Variation in population sex ratio is particularly pronounced in gynodioecious angiosperms. Extremely high female frequencies in gynodioecious populations cannot be readily explained by selective forces alone. To assess the contributions of drift and gene flow to extreme sex-ratio variation, we documented sex ratio and population size in 92 populations of Lobelia siphilitica across its range and genotyped plants using plastid and nuclear genetic markers. Similarity in spatial patterns of genetic and demographic variables may suggest that drift and/or gene flow have contributed to population sex-ratio variation in L. siphilitica. We found strong spatial structuring of extremely high female frequencies: populations with >50% female plants are restricted to the south–central portion of the range. However, we did not detect any spatial structuring in population size nor metrics of genetic diversity, suggesting that extreme variation in female frequency is not strongly affected by drift or gene flow. Extreme sex-ratio variation is frequently observed in gynodioecious plants, but its causes are difficult to identify. Long‐distance dispersal (LDD) is an essential event for species colonization and expansion in oceanic island ecosystems. Endozoochory by birds is an important factor promoting the LDD of plant seeds, but its contribution to interisland seed dispersal is still unclear. Here, we show possible seed dispersal by a seed predator pigeon, the Japanese wood pigeon Columba janthina, among oceanic islands in the Izu archipelago, Japan. Although some previous studies showed that most seeds swallowed by this pigeon are crushed, intact seeds were found in 44.5% of pigeon feces, indicating the contribution of these birds to seed dispersal. Seasonal population fluctuations and movements by pigeons between neighbouring islands which are 4 km apart by pigeons can promote interisland seed dispersal. We found seeds from fruiting trees not located on the islands where the pigeon feces were collected, which could have been transported from another island. Although many tree species overlap their fruiting phenology with pigeon movement between islands, the seeds of only six species were dispersed by pigeons, and most of the dispersed seeds were from species with small seeds less than 1.5 mm in diameter. Fruit consumption and frequent interisland movement of Japanese wood pigeons might homogenize the distribution of plants with small seeds among neighboring islands. In contrast, their intensive seed consumption might disturb the recruitment of plants with large seeds and make their expansion to other islands difficult. Such effects of mutualistic (dispersal) and antagonistic (predation) interactions with highly mobile seed predators may modify plant distributions and gene flow in oceanic archipelagos. Surveys of patterns of genetic variation in natural sympatric and allopatric populations of recently diverged species are necessary to understand the processes driving intra‐ and interspecific diversification. The South American moths Cactoblastis cactorum, Cactoblastis doddi and Cactoblastis bucyrus are specialized in the use of cacti as host plants. These species have partially different geographic ranges and differ in patterns of host plant use. However, there are areas that overlap, particularly, in northwestern Argentina, where they are sympatric. Using a combination of genome‐wide SNPs and mitochondrial data we assessed intra and interspecific genetic variation and investigated the relative roles of geography and host plants on genetic divergence. We also searched for genetic footprints of hybridization between species. We identified three well delimited species and detected signs of hybridization in the area of sympatry. Our results supported a hypothetical scenario of allopatric speciation in the generalist C. cactorum and genetic interchange during secondary geographic contact with the pair of specialists C. bucyrus and C. doddi that probably speciated sympatrically. In both cases, adaptation to new host plants probably played an important role in speciation. The codling moth (Cydia pomonella (L.)) is an invasive pest of pome fruits introduced to the Americas in the 19–20th centuries. This pest is widespread on both sides of the Andes range separating Argentina and Chile. We performed an analysis of the population genetic variability and structure of C. pomonella in Argentina and Chile using 13 microsatellite markers and sampled C. pomonella from apple as the main host plant along its distribution area (approx. 