In this study, three acidic polysaccharides from different plant parts of Codonopsis pilosula var. Modesta (Nannf.) L. T. Shen were obtained by ion exchange chromatography and gel filtration chromatography, and the yields of these three polysaccharides were different. According to the preliminary experimental results, the antioxidant activities of the polysaccharides from rhizomes and fibrous roots (CLFP-1) were poor, and was thus not studied further. Due to this the structural features of polysaccharides from roots (CLRP-1) and aerial parts (CLSP-1) were the object for this study and were structurally characterized, and their antioxidant activities were evaluated. As revealed by the results, the molecular weight of CLRP-1and CLSP-1 were 15.9 kDa and 26.4 kDa, respectively. The monosaccharide composition of CLRP-1 was Ara, Rha, Fuc, Xyl, Man, Gal, GlcA, GalA in a ratio of 3.8: 8.4: 1.0: 0.8: 2.4: 7.4: 7.5: 2.0: 66.7, and Ara, Rha, Gal, GalA in a ratio of 5.8: 8.9: 8.0: 77.0 in for CLSP-1. The results of structural elucidation indicated that both CLRP-1 and CLSP-1 were pectic polysaccharides, mainly composed of 1, 4-linked galacturonic acid with long homogalacturonan regions. Arabinogalactan type I and arabinogalactan type II were presented as side chains. The antioxidant assay in IPEC-J2 cells showed that both CLRP-1 and CLSP-1 promoted cell viability and antioxidant activity, which significantly increase the level of total antioxidant capacity and the activity of superoxide dismutase, catalase, and decrease the content of malondialdehyde. Moreover, CLRP-1 and CLSP-1 also showed powerful antioxidant abilities in Caenorhabditis elegans and might regulate the nuclear localization of DAF-16 transcription factor, induced antioxidant enzymes activities, and further reduced reactive oxygen species and malondialdehyde contents to increase the antioxidant ability of Caenorhabditis elegans. Thus, these finding suggest that CLRP-1 and CLSP-1 could be used as potential antioxidants.

207. Effects of dietary L-leucine supplementation on testicular development and semen quality in boars

Sperm and seminal plasma are rich in leucine, and leucine can promote the protein synthesis. This property makes it an interesting amino acid to increase sperm quality of human and livestock spermatogenesis. The goal of this study was to explore the effects of dietary leucine supplementation on testicular development and semen quality in boars from weaning to 10 months of age. 30 pure-bred, weaned Duroc boars (8.0 ± 1.0 kg) were randomly divided into two groups: control group (CON; fed the basal diet) and leucine group (LEU; fed the basal diet supplemented with 1.2% leucine); then, their body weight and testicular volume were recorded every 4 weeks. Testes were collected for histological and genes expression analysis from 150-day-old boars. Semen was collected and analyzed. Amino acids contents of blood plasma, seminal plasma, sperm, and testes were determined. Dietary supplementation with leucine increased the testicular volume and weight of boars, compared with CON. Sperm viability, sperm count per ejaculation, and average curve speed of sperm in leucine-supplemented boars were increased. Furthermore, leucine supplementation increased the blood plasma and seminal plasma leucine concentrations, and enhanced the gene expressions of branch chain amino acid transaminase, protein kinase B, mammalian target of rapamycin (mTOR), and cyclinb1 in the testes. Interestingly, the expressions of the p-mTOR and mTOR proteins in the testes were also upregulated. Thus, dietary leucine supplementation increased leucine absorption and utilization in the testes, promoted testicular development, and improved semen quality of boars, partly through the mTOR signaling pathway.

Abstract In this paper, we investigate a six-dimensional smooth thick braneworld model which contains a compact extra dimension and an infinite large one. The braneworld is generated by a real scalar field with a ϕ 6 potential and the bulk is an asymptotically AdS6 spacetime. The geometry achieves the localization of the free U(1) gauge field, which is a problem in five-dimensional Randall-Sundrum-like models. In addition, we analyze the stability of the braneworld system and the localization of the graviton.

Abstract Background Replacement gilts are typically fed ad libitum, whereas emerging evidence from human and rodent studies has revealed that time-restricted access to food has health benefits. The objective of this study was to investigate the effect of meal frequency on the metabolic status and ovarian follicular development in gilts. Methods A total of 36 gilts (Landrace × Yorkshire) with an age of 150±3 d and a body weight of 77.6±3.8 kg were randomly allocated into one of three groups (n = 12 in each group), and based on the group allocation, the gilts were fed at a frequency of one meal (T1), two meals (T2), or six meals per day (T6) for 14 consecutive weeks. Abstract Background Congenital heart disease is a leading cause of death in newborns and infants. The feasibility of fetal cardiac surgery is linked to extracorporeal circulation (ECC); therefore, cardioplegic solutions need to be effective and long-lasting. Methods Eighteen pregnant sheep were divided into an ECC-only group, St. Thomas’ Hospital cardioplegic solution (STH1) group (STH group), and HTK preservation solution (Custodiol®) group (HTK group). Markers of myocardial injury including troponin I (cTnI), troponin T (cTnT) and creatine kinase myocardial band (CKMB) were measured at specific time points (T1: pre-ECC, T2: 30 min of ECC, T3: 60 min of ECC, T4: 60 min post-ECC, T5: 120 min post-ECC). Myocardial tissue was removed from the fetal sheep at T5, and apoptosis was detected by TUNEL staining. Results Changes in the serum cTnI, cTnT and CKMB concentrations were not significantly different among the three groups before and during the ECC(T1,T2,T3). At 60 min after ECC shutdown(T4), cTnI and cTnT concentrations were significantly higher in the STH group than before the start of ECC. The concentration of cTnI was higher in the STH group than in the HTK and ECC-only groups. The concentration of cTnT was higher in the STH group than in the ECC-only group. At 120 min after ECC shutdown(T5), cTnI and cTnT concentrations were significantly higher in the ECC and HTK groups than before the start of ECC, and CKMB concentration was significantly higher in STH and HTK groups. The concentrations of cTnT, cTnI and CKMB was higher in the STH group than in the HTK and ECC-only groups. The number of TUNEL-positive cells in the HTK and STH groups was higher than in the ECC-only group. The number of TUNEL-positive cells in the STH group was higher than in the HTK group. There was no statistically significant difference among the groups in the heart rate and mean arterial pressure after ECC. Conclusion The HTK preservation solution was significantly better than STH1 in reducing the release of cardiomyocyte injury markers and the number of apoptotic cells in fetal sheep ECC. Fetal sheep receiving ECC-only had an advantage in all indicators, which suggests ECC-only fetal heart surgery is feasible.

