The coupling of PHB generation with NADH reoxidation is required to generate PHB as a fermentation product. A fundamental trait to accomplish this feature is to express a functional NADH-preferring acetoacetyl-CoA reductase, engaged in PHB accumulation. One way to obtain such a reductase is by engineering the cofactor preference of the acetoacetyl-CoA reductase encoded by the phaB1 gene from Cupriavidus necator (AARCn1). Aiming to have a deeper understanding of the structural determinants of the cofactor preference in AARCn1, and to obtain an NADH-preferring acetoacetyl-CoA reductase derived from this protein, some engineered enzymes were expressed, purified and kinetically characterized, together with the parental AARCn1. One of these engineered enzymes, Chimera 5, experimentally showed a selectivity ratio ((kcat/KM)NADH/(kcat/KM)NADPH) ≈ 18, which is 160 times higher than the selectivity ratio experimentally observed in the parental AARCn1. A thermodynamic-kinetic approach was employed to estimate the cofactor preference and flux capacity of Chimera 5 under physiological conditions. According to this approach, Chimera 5 could prefer NADH over NADPH between 25 and 150 times. Being a derivative of AARCn1, Chimera 5 should be readily functional in Escherichia coli and C. necator. [ES] En el presente trabajo se define el diseño e implementación de un sistema de desinfección mediante ozono con monitorización IoT, capaz de eliminar patógenos presentes en una estancia. Además, adquiere y gestiona datos sobre el lugar en tiempo real, de manera que posibilita la consulta tiempo después. Para el control del sistema se ha elegido la placa de desarrollo ESP32, que dispone de conectividad WIFI y bluetooth. En esta aplicación se utiliza una conexión WIFI para obtener acceso a la red. Para el proceso de desinfección, utilizaremos cuatro actuadores: generador de ozono para producir la molécula O3 la cual se encarga de la desinfección, ventiladores para proporcionar al sistema de la ventilación adecuada, pantalla LCD para la visualización del temporizador y zumbador para advertir del fin del proceso. Los sensores empleados serán tres: MQ-131 para la adquisición de la concentración de ozono, MQ-135 para cuantificar la cantidad de CO2 en el ambiente y DHT11 para obtener valores de temperatura y humedad. Durante este trabajo explicaremos tanto sus principios de funcionamiento como sus comunicaciones con el micro para poder gestionar los datos. Por último, se realizará el montaje del sistema con las conexiones de cada uno de los distintos sensores y se explicará cómo observar los datos obtenidos en la aplicación móvil Blynk y en el servidor web ThingSpeak., [CA] En el present treball es defineix el disseny i implementació d'un sistema de desinfecció mitjançant ozó amb monitoratge IoT que és capaç d'eliminar patògens com a bacteris, fongs, protozous o virus presents en una estada, en aquest projecte se centra en el virus Sars-Cov-2. A més, adquireix i gestiona dades sobre el lloc en temps real, de manera que possibilita la consulta temps després. Per al control del sistema s'ha triat la placa de desenvolupament ESP32, que disposa de connectivitat WIFI i bluetooth. En aquesta aplicació s'utilitza una connexió WIFI per a obtindre accés a la xarxa. Per al procés de desinfecció, utilitzarem quatre actuadors: generador d'ozó per a produir la molècula la qual s'encarrega de la desinfecció, ventiladors per a proporcionar al sistema de la ventilació adequada, pantalla LCD per a la visualització del temporitzador i brunzidor per a advertir de la fi del procés. Els sensors emprats seran tres: MQ131 per a l'adquisició de la concentració d'ozó, MQ135 per a quantificar la quantitat de en l'ambient i DHT11 per a obtindre valors de temperatura i humitat. Durant aquest treball explicarem tant els seus principis de funcionament com les seues comunicacions amb el micro per a poder gestionar les dades. Finalment, es realitzarà el muntatge del sistema amb les connexions de cadascun dels diferents sensors i s'explicarà com observar les dades obtingudes en l'aplicació mòbil Blynk i en el servidor web ThingSpeak., [EN] This work defines the design and implementation of an ozone disinfection system with IoT monitoring that is capable of eliminating pathogens such as bacteria, fungi, protozoa or viruses present in a room, in this project it focuses on the Sars-Cov virus -2. In addition, it acquires and manages data about the place in real time, so that it can be consulted later. To control the system, the ESP32 development board has been chosen, which has WIFI and bluetooth connectivity. In this application a WIFI connection is used to gain access to the network. For the disinfection process, we will use four actuators: ozone generator to produce the molecule which is responsible for disinfection, fans to provide the system with adequate ventilation, LCD screen to display the timer and buzzer to warn of the End of the process. The sensors used will be three: MQ131 to acquire the ozone concentration, MQ135 to quantify the amount of in the environment and DHT11 to obtain temperature and humidity values. During this work we will explain both its operating principles and its communications with the micro in order to manage the data. Finally, the assembly of the system will be carried out with the connections of each of the different sensors and it will be explained how to observe the data obtained in the Blynk mobile application and in the ThingSpeak web server.

he purpose of this study was to determine the gyms of the Municipality of Rioverde S.L.P. positioning based on their attributes. The study was of a quantitative, descriptive, correlational and cross-sectional design. The study population for this study was composed of people who attend the gyms in the municipality of Rioverde, such as Chino's Gym, Fitness Ten Gym, Geras Gym, Marmak Gym and Optimus Gym which were selected to carry out this research. The population that was determined is infinite since the exact number of people who attend the gyms is unknown, the technique of sampling was probabilistic by conglomerates, also called cluster or cluster sampling. To collect the information, a questionnaire was required which was structured based on the information needs for this study. Once the information was analyzed, it was possible to conclude that the best positioned gym according to its attributes was Fitness Ten Gym, since this gym has very well qualified attributes such as location, instructors, schedules and customer service, these attributes make it the leading brand. In second place was Marmak Gym, closely followed by Geras Gym in third place, Optimus Gym in fourth place and finally Chinos Gym., El objetivo de la investigación fue determinar el posicionamiento de los gimnasios del Municipio de Rioverde S.L.P. en base a sus atributos. El estudio fue de tipo cuantitativo, descriptivo, correlacional y de diseño transversal. La población para estudiar para este proyecto estuvo constituida por personas que asisten a los gimnasios del municipio de Rioverde que fueron seleccionados para desarrollar esta investigación como son Chino´s Gym, Fitness Ten Gym, Geras Gym, Marmak Gym y Optimus Gym. La población que se determinó es infinita ya que se desconoce la cantidad exacta de las personas que acuden a los gimnasios, el muestreo utilizado fue probabilístico por conglomerados, también llamado clúster o muestreo por racimos. Para la recopilación de la información se requirió un cuestionario que se estructuró con base en lo que se necesitaba saber. Con los datos obtenidos, se pudo concluir que el gimnasio mejor posicionado ante sus competidores de acuerdo con sus atributos fue Fitness Ten Gym, ya que, este gimnasio posee atributos muy bien calificados como son, ubicación, instructores, horarios y atención al cliente, estos atributos lo hacen la marca líder. En segundo lugar, se encuentra Marmak Gym, seguido muy de cerca por el tercer lugar que se lo lleva Geras Gym, en la cuarta posición se encuentra Optimus Gym y por último encontramos a Chinos Gym.
