Here, the ability of human organoids derived from normal gastric, pancreas, liver, colon and kidney tissues to metabolise the environmental carcinogen benzo[a]pyrene (BaP) was investigated. While organoids from the different tissues showed varied cytotoxic responses to BaP, with gastric and colon organoids being the most susceptible, the xenobiotic-metabolising enzyme (XME) genes, CYP1A1 and NQO1, were highly upregulated in all organoid types, with kidney organoids having the highest levels. Furthermore, the presence of two key metabolites, BaP-t-7,8-dihydrodiol and BaP-tetrol-l-1, was detected in all organoid types, confirming their ability to metabolise BaP. BaP bioactivation was confirmed both by the activation of the DNA damage response pathway (induction of p-p53, pCHK2, p21 and γ-H2AX) and by DNA adduct formation. Overall, pancreatic and undifferentiated liver organoids formed the highest levels of DNA adducts. Colon organoids had the lowest responses in DNA adduct and metabolite formation, as well as XME expression. Additionally, high-throughput RT-qPCR explored differences in gene expression between organoid types after BaP treatment. The results demonstrate the potential usefulness of organoids for studying environmental carcinogenesis and genetic toxicology.

Microplastics are a widespread contaminant found not only in various natural habitats but also in drinking waters. With spectroscopic methods, the polymer type, number, size, and size distribution as well as the shape of microplastic particles in waters can be determined, which is of great relevance to toxicological studies. Methods used in studies so far show a huge diversity regarding experimental setups and often a lack of certain quality assurance aspects. To overcome these problems, this critical review and consensus paper of 12 European analytical laboratories and institutions, dealing with microplastic particle identification and quantification with spectroscopic methods, gives guidance toward harmonized microplastic particle analysis in clean waters. The aims of this paper are to (i) improve the reliability of microplastic analysis, (ii) facilitate and improve the planning of sample preparation and microplastic detection, and (iii) provide a better understanding regarding the evaluation of already existing studies. With these aims, we hope to make an important step toward harmonization of microplastic particle analysis in clean water samples and, thus, allow the comparability of results obtained in different studies by using similar or harmonized methods. Clean water samples, for the purpose of this paper, are considered to comprise all water samples with low matrix content, in particular drinking, tap, and bottled water, but also other water types such as clean freshwater., B. In this paper the authors adopt a constructivist approach to explain the efforts of reborderisation following the so-called ‘Refugee Crisis’ unfolding in the European Union after a sharp influx of refugees in 2015. One of the core principles of the European Union, the freedom of movement, is heavily challenged, through the perception of security threats and economic burden that is associated with the arrival of people seeking asylum in large numbers. Through a discourse analysis centring around the label ‘refugee’, which experienced a shift in meaning, this paper aims to display the driving social force that catalysed political actions to reintroduce borders between European Union Member States as a tool to recreate the illusion of control over the influx of people. Germany and France, as pioneers of the principle of freedom of movement in Europe, serve as empirical case studies for the efforts to reinstate control through reborderisation.

Determining microplastics in environmental samples quickly and reliably is a challenging task. With a largely automated combination of optical particle analysis, Fourier transform infrared (FT-IR), and Raman microscopy along with spectral database search, particle sizes, particle size distributions, and the type of polymer including particle color can be determined. We present a self-developed, open-source software package for realizing a particle analysis approach with both Raman and FT-IR microspectroscopy. Our software GEPARD (Gepard Enabled PARticle Detection) allows for acquiring an optical image, then detects particles and uses this information to steer the spectroscopic measurement. This ultimately results in a multitude of possibilities for efficiently reviewing, correcting, and reporting all obtained results.