1,800 km). A total of 22 locations (11 from Chile and 11 from Argentina) were sampled. Significant genetic differentiation was observed among samples from Argentina and Chile (FSC = 0.045) and between all localities (FST = 0.085). Significant isolation by distance (IBD) was found for each country and when samples from both sides of the Andes range were pooled, although a lower correlation coefficient was observed. The Mantel test showed that the geographic distance and highest altitude of the mountains between locations were significantly associated with the pairwise FST when samples from both sides of the Andes range were pooled. According to a Bayesian assignment test (STRUCTURE), samples from Argentina and Chile conformed to two distinct genetic clusters. Our results also suggest that the recent invasion of C. pomonella in the southernmost localities (Aysén Region in Chile and Santa Cruz Province in Argentina) originated in populations from the respective sides of the Andes range. Our results indicate a genetic exchange of C. pomonella within each country and significant genetic differentiation between countries, which could be explained by dispersal mediated by human activities related to fruit production within each country with little exchange between them. A possible explanation is that the Andes range could be a significant barrier for dispersal by flight, and quarantine barriers could prevent the movement of plant material or infested fruit between countries. The spatial distribution of plants is constrained by demographic and ecogeographic factors that determine the range and abundance of the species. Wild grapevine (Vitis vinifera ssp. sylvestris) is distributed from Switzerland in the north to Israel in the south. However, little is known about the ecogeographic constraints of this species and its genetic and phenotypic characteristics, especially at the southern edge of its distribution range in the Levant region. In this study, we explore the population structure of southern Levantine wild grapevines and the correlation between demographic and ecogeographic characteristics. Based on our genetic analysis, the wild grapevine populations in this region can be divided into two major subgroups in accordance with a multivariate spatial and ecogeographical clustering model. The identified subpopulations also differ in morphological traits, mainly leaf hairiness which may imply adaptation to environmental stress. The findings suggest that the Upper Jordan River population was spread to the Sea of Galilee area and that a third smaller subpopulation at the south of the Golan Heights may represent a distinguished gene pool or a recent establishment of a new population. A spatial distribution model indicated that distance to water sources, Normalized difference vegetation index, and precipitation are the main environmental factors constraining V. v. sylvestris distribution at its southern distribution range. Pre-commercialization studies on environmental biosafety of genetically modified (GM) crops are necessary to evaluate the potential for sexual hybridization with related plant species that occur in the release area. The aim of the study was a preliminary assessment of factors that may contribute to gene flow from sugarcane (Saccharum hybrids) to indigenous relatives in the sugarcane production regions of Mpumalanga and KwaZulu-Natal provinces, South Africa. In the first instance, an assessment of Saccharum wild relatives was conducted based on existing phylogenies and literature surveys. The prevalence, spatial overlap, proximity, distribution potential, and flowering times of wild relatives in sugarcane production regions based on the above, and on herbaria records and field surveys were conducted for Imperata, Sorghum, Cleistachne, and Miscanthidium species. Eleven species were selected for spatial analyses based on their presence within the sugarcane cultivation region: four species in the Saccharinae and seven in the Sorghinae. Secondly, fragments of the nuclear internal transcribed spacer (ITS) regions of the 5.8s ribosomal gene and two chloroplast genes, ribulose-bisphosphate carboxylase (rbcL), and maturase K (matK) were sequenced or assembled from short read data to confirm relatedness between Saccharum hybrids and its wild relatives. Phylogenetic analyses of the ITS cassette showed that the closest wild relative species to commercial sugarcane were Miscanthidium capense, Miscanthidium junceum, and Narenga porphyrocoma. Sorghum was found to be more distantly related to Saccharum than previously described. Based on the phylogeny described in our study, the only species to highlight in terms of evolutionary divergence times from Saccharum are those within the genus Miscanthidium, most especially M. capense, and M. junceum which are only 3 million years divergent from Saccharum. Field assessment of pollen viability of 13 commercial sugarcane cultivars using two stains, iodine potassium iodide (IKI) and triphenyl tetrazolium chloride, showed decreasing pollen viability (from 85 to 0%) from the north to the south eastern regions of the study area. Future work will include other aspects influencing gene flow such as cytological compatibility and introgression between sugarcane and Miscanthidium species.

