Oxidative stress is one of the important mechanisms of inner ear cell damage, which can lead to age-related hearing loss (ARHL). LncRNA H19 is significantly downregulated in the cochlea of old mouse, however, the role of H19 in the development of ARHL remains unclear. In this study, we aim to investigate the expression and function of H19 in oxidative stress injury of cochlear hair cells induced by H2O2. RT-qPCR and western blot analysis confirms that HEI-OC1 cells stimulated with H2O2 decreases the expressions of H19 and SIRT1, but increases the expression of miR-653-5p. Overexpression of H19 could increase cell viability, ATP level and mitochondrial membrane potential, but reduce mitochondrial ROS generation and cell apoptosis ratio in H2O2-stimulated HEI-OC1 cells. MiR-653-5p is a target of H19, which can bind to the 3′-UTR of SIRT1. H19 is found to regulate the expression of SIRT1 through miR-653-5p. Further experiments demonstrates that H19 regulates HEI-OC1 cell viability, ATP level, mitochondrial membrane potential, mitochondrial ROS generation, and cell apoptosis ratio via the miR-653-5p/SIRT1 axis. In conclusion, lncRNA H19 inhibits oxidative stress injury of cochlear hair cells via the miR-653-5p/SIRT1 axis.

Allergic rhinitis (AR) is a condition with limited treatment options. This study investigates the potential use of mesenchymal stem cell (MSC) nanovesicles as a novel therapy for AR. Specifically, the study explores the underlying mechanisms of MSC nanovesicle therapy by targeting dendritic cells (DCs). The researchers fabricated DC-targeted P-D2-EVs nanovesicles and characterized their properties. Transcriptomic sequencing and single-cell sequencing analyses were performed to study the impact of P-D2-EVs on AR mice, identifying core genes involved in the treatment. In vitro cell experiments were conducted to validate the effects of P-D2-EVs on DC metabolism, Th2 differentiation, and ILC2 activation. The results showed that P-D2-EVs efficiently targeted DCs. Transcriptomic sequencing analysis revealed differential expression of 948 genes in nasal tissue DCs of mice treated with P-D2-EVs. Single-cell sequencing further revealed that P-D2-EVs had inhibitory effects on DC activation, Th2 differentiation, and ILC2 activation, with Fut1 identified as the core gene. Validation experiments demonstrated that P-D2-EVs improved IL10 metabolism in DCs by downregulating Fut1 expression, thereby suppressing Th2 differentiation and ILC2 activation. Animal experiments confirmed the inhibitory effects of P-D2-EVs and their ability to ameliorate AR symptoms in mice. The study suggests that P-D2-EVs reshape DC metabolism and suppress Th2 differentiation and ILC2 activation through the inhibition of the Fut1/ICAM1/P38 MAPK signaling pathway, providing a potential therapeutic approach for AR. Nasopharyngeal carcinoma (NPC) is a tumor that occurs in the nasopharynx. Although advances in detection and treatment have improved the prognosis of NPC the treatment of advanced NPC remains challenging. Here, we explored the effect of microRNA (miR)-122-5p on erastin-induced ferroptosis in NPC cells and the role of ferroptosis in the development of NPC. The effect of miR-122-5p silencing and overexpression and the effect of citrate synthase on erastin-induced lipid peroxidation in NPC cells was analyzed by measuring the amounts of malondialdehyde, Fe2+, glutathione, and reactive oxygen species and the morphological alterations of mitochondria. The malignant biological behavior of NPC cells was examined by cell counting kit-8, EDU, colony formation, Transwell, and wound healing assays. The effects of miR-122-5p on cell proliferation and migration associated with ferroptosis were examined in vivo in a mouse model of NPC generated by subcutaneous injection of NPC cells. We found that erastin induced ferroptosis in NPC cells. miR-122-5p overexpression inhibited CS, thereby promoting erastin-induced ferroptosis in NPC cells and decreasing NPC cell proliferation, migration, and invasion. Sulfur (S) fertilization and rhizobium inoculation are important agronomic practices for improving soybean growth and yield. However, their combined effects on yield components and the resultant impacts on yield need further investigation. Our study aimed to verify the combined effects of S fertilizer and rhizobium inoculant on the yield components and seed yield of soybeans, as well as photosynthesis dynamics during the V5–R6 growth stages. A randomized block design incorporating two factors was employed for field experiments on soybean, involving the application of five rates of sulfur (0, 5.0, 9.8, 