微生物发酵法生产番茄红素是直接提取和化学合成的有效替代策略。以油酸为外源脂肪酸，探究其对粗糙脉孢菌发酵产番茄红素的影响，通过响应面法优化确定最佳培养条件，并进一步探究油酸对番茄红素合成相关酶的影响。结果表明：在含油酸的培养基中，粗糙脉孢菌的番茄红素产量显著增加；在油酸质量浓度0.90 g/L、振荡速率100 r/min、振荡培养温度34 ℃、静置培养温度30 ℃条件下，番茄红素产量达到4.21 mg/L，是空白对照组的2.77倍。培养基中油酸的添加不仅促进了粗糙脉孢菌生物量的积累，菌体内乙酰辅酶A的水平也显著增加；同时，与番茄红素合成相关酶（HMG-CoA合成酶、HMG-CoA还原酶、香叶基香叶基焦磷酸合酶）的水平亦显著升高。结果表明油酸的添加可供利用更多的乙酰辅酶A进入甲羟戊酸途径，并进一步在上述酶的诱导作用下促进番茄红素的合成。 The lycopene production through microbial fermentation method is an effective alternative strategy for direct extraction and chemical synthesis. With oleic acid as exogenous fatty acid, the effect of oleic acid on lycopene production in Neurospora crassa was studied, and the response surface methodology was used to determine the optimal fermentation conditions, and the levels of enzymes associated with lycopene synthesis were further studied in the presence or absence of oleic acid. The results showed that adding oleic acid to the medium significantly increased the lycopene yield in N.crassa, and the optimal fermentation conditions were obtained as follows: oleic acid mass concentration 0.90 g/L, shaking rate 100 r/min, shaking culture temperature 34 ℃ and static culture temperature 30 ℃. Under these conditions, lycopene yield was 4.21 mg/L, which was 2.77 times that of the control group. Moreover, oleic acid added in the medium could not only increase the biomass of N. crassa, but also significantly increase the acetyl-CoA content compared with the control group. Meanwhile, the levels of enzymes associated with lycopene synthesis (HMG-CoA reductase, HMG-CoA synthase and geranylgeranyl pyrophosphate synthase) also significantly increased. These results suggested that more available acetyl-CoA could enter the mevalonate pathway and further promote the synthesis of lycopene under the action of the above enzymes by adding oleic acid.

Objective: To investigate the lower limb muscle strength characteristics in amateur runners with patellofemoral pain (PFP) . Method: Thirty PFP amateur runners (20 males and 10 females) were recruited as the knee pain group, and 30 matched non-injured amateur runners were recruited as the non-knee pain group. The hip abductor strength, quadriceps strength, hamstring strength, hamstring/quadriceps ratio, and bilateral limb symmetry index were collected for all participants. Independent sample t-tests were used to compare the differences between the groups. Result: The hip abductor strength and bilateral limb symmetry index, quadriceps strength and bilateral limb symmetry index of male PFP amateur runners were significantly lower than those of the non-knee pain group, while the hamstring/quadriceps ratio was significantly higher than that of the non-knee pain group. The hip abductor strength and bilateral limb symmetry index of female PFP amateur runners were significantly lower than those of the non-knee pain group. Conclusion: PFP amateur runners exhibit different lower limb muscle strength characteristics compared to non-knee pain runners, and the lower limb strength characteristics of male and female PFP amateur runners are not consistent. 【Objective】The present study aimed to reveal differences in the response of fir seedlings to different light qualities by studying chloroplast ultrastructure, photosynthetic pigment content and photosynthesis-related parameters of fir seedlings under different light qualities to provide a theoretical basis for fir cultivation. 【Method】 The 1-year-old excellent clone 020 cuttings were used as the material, and LED intelligent dimming table was used as the artificial light source. Four treatments were set up, including white light (CK), red light (R), blue light (B) and red and blue combined light (R∶B=1∶1), with three replicates in each treatment. The chloroplast structure was observed by transmission electron microscope, and the related photosynthetic parameters and photosynthetic pigments were determined.【Result】The net photosynthetic rate, stomatal conductance, and transpiration rate under white light and red-blue composite light treatments were higher than those under red light treatment and blue light treatment. Among them, the white light treatment was 130.61%, 204.05% and 112.14% higher than that of the red-light treatment, 37.80%, 31.34% and 49.91% higher than that of the blue light treatment, respectively, while the red-blue composite light treatment was 89.02%, 176% and 51.09% higher than that of the red light, 13.07%, 18.96% and 6.81% higher than that of the blue light treatment, respectively. The intercellular CO2 of seedling leaves under white light treatment and red-light treatment was higher than that of blue light treatment and red-blue composite light treatment. Among them, the white light treatment was 6.32% and 5.10% higher than that of the blue light and red-blue composite light treatments, and the red-light treatment was 4.18% and 3.01% higher than that of the blue light and red-blue composite light treatments. Photosynthetic pigments were the lowest in the red and blue combined light treatment, 27.81%, 26.51% and 15.52% lower than that of the remaining three treatments, respectively. Chlorophyll ratios were the lowest under red light treatment, 3.32%, 1.90% and 3.79% lower than that of the remaining three treatments, respectively, while they were maximum under red-blue complex light treatment, significantly higher than that of the remaining three treatments. The observation of chloroplast ultrastructure yielded that the cell wall was more complete and smoother under red and blue combined light, and the basal granules, stromal lamellae and their structures were more complete.