Oaks are important timber trees with wide distributions in China, but few genetic studies have been conducted on a fine scale. In this study, we seek to investigate the genetic diversity and differentiation of three sympatric oak species (Quercus aliena Blume, Quercus dentata Thunb. ex Murray, and Quercus variabilis Blume) in their northern distribution in China using 17 bi-parentally inherited nSSRs markers and five maternally inherited chloroplast DNA (cpDNA) fragments. Both the cpDNA and the nSSRs show a high level of genetic differentiation between different oak sections. The chloroplast haplotypes are clustered into two lineages. Clear species boundaries are detected between Q. variabilis and either Q. aliena or Q. dentata. The sharing of chloroplast haplotype H1 between Q. aliena and Q. dentata suggests very recent speciation and incomplete lineage sorting or introgression of H1 fromone species to another. The nSSRs data indicate a complete fixation of variation within sites for all three oak species, and that extensive gene flow occurs within species whereas only limited gene flow is detected between Q. aliena and Q. dentata and nearly no gene flow can be detected between Q. aliena and Q. variabilis and between Q. dentata and Q. variabilis. Aim The hypotheses proposed to explain the high percentage of bipolar lichens in Antarctica have never been explicitly tested. We used the strictly bipolar, coastal lichenized fungus Mastodia tessellata ( Verrucariaceae, Ascomycota) and its photobionts ( Prasiola, Trebouxiophyceae, Chlorophyta) as model species to discern whether this extraordinary disjunction originated from vicariance or long-distance dispersal. Location Coasts of Antarctica, Tierra del Fuego (Chile), Alaska ( USA) and British Columbia (Canada). Methods Based on a comprehensive geographical (315 specimens and 16 populations from Antarctica, Tierra del Fuego and North America) and molecular sampling (three and four loci for the fungus and algae respectively), we implemented explicit Bayesian methods to compare alternative hypotheses of speciation and migration, and performed dating analyses for the fungal and algal partner, in order to infer the timing of the colonization events and the direction of gene flow among distant, disjunct areas. Results Mastodia tessellata comprises two fungal species which in turn associate with three photobiont lineages along the studied distribution range. Independent estimation of divergence ages for myco- and photobionts indicated a middle to latest Miocene species split in the Southern Hemisphere, and a late Miocene to Pleistocene acquisition of the bipolar distribution. Comparison of migration models and genetic diversity patterns suggested an austral origin for the bipolar species. Main conclusions The complex evolutionary history of Mastodia tessellata s.l. can be explained by a combination of vicariant and long-distance dispersal mechanisms. We provide novel evidence of a pre-Pleistocene long-term evolution of lichens in Antarctica as well as for bipolar distributions shaped by Southern to Northern Hemisphere migratory routes without the need for stepping stones. Aim We examine the influence of fluctuating sea levels in a land-bridge archipelago on the apportioning of intraspecific genetic diversity and divergence in the widespread Puerto Rican crested anole ( Anolis cristatellus). We compare three alternative scenarios for genetic diversification in an archipelagic species that contrast the relative influences of periodic isolation versus island connectedness driven by fluctuating sea levels. Our approach combines information from geography and population genetics to assess the influence of island size, island isolation, island historical geography, and population genetic processes such as drift on the contemporary distribution of genetic variation within and among islands. Location The Puerto Rico Bank in the Caribbean focusing primarily on the Spanish, British and U.S. Virgin Islands. Methods We used nuclear and mitochondrial DNA sequences and microsatellite genotypes sampled from A. cristatellus populations to investigate: (1) the broad-scale pattern of phylogeographical divergence across Puerto Rico Bank islands and (2) diversification within the Virgin Islands archipelago. For the first component, we used sequence data to reconstruct the relationships among 542 samples from across the species range. For the second component, we examined the relative influences of island size, isolation, and population genetic processes on the distribution of genetic diversity across the Virgin Islands. Results In the Virgin Islands, A. cristatellus is represented by a monophyletic clade except on the island of Vieques, where two divergent clades coexist. We found evidence for non-equilibrium dynamics in the Virgin Islands, suggesting spatial population expansion during intraglacial periods of low sea level. Main conclusions We found limited evidence that periods of island isolation affected patterns of genetic diversity and differentiation. Instead, we found that the patterns of genetic diversity and divergence in A. cristatellus in the Virgin Islands archipelago are likely shaped by long-term persistence in the region and periods of population spatial expansion. Machilus pauhoi Kanehira is an important timber species in China. A provenance trial was recently set up to