14.8, and 19.6 kg S ha−1) with rhizobium inoculation or without. A reduction of 50% in mineral fertilizer according to farmer practice (FP) was applied alongside different rates of sulfur for field experiments on soybeans. The findings indicated that the application of sulfur fertilization significantly enhanced soybean yield by increasing the number of grains per plant (NG) and the number of pods per plant (NP). Maximum grain yield was reached under treatment with an S rate of 19.6 kg S ha−1, which increased NG by 39% more than an S rate of 0 kg S ha−1. Correlation analysis indicated that higher photosynthesis was closely associated with increased yield components. Horses were domesticated 5500 years ago, thousands of years later than other domestic animals; however, in this relatively short period, domestic horses have had a great impact on human history by accelerating civilization, revolutionizing warfare and advancing agricultural production. Modern breeding using marker-assisted selection has greatly accelerated breeding progress. Therefore, identification of genetic markers underlying the traits of interest in domestic horses is the basis for the modern breeding system. In this review, we present an overview of genetic mapping studies and genome wide analyses to identify the genomic regions targeted by positive selection for four important aspects of horses, coat color, racing performance, gait and height at withers. The MC1R locus, for example, has been shown to be the main gene responsible for chestnut color, and the MSTN locus has been shown to control the muscle fiber growth in racing breeds. The missense mutation in DMRT3 is the causal mutation for the alternate gaits in horses. Height at withers, a quantitative trait, was mapped to four major loci (3:105547002, 6:81481064, 9:75550059 and 11:232597 32) that can explain 83% of the height variations in domestic horses.

D-glucaric acid is an important bio-based building block of polymers and is a high value-added chemical that can be used in a variety of applications. In the present study, the Udh target genes from Pseudomonas putida and Pseudomonas syringae were used together to construct the expression vector pETDuet-2 × Udh. The transformants of BL21 (DE3) with vector pETDuet-2 × Udh were applied to produce glucaric acid from glucuronic acid. After optimizing the induction conditions, the highest Udh expression was achieved when 0.4 mmol·L−1 isopropyl-β-d–thiogalactoside (IPTG) was added to the cell cultures at an OD600 value of 0.6 followed by culturing at 26 °C for 6 h. The production of glucaric acid substantially reached 5.24 ± 0.015 g·L−1 in fed-batch cultures in a 30 L tank. The high energy density, low cost, and low toxicity of LiNi0.8Co0.1Mn0.1O2 (NCM811) cathodes has led to their large‐scale mass production. However, the poor interfacial stability between NCM811 and organic electrolytes impairs the long‐cycle performance of lithium ions batteries. In this study, carbonized polymer dots (CPDs) are successfully introduced onto the surface of NCM811 (NCM811@CPDs) via a simple physical mixing process. CPDs with rich surface oxygen functional groups form strong covalent bonds with transition metal (TM) ions at the NCM811 surface, distinctly restraining surface structure degradation, and transition metal ion dissolution. Furthermore, CPDs facilitate the formation of a compact and steady cathode electrolyte interface (CEI) layer during electrochemical cycling. As a result, the NCM811@1 wt% CPDs exhibit enhanced cycling performance with a capacity retention of 89.77% after 100 cycles at 0.5 C compared to 55.39% of the bare NCM811. Colloidal semiconductor II–VI metal chalcogenide (ME) magic‐size clusters (MSCs) exhibit either an optical absorption singlet or doublet. In the latter case, a sharp photoluminescence (PL) signal is observed. Whether the PL‐inactive MSCs transform to the PL‐active ones is unknown. We show that PL‐inactive CdS MSC‐322 transforms to PL‐active CdS MSC‐328 and MSC‐373 in the presence of acetic acid (HOAc). MSC‐322 displays a sharp absorption at ≈322 nm, whereas MSC‐328 and MSC‐373 both have broad absorptions respectively around 328 and 373 nm. In a reaction of cadmium myristate and S powder in 1‐octadecene, MSC‐322 develops; with HOAc, MSC‐328 and MSC‐373 are present. We propose that the MSCs evolve from their relatively transparent precursor compounds (PCs). The PC‐322 to PC‐328 quasi‐isomerization involves monomer substitution, while monomer addition occurs for the PC‐328 to PC‐373 transformation. Simple Summary: In order to increase the growth performance and make up for the lack