Soil structure is closely linked to biological activities. However, identifying, describing and quantifying soil aggregates remain challenging. X‐ray micro computed tomography (X‐ray μCT) provides a detailed view of the physical structure at a spatial resolution of a few microns. It could be a useful tool to discriminate soil aggregates, their origin and their formation processes for a better comprehension of soil structure properties and genesis. Our study aims to (a) determine different X‐ray μCT‐based aggregate parameters for differentiating earthworm casts belowground (earthworm aggregates) from aggregates that are not formed by earthworms (non‐earthworm aggregates), and (b) to evaluate if these parameters can also serve as specific “tomographic signatures” for the studied earthworm species. For this purpose, we set up a microcosm experiment under controlled conditions during 8 weeks, including three species of earthworms tested separately: the epigeic Lumbricus rubellus, the anecic Lumbricus terrestris and the endogeic Allolobophora chlorotica. Our results show that X‐ray μCT analysis helps distinguish earthworm aggregates from non‐earthworm ones using (a) the relative volume of the components within aggregates and (b) the volumetric mass of aggregates and their global volume. In particular, the volume ratio of mineral grains within the aggregates is significantly different according to earthworm species. So, X‐ray μCT is a powerful and promising tool for studying the composition of earthworm casts and their formation. However, future research is needed to take into account the shapes and spatial distribution of the aggregates' components, in particular the different states of organic matter decomposition. HIGHLIGHTS: Can earthworm belowground casts be differentiated from other soil aggregates using X‐ray μCT? The use of X‐ray μCT for characterizing soil aggregates at a spatial resolution of a few microns. A combination of X‐ray μCT variables discriminates earthworm casts from non‐earthworm aggregates. X‐ray μCT, used alone, is relevant for defining species‐specific signatures of earthworm casts.

Microplastic (MP) appears to be omnipresent in the atmosphere, raising concerns about dispersion across environmental compartments, ecological consequences and human health risks by inhalation. To date, data on the sources of atmospheric MP and deposition to river catchment areas are still sparse. We, therefore, took aerosol and total atmospheric deposition samples in the catchment area of the large German river Weser to estimate microplastic deposition fluxes (DFs) at six specific sites and airborne MP concentrations. Sampling in rural, suburban, and urban environments and wastewater treatment plants (WWTPs) was performed, aiming at a variation in airborne MP pollution and elucidating potential MP source areas. Aerosol samples were taken twice in April and October while monthly total deposition samples were collected over a period from March to October. Microplastics were detected in all analysed aerosol samples by Raman spectroscopy down to 4 μm, and in all 32 total deposition samples by μFT-IR down to 11 μm. Average MP number concentrations of 91 ± 47 m−3 were found in aerosol samples. The measured total MP number DFs ranged between 10 and 367 N m−2 day−1 (99 ± 85 mean ± SD) corresponding to total deposition of 0.05 ± 0.1 kg ha−1 per year and to an estimated 232 metric tons of plastic being deposited in the Weser River catchment annually. MP number DFs were higher in urban than rural sites. An effect of WWTPs on the MP abundance in air was not observed. Polypropylene, polyethylene, polyethylene terephthalate, polyvinyl chloride, polystyrene, and silicone fragments were found as the predominant polymer types in total deposition samples, while polyethylene particles dominated in aerosol samples. The results suggest that proximity to sources, especially to cities, increase the numbers of MP found in the atmosphere. It further indicates that atmospheric MP considerably contributes to the contamination of both aquatic and terrestrial habitats.

