Abstract

Abstract Background According to the required reorganization of all hospital activities, the recent COVID-19 pandemic had dramatic consequences on the orthopedic world. We think that informing the orthopedic community about the strategy that we adopted both in our hospital and in our Department of Orthopedics could be useful, particularly for those who are facing the pandemic later than Italy. Methods Changes were done in our hospital by medical direction to reallocate resources to COVID-19 patients. In the Orthopedic Department, a decrease in the number of beds and surgical activity was stabilized. Since March 13, it has been avoided to perform elective surgery, and since March 16, non-urgent outpatient consultations were abolished. This activity reduction was associated with careful evaluation of staff and patients: extensive periodical swab testing of all healthcare staff and swab testing of all surgical patients were applied. Results These restrictions determined an overall reduction of all our surgical activities of 30% compared to 2019. We also had a reduction in outpatient clinic activities and admissions to the orthopedic emergency unit. Extensive swab testing has proven successful: of more than 160 people tested in our building, only three COVID-19 positives were found, and of over more than 200 surgical procedures, only two positive patients were found. Conclusions Extensive swab test of all people (even if asymptomatic) and proactive tracing and quarantining of potential COVID-19 positive patients may diminish the virus spread.

Abstract

Identify the predictors of common mental disorders (CMD) and psychiatric medication use by faculty members in different teaching units of a public university.A cross-sectional study on 253 faculty.Being single, not having children, and not practicing physical activity were identified as predictors of CMD. Relative to psychiatric medication use, the predictors were the following: being homosexual, teaching classes in the biological sciences field, using medication for the urinary tract and sexual hormones, presenting mental disorder/sleep disorder diagnoses, and being positive for CMD.The findings of this study may subsidize more effective interventions aimed toward protecting the health and preventing aggravation in these professionals, as well as measures for promoting changes in the university work context.A cross-sectional study on 253 faculty.Being single, not having children, and not practicing physical activity were identified as predictors of CMD. Abstract

Context The COVID-19 outbreak in Italy is the major concern of Public Health in 2020: measures of containment were progressively expanded, limiting Outpatients’ visit. Objective We have developed and applied an emergency plan, tailored for Outpatients with endocrine diseases. Design Cross-sectional study from March to May 2020. Setting Referral University-Hospital center. Patients 1262 patients in 8 weeks. Interventions The emergency plan is based upon the endocrine triage, the stay-safe procedures and the tele-Endo. During endocrine triage every patient was contacted by phone to assess health status and define if the visit will be performed face-to-face (F2F) or by tele-Medicine (tele-Endo). In case of F2F, targeted stay-safe procedures have been adopted. Tele-Endo, performed by phone and email, is dedicated to COVID-19-infected patients, to elderly or frail people, or to those with a stable disease. Main outcome measure To assess efficacy of the emergency plan to continue the follow-up of Outpatients. Results The number of visits cancelled after endocrine triage (9%) is lower than that cancelled independently by the patients (37%, p p = 0.032). 86% of patients contacted by endocrine-triage received a clinical response (F2F and tele-Endo visits). F2F visit was offered especially to young patients; tele-Endo was applied to 63% of geriatric patients (p Conclusions The emergency plan respects the WHO recommendations to limit viral spread and is useful to continue follow-up for outpatients with endocrine diseases.

Abstract

Blazars are a subset of active galactic nuclei (AGN) with jets that are oriented along our line of sight. Variability and spectral energy distribution (SED) studies are crucial tools for understanding the physical processes responsible for observed AGN emission. Aims. We report peculiar behavior in the bright gamma-ray blazar PKS 1424-418 and use its strong variability to reveal information about the particle acceleration and interactions in the jet. Methods. Correlation analysis of the extensive optical coverage by the ATOM telescope and nearly continuous -ray coverage by the Fermi Large Area Telescope is combined with broadband, time-dependent modeling of the SED incorporating supplemental information from radio and X-ray observations of this blazar. Results. We analyse in detail four bright phases at optical-GeV energies. These flares of PKS 1424-418 show high correlation between these energy ranges, with the exception of one large optical flare that coincides with relatively low -ray activity. Although the optical/gamma-ray behavior of PKS 1424-418 shows variety, the multiwavelength modeling indicates that these differences can largely be explained by changes in the flux and energy spectrum of the electrons in the jet that are radiating. We find that for all flares the SED is adequately represented by a leptonic model that includes inverse Compton emission from external radiation fields with similar parameters. Conclusions. Detailed studies of individual blazars like PKS 1424-418 during periods of enhanced activity in different wavebands are helping us identify underlying patterns in the physical parameters in this class of AGN.

