Abstract

Extracellular vesicles (EVs)‐based intercellular communication (through exosomes, microvesicles, and apoptotic bodies) is conserved across all kingdoms of life. In recent years, exosomes have gained much attention for targeted pharmaceutical administration due to their unique features, nanoscale size, and capacity to significantly contribute to cellular communication. As drug delivery vehicles, exosomes have several advantages over alternative nanoparticulate drug delivery technologies. A key advantage lies in their comparable makeup to the body's cells, which makes them non‐immunogenic. However, exosomes vesicles face several challenges, including a lack of an effective and standard production technique, decreased drug loading capacity, limited characterization techniques, and underdeveloped isolation and purification procedures. Exosomes are well known for their long‐term safety and natural ability to transport intercellular nucleic acids and medicinal compounds across the blood‐brain‐barrier (BBB). Therefore, in addition to revealing new insights into exosomes' distinctiveness, the growing availability of new analytical tools may drive the development of next‐generation synthetic systems. Herein, light is shed on exosomes as drug delivery vehicles in anti‐infective therapy by reviewing the literature on primary articles published between 2002 and 2023. Additionally, the benefits and limitations of employing exosomes as vehicles for therapeutic drug delivery are also discussed. [ABSTRACT FROM AUTHOR]

Abstract

Background: Urolithiasis, which is acquiring clinical relevance, is one of the most prevalent diseases seen in urology clinics. The most often reported uroliths in humans are oxalate, struvite, urate, brushite and cystine. The globally rising occurrence and severity of urolithiasis has made it an issue of demanding medical attention. Medicinal herbs and natural substances have been shown to be useful in the treatment and management of urolithiasis across various recent investigations. Calamus floribundus Griff. belongs to the Aracaceae family and its urolithiatic potential has yet to be established. Objectives: The objective of the study is to evaluate the antiurolithiatic potential of methanolic extract Calamus floribundus Griff. utilizing in vitro and in vivo methods. Materials and Methods: Nucleation assay and aggregation assay was done to evaluate the in vitro antiurolithiatic activity. In the in vivo studies ethylene glycol induced and glycolic acid induced model were used. MTT assay was done to evaluate the cytotoxic effect of the methanolic extract of Calamus floribundus Griff. Results: The IC50 value of the extract was found to be 767.394±0.22 µg/mL and 759.435±0.29 µg/mL in the nucleation and aggregation assay respectively. In the in vivo study all the biochemical parameters were normalised in dose dependent manner. Methanolic extract exhibited cytotoxic effect on NRK-52E cell lines with a IC50 value of 11.1 µg/mL. Conclusion: The obtained results indicates for the antiurolithiatic potential of Calamus floribundus Griff. [ABSTRACT FROM AUTHOR]

Abstract

Cold atmospheric plasma (CAP) is a novel technology with boundless significance that can be used in the medical sector that offers noninvasive in-vivo applications without damaging the living tissues. CAPs can be obtained by curtailing the concentration of high-energetic electrons per phase and by freezing molecules/atoms (devoid of charge) in plasma utilizing gas circulation and atmospheric air, which includes a variety of charged and neutral reactive entities, UV rays, electric currents, and fields, etc. that have an influence on cellular material in a multitude of diverse manners. Reactive oxygen species (ROS) and reactive nitrogen species (RNS), produced by the plasma, essentially cause biological and therapeutically advantageous plasma effects. CAP plasma has several important biological functions, including the deactivation of pathogens, induction of tissue restoration and cell propagation, the annihilation of cells by triggering apoptosis, etc. Several fundamental concepts are defined, even if the precise process of the effect of plasma on biomolecules is still not properly identified. Depending on the biological synthesis of RNS and ROS in reactions to plasma emissions, the present review described several aspects of plasma therapy in neuroscience, particularly in anti-glioblastoma, neuro-differentiation, and neuroprotection and also the various applications of CAP in medical fields where it is used in the therapy of SARS-CoV-2, cancer therapy, and chronic and acute wounds. Furthermore, the proliferation in stem cells, dental medicines, dermatology, and a brief insight into CAP devices and their risk factors was highlighted.
