Cellular senescence contributes to inflammatory kidney disease via the secretion of inflammatory and profibrotic factors. Protease‐activating receptor 2 (PAR2) is a key regulator of inflammation in kidney diseases. However, the relationship between PAR2 and cellular senescence in kidney disease has not yet been described. In this study, we found that PAR2‐mediated metabolic changes in renal tubular epithelial cells induced cellular senescence and increased inflammatory responses. Using an aging and renal injury model, PAR2 expression was shown to be associated with cellular senescence. Under in vitro conditions in NRK52E cells, PAR2 activation induces tubular epithelial cell senescence and senescent cells showed defective fatty acid oxidation (FAO). Cpt1α inhibition showed similar senescent phenotype in the cells, implicating the important role of defective FAO in senescence. Finally, we subjected mice lacking PAR2 to aging and renal injury. PAR2‐deficient kidneys are protected from adenine‐ and cisplatin‐induced renal fibrosis and injury, respectively, by reducing senescence and inflammation. Moreover, kidneys lacking PAR2 exhibited reduced numbers of senescent cells and inflammation during aging. These findings offer fresh insights into the mechanisms underlying renal senescence and indicate that targeting PAR2 or FAO may be a promising therapeutic approach for managing kidney injury. Organic photodetectors (OPDs) based on non‐fullerene acceptors (NFAs) have received considerable attention because of their potential for use in various commercial applications as near‐infrared (NIR) light sensing platforms. However, recent OPDs suffer from low NIR photoresponse and large dark/noise currents with narrow bandgap organic photoactive materials. Herein, a π‐bridge molecular engineering strategy replacing alkoxythienyl with benzothiadiazole for ultra‐narrow bandgap (ultra‐NBG) NFAs is designed to achieve simultaneously high photoresponse at NIR region and low noise current density, thereby leading to excellent NIR (≈1050 nm) detectivity (D*). The newly synthesized ultra‐NBG NFAs, namely COB and CBT with optical bandgaps below 1.14 eV, present high responsivity (R) with 0.369 and 0.080 A W−1, respectively, at a wavelength of 1050 nm. Especially, with effectively suppressed noise current density, COB‐based OPD exhibits a high NIR (≈1050 nm) D* value of 2.18 × 1011 cm Hz1/2 W−1 at −0.5 V bias. The obtained R and D* values for these NFAs exceed or are comparable to those of a commercial Si photodetector at 1050 nm. This work provides important insight into the π‐bridge molecular engineering strategy for ultra‐NBG NFAs, which facilitate achieving highly sensitive NIR OPDs with high NIR photoresponse and low dark/noise current. TiO2 polymorphs have distinct properties that are widely employed in various applications. However, mechanisms of transformations between these polymorphs are not fully understood, especially at atomic scale, inhibiting advancing the design and application of the transitional phases. Here, based on results from semi‐in situ transmission electron microscopy, density functional theory, and X‐ray photoemission experiments, a physical picture of transitional structures is discovered, in which continuous variations in atomic positions form along a previously unreported anatase‐to‐rutile phase transformation path of [010]A–to–[1¯1¯1$\overline 1 \overline 1 1$]R and (004)A–to– (011)R. These gradient structures give rise to continuous band bending, which promotes electron‐hole separation and inhibits their recombination across the bulk of the particles, leading to a large functionally active volume fraction and resulting in high photoactivity. Understanding structure–function relationships enables the design of materials with tailored functionalities. The long‐standing challenge is to design materials with high active volume to improve efficiency. Tailored grain boundaries and lattice defects are traditionally used to tune the electronic structure near interfaces or defects to promote electron and hole separation. However, the active volume of point defect sites or interfaces in these traditional photocatalysts is extremely low. This study reports a structure with continuous atomic positional deformation across the bulk, altering the electronic structure in 3D and creating a significantly higher active volume. Such a structure in anatase is obtained and tuned by phase transformation during the heating process. Transmission electron microscopy and density functional theory results reveal that atomic deformations result in continuous band bending across the particles, facilitating electron–hole separation, inhibiting their recombination, and inducing dramatically enhanced photoactivity. The effect of the cross‐coupling catalyst tetrakis(triphenylphosphine)palladium(0) (Pd(PPh3)4) on the performance of a model organic bulk‐heterojunction solar cell composed of a blend of poly([2,6′‐4,8‐di(5‐ethylhexylthienyl)benzo[1,2‐b;3,3‐b]dithiophene]{3‐fluoro‐2[(2‐ethylhexyl)carbonyl]thieno[3,4‐b]thiophenediyl}) (PTB7‐Th) donor