Abstract

Purpose or Objective: On 30 January 2020,WHO declared the outbreak of coronavirus disease 2019(COVID19).As cancer patients are already considered at increased risk of persistent depressive and anxiety disorders, the aim of this study was to evaluate psycological impact of the COVID19 pandemic in these patients.Psychological counseling services were offered to patients who showed anxiety and stress above selected cut off levels. Materials and Methods: All consecutive adult cancer outpatients between April and May 2020 were prospectively enrolled in this study and therefore received structured and validated ad-hoc questionnaires.Emotional distress was measured by means of visual scale.The structured questionnaire consisted of questions covering several areas such as knowledge and concerns about COVID19,the psychological impact of the COVID19 outbreak and mental health.Validated self-assessment questionnaires for the evaluation of psychological functioning and resilience skills were:1)Connor-Davidson Resilience Scale(CD-RISC-25) investigating five factors of resilience;2)Beck Depression Inventory-II(BDI-II)consisting of 21 items evaluating the severity of depression;3)State and Trait Anxiety Inventory(STAI-¬TRAIT-Y) investigating the anxiety-trait and anxiety-state. Results: A total of 208 patients completed the survey with a participation rate of 99%. Most of patients(79.3%) were in treatment and 20.7% in follow up.70 patients(42.7%) received palliation treatment and 94(57.3%) curative treatment. Patients who cohabited with others declared themselves to be more resilient;the COVID related physical concern is worse in those with lung cancer than in other cancers and in those on active treatment compared to follow up. COVID-related physical concern correlates significantly with depression, distress, and state and trait anxiety. (Figure Presented) Conclusion: During COVID19,depression and emotionality were limited in our patients.Physical concern,as expected due to the cancer diagnosis,was present in this cohort of patients and was higher in treatment compared with follow up patients.COVID19 worsens Trait-Anxiety in all patients.

The chapter describes how an understanding of the ecology of a tailings storage facility can be used to manage risks to wildlife associated with weak-acid dissociable cyanide concentrations above the Code's (Code) limit of 50 mg/L. Collection of the appropriate chemistry and ecological data associated with understanding the risks of a system is summarized, and how this information can then be used to make a cost-effective and informed decision on the most appropriate approach for achieving compliance with the Code is outlined.

Abstract

We report Fermi-LAT observations of the radio-loud AGN SBS 0846+513 (z=0.5835), optically classified as a Narrow-Line Seyfert 1 galaxy, together with new and archival radio-to-X-ray data. The source was not active at gamma-ray energies during the first two years of Fermi operation. A significant increase in activity was observed during 2010 October-2011 August. In particular a strong gamma-ray flare was observed in 2011 June reaching an isotropic gamma-ray luminosity (0.1-300 GeV) of 1.0x10^48 erg/s, comparable to that of the brightest flat spectrum radio quasars, and showing spectral evolution in gamma rays. An apparent superluminal velocity of (8.2+/-1.5)c in the jet was inferred from 2011-2012 VLBA images, suggesting the presence of a highly relativistic jet. Both the power released by this object during the flaring activity and the apparent superluminal velocity are strong indications of the presence of a relativistic jet as powerful as those of blazars. In addition, variability and spectral properties in radio and gamma-ray bands indicate blazar-like behaviour, suggesting that, except for some distinct optical characteristics, SBS 0846+513 could be considered as a young blazar at the low end of the blazar's black hole mass distribution.

The helium pycnometer allows us to measure the cell-wall density of dry woods and the basic density of wood samples soaked with water and/or a consolidant solution if a non-volatile solvent is used. These parameters were correlated to the porosity, which for degraded waterlogged wood is related to the maximum water content. Moreover, this has revealed the possibility of investigating, by means of accurate cell-wall density determination, the efficacy of several consolidants in the treatment of waterlogged woods.