of high-quality roughage, farmers are accustomed to adopting high-concentration diets during the fattening period of ruminants. The long-term consumption of high-concentration diets induces a series of nutritional metabolic diseases. In production practice, the major mutton sheep fattening region (e.g., Jiangsu, China) generally uses diets with a concentrate ratio of up to 80% to 90% during the fattening period, and reports of cases of nutritional metabolic diseases are also rare. This seems to indicate that sheep and goats are more adaptable to high-concentration diets than cattle. Our work shows that high-concentrate diets can significantly promote the digestion of nutrients; the liver enhances the adaptability of goats to high-concentration diets by regulating the expression of genes involved in nutrient metabolism and toxin clearance. Therefore, the liver plays a vital role in the adaptation of ruminants to high-concentration diets. The results here can lay the foundation for the rational application of high-concentration diets in production. The objective of this study was to determine the effect of high-concentrate diets on the blood parameters and liver transcriptome of goats. Eighteen goats were allocated into three dietary treatments: the high level of concentrate (HC) group, the medium level of concentrate (MC) group, and the low level of concentrate (LC) group. The blood parameters and pathological damage of the gastrointestinal tract and liver tissues were measured. In hepatic portal vein blood, HC showed higher LPS, VFAs, and LA; in jugular vein blood, no significant differences in LPS, VFAs, and LA were recorded among groups (p > 0.05). Compared to the LC and MC groups, the HC group showed significantly increased interleukin (IL)-1β, IL-10, TNF-α, and diamine oxidase in jugular vein blood (p < 0.05). Liver transcriptome analysis discovered a total of 1269 differentially expressed genes (DEGs) among the three groups and most of them came from the HC vs. LC group. There were 333 DEGs up-regulated and 608 down-regulated in the HC group compared to the LC group. The gene ontology enrichment analysis showed that these DEGs were mainly focused on the regulation of triacylglycerol catabolism, lipoprotein particle remodeling, and cholesterol transport. The Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed that the liver of the HC group enhanced the metabolism of nutrients such as VFAs through the activation of AMPK and other signaling pathways and enhanced the clearance and detoxification of LPS by activating the toll-like receptor signaling pathway. This article aimed to explore whether the regulation of Th1/Th2 immune responses by FOXD3-AS1 is associated with dendritic cells (DCs) in allergic rhinitis (AR). HE staining was performed to assess the pathological changes in the nasal mucosa; ELISA was performed to measure the levels of Th1/Th2-related cytokines; flow cytometry was performed to analyze Th1/Th2 cells and MHC-II-, CD80-, and CD86-positive DCs; and qRT‒PCR and western blotting were performed to measure mRNA and protein expression levels, respectively. Our data revealed that LV-FOXD3-AS1 improved AR and increased the Th1/Th2 cell ratio in AR model mice. LV-FOXD3-AS1 further inhibited DC maturation both in vivo and in vitro. Moreover, the coculture system of DCs and CD4+ T cells demonstrated that LV-FOXD3-AS1 increased the Th1/Th2 cell ratio by inhibiting the maturation of DCs. In addition, LV-FOXD3-AS1 reduced the level of phosphorylated STAT6 in DCs derived from healthy mice, and STAT6 overexpression eliminated the inhibitory effect of LV-FOXD3-AS1 on the maturation of DCs. In summary, LV-FOXD3-AS1 ameliorated AR by increasing the Th1/Th2 cell ratio by inhibiting DC maturation via the inhibition of STAT6 phosphorylation. Laryngocarcinoma is one of the most frequent malignancies occurring in the head and neck. The roles of spindle- and kinetochore-associated complex 1 (SKA1) in the malignant progression of several cancers have already been discussed. However, the precise significance and action's mechanism of SKA1 in laryngocarcinoma remain largely unknown. In this study, SKA1 was shown to be strongly expressed in laryngocarcinoma tissues and cells, and higher expression of SKA1 was associated with more severe tumor infiltration, larger tumor diameter, higher risk of lymphatic metastasis and later pathological stage. Additionally, loss-of-function assays in vitro suggested that SKA1 depletion caused a reduction in cell proliferation, migration, and colony formation as well as an increase in apoptosis. In animal experiments, tumors generated from