Tese de doutoramento em Ciências - Especialidade de Química, Wound repair is a highly complex process that leads to the formation of new tissue after injury. Under an inflammatory process, neutrophils form a primary line of defence against bacterial infections using complementary oxidative and non-oxidative pathways to destroy phagocytised pathogens. Neutrophil activation results in the release of proteases, such as elastase, into the extracellular medium as proteolytically active enzymes. The excessive and uncontrolled release of proteolytic enzymes found in chronic wounds leads to considerably reduced amounts of growth factors and protease inhibitors and to an extensive degradation of the extracellular matrix. Several therapeutic approaches have been investigated to neutralise the deleterious effect of high protease levels in chronic wounds. Within this research work two distinct approaches have been proposed to promote the controlled release of novel elastase inhibitor-peptides to the wound medium. In the first approach, the inhibitors activity was tuned by a posttranslational modification (phosphorylation) that induced differential inhibitory activity towards elastase. The biomatrix system designed to incorporate this system was a protease-degradable biomatrix composed of silk fibroin and keratin. The second approach relies in the incorporation of these synthetic elastase inhibitor-peptides in a biodegradable hydrogel based on hyaluronic acid (HA). This biomimetic material was applied as a model drug delivery system due to its triple function: sequestration of elastase from the wound environment; inhibition of elastase prompted by the elastase inhibitor-peptides and the ability of HA to promote natural healing and regeneration. These novel elastase inhibitor-peptides were selected from specific sequences of endogenous proteins, like secretory leucocyte protease inhibitor (SLPI), elafin, eosinophil cationic protein (ECP) and surfactant protein D (SP-D). The peptide sequences, KRCCPDTCGIKCL (Pep4) and KRMMPDTMGIKML (Pep4M), revealed both the ability to moderately inhibit elastase activity (a decrease on elastase activity of ~40%) and to be phosphorylated by an appropriated protein kinase (casein kinase Iδ, CKI). The phosphorylation of these peptides on-chip (micro-array technology) and in solution (PKLight HTS assay) allowed to attain the optimal phosphorylation conditions for the current peptides and in combination with the elastase activity assays, was thus possible to perceive a decrease in the inhibitory capacity of these peptides towards elastase, when in the phosphorylated form. Furthermore, the stability of the peptides and casein kinase was evaluated in presence of the proteolytic enzyme elastase by mass spectrometry and electrophoresis (SDS-PAGE). Both the peptides and protein kinase have revealed to be elastase degradable materials (after 5, 15 and 60 minutes for CKI, Pep4 and Pep4M, respectively), therefore appropriated materials to be incorporated in a wound dressing applications. To determine the nature of the interaction between elastase and the inhibitor-peptides, it was assessed the kinetic parameters for the substrate hydrolysis by elastase (PPE-porcine pancreatic elastase and HNE-human neutrophil elastase) in presence of both peptides. According to the kinetic studies, these peptides behave as uncompetitive and noncompetitive inhibitors of HNE and PPE, respectively. Additionally, it was also evaluated the influence of the reactive oxygen species (ROS), which are known to be present under inflammatory conditions, on the inhibitory capacity of the current peptides relatively to elastase. Unexpectedly, the presence of ROS and also albumin prompted an increase in the inhibitory activity of both peptides towards elastase (a maximum increase of 27 and 20% for Pep4 and Pep4M, respectively). The structure of these peptides in solution was determined by two dimensional NMR and the interactions of these peptides, in the phosphorylated and non-phosphorylated form, with the serine protease (PPE) was evaluated by molecular modelling. These studies, evidenciated the differential conformations adopted by the peptides, prompted by to interresidue and inter-molecular interactions, when these peptides are in the phosphorylated and non-phosphorylated form. Lastly, a preliminary study on novel elastase inhibitor-peptides based on the binding segment of the endogenous elastase inhibitors SLPI (YGQCLML and LGQCLML) and elafin (LIRCAML, LIRCLML and LIRCVML) was also comprised within this research work. Unexpectedly, these peptides do not behave as PPE inhibitors (higher PPE inhibition levels of 19%), although they behave as potential HNE inhibitors (higher HNE inhibition levels near 77%). Thus, the incorporation of the peptides reported within this research work (exhibiting moderated elastase inhibition) into biomimetic materials (hyaluronic acid hydrogels), results in wound dressing formulations with potential application to wounds under severe inflammatory conditions (with high