Abstract

Using the Parkes radio telescope we have carried out deep observations of eleven unassociated gamma-ray sources. Periodicity searches of these data have discovered two millisecond pulsars, PSR J1103-5403 (1FGL J1103.9-5355) and PSR J2241-5236 (1FGL J2241.9-5236), and a long period pulsar, PSR J1604-44 (1FGL J1604.7-4443). In addition we searched for but did not detect any radio pulsations from six gammaray pulsars discovered by the Fermi satellite to a level of - 0.04 mJy (for pulsars with a 10% duty cycle). Timing of the millisecond pulsar PSR J1103-5403 has shown that its position is 9' from the centroid of the gamma-ray source. Since these observations were carried out, independent evidence has shown that 1FGL J1103.9-5355 is associated with the flat spectrum radio source PKS 1101-536. It appears certain that the pulsar is not associated with the gamma-ray source, despite the seemingly low probability of a chance detection of a radio millisecond pulsar. We consider that PSR J1604-44 is a chance discovery of a weak, long period pulsar and is unlikely to be associated with 1FGL J1604.7-4443. PSR J2241-5236 has a spin period of 2.2 ms and orbits a very low mass companion with a 3.5 hour orbital period. The relatively high flux density and low dispersion measure of PSR J2241-5236 makes it an excellent candidate for high precision timing experiments. The gamma-rays of 1FGL J2241.9-5236 have a spectrum that is well modelled by a power law with exponential cutoff, and phasebinning with the radio ephemeris results in a multi-peaked gamma-ray pulse profile. Observations with Chandra have identified a coincident X-ray source within 0.1" of the position of the pulsar obtained by radio timing

Abstract

We report on a multiwavelength campaign for the radio-loud narrow-line Seyfert 1 (NLS1) galaxy PMN J0948+0022 (z= 0.5846) performed in 2010 July–September and triggered by a high-energy γ-ray outburst observed by the Large Area Telescope onboard the Fermi Gamma-ray Space Telescope. The peak flux in the 0.1–100 GeV energy band exceeded, for the first time in this type of source, the value of ~10^(−6) photon cm^(−2) s^(−1), corresponding to an observed luminosity of ~10^(48) erg s^(−1). Although the source was too close to the Sun position to organize a densely sampled follow-up, it was possible to gather some multiwavelength data that confirmed the state of high activity across the sampled electromagnetic spectrum. The comparison of the spectral energy distribution of the NLS1 PMN J0948+0022 with that of a typical blazar – such as 3C 273 – shows that the power emitted at γ-rays is extreme.

Abstract

Wildlife deaths associated with cyanide-bearing tailings dams are a significant environmental issue that has affected the gold mining industries for many years and still characterized by little knowledge about how to measure, monitoring, reduce or eliminate those deaths. The purpose of this paper is statistically to determine: the potential for establishing causal relations between exposure to cyanide (in its most common species relevant to tailings) and response (measured by death counts), to develop a protocol of data analysis, the understanding of the significance of data gaps, and the effect of likely risk management interventions to achieve the goals of the International Cyanide Management Code (ICMC); [ICMC The International Cyanide Management Institute. However, operator's certification under the ICMC is difficult because of the limited data and potentially serious under-estimation of the death counts. This is due to observational skill and monitoring frequency, the small size of the carcasses, large extent of tailings facilities, carcasses loss by; entombment in tailings, sink, or taken by scavenging wildlife. This (1st order or bounding) assessment results focus on bird-deaths, which appear to be most frequent at sites where elevated cyanide concentrations are found. Those results indicate that the empirical causal associations we generate support the hypotheses that: • Cyanide intake and bird-deaths is not linear, and that relationship is characterized by a threshold at approximately 50 mg weak-acid-dissociable cyanide (WADCN) per litre of tailings waste (50 mg/L); and • The counts of either wildlife deaths or visitations on tailings dams decrease as the surface area of the supernatant decreases.This paper also develops the basis for a complete risk assessment study to be based on additional data gathering activities and detailed statistical analyses. These two activities, combined with a risk management plan also being developed, will provide a tool for compliance with the ICMC. Abstract