The treatment of samples for microplastic (MP) analysis requires purification steps that sufficiently reduce the non-MP content while preserving the targeted particles integrity. Besides their macromolecular structure this also encompasses their in situ numbers and sizes. However, any step of sample manipulation will come at a cost: particle loss, fragmentation, coagulation or degradation may lead to distorted results, predominantly in the smaller fraction of the MP size range. Therefore, the evaluation of MP resistivity against applied methods such as chemical digestions is a vital criterion for obtaining meaningful results on MP content of a sample. We developed a framework to test the applicability of MP purification methods and apply it to four protocols commonly used to prepare environmental samples for MP particle identification. The approach was designed for MP particles being too small to be handled manually (i.e. 10–70 μm). The evaluation consists of a two-tiered assay: a simple particle suspension approach is used to confirm a post-treatment qualitative recognisability of the target polymers by the analysis method of choice (here Raman and FTIR). In a following quantitative part, immobilised particles are used to evaluate the preservation of particle numbers and areas after the treatment on an individual particle level. A Python image analysis package was written that identifies, matches and measures particles on pairs of pre- and post-treatment images, and is available as open source software. Our results show that the chemical digestions using hydrogen peroxide, cooled Fenton’s and a combined alkaline / oxidative treatment using potassium hydroxide and sodium hypochlorite are suitable methods for preparing MP samples for a microspectroscopic analyses. Also acidic sodium polytungstate solution used for MP density separations and a pentane based protocol for lipid removal were found applicable for small sized MP. Certain degradative effects were found when acrylonitrile butadiene styrene is exposed to acidic treatments, as well as for MP from acrylate and epoxy based paint resins in strong oxidative regimes. Several paint resins tested here were spectroscopically not identifiable by polymer attributed bands even before treatment, indicating that these materials might slip through analyses of environmental samples and consequently being underreported. While it is now appreciated that the millions of tons of plastic pollution travelling through marine systems carry complex communities of microorganisms, it is still unknown to what extent these biofilm communities are specific to the plastic or selected by the surrounding ecosystem. To address this, we characterized and compared the microbial communities of microplastic particles, nonplastic (natural and wax) particles, and the surrounding waters from three marine ecosystems (the Baltic, Sargasso and Mediterranean seas) using high-throughput 16S rRNA gene sequencing. We found that biofilm communities on microplastic and nonplastic particles were highly similar to one another across this broad geographical range. The similar temperature and salinity profiles of the Sargasso and Mediterranean seas, compared to the Baltic Sea, were reflected in the biofilm communities. We identified plastic-specific operational taxonomic units (OTUs) that were not detected on nonplastic particles or in the surrounding waters. Twenty-six of the plastic-specific OTUs were geographically ubiquitous across all sampled locations. These geographically ubiquitous plastic-specific OTUs were mostly low-abundance members of their biofilm communities and often represented uncultured members of marine ecosystems. These results demonstrate the potential for plastics to be a reservoir of rare and understudied microbes, thus warranting further investigations into the dynamics and role of these microbes in marine ecosystems. IMPORTANCE This study represents one of the largest comparisons of biofilms from environmentally sampled plastic and nonplastic particles from aquatic environments. By including particles sampled through three separate campaigns in the Baltic, Sargasso, and Mediterranean seas, we were able to make cross-geographical comparisons and discovered common taxonomical signatures that define the plastic biofilm. For the first time, we identified plastic-specific bacteria that reoccur across marine regions. Our data reveal that plastics have selective properties that repeatedly enrich for similar bacteria regardless of location, potentially shifting aquatic microbial communities in areas with high levels of plastic pollution. Furthermore, we show that bacterial communities on plastic do not appear to be strongly influenced by polymer type, suggesting that other properties, such as the absorption and/or leaching of chemicals from the surface, are likely to be more important in the selection and enrichment of specific microorganisms.

Microplastic (MP) has been detected ubiquitously in freshwater systems. Until now MP sampling, however, is predominantly based on short-term net or pumping and filtration systems which can only provide snapshots of MP abundance; especially in flowing water bodies. To improve representativeness in the determination of MP occurrences in these aquatic compartments, an integrative approach that covers larger water volumes for a longer period of time is required. In this regard, surface water supplied drinking water treatment plants (DWTPs) represent an opportunity. In DWTPs, suspended solids from thousands of cubic metres of raw water are continuously removed over several hours and enriched in coagulation/flocculation and filtration processes. Our hypothesis was that MP is also removed to a full extent, like suspended solids, and that an integrative approach for identification and quantification in raw water can be derived from the analysis of MP in the treatment sludge. To prove this hypothesis, treatment sludge from a riverside DWTP (Warnow river, North-Eastern Germany) was analysed for MP > 50 μm. A sample purification protocol overcoming potential matrix effects caused by coagulants and flocculants was developed and validated. MP was analysed using micro-Raman spectroscopy. MP occurrence determined for the Warnow river was compared with in situ reference sampling using an established pumping and filtration system at relatively stable flow conditions. As result, the number of MP particles derived from treatment sludge was extrapolated to 196 ± 42 m−3 for the Warnow river and is statistically insignificantly different from 233 ± 36 m−3 identified by conventional water sampling. In addition, the polymer distribution and particles shape indicated the validity of the integrative concept. Consequently, the determination of MP abundance for freshwater systems based on DWTP treatment sludge represents an adequate method to estimate MP concentrations in flowing waters in an integrative way.

