Abstract

CUORE is a 741 kg array of TeO2bolometers for the search of neutrinoless double beta decay of130Te. The detector is being constructed at the Laboratori Nazionali del Gran Sasso, Italy, where it will start taking data in 2015. If the target background of 0.01 counts/(keV.kg.y) will be reached, in five years of data taking CUORE will have a 1σ half life sensitivity of 1026y. CUORE-0 is a smaller experiment constructed to test and demonstrate the performances expected for CUORE. The detector is a single tower of 52 CUORE-like bolometers that started taking data in spring 2013. The status and perspectives of CUORE will be discussed, and the first CUORE-0 data will be presented.

Abstract

The CUORE (Cryogenic Underground Observatory for Rare Events) experiment is an array of 741 kg of TeO2bolometers to search for neutrinoless double beta decay (ββ0ν) of130Te. The detector is being constructed at the Laboratori Nazionali del Gran Sasso (Italy) where it will start operation in 2015. To test and demonstrate the possibility of realising such a large scale bolometric detector, a prototype (CUORE-0) has been realised. The CUORE-0 detector is a single tower of 52 CUORE-like bolometers. CUORE-0 data taking started in Spring 2013. The status of CUORE and the first CUORE-0 data are here reported.

Abstract

With 741 kg of TeO2crystals and an excellent energy resolution of 5 keV (0.2%) at the region of interest, the CUORE (Cryogenic Underground Observatory for Rare Events) experiment aims at searching for neutrinoless double beta decay of130Te with unprecedented sensitivity. Expected to start data taking in 2015, CUORE is currently in an advanced construction phase at LNGS. CUORE projected neutrinoless double beta decay half-life sensitivity is 1.6 × 1026y at 1σ (9.5 × 1025y at the 90 % confidence level), in five years of live time, corresponding to an upper limit on the effective Majorana mass in the range 40-100 meV (50-130 meV). Further background rejection with auxiliary bolometric detectors could improve CUORE sensitivity and competitiveness of bolometric detectors towards a full analysis of the inverted neutrino mass hierarchy. CUORE-0 was built to test and demonstrate the performance of the upcoming CUORE experiment. It consists of a single CUORE tower (52 TeO2bolometers of 750 g each, arranged in a 13 floor structure) constructed strictly following CUORE recipes both for materials and assembly procedures. An experiment its own, CUORE-0 is expected to reach a sensitivity to the ββ(0ν) half-life of130Te around 3×1024y in one year of live time. We present an update of the data, corresponding to an exposure of 18.1 kg y. An analysis of the background indicates that the CUORE performance goal is satisfied while the sensitivity goal is within reach.

Abstract

The CUORE (Cryogenic Underground Observatory for Rare Events) experiment will search for neutrinoless double beta decay of 130Te. With 741 kg of TeO2 crystals and an excellent energy resolution of 5 keV (0.2%) at the region of interest, CUORE will be one of the most competitive neutrinoless double beta decay experiments on the horizon. With five years of live time, CUORE projected neutrinoless double beta decay half-life sensitivity is 1.6 × 1026 y at 1σ (9.5 × 1025 y at the 90% confidence level), which corresponds to an upper limit on the effective Majorana mass in the range 40-100 meV (50-130 meV). Further background rejection with auxiliary light detector can significantly improve the search sensitivity and competitiveness of bolometric detectors to fully explore the inverted neutrino mass hierarchy with 130Te and possibly other double beta decay candidate nuclei.

Abstract

The CUORE (Cryogenic Underground Observatory for Rare Events) experiment will search for neutrinoless double beta decay in {Mathematical expression}Te. Observation of the process would unambiguously establish that neutrinos are Majorana particles as well as provide information about the absolute neutrino mass scale and mass hierarchy.The CUORE setup will consist of an array of 988 tellurium dioxide crystals (containing 206 kg of {Mathematical expression}Te in total), operated as bolometers at a temperature of {Mathematical expression}10 mK. The experiment is now under construction at the Gran Sasso National Laboratory in Italy. As a first step towards CUORE, a tower (CUORE-0) has been assembled and is taking data. Here a detailed description of the CUORE-0 tower and its performance is reported. The status of the CUORE experiment and its expected sensitivity will then be discussed. © 2014 Springer Science+Business Media New York.

