O presente trabalho demonstra os efeitos e resultados da utilização da ferramenta Single Minute Exchange of Die, em português, Troca Rápida de Ferramenta, na redução do tempo de setup em uma linha de produção de envase de produtos em uma fábrica de cosméticos. A linha em questão apresenta alto tempo de setup, além de possuir alto índice de trocas ao longo dos turnos de trabalho, devido à grande variação de produtos. Para o início do processo de coleta de dados foi realizada uma entrevista com a analista de produtividade das linhas do ramo de coloração da fábrica onde foi passado uma primeira impressão da atual situação das linhas de produção. Em seguida foi realizada a primeira visita à fábrica com o objetivo de aprimorar o conhecimento em relação à linha de produção. Em uma segunda visita, foi realizado o acompanhamento de todo o processo de setup da linha de produção, onde foram mapeadas todas as atividades realizadas ao longo da troca da linha e foi, também, realizada a medição de tempo para execução de cada uma delas. Por fim, com objetivo de consolidar as informações levantadas nas visitas anteriores e tirar possíveis dúvidas, foi realizada uma segunda entrevista com a analista responsável. A partir do mapeamento das atividades realizadas ao longo do setup da linha, utilizou-se a ferramenta SMED para alcançar melhorias. Com a implementação destas melhorias foi obtido uma redução de 32,93% do tempo total de setup, estimando um ganho considerável para a empresa dentro dos próximos 12 meses.

Clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated gene (Cas) system and RNA interference (RNAi)-based non-transgenic approaches are powerful technologies capable of revolutionizing plant research and breeding. In recent years, the use of these modern technologies has been explored in various sectors of agriculture, introducing or improving important agronomic traits in plant crops, such as increased yield, nutritional quality, abiotic- and, mostly, biotic-stress resistance. However, the limitations of each technique, public perception, and regulatory aspects are hindering its wide adoption for the development of new crop varieties or products. In an attempt to reverse these mishaps, scientists have been researching alternatives to increase the specificity, uptake, and stability of the CRISPR and RNAi system components in the target organism, as well as to reduce the chance of toxicity in nontarget organisms to minimize environmental risk, health problems, and regulatory issues. In this review, we discuss several aspects related to risk assessment, toxicity, and advances in the use of CRISPR/Cas and topical RNAi-based technologies in crop management and breeding. The present study also highlights the advantages and possible drawbacks of each technology, provides a brief overview of how to circumvent the off-target occurrence, the strategies to increase on-target specificity, the harm/benefits of association with nanotechnology, the public perception of the available techniques, worldwide regulatory frameworks regarding topical RNAi and CRISPR technologies, and, lastly, presents successful case studies of biotechnological solutions derived from both technologies, raising potential challenges to reach the market and being social and environmentally safe.

As espécies de Meloidogyne (Nematoide das Galhas NG) são responsáveis por muitos danos na agricultura em todo o mundo. Apesar de sua ampla distribuição mundial, as epidemias causadas pelos NGs são mais intensas nas regiões tropicais. Meloidogyne incognita, M. javanica e M. arenaria, o grupo M. incognita ou grupo MIG, estão amplamente distribuídas nessa região e causam danos em uma elevada gama de hospedeiros. As principais limitações para o controle eficiente dessas espécies são: (i) a dificuldade de identificação das espécies em campo e (ii) a falta de conhecimento sobre a variabilidade genética destas espécies. Por isso, este estudo teve como objetivo identificar, por meio de uma abordagem polifásica, as espécies de Meloidogyne predominantes nos campos de cultivo de soja na região Centro-Oeste do Brasil, bem como estudar a variabilidade genética de isolados de M. incognita. Além disso, avaliou-se a possibilidade de identificação das espécies do grupo MIG por espectrometria de massa do tipo MALDI-TOF (matriz assistida por dessorção e ionização por tempo de voo, do inglês matrix-assisted dessortion/ionization time-of-flight). A coleta foi realizada em 49 campos de soja com o histórico da ocorrência de espécies de Meloidogyne, localizados em cinco municípios do estado de Mato Grosso (MT) e 10 de