1. Comparative analysis of the metabolites in Pinellia ternata from two producing regions using ultra-high-performance liquid chromatography–electrospray ionization–tandem mass spectrometry

Abstract

The rhizomes of Pinellia ternata have a long history of being used as a traditional Chinese herb. To determine the chemical constituents of Pinelliae rhizome, Xi banxia and Jing banxia were collected from two different producing regions and subjected to analysis using ultra-high-performance liquid chromatography–electrospray ionization–tandem mass spectrometry. A total of 573 metabolites were identified in 12 different categories. Next, cluster analysis and principal component analysis were performed, which revealed a distinct separation between the two species. The analysis of the differential metabolites revealed that among the 155 metabolites in Xi banxia, 68 metabolites were upregulated, and 87 metabolites were downregulated. The relative concentration of flavonoids in Xi banxia was higher than that in Jing banxia, while the relative concentrations of alkaloids, phenolic acids, and terpenoids were higher in Jing banxia. These results would provide a theoretical basis for the pharmacological activity analysis and functional study of P. ternata.

CUR-M was successfully prepared by dialysis, and the formulation was evaluated for particle size, zeta potential, surface morphology, entrapment effciency (EE%), drug loading (DL), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR) and the in vitro drug release. Additionally, in vitro skin permeation and retention, in vivo topical analgesic and anti-inflammatory activity, and skin irritation were assessed.Results: The mean drug loading (DL), drug entrapment efficiency (EE), hydrodynamic diameter and zeta potential of CUR-M were 8.26%, 90.86%, 165.64 nm and − 26.85 mV, respectively. CUR-M was characterized by X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR), it was found that there was an interaction between CUR and OHA, and CUR existed in CUR-M in an amorphous form. CUR-M exhibited sustained release in 48 h and good stability at 4 °C for 21days. Abstract

Implantation with low-energy ion beam can induce various plant biological effects including stimulation, damage and mutation. But the adaptive mechanism altering gene expression in response to this stressor in plant is not fully understood. A highly thorough and comprehensive analysis to investigate the general-key genes responsive to implantation with low-energy ion beam in plant is important to understand the diverse mechanism. We used Agilent Rice Gene Expression Microarray to investigate the overlap differentially expressed genes between two independent implantation experiments underlying three ion fluences. Our microarray data showed that 26 up-regulated overlap genes and 6 down-regulated overlap genes were observed. The Gene Ontology terms analysis showed that genes with functions including kinase, transporter, acceptor, transcription regulation were involved in signaling and response biological processes in rice responding to the implantation with ion beam. Our data also implied that ABA signal transduction pathway is involved in response to ion-beam implantation in rice. The RiceNet co-expressed network analysis showed the direct and indirect co-expressed linkages among these up-regulated genes, which suggested some signal transduction pathways should be involved in this response and remained unclear. Our microarray data revealed the general and key genes responsive to the stress of implantation with low-energy ion beam during the rice germination. These data provide information on the candidate genes for further elucidation regarding the molecular mechanisms of the biological effects of ion-beam implantation and its application for improvement of corn. Abstract

In order to understand the molecular mechanism of the ion-implantation biological effects, we analyzed genes expression profiles in rice seedlings from the seeds implanted with the nitrogen ion beam, using the Agilent Rice Oligo Microarray (4×44) Genome Array. Rice seeds were implanted by the nitrogen ion beam with 40 Kev energy in dose 6×1017 N+/cm2. Total RNA, from the seedlings 96h after the germination, was used to genechip analysis. 16. Gene expression profiles in promoted-growth rice seedlings that germinated from the seeds implanted by low-energy N+ beam

Abstract

The stimulation effect that some beneficial agronomic qualities have exhibited in present-generation plants have also been observed due to ion implantation on plants. However, there is relatively little knowledge regarding the molecular mechanism of the stimulation effects of ion-beam implantation. In order to extend our current knowledge about the functional genes related to this stimulation effect, we have reported a comprehensive microarray analysis of the transcriptome features of the promoted-growth rice seedlings germinating from seeds implanted by a low-energy N+ beam. The results showed that 351 up-regulated transcripts and 470 down-regulated transcripts, including signaling proteins, kinases, plant hormones, transposable elements, transcription factors, non-coding protein RNA (including miRNA), secondary metabolites, resistance proteins, peroxidase and chromatin modification, are all involved in the stimulating effects of ion-beam implantation. The divergences of the functional catalog between the vacuum and ion implantation suggest that ion implantation is the principle cause of the ion-beam implantation biological effects, and revealed the complex molecular networks required to adapt to ion-beam implantation stress in plants, including enhanced transposition of transposable elements, promoted ABA biosynthesis and changes in chromatin modification. Our data will extend the current understanding of the molecular mechanisms and gene regulation of stimulation effects. Further research on the candidates reported in this study should provide new insights into the molecular mechanisms of biological effects induced by ion-beam implantation.