Abstract Ankylosing spondylitis (AS) is chronic inflammatory arthritis with a progressive fusion of axial joints. Anti-inflammatory treatments such as anti-TNF-α antibody therapy suppress inflammation but do not effectively halt the progression of spine fusion in AS patients. Here we report that the autoimmune inflammation of AS generates a microenvironment that promotes chondrogenesis in spine ligaments as the process of spine fusion. Chondrocyte differentiation was observed in the ligaments of patients with early-stage AS, and cartilage formation was followed by calcification. Moreover, a large number of giant osteoclasts were found in the inflammatory environment of ligaments and on bony surfaces of calcified cartilage. Resorption activity by these giant osteoclasts generated marrow with high levels of active TGF-β, which induced new bone formation in the ligaments. Notably, no Osterix+ osteoprogenitors were found in osteoclast resorption areas, indicating uncoupled bone resorption and formation. Even at the late and maturation stages, the uncoupled osteoclast resorption in bony interspinous ligament activates TGF-β to induce the progression of ossification in AS patients. Osteoclast resorption of calcified cartilage-initiated ossification in the progression of AS is a similar pathologic process of acquired heterotopic ossification (HO). Our finding of cartilage formation in the ligaments of AS patients revealed that the pathogenesis of spinal fusion is a process of HO and explained why anti-inflammatory treatments do not slow ankylosing once there is new bone formation in spinal soft tissues. Thus, inhibition of HO formation, such as osteoclast activity, cartilage formation, or TGF-β activity could be a potential therapy for AS.

This study was conducted to investigate the effects of dietary multi-enzyme (multi-carbohydrase and phytase complex, MCPC) supplementation on digestibility, growth performance, bone mineralization, and carcass yield and traits in growing–finishing pigs fed diets with adequate or deficient net energy (NE), amino acids (AA), calcium (Ca) and phosphorus (P) levels. A total of 576 crossbred [Duroc × (Landrace × Yorkshire)] barrows (~25 kg) were fed one of the six diets till live weight approached 130 kg. Basal diets included a positive control (PC), negative control 1 (NC1) and 2 (NC2), while another three diets were prepared by adding MCPC to the three basal diets. The final body weight was lower (p < 0.05) in NC2 than in NC1 and PC treatments, while overall feed intake and feed-gain ratio were higher (p < 0.05) in NC1 and NC2 than in PC treatment. The NC2 treatment showed lower (p < 0.05) carcass weight but higher (p < 0.05) lean meat percentage than the PC treatment. The apparent ileal digestibility (AID) of gross energy (GE), crude protein (CP) and AA was decreased (p < 0.05) or tended (p < 0.10) to decrease in NC1 and/or NC2 diets compared with a PC diet. MCPC supplementation improved (p < 0.05) AID of Ca, P and AA (Lys, Leu, Val, Phe, Gly, Tyr and Pro), apparent total-tract digestibility (ATTD) of GE, CP, bone strength, Ca, and P retention. In conclusion, MCPC supplementation improved nutrient digestibility, bone mineralization, and growth performance of fattening pigs, regardless of the nutritional level of the basal diet.

213. Experimental Study of Oxygen Depletion Effects on Soot Morphology and Nanostructure in Coflow Diffusion Aviation Fuel (RP-3) Flames

Oxygen concentration is a significant factor affecting soot formation and oxidation. However, there are few studies that have focused on the morphology and nanostructure characteristics of soot in aviation kerosene, oxygen-depleted combustion flames. In the present paper, five coflow flames under initial oxygen volume concentrations of 18.5%, 19%, 20%, 21%, and 23.5% were studied. The pneumatic probe sampling method and high-resolution transmission electron microscopy (HRTEM) analysis were conducted to quantify the morphology and nanostructure parameters, and laser extinction (LE) was applied to determine the soot volume fraction. Among the cases of different oxidizer oxygen concentrations (23.5% to 18.5%), the change in soot volume fraction was quantified, and the degree of graphitization of soot particles, i.e., the maturity, were compared. The results show that the peak value of soot volume fraction of the flames increased by 0.73 ppm as the oxygen concentration increased from 21% to 23.5%, and decreased by 1.25 ppm as the oxygen concentration decreased from 21% to 18.5%. When the oxygen concentration decreased from 23.5% to 18.5%, the soot primary particle diameter at the same dimensionless height decreased and then increased, which was attributed to the competition between the changes in the residence time and the growth rate of the soot particles. The quantitative analysis results of the soot nanostructure suggested that reduced oxygen concentration inhibited the graphitization process of carbon lattices and decreased the maturity and oxidation resistance of soot. When the oxygen concentration decreased from 23.5% to 18.5% at the same dimensionless height, the mean fringe length decreased by an average of 0.18 nm, and the mean value of fringe tortuosity and spacing increased by an average of 0.053 and 0.035 nm.