Wildlife deaths associated with cyanide-bearing mine waste solutions have plagued the gold mining industries for many years, yet there is little published data showing the relationship between wildlife mortality and cyanide toxicity. A gap of knowledge exists in monitoring, understanding the causal relationships and managing risks to wildlife from cyanide-bearing waste solutions and tailings. There is a need for the gold industry to address this issue and to meet the International Cyanide Management Code (ICMC) guidelines.The perceived extent of the issue varies, with one study finding the issue inadequately monitored and wildlife deaths grossly underestimated. In Nevada, USA during 1990 and 1991, 9512 carcasses were reported of over 100 species, although there was underestimation due to reporting being voluntary. Of these, birds comprised 80–91% of vertebrate carcasses reported annually. At Northparkes, Australia in 1995, it was initially estimated that 100 bird carcasses were present by mine staff following a tailings incident; when a thorough count was conducted, 1583 bird carcasses were recorded. Eventually, 2700 bird deaths were documented over a four-month period. It is identified that avian deaths are usually undetected and significantly underestimated, leading to a perception that a risk does not exist.Few guidelines and information are available to manage the risks of cyanide to wildlife, although detoxification, habitat modification and denying wildlife access have been used effectively. Hazing techniques have proven ineffective. Apparently no literature exists that documents accurate wildlife monitoring protocols on potentially toxic cyanide-bearing mine waste solutions or any understanding on the analysis of any derived dataset. This places the onus on mining operations to document that no risk to wildlife exists.Cyanide-bearing tailings storage facilities are environmental control structures to contain tailings, a standard practice in the mining industry. Cyanide concentrations below 50 mg/L weak-acid-dissociable (WAD) are deemed safe to wildlife but are considered an interim benchmark for discharge into tailings storage facilities (TSFs).Cyanide is a fast acting poison, and its toxicity is related to the types of cyanide complexes that are present. Cyanide in biota binds to iron, copper and sulfur-containing enzymes and proteins required for oxygen transportation to cells. The accurate determination of cyanide concentrations in the field is difficult to achieve due to sampling techniques and analytical error associated with loss and interferences following collection.The main WAD cyanide complexes in gold mine tailings are stable in the TSF environment but can release cyanide ions under varying environmental conditions including ingestion and absorption by wildlife. Therefore distinction between free, WAD and total cyanide forms in tailings water for regulatory purposes is justified.From an environmental perspective, there is a distinction between ore bodies on the basis of their copper content. For example, wildlife deaths are more likely to occur at mines possessing copper–gold ores due to the formation of copper–cyanide complexes which is toxic to birds and bats. The formation of copper–cyanide complex occurs preferentially to gold cyanide complex indicating the relative importance of economic vs. environmental considerations in the tailings water.Management of cyanide to a perceived threshold has inherent risks since cyanide has a steep toxicity response curve; is difficult to accurately measure in the field; and is likely to vary due to variable copper content of ore bodies and ore blending. Consequently, wildlife interaction needs to be limited to further reduce the risks. A gap in knowledge exists to design or manage cyanide-bearing mine waste solutions to render such facilities unattractive to at-risk wildlife species. Abstract

Introduction. Deep infiltrating endometriosis (DIE) is a form of endometriosis in which the lesion penetrates for more than 5 mm under the peritoneal surface. It is a chronic disease which can impair women's sexual function. There is a growing body of evidence supporting combined surgical/medical treatment in the management of DIE. Aims. The aims of this article are to evaluate the impact of the laparoscopic full excision of endometriosis and postoperative combined oral contraceptives (COC) administration on sexual function in patients with DIE and to compare sexual function outcomes of women submitted to intestinal resection and nodule excision. Methods. It is a prospective cohort study in a tertiary care university hospital on 106 sexually active women, with histologically confirmed DIE, managed by laparoscopy and subsequent COC therapy for 6 months. Patients filled preoperatively and 6-month postoperatively a quality of sexual life questionnaire, the Sexual Health Outcomes in Women Questionnaire (SHOW-Q) and they ranked their symptom intensity using a 10-point visual analogue scale (VAS). Main Outcome Measures. Sexual function was measured through the SHOW-Q scores and pain symptoms through VAS scores. Intraoperative details, type of intervention and perioperative complications were noted. Results. Six months after surgery and postoperative COC treatment, a significant improvement was observed in the SHOW-Q domains of pelvic problem interference, sexual satisfaction and desire (P 0.05). Conclusions. Sexual desire, satisfaction with sex and pelvic problem interference with intercourse are significantly improved after 6 months from laparoscopic excision of DIE combined with postoperative COC therapy. We present the KLOE measurements on KS , KL , K �� semileptonic decays from which we find |Vus | = 0.2237(13). From the KLOE measurement of BR(K+��� ��+ �� ) we obtain |Vus & Vud | = 0.2323(15). Abstract