Proline has been widely used for various cocrystallization applications, including pharmaceutical cocrystals. Combining enantiopure and racemic flurbiprofen and proline, we discovered 18 new crystal structures. Liquid-assisted grinding proved highly efficient to explore all the variety of crystal forms. A unique combination of state-of-the-art characterization techniques, comprising variable temperature in situ X-ray diffraction and in situ ball-milling, along with other physicochemical methods and density functional theory calculations, was indispensable for identifying all the phases. Analyzing the results of in situ ball-milling, we established a stepwise mechanism for the formation of several 1:1 cocrystals via an intermediate 2:1 phase. The nature of the solvent in liquid-assisted grinding was found to significantly affect the reaction rate and, in some cases, the reaction pathway.

Child-to-parent violence (CPV) is a social problem that remains vastly understudied compared with other forms of family violence. The aim of this study is to identify family and child risk and protective factors of CPV, and to investigate whether they differentially predict physical and verbal parent-directed violence among boys and girls. Predictors include parenting behavior during childhood (physical and verbal violence, warmth, monitoring) and respondents’ individual characteristics (suicidal ideation, self-control, problematic substance use). Data were examined from a large representative sample of ninth graders ( N = 6,444) in Lower Saxony, Germany. Bivariate analyses showed that female adolescents were more likely to aggress verbally, while no gender differences were found for physical CPV. Multilevel logistic regression models revealed that direct experiences of parental physical and verbal violence during childhood were among the strongest predictors of physical and verbal CPV, both among males and females. While parental monitoring was not significantly associated with CPV, parental warmth protected girls from physical parent-directed aggression. Furthermore, high self-control was protective against verbal CPV as well as boys’ physical CPV, while problematic substance use predicted physical violence toward parents in both sexes but only boys’ verbal CPV. Suicidal ideation was a risk factor of aggression in males only. Except for parental warmth, the importance of risk and protective factors did not substantially vary across child gender. These findings broaden our understanding of different family and child-related factors that either promote or prevent CPV. Specifically, they point to the importance of the parenting context and especially harsh discipline practices for the occurrence of both physical and verbal CPV.

The mechanochemical Knoevenagel condensation of malononitrile with p-nitrobenzaldehyde was studied in situ using a tandem approach. X-ray diffraction and Raman spectroscopy were combined to yield time-resolved information on the milling process. Under solvent-free conditions, the reaction leads to a quantitative conversion to p-nitrobenzylidenemalononitrile within 50 minutes. The in situ data indicate that the process is fast and proceeds under a direct conversion. After stopping the milling process, the reaction continues until complete conversion. The continuous and the stopped milling process both result in crystalline products suitable for single crystal X-ray diffraction.

