The utilization of genotyping has gained significant popularity in tree improvement programs, aiding in enhancing the precision of breeding values, removing pedigree errors, the assessment of genetic diversity, and evaluating pollen contamination. Our study explores the impact of utilizing 5308 SNP markers to genotype seed orchard parents (166), progeny in progeny trials (667), and seedlot orchard seedlings (780), to simultaneously enhance variance components, breeding values, genetic diversity estimates, and pollen flow in the Region I white spruce (Picea glauca) breeding program in central Alberta (Canada). We compared different individual tree mixed models, including pedigree-based (ABLUP), genomic-based (GBLUP), and single-step pedigree-genomic-based (ssGBLUP) models, to estimate variance components and predict breeding values for the height and diameter at breast height traits. The highest heritability estimates were achieved using the ssGBLUP approach, resulting in improved breeding value accuracy compared to the ABLUP and GBLUP models for the studied growth traits. In the six orchard seedlots tested, the genetic diversity of the seedlings remained stable, characterized by an average of approximately 2.00 alleles per SNP, a Shannon Index of approximately 0.44, and an expected and observed heterozygosity of approximately 0.29. The pedigree reconstruction of seed orchard seedlings successfully identified consistent parental contributions and equal genotype contributions in different years. Pollen contamination levels varied between 11% and 70% using SNP markers and 8% to 81% using pollen traps, with traps both over- and under-estimating contamination. Overall, integrating genomic information from parents and offspring empowers forest geneticists and breeders in the Region I white spruce breeding program to correct errors, conduct backward and forward selections with greater precision, gain a deeper understanding of the orchard's genetic structure, select superior seedlots, and accurately estimate the genetic worth of each orchard lot, which can ultimately result in increased and more precise estimates of genetic gain in the studied growth traits. A significant portion of the harvested land base in western Canada is becoming old enough or entering a phase where thinning is a legitimate forest management option. A comprehensive review of the existing knowledge of commercial thinning (CT) treatments applied to pine and spruce-dominated stands in Alberta was conducted, with particular regard to the intensity, timing of interventions, method, and impacts on crop tree growth responses. Although the geographical focus of this review is Alberta, information on this topic is more complete in other areas of North America and Europe, where there is a long history of density management. In areas of eastern North America, our review revealed that CT from below, with tree removal levels from 27 to 43% of the basal area, could increase total merchantable wood produced from 11 to 60 m3 ha−1 over a rotation, depending on stand age and intensity of thinning. For Alberta conditions, and considering the risks, we conclude that commercial thinning basal area removal should be in the range of 25 to 40%, depending on a variety of factors such as species, wind firmness, and insect or disease incidence and risk. Thinning too aggressively and/or too late will increase the blowdown risk but the literature is fairly consistent in suggesting that live crown ratios should be >40% to maximize the chance of growth response and minimize the blowdown risk. In cases where stands are also threatened by stressors such as drought, wind, and insect or disease outbreaks, CT treatments likely offer the potential at limiting the overall risk, but localized knowledge and experience are critical. It is intended that the information presented may support ongoing and future research trials and growth and yield (G&Y) model development about potential CT treatments to apply and the likely results of practical application to commercial forestry. Tree improvement programs select genotypes for faster growth, at both early and late stages, to increase yields over unimproved material, and the improvement is frequently attributed to genetic control in growth parameters among genotypes. Underutilized genetic variability among genotypes also has the potential to ensure future gains are possible. However, the genetic variation in growth, physiology and hormone control among genotypes generated from different breeding strategies has not been well characterized in conifers. We assessed growth, biomass, gas exchange, gene expression and hormone levels in white spruce seedlings obtained from three different breeding strategies (controlled crosses, polymix pollination, open pollination) using parents grafted into a clonal seed orchard in Alberta, Canada. A pedigree-based best linear unbiased prediction (ABLUP) mixed model was implemented to quantify variability and narrow-sense heritability for target traits. The levels of several hormones and expression of gibberellinrelated genes in apical internodes were also determined. Over the first two years of development, the estimated heritabilities for height, volume, total dry biomass, above ground dry biomass, root:shoot ratio and root length, varied between 0.10 and 0.21, with height having the highest value. The ABLUP values showed large genetic variability in growth and physiology traits both between families from different breeding strategies, and within families. The principal component analysis showed that developmental and