The Fermi Gamma-ray Space Telescope has been detecting a wealth of sources where the multi-wavelength counterpart is either inconclusive or missing altogether. We present a combination of factors that can be used to identify multi-wavelength counterparts to these Fermi unassociated sources. This approach was used to select and investigate seven bright, high-latitude unassociated sources with radio, UV, X-ray and gamma-ray observations. As a result, four of these sources are candidates to be active galactic nuclei (AGN), and one to be a pulsar, while two do not fit easily into these known categories of sources. The latter pair of extra-ordinary sources might reveal a new category subclass or a new type of gamma-ray emitters. These results altogether demonstrate the power of a multi-wavelength approach to illuminate the nature of unassociated Fermi sources., 9 pages, 2 figures, 1 Table. Accepted for publication in ApJ

Abstract

Introduction The aim of this prospective observational registry was to study the outcome of symptomatic patients presenting with recent TIA or minor stroke and severe carotid stenosis, submitted to early percutaneous treatment by stenting. A secondary aim was to evaluate the biological activity of the symptomatic carotid plaques by serial serum and urinary markers (PAPP-A, hs-CRP, MMP-2/MMP-9, IL-6/IL-8, TNF alpha, CD40L) measured by enzyme-linked immunosorbent assay before and after treatment. Methods From May 2005 to June 2006, 57 patients were enrolled in this prospective registry. All patients underwent carotid stenting using a concentric filter for cerebral protection. The procedure was performed within 24–48hrs of the last attack in patients with TIA ( n =24, 42%) and between 14 and 30 days in patients with stroke ( n =33, 58%). Results Successful stent implantation was achieved in all cases (100%). Adverse events at 1 month were 1 death (1.7%) and 2 TIAs (3.5%). Some of the vulnerability markers, in particular those reflecting an active systemic inflammatory process of the plaque (PAPP-A, hs-CR, and IL-6), were significantly elevated at the time of enrolment, increased after stenting and decreased after 30 days. Conclusion Deferred CAS is feasible and safe in selected patients with symptomatic carotid stenosis. This preliminary study in a limited series of patients with unstable carotid plaques revealed that endovascular treatment has a satisfactory outcome considering the very high risk profile of the patient population. The evaluation of some biomarkers suggested an inflammatory role in the process of an unstable carotid plaque generating an acute cerebral event.

Abstract

We present results of a direct search for the decay KS → e +e- with the KLOE de- tector, obtained with a sample of e+e- → φ → KSKL events produced at DAPdblNE, the Frascati φ-factory, for an integrated luminosity of 1.3 fb-1. The StandardModel prediction for this decay is BR(KS → e+e -) = 1.6 × 10-15. The search has been performed tagging the KS decays by simultaneous detection of a KL interaction in the calorimeter. Background rejection has been optimized by using both kine- matic cuts and particle identification. At the end of the analysis chain we find BR(KS → e+e-) < 2.1 × 10-8 at 90% CL, which improves by a factor of ∼ 7 on the previous best result, obtained by CPLEAR experiment.

Abstract

An integrated luminosity of 410 pb-1, corresponding to ��� 17 million of �� events, has been analyzed to measure the �� mass using the decay �� ��� ����. The measurement is insensitive to the calorimeter energy calibration and the systematic error on the measurement is dominated by the uniformity of the detector response. As a cross check of the method the ��0 mass from the decay �� ��� ��0, ��0 ��� has been measured and it is in agreement with the most accurate previous determinations. The result obtained is m �� = 547.873��0.007stat. ��0.031syst. MeV, that is today best measurement of the �� mass.

The KLOE experiment has been collecting data since april 1999 at the DAΦNE collider in Frascati. A statistics of about 0.45 f b −1 has been analyzed. The latest results concerning the study of the ϕ radiative decays and of the kaon decays are illustrated in this paper.

