Landfills are always the most important part of solid waste management and bear diverse metabolic activities involved in element biogeochemical cycling. There is an increasing interest in understanding the microbial community and activities in landfill cover soils. To improve our knowledge of landfill ecosystems, we determined the microbial physiological profiles and communities in three landfill cover soils (Ninghai: NH, Xiangshan: XS, and Fenghua: FH) of different ages using the MicroResp, phospholipid fatty acid (PLFA), and high-throughput sequencing techniques. Both total PLFAs and glucose-induced respiration suggested more active microorganisms occurred in intermediate cover soils. Microorganisms in all landfill cover soils favored l-malic acid, ketoglutarate, and citric acid. Gram-negative bacterial PLFAs predominated in all samples with the representation of 16:1ω7, 18:1ω7, and cy19:0 in XS and NH sites. Proteobacteria dominated soil microbial phyla across different sites, soil layers, and season samples. Canonical correspondence analysis showed soil pH, dissolved organic C (DOC), As, and total nitrogen (TN) contents significantly influenced the microbial community but TN affected the microbial physiological activities in both summer and winter landfill cover soils. Although reductive soil disinfestation (RSD) is increasingly used for the control of soil-borne diseases, its impact on the soil microbial community during and after RSD remains poorly understood. MiSeq pyrosequencing, real-time PCR and denaturing gradient gel electrophoresis were performed to investigate the changes of microbial community in the Fusarium oxysporum f. sp. cubense (FOC) infected soil during RSD and at the simulative banana cultivation after RSD. The results showed that RSD significantly increased soil microbial populations and a different microbial community with the pathogenic soil was established after RSD. Specifically, the number of Firmicutes mainly containing Ruminococcus and Coprococcus followed by a small part of Clostridium which were the dominant bacterial genera significantly increased during RSD. In contrast, Symbiobacterium and Flavisolibacter were the dominant genera in the flooding soil. When the soils were recovered under aerobic condition, the relative abundances of the bacteria belonging to the phylum Bacteroidetes , Acidobacteria , Planctomycetes increased as alternatives to the reducing Firmicutes . For fungi, the population of F. oxysporum significantly decreased during RSD accompanied with the pH decline, which resulted in the significant decrease of relative abundance in the phylum Ascomycota . Alternatively, the relative abundances of some other fungal species increased, such as Chaetomium spp. and Penicillium spp. belonging to Ascomycota and the family Clavulinaceae belonging to Basidiomycota . Then, the relative abundance of Ascomycota re-increased after RSD with Podospora and Zopfiella as dominant genera, whereas the relative abundance of Fusarium further decreased. Overall, the microbial populations and community re-established by RSD made the soil more disease-suppressive and beneficial to the soil nutrient cycling and plant growth compared with the previous pathogenic soil. Biochar is a promising novel material for mitigating phosphorus (P) loss and enhancing P retention in chemical-amended agricultural soils. However, the optimal application rate for aforesaid effectiveness and potential drivers of the process are not well understood. Herein, a column-based pot experiment was carried out to investigate how and to what extent reed-biochar is effective in positively triggering P loss and availability in paddy soils treated by chemical fertilizer. Compared with chemical-only treatment, the accumulated leakage of total P, dissoluble P, and particulate P in chemical fertilizer coupled with 1–4% reed-biochar treatment decreased by 5.3–13.3%, 8.3–10.4%, and 3.0–15.4%, respectively. The accumulated leakage of total P and dissoluble P in 6–8% rate treatments was increased by 5.6–7.5% and 18.3–32.9%, respectively. Increasing reed-biochar rate from 1% to 8% caused an enhancement in soil total P and available P content and P activation coefficient, and the 4% rate achieved a similar effectiveness to the higher rate. Reed-biochar application increased the abundance and diversty of soil phoD-harboring microbes (P < 0.05), while the increment had little to do with the application rate. Soil phoD-harboring community composition and total C content were the main predictors of the P leaching losses, and meanwhile, the total C content was the dominated predictor of soil P retention and availability. These results suggest that adding 1–4% reed-biochar was more beneficial to mitigate paddy P loss and to enhance soil P availability. This study highlights the importance of understanding how microbial populations mediate P transformation to decipher the biochar-driven improvement of soil P utilization. Abstract: Continuous cropping is not considered to be a long-term sustainable agricultural practice. How soil microbial properties change under continuous cropping is not yet fully understood. Dynamics of cucumber rhizosphere soil microbial communities were monitored in a continuous cropping system, in which cucumber was monocropped in pots filled with field soils for nine successive croppings under greenhouse conditions. Results showed that rhizosphere soil dehydrogenase activity, microbial biomass carbon (MBC) content and active bacterial community abundance were significantly affected by the number of croppings (P < 0.05). Soil dehydrogenase activity was the lowest in the seventh cropping and increased in the ninth cropping. Soil MBC content and active bacterial community abundance increased in the third cropping and tended to decrease with croppings from the third to the ninth cropping. PCR-denaturing