Goiás (GO). A identificação dos isolados foi realizada com marcadores moleculares do tipo Sequence Characterized Amplified Regions (SCAR)-PCR, perfil isoenzimático de esterase e sequenciamento da região mitocondrial citocromo oxidase I (COI) e NADH desidrogenase subunidade 5 (NADH5). A presença de isolados de Meloidogyne foi detectada em 18 amostras. Meloidogyne incognita, M. javanica e M. enterolobii foram detectadas e observou-se a presença de populações mistas em 50% das amostras. A análise filogenética do gene NADH5 apresentou resolução suficiente para separar os isolados de M. javanica e M. incógnita a nível de espécie. A análise da variabilidade de M. incognita indica tratar-se de uma população clonal com um reduzido número de haplótipos e com baixa diversidade haplotípica e nucleotídica. Este estudo demonstrou que M. incognita é a principal espécie encontrada na cultura da soja, podendo ocorrer em associação com M. javanica e/ou M. enterolobii. A avaliação do uso da espectrometria do tipo MALDI-TOF para a identificação taxonômica dos fitonematoides do grupo MIG foi realizada com 18 isolados de M. incognita, 20 isolados de M. javanica e cinco isolados de M. arenaria. Os isolados foram identificados previamente por perfil de esterase e os espectros foram obtidos de proteínas extraídas de cinco fêmeas maceradas em solução de acetonitrila: água Milli Q (50:50). A ionização das proteínas foi realizada em matriz de αciano-4-hidroxicinâmica (CHCA). Após a geração de dendrograma, observou-se que a técnica MALDI-TOF MS tem sensibilidade para separar isolados de M. javanica, M. incognita e M. arenaria em grupos distintos, o que possibilita seu uso para a identificação das espécies do grupo MIG. Species of Meloidogyne species (Root-Knot Nematode – RKN) are responsible for much damage to agriculture worldwide. Despite their wide worldwide distribution, epidemics caused by RKN are more intense in tropical regions. Meloidogyne incognita, M. javanica, and M. arenaria, the M. incognita group or MIG group, are widely distributed in this region and cause damage in a wide range of hosts. The main limitations for the efficient control of these species are: (i) the difficulty of identifying the species in the field, and (ii) the lack of knowledge on the genetic variability of these species. Therefore, this study aimed to identify, through a polyphasic approach, which Meloidogyne species are predominant in soybean fields in the Midwest region of Brazil, as well as to study the genetic variability of M. incognita. In addition, the possibility of identifying the species of the MIG group was evaluated by Matrixassisted desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). The sample collection was carried out in 49 soybean fields with a history of the occurrence of Meloidogyne species located in five municipalities in the states of Mato Grosso (MT) and 10 of Goiás (GO). The identification of the isolates was performed with molecular markers such as Sequence Characterized Amplified Regions (SCAR)-PCR, with an isoenzymatic profile of esterase and sequencing of the mitochondrial region cytochrome oxidase I (COI) and NADH dehydrogenase subunit 5 (NADH5). The presence of Meloidogyne isolates was detected in 18 samples. Meloidogyne incognita, M. javanica, and M. enterolobii were detected, and the presence of mixed populations was observed in 50% of the samples. Phylogenetic analysis of the NADH5 gene showed sufficient resolution to separate M. javanica and M. incognita isolates into individual groups at the species level. The variability analysis of M. incognita indicates that it is a clonal population, with a reduced number of haplotypes with low haplotypic and nucleotide diversity. This work demonstrated that M. incognita is the main species found in soybean and may occur in association with M. javanica and/or M. enterolobii. The evaluation of the use of MALDI-TOF MS for taxonomic identification of phytonematodes of the MIG group was carried out with 18 isolates of M. incognita, 20 isolates of M. javanica, and five isolates of M. arenaria. The isolates were previously identified by esterase profile and the spectra were obtained from proteins extracted from five females macerated in a solution of acetonitrile: Milli Q water (50:50). Protein ionization was performed in an α-cyano-4-hydroxycinnamic (CHCA) matrix. After generation of the dendrogram, it was observed that the MALDI-TOF MS technique is sensitive to separate isolates of M. javanica, M. incognita, and M. arenaria into distinct groups, which allows its use for the identification of species of the MIG group.