Abstract

We present a summary of Swift and Suzaku X-ray observations of the 2010 nova outburst of the symbiotic star, V407 Cyg. The Suzaku spectrum obtained on day 30 indicates the presence of the supersoft component from the white dwarf surface, as well as optically thin component from the shock between the nova ejecta and the Mira wind. The Swift observations then allow us to track the evolution of both components from day 4 to day 150. Most notable is the sudden brightening of the optically thin component around day 20. We identify this as the time when the blast wave reached the immediate vicinity of the photosphere of the Mira. We have developed a simple model of the blast wave-wind interaction that can reproduce the gross features of the X-ray evolution of V407 Cyg, and explore a parameter space of ejected mass, binary separation and Mira mass loss rate. A search for the weak production of charginos and neutralinos decaying to a final state with three leptons (electrons or muons) and missing transverse momentum is presented. The analysis uses 2.06??????fb???1 of s???=7??????TeV proton-proton collision data delivered by the Large Hadron Collider and recorded with the ATLAS detector. Observations are consistent with standard model expectations in two signal regions that are either depleted or enriched in Z-boson decays. Upper limits at 95% confidence level are set in R-parity conserving phenomenological minimal supersymmetric and simplified models. For the simplified models, degenerate lightest chargino and next-to-lightest neutralino masses up to 300 GeV are excluded for mass differences from the lightest neutralino up to 300 GeV.

Abstract

Some acetone-carried consolidants for waterlogged archaeological wood were tested in order to evaluate treatments able to save time and energy. In details, colophony (rosin), two esterified colophonies (Rosin 100® and Rosin 459®), a mixture of colophony with PEG 3400 and a vinyl acetate - vinyl versate copolymer (Vinavil 8020S®) were tested. The treatments were carried out at temperatures of 20 and 35 °C on waterlogged maritime pine, elm, oak and beech. The materials came from the archaeological Site of the ancient ships of Pisa (Tuscany, Italy) and were dated back to VII cent. BC – II cent. AD. To evaluate the processes, equilibrium moisture content and dimensional stability of treated wood samples at different relative humidity and retention of impregnating products were measured; moreover macroscopic and microscopic examination were also run to respectively assess the shape and appearance of treated wood and the way of deposition of consolidants. The results highlighted that natural and modified colophony treatments gave the most satisfactory results both in the maintenance of shape and dimensions of samples and in the stabilization with respect to RH variations. Moreover, the equilibrium moisture contents of samples treated with R100 and R459 were much reduced in comparison to the other consolidants and to untreated archaeological wood. This fact was related to the high retention values of those products that occluded most of the porosity including the microporosity of cell walls. Therefore, in terms of higroscopicity treated wood was more similar to impregnating substances rather than to decayed wood. This fact was considered helpful in contrasting the moisture-related negative effects in cases of eventual faults in the climate control during e.g. exhibition and in protecting treated wood from the risks of new fungal attacks.

Abstract

The prognosis of adenosquamous carcinoma of the cervix compared to the pure cell types of this disease is a controversial issue. Survival rates vary widely among published series, with some authors finding the prognosis to be much worse and others finding it to be equal. We have studied a group of 290 patients, all of whom had the diagnosis of stage IB or IIA cervical cancer, and all of whom were treated by radical hysterectomy and bilateral pelvic and paraaortic lymphadenectomy. The pathology specimens were reviewed for every case. Median follow-up for all living patients was 73 months. Forty-five patients (15.5%) had adenosquamous histology, 220 (75.9%) had squamous cell, and 25 (8.6%) had adenocarcinoma. By X2, there was no significant difference among the three groups with regard to race, economic status, number of grade 3 lesions, number with positive pelvic lymph nodes, number with positive margins, stage distribution (IB vs IIA), or number of recurrences. Using Student's t test, there was no significant difference between any combination of two groups with regard to mean patient age, mean depth of invasion, or mean tumor size. Estimated disease-free survival and overall survival were not different among the three cell types. We conclude that for early stage cervical cancer treated by radical hysterectomy, the adenosquamous cell type does not carry a worse prognosis than either of the pure cell types.

Abstract

Most patients with ovarian cancer currently are treated primarily with surgery and chemotherapy. Drug selection usually is not based on individualized in vitro sensitivity studies but on reported response rates of clinical trials. Attempts to include in vitro chemosensitivity testing into the management of ovarian cancer have been disappointing to clinicians. Tumor cells from fresh human ovarian cancer do not grow well under artificial in vitro growth conditions. The selection of cells that happen to proliferate in vitro (e.g., human tumor clonogenic assay) has resulted in low plating efficiencies (0.001-0.1% of plated cells). The vigorous mechanical and enzymatic tumor disaggregation, done to obtain a single-cell suspension, further reduces the number of cells that grow in vitro, resulting in low overall evaluability rates of 40-70% for the human tumor clonogenic assay. At the University of Miami, a new in vitro chemosensitivity assay was developed that detected the decrease in total tumor cell viability by measuring intracellular adenosine triphosphate as a function of in vitro drug response. Preliminary data on 31 tumor tissues from patients, which was evaluated with this method, showed a sensitivity of 92% and a specificity of 100%. Since these initial studies, data were gathered on more than 150 fresh gynecologic tumor specimens to evaluate single drugs and drug combinations at five concentrations (range, 10-500% of reported peak plasma concentrations). The evaluability rate for ovarian tumors was more than 90%. Some tumors showed almost complete cell kill at the lowest drug concentration; others had only a limited response at the highest level. Drug-response patterns also were variable for combined drug exposure. These findings underscore the heterogeneity of drug response in morphologically similar tumors and the importance of characterizing individual chemosensitivity profiles for patients before drug treatment.

