Ethnopharmacological relevance Jiang-Xian HuGan (JXHG) formulated by five natural products including Freshwater clam (Corbicula fluminea) , Curcuma longa L., Ligustrum lucidum , Eclipta prostrata (L.) L. and Paeonia lactiflora Pall. , has exhibited a great hepatoprotective effect. Aim of this study We investigated the effect of JXHG on concanavalin A (ConA)-induced acute live injury in mice, and to elucidate its underlying molecular mechanisms. Materials and methods Jiangkanling Capsule (900 mg/kg), low-dose JXHG (LJXHG, 700 mg/kg), high-dose JXHG (HJXHG, 1400 mg/kg) were administered to mice by oral gavage daily for 20 days prior to a single intravenous injection of ConA (20 mg/kg). Liver injury was evaluated by measuring the serum levels of enzymes and cytokines as well as liver histological analysis. We also measured the hepatic expression of cytokines at mRNA levels and the proteins related to NF-κB and Nuclear factor (erythroid-derived 2)-like 2 (Nrf2) signaling pathways. Result Our results showed that JXHG pretreatment significantly alleviated ConA-induced live injury as evidenced by decreased serum levels of glutamic-pyruvic transaminase (ALT) and glutamic oxalacetic transaminase (AST), and reduced hepatocyte apoptosis and mortality. Furthermore, JXHG was able to significantly reduce the serum levels of proinflammatory cytokines, down-regulate the mRNA expression of interleukin-6 (IL-6) and interferon-γ (IFN-γ), and up-regulate IL-10 as well as superoxide-dimutase-1 (SOD1), glutathione reductase (GSR) and Glutathione peroxidase 2 (GPX2) mRNA in the liver tissues after Con A injection. In addition, JXHG pretreatment dramatically suppressed the phosphorylation of NF-κB p65 (p65), increased Nrf2 expression, and decreased the expression ratio of cleaved caspase-3/caspase-3 in liver tissues. Conclusion These results suggest that JXHG protects against ConA-induced acute live injury through inhibiting NF-κB mediated inflammatory pathway and promoting Nrf2 mediated anti-oxidative stress signaling pathway. Abstract: Chinese grasslands have undergone great changes in land use in recent decades. Approximately 18.2% of the present arable land in China originated from the cultivation of grassland, but its impact on the carbon cycle has not been fully understood. This study was conducted in situ for 3 years to assess the comprehensive effects of cultivation of temperate steppe on soil organic carbon (SOC) and soil respiration rates as well as ecosystem respiration. As compared with those in the Stipa baicalensis steppe, the SOC concentrations at depths of 0–10 and 10–20cm in the spring wheat field were found to have decreased by 38.3 and 17.4% respectively from 29.5 and 21.9gkg−1 to 18.2 and 18.1gkg−1 after a cultivation period of 30 years. Accordingly, the total amounts of soil respiration through the growing season (from April to September) in 2002, 2003 and 2004 were 265.2, 282.2 and 237.4gCm−2 respectively in the spring wheat field, which were slightly lower than the values of 342.2, 412.0 and 312.1gCm−2 in the S. baicalensis steppe, while ecosystem respiration of 690.9, 991.2 and 569.6gCm−2 respectively in the spring wheat field were markedly higher than those of 447.0, 470.9 and 429.7gCm−2 in the steppe plot. Similar seasonal variations of ecosystem respiration and soil respiration existed in both sample sites. Respiration rates were higher and greater differences existed in both ecosystem respiration and soil respiration during the exuberant growth stage of plants (from mid-June to mid-August). However, in the slower-growth period of the growing season (before late May and after late August), the CO2 effluxes of the two sample sites were similar and remained at a relatively low level. The results also showed that ecosystem respiration and soil respiration were under similar environmental controls in both sample