【Conclusion】The photosynthetic characteristics of fir seedlings differed greatly under different light quality treatments. Objective To explore the effect of 2,3,5,4'-tetrahydroxystilbene-2-O-β-D- glucoside (TSG) and Emodin, the main components of polygonum multiflorum, on the apoptosis of hippocampal neuron HT-22 cells induced by high glucose. Methods The mouse hippocampal neuron HT-22 cells were divided into 4 groups: the control medium group (NC group, glucose concentration 25 mmol/L), the high-glucose medium group (HG group, glucose concentration 55 mmol/L), the HG+TSG group and the HG+Emodin group. The HG+TSG group and the HG+Emodin group were treated with TSG and Emodin at the concentrations of 50, 100 and 200 μmol/L for 12, 16, 20, 24 and 48 h, respectively. The cell viability was detected by CCK-8 method after treatment. Histone acetyltransferase (HAT) and histone deacetylase (HDAC) enzyme activities were detected by enzyme-linked immunosorbent assay (ELISA). Flow cytometry was used to detect the apoptosis rate. Western blotting was used to detect the expression of apoptosis-related proteins: B-lymphoma-2 (Bcl-2), Bcl-2-related X protein (Bax) and Caspase-3. Results The 200 μmol/L of TSG for 48 h, and 100 μmol/L of Emodin for 48 h were the optimal condition for culturing cells after testing. Flow cytometry results showed that TSG and Emodin significantly down-regulated cell apoptosis rate after 48 h of treatment. ELISA results showed that the activity of HAT was significantly down-regulated, HDAC activity was up-regulated in the HG+TSG group. The activities of HAT and HDAC were significantly down-regulated in the HG+Emodin group. Western blot results showed that the expressions levels of Bax and Caspase-3 were significantly down-regulated, and the expression of Bcl-2 was significantly up-regulated after TSG and Emodin treated cells for 48 h. Conclusion TSG and Emodin may regulate the activities of HAT and HDAC, thereby downregulate the apoptosis of hippocampal neurons induced by high glucose. [ABSTRACT FROM AUTHOR]

Lunar sampling manipulator has large span and a low stiffness. Its links and joints are prone to large flexible deformation under static external force, which affects the absolute end positioning accuracy of the manipulator. At the same time, the kinematics error caused by manufacturing assembly error also increases the difficulty to the improvement of the manipulator’s accuracy. In order to improve the absolute positioning accuracy of the end, a method for robot kinematics parameter calibration based on elastic error model is proposed. Firstly, based on the modified Denavit-Hartenberg(MDH)model, the kinematics model and the geometric error model are established. Secondly, based on the force analysis of the joints and links of the manipulator, the elastic error model is established to reflect the relation between the elastic error and the end positioning error of the manipulator. Thirdly, the kinematics error model is developed by combining the geometric error model with the elastic error model. Finally, a method for kinematic calibration is designed and kinematic parameters is identified. BACKGROUND: Bone marrow mesenchymal stem cells have been extensively applied in animal experiments and clinical studies. The cell concentration, treatment times and results in each study are different, and there is no standard for optimal cell concentration. OBJECTIVE: To investigate the optimal concentration of bone mesenchymal stem cells injected into articular cavity in the treatment of rabbit cartilage defects. METHODS: Thirty 6-month-old New Zealand white rabbits were selected and randomly divided into control, 1×108, 1×109, 1×1010, and 1×1011/L groups. Cartilage defect models with diameter of 3 mm and depth of 2 mm were established in femoral trochlea in each group. One week after modeling, 1 mL of normal saline was injected into the rabbit’s knee of the control group. The other groups were injected with bone marrow mesenchymal stem cells at corresponding concentrations. After 6 and 12 weeks, gross observation, hematoxylin-eosin staining, Safranin-O-fast green-staining, type I and II collagen staining were performed to assess the cartilage regeneration. RESULTS AND CONCLUSION: In the control group, the defect area was obvious with no cartilage regeneration. The 1×108, 1×109, and 1×1010/L groups showed cartilage regeneration. The repairing effect was increased with the cell concentration increasing. The effect of cartilage repair in the 1×1011/L group was similar to that in the 1×1010/L group (P > 0.05). Therefore, 1×1010/L is the optimal concentration for intra-articular injection of bone marrow mesenchymal stem cells for treating cartilage defects, and higher concentration cannot enhance the repairing effect. To evaluate the effects of periodontal basic therapy on hemoglobin (HbA1c) and periodontal status of diabetic patients, a total of 102 patients with type 2 diabetes mellitus combined chronic periodontitis, which included male 58 and female 44 with average age(45±9)yrs, were enrolled from January 2011 to December 2013. The patients were randomly divided into two groups. The intervention group of 51 patients received hypoglycemic therapy and systemic periodontal therapy. The control group only received hypoglycemic therapy. Periodontal clinical examination indexes included the probing depth(PD), clinical attachment loss(CAL) and aspartate transaminase(AST) in gingival crevicular fluid. Glycemic control was measured as glycosylated hemoglobin(HbA1c). After supragingival scaling for 3 months and subgingival curettage for 3 months, periondontal indexes of fixed tooth position in the same site and the corresponding HbA1c were recorded in periodontal treatment group. Same indexes were recorded in the control group, too. Independent sample t test was used for the difference between groups. Chi square test was used for the comparison of categorical data. P<0.05 was considered as statistically significant difference. After subgingival curettage for 3 months, PD(1.92 ± 0.62)mm, CAL(1.79 ± 0.72)mm, AST(364.0 ± 105.2)mmol/L, in intervention group were significantly lower than those in the control group:PD(2.56±0.65)mm, CAL (2.15± 0.57)mm, AST (532.3 ± 112.5)mmol/L (t=2.438, 3.375, 3.591, all P<0.05). In addition, HbA1c (7.3 ± 1.9)%of intervention group was much lower than that (8.1 ± 1.9)% of control group (χ2=2.831, P<0.05). The results suggest supragingival scaling combined with subgingival curettage not only alleviated significantly inflammation of periodontal tissues, but also improved glycemic control of type 2 diabetic patients.