Abstract

Since it is not possible to predict when a Gamma-Ray Burst (GRB) will occur or when Active Galactic Nucleus (AGN) flaring activity starts, follow-up/monitoring ground telescopes must be located as uniformly as possible all over the world in order to collect data simultaneously with Fermi and Swift detections. However, there is a distinct gap in follow-up coverage of telescopes in the eastern U.S. region based on the operations of Swift. Motivated by this fact, we have constructed a 14" fully automated optical robotic telescope, Goddard Robotic Telescope (GRT), at the Goddard Geophysical and Astronomical Observatory. The aims of our robotic telescope are 1) to follow-up Swift/Fermi GRBs and 2) to perform the coordinated optical observations of Fermi Large Area Telescope (LAT) AGN. Our telescope system consists of off-the-shelf hardware. With the focal reducer, we are able to match the field of view of Swift narrow instruments (20' x 20'). We started scientific observations in mid-November 2008 and GRT has been fully remotely operated since August 2009. The 3 sigma upper limit in a 30-second exposure in the R filter is ~15.4 mag; however, we can reach to ~18 mag in a 600-second exposures. Due to the weather condition at the telescope site, our observing efficiency is 30-40% on average., 14 pages, 14 figures, accepted for publication in ASR special issue on Neutron Stars and Gamma Ray Bursts

Abstract

The aim of the work is to set up a methodological approach to verify the effectiveness of the treatments of decayed waterlogged archaeological wood and to point out the proper thermo-hygrometric conditions for its preservation after treatment. The treatments were performed on wood samples of maritime pine (Pinus pinaster Aiton), oak (Quercus sp. caducifolia), elm (Ulmus cf. minor) and strawberry tree (Arbutus unedo L.), obtained from stems pertaining to the original vegetation found in the excavation site of the Ancient Ships in Pisa (Italy), and dated from seventh century BC to second century AD The utilised products were: Polyethylene Glycols (PEG) of various molecular weights, a Polypropylene Glycol (PPG 425), Trehalose (α-D-glucopyranosyl-α-D-glucopyranoside), and their mixtures, and also a Hydroxypropylcellulose (Klucel). The main objective to be pursued by the treatments was considered the stabilisation of the original size and shape of samples. The various steps of this approach were: the execution of a preliminary diagnostic survey on untreated samples; the characterisation of treatment solutions ‘as such’ to establish the property to be monitored during the treatment; the evaluation of the main physical characteristics of wood after the treatment were determined. Among the latter, the coefficient of dimensional stability during the exposure to a series of selected thermo-hygrometric conditions and the retention of consolidants after the treatment. These two measurements allowed the definition of the ‘efficacy of a treatment’, φT, a new parameter firstly utilised in this work. It measures the stabilisation capability of the percent unit of retained product, and its value permits to put in evidence the consolidants that stabilise wood with the lowest amount of product. Conversely, it was not possible to measure the Anti-Shrink Efficiency (ASE), one of the most utilised parameters for the evaluation of treatments, because of the serious distortions and fractures observed in all the heavily degraded untreated samples.