evaluate the growth performance of trees from different localities, with the aim of designing seed transfer guidelines. Here, we tested twelve nuclear microsatellite markers derived from other species of the Lauraceae family and investigated population genetic structure in M. pauhoi. Both the number of observed alleles per locus (Na) and the polymorphic information content (PIC) significantly decreased against the latitude, but showed an insignificant decrease against the longitude. Heterozygosity (Ho) and gene diversity (h) exhibited a weak correlation with geographic location. Private alleles were present in multiple populations, and a moderate level of population genetic differentiation was detected (Gst = 0.1691). The joint pattern of genetic diversity (Na, PIC, Ho, and h) suggests that general northeastward dispersal led to the current distribution of M. pauhoi. Significant but weak effects of isolation-by-distance (IBD) occurred, implicating the mountain ranges as the major barrier to gene flow. Both STRUCTURE and hierarchical clustering analyses showed three distinct groups of populations related to the physical connectivity among mountain ranges. A priority in designing genetic conservation should be given to the populations at the southwest side of the species’ distribution. This conservation strategy can also be combined with the pattern of adaptive genetic variation from the provenance trial for comprehensive genetic resource management of native M. pauhoi. The influence of geographic range on species persistence has long been of interest and there is a need for a better understanding of the genetic consequences for species with restricted distributions, particularly with the increasing rate of global species extinctions. However, the genetic effects of restricted range are often confounded by the impacts of population distribution. We compared chloroplast and nuclear genetic diversity and differentiation in two acacias, the restricted, patchily distributed Acacia atkinsiana and the widespread, semi-continuously distributed A. ancistrocarpa. Lower intra-population diversity and higher differentiation between populations were seen in A. atkinsiana compared to its widespread congener, A. ancistrocarpa. There was little evidence of geographical influences on population genetic structure in A. ancistrocarpa whereas A. atkinsiana exhibited nuclear genetic structure with isolation by distance, differentiation of near-coastal populations from those in the ranges, and differentiation of peripheral populations from those in the centre of the distribution. These results are consistent with expectations of the effect of geographic range and population distribution on genetic diversity, but indicate that distribution of populations rather than geographic range has influenced the observed genetic structure. The contrasting patterns observed here demonstrate that conservation approaches for species management and ecological restoration need to consider the distribution of populations in geographically restricted species. Background: Habitat fragmentation, water resources and biological characteristics are important factors that shape the genetic structure and geographical distribution of desert plants. Analysis of the relationships between these factors and population genetic variation should help to determine the evolutionary potential and conservation strategies for genetic resources for desert plant populations. As a traditional Chinese herb, Glycyrrhiza inflata B. (Fabaceae) is restricted to the fragmented desert habitat in China and has undergone a dramatic decline due to long-term over-excavation. Determining the genetic structure of the G. inflata population and identifying a core collection could help with the development of strategies to conserve this species. Results: We investigated the genetic variation of 25 G. inflata populations based on microsatellite markers. A high level of population genetic divergence (FST = 0.257), population bottlenecks, reduced gene flow and moderate genetic variation (HE = 0.383) were detected. The genetic distances between the populations significantly correlated with the geographical distances, and this suggests that habitat fragmentation has driven a special genetic structure of G. inflata in China through isolation by distance. STRUCTURE analysis showed that G. inflata populations were structured into three clusters and that the populations belonged to multiple water systems, which suggests that water resources were related to the genetic structure of G. inflata. In addition, the biological characteristics of the perennial species G. inflata, such as its long-lived seeds, asexual reproduction, and oasis ecology, may be related to its resistance to habitat fragmentation. A core collection of G. inflata, that included 57 accessions was further identified, which captured the main allelic diversity of G. inflata. Conclusions: Recent habitat fragmentation has accelerated genetic divergence. The population genetic structure of G. inflata has been shaped by habitat fragmentation, water resources and biological characteristics. To explore the influence of vicariance on differentiation patterns of taxa in arid regions, we systematically assessed the genetic diversity and variability of Capparis spinosa , a typical xerophyte that is widely distributed in the Tianshan Mountains