Abstract

Among tunicates, gamete morphology and sperm-egg interactions have been extensively investigated in ascidians, and to a lesser extent in appendicularians and thaliaceans. Sperm-egg interaction has been studied in only one salp, Pegea socia (Bosc, 1802). To determine if the pattern of internal fertilization of P. socia is generally applicable to salps, we performed an ultrastructural study on blastozooids of Thalia democratica (Forsskal, 1775). The ovary, located in the mantle near the gut, consists of a single oocyte connected to the atrial chamber wall by a 'fertilization duct', resembling a stack of single cells without a lumen. The flagellate sperm has a long corkscrew-like head with the single mitochondrion twisted around the nucleus. Fertilization is internal, and sperm actively penetrate the atrial wall and bore through the cells of the fertilization duct. During this process, the fertilization duct shortens as the cells move apart, one to one side and the next to the other, and rejoin to form a central lumen, which contains many sperm. At the same time a few sperm reach the periovular space for fertilizing the oocyte. Comparisons with P. socia indicate that this singular mode of internal fertilization with a complex corkscrew sperm actively penetrating the fertilization duct cells, probably evolved in the salp ancestor and has been modified to some extent in various genera. Chez les tuniciers, la morphologie des gametes et les interactions spermatozoides-oeufs ont ete bien etudiees chez les ascidies, mais moins bien chez les appendiculaires et les thaliaces. Les interactions entre les spermatozoides et les oeufs sont connues chez une seule salpe, Pegea socia (Bosc, 1802). Afin de determiner si le patron de fecondation interne de P. socia s'applique de facon generale aux salpes, nous avons etudie l'ultrastructure des blastozooides de Thalia democratica (Forsskal, 1775). L'ovaire, situe dans le manteau pres du tube digestif, consiste en un seul oocyte relie au mur de la chambre de l'atrium par un << conduit de fecondation >> ressemblant a un amoncellement de cellules individuelles sans lumiere. Le spermatozoide flagelle possede une longue tete en tire-bouchon avec une seule mitochondrie tournee autour du noyau. La fecondation est interne et les spermatozoides penetrent activement dans la paroi de l'atrium et percent les cellules du conduit de fecondation. Durant ce processus, le conduit de fecondation raccourcit, alors que les cellules se detachent, les unes d'un cote, les autres de l'autre, et se rejoignent pour former une lumiere centrale qui contient de nombreux spermatozoides. Au meme moment, quelques spermatozoides atteignent l'espace periovulaire pour feconder l'oocyte. Les comparaisons avec P. socia indiquent que ce mode particulier de fecondation interne, dans lequel un spermatozoide complexe en tire-bouchon penetre activement dans les cellules du conduit de fecondation, a probablement evolue chez l'ancetre des salpes et s'est modifie jusqu'a un certain degre dans les differents genres. Abstract

Background In tunicates, the capacity to build an adult body via non-embryonic development (NED), i.e., asexual budding and whole body regeneration, has been gained or lost several times across the whole subphylum. A recent phylogeny of the family Styelidae revealed an independent acquisition of NED in the colonial species Polyandrocarpa zorritensis and highlighted a novel budding mode. In this paper, we provide the first detailed characterization of the asexual life cycle of P. zorritensis. Results Bud formation occurs along a tubular protrusion of the adult epidermis, the stolon, in a vascularized area defined as budding nest. The bud arises through a folding of the epithelia of the stolon with the contribution of undifferentiated mesenchymal cells. This previously unreported mode of bud onset leads to the formation of a double vesicle, which starts to develop into a zooid through morphogenetic mechanisms common to other Styelidae. The budding nest can also continue to accumulate nutrients and develop into a round-shaped structure, designated as spherule, which represents a dormant form able to survive low temperatures. Conclusions To understand the mechanisms of NED and their evolution, it is fundamental to start from a robust phylogenetic framework in order to select relevant species to compare. The anatomical description of P. zorritensis NED provides the foundation for future comparative studies on plasticity of budding and regeneration in tunicates. 18. Evolutionary conservation of the placodal transcriptional network during sexual and asexual development in chordates

Abstract

Background: An important question behind vertebrate evolution is whether the cranial placodes originated de novo, or if their precursors were present in the ancestor of chordates. In this respect, tunicates are of particular interest as they are considered the closest relatives to vertebrates. They are also the only chordate group possessing species that reproduce both sexually and asexually, allowing both types of development to be studied to address whether embryonic pathways have been co-opted during budding to build the same structures. Results: We studied the expression of members of the transcriptional network associated with vertebrate placodal formation (Six, Eya, and FoxI) in the colonial tunicate Botryllus schlosseri. During both sexual and asexual development, each transcript is expressed in branchial fissures and in two discrete regions proposed to be homologues to groups of vertebrate placodes. Discussion: Results reinforce the idea that placode origin predates the origin of vertebrates and that the molecular network involving these genes was co-opted in the evolution of asexual reproduction. Considering that gill slit formation in deuterostomes is based on similar expression patterns, we discuss possible alternative evolutionary scenarios depicting gene co-option as critical step in placode and pharynx evolution. Abstract

The colonial tunicate Botryllus schlosseri is a widespread filter-feeding ascidian that lives in shallow waters and is easily reared in aquaria. Its peculiar blastogenetic cycle, characterized by the presence of three blastogenetic generations (filtering adults, buds, and budlets) and by recurrent generation changes, has resulted in over 60 years of studies aimed at understanding how sexual and asexual reproduction are coordinated and regulated in the colony. The possibility of using different methodological approaches, from classical genetics to cell transplantation, contributed to the development of this species as a valuable model organism for the study of a variety of biological processes. Here, we review the main studies detailing rearing, staging methods, reproduction and colony growth of this species, emphasizing the asymmetry in sexual and asexual reproduction potential, sexual reproduction in the field and the laboratory, and self- and cross-fertilization. These data, opportunely matched with recent tanscriptomic and genomic outcomes, can give a valuable help to the elucidation of some important steps in chordate evolution.