Abstract

Although progesterone receptor status has been shown to correlate with response to hormonal therapy, not all progesterone receptor-positive patients respond to this treatment and additional biologic assays are needed to help better predict clinical response to hormonal therapy. This study explored the potential of the ATP bioluminescence assay and flow cytometry as biological assays of hormonal response. Five uterine cancer cell lines were used: AE7, ECC-1, HEC1A, AN3, and SKUT1B. Cells were exposed to Provera or tamoxifen at 0.1, 0.2, 0.5, 1, 2, and 5X (X equal to peak plasma concentrations: 1.0 micrograms/ml Provera and 0.1 micrograms/ml tamoxifen). For correlation, estrogen and progesterone receptors were determined by the standard dextran-coated charcoal method. Only AE7 and ECC-1 were positive for progesterone receptors (501 fmol/mg AE7, 194 fmol/mg ECC-1) and the rest were negative (less than 8 fmol/mg). Tamoxifen exerted no inhibition to the above cell lines. Meanwhile, Provera exerted dose-response inhibition on both AE7 and ECC-1 cell lines. The effects of accumulation of G0-G1 phase and reduction of S, G2 cells (P less than 0.05), but not on the HEC1A cell line (P = 0.4). These changes confirmed the antiproliferative property of Provera. Further studies are needed to establish the role of the ATP bioluminescence assay and flow cytometry as biological assays of hormonal response.

Abstract

Pirarubicin (PIRA) has been shown to have improved potency with less cardiac toxicity in several phase I and II clinical trials in Japan and Europe. Since Adriamycin (DXR) remains one of the most potent drugs in treatment of gynecologic cancers, this derivative has the potential to become an important chemotherapeutic agent. In this study, we compared the performance of these two drugs against a panel of 10 gynecologic cancer cell lines. The ATP chemosensitivity assays were used to determine dose-response curves. Flow cytometry was used to study cell kinetic response to both drugs. Using an IC50 value of 0.2 micrograms/ml as a cutoff for drug sensitivity, 4 cell lines, ECC1, HEC1B, BG1, and SKOV3, were considered resistant to DXR. By comparing IC50s, PIRA was 3.4 +/- 0.4 times more potent than DXR (P = 0.05). The other 6 cell lines, AN3, AE7, HEC1A, CAOV3, SKUT1B, and ME180, were considered sensitive to DXR. In this group of cell lines, PIRA was 1.6 +/- 0.3 times more potent than DXR (P = 0.5). Both PIRA and DXR elicited a spectrum of cell kinetics. By comparing the magnitude of G2 blocks at 0.1 micrograms/ml, PIRA was approximately 2-5 times more potent than DXR in SKUT1B, HEC1A, and BG1 cell lines. PIRA also displayed a reverse dose-response pattern of G2 block so that at high dose, cell cycle kinetics would mirror those of untreated controls. This observation supports the presence of a resistant tumor subpopulation and the concept of tumor heterogeneity.

Abstract

The ATP bioluminescence assay has demonstrated a strong potential to become a clinical assay for chemosensitivity testing. Currently, chemotherapy of gynecologic cancers remains controversial and empirical. To optimize the patient's chance of survival and to justify related toxicities, the chemoregimen should be individualized and based on the patient's chemosensitivity profiles. This study was performed to identify a panel of active drugs against uterine cancer cell lines for possible use in future chemosensitivity testing. We used the ATP chemosensitivity assays to screen 12 common cytotoxic agents against six uterine cancer cell lines. Drug concentrations required for a 50% surviving fraction were defined as IC50s. When using an IC50 of 0.21 PPC (peak plasma concentration) as a cutoff value for sensitivity, the following 8 drugs were considered effective for uterine cancer cell lines: actinomycin D, Adriamycin, vinblastine, etoposide, 5-fluorouracil, methotrexate, cytosine arabinoside, and mitomycin-C. Meanwhile, 4 drugs, cisplatin, 4OH-Cytoxan, bleomycin, and Alkeran with mean IC50s of 2.1 +/- 0.7, 0.8 +/- 0.1, greater than 5.0, and 0.75 +/- 0.36 PPC, respectively, were considered inactive or partially active with higher IC50s than peak plasma concentrations. In conclusion, the above panel of promising drugs can be further tested in animal models or human cancer specimens for possible use in chemosensitivity testing of uterine cancer patients.

Abstract

Densities and viscosities of sodium bis(2-ethylhexyl) sulfosuccinate (AOT in-heptane system containing light and heavy water, as a function of the molar ratio R (R=[H2O or D2O]/[AOT]) were measured at 0, 5, 25 and 40°C. At low R values, the apparent molar volume of deuterium oxide is smaller than that of light water. The difference is related to the strength of the hydrogen bonding H2O and D2O. The viscosities of both H2O-AOT-n-heptane and D2O-AOT-n-heptane systems were explained in terms of intermicellar interactions mainly governed by hydration of the head groups of AOT.
