1. Determining Magnetic Nanoparticle Size Distributions from Thermomagnetic Measurements

Abstract

Thermomagnetic measurements are used to obtain the size distribution and anisotropy of magnetic nanoparticles. An analytical transformation method is described which utilizes temperature-dependent zero-field cooling (ZFC) magnetization data to provide a quantitative measurement of the average diameter and relative abundance of superparamagnetic nanoparticles. Applying this method to self-assembled MnAs nanoparticles in MnAs-GaAs composite films reveals a log-normal size distribution and reduced anisotropy for nanoparticles compared to bulk materials. Abstract

Background & Aims: Continuing recalcitrance to therapy cements pancreatic cancer (PC) as the most lethal malignancy, which is set to become the second leading cause of cancer death in our society. The study aim was to investigate the association between DNA damage response (DDR), replication stress, and novel therapeutic response in PC to develop a biomarker-driven therapeutic strategy targeting DDR and replication stress in PC. Methods: We interrogated the transcriptome, genome, proteome, and functional characteristics of 61 novel PC patient–derived cell lines to define novel therapeutic strategies targeting DDR and replication stress. Validation was done in patient-derived xenografts and human PC organoids. Results: Patient-derived cell lines faithfully recapitulate the epithelial component of pancreatic tumors, including previously described molecular subtypes. Biomarkers of DDR deficiency, including a novel signature of homologous recombination deficiency, cosegregates with response to platinum (P <.001) and PARP inhibitor therapy (P <.001) in vitro and in vivo. We generated a novel signature of replication stress that predicts response to ATR (P <.018) and WEE1 inhibitor (P <.029) treatment in both cell lines and human PC organoids. Replication stress was enriched in the squamous subtype of PC (P <.001) but was not associated with DDR deficiency. Conclusions: Replication stress and DDR deficiency are independent of each other, creating opportunities for therapy in DDR-proficient PC and after platinum therapy.

Abstract

A combination of semiconductor photolithography with DNA sequencing technology gives high-density oligonucleotide arrays that provide rapid access to large scale genetic information. The desired arrays may be obtained using either polymeric photoresists or photogenerated acids. The photoresist process uses standard dimethoxytrityl (DMT) -protected nucleoside phosphoramidites along with traditional semiconductor lithography tools. The use of photogenerated acid in solid polymer thin-film matrices provides spatially directed detritylation of DMT-protected oligonucleotides.

Abstract

One of the fundamental challenges for a hyperspectral imaging surveillance system is the detection of sub-pixel objects in background clutter. The background surrounding the object, which acts as interference, provides the major obstacle to successful detection. Two additional limiting factors are the spectral variabilities of the background and the object to be detected. In this paper, we evaluate the performance of detection algorithms for sub-pixel objects using a replacement signal model, where the spectral variability is modeled by multivariate normal distributions. The detection algorithms considered are the classical matched filter, the matched filter with false alarm mitigation, the mixture tuned matched filter and the finite target matched filter. Abstract

Nonvolatile resistive memory consisting of gold nanoparticles embedded in the conducting polymer poly(4-n-hexylphenyldiphenylamine) examined using admittance spectroscopy. The frequency dependence of the devices indicates space-charge-limited transport in the high-conductivity "on" state, as well as evidence for similar transport in the lower-conductivity "off" state. Furthermore, the larger dc capacitance of the on state indicates that a greater amount of filling of the midgap nanoparticle trap levels increases the overall device conductivity, leading to the memory effect.

Abstract

As the feature sizes of semiconductor devices continue to shrink, there is an increasing interest in thin film imaging approaches such as silicon-based bilayer resists. We have developed such a resist based on a copolymer of 4-hydroxystyrene with a silicon-containing monomer, which functions simultaneously as the acid-sensitive component and a source of O2 etch resistance. In an attempt to understand the reactions that occur in the photoresist film, the acidolysis reactions of the 2-[tris(trimethylsilyl)silyl]ethyl moiety have been studied in solution. Acid-catalyzed cleavage of the model 2-trimethylsilylethyl acetate in solution proceeds via a nucleophilic attack on the silicon atom of the protonated acetate. Protonation of 2-[tris(trimethylsilyl)silyl]ethyl acetate is postulated to lead to a bridged siliconium cation, which reacts with nucleophiles along three pathways and yields products in which a nucleophile is attached to a silicon atom. This mechanism is consistent with the silylation of phenolic hydroxyl groups in the photoresist film consisting of a copolymer of 4-hydroxystyrene with 2-[tris(trimethylsilyl)silyl]ethyl methacrylate, observed during photolithographic processing.

41. Nanoporous Polyimides Derived from Highly Fluorinated Polyimide/Poly(propylene Oxide) Copolymers

Abstract

Porous, low dielectric constant polyimide films have been made by a “nanofoam” approach. The pore sizes generated in the polymer films are in the tens of nanometers range, hence the term “nanofoams”. The nanoporous foams are generated by preparing triblock copolymers with the majority phase comprising polyimide and the minor phase consisting of a thermally labile block. Films of the copolymers are cast and then heated to effect solvent removal and annealing, resulting in microphase separation of the two dissimilar blocks. The labile blocks are selectively removed via thermal treatments, leaving pores the size and shape of the original copolymer morphology. The polyimide derived from 2,2-bis(4-aminophenyl)hexafluoropropane (6FDAm) and 9,9-bis(trifluoromethyl)xanthenetetracarboxylic dianhydride (6FXDA) was used as the matrix material for the generation of nanofoams, and specially functionalized poly(propylene oxide) oligomers were used as the thermally labile constituent. The synthesis and characterization of the copolymers were performed and the process for obtaining nanofoams was optimized. The foams were characterized by a variety of techniques including thermal gravimetric analysis (TGA), transmission electron microscopy (TEM), dynamic mechanical thermal analysis (DMTA), density, small-angle X-ray scattering (SAXS), refractive index, and dielectric constant measurements. Thin-film, high-modulus nanoporous films with good mechanical properties and dielectric constants ~2.3 have been synthesized by the copolymer/nanofoam approach.

44. Polyimide Nanofoams Based on Ordered Polyimides Derived from Poly(amic alkyl esters):  PMDA/4-BDAF

Abstract

A means of generating foams of high-temperature polymers, polyimides, has been developed for use in dielectric layers in microelectronics. In these systems, the pore sizes generated are in the tens of nanometers range, thus the term “nanofoams”. The foams are generated by preparing phase-separated block copolymers with the majority phase comprised of polyimide and the minor phase consisting of a thermally labile block. Films are cast, solvent is removed, and the copolymers are cured, causing phase separation of the two blocks. The labile blocks are subsequently removed via thermal treatments leaving pores having a size and shape commensurate with the size and shape of the original copolymer morphology. The polyimide derived from pyromellitic dianhydride (PMDA) and 2,2-bis[4-(4-aminophenoxy)phenyl]hexafluoropropane (4-BDAF) was used as the matrix materials for the generation of nanofoams, and poly(propylene oxide) oligomers were used as the thermally labile constituent. The synthesis and characterization of the copolymers were performed, and the process for obtaining nanofoams was optimized. The foams were characterized by a variety of techniques including TEM, SAXS, WAXD, DMTA, density, and refractive index measurements. Thin-film, high-modulus foams with good mechanical properties can be synthesized using the copolymer/nanofoam approach.