Abstract

CUORE-0 is a cryogenic detector that uses an array of tellurium dioxide bolometers to search for neutrinoless double-beta decay of (Formula presented.). We present the first data analysis with (Formula presented.) of total (Formula presented.) exposure focusing on background measurements and energy resolution. The background rates in the neutrinoless double-beta decay region of interest (2.47 to (Formula presented.)) and in the (Formula presented.) background-dominated region (2.70 to (Formula presented.)) have been measured to be (Formula presented.) and (Formula presented.), respectively. The latter result represents a factor of 6 improvement from a predecessor experiment, Cuoricino. The results verify our understanding of the background sources in CUORE-0, which is the basis of extrapolations to the full CUORE detector. The obtained energy resolution (full width at half maximum) in the region of interest is (Formula presented.). Based on the measured background rate and energy resolution in the region of interest, CUORE-0 half-life sensitivity is expected to surpass the observed lower bound of Cuoricino with one year of live time.

Abstract

The CUORICINO experiment was an array of 62 TeO2 single-crystal bolometers with a total 130Te mass of 11.3kg. The experiment finished in 2008 after more than 3 yr of active operating time. Searches for both 0ν and 2ν double-β decay to the first excited 0+ state in 130Xe were performed by studying different coincidence scenarios. The analysis was based on data representing a total exposure of N(130Te)•t=9.5×1025yr. No evidence for a signal was found. The resulting lower limits on the half-lives are T122ν(130Te→130Xe*)>1. 3×1023yr (90% C.L.), and T120ν(130Te →130Xe*)>9.4×1023yr (90% CL). ©2012 American Physical Society.

Abstract

We present a study of the sensitivity and discovery potential of CUORE, abolometric double-beta decay experiment under construction at the LaboratoriNazionali del Gran Sasso in Italy. Two approaches to the computation ofexperimental sensitivity for various background scenarios are presented, and anextension of the sensitivity formulation to the discovery potential case isalso discussed. Assuming a background rate of 10^-2 cts/(keV kg y), we findthat, after 5 years of live time, CUORE has a 1 sigma sensitivity to theneutrinoless double-beta decay half-life of T_1/2(1 sigma) = 1.6 \times 10^26 yand thus a potential to probe the effective Majorana neutrino mass down to40-100 meV; the sensitivity at 1.64 sigma, which corresponds to 90% C.L., willbe T_1/2(1.64 sigma) = 9.5 \times 10^25 y. This range is compared with theclaim of observation of neutrinoless double-beta decay in 76Ge and thepreferred range of the neutrino mass parameter space from oscillation results.

Abstract

We report the final result of the CUORICINO experiment. Operated between 2003 and 2008, with a total exposure of 19.75 kg · y of 130Te, CUORICINO was able to set a lower bound on the 130Te 0νββ half-life of 2.8 × 1024 years at 90% C.L. The limit here reported includes the effects of systematic uncertainties that are examined in detail in the paper. The corresponding upper bound on the neutrino Majorana mass is in the range 300-710 meV, depending on the adopted nuclear matrix element evaluation. © 2011 Elsevier B.V. All rights reserved.

Abstract

We present a search for β+/EC double beta decay of 120Te performed with the CUORICINO experiment, an array of TeO2 cryogenic bolometers. After collecting 0.0573 kg · y of 120Te, we see no evidence of a signal and therefore set the following limits on the half-life: T1/20ν>1.9·1021 y at 90% C.L. for the 0ν mode and T1/22ν>7.6·1019 y at 90% C.L. for the 2ν mode. These results improve the existing limits by almost three orders of magnitude (four in the case of 0ν mode). © 2011 Published by Elsevier B.V.