Aiming at the problem of shadow interference in UAV's ground reconnaissance, a color and polarization synergistic target detection method is proposed for anti-shadow interference, based on the influence of two physical characteristics (wavelength and polarization) under different illuminations. A valid fusion strategy is proposed via integrating two separate detection results on color and polarization images. Moreover, a local enhancement and recognition module (LER) is introduced to boost the performance. Based on our built dataset, experimental results show that our method achieves mAP of 87.76%, and 12.37% higher than that of color image and 14.68% higher than that of polarization image. The presence of side-lobe noise degrades the image quality and adversely affects the performance of inverse synthetic aperture radar (ISAR) image understanding applications, such as automatic target recognition (ATR), target detection, etc. However, methods reliant on data processing, such as windowing, inevitably encounter resolution reduction, and current deep learning approaches under-appreciate the sparsity inherent in ISAR images. Taking the above analysis into consideration, a convolutional neural network-based process for ISAR side-lobe noise training is proposed in this paper. The proposed processing, based on the ISAR images sparsity characteristic analysis, undergoes enhancements in three core ideas, dataset construction, prior network design, and loss function improvements. In the realm of dataset construction, we introduce a bin-by-bin fast clean algorithm and accelerate the processing speed significantly on the basis of image complete information. Subsequently, a spatial attention layer is incorporated into the prior network designed to augment the network's focus on the crucial regions of ISAR images. In addition, a loss function featuring a weighting factor is devised to ensure the precise recovery of the strong scattering point. Simulation experiments demonstrate that the proposed process achieves significant improvements in both quantitative and qualitative results over the classical denoise methods. [ABSTRACT FROM AUTHOR]

Background: Neuro-inflammation is regarded as one of the critical pathogenesis in neurodegenerative diseases, which is characterized by the activated microglial cells. Pectolinarin (Pec), a natural flavonoid that exists in many Chinese herbal medicines, has been reported to have various biological activities. However, the effects and mechanisms on neuro-inflammation are not clear. Methods: In this study, the inhibitory effects and mechanisms of Pec on neuro-inflammation were investigated in the LPS-stimulated microglial BV2 cells. BV2 microglial cells were treated with Pec or vehicle, followed by LPS. Enzyme-linked immunosorbent assay, real-time quantitative PCR, nitric oxide and reactive oxygen species assay, and western blot were performed to examine the effects of Pec on neuro-inflammatory responses. Results: We showed that Pec significantly inhibited the expression of tumor necrosis factor α and interleukin 6 in mRNA and protein levels induced by LPS. Moreover, the production of nitric oxide, iNOS, reactive oxygen species, and COX-2 were suppressed by Pec in LPS-stimulated microglial BV2 cells. In addition, Pec inhibited LPS-induced inflammation via nuclear factor kappa B signaling pathway, as evidenced by the reduction of the phosphorylation of inhibitor of nuclear factor kappa-B kinase, the degradation of IκBα, and the nuclear translocation of p65. Conclusion: Taken together, Pec exhibited anti-inflammatory effects in LPS-stimulated microglial BV2 cells via nuclear factor kappa B signaling pathway, which might provide therapeutic potential for neuro-inflammation and neurodegenerative diseases. 217. Usefulness of a Simple Preoperative Planning Technique using Plain X‐rays for Direct Anterior Approach for Total Hip Arthroplasty