hormonal traits explained 44.2% and 29.4% of the total phenotypic variation between the three different breeding strategies and two growth groups. In general, controlled crosses from the fast growth group showed the best apical growth, with more accumulation of indole-3-acetic acid, abscisic acid, phaseic acid, and a 4-fold greater gene expression of PgGA3ox1 in genotypes from controlled crosses versus those from open pollination. However, in some cases, open pollination from the fast and slow growth groups showed the best root development, higher water use efficiency (iWUE and d13C) and more accumulation of zeatin and isopentenyladenosine. Inconclusion, tree domestication can lead to trade-offs between growth, carbon allocation, photosynthesis, hormone levels and gene expression, and we encourage the use of this phenotypic variation identified in improved and unimproved trees to advance white spruce tree improvement programs. Stand tending using glyphosate to promote coniferous overstory trees has been a common practice in the boreal forests of Alberta. However, there are concerns about the impact of this practice on biodiversity of understory species. This study examined the impact of broadcast glyphosate application during the active reforestation phase, two to several years post-harvest, on forest plant diversity 25 to 40 years post-harvest. Herbicide treatments had the desired effect of shifting tree layer dominance from deciduous to coniferous species, driven by a 25-fold reduction in the density of trembling aspen (3927 vs. 154 stems·ha−1, untreated and treated, respectively). However, understory plant diversity was not significantly different between treated and untreated sites as examined by the Shannon–Wiener (H) and evenness (E) indices. Shared plant species (beta) across sites was high. Of the seven site-indicator species examined, three had significantly lower cover on treated sites: Wild sarsaparilla, low bush-cranberry and oak fern. Total understory plant cover was significantly greater in the treated portion (98.0%) versus untreated (71.4%); however, this difference was not significant when bryophytes were excluded in the analysis. Understory protection harvesting is a form of partial cutting that can be used in aspen (Populus tremuloides Michx.)-dominated stands that have understories of white spruce (Picea glauca (Moench) Voss). This practice involves removing 75% to 85% of the merchantable aspen while minimizing damage to the advance spruce regeneration, in addition to leaving 15% to 25% of the aspen standing to reduce potential windthrow of the spruce understory. In this paper, we summarize results from 18 stands measured 10 to 12 years after understory protection harvest. Diameter growth of spruce increased during the first five years after harvest while height growth increased during the second five-year period (5 to 10 or 7 to 12 years after release). Consistent with other studies, mortality rates of aspen trees ≥7.1 cm DBH (diameter breast height, 1.3 m) averaged 45.0% over the 10–12 year period following harvesting. Spruce mortality averaged 27.5% over the same 10–12 year period. Substantial aspen regeneration was evident across most harvested blocks, with aspen sapling densities 10–12 years from harvest being higher in removal (14,637 stems⋅ha−1) than in buffer areas (6686 stems⋅ha−1) and in extraction trails (7654 stems⋅ha−1). Spruce sapling (>1.3 m height and <4 cm DBH) densities averaged 1140 stems⋅ha−1 in removal areas at ages 10–12, with these trees likely being present as seedlings at the time of harvest. Mixedwood Growth Model projections indicate merchantable volumes averaging 168 m3⋅ha−1 (conifer) and 106 m3⋅ha−1 (deciduous) 70 years from harvest, resulting in MAI (mean annual increment) for this period averaging 2.0 m3⋅ha−1⋅y−1 with MAI for a full 150-year rotation of approximately 2.5 m3⋅ha−1⋅y−1. Nine open-pollinated families of Picea glauca (Moench) Voss from the Region D1 Controlled Parentage Program (Alberta, Canada) were systematically chosen from fast, medium and slow-growth rankings based on breeding values for height from field progeny tests at age 30 years. Seeds from these families were sown and grown to age 3 years to analyze the performance and correlations of growth, physiological traits and expression of gibberellin-related genes, with and without elevated gibberellic acid 3 (GA3) application, under greenhouse conditions. We observed a significant interaction effect between families and growth groups subjected to 50 μg μl−1 of GA3 treatment, causing a decrease in apical internode length, diameter, volume and absolute transcript level for fast-growing families but an increase for families in the slow-growth group for the same traits. We also observed that in the apical internode, the gene PgGA20ox1 had significantly more relative expression under the elevated GA3 treatment than the control trees. In the stem, PgGA3ox1 showed a significantly higher relative expression under elevated GA3 treatment compared with control trees. Also, the slow-growth group showed more relative expression of PgGA20ox1 (in the apical internode) and PgGA3ox1 (in the stem) than the fast-growth group. The apical internode length and diameter significantly increased by 24% and 16%, respectively, with the hormone treatment in the slow growing group. In general, the PgGID1 and PgDELLA1 genes were upregulated and downregulated respectively, in spruce shoots under the GA3 treatment, meaning a positive feedback regulation by those genes were influencing PgGA20ox1 and PgGA3ox1 expression in that tissue