and adjacent areas. In total, 300 individuals from 25 populations were sampled, and 14 haplotypes were identified using two cpDNA sequences ( rpS 12- rpL 20 and ndh F). A high level of total genetic diversity ( H T = 0.859) was detected, and this was attributed to the extensive distribution range, which included numerous large populations. The SAMOVA results suggested that the 25 populations were clustered into 4 major geographical groups; a similar divergence trend was found by constructing a BEAST phylogenetic tree and a network diagram. The AMOVA results revealed that significant genetic differentiation occurred among groups. Our results indicated a considerable correlation between genetic divergence and geographical distribution. Isolation due to complex mountain and desert geography might limit gene exchange between disjunct populations, resulting in high differentiation between geographical groups. [ABSTRACT FROM AUTHOR]

Spatial genetic structure (SGS) is the non-random distribution of genotypes in space, which is usually correlated with life history traits and is driven by ecological and demographic processes. This study aimed to use spatial statistical analysis methods to assess genetic diversity and spatial genetic structure in three known disjunct localities where Tibouchina papyrus occurs, varying in aggregation and density of individuals and in landscape characteristics. This small tree species is endemic to the 'Cerrado' (savannas) region of Central Brazil and is also restricted to elevation rock fields ('campo rupestre'). Therefore, it can be considered a model species to help guide studies on Cerrado endemic plants that are isolated with geographic distributions restricted to this highland habitat. The analyses were based on the polymorphisms at ten microsatellite loci. Our working hypothesis was that wind-dispersed species, such as T. papyrus, present no or low SGS because of the long distance seed gene flow. The spatial genetic structure was indeed weak for two of the three populations. The intensity and shape of the SGS are related to density and aggregation (defined by Ripley's K statistic of spatial aggregation) of individuals in the three localities. Low SGS occurs in populations with aggregation patterns, corroborating the overall hypothesis, although in this species, this pattern depends on the landscape characteristics of each local population. We examined the genetic diversity, population structure and gene flow in a dominant mangrove tree ( Rhizophora stylosa) at its northern biogeographical limit in Sakishima islands of the Japanese archipelago. Simple sequence repeat (SSR) markers from chloroplast (cpSSR) and nuclear DNA were used to analyze 16 populations recovered from various river basins across the chain of three Sakishima islands-Iriomote, Ishigaki and Miyako. The average number of alleles (1.7-2.7) and observed heterozygosities (0.031-0.216) at nuclear SSR and haploid diversity (0.000-0.489) at cpSSR across the populations suggested low genetic diversity in R. stylosa in Sakishima islands. cpSSR analysis identified two haplotypes, and Bayesian clustering analysis (nuclear SSR) revealed two genetic clusters. Analysis of molecular variance (nuclear SSR) showed significant population differentiations. Pairwise tests consistently revealed significant differentiation between most of the population pairs; however, the degrees of differentiations are generally correspondent to the relative geographical distances as suggested from pairwise F and cpSSR genetic distances. Moreover, Mantel tests showed some signals of correlations between genetic distances (nuclear and chloroplast) and geographical distances. These results suggest that combined contribution of gene flow via pollen and propagule dispersal in R. stylosa mostly occurred between neighboring river basins. The appearances of two cpSSR haplotypes (maternal lineages) as well as two nuclear genetic clusters (putative ancestral lineages) at various river basins support the hypothesis that present-day R. stylosa populations across the Sakishima islands were established from few identical founders; however, significant differentiations among various river basins most likely resulted from the limited gene flow and high inbreeding. Background and Aims Clonal growth is a common feature in flowering plants. As clone size increases, the selfing rate in self-compatible species is likely to increase due to more frequent geitono-pollination events (i.e. pollination among flowers within the same genet). This study investigated the breeding system of the marsh cinquefoil (Comarum palustre) and assessed spatial distribution of clones, clone size and architecture, and their effects on realized outcrossing rates. In addition, pollen dispersal was investigated in two patchy populations. Methods The species' breeding system was investigated under controlled conditions through hand pollinations (self- vs. cross-pollination). Using microsatellite markers, an assessment was made of the realized outcrossing rates and the genetic diversity in four natural populations, the clonal structure in two populations within five 15 × 15 m sampling plots following 0·5 × 0·5 m grids, and the pollen dispersal through paternity assignment tests in those two populations. Key Results Comarum palustre is a self-compatible species but only presents a low rate of