Objective To examine the accuracy, reliability, and reproducibility of a simple preoperative planning technique using plain X‐rays. Methods A retrospective analysis of 96 consecutive cases of primary direct anterior approach (DAA)‐total hip arthroplasty (THA) from July 2015 to December 2018 was performed. The 96 patients included 24 males and 72 females, with an average age of 70 years. The standard AP pelvis radiographs with the patients' hips extended and internally rotated were obtained pre‐ and postoperatively. The preoperative planning was also completed on the standardized AP pelvic radiographs. The prearranged cup positioning was radiologically measured intraoperatively using fluoroscopy. The correct leg length was assessed intraoperatively, which was compared with the preoperative planning. The component positioning was measured by three independent researchers. Two of the researchers completed the measurements three times, and intra‐observer and inter‐observer reliability were calculated. All patients received at least 6 months follow‐up (6 months–4 years). Results In all cases, the median leg length discrepancy (LLD) was 4.4 mm (range 1.6–15.9 mm), and 84 patients had an LLD smaller than 10 mm, of which 58 patients had an LLD of less than 5 mm. None of the patients had a critical LLD of 2 cm or larger. The multivariable logistic regression for LLD (safe range: yes/no) with the co‐variables including gender, ASA classification, type of cup, the surgeon's experience level, and the presence of a total hip arthroplasty (THA) on the contralateral side did not present statistical significance. The median angle of the inclination of the acetabular component (IA) was 42.3° (range: 28.7°–52.2°). Ninety‐one patients were within the defined safe range. Objective Venous thromboembolism (VTE) is a significant concern post total joint arthroplasty (TJA). However, the optimal prevention method of VTE remains controversial at present. This study aims to evaluate a risk‐stratified VTE prophylaxis protocol for patients undergoing TJA. Methods A total of 891 TJA patients from January 2011 to November 2019 were retrospectively investigated. The study was divided into two cohorts. In cohort 1, 410 patients (250 females and 160 males, mean age 64.32 years) were treated with an aggressive VTE chemoprophylaxis protocol. In cohort 2, 481 patients were treated with a risk‐stratified protocol that utilized low molecular weight heparins (LMWH) and sequential aspirin (ASA) for standard‐risk patients (a total of 288 containing 177 females and 111 males, mean age 65.4 years), and targeted anticoagulation for high‐risk patients (a total of 193 containing 121 females and 72 males, mean age 66.8 years). The patients were followed up at 2–4 weeks for an initial visit and at 6–10 weeks for a subsequent visit after surgery. A chart review of all patient medical records was performed to record the demographics, comorbidities, deep vein thrombosis, pulmonary embolus, superficial infection, deep infection, bleeding complications, and 90‐day readmissions. Results The VTE rate was 1.71% (7/410) in cohort 1 and 1.46% (7/481) in cohort 2 respectively. For cohort 2, the VTE rate was 2.07% (4/193) in high‐risk group and 1.04% (3/288) in standard‐risk group. The readmission rate was 2.44% (10/410) in cohort 1 and 2.08% (10/481) in cohort 2. For cohort 2, the readmission rate was 2.07% (4/193) in high‐risk group and 2.08% (6/288) in standard‐risk group. The reasons for readmission were as follows: infection, 1.3% (5/410) in cohort 1 and 1.3% (6/481) in cohort 2; wound or bleeding complications, 0.48% (2/410) in cohort 1 and 0.2% (1/481) in cohort 2; trauma, 0.2% (1/410) in cohort 1 and 0.2% (1/481) in cohort 2; VTE, 0.2% (1/410) in cohort 1 and 0.2% (1/481) in cohort 2; others, 0.2% (1/410) in cohort 1 and 0.6% (3/481) in cohort 2. There was a decrease in VTE events and readmissions in the risk‐stratified cohort, although this did not reach statistical significance. Abstract. Background:. Aspirin has demonstrated safety and efficacy for venous thromboembolism (VTE) prophylaxis following total hip arthroplasty (THA); however, inconsistent dose regimens have been reported in the literature. This study aimed to evaluate and compare the safety and efficacy of 100 mg aspirin twice daily with rivaroxaban in VTE prophylaxis following THA. Methods:. Patients undergoing elective unilateral primary THA between January 2019 and January 2020 were prospectively enrolled in the study and randomly allocated to receive 5 weeks of VTE prophylaxis with either oral enteric-coated aspirin (100 mg twice daily) or rivaroxaban (10 mg once daily). Medication safety and efficacy were comprehensively evaluated through symptomatic VTE incidence, deep vein thrombosis (DVT) on Doppler ultrasonography, total blood loss (TBL), laboratory bloodwork, Harris hip score (HHS), post-operative recovery, and the incidence of other complications. Results:. We included 70 patients in this study; 34 and 36 were allocated to receive aspirin and rivaroxaban prophylaxis, respectively. No cases of symptomatic VTE occurred in this study. The DVT rate on Doppler ultrasonography in the aspirin group was not significantly different from that in the rivaroxaban group (8.8% vs. 8.3%, χ2 = 0.01, P = 0.91), confirming the non-inferiority of aspirin for DVT prophylaxis (χ2 = 2.29, P = 0.01). The calculated TBL in the aspirin group (944.9 mL [658.5–1137.8 mL]) was similar to that in the rivaroxaban group (978.3 mL [747.4–1740.6mL]) (χ2 = 1.55, P = 0.12). However, there were no significant inter-group differences in HHS at post-operative day (POD) 30 (Aspirin: 81.0 [78.8–83.0], Rivaroxaban: 81.0 [79.3–83.0], χ2 = 0.43, P = 0.67) and POD 90 (Aspirin: 90.0 [89.0–92.0], Rivaroxaban: 91.5 [88.3–92.8], χ2 = 0.77, P = 0.44), the incidence of bleeding events (2.9% vs. 8.3%, χ2 = 0.96, P = 0.33), or gastrointestinal complications (2.9% vs. 5.6%, χ2 = 1.13, P = 0.29). Conclusion:. In terms of safety and efficacy, the prophylactic use of 100 mg aspirin twice daily was not statistically different from that of rivaroxaban in preventing VTE and reducing the risk of blood loss following elective primary THA. This supports the use of aspirin chemoprophylaxis following THA as a less expensive and more widely available option for future THAs. Trial Registration:. The red swamp crayfish, Procambarus clarkii, is a native species in north-eastern Mexico and south-central USA. P. clarkii was introduced to China in 1929 and has been used as an aquaculture species in China since 1983. It currently exists in most of the provinces of China, but threatens local fish, crustaceans, aquatic plants and local freshwater ecosystems. We examined the genetic variation in partial mitochondrial ND2 gene of 831 individuals collected from 25 P. clarkii populations in 13 provinces of China to infer the expansion pathways and mechanisms. Six haplotypes were detected. All six haplotypes appeared in four populations in Nanjing and a population located near Nanjing whereas only 1–5 of the six haplotypes were present in other populations. These data suggest that the populations in Nanjing are probably the source of all other populations in China. There were no significant relationships between geographic distances and genetic distances in 25 populations, whereas significant relationship was found in four populations in Qinhuai River covering 50 km in Nanjing. These data suggest that the expansion mainly be human-mediated in large scale, and active disposal or non-anthropogenic passive dispersal might have played an important role in expansion at a smaller scale. In some places far away from Nanjing, several haplotypes existed, suggested multiple introduction events may have happened. Although aquaculture of this species could bring huge economic benefit, its potential to negatively affect native biota and entire ecosystems should not be ignored.