This work aimed to report an experience involving the application of a Didactic Sequence (DS) integrating chemistry contents to environmental themes for High School Students (HSS). The activities were developed in a public school, located in Encanto City, High West' region, in the State of Rio Grande do Norte, with students enrolled in the 3rd grade of HSS. Aiming to provide an effective and satisfactory in organic functions' contents and their relationship with the environment, the didactic sequence was structured in 05 steps: I) Survey of students' prior knowledge; II); Field lesson in Encanto’ River; III) Expositive-dialogical classes addressing contents of chemistry and the environment; IV) Elaboration of educational-environmental posters; V) Socialization of elaborated materials. Through this lived experience, it is believed that the inclusion of environmental themes associated with Chemistry classes, allows students to be sensitized to local environmental issues, thus becoming critical and conscientious citizens, intervening in the social environment of their experience with social and environmental practices, innovative, transforming and sustainable. El objetivo de este trabajo fue reportar una experiencia de aplicación de una Secuencia Didáctica (SD) integrando contenidos de química a temas ambientales para estudiantes de secundaria (ES). Las actividades se desarrollaron en una escuela pública, ubicada en la Ciudad de Encanto, Región Alto Oeste, en el Estado de Rio Grande do Norte, con estudiantes matriculados en el 3er grado de ES. Para brindar un desempeño efectivo y satisfactorio en las funciones orgánicas y en su relación con el medio, se estructuró una secuencia didáctica en 5 etapas: I) Encuesta de conocimientos previos de los estudiantes; II); Clase de campo en Rio do Encanto; III) Clases expositivo-dialógicas que abordan contenidos de Química y medio ambiente; IV) Elaboración de carteles educativos-ambientales; V) Socialización de materiales elaborados. A través de esta experiencia, se cree que la inclusión de temas ambientales asociados a las clases de Química permite que los estudiantes tomen conciencia de los problemas ambientales locales, convirtiéndose así en ciudadanos críticos y conscientes, interviniendo en el entorno social de su experiencia. innovador, transformador y sostenible. Esse trabalho teve como objetivo relatar uma experiência envolvendo a aplicação de uma Sequência Didática (SD) integrando conteúdos de química à temas ambientais para estudantes do ensino médio (EM). As atividades foram realizadas em uma escola pública, localizada na Cidade de Encanto, região do Alto Oeste, no Estado do Rio Grande do Norte, junto aos estudantes matriculados na 3ª série do EM. Visando proporcionar uma efetiva e satisfatória em funções orgânicas e suas relações com o meio ambiente, estruturou-se uma sequência didática em 05 etapas: I) Sondagem dos conhecimentos prévios dos estudantes; II); Aula de campo no Rio encanto; III) Aulas expositivas-dialógicas abordando conteúdos de Química e meio ambiente; IV) Elaboração de cartazes educativos-ambientais; V) Socialização dos materiais elaborados. Por meio desta experiência vivenciada, acredita-se que a inserção de temas ambientais associadas às aulas de Química, possibilitam que os discentes sejam sensibilizados quanto as questões ambientais locais, tornando-se assim, cidadãos críticos e conscientes, intervindo no meio social de sua vivência com práticas socioambientalmente, inovadoras, transformadoras e sustentáveis.