sites. Soil water content at a depth of 0–10cm and soil temperatures at 5 and 10cm were the main factors affecting the variations in ecosystem respiration and soil respiration rates in droughty years of 2002 and 2004 and in the rainy 2003, respectively. This study suggests that the conversion of the grassland to the spring wheat field has increased the carbon loss of the whole ecosystem due to the change of vegetation cover type and significantly reduced the carbon storage of surface soil. In addition, the tillage of grassland had different effects on ecosystem respiration and soil respiration. The effects were also dissimilar in different growth stages, which should be fully considered when assessing and predicting the effects of cultivation on the net CO2 balance of grassland ecosystems. Background CH4 concentrations were continuously measured at the 4 WMO/GAW stations [Waliguan in Qinghai (WLG), Lin'an in Zhejiang (LAN), Shangdianzi in Beijing (SDZ), and Longfengshan in Heilongjiang (LFS)] by Cavity Ring Down Spectroscopy system. From 2009 to 2010, the diurnal cycle of hourly average CH4 concentration at LAN was found to be similar in all four seasons, with the highest level detected at 05:00 (Beijing Time) and the lowest at about 14:00. Similar CH4 diurnal cycles were observed at LFS in the summer time. However, the daily amplitude was much higher than that at LAN and reached 216. 8 x 10(-9) (molar ratio). For SDZ station, there were similar trends in spring, autumn and winter. The daily average concentration in the summer was much higher than those of the other seasons and reached the highest at about 20:00. No apparent CH4 diurnal cycle was observed at the WLG station during the whole year. The seasonal variations were obvious at the three regional stations (LAN, SDZ, LFS). The background concentration was the lowest in July at LAN while reached the highest level in August at LFS. The yearly background concentration variation at LFS displayed a "W" pattern. At LFS and SDZ, the wintertime CH4 concentrations were higher than those in spring and autumn. WLG represented a clean area and its CH4 value was the lowest among the four stations with the monthly average amplitude to be about 11.5 x 10(-9). At all three regional stations, non-background data accounted for more than 70% of the whole data. Cluster analysis of 3 day backward trajectories corresponding to the high CH4 concentration (WLG: CH4 > 1 870 x 10(-9), LFS: CH4 > 2100 x 10(-9), LAN: CH4 > 2 150 x10(-9), SDZ: CH4 > 2050 x 10(-9)) data points suggested that the high CH4 level measured in summer might be associated with the air mass transportation.

In-situ GC-FID system for atmospheric CH4 and CO mixing ratio measurements at the Shangdianzi (SDZ) GAW regional station in Beijing was designed and optimized in 2009 based on a comparable system at the Waliguan GAW global station in Qinhai. Results from this study indicate that the system's precisions for CH4 and CO are higher than 0.03% and 0.45% respectively, which can meet the quality target on background greenhouse gas observations by the World Meteorology Organization's Global Atmosphere Watch (WMO/GAW) program. The selection method of working standards for this system was established: two working standards (WH for the high concentration and WL for the low concentration) were selected, the concentrations of CH4 and CO in these two standards can cover the ambient mixing ratios of CH4 (2 007.1 x 10(-9) and 1 809.5 x 10(-9)) and CO (405.6 x 10(-9) and 123.8 x 10(-9)), an injection sequence was programmed so that the two standards were analyzed alternatively for every three runs. The measurement accuracies are high, as shown by the standard deviations less than 1.7 x 10(-9) and 1 x 10(-9), for CH4 and CO, respectively. This method has been applied to in-situ measurement of atmospheric CH4 and CO in North China.