gradient gel electrophoresis (DGGE) analysis of bacterial 16S rRNA gene fragments demonstrated that continuous cropping caused shifts in bacterial community structures at both DNA and RNA levels, and the active bacterial communities were more sensitive to continuous cropping. Overall, our results revealed that soil microbial communities changed during continuous cropping of cucumber, which may be both a cause and a reflection of the poor plant growth performance. There is growing evidence of a direct relationship between microbial community composition and function, which implies that distinct microbial communities vary in their functional properties. The aim of this study was to determine whether differences in initial substrate utilization between distinct microbial communities are due to the activities of certain microbial groups. We performed a short-term experiment with beech forest soils characterized by three different microbial communities (winter and summer community, and a community from a tree-girdling plot). We incubated these soils with different 13C-labelled substrates with or without inorganic N addition and analyzed microbial substrate utilization by 13C-phospholipid fatty acid ( PLFA) analysis. Our results revealed that the fate of labile C (glucose) was similar in the three microbial communities, despite differences in absolute substrate incorporation between the summer and winter community. The active microbial community involved in degradation of complex C substrates (cellulose, plant cell walls), however, differed between girdling and control plots and was strongly affected by inorganic N addition. Enhanced N availability strongly increased fungal degradation of cellulose and plant cell walls. Our results indicate that fungi, at least in the presence of a high N supply, are the main decomposers of polymeric C substrates. [ABSTRACT FROM AUTHOR]

The Po river plain lowland springs represent unique paradigms of managed environments. Their current locations used to be swamps that were drained 6-7 centuries ago, and they have been in constant use ever since. Our aims were to identify the effects of land use on the microbial communities of these soils, look for associated diversity drivers, and assess the applicability of ecology theories with respect to identified patterns. We screened the microbial diversity across a land use transect via high-throughput sequencing of partial 16 S r rRNA gene amplicons. Land use had a major effect on soil properties and microbial community structures. Total organic carbon and p H were major diversity drivers for Bacteria, and p H was important for Archaea. We identified the potential contribution of soil amendments to the indigenous microbial communities, and also gained insights into potential roles of taxa in the organic carbon turnover. Verrucomicrobia coincided with the higher values of the recalcitrant organic carbon. Actinobacteria and Acidobacteria correlated with the more labile organic carbon. Finally, the higher diversity found in the soils less enzymatically active and relatively poorer in nutrients, may be explained to an extent by niche-based theories such as the resource heterogeneity hypothesis and Connell's intermediate disturbance hypothesis. Vermicompost, the digestion product of organic material by earthworms, has been widely reported to have a more positive effect on plant growth and plant health than conventional compost. A study was conducted to investigate the effects of different vermicompost elutriates (aerated compost teas) on soils and plant growth. The teas were analyzed by chemical, microbiological, and molecular methods accompanied by plant growth tests at laboratory and field scale. The number of microorganisms in the teas increased during the extraction process and was affected by substrate addition. The vermicompost tea found to increase plant growth best under laboratory tests was applied to cereals (wheat and barley) and vegetables ( Raphanus sativus, Rucola selvatica, and Pisum sativum) in a field study. The results revealed no effects of tea application on plant yield; however, sensoric tests indicated an improvement in crop quality. The soils from laboratory and field studies were investigated to detect possible microbial or chemical changes. The results indicated that minor changes to the soil microbial community occurred following tea application by foliar spray in both the laboratory-scale and field-scale experiments. Abstract: Wetlands may be constructed to provide several ecosystem functions. A constructed wetland receiving agricultural runoff water was observed prior to, and for more than two years after, establishment. The excavated portion of this wetland was compared to an undisturbed, upland area and to an adjacent, natural, depressional, reference wetland. After construction the excavated cell was rapidly colonized by wetland plant species, including some exotic invasive weeds. Flourescein diacetate (FDA) hydrolysis and triphenyl tetrazolium chloride (TTC) dehydrogenase assays indicated that the soil microbial community was more active in the excavated wetland cell than either of the two reference areas. In 2003 and 2004 TTC activity was greater than twice as high in the constructed wetland than the reference wetland. FDA was greatly stimulated in the constructed wetland immediately after construction, but by 2004 the three systems were not significantly different. The soil microbial community of the constructed wetland rapidly decreased in the abundance of fungi and gram-negative bacteria and increased in gram-positive bacteria and overall bacteria, as measured by fatty acid methyl esters. These shifts in the microbial community were consistent with the differences noted between the communities of the upland system and the reference wetland. During the time of observation