A novel approach for the integration of π-conjugated polymers (CPs) into DNA-based nanostructures is presented. Using the controlled Kumada catalyst-transfer polycondensation, well-defined thiophene-based polymers with controllable molecular weight, specific end groups, and water-soluble oligoethylene glycol-based side chains were synthesized. The end groups were used for the easy but highly efficient click chemistry-based attachment of end-functionalized oligodeoxynucleotides (ODNs) with predesigned sequences. As demonstrated by surface plasmon resonance spectroscopy, the prepared block copolymers (BCPs), P3(EO)3T-b-ODN, comprising different ODN lengths and specific or repetitive sequences, undergo specific hybridization with complementary, thiol-functionalized ODNs immobilized on a gold surface. Furthermore, the site-specific attachment of the BCPs to DNA origami structures is studied. Mechanochemistry is increasingly used as a ‘green alternative’ for synthesizing various materials including pharmaceutical cocrystals. Herein, we present the mechanochemical synthesis of three new cocrystals containing the API carbamazepine (cocrystals CBZ:Indometacin 1:1, CBZ:Benzamide 1:1, and CBZ:Nifedipine 1:1). The mechanochemical reaction was investigated in situ documenting a fast and complete reaction within one minute. Online NMR spectroscopy proved the direct influence of the dissolution behaviour of the coformers to the dissolution behaviour of the API carbamazepine. The dissolution behaviour of the organic cocrystals is compared to the behaviour of the pure drug indicating a general applicability of this approach for detailed cocrystal dissolution studies.

A newly established in situ technique using Raman spectroscopy was employed for the detailed kinetic investigation of mechanochemical reaction pathways. This approach was applied for the systematic investigation of the direct influence of colliding balls on the reaction rate constants of a mechanochemical cocrystallization reaction. As a model reaction, the mechanochemical cocrystallization of felodipine and the coformer imidazole was investigated. Keeping the total ball mass constant by varying the number of milling balls, our study reveals that the impact of each single collision has a more significant influence on the reaction kinetics than expected.

The effect of the reactant powder mass on reaction times for the mechanochemical formation of a soft matter model system was studied by in situ PXRD. The syntheses were performed at a constant ball mass in a shaker mill with and without glassy SiO2 as an inert additive. Reaction times decreased with the increase of the ball to reactant ratio (BRR). The kinetic influence of the SiO2 powder was excluded. The decrease in the reaction time with decreasing mass of reactants was related to the rise in the stress energy transferred to the powder by a higher ball impact. The BRR had no effect on the induction time. But the product conversion was accelerated by raising the BRR. While a certain temperature is needed for the activation of reactants in the induction phase, the conversion of soft matter reactants is rather controlled by impact than temperature.

DNA origami method aims at the construction of nanometer sized bio-templates from a long single-stranded DNA molecule [1] , [2] . It allows to build a variety of different structures and functionalities arranged with a resolution of about 1 nm for devices like plasmonic antennas or optical waveguides [3] . Our contribution deals with the immobilization of DNA origami templates in well-defined micrometer structures on 6-inch silicon wafers aiming a large-scale fabrication. We present a process of surface structuring of hydrophobic diamond-like carbon and fluorocarbon–polymer layers onto hydrophilic SiO2. Further we introduce a procedure for selective DNA Origamis self-immobilization in these hydrophilic cavities

We present a first direct measurement of the temperature during milling combined with in situ Raman spectroscopy monitoring. The data reveal a low temperature increase due to the mechanical impact and clear temperature increases as a consequence of the reaction heat. Based on the data, temperature rises as postulated in the magma plasma and hot spot theory can be excluded for soft matter milling syntheses.

Microplastics in food is a relatively new research field with only few studies available so far. Scientists have been pointing out that some of these studies apply questionable analytical methods. Nevertheless, media often use such results to gain attention of the readers. It is therefore of particular significance, that only those scientific studies are published, clearly presenting valid data on the content of microplastics in food. Unfortunately, the study by Zuccarello et al. shows very critical aspects regarding analytical methods used and conclusions made. The applied procedure is not described and, therefore, does not allow any assessment by other groups, which is indispensable prerequisite of any scientific publication. Moreover, the analytical method used for the identification and quantification of microplastic particles – SEM-EDX – is not sound and not validated. Therefore, in our opinion the results on the contamination of bottled mineral water with microplastics published by Zuccarello et al. are more than questionable.