In this paper, a wide-angle frequency-scanning array antenna is presented for three-dimensional (3D) radar. Initially, a modified composite right/left-handed (CRLH) unit cell based on a dual-layer finger-connected interdigital capacitor (FCIDC) is analyzed. Compared with its conventional counterpart, the proposed unit cell eliminates the undesired high-order resonances and has a lower right/left-handed passband. Then, a broadband frequency-scanning feeding network, which has a large scanning angle, is designed using the proposed CRLH unit cell. The responses of the four output ports indicate that the network can offer desired amplitude and phase difference to scan towards both negative and positive elevation angles. To validate its performance, a tightly coupled planar quasi-Yagi antenna array is fed to synthesize an array antenna. Functionally graded materials (FGMs) used in electrical insulation have spatially inhomogeneous dielectric properties (i.e., permittivity or conductivity), which can be applied to relieve localized electric field (E-field) intensification and improve insulation performance. However, previous research shows some limitations in inadequate considerations on the material feasibility, and insufficient universality on material grading type and voltage form. In this study, a material study of silicone rubber (SiR) nanocomposites containing carbon nanotubes (CNTs) is conducted to determine the practical variation range of dielectric properties (both permittivity and conductivity) in joint insulation materials. Then, the optimal design of both multilayer and pointwise FGM joint insulation is investigated under both AC and DC voltage, in which the lower and upper limits of permittivity and conductivity were derived from the experimental results. Experimental results on SiR/CNT nanocomposites indicate that low-amount doping of the CNTs (0-0.5% wt%) can effectively increase the permittivity and conductivity of joint insulation materials. The successive simulation study indicates that compared to uniform joints, cable joints employing optimally designed FGM insulation show a considerable increase in the E-field utilization factor (from 0.08-0.17 to 0.23-0.56), indicating elevated E-field uniformity. Finally, the optimal range of CNT doping ratios is determined to be 0-0.3 wt% from both experimental and simulation results. This study systematically verifies the applicability of FGMs in enhancing the performance of power cable accessories, which can show some guidance to the design and fabrication of advanced power cable systems.

Various studies have evaluated the relationship between cholecalciferol (vitamin D3) and reproductive performance. This study aimed at investigating the effects of maternal D3 supplementation during gestation on reproductive performance and antioxidant capacities of gilts and their offsprings. Twenty-three Landrace × Yorkshire gilts were randomly allocated into two groups and fed on one of the following two diets during gestation: control diet or D3 supplemented diet. It was found that D3 supplementation had a tendency to increase the number of total born and born alive piglets. Moreover, it elevated serum 25-hydroxycholecalciferol concentrations in gilts and newborn piglets. Besides, D3 supplementation improved the activities of antioxidant enzymes (GSH-Px, T-AOC and T-SOD) in the blood of gilts, umbilical cord and newborn piglets and piglets’ liver, while serum malondialdehyde concentrations of gilts and umbilical cord were reduced in the D3 group. In addition, D3 supplementation upregulated the expression of antioxidant related genes in the placenta and piglets’ liver. In conclusion, D3 supplementation during gestation might improve antioxidant capacities in gilts, placenta and in newborn piglets. Therefore, D3 supplementation has the potential to enhance the reproductive performance of gilts.HIGHLIGHTS D3 supplementation has a significant role in the nutrition of gilts. Maternal D3 supplementation during gestation increased the number of born and born alive piglets. Maternal D3 supplementation during gestation improved the antioxidant capacities of gilts, placenta and newborn piglets.

Abstract Background Previous evidence suggested that perioperative anti-rheumatic therapy for patients receiving total knee arthroplasty (TKA) helped improve postoperative rehabilitation for rheumatoid arthritis (RA), yet long-term effects and outcomes of perioperative drug therapy in TKA presently remain unclear. This study investigated whether perioperative treatment with glucocorticoids (GC) and disease-modifying anti-rheumatic drugs (DMARDs) can improve clinical outcomes for patients with RA undergoing TKA. Methods Patients between January 2000 and December 2011 were allocated into three groups based on perioperative drug therapy: A, control group (no GC or DMARDs), B, DMARD group (DMARDs given without GC), and C, co-therapy group (DMARDs plus GC). The patients were followed up for average 11.4 years. Baseline characteristics, pre- and post-operative Hospital for Special Surgery score (HSS), laboratory parameters, and complications were recorded by follow-up. Results Fifty-six RA patients undergoing 91 TKAs were included in this study. Patients who received perioperative GC with DMARDs (group C) achieved larger/increased range of motion (ROM) (C:122.17 vs A:108.31 vs B:108.07, p = 0.001, partial eta squared (η 2 p) = 0.18) at 1 year, better HSS score (C, 83.01 vs A, 79.23 vs B, 77.35, p = 0.049, η 2 p = 0.067), pain relief (C, 1.09 vs A, 1.17 vs B, 1.75, p = 0.02, η 2 p = 0.094), and ROM (C, 130.81 vs A, 112.82 vs B, 113.58, p = 0.001, η 2 p = 0.142) at latest follow-up comparing with the other treatment groups. No differences were noted in laboratory tests, blood loss, volume of transfusion, or complications among groups. Conclusions Compared with the other perioperative anti-rheumatic treatments, the combination of GC and DMARDs results in improved HSS score, better function, larger range of motion, and reduced postoperative pain for TKA patients with RA in the long term. Further investigation is warranted to look for a better understanding of more specific medication effects and strike a good balance between the benefits and complications for long-term pharmacotherapy.