Abstract

The search for neutrinoless double beta decay (DBD-0ν) is a powerful tool to establish the correct neutrino mass hierarchy and whether the neutrino is a Majorana or Dirac particle. The Milano group has run several experiments using thermal detectors to search for the 130Te DBD-0ν. The Cuoricino experiment consists of an array of 62 TeO2 thermal detectors for a total mass of about 40 kg, by far the largest cryogenic experiment in the world. The detector installation in the Gran Sasso Underground Laboratory has been recently completed. After a test phase the experiment is now taking data and we report here the preliminary results. Cuoricino is the first step toward the CUORE experiment, which will consists of 1000 TeO2 thermal detectors for a total mass of about 760 kg: in this paper we discuss also the physics potential of both stages for what concerns the DBD-0ν search. Abstract

The Cryogenic Underground Observatory for Rare Events (CUORE) is an experiment to search for neutrinoless double beta decay ($0\nu\beta\beta$) in $^{130}$Te and other rare processes. CUORE is a cryogenic detector composed of 988 TeO$_2$ bolometers for a total mass of about 741 kg. The detector is being constructed at the Laboratori Nazionali del Gran Sasso, Italy, where it will start taking data in 2015. If the target background of 0.01 counts/(keV$\cdot$kg$\cdot$y) will be reached, in five years of data taking CUORE will have an half life sensitivity around $1\times 10^{26}$ y at 90\% C.L. As a first step towards CUORE a smaller experiment CUORE-0, constructed to test and demonstrate the performances expected for CUORE, has been assembled and is running. The detector is a single tower of 52 CUORE-like bolometers that started taking data in spring 2013. The status and perspectives of CUORE will be discussed, and the first CUORE-0 data will be presented.

Abstract

The CUORICINO experiment was an array of 62 TeO2single-crystal bolometers with a total 130Te mass of 11.3kg. The experiment finished in 2008 after more than 3 yr of active operating time. Searches for both 0ν and 2ν double-β decay to the first excited 0+state in 130Xe were performed by studying different coincidence scenarios. The analysis was based on data representing a total exposure of N(130Te)•t=9.5×1025yr. No evidence for a signal was found. The resulting lower limits on the half-lives are T122ν(130Te→130Xe*)>1. 3×1023yr (90% C.L.), and T120ν(130Te→130Xe*)>9.4×1023yr (90% CL). ©2012 American Physical Society.

Abstract

A detailed description of the CUORICINO Te130 neutrinoless double-beta (0 νββ) decay experiment is given and recent results are reported. CUORICINO is an array of 62 tellurium oxide (TeO2) bolometers with an active mass of 40.7 kg. It is cooled to ~8-10 mK by a dilution refrigerator shielded from environmental radioactivity and energetic neutrons. It is running in the Laboratori Nazionali del Gran Sasso (LNGS) in Assergi, Italy. These data represent an exposure of 11.83 kg yr or 91 mole-years of Te130. No evidence for 0νββ-decay was observed and a limit of T1/20ν(130Te)≥3. 0×1024y (90% CL) is set. This corresponds to an upper limit on the effective mass, mν, between 0.19 and 0.68 eV when analyzed with the many published nuclear structure calculations. In the context of these nuclear models, the values fall within the range corresponding to the claim of evidence of 0νββ-decay by H. V. Klapdor-Kleingrothaus The experiment continues to acquire data.

Abstract

Thermal detectors are introduced and their possible impact on neutrinoless double-beta-decay (2β0ν) search is discussed. The thermal experiment MIBETA is described, reporting the up-to-date limits on 130Te and 128Te. A new generation experiment, consisting in a large expansion of MIBETA and known as CUORE, is presented and discussed.

Abstract

A large body of evidence suggests that the production of reactive oxygen species (ROS) can play an important role in ischemic neuronal injury. However any studies has been performed in hypoxic conditions. In the present experiments we studied using electron spin resonance (ESR) techniques the ROS release in neostriatum of newborn rats subjected to acute perinatal asphyxia (PA) followed by various periods of reoxygenation. Pregnant rats' uteri still containing foetuses were taken out and subjected to PA by immersion in a 37 degrees C water bath during the following periods of time: 5, 10, 15, 19 and 20 min. After performing PA, animals were recovered and ROS measured after 0, 5, 15, 30 or 60 min of reoxygenation. Then, pups were sacrificed, their neostriatum removed and homogenised with N-tert.-butyl-alpha-phenylnitrone (PBN) and diethylenetriamine-pentacetic acid (DPTA) in phosphate-buffered saline (PBS) and the formed complexes were extracted with ethyl acetate an analysed using an X-band ESR spectrometer. A significant release of ROS was detected at 19 and 20 min of PA after 5 min of reoxygenation. These data provide strong evidence that ROS could be involved in neuronal damage during PA.