and 3,9‐bis(2‐methylene‐((3‐(1,1‐dicyanomethylene)‐6,7‐difluoro)‐indanone))‐5,5,11,11‐tetrakis(4‐hexylphenyl)‐dithieno[2,3‐d:2′,3′‐d′]‐s‐indaceno[1,2‐b:5,6‐b′]dithiophene (IOTIC‐4F) non‐fullerene acceptor is investigated. The effect of intentional addition of different amounts of Pd(PPh3)4 on morphology, free charge carrier generation, non‐geminate bulk trap‐ and surface trap‐assisted recombination as well as bimolecular recombination and charge extraction is quantified. This work shows that free charge carrier generation is affected significantly, while the impact of Pd(PPh3)4 on non‐geminate recombination processes is limited because the catalyst does not facilitate efficient trap‐assisted recombination. The relationship of the temperature–light intensity dependence of open‐circuit voltage Voc in nonfullerene‐based organic solar cells with their material characteristics and multimechanism recombination parameters is described. The systematic variation of the effective bandgap Eg,eff and the electrode layers allows the observation of different relative contributions of bimolecular, bulk, and surface trap‐assisted recombination mechanisms. The complementary advantages of the analytical model and the established voltage‐impedance spectroscopy technique provide a useful tool to quantify multimechanism recombination parameters, effective density of states Nc, and energetic disorder σ in organic solar cells under operating conditions. The validity of the proposed model to understand the temperature and light intensity dependent of Voc is shown by applying it to four different donor:nonfullerene acceptor blend systems with conventional or inverted device architectures. The short‐circuit current (Jsc) of organic solar cells is defined by the interplay of exciton photogeneration in the active layer, geminate and non‐geminate recombination losses and free charge carrier extraction. The method proposed in this work allows the quantification of geminate recombination and the determination of the mobility‐lifetime product (µτ) as a single integrated parameter for charge transport and non‐geminate recombination. Furthermore, the extraction efficiency is quantified based on the obtained µτ product. Only readily available experimental methods (current‐voltage characteristics, external quantum efficiency measurements) are employed, which are coupled with an optical transfer matrix method simulation. The required optical properties of common organic photovoltaic (OPV) materials are provided in this work. The new approach is applied to three OPV systems in inverted or conventional device structures, and the results are juxtaposed against the µτ values obtained by an independent method based on the voltage–capacitance spectroscopy technique. Objectives: This study focuses on the impact of race and ethnicity on intergenerational mobility between mothers and children. Background: Most studies have examined intergenerational mobility between fathers and their children. Racial and ethnic disparities might explain the high rates of intergenerational inequality in mobility. Methods: The current study used data from the National Longitudinal Survey of Youth 1979 and the National Longitudinal Survey of Youth 1979 for Children and Young Adults. The two data sets were merged based on mother and child's identification number. The final sample consisted of 1,245 non‐Black/non‐Hispanic, 740 Black, and 538 Hispanic dyads. Multiple linear regression and logistic regression analyses were conducted. Results: There are ethnic and racial disparities in net worth and poverty among mothers. Black and Hispanic mothers were less likely to accumulate net worth compared with non‐Black/non‐Hispanic mothers, and they were also at greater risk of being in poverty compared with non‐Black/non‐Hispanic mothers. For intergenerational economic mobility, net worth and not living in poverty across mothers were associated with higher income among young adult children. Black race moderated the relationship between maternal net worth and young adult children's income. Conclusion: A new perspective is necessary to explore intergenerational economic mobility, considering relations between mothers and children rather than fathers and children. Implications: Eliminating discrimination in the labor market is critical to addressing racial and ethnic disparities in economic resources. Service programs to help Black women with children to save money or increase their job opportunities should be in place to improve their potential for upward economic mobility across generations. This study examines whether the daughters' educational attainment mediates the intergenerational transmission of economic mobility between mothers and their young adult daughters. To create mother–daughter dyads, two data sets were combined: The National Longitudinal Survey of Youth 1979 (NLSY79) and the National Longitudinal Survey of Youth 79 for Children and Young Adults (NLSY79 CY). A total of 2,456 dyads were included for analysis. We used a mediation model to explore the relationship between