9. Knockdown of miR-155 alleviates skin damage in rats with chronic spontaneous urticaria by modulating the JAK/STAT signaling pathway

Abstract

Abstract Objective The aim of this study was to investigate the role and mechanisms of miR-155 in chronic spontaneous urticaria (CSU). Methods The expression level of miR-155 in the skin tissues of patients with CSU and experimental rats were detected by RT-qPCR, followed by the measurement of the histamine release rate in the serum through the histamine release test. Besides, hematoxylin & eosin staining was used to observe the pathological changes of the skin tissues; Corresponding detection kits and flow cytometry to measure the changes of immunoglobulins, inflammatory cytokines and T cell subsets in the serum of rats in each group; and western blot to check the expression level of proteins related to JAK/STAT signaling pathway in the skin tissues. Results Knockdown of miR-155 reduced the number and duration of pruritus, alleviated the skin damage, and decreased the number of eosinophils in CSU rats. Moreover, knockdown of miR-155 elevated the serum levels of IgG and IgM, decreased the levels of IgA and inflammatory cytokines, and reduced the proportion of CD4 + and CD4 + CD25 + T cells, as well as the CD4+/CD8 + ratio in CSU rats. However, Tyr705 intervention could reverse the effects of knockdown of miR-155 on CSU model rats. Furthermore, we found that knockdown of miR-155 significantly reduced the protein expression of IRF-9, as well as the P-JAK2/JAK2 and P-STAT3/STAT3 ratios in the skin tissues of CSU rats. Conclusion Knockdown of miR-155 can alleviate skin damage and inflammatory responses and relieve autoimmunity in CSU rats by inhibiting the JAK/STAT3 signaling pathway.

Abstract

The effect of hot isostatic pressing (HIP) on the microstructure and properties of hot dip aluminum coating cooled in a magnetic field was investigated in this study. In order to improve the microstructure and properties of magnetic dip aluminum coating, hot isostatic pressing technology was used for post-treatment. Initially, a traditional aluminum-impregnated coating was prepared on the surface of titanium alloy TA15, an alternating electromagnetic field was applied during the forming and solidification process of the coating. Finally, the coating was treated with hot isostatic pressing technology. Analyzed three different coatings of the microstructure and element distribution, and tested the microhardness of the coatings at various positions. The test results revealed that the TA15 titanium alloy hot-dip aluminum coatings obtained through the three different processes exhibited a gradient structure. Compared with the traditional hot-dipped aluminum air-cooled coating, when an appropriate intensity of alternating electromagnetic field was applied during the coating solidification process, the outer coating structure was enhanced, the number of holes was reduced, the microstructure density increased, and the number of cracks significantly decreased. The defects of the 800 °C hot isostatic magnetic cold and hot dip aluminum coating were repaired under high temperature and pressure, resulting in a uniform and fine microstructure. The comprehensive properties of the magnetic cold and hot dip aluminum coating on the surface of the titanium alloy were effectively enhanced through hot isostatic pressing.

13. Effect of pressure on microstructure and properties of Ti–Al coating on titanium alloy surface

Abstract

The effect of pressure on the microstructure and properties of Ti–Al coating on titanium alloy surface is studied. Ti–Al coating is prepared on the surface of titanium alloy by pressure-assisted high-temperature thermal diffusion. The manufacturing method is simple and easy to operate. The microstructure and element distribution of the coating are analyzed under different processes, and the composition of the coating is tested. The test results show that the substrate alloy and the coating alloy are not connected when the coating is diffused at 650 °C. The coating diffused at 750 °C without pressure is mostly connected to the substrate, but there are microscopic gaps in some areas. The main components of the coating diffused at 750 °C under 80 Pa pressure are TiAl, Ti3Al, and other intermetallic compounds. The boundary between the substrate and the coating is flat, and there is no obvious micro gap, so the metallurgical connection is fundamentally realized. In addition, the maximum hardness can reach 334.9HV0.2. The thickness of the coating diffused at 750 °C under 160 Pa pressure was greatly reduced, showing a gradient form. The surface quality of the outer coating is poor, the structure density of the binding layer is also poor, and there are no obvious penetrating cracks. The bonding between the coating and the substrate is effectively enhanced by an auxiliary action under certain pressure, and then the microstructure and properties of the coating are improved.

Abstract

Abstract Identification and validation of bioactive small-molecule targets is a significant challenge in drug discovery. In recent years, various in-silico approaches have been proposed to expedite time- and resource-consuming experiments for target detection. Herein, we developed several chemogenomic models for target prediction based on multi-scale information of chemical structures and protein sequences. By combining the information of a compound with multiple protein targets together and putting these compound-target pairs into a well-established model, the scores to indicate whether there are interactions between compounds and targets can be derived, and thus a target prediction task can be completed by sorting the outputted scores. To improve the prediction performance, we constructed several chemogenomic models using multi-scale information of chemical structures and protein sequences, and the ensemble model with the best performance was used as our final model. The model was validated by various strategies and external datasets and the promising target prediction capability of the model, i.e., the fraction of known targets identified in the top-k (1 to 10) list of the potential target candidates suggested by the model, was confirmed. Compared with multiple state-of-art target prediction methods, our model showed equivalent or better predictive ability in terms of the top-k predictions. It is expected that our method can be utilized as a powerful computational tool to narrow down the potential targets for experimental testing. Graphical Abstract
