Abstract

To verify the fatigue life under high-low cycle combined loads of the blade the combined fatigue experimental technique was established. The orthogonal loads decoupling method was proposed based on the cylindrical rolled bearing and linear slide rail,and the coordinated loading control technique of a combined loads was presented by means of an external signal trigger. Then,the fatigue experiment of a certain type of ducted tail rotor fan blade under centrifugal load/Low Cycle Fatigue（ LCF） and aerodynamic load/High Cycle Fatigue（ HCF） was developed. The control precision of complex loads is superior to than ±1%,and the coordinated loading time interval is less than 0. 3 second. The experimental result indicates that the high-low cycle combined fatigue experimental technique can solve the technical problems of orthogonal loads decoupling and coordinated loading control,which provides the technical foundation for improving the high-low cycle combined fatigue life prediction precision of the blade.

Abstract

Drought-induced stress poses a significant challenge to wheat throughout its growth, underscoring the importance of identifying drought-stable quantitative trait loci (QTLs) for enhancing grain yield. Here, we evaluated 18 yield-related agronomic and physiological traits, along with their drought tolerance indices, in a recombinant inbred line population derived from the XC7 × XC21 cross. These evaluations were conducted under both non-stress and drought-stress conditions. Drought stress significantly reduced grain weight per spike and grain yield per plot. Genotyping the recombinant inbred line population using the wheat 90K single nucleotide polymorphism array resulted in the identification of 131 QTLs associated with the 18 traits. Drought stress also exerted negative impacts on grain formation and filling, directly leading to reductions in grain weight per spike and grain yield per plot. Among the identified QTLs, 43 were specifically associated with drought tolerance across the 18 traits, with 6 showing direct linkages to drought tolerance in wheat. These results provide valuable insights into the genetic mechanisms governing wheat growth and development, as well as the traits contributing to the drought tolerance index. Moreover, they serve as a theoretical foundation for the development of new wheat cultivars having exceptional drought tolerance and high yield potentials under both drought-prone and drought-free conditions. 247. HIF‐1α Contributes to Hypoxia‐induced VSMC Proliferation and Migration by Regulating Autophagy in Type A Aortic Dissection.

Abstract

Type A aortic dissection (AD) is a catastrophic cardiovascular disease. Hypoxia‐inducible factor‐1α (HIF‐1α) and autophagy are reported to be upregulated in the AD specimens. However, the interaction between HIF‐1α and autophagy in the pathogenesis of AD remains to be explored. HIF‐1α and LC3 levels are evaluated in 10 AD and 10 normal aortic specimens. MDC staining, autophagic vacuoles, and autophagic flux are detected in human aortic smooth muscle cells (HASMCs) under hypoxia treatment. CCK‐8, transwell, and wound healing assay are used to identify proliferation and migration under hypoxia treatment. Furthermore, 3‐MA is used to inhibit autophagy in hypoxia‐treated HASMCs. This study reveals that AD tissues highly express HIF‐1α and the LC3. Autophagy is induced under hypoxia in a time‐dependent manner, and autophagy is positively related to HIF‐1α in HASMCs. Moreover, the proliferation and migration of HASMCs are enhanced by hypoxia, whereas the knockdown of HIF‐1α attenuates this effect. Additionally, inhibiting autophagy with 3‐MA ameliorates hypoxia‐induced proliferation and migration of HASMCs. In summary, the above results indicate that HIF‐1α facilitates HASMC proliferation and migration by upregulating autophagy in a hypoxic microenvironment.