mothers' income and their young adult daughters' income. Mothers' income was associated with their young adult daughters' educational attainment and income. The mediation model indicated partial mediation of the relationship between mothers' income and their young adult daughters' income via their young adult daughters' educational attainment. Addressing issues of income inequality among mothers may serve as a buffer against the low upward mobility across generations for their young adult daughters raised by low‐income mothers. It is imperative to provide programs and financial assistance for mothers to bolster their income and thereby their daughters' educational attainment and income in young adulthood and therefore improve economic mobility from mothers to daughters. In this work, environmentally friendly photocatalysts with attractive catalytic properties are reported that have been prepared by introducing SnO2 quantum dots (QDs) directly onto ZnSe(N2H4)0.5 substrates to induce advantageous charge separation. The SnO2/ZnSe(N2H4)0.5 nanocomposites could be easily synthesized through a one‐pot hydrothermal process. Owing to the absence of capping ligands, the attached SnO2 QDs displayed superior photocatalytic properties, generating many exposed reactive surfaces. Moreover, the addition of a specified amount of SnO2 boosted the visible‐light photocatalytic activity; however, the presence of excess SnO2 QDs in the substrate resulted in aggregation and deteriorated the performance. The spectroscopic data revealed that the SnO2 QDs act as a photocatalytic mediator and enhance the charge separation within the type II band alignment system of the SnO2/ZnSe(N2H4)0.5 heterojunction photocatalysts. The separated charges in the heterojunction nanocomposites promote radical generation and react with pollutants, resulting in enhanced photocatalytic performance. [ABSTRACT FROM AUTHOR]

Even though significant breakthroughs with over 18% power conversion efficiencies (PCEs) in polymer:non‐fullerene acceptor (NFA) bulk heterojunction organic solar cells (OSCs) have been achieved, not many studies have focused on acquiring a comprehensive understanding of the underlying mechanisms governing these systems. This is because it can be challenging to delineate device photophysics in polymer:NFA blends comprehensively, and even more complicated to trace the origins of the differences in device photophysics to the subtle differences in energetics and morphology. Here, a systematic study of a series of polymer:NFA blends is conducted to unify and correlate the cumulative effects of i) voltage losses, ii) charge generation efficiencies, iii) non‐geminate recombination and extraction dynamics, and iv) nuanced morphological differences with device performances. Most importantly, a deconvolution of the major loss processes in polymer:NFA blends and their connections to the complex BHJ morphology and energetics are established. An extension to advanced morphological techniques, such as solid‐state NMR (for atomic level insights on the local ordering and donor:acceptor ππ interactions) and resonant soft X‐ray scattering (for donor and acceptor interfacial area and domain spacings), provide detailed insights on how efficient charge generation, transport, and extraction processes can outweigh increased voltage losses to yield high PCEs. Three symmetrically difluorinated organic semiconductors (namely D5T2F‐P, D5T2F‐S, and D5T2F‐T) containing rhodanine‐flanked pentathiophene structures are synthesized and used as donors in all‐small‐molecule organic solar cells (ASM‐OSCs) prepared with the small‐molecule acceptor 2,2′‐((2Z,2′Z)‐((4,4,9,9‐tetrahexyl‐4,9‐dihydro‐s‐indaceno[1,2‐b:5,6‐b′]dithiophene‐2,7‐diyl)bis(methanylylidene))bis(5,6‐difluoro‐3‐oxo‐2,3‐dihydro‐1H‐indene‐2,1‐diylidene))dimalononitrile (IDIC‐4F). The different substitutional positions of fluorine atoms (–F) in the conjugated backbone of the donor molecule lead to various material and photovoltaic properties being exhibited. Among the three isomers, the centrally fluorinated D5T2F‐P exhibits a redshifted absorption spectrum, downshifted highest occupied molecular orbital (HOMO) energy level, and improved miscibility with IDIC‐4F in the blend films, all of which result in superior device performance. The power conversion efficiency (PCE) of the ASM‐OSCs based on D5T2F‐P:IDIC‐4F reaches an impressive value of 9.36% with an open‐circuit voltage (VOC) value of 0.86 V and a short‐circuit current density (JSC) value of 16.94 mA cm−2, whereas those of D5T2F‐S (6.11%) and D5T2F‐T (5.42%) are much lower. In comparison, an ASM‐OSC based on the nonfluorinated analogue DRCN5T fabricated under the same conditions exhibits poorer performance (8.03% with IDIC‐4F), revealing 16% enhancement in the PCE achieved through backbone fluorination. The PCE of 9.36% may be one of the highest efficiencies of oligothiophene‐based ASM‐OSCs reported in the literature to date. [ABSTRACT FROM AUTHOR]