content in ROS), once the inhibitory capacity of these peptides will be enhanced under such conditions., A cicatrização de uma ferida é um processo muito complexo, que leva à formação de um novo tecido após a lesão. Perante um processo inflamatório, os neutrófilos formam a primeira linha de defesa contra as infeções bacterianas, usando processos complementares oxidativos ou não-oxidativos, para destruir os agentes patogénicos fagocitados. A ativação dos neutrófilos desencadeia a libertação de proteases, como a elastase, para o meio extracelular sob a forma de enzimas proteoliticamente ativas. Em feridas crónicas observa-se uma libertação excessiva e incontrolada de enzimas proteolíticas, que conduzem a uma redução dos níveis dos fatores de crescimento e dos inibidores endógenos das proteases e a uma considerável degradação da matriz extracelular. Até à data, foram investigadas diversas abordagens terapêuticas com o objetivo de neutralizar os efeitos nefastos provocados pelos níveis elevados de proteases em feridas crónicas. No âmbito deste trabalho, foram propostas duas abordagens distintas com o intuito de promover a libertação controlada de novos inibidores peptídicos da elastase em feridas crónicas. Na primeira abordagem, a atividade dos inibidores é regulada através de uma modificação pós-traducional (fosforilação), a qual induz uma inibição diferencial da elastase por parte do péptido. A segunda abordagem, baseou-se na incorporação dos inibidores peptídicos sintéticos da elastase, num hidrogel biodegradável à base de ácido hialurónico (HA). Este material biomimético foi aplicado como sistema de libertação controlada de agentes ativos, pela sua tripla função: sequestrar a elastase da zona lesionada; inibição promovida pelos inibidores peptídicos da elastase e a capacidade do HA em promover a cicatrização e regeneração dos tecidos. Estes novos inibidores peptídicos da elastase foram selecionados a partir de sequências específicas de proteínas endógenas, como o secretory leucocyte protease inhibitor (SLPI), elafin, eosinophil cationic protein (ECP) e surfactant protein D (SP-D). As sequências peptídicas, KRCCPDTCGIKCL (Pep4) e KRMMPDTMGIKML (Pep4M), demonstraram uma capacidade moderada de inibição da atividade da elastase (diminuição de ~40%). A fosforilação dos péptidos Pep4 e Pep4M on-chip (tecnologia de micro-array) e em solução (ensaio PKLight HTS) por uma proteína cinase específica (caseína cinase Iδ, CKI), permitiram estabelecer as condições ótimas de fosforilação para estes péptidos. A combinação destes ensaios, com os de medição da atividade da elastase, permitiu compreender a diminuição da capacidade de inibição destes péptidos quando se encontram na forma fosforilada. Além disso, a estabilidade dos péptidos e da caseína cinase foi avaliada na presença da enzima proteolítica elastase por espectrometria de massa e eletroforese (SDS-PAGE). Os péptidos selecionados e a proteína cinase revelaram ser degradados pela elastase (após 5, 15 e 60 minutos para a CKI, Pep4 e Pep4M, respetivamente), demonstrando serem materiais apropriados para incorporação em compressas. Para determinar a natureza das interações entre a elastase e os inibidores peptídicos, foram obtidos parâmetros cinéticos para a hidrólise do substrato (N-succinyl-Ala-Ala- Ala-p-nitroanilide e MeO-Succinyl-(Ala)2-Pro-Val-p-nitroanilide) pela enzima (PPEporcine pancreatic elastase e HNE- human neutrophil elastase), na presença dos dois péptidos. De acordo com os estudos cinéticos, estes péptidos comportaram-se como inibidores incompetitivos e não-competitivos da HNE e PPE, respetivamente. Para além disso, determinou-se a influência das espécies reativas de oxigénio (ROS), presentes em condições inflamatórias, na capacidade inibitória dos péptidos estudados. Surpreendentemente, a presença de ROS e de albumina induzem um aumento da atividade inibitória de ambos os péptidos para a elastase (um aumento máximo de 27 e 20% para o Pep4 e Pep4M, respetivamente). A estrutura dos péptidos Pep4 e Pep4M em solução foi determinada pela técnica de RMN bidimensional e a interação destes péptidos, na forma fosforilada e não-fosforilada, com a protease de serina (PPE) foi avaliada por modelação molecular. Estes estudos evidenciaram as diferentes conformações adotadas pelos péptidos, induzidas por interações inter-resíduo e intermoleculares, quando estes se encontram na forma fosforilada e não-fosforilada. Por fim, foi realizado um estudo preliminar sobre novos inibidores peptídicos da elastase, baseados na sequência de ligação dos inibidores endógenos SLPI (YGQCLML e LGQCLML) e elafin (LIRCAML, LIRCLML e LIRCVML). Contrariamente ao esperado, estes péptidos não se comportaram como inibidores da PPE (máximo de inibição 19%) porém, comportaram-se como potenciais inibidores da HNE (máximo de inibição ~77%). A incorporação dos péptidos envolvidos neste estudo, que exibem moderada inibição da elastase, em hidrogéis de ácido hialurónico resultou em materiais biomiméticos com potencial aplicação terapêutica em compressas para feridas crónicas.