Abstract

To better understand the contribution of cosmic ray muons to the CUORICINO background, 10 plastic scintillator detectors were installed at the CUORICINO site and operated during the final 3 months of the experiment. From these measurements, an upper limit of 0.0021 counts/(keV kg yr) (95% CL) was obtained on the cosmic ray-induced background in the neutrinoless double beta decay region of interest. The measurements were also compared to GEANT4 simulations.

Abstract

In the last 10 years bolometers have become extremely powerful detectors in the search for rare events, due to their very good resolution. Cuoricino, a 62 bolometer array, in the first months of activity reached a resolution (average in all detectors) in the energy region of interest (2528 keV) of 7 keV and a limit on the t1/2 of the process of 7.5·1023y. Cuoricino’s 62 detectors constitute the biggest number of macrobolometers (790 g each) ever cooled and demonstrate the feasibility of large arrays of bolometers. Following the indication of Cuoricino in CUORE R&D a new detector has been developed trying to improve the reproducibility of the detector’s performance, to increase the single detector mass (to reduce the number of channels) and to simplify the experiment assembly. The latest results are presented.

Abstract

After an introduction on the various experimental techniques to be adopted in searches for double beta decay, the new approach based on the use of cryogenic low temperature detectors is described. The present results are reported on the limit for neutrinoless double beta decay of 130Te obtained with the large bolometric detector CUORICINO. This setup consists of 44 cubic crystals of natural TeO2 with 5 cm sides and 18 crystals of 3×3×6 cm3. Four of these latter crystals are made with isotopically enriched materials: two in 128Te and two others in 130Te. With a sensitive mass of 40.7 kg, this array is by far the most massive running cryogenic detector. The array is operated at a temperature of ∼10 mK in a dilution refrigerator under a heavy shield in the Gran Sasso Underground Laboratory at a depth of about 3500 m.w.e. The counting rate in the region of neutrinoless double beta decay is 0.18±0.02 counts keV -1 kg-1 y-1, among the lowest in this type of experiment. No evidence for neutrinoless double beta decay is found. The corresponding lower limit for the lifetime of this process is 2×1024 years at 90% C.L. The resultant upper limit on the effective neutrino mass ranges between 0.2 and 1.0 eV, depending on the theoretically calculated nuclear matrix elements. This constraint is the most restrictive one, except for those obtained with Ge diodes, and is comparable to them. The second part of this report is devoted to the present status of the construction of the larger experiment CUORE (Cryogenic Underground Observatory for Rare Events) formed from 988 bolometers with a cubic TeO2 absorber of size 5×5×5 cm3, with a total mass of ∼750 kg. We present technical details of the CUORE setup as well as of its location and our efforts to reduce radioactive backgrounds. © 2006.

Abstract

CUORE is a 0.75 ton experiment to search for neutrinoless double beta decay of Te130 using 988 TeO2 bolometers. It aims at reaching a sensitivity on the effective neutrino mass of the order of few tens of meV. CUORICINO, a single CUORE tower running since 2003, plays an important role as a stand alone experiment and for developing the future CUORE setup. Present results already achieved and studies that are underway are here presented and discussed.

Abstract

We report the present results of the CUORICINO experiment, a cryogenic experiment on neutrinoless Double Beta Decay (DBD) of 130Te consisting of an array of 62 TeO2 crystals with a total active mass of 40:7 kg. The array is framed inside a dilution refrigerator, heavily shielded against environmental radioactivity and high-energy neutrons, and it is operated at a temperature of 8mK in the Gran Sasso Underground Laboratory. After several improvements the live time of the experiment is near 75%.

Abstract

CUORICINO is a bolometric experiment on Neutrinoless Double Beta Decay (DBDOv) of 130Te. It consists of an array of 62 Te02 crystals with a total mass of - 40.7 kg. While being a self consistent experiment CUORICINO is also a good test for the feasibility of the next generation experiment CUORE, -750 kg of Te02 bolometric mass. In this paper last results from CUORICINO and prospects for the future CUORE experiment will be reported.