Endoplasmic reticulum (ER) stress, which can be induced by reactive oxygen species (ROS) and multiple factors, is associated with numerous intestinal diseases. The organic selenium source 2-hydroxy-4-methylselenobutanoic acid (HMSeBA), has been proved to decrease intestinal inflammation and autophagy by improving the expression of selenoproteins. However, it remains unclear whether HMSeBA could alleviate intestinal ER stress by decreasing excessive production of ROS products. This study was conducted to investigate the effect of maternal HMSeBA supplementation on the regulation of intestinal ER stress of their offspring and the regulatory mechanism. Sows were supplemented with HMSeBA during gestation and jejunal epithelial (IPEC-J2) cells were treatment with HMSeBA. Results showed that maternal HMSeBA supplementation significantly upregulated mRNA level of selenoprotein S (SELS) in the jejunum of newborn and weaned piglets compared with the control group, while decreased the gene expression and protein abundance of ER stress markers in the jejunum of LPS challenged weaned piglets. In addition, HMSeBA treatment significantly increased the expression of glutathione peroxidase 4 (GPX4) and SELS, while decreased ROS level and the expression of ER stress markers induced by hydrogen peroxide (H2O2) in IPEC-J2 cells. Furthermore, knockdown of GPX4 did not enhance the ERS signal induced by H2O2, but the lack of GPX4 would cause further deterioration of ER stress signal in the absence of SELS. In conclusion, maternal HMSeBA supplementation might alleviate ROS induced intestinal ER stress by improving the expression of SELS and GPX4 in their offspring. Thus, maternal HMSeBA supplementation might be benefit for the intestinal health of their offspring.

Selenium (Se) is assumed to promote the follicle development by attenuating oxidative stress. The current study was developed to evaluate the effects of dietary 2-hydroxy-4-methylselenobutanoic acid (HMSeBA) supplementation on the follicle development in vivo and on the function of ovarian granulosa cells (GCs) in vitro. Thirty-six gilts were randomly assigned to fed control diet (CON), Na2SeO3 diet (0.3 mg Se/kg) or HMSeBA diet (0.3 mg Se/kg). The results showed that HMSeBA and Na2SeO3 supplementation both increased the total selenium content in liver and serum compared with control, while HMSeBA increased the total selenium content in liver compared with Na2SeO3 group. HMSeBA tended to increase the total selenium content in ovary compared with control. HMSeBA and Na2SeO3 supplementation both increased the weight of uteri in gilts at the third estrus. Moreover, HMSeBA supplementation down-regulated the gene expression of growth differentiation factor-9 (GDF-9) and bone morpho-genetic protein-15 (BMP-15) in cumulus-oocyte complexes (COCs). HMSeBA supplementation decreased malondialdehyde (MDA) content in serum, liver and ovary, increased activity of T-AOC in liver, TXNRD in ovary and GPX in serum, liver and ovary, while up-regulated the liver GPX2, SOD1 and TXNRD1, ovarian GPX1 gene expression. In vitro, HMSeBA treatment promoted GCs' proliferation and secretion of estradiol (E2). HMSeBA treatment increased the activity of T-AOC, T-SOD, GPX, TXNRD and decreased MDA content in GCs in vitro. Meanwhile, HMSeBA treatment up-regulated SOD2 and GPX1 gene expression in GCs in vitro. In conclusion, HMSeBA supplementation is more conducive to promoting follicle development by antioxidant pathway.

The experiment was conducted to investigate the effects of dietary fiber (DF) supplementation in gestation diet on fetal growth and placental development and function and explore the possible mechanism of DF improving sow reproductive performance. A total of 16 Large White × Landrace crossbred gilts were randomly allotted to two groups and fed a semi-purified basal diet [non-fiber (NF) group, 0.1% total DF] or a basal diet supplemented with 8.33 g/kg inulin and 200 g/kg cellulose [Fiber (F) group] during the gestation period. On day 106 of gestation, five sows per group were chosen and slaughtered for sample collection. Results showed that DF supplementation during gestation increased the total fetal weight and placental weight on day 106 of gestation; elevated serum serotonin concentration; increased concentrations of serotonin and short-chain fatty acids (acetate, propionate, and butyrate), as well as tryptophan hydroxylase 1 expression, in colon; elevated serotonin and progesterone concentrations and up-regulated the serotonin transporter, cytochrome P450 11A1, and insulin-like growth factor 2 expressions in the placenta. Besides, the sows in the F group had microbial community structures distinct from those in the NF group. Supplementation of DF in gestation diet increased the Coprococcus 3 abundance that was positively correlated with colonic serotonin concentration, while significantly decreasing the Family XIII AD3011 group abundance which was negatively correlated with colonic serotonin concentration. Above all, DF supplementation in the gestation diet could increase placental serotonin levels by promoting maternal serotonin synthesis in the colon and the transport from the mother to the placenta in sows, and then improve placental development and function, finally promoting fetal growth. Our findings provided insight into the mechanisms of DF improving sow reproductive performance.