AMC-HN-8 cells with SKA1 depletion exhibited declined tumor volume and weight. Similarly, the detection of Ki67 protein in xenograft tumor tissues reflected that knocking down SKA1 curbed tumor growth in vivo. Further exploration on downstream mechanism revealed that after treatment with Pifithrin-α, the suppression in proliferation level caused by SKA1 knockdown was reversed, while the increase of cell apoptosis was withdrawn; at the molecular level, Pifithrin-α treatment caused p-P53 and Bax diminished, while Bcl-2 ameliorated. A compilation of new advances made in the research field of laboratory reaction kinetics in China's Key Development Project for Air Pollution Formation Mechanism and Control Technologies was presented. These advances are grouped into six broad, interrelated categories, including volatile organic compound (VOC) oxidation, secondary organic aerosol (SOA) formation, new particle formation (NPF) and gas-particle partitioning, ozone chemistry, model parameters, and secondary inorganic aerosol (SIA) formation, highlighting the laboratory work done by Chinese researchers. For smog chamber applications, the current knowledge gained from laboratory studies is reviewed, with emphasis on summarizing the oxidation mechanisms of long-chain alkanes, aromatics, alkenes, aldehydes/ketones in the atmosphere, SOA formation from anthropogenic emission sources, and oxidation of aromatics, isoprene, and limonene, as well as SIA formation. For flow tube applications, atmospheric oxidation mechanisms of toluene and methacrolein, SOA formation from limonene oxidation by ozone, gas-particle partitioning of peroxides, and sulfuric acid-water (H 2 SO 4 -H 2 O) binary nucleation, methanesulfonic acid-water (MSA-H 2 O) binary nucleation, and sulfuric acid-ammonia-water (H 2 SO 4 -NH 3 -H 2 O) ternary nucleation are discussed. Droplet removal from solid surfaces is particularly important for heat and mass transfer, corrosion protection, and certain technological requirements in production. In this study, we investigate droplet removal from a solid surface using an electric field. First, a visual platform was established to capture a video of the droplet deforming and jumping motion in an electric field, and a deformed ellipse equation was applied to fit the liquid droplet profiles. Second, the electric charge distribution was obtained, and the electric forces on the droplet surface before and after jumping were calculated. The result indicates that the charge only accumulates on the upper surface of the droplet, mostly at the top point, and the maximum charge of the 7 μl droplet is about 2 × 10−4 μC in this experiment. The forces on the droplet are almost constant and maintain a constant acceleration (greater than 10 m/s2) after leaving the surface. Third, the effects of droplet volume, electric field intensity, and electrode plate distance on droplet jumping were quantitatively studied. The experiments show that the electric field intensity required for droplet jumping is independent of the droplet volume but positive with the distance between the plates, when the distance between plates increases from 10 to 18 mm, the critical jumping electric field intensity increases by 0.1 kV/mm. The droplet acceleration decreases by about 20% with the increase in volume (5–10 μl) but increases with the increase in electric field intensity. The charge increases with the increase in electric field intensity, but the charge–mass ratio decreases by about 30% with the increase in volume (5–10 μl). Finally, the results show that a small volume and plate distance are more favorable to stimulating the droplets jumping under the electric field. [ABSTRACT FROM AUTHOR]

Although crop rotations have been widely shown as an effective approach for improving yield or soil quality in the long term, the relationship between crop rotations and quality traits of crop or biochar-based fertilization is still unclear. To address this, we conducted a long-term field experiment in the Heilongjiang province of China to investigate the effects of crop rotation and biochar-based fertilizer application on the crop yield, soil parameters, crop quality, and agronomic traits in 2014–2020. The effects of rotations on crop production and soil properties were evaluated with the average yield variability during the 7 years of this study. Our results showed that the diversified corn and soybean rotations had a significant positive effect on average crop yield compared with their monocultures. The corn yield was enhanced by 0.6 Mg ha−1 (5.4%) in the corn–soybean–corn (CSC) crop sequence compared with monoculture