Esta pesquisa teve como objetivo geral identificar a relação existente entre os riscos destacados pelos auditores independentes por meio dos principais assuntos de auditoria (PAAs) e os fatores de risco evidenciados nos formulários de referência. Para alcançar os resultados, utilizou-se a metodologia de análise de conteúdo, cuja base foi Bardin (2016). Pesquisa de natureza qualitativa, a amostra foi constituída por 16 Companhias do setor de construção civil, de 2016 a 2018. Esse período culminou com vários fatores que influenciaram de forma negativa o ramo de construção civil no Brasil, como o impacto da operação Lava Jato, a redução de obras públicas e na venda de imóveis. O resultado geral indicou relação total existente de 42,96%, relação parcial de 14,07% e relação nula de 42,96%. As empresas Direcional Engenharia, EZ TEC, MRV e Construtora Tenda foram as que mais divulgaram os riscos identificados pela auditoria. Por outro lado, as companhias Adolpho Lindenberg, CR2, JHSF e João Fortes foram as que menos divulgaram. Exceto a JHSF, as empresas com as menores notas não pertencem a nenhum segmento de governança corporativa da B3 e possuem os Totais de Ativos menores. Identificou-se oito categorias de PAAs entre os 135 divulgados e uma média geral de 2,81 PAAs por companhia. Os mais considerados foram reconhecimento de receitas, impairment e provisões, que juntos representam 80,74%. Entre os que encontraram correspondência nos fatores de risco, provisões e reconhecimento de receitas totalizaram 84,48%. Constatou-se que os auditores usam redação semelhante para descreverem os PAAs, e que mais de 80% deles são repetidos, além do uso pelas companhias de cópias de trechos inteiros já evidenciados por outras empresas nos formulários de referência. Ao considerar que não se identificou pesquisas ligando os PAAs aos fatores de risco, esta pesquisa contribui ao oferecer novos conhecimentos aos auditores, às companhias, ao público investidor e à academia, possibilitando que os stakeholders compreendam melhor esses tipos de riscos nas organizações. This research aimed to identify the relationship between the risks highlighted by the independent auditors through the main audit matters (PAAs) and the risk factors evidenced in the reference forms. To achieve the results, the content analysis methodology was used, based on Bardin (2016). Qualitative research, the sample consisted of 16 companies in the civil construction sector, from 2016 to 2018. This period culminated with several factors that negatively influenced the civil construction industry in Brazil, such as the impact of the Lava Jato operation, the reduction of public works and the sale of real estate. The general result indicated an existing total ratio of 42.96%, a partial ratio of 14.07% and a null ratio of 42.96%. Direcional Engenharia, EZ TEC, MRV and Construtora Tenda were the companies that most disclosed the risks identified by the audit. On the other hand, the companies Adolpho Lindenberg, CR2, JHSF and João Fortes were the least publicized. Except for JHSF, the companies with the lowest scores do not belong to any corporate governance segment of B3 and have the lowest total assets. Eight categories of PAAs were identified among the 135 disclosed and an overall average of 2.81 PAAs per company. The most considered were revenue recognition, impairment and provisions, which together represent 80.74%. Among those that found correspondence in risk factors, provisions and revenue recognition totaled 84.48%. It was found that the auditors use similar wording to describe the PAAs, and that more than 80% of them are repeated, in addition to the companies' use of copies of entire excerpts already evidenced by other companies in the reference forms. When considering that no research linking PAAs to risk factors has been identified, this research contributes by offering new knowledge to auditors, companies, the investing public and the academy, enabling stakeholders to better understand these types of risks in organizations.

In this article we aim to demarcate the interchanges of the ethics of Haberma’s discourse and the genetic psychology of Lawrence Kohlberg. If, on the one hand, for the German philosopher his ethical proposal can contribute to fill the deficits of genetic psychology in relation to moral development, on the other hand, discourse ethics is based on the empirical researches carried out by psychology theorists and, with this, reinforce the thesis of the ethical universality defended by Habermas and Carl Otto-Apel. However, there are limits on the demarcation of the philosophy and the psychology because the are fields in which the methodologies are not identified and and because they assume different epistemologics perspectives, descriptive for the psychology of development and prescriptive for Habermas’ philosophy intends to establish a relation with different fields of knowledge, from which it ensures interdisciplinarity and openness to the constant dialogue between diverse knowledge. Neste artigo visa-se demarcar os intercursos da ética do discurso de Habermas e da psicologia genética de Lawrence Kohlberg. Se, por um lado, para o filósofo alemão a sua proposta ética pode contribuir para suprir os déficits da psicologia genética em relação ao desenvolvimento moral, por outro lado, a ética do discurso se apoia nas pesquisas empíricas realizadas pelos teóricos da psicologia e, com isso, reforçam a tese da universalidade ética defendida por Habermas e Carl Otto-Apel. No entanto, há limites na demarcação da filosofia e da psicologia por se tratar de campos nos quais as metodologias não se identificam e por assumirem perspectivas epistemológicas distintas, descritiva para a psicologia do desenvolvimento e prescritiva para a ética do discurso. Ainda assim, a filosofia de Habermas pretende estabelecer uma relação com campos distintos de saberes, a partir dos quais garante a interdisciplinaridade e a abertura para o diálogo constante entre saberes diversos.