Although fiber-rich diets have been positively associated with sperm quality, there have not been any studies that have examined the effects of dietary fiber and its metabolites on sperm quality in young or pre-pubescent animals. In this study, we aimed to explore the effect of dietary fiber supplementation on semen quality and the underlying mechanisms in a boar model. Sixty purebred Yorkshire weaning boars were randomly divided into the four groups (T1–T4). Groups T1, T2, and T3 boars were fed diets with different levels of fiber until reaching 160 days of age and were then fed the same diet, while group T4 boars were fed a basal diet supplemented with butyrate and probiotics. Compared with T1 boars, sperm motility and effective sperm number were significantly higher among T3 boars. Meanwhile, at 240 days of age, the acetic acid and total short-chain fatty acid (SCFA) contents in the sera of T3 and T4 boars were significantly higher than those in T1 boars. The abundance of microbiota in T2 and T3 boars was significantly higher than that in T1 boars (P < 0.01). Moreover, dietary fiber supplementation increased “beneficial gut microbes” such as UCG-005, Rumenococcus, Rikenellaceae_RC9_gut_group and Lactobacillus and decreased the relative abundance of “harmful microbes” such as Clostridium_sensu_stricto_1, Romboutsia and Turicibacter. Collectively, the findings of this study indicate that dietary fiber supplementation improves gut microbiota and promotes SCFA production, thereby enhancing spermatogenesis and semen quality. Moreover, the effects of dietary fiber are superior to those of derived metabolites.

Dietary yeast culture supplementation can contribute to the performance and health of sows and piglets, but few studies have focused on the relationships between the effects of yeast culture and gut microbiota. This study investigated the effect of yeast culture (Saccharomyces cerevisiae) supplementation from late gestation to weaning on the reproductive performance of lactating sows and their faecal microbiota. One hundred and six purebred Landrace sows, of parities two to six were selected and randomly assigned to a control (CON) and yeast culture supplementation (YC) groups based on parity and back fat thickness. The YC sows were individually fed with yeast culture at a dose of 24 g/d from day 90 of gestation to parturition and 40 g/d during lactational period. Blood samples were collected from sows on d 110 of gestation and at weaning at day 21 of lactation for plasma hormone and immunoglobulin analysis. Colostrum and milk on day 20 of lactation were collected for composition analysis. Faecal samples from sows on day 110 of gestation and day 20 of lactation were collected for short-chain fatty acid and faecal microbial analysis. Results showed that the farrowing performance of YC sows did not differ significantly from the CON group (P > 0.05). The average daily feed intake by the YC group during the lactation period was significantly increased by 9.98% (P = 0.004), the weaning-to-oestrus interval was shortened by 0.96 d (P = 0.046) and average daily weight gain of piglets increased by 7.14% (P = 0.036) compared with the CON group. How to evaluate the health status of OPGW optical cable according to different parameters and repair or replace it before failure is the main problem. In this paper, a health evaluation method of OPGW optical cable based on subjective and objective weight assignment method and Principal Component Analysis (PCA) method is proposed with detailed case analysis. The data analysis results show that this method can effectively reflect the health status of OPGW optical cable.

234. Dietary Fiber Supplementation in Replacement Gilts Improves the Reproductive Performance From the Second to Fifth Parities

This study examined the effects of soluble fiber (SF) supplementation (0.8%), containing 17.4% rhamnose, 4.1% fucose, 11.1% arabinose, 30.6% xylose, and 16.4% galactose during the prepubescent phase on the subsequent performance from the second to fifth parities. After the first parity, 56 and 55 post-weaning sows in the control (CON) and SF groups had their reproductive performance monitored in succeeding parities. Circulating concentrations of anti-mullerian hormone (AMH) were greater in the SF group than in the CON group at 205 d of age and the first post-weaning day (p < 0.05). The SF treatment at the prepubescent phase resulted in an enhanced reproductive performance from parities three to five. In Parity three, the SF sows had 1.32 total born (p = 0.044), 1.43 born alive (p = 0.023) and 1.40 born effective, which was significantly more than in the CON group (p = 0.022). In Parity four, the SF sows had 1.1 total born (p = 0.058), 1.28 born alive (p = 0.019), and 1.06 born effective, significantly more than in the CON group (p = 0.049). In Parity five, the SF gilts had 1.43 total born (p = 0.075), 1.53 born alive (p = 0.067) and 1.65 born effective, significantly more than in the CON group (p = 0.020). No effects were observed for the removal of sows and backfat thickness at the mating in each parity between groups (p > 0.05). Collectively, gilts that received an extra 0.8% SF during the prepubescent phase increased their subsequent litter size as breeding sows. These results showed that nutritional decisions at the replacement phase could influence lifetime fertility.