Abstract

CUORE is a proposed tightly packed array of 1000 TeO2 bolometers, each being a cube 5 cm on a side with a mass of 760 g. The array consists of 25 vertical towers, arranged in a square of 5 towers×5 towers, each containing 10 layers of four crystals. The design of the detector is optimized for ultralow-background searches: for neutrinoless double-beta decay of 130Te (33.8% abundance), cold dark matter, solar axions, and rare nuclear decays. A preliminary experiment involving 20 crystals 3×3×6 cm3 of 340 g has been completed, and a single CUORE tower is being constructed as a smaller-scale experiment called CUORICINO. The expected performance and sensitivity, based on Monte Carlo simulations and extrapolations of present results, are reported. © 2003 Elsevier B.V. All rights reserved.

Abstract

After 3 years of project study and development, a 40 kg, 62-crystal, thermal detector array CUORICINO started last February. It is supposed to test the two claimed, but still not confirmed, evidences in Dark Matter and in Double Beta Decay (DBD) reported in the last few years and it will have a first glance on Solar Axion. The technical goals that were forseen for this experiment would be the confirmation on the feasibility of the CUORE experiment, a 1 ton multipurpose experiment with bolometers, that could open a new era in DBD physics. © 2003 Elsevier B.V. Abstract

The first results are reported on the limit for neutrinoless double decay of 130Te obtained with the new bolometric experiment CUORICINO. The set-up consists of 44 cubic crystals of natural TeO2, 5 cm on the side and 18 crystals of 3×3×6 cm3. Four of these latter crystals are made with isotopically enriched materials: two in 128Te and two others in 130Te. With a sensitive mass of ∼ 40 kg, our array is by far the most massive running cryogenic detector to search for rare events. The array is operated at a temperature of ∼ 10 mK in a dilution refrigerator under a heavy shield in the Gran Sasso Underground Laboratory at a depth of about 3500 m.w.e. The counting rate in the region of neutrinoless double beta decay is ∼ 0.2 counts keV−1 kg−1 y−1, among the lowest in this type of experiment. No evidence for neutrinoless double beta decay is found with the present statistics obtained in about three months with a live time of 72%. The corresponding lower limit for the lifetime of this process is of 5.5×1023 years at 90% C.L. The corresponding limit for the effective neutrino mass ranges between 0.37 to 1.9 eV depending on the theoretically calculated nuclear matrix elements used. This constraint is the most restrictive one except those obtained with Ge diodes, and is comparable to them.

Abstract

Neutrino physics represents today a hot topic in elementary particle physics, due to the observations of flavour oscillations both in the atmospheric and in the solar sector. This proves the existence of finite neutrino masses. In order to fix their absolute values, sensitive experiments on Neutrinoless Double Beta Decay (a rare nuclear process) must be carried on. The experiment here described, named Cryogenic's Underground Observatory for Rare Events (CUORE), can extend the neutrino mass sensitivity down to 30 meV. CUORE will consist of a large, closely packed, high-granularity array of 1000 tellurite (TeO2) IOW-temperature calorimeters, operated at 10 mK and with a total mass of 800 kg. The final structure of the detector and the preliminary tests are presented and discussed

Abstract

The Cryogenic Underground Observatory for Rare Events (CUORE) will be a large-scale, multi-purpose observatory that is planned for construction in the Gran Sasso underground laboratory in Italy. Its primary focus will be on a search for the neutrinoless double-beta decay of 130Te. The source/detector will be composed of one thousand 5-cm×5-cm×5-cm single crystals of TeO2 all housed in a common dilution refrigerator. Attached to each crystal will be one or more neutron-transmutation-doped (NTD) germanium thermistors that will measure the small temperature rise produced in a crystal when radiation is absorbed. The high natural isotopic abundance of 130Te (33.8%) makes the use of isotopically enriched material unnecessary. In addition, the high Q-value for the double-beta decay of 130Te means that the peak in the summed electron energy spectrum expected from the neutrinoless mode will occur in an energy region of very little natural background. Once constructed, CUORE can also be used in searches for dark matter, solar axions, and other rare decay processes in nuclear physics.