Benzamide has been known for its polymorphism for almost 200 years. Three polymorphic forms are described. To date, it was only possible to crystallize a metastable form in a mixture together with the thermodynamically most stable form I. A complete transformation of form I into the metastable form III by mechanochemical treatment has been achieved. Catalytic amounts of nicotinamide seeds were used to activate the conversion by mechanochemical seeding. NMR experiments indicated that the nicotinamide molecules were incorporated statistically in the crystal lattice of benzamide form III during the conversion. The transformation pathway was evaluated using in situ powder X-ray diffraction.

The stability of different theophylline cocrystals under milling conditions was investigated by competitive cocrystal reactions. To determine the most stable cocrystal form under milling conditions, the active pharmaceutical ingredient theophylline was either ground with two similar coformers (benzoic acid, benzamide, or isonicotinamide), or the existing theophylline cocrystals were ground together with a competitive coformer. All competitive reactions were investigated by in situ powder X-ray diffraction disclosing the formation pathway of the milling processes. On the basis of these milling reactions, a stability order (least to most stable) was derived: tp/bs < tp/ba < tp/ina < bs/ina.

Chronic central serous chorioretinopathy (CSC) is a vision-threatening eye disease for which there is still no approved treatment. Recent studies suggest that the corticosteroid pathway in the choroid is implicated in CSC pathogenesis, and that therapy with the aldosterone antagonist eplerenone improves clinical outcomes. However, there is still little clinical data to support this hypothesis. We performed a retrospective chart review to further investigate the clinical value of eplerenone treatment in patients with chronic CSC and to identify possible response predictors. Twenty-four patients with chronic CSC resistant to conventional therapy over at least 4 months were included in this retrospective study. Patients were initially treated with 25 mg/day of eplerenone administered orally for 1 week, followed by a sustained daily dose of 50 mg. The primary outcome measure was percentage of eyes achieving complete resolution of subretinal fluid (SRF), recorded by spectral domain optical coherence tomography (SD-OCT). Secondary outcomes included changes in central macular thickness (CMT) and best-corrected visual acuity (BCVA). Baseline SD-OCT images were also evaluated as possible predictors of treatment response. Twenty-nine percent of patients experienced complete resolution of SRF after a median of 106 days of treatment, while 33 % of patients showed a transient initial decrease in SRF, and 25 % failed to respond to treatment. Treatment had to be stopped in 13 % of patients because of adverse effects of the eplerenone treatment. In the study population, CMT decreased from 342 to 275 μm after treatment, which was associated with a modest improvement in mean BCVA from 0.35 to 0.3 logMar. The integrity of the ellipsoid zone and the retinal pigment epithelium (RPE) at baseline were associated with a tendency towards a favourable visual outcome. This study confirms the proposed clinical value of eplerenone for treating patients with therapy-resistant CSC. However, patients presenting widespread RPE changes are less likely to benefit from eplerenone treatment, which may argue for an earlier intervention. Larger studies are needed to characterise patient subgroups that may benefit the most from eplerenone treatment.

Uber die zugrundeliegenden Prozesse ist bislang wenig bekannt.

The liquid-assisted grinding cocrystallization of theophylline with benzamide leading to polymorphic compounds was investigated. A solvent screening with 17 different solvents was performed. The dipole moment of the solvent used in the synthesis determines the structure of the polymorphic product. A detailed investigation leads to the determination of the kinetically and thermodynamically favored product. In situ observations of the formation pathway during the grinding process of both polymorphs show that the thermodynamically favored cocrystal is formed in a two-step mechanism with the kinetic cocrystal as an intermediate.

The crystalline cadmium phenylphosphonates Cd(O3PPh)·H2O (1), Cd(HO3PPh)2 (2), and Cd(HO3PPh)2(H2O3PPh) (3) can be synthesized via solid state reactions by grinding together cadmium acetate with the respective equivalents of phenylphosphonic acid. Phosphonates (2) and (3) have not been obtained via any other synthesis method so far. The determination of the crystal structure of the new compounds (2) and (3) based on powder X-ray diffraction (PXRD) data is reported. The milling reactions were investigated using in situ synchrotron PXRD. Based on these data, an identification of intermediates and a detailed analysis of the underlying mechanisms were possible.