Hepatocellular carcinoma (HCC) is a high mortality liver cancer. The existing treatments (transplantation, chemotherapy, and individualized treatment) with limitations. However, drug combination provides a viable option for hepatocellular carcinoma treatment. A Chinese patent medicine, ShuangDan Capsules (SDC), has been clinically prescribed to hepatocellular carcinoma patients as adjuvant therapy and has shown good antitumor activity. The purpose of this study was to investigate whether SDC could improve the anti-cancer effect and mitigate adverse reactions of sorafenib on HCC in vivo. Magnetic resonance imaging (MRI), immunohistochemistry, and western blot were executed to reveal the potential mechanisms of the combination of SDC and sorafenib on HCC. Tumors appeared hyperintense on T2 sequence images relative to the adjacent normal liver in MRI. Combination of SDC and sorafenib inhibited the progression of DEN (Diethylnitrosamine)-induced HCC. In the HepG2 xenografts model, sorafenib plus SDC exhibited greater suppression on tumor growth than individual treatment accompanied with decreased expression of VEGF, VEGFA, Ki67, CD31 and increased expression of caspase-3. Furthermore, PI3K/Akt/mTORC1 pathway was inhibited by co-administration. Sorafenib monotherapy elicited hepatotoxicity for specific expression in the up-regulated level of aspartate transaminase (AST) and AST/glutamic-pyruvic transaminase (ALT) ratio, but the co-administration could remedy this adverse effect. These dates indicated that the combination of SDC and sorafenib might offer a potential therapy for HCC.

To study the effects of energy and dietary fiber on breast development in gilts and its possible mechanisms, 32 gilts (Landrace × Yorkshire) were randomly allocated into a 2 × 2 factorial design to receive a diet with low or high energy [LE: 33.37 MJ/d digestible energy (DE); HE: 41.87 MJ/d DE] and low or high fiber (LF: 0.3 kg/d dietary fiber, HF: 0.6 kg/d dietary fiber). The weight of breast tissue was recorded. The mammary glands were collected for further analyses. The high energy intake increased the relative weight of breast tissue (p < 0.05) and the content of breast fat (p < 0.05). At the same time, the oil red staining of breast slices also showed an increase in breast fat content in high-energy treatment. High energy intake increased the DNA concentration in breast tissues (p < 0.05). In addition, high energy intake increased the concentration of triglycerides, free fatty acids, and total cholesterol in the blood of gilts (p < 0.05), and the supplementation of high fiber tended to reduce free fatty acids, total cholesterol, and estradiol (p < 0.1). Proteomic analysis suggested that there were notable differences in the cytoskeleton, intracellular non–membrane-bounded organelle, apoptosis, receptor activity, and endopeptidase inhibitor activity in molecular function between the energy and fiber effects (p < 0.05). High fiber intake also decreased the mRNA expression of 5-HT7, Bax, and caspase-3 in the breast tissue of gilts (p < 0.05), which further confirmed the importance of fiber in regulating breast development in gilt. Our results indicate that increasing gilt energy intake improved breast weight and fat deposition and increased breast cell apoptosis. Increased fiber intake reduced breast fat deposition and breast cell apoptosis at high energy intake in gilts. These results provide a potential strategy for dietary intervention against high energy intake in gilts and even in humans.

Testicular development is imperative to spermatogenesis, and pre-puberty is the key period for testis development. This study, therefore, investigated the effects of fibre supplementation on testis development and its possible mechanism in a growing boar model. Thirty Yorkshire boars were randomly divided into a control group (Control) and a fibre group (Fibre) from day 0 to 90 after weaning, with three pigs per pen and five pens per treatment. Blood and testes were collected for analysis. Dietary fibre supplementation had no significant effect on growth performance, testicular volume, or libido but increased the semen production of boars. Boars fed with fibre had lower serum cholesterol (CHO) and low-density lipoprotein (LDL) levels compared to those on the Control diet; however, testicular CHO, triglyceride (TG), and LDL concentration in the Fibre group were significantly higher than the Control group (P < 0.01). Testicular histological analysis showed that seminiferous tubules and testicular germ cells of 120-day-old boars were densely arranged in the Fibre group, and the number of Leydig cells was significantly higher than that of the Control group (P < 0.001). Furthermore, the diet supplemented with fibre significantly decreased leptin, leptin receptor (Leptor), and luteinising hormone (LH) concentrations in boar serum (P < 0.05), whereas follicle-stimulating hormone (FSH) and testosterone concentrations were significantly increased (P < 0.05). Meanwhile, the expression of AMH, AMHR2, and SYCP3 genes related to proliferation and differentiation, and hormone-related genes STAR and SOCS3, were significantly up-regulated (P < 0.05). OCCLUDIN expression was up-regulated, whereas CDH2 expression was down-regulated. In conclusion, increased fibre intake during the pre-puberty period in growing boar is crucial for Leydig cell proliferation, up-regulating the expression of genes related to hormone synthesis and thereby promoting the secretion of testosterone and semen production.

Abstract Background The prevalence of peripheral artery disease (PAD) is obviously increased in patients with diabetes. Existing evidence shows that cysteine-rich angiogenic inducer 61 (Cyr61), a 40-kD secreted protein, plays important roles in regulating cellular physiological processes. Recent studies have demonstrated a significant correlation between serum Cyr61 and atherosclerosis. However, the relationship between Cyr61 levels and PAD in patients with type 2 diabetes (T2DM) remains obscure. Methods Data from a total of 306 subjects with T2DM were cross-sectionally analysed. The extent of PAD was determined by using the Fontaine classification, which defines four stages. We measured serum Cyr61 concentrations by ELISA in subjects with and without PAD at Fontaine’s stage II, III, or IV. Logistic regression models were used to examine the independent association of Cyr61 with PAD. Results Out of the 306 subjects enrolled, 150 were free from PAD, while 156 had clinically significant PAD. In subjects with PAD, the prevalences of Fontaine classification stages II, III and IV were 48.7%, 32.1%, and 19.2%, respectively.