Abstract

We report the present results of CUORICINO, a search for neutrinoless double-beta (0nu betabeta) decay of 130Te. The detector is an array of 62 TeO2 bolometers with a total active mass of 40.7 kg. The array is cooled by a dilution refrigerator shielded from environmental radioactivity and energetic neutrons, operated at approximately 8 mK in the Gran Sasso Underground Laboratory. No evidence for (0nu betabeta) decay was found and a new lower limit, T(1/2)(0nu) > or = 1.8 x 10(24) yr (90% C.L.) is set, corresponding to [m(nu)] < or = 0.2 to 1.1 eV, depending on the theoretical nuclear matrix elements used in the analysis.

Abstract

A modification of the Asakawa-Matsushita iodometric assay method for the determination of the content of lipid hydroperoxides was developed which permits the simultaneous processing of many samples of high lipid content. The method has the advantages of simplicity as well as good reproducibility, so it is not necessary to process standards with each determination. Our technique exceeds the sensitivity attained with other spectrophotometric determinations reported in the literature. The method requires the total elimination of water from the samples, and this was accomplished using an azeotropic mixture of ethanol:water of 96:4. The results obtained with liposomes indicate that the method is applicable to biological material limited to small volume samples, ranging 5-50 microliters. We want to emphasize that this method permits the study of the peroxidation process as function of time.

Abstract

A study of oxidative damage was made in elderly noninsulin-dependent diabetes mellitus (NIDDM) patients. A statistically significant increase in glucose and fructosamine was found in fasting NIDDM patients, as well as an increase in the oxidation induced by tert-butyl hydroperoxide. The Total Reactive Antioxidant Potential (TRAP) of the plasma was much reduced (p < .02) and the uricemia was unchanged. The erythrocytes of diabetic patients show greater basal oxidation products (p < .05), and the susceptibility of the diabetic erythrocytes to oxidation injury was also shown to increase in the oxidation induced by t-BOOH (p < .05). Linear regression studies showed that TRAP was associated directly with uric acid (p < .05) and inversely with fructosamine and with glucose (p < .03 and p < .05 respectively) in patients with NIDDM, but not in the controls. The levels of fructosamine were found to be related to the basal damage of the red blood cells (direct correlation, p < .001). This study suggest an useful approach to diabetic oxidative stress for clinical settings.

Abstract

In order to investigate the implications of oxidative disturbances in the hemolysis associated with the Hemolytic Uremic Syndrome (HUS), basal levels of lipid peroxidation products, the response to t-butyl hydroperoxide induced damage and membrane fluidity were assayed by the technique of electron spin resonance in erythrocytes spin labeled with 5-Doxyl stearic acid obtained from eight children with HUS, during the 1st, 2nd, 4th and 12th weeks after diagnosis. During the acute phase of the disease, red blood cells (RBC) showed increased initial lipid peroxidation products, a higher susceptibility to oxidative insult and a lower membrane fluidity. All parameters reached control values the 12th week after diagnosis. The results suggest that in the acute phase of HUS, RBCs are exposed to an oxidative imbalance that could contribute to hemolysis directly through oxidative damage and/or by decreasing membrane fluidity.

Abstract

Two vitamin B6 derivatives, N-bromoacetylpyridoxamine (BAPM) and its phosphate ester have been found to be affinity-labeling reagents for mitochondrial aspartate aminotransferase (EC 2.6.1.1). These derivatives were first shown to react with a critical sulfhydryl group in tryptophan synthase (Higgins, W., and Miles, E. W. (1978) J. Biol. Chem. 253, 4648-4652). In the apoaminotransferase, BAPM has now been found to inactivate by covalently modifying a critical lysyl residue, preventing reconstitution of the apoenzyme by pyridoxal 5'-phosphate. The dependence of the rate of inactivation upon the concentration of the reagent is consistent with a rapid equilibrium binary complex formation prior to the inactivation reaction. Both the dissociation constant for this complex and the rate of the reaction leading to inactivation are dependent on pH. BAPM binds best from pH 7.5 to 8.5. The rate of inactivation increases from pH 6 to 9. Succinate and phosphate competitively bind to the apoenzyme, protecting against BAPM inactivation. The C-5'-phosphorylated derivative is rapidly and tightly bound by the apotransaminase to form an inactive, noncovalent adduct. This bound reagent subsequently alkylates Lys-258. The rate of this covalent incorporation increases from pH 6 to 9 and is greater than the rate of BAPM modification at all pH values. The effect of pH on the reaction rates of both pyridoxal derivatives is interpreted to indicate protonation of Lys-258 at neutral pH values. These derivatives may also be analogs to a reaction intermediate different from those observed in other affinity-labeling studies. The ionization states of the Lys-258 epsilon-amino group apparently vary with the nature of the affinity label. These variations can be explained in terms of changing ionization states of Lys-258 in the steps of catalysis as well as in terms of the occupancy of charged sites on the protein by active site-directed substrates or inhibitory compounds.