To strengthen scientific management and sharing of greenhouse gas data obtained from atmospheric background stations in China, it is important to ensure the standardization of observations and establish the data treatment and quality control procedure so as to maintain consistency in atmospheric carbon dioxide (CO2) and methane (CH4) measurements from different background stations. An automated gas chromatographic system (Hewlett Packard 5890GC employing flame ionization detection) for in situ measurements of atmospheric CO2 and CH4 has been developed since 1994 at the China Global Atmosphere Watch Baseline Observatory at Mt. Waliguan, in Qinhai. In this study, processing and quality control flow of CO2 and CH4 data acquired by HP ChemStation are discussed in detail, including raw data acquisition, data merge, time series inspection, operator flag, principal investigator flag, and the comparison of the GC measurement with the flask method. Atmosphere CO2 and CH4 mixing ratios were separated as background and non-background data using a robust local regression method, approximately 72% and 44% observed values had been filtered as background data for CO2 and CH4, respectively. Comparison of the CO1 and CH, in situ data to the flask sampling data were in good agreement, the relative deviations are within +/- 0.5% for CO2 and for CH4. The data has been assimilated into global database (Globalview-CO2, Globalview-CH4), submitted to the World Data Centre for Greenhouse Gases (WDCGG), and applied to World Meteorological Organization (WMO) Greenhouse Gas Bulletin and assessment reports of the United Nations Intergovernmental Panel on Climate Change (IPCC).

Objective: To study the correlations of mannose-binding lectin (MBL) gene promoter polymorphisms and plasma MBL concentrations to the susceptibility to HIV infection in northern Chinese Han population., Methods: This case-control study included 115 HIV-infected patients and 115 non-infected healthy individuals, in whom the MBL gene promoter polymorphisms were detected using pyrosequencing technique and plasma MBL concentrations measured using enzyme-linked immunosorbent assay (ELISA)., Results: The MBL promoter genotypes of LY/LY, LY/LX, HY/LY, HY/HY and LX/LX were detected in 66 (57.40%), 25 (21.70%), 17 (14.80%), 5 (4.30%) and 2 (1.70%) among the HIV-infected patients, and in 77 (67.00%), 23(20.00%), 12 (10.40%), 0 (0.00%), and 3 (2.60%) among the healthy individuals, respectively. The frequencies of haplotypes LY, HY and LX were 75.70%, 11.70% and 12.60% among the patients, and 82.20%, 5.20% and 12.60% among the healthy individuals, respectively, showing significant difference in the halpotype between the two groups (P=0.041). The average plasma MBL concentration was significantly lower in HIV-infected group than in the healthy individuals (1775.14-/+786.31 vs 3672.21-/+597.13 microg/L, P=0.001)., Conclusion: The genotypes of LY/LY and LY/LX and the haplotypes of LY and HY are predominant in northern Chinese Han population, and the plasma MBL concentration in HIV infected patients is generally only 50% of that in healthy individuals. We therefore presume that MBL promoter polymorphisms and plasma MBL concentration can be associated with the susceptibility to HIV infection in this population, and individuals with low plasma MBL concentration are more susceptible to HIV infection.

Using the static opaque chamber method, the soil respiration rates (SR) were measured through the continuous experiments in situ in semiarid Stipa grandis steppe in Xilin River Basin of Inner Mongolia, China from June 2001 to June 2003, in parallel, the difference between the SR and the ecosystem respiration rates (TER) were compared. The results indicated that the seasonal variations of the SR and TER were obvious with higher emissions in growing season and a relatively low efflux level in non-growing season, furthermore, the negative effluxes were found in the observation site in winter; the annual CO2 efflux of total ecosystem ranged from 160.5 gC/(m2 x a) to 162.8 gC/(m2 x a) and that of soil ranged from 118.7 gC/(m2 x a) to 152.3 gC/(m2 x a). The annual SR accounted for about 74.0% to 93.5% of the annual TER, but the results of Analysis of Variance (ANOVA) indicated that the difference between the annual average TER and SR did not reach the significance level of 0.05. The TER was under similar environmental controls as SR, in growing seasons of drought years, the variations of soil moisture at 0-10 cm and 10-20 cm depth could account for 79.1%-95.6% of the changes of the SR and TER, but in non-growing season, more than 75% of the variations of the SR and TER could be explained by the changes of the ground temperature of soil surface layers.