the constructed wetland did not sequester carbon relative to the upland system (P =0.11) and did not significantly increase in carbon content (P =0.41). Abstract: Electrokinetic remediation has been successfully used to remove organic contaminants and heavy metals within soil. The electrokinetic process changes basic soil properties, but little is known about the impact of this remediation technology on indigenous soil microbial activities. This study reports on the effects of electrokinetic remediation on indigenous microbial activity and community within diesel contaminated soil. The main removal mechanism of diesel was electroosmosis and most of the bacteria were transported by electroosmosis. After 25days of electrokinetic remediation (0.63mAcm−2), soil pH developed from pH 3.5 near the anode to pH 10.8 near the cathode. The soil pH change by electrokinetics reduced microbial cell number and microbial diversity. Especially the number of culturable bacteria decreased significantly and only Bacillus and strains in Bacillales were found as culturable bacteria. The use of EDTA as an electrolyte seemed to have detrimental effects on the soil microbial activity, particularly in the soil near the cathode. On the other hand, the soil dehydrogenase activity was enhanced close to the anode and the analysis of microbial community structure showed the increase of several microbial populations after electrokinetics. It is thought that the main causes of changes in microbial activities were soil pH and direct electric current. The results described here suggest that the application of electrokinetics can be a promising soil remediation technology if soil parameters, electric current, and electrolyte are suitably controlled based on the understanding of interaction between electrokinetics, contaminants, and indigenous microbial community. Abstract: Soil and rhizosphere microbial communities in agroecosystems may be affected by soil, climate, plant species, and management. The management and environmental factors controlling microbial biomass and community structure were identified in a three-year field experiment. The experiment consisted of a tomato production agroecosystem with the following nine treatments: bare soil, black polyethylene mulch, white polyethylene mulch, vetch cover crop, vetch roots only, vetch shoots only, rye cover crop, rye roots only, and rye shoots only. The following hypotheses were tested: (1) Temperature and moisture differences between polyethylene-covered and cover-cropped treatments are partly responsible for treatment effects on soil microbial community composition, and (2) Different species of cover crops have unique root and shoot effects on soil microbial community composition. Microbial biomass and community composition were measured by phospholipid fatty acid analysis. Microbial biomass was increased by all cover crop treatments, including root only and shoot only. Cover cropping increased the absolute amount of all microbial groups, but Gram-positive bacteria decreased in proportion under cover crops. We attribute this decrease to increased readily available carbon under cover-cropped treatments, which favored other groups over Gram-positive bacteria. Higher soil temperatures under certain treatments also increased the proportion of Gram-positive bacteria. Vetch shoots increased the amount and proportion of Gram-negative bacteria, fungi, and arbuscular mycorrhizal fungi in the rhizosphere of tomato plants. The imposed treatments were much more significant than soil temperature, moisture, pH, and texture in controlling microbial biomass and community structure. [Copyright &y& Elsevier]

Abstract: Eucalyptus is one of the fastest growing woody plants in the world, but few studies have reported the soil microbial community composition in Eucalyptus ecosystems. This study investigated the soil microbial communities in plantations of 3-, 7-, 10- and 13-year-old Eucalyptus in subtropical China based on phospholipid fatty acids (PLFA) analysis. The variation in soil microbial biomass and community compositions were influenced by sampling site and season and the interaction of both, which were consistent with the variation in soil total nitrogen (TN), soil organic carbon (SOC) and soil moisture. The number and abundances of PLFAs, and the amount of soil TN and SOC were higher in plantation of 13-year-old Eucalyptus than those in other younger plantations, suggesting that the soil properties and the soil microbial community composition is not negatively affected by the planting of Eucalyptus. The ratio of monounsaturated-to-branched fatty acids, the proportional abundance (mol%) of bacterial PLFA and fungal PLFA varied significantly with Eucalyptus plantations of different age, suggesting that the individual PLFA signatures might be sensitive indicators of soil properties associated with forest plantations. The effect of pulp mill sludge addition (10–30Mg/ha) to soil derived from volcanic ash (Andisol) on soil characteristics, microbial community and Lolium perenne L. cv quartet. biomass production was evaluated in field assays. Soil without sludge was used as a control treatment. The sludge addition improved the chemical properties of the soil. Organic matter and phosphorous content increased in the soil with increasing amounts of sludge, obtaining 35% more organic matter content with the application of 30Mg/ha than the control soil. The phosphorous was accumulated into the soil after the end of cultivation improving the phosphorous pool in the soil. When 30Mg/ha sludge was added to the soil, a biomass of Lolium perenne, was 60% more than the control soil at the end of the experiment. The analysis of soil microbial community showed that the application of sludge did not modify greatly the microbial community of fungi and bacteria even when high doses were applied.