Abstract

An essential initial step in fertilization in the sea urchin Strongylocentrotus purpuratus is an intracellular membrane fusion event in the sperm known as the acrosome reaction. This Ca2+-dependent, exocytotic process involves fusion of the membrane of the acrosomal vesicle and the plasma membrane. Recently, metalloendoproteases requiring divalent metals have been implicated in several Ca2+-dependent membrane fusion events in other biological systems. In view of the suggested involvement of Zn2+ in the sea urchin sperm acrosome reaction (Clapper, D.L., Davis, J.A., Lamothe, P.J., Patton, C., and Epel, D. (1985) J. Cell Biol. 100, 1817-1824) and the fact that Zn2+ is a metal cofactor for metalloendoproteases, we investigated the potential role of this protease in the acrosome reaction. A soluble metalloendoprotease was demonstrated and characterized in sperm homogenates using the fluorogenic protease substrate succinyl-alanine-alanine-phenylalanine-4-aminomethylcoumarin. The protease was inhibited by the metal chelators EDTA and 1,10-phenanthroline, and activity of the inactive apoenzyme could be reconstituted with Zn2+. The metalloendoprotease substrate and inhibitors blocked the acrosome reaction induced either by egg jelly coat or by ionophore, but had no effect on the influx of Ca2+. These observations suggest that inhibition occurs at a step independent of Ca2+ entry. Overall, the results of this study provide strong indirect evidence that the acrosome reaction requires the action of metalloendoprotease.

alpha-Bungarotoxin (alpha-Bgt) is a potent postsynaptic neurotoxin which blocks neurotransmission by binding very tightly to the acetylcholine-receptor (AcChR) protein. We have previously shown (P. Calvo-Fernandez, and M. Martinez-Carrion (1981) Arch. Biochem. Biophys. 208, 154-159) that alpha-Bgt free in its native solution conformation incorporates 12 methyl groups when reductively methylated using formaldehyde and sodium cyanoborohydride. We now show that when the alpha-Bgt molecule is bound to the AcChR contained in native membranes prepared from Torpedo californica electroplax, the number of accessible methylation sites is significantly reduced. This favors a model of alpha-Bgt-AcChR interaction involving significant numbers of lysyl moieties distributed over a reasonably large surface of the toxin molecule. In addition, this paper presents a novel procedure for the rapid and nondestructive dissociation of the toxin-AcChR membrane complex which takes advantage of the thermal instability of the complex.

Abstract

The cofactor analogue N-(bromoacetyl)pyridoxamine (BAPM) has been employed to inactivate the cytosolic isozyme of apo-aspartate aminotransferase. Inactivation is the result of covalent bond formation in the (bromoacetyl)-pyridoxamine-transferase complex, via the epsilon-amino group of a lysyl residue at the active site. The stoichiometry of this inactivation is one molecule of (bromoacetyl)pyridoxamine per subunit of the transaminase dimer. Trace amounts of inorganic phosphate protect the enzyme from BAPM inactivation. In the absence of phosphate, inactivation demonstrates time, concentration, and pH dependence with an apparent pK for the target group of about 8.5 or higher. A tryptic peptide from the alpha subform has been obtained containing the carboxymethyl derivative of lysine-258, identifying this particular residue as the reactive group in the region of cofactor binding. Evidence is presented indicating that the pK of Lys-258 appears to be highly dependent upon the electrostatic state of neighboring groups in the active site region. Hence, experimentally obtained values vary according to the chemical nature and charge of the modifying agent or probe.