spontaneous self-pollination. The occurrence of inbreeding depression was not detected at the seed set stage (δSS = 0·04). Clones were spatially clumped (AC = 0·60–0·80), with intermediate to no intermingling of the ramets (DC = 0·40–1·00). Genet size ranged from one to 171 ramets. Patchy populations had low outcrossing rates (tm = 0·33–0·46). Large clones showed lower outcrossing rates than small clones. Pollen dispersal mainly occurred within patches as only 1–7 % of the pollination events occurred between patches of >25 m separation. Seedling recruitment events were detected. Conclusions Genet size together with distances between patches, through increasing geitono-pollination events, appeared to be important factors influencing realized outcrossing rates. The study also revealed seed flow allowing seedling recruitment, which may contribute to increasing the number of new patches, and potentially further enhance gene flow within populations. We found both continuous genetic differentiation over the majority of its range, and discrete isolated local clusters. The discrete differentiation between two genetic groups in the west and east peripheries is consistent with niche divergence and geographical isolation of these groups. In the central area of the species’ range, population structure was shaped mainly by neutral processes and geography rather than by ecological selection. These results show that geographical and environmental factors together created stronger and more discrete genetic differentiation than isolation by distance alone, and illustrate the importance of ecological factors in forming or maintaining genetic divergence across a complex landscape. Extensive studies of genetic diversity and population structure important for conservation of wild sorghum are yet lacking in Ethiopia, the centre of origin for cultivated sorghum. To assess both genetic diversity and the probability of gene flow between wild and cultivated types, collections of wild Sorghum bicolor were made from regions in Ethiopia where wild and cultivated sorghum coexist. Morphological data were recorded in situ for both quantitative and qualitative characters from 30 populations in five diverse geographical regions and eight agroecologies. High phenotypic diversity was observed among the wild and weedy sorghum populations. The overall standardized Shannon–Weaver diversity index (H′), computed from the frequencies of all qualitative traits, ranged from 0.47 to 0.98 with an average value of 0.76. Moreover, warm semi-arid lowland (SA2) agroecologies, which contain Tigray populations, supported the highest diversity for these traits. Subspecies verticilliflorum and drummondii (the two major subspecies of wild S. bicolor) were observed in diverse habitats throughout northern and central Ethiopia. In some areas, weedy types showed domestication traits including the absence of awns and reduced seed shattering. The existence of morphologically intermediate forms indicates that gene flow between cultivated and wild forms has likely occurred. Deployment of transgenic crop sorghum, therefore, would pose a distinct risk for transgene movement into wild Ethiopian populations. [ABSTRACT FROM AUTHOR]

Simple Summary: Pest management can be greatly enhanced by basic knowledge about pest dispersal patterns in agroecosystems, which for insects often relies on comparisons of genetic variation among populations. The globally invasive sweet potato whitefly Bemisia tabaci is one such pest for which a large body of research has examined patterns of genetic variation. We review this literature to address the question: What spatial scales define B. tabaci populations? These studies are global in coverage and draw from a variety of genetic marker types. We found that genetic differentiation among populations is typically low, and that patterns of genetic diversity suggest that groups of migrating whiteflies from divergent populations are typically being sampled together. Overall, these results suggest that there is high ongoing gene flow over large spatial extents, but recent invasion by most populations could obscure genetic markers' ability to detect geographic isolation. Genome-wide data collected across finer spatial and temporal scales hold great promise to clarify the spatial extent of a B. tabaci population, and could reveal whether insecticide rotations can be tailored to specific commodities or if coordination across commodities and regions linked by B. tabaci gene flow is justified. We find that within-species genetic differentiation among sites is often low, and evidence of population structuring by host plant or geography is rare. Heterozygote deficits prevail among populations, indicating that migrants from divergent populations are frequently sampled together. Overall, these results suggest that there is high ongoing gene flow over large spatial extents. However, genetic homogeneity typical of recently invading populations could obscure power to detect real isolation among populations. Genome-wide data collected systematically across space and time could distinguish signatures of invasion history from those of ongoing gene flow. Characterizing the spatial extent of B. tabaci populations could reveal whether insecticide rotations can be tailored to specific commodities or if coordination across linked commodities and regions is justified.
