models. Study Three tested the competing models from the first two studies using Confirmatory Factor Analysis (N=441), finding the four-factor structure to be the best model out of the three, but with the scale requiring further work. Although none of the fit indices’ statistics were ideal, this is the first attempt to design a higher education bullying scale based on a multi-phase approach, which shows potential as a useful tool for measuring victimisation following further research.