corn. Similarly, soybean yield was enhanced by 0.21 Mg ha−1 (9.7%) in the soybean–corn–corn (SCC) crop sequence compared with monoculture soybean. However, a negative effect of crop rotations was detected on the protein content of soybean compared with the monoculture soybean, while a positive effect was detected on oil content. Additionally, no differences were detected in crop yield between biochar-based fertilization and mineral fertilization treatments, but a significant positive effect of biochar-based fertilization was observed for any crop on both protein and oil content. A significant effect of crop rotation was found on the percentage of total soil N (TN), available soil N (AN), and available soil K (AK) content. The SSS crop sequence treatment illustrated the highest TN values at 0.18%. The CCC crop sequence treatment increased AN and AK content by 9.1% and 7.8%, respectively, compared with SSS (p < 0.05). We conclude that crop rotations increase crop yield and biochar-based fertilizer application, improving crop quality traits in the long term. Thus, the addition of biochar-based fertilizer could efficiently enhance the yield and quality of crop in the rotation cropping system. Background. lncRNA AFAP1-AS1 has been linked to the pathogenesis of a wide range of tumors. Nevertheless, whether it plays a role in laryngeal carcinoma (LC) remains unclear. Methods. Twenty-nine pairs of LC and related normal tissues were collected for the detection of lncRNA AFAP1-AS1 using qRT-PCR. Correlation of lncRNA AFAP1-AS1 level and clinicopathological characters was assessed by the chi-square test. Impacts of lncRNA AFAP1-AS1 silencing on LC phenotypes were tested in vitro via CCK-8, clone formation, EdU staining, wound healing, flow cytometry, and Transwell assay. Results. Herein, a remarkable elevation of lncRNA AFAP1-AS1 was observed in LC patients. And higher lncRNA AFAP1-AS1 level was correlated to worse clinical pathological characteristics. Moreover, lncRNA AFAP1-AS1 silencing was revealed to repress TU212 malignant phenotypes. Conclusion. Our data suggested that lncRNA AFAP1-AS1 acts as an oncogene of LC in vitro. The development of deep learning provides a new way for solving the colorization problem on the grayscale image. Excellent coding‐based methods appear in the automatic image colorization task, avoiding the unsaturated colour effect problem of previous methods based on the L2 loss function. Traditional neural networks come with high computational costs and a large number of parameters. Considering the limitation of memory and computing resources and aiming at lightweight, a novel grey image automatic colorization network is proposed. The basic idea of coding‐based methods is used, regarding the colorization task as a pixel‐level classification problem, meanwhile redesign and improve the colour encoding and decoding process. This network architecture leverages a lightweight convolution to reduce the computation and combines an efficient attention model to form a residual block as the kernel of the backbone network. Furthermore, an efficient image self‐attention mechanism placed at the end of the network is applied to enhance the ultimate colouring results. The purpose of this study is to propose a methodology that reflects the impact of interest rate risk on firms in supply chain network under bank financing and trade credit and further describe how trade credit improves the impact of interest rate risk on supply chain network through a financial flow equilibrium. A mean-variance framework and a network equilibrium analysis are integrated to provide a modeling framework. The model allows for the investigation of how bank credit financing (BCF) and trade credit financing (TCF) affect the payment strategy and financial flow of interconnected firms in supply chain networks and how they are affected by interest rate risks. The optimal behavior of manufacturers and retailers is described through variational inequality. We construct a supply chain network equilibrium model and derive qualitative properties of the solution and the function that becomes assimilated to the variational inequality problem. Additionally, variational inequality is solved using the modified projection method. This study extends the research on the impact of interest rate risk on the decision in supply chain network of firms. While other studies focus on the game between banks and firms, only a few authors have made attempts to examine the game between one manufacturer and one retailer in supply chain. An effective trade credit strategy is obtained by balancing cash and credit transactions.