type. Moreover, there was a significant correlation between absolute transcript levels of PgGA20ox1 in the apical internode and apical internode length, and absolute transcript levels of PgGA3ox1 in the stem and the diameter, in the fast-growth group families. This study shows that expression of GA genes is a limiting factor for growth in certain white spruce families with a complex feedback mechanism. Finally, absolute transcript levels of endogenous GA relative to growth parameters in juvenile seedlings could potentially be used to accelerate the early selection of families with inherently rapid apical and radial growth expansion. Genomics-assisted tree breeding (GATB) is an emerging biotechnology method that has the potential to produce improved planting stock in selected traits, such as greater volume or higher wood quality, more quickly and effectively compared with traditional field-based breeding (TB) methods. In this study, we conducted an ex-ante stand-level financial benefit-cost analysis that is linked to a provincial tree growth and yield projection model to investigate the potential financial benefits of using improved white spruce (Picea glauca (Moench) Voss) and lodgepole pine (Pinus contorta Dougl. var. latifolia Engelm.) planting stock identified via alternative tree improvement strategies (i.e. GATB vs. TB methods) for forest companies in the province of Alberta, Canada. With consideration of major tree breeding and establishment costs, expected sawlog price premiums at harvest, a shorter breeding cycle time, and additional genetic gain in volume achieved from using the GATB method relative to the TB method in the financial analysis; this study found that the use of GATB-selected third-generation planting stock was difficult to justify economically alone compared with TB-selected seedlings under the current sawlog market value and the current planting rate with improved stock in the province. However, results of our sensitivity analysis also revealed that the GATB method is: (1) more financially supported at lower discount rates; (2) strongly supported through decreased seedling costs with increased areas planted with improved stock; and (3) strongly supported through an increase in log price premiums at harvest resulting from potentially better wood quality and higher volume production relative to TB-selected stock. A variable-exponent stem profile model was developed for white spruce (Picea glauca (Moench) Voss) trees in Alberta using a nonlinear mixed model approach. Between-tree variation in stem diameter was best captured by incorporating three random parameters into the model. Heterogeneous residual variances were modelled as a power function of tree breast height diameter. Within-tree residual autocorrelation was modelled through a covariance structure. Based on model fitting statistics, the four-banded toeplitz (TOEP(4)) and the spatial power structures were both selected for further evaluation. An independent validation dataset was used for evaluating the calibration accuracy of the final model in stem diameter and volume predictions. To make tree-specific calibrations, one or more prior diameter measures should be available from each tree. For this study, four scenarios were evaluated, in which one, two, three and four prior diameters were randomly selected for predicting random parameters by an approximate Bayesian estimator. Tree-specific calibrations were subsequently derived. Population-level predictions were also produced for comparison. Fungi colonizing fine roots of containerized Picea glauca seedlings were assessed in four large conifer nurseries in northern Alberta. PCR amplification of fungal rDNA (internal transcribed spacer and a portion of the 5? end of the large subunit gene) from random samples of fine feeder roots gave between 1 and 4 amplicons per seedling. Amplicons were either separated by electrophoresis and sequenced directly, or cloned and sequenced. The resulting sequences were compared to sequences obtained from cultures established from seedling roots and from GenBank by maximum parsimony analysis. ITS sequences formed 11 distinct clades, each including at least one reference sequence. The ectomycorrhizal basidiomycetes Thelephora americana and Amphinema byssoides were dominant, whereas ascomycetes were less common. Fungi with sequences similar to members of the Heleotiales which form ericoid mycorrhizas were also present. Correspondence analysis revealed strong positive and negative associations among fungal taxa as well as an influence of applied fertilizer level on fungal diversity and species composition. A few studies in young mixedwood stands demonstrate that precommercial thinning of aspen at early ages can improve the growth of spruce and increase stand resilience to drought. However, information on tree and stand responses to thinning in older mixedwood stands is lacking. To address this need, a study was initiated in 2008 in Alberta, Canada in 14 boreal mixedwood stands (seven each at ages 17 and 22). This study investigated growth responses following thinning of aspen to five densities (0, 1000, 2500, 5000 stems ha−1 and unthinned (control)). Measurements were collected in the year of establishment, and three and eight years later. Mortality of aspen in the unthinned plots was greater than in the thinned plots which were not significantly different amongst each other. Eight years following treatment, aspen diameter was positively influenced by thinning, while there was no effect on aspen height. The density of aspen had no significant effect on the survival of planted