The article presents a discussion about the process of valuation of landscapes that base the activation of material and immaterial cultural heritage patronized by UNESCO in the Amazon, relating it to the subaltern patrimonies analyzed by the dialectical perspective, seeking to give visibility to local patrimonies not recognized by global hegemonic logic. The research has a qualitative, bibliographic character and the results demonstrated that the Amazonian heritage inscribed in the dynamics of space production and landscapes, are not recognized by the organs of world patrimonialization, but must be the foundation of patrimonialisation policies, since they are not limited to dominant idea of ​​being made of stone and lime. O artigo apresenta uma discussão sobre o processo de valoração de paisagens que fundamentem a ativação de patrimônios culturais, materiais e imateriais chancelados pela UNESCO na Amazônia, relacionando-os aos patrimônios subalternos analisados pela perspectiva dialética, buscando dar visibilidade aos patrimônios locais não reconhecidos pela lógica hegemônica global. A pesquisa tem um caráter qualitativo, bibliográfico e os resultados demonstraram que os patrimônios amazônicos inscritos na dinâmica de produção do espaço e paisagens, não são reconhecidos pelos órgãos da patrimonialização mundial, mas devem ser o fundamento das políticas de patrimonialização, pois não se resumem a ideia dominante de serem feitos de pedra e cal.

The last decades have witnessed an intensification of research on positive affective experiences in the workplace. Happiness in the workplace is paramount to improve productivity in any organization since happy people are naturally people who care about the quality of the work they develop and therefore, they are more productive and more efficient. The aim of this study was to measure the impact of the factors motivation at work and work environment on happiness in the workplace. The study population involved collaborators from various professional categories from an institution for social solidarity, located in the municipal district of Bragança, Portugal. Among a total of 353 workers, 186 were selected randomly and a response rate of 52.7% was obtained. The results showed that motivation at work and work environment are both predictors of happiness at work. However, it is motivation at work that most contributes to happiness at work. Within this context, keeping work environments that promote positive and healthy relationships among professionals and investing in workers’ motivation and wellbeing improves their professional performance, thus contributing to the success of the organization. This study aimed to determine the level of entrepreneurial self-efficacy in higher education students and to verify if there were differences statistically significant, taking into account the scientific area of the course attended by the student, in the five dimensions of the entrepreneurial self-efficacy scale, namely: Developing new products or new market opportunities; Developing critical human resources; Initiating investor relations; Building innovative environments; and Working under stress. A total of 795 students attending the 1st year at a Portuguese higher education institution, aged between 17 and 52 years old, participated in the study. The students registered a medium level of entrepreneurial self-efficacy. Statistically significant differences were found in entrepreneurial self-efficacy taking into account the students’ scientific area. It was the Technology and Management students who had the highest levels of entrepreneurial self-efficacy and the students in the Education area who had the lowest levels. Entrepreneurial self-efficacy contributes to the generation of ideas and the creation of new businesses with success. Therefore, higher education institutions should be able to improve the entrepreneurial skills and increase the self-efficacy of their students. Utilizamos o sistema conceitual espaço, paisagem e lugar definido nas diversas obras do geógrafo Milton Santos para analisarmos, traduzirmos e interpretarmos as relações socioespaciais que ocorrem nas ilhas de Ananindeua-Pará. O sistema conceitual selecionado permite-nos traduzir processos que ocorrem no interior da Amazônia, em comunidades extrativistas, ribeirinhas e próximas a Região metropolitana de Belém que sofrem pressões da ordem globalizada, mas resistem pelo lugar e paisagem, ratificando o cotidiano e tradições. Temos o entendimento de que nenhuma teoria dá conta de explicar totalmente a realidade, mas o sistema conceitual adotado permite-nos entender, a partir dos fundamentos do Materialismo Dialético, tais como, as relações que envolvem as partes e o todo, o visível e o invisível e os processos de afirmação e negação, como a dinâmica socioespacial em espaços singulares da Amazônia, estão intimamente relacionadas não podendo ser compreendidas sem a associação das variáveis locais e globais.Palavras-Chave: Espaço; paisagem; lugar; produção local, dialética.