We present naproxen/proline cocrystals discovered when combining enantiopure and racemic naproxen and proline. Using liquid-assisted grinding as the main method to explore the variety of crystal forms in this system, we found 17 cocrystals, of which the structures of only four of them were previously known. The naproxen/proline system exhibited multiple polymorphs of 1 : 1 stoichiometry as well as more rare cocrystals with 1 : 2 and 2 : 3 stoichiometries, two cocrystal hydrates and one cocrystal solvate. In situ ball-milling, used to monitor liquid-assisted grinding reactions, revealed that the solvent dictates the reaction intermediates even if the final reaction product stays the same. Synchrotron X-ray diffraction data collected in situ upon heating allowed us to monitor directly the phase changes upon heating and gave access to pure diffraction patterns of several cocrystals, thus enabling their structure determination from powder X-ray diffraction data; this method also confirmed the formation of a conglomerate in the RS-naproxen/dl-proline system. Proline in cocrystals kept its ability to form charge-assisted head-to-tail N-H...O hydrogen bonds, typical of pure crystalline amino acids, thus increasing the percentage of strong charge-assisted interactions in the structure and consequently providing some of the cocrystals with higher melting points as compared to pure naproxen. The majority of drugs are chiral, and hence, these data are of importance to the pharmaceutical industry as they provide insight into the challenges of chiral cocrystallization.

The driving forces triggering the formation of co-crystals under milling conditions were investigated by using a set of multicomponent competitive milling reactions. In these reactions, different active pharmaceutical ingredients were ground together with a further compound acting as coformer. The study was based on new co-crystals including the coformer anthranilic acid. The results of the competitive milling reactions indicate that the formation of co-crystals driven by intermolecular recognition are influenced and inhibited by kinetic aspects including the formation of intermediates and the stability of the reactants.

We describe the design, synthesis and aggregation behaviour of a biomolecular breadboard in the form of a DNA origami frame. Our findings focus on determining the shortest thermal annealing protocol, yielding correctly folded frames. In addition, we investigate the frames’ chain formation behaviour due to base stacking. By minor alterations in the origami design, we can deliberately avoid or induce this behaviour and influence the relative orientation of frames in the chains. Aggregate formation of DNA origami frames using nucleic acid base stacking.

The structure and the formation pathway of a new theobromine:oxalic acid (2:1) cocrystal are presented. The cocrystal was synthesised mechanochemically and its structure was solved based on the powder X-ray data. The mechanochemical synthesis of this model compound was studied in situ using synchrotron XRD. Based on the XRD data details of the formation mechanism were obtained. The formation can be described as a self-accelerated (‘liquid like’) process from a highly activated species.

The combination of two analytical methods including time-resolved in situ X-ray diffraction (XRD) and Raman spectroscopy provides a new opportunity for a detailed analysis of the key mechanisms of milling reactions. To prove the general applicability of our setup, we investigated the mechanochemical synthesis of four archetypical model compounds, ranging from 3D frameworks through layered structures to organic molecular compounds. The reaction mechanism for each model compound could be elucidated. The results clearly show the unique advantage of the combination of XRD and Raman spectroscopy because of the different information content and dynamic range of both individual methods. The specific combination allows to study milling processes comprehensively on the level of the molecular and crystalline structures and thus obtaining reliable data for mechanistic studies.

The synthesis of the polymorphic cocrystal caffeine:anthranilic acid was investigated to obtain a better understanding of the processes leading to the formation of different polymorphic forms. In the case of these cocrystal polymorphs synthesized by liquid-assisted grinding a distinct influence of the dipole moment of the solvent was found. A pre-coordination between the solvent molecules and the caffeine:anthranilic acid cocrystal could be identified in the formation of form II. In the case of form II the solvent can be regarded as a catalyst. The formation pathway of each polymorph was evaluated using synchrotron X-ray diffraction.