spruce. Spruce height and diameter growth increased with both aspen thinning intensity and time since treatment. Differentiation among treatments in spruce diameter growth was evident three years from treatment, while differentiation in height was not significant until eight years following treatment. Yield projections using two growth models (Mixedwood Growth Model (MGM) and Growth and Yield Projection System (GYPSY)) were initialized using data from the year eight re-measurements. Results indicate that heavy precommercial aspen thinning (to ~1000 aspen crop trees ha−1) can result in an increase in conifer merchantable volume without reducing aspen volume at the time of harvest. However, light to moderate thinning (to ~2500 aspen stems ha−1 or higher), is unlikely to result in gains in either deciduous or conifer merchantable harvest volume over those of unthinned stands. Photosynthetic light and temperature response curves were measured seasonally in seedlings of white spruce (Picea glauca (Moench.) Voss) grown for two years in the understory of aspen (Populus tremuloides Michx.) or in the open in central Alberta. Light-saturated rate of net photosynthesis, the optimum temperature for net photosynthesis, transpiration rate, photochemical efficiency, and stomatal and mesophyll conductances increased from spring to summer and declined thereafter, whereas dark respiration rate and compensation and saturation points were highest in spring. Depression of photosynthetic parameters was greater in open-grown seedlings than in understory seedlings during the periods in spring and autumn when night frosts were common. Net photosynthetic rates were similar in understory and open-grown seedlings in summer, but they were significantly lower in open-grown seedlings in spring and autumn. Significantly lower transpiration rates and stomatal conductances in open-grown seedlings than in understory seedlings were also observed at 15 and 25 °C in the autumn. Shoot and needle growth were less in open-grown seedlings than in understory seedlings. In summer, when irradiances were low in the aspen understory, understory white spruce seedlings maintained a positive carbon balance by decreasing their compensation and saturation points and increasing their photochemical efficiency compared to spring and autumn. Gateway Hill is a 104-ha area in the oil sands region of Alberta, which was operationally reclaimed and reforested in the late 1980s and certified as reclaimed in 2007/08. In 2014, ∼30 years after reclamation was completed, we began studies with the main objective to determine the soil fungi present at the site. Sporocarp surveys in 2014 and 2016 were used to supplement metagenomic analyses of soil fungi in jack pine (Pinus banksiana), white spruce (Picea glauca), and Siberian larch (Larix sibirica) stands. Fungal internal transcribed spacer amplicons of DNA extracted from forest floor, root, and fine soil fractions were sequenced using Roche 454 pyrosequencing in 2014. Coarse soil reclamation samples and soil samples from a nearby unmined spruce stand were sequenced with Illumina in 2016. Sequenced community assemblages were first assessed using unconstrained multivariate analyses, and then using (partially) constrained ordination techniques to directly account for the effects of tree species and other confounding factors. Based on soil DNA analyses the relative abundance of ectomycorrhizal taxa was greater in spruce stands, saprotrophs were more abundant in pine stands, and pathogens were more abundant in larch stands. Associations of fungal taxa, mostly saprotrophs and ectomycorrhizae, with tree species were especially prominent in root fractions. The sporocarp survey found sporcarp abundance was greatest in pine stands and they tended to have more ectomycorrhizal taxa and spruce and larch stands more saprotrophic taxa. The effects of tree species on fungal taxa differed between the sporocarp surveys and soil DNA analyses: many of the fungi identified by sporocarp presence were not detected by soil DNA sequencing, whereas the latter method detected many more species in all trophic groups. Both DNA sequencing methods showed that many operational taxonomic units were preferentially associated with either spruce or pine stands. Though not definitive, preliminary examination of soil fungi using the Illumina platform showed that a natural spruce stand at the base of Gateway Hill had greater species richness than did reclaimed spruce stands. The results of this study show that communities of soil fungi have become established since the initiation of reclamation. This paper presents a scaling approach for estimating sapwood area at the stand level using knowledge obtained for individual trees of five boreal species: Populus tremuloides (Michx.), Pinus contorta (Doug. ex Loud. var. latifolia Engelm.), Pinus banksiana (Lamb.), Picea mariana (Mill.) BSP, and Picea glauca (Moench) Voss. Previously developed allometric models for sapwood depth and diameter at breast height for individual tree species were used to build stand level sapwood area estimates as well as stand level leaf area estimates, in pure and mixed vascular vegetation stands. A stand's vegetation heterogeneity is considered in the scaling approach by proposing regression models for each species. The new combined scaling approach drew strong linear correlations at the stand scale between sapwood area and leaf area using observations taken in mixed stands of Southern Alberta, Canada. This last outcome suggests a good linear relationship between stand sapwood area and stand leaf area.