In the last decades, especially since 2000, the natural vulnerability of Earth’s climate system has been a cause of great concern as to the status of global climate change due to the interference of natural and/or anthropic activities. Instituto Nacional de Meteorologia (Inmet), the government body officially responsible for monitoring weather and climate in Brazil, and also a member of the World Meteorological Organization (WMO), is proud to release the new edition of the climate normals for the period of 1981-2010. The new edition aims to analyze and register the climate changes that occurred during the two decades following the previous edition of 1961-1990. For that purpose, Inmet created a working group to prepare and edit these normals, as a basis of knowledge for different spheres of meteorology, aiming to strengthen the study and research of climate variability, as well as the Paris Agreement, which limits global warming to 1.5°C above pre-industrial levels. Finally, the publication intends to offer guidance, information, and assistance to the communities of climate sciences, agribusiness, and public and private institutions, both national and international. Overall, activities related to climate have expanded in practically every sphere of human life, especially in the fields of science and public policies. Resumo: Nas últimas décadas, principalmente desde 2000, a vulnerabilidade natural do sistema climático da Terra tem causado preocupações quanto à mudança climática global devido a processos naturais e/ou atividades antropogênicas. O Instituto Nacional de Meteorologia (Inmet), responsável por monitorar o tempo e o clima no Brasil, e também membro da Organização Meteorológica Mundial (OMM), tem o orgulho de lançar a nova edição das normais climatológicas 1981-2010. Esta nova edição visa analisar e registrar as mudanças climáticas que ocorreram durante as duas décadas após a edição anterior de 1961-1990. Com esse propósito, o Inmet criou um grupo de trabalho para preparar e editar essas normais, como base para conhecimento em diferentes esferas da meteorologia, para fortalecer o estudo e a pesquisa da variabilidade climática, bem como o Acordo de Paris, que limita o aquecimento global a 1,5°C acima dos níveis da era pré-industrial. Por fim, a publicação visa oferecer diretrizes, informações e assistência a comunidades de ciências climáticas, agronegócios, instituições públicas e privadas, tanto no âmbito nacional quanto internacional. No geral, as atividades relacionadas ao clima têm expandido em praticamente todas as esferas da vida humana, sobretudo nas áreas de ciências e políticas públicas.

A regulação de urgências/emergências e a regulação médica, como um instrumento de gerenciamento deste cuidado, vem sendo utilizada em outros países com muito sucesso. Para o Ministério da Saúde brasileiro, este é um meio de garantir a integralidade da assistência e a equidade no acesso dos clientes ao Sistema. Este artigo busca relacionar a organização e a estrutura dos serviços de saúde com a regulação médica das urgências e emergências, apresentando suas possibilidades e dificuldades, levando ”“ se em conta as atribuições do médico regulador - equidade do acesso, autonomia médica e efetividade da regulação. Para tanto, analisa através de entrevistas semi-estruturadas complementadas por um grupo focal com médicos reguladores, as condições do exercício da regulação na atual rede de saúde do Rio de Janeiro, considerando a legislação vigente. O estudo permitiu relacionar diretamente uma adequada organização/estrutura dos serviços de saúde, particularmente, os de urgência/ emergência, com a melhor prática da regulação médica e as suas atribuições. Desta forma, evidenciaram - se os problemas e as insuficiências da rede de saúde, em particular a de urgências/emergências no município estudado. Os resultados apontaram para a importância e urgência no gerenciamento e tratamento destas questões e os desafios para futuras pesquisas.

Honey is considered the only food of animal origin that can be consumed without being processed. The honey is, also, considered a natural and healthy product with dietary, nutritional and medicinal characteristics. Quality, geographical region of production, information available on the products’ label, brand's reputation, variety, texture, taste, aroma, appearance, packaging and price are important factors when consumers choose to purchase honey. Thus, this research intends to identify determinant factors on which consumers base their purchasing decision. A cross-sectional study was developed based on a non-probabilistic sample of 474 individuals, of whom 399 were honey consumers. Data collection took place from March to May 2016 and was based on a questionnaire developed by Ribeiro et al. (2009), which was applied directly to consumers in the city of Bragança. Later, data was analysed with SPSS 23.0 software. The data analysis includes a univariate descriptive analysis and a multivariate analysis that involved the estimation of a binary logistic regression. The significant parameters of the logit model were taste, colour, origin and certification label, at a significance level of 1%. These characteristics explained 68.9% of the consumer's decision to purchase honey since consumers recognize authenticity and distinctive attributes of the product. info:eu-repo/semantics/publishedVersion