Abstract: A series of narrow bandgap electron acceptors is designed and synthesized for efficient near‐infrared (NIR) organic solar cells. Extending π‐conjugation of donor frameworks leads to an intense intramolecular charge transfer, resulting in broad absorption profiles with band edge reaching 950 nm. When blended with an electron donor polymer PTB7‐Th, IOTIC‐2F exhibits efficient charge transfer even with a small energetic offset, so as to achieve a large photogenerated current over 22 mA cm−2 with small energy losses (≈0.49 eV) in solar cell devices. With an intense NIR absorbance, PTB7‐Th:IOTIC‐2F‐based cells achieve a power conversion efficiency of 12.1% with good visible transparency (52% transmittance from 370 to 740 nm). Analysis of film morphology reveals that processing with solvent additives enhances crystalline features of acceptor components, while keeping an appropriate level of donor:acceptor intermixing in the binary blends. The incorporation of the third component, ITIC‐2F, into the PTB7‐Th:IOTIC‐2F blends increases the device efficiency up to 12.9%. The improvement is assigned to the cascaded energy‐level structure and desirable nanoscale phase separation of the ternary blends, which is beneficial to the photocurrent generation. Abstract: Two narrow bandgap non‐fullerene acceptors (NBG‐NFAs), namely, COTIC‐4F and SiOTIC‐4F, are designed and synthesized for the fabrication of efficient near‐infrared organic solar cells (OSCs). The chemical structures of the NBG‐NFAs contain a D′‐D‐D′ electron‐rich internal core based on a cyclopentadithiophene (or dithienosilole) (D) and alkoxythienyl (D′) core, end‐capped with the highly electron‐deficient unit 2‐(5,6‐difluoro‐3‐oxo‐2,3‐dihydro‐1H‐inden‐1‐ylidene)malononitrile (A), ultimately providing a A‐D′‐D‐D′‐A molecular configuration that enhances the intramolecular charge transfer characteristics of the excited states. One can thereby reduce the optical bandgap (Egopt) to as low as ≈1.10 eV, one of the smallest values for NFAs reported to date. In bulk‐heterojunction (BHJ) OSCs, NBG‐NFA blends with the polymer donor PTB7‐Th yield power conversion efficiencies (PCE) of up to 9.0%, which is particularly high when compared against a range of NBG BHJ blends. Most significantly, it is found that, despite the small energy loss (Egopt − eVOC) of 0.52 eV, the PTB7‐Th/NBG‐NFA bulk heterojunction blends can yield short‐circuit current densities of up to 22.8 mA cm−2, suggesting that the design and application of NBG‐NFA materials have substantial potential to further improve the PCE of OSCs. Abstract: DNA origami can provide programmed information to guide the self‐assembly of gold nanospheres (Au NSs) into higher‐order supracolloids. Molecularly precise and truly 2D/3D integration of Au NSs is possible using DNA origami‐enabled assembly, and the resulting assemblies have potential applications in plasmonics and metamaterials. However, the relatively small size (<60 nm) and randomly faceted Au NSs that have been used thus far in DNA origami‐enabled assembly have limited their nanophotonic applications. Here, the robust self‐assembly of the 60–100 nm roundest Au NSs into metamolecular assemblies using 3D DNA origami is described. These Au NSs are successfully conjugated with DNA oligonucleotides and are therefore stable at high salt concentrations even without backfilling using organic ligands. The roundest Au NSs are successfully assembled into supracolloidal metamolecules and chains via 3D DNA origami. These plasmonic metamolecules and chains display strong electric and unnatural magnetic resonances that can be deterministically controlled. [ABSTRACT FROM AUTHOR]

This study examined associations among Head Start attendance, individualized education programmes (IEPs), parental behaviours and child outcomes in a sample of five-hundred and seventy 3- to 4-year-old children with disabilities. Home language, number of disabilities and Head Start enrollment were associated with having an IEP. Parents of children with IEPs and those who participated in Head Start used more social services, while social support was more prevalent for parents of non-Head Start children with IEPs. For all children, frequent parental book reading, greater number of books in the home and greater perceived social support among parents were associated with favourable cognitive and social-emotional outcomes. Greater social service use was adversely associated with reading scores only for non-Head Start children. Findings highlight the need for inclusive IEP policies and Head Start programmes for parents regarding access to special education supports for children who demonstrate developmental concerns. Participation in Head Start may buffer negative effects of social service use on children's reading skill development, although more research is needed to uncover the specific mechanisms responsible for this association. Interfacial energetics determines the performance of organic photovoltaic (OPV) cells based on a thin film of organic semiconductor blends. Here, an approach to modulating the 'carrier selectivity' at the charge collecting interfaces and the consequent variations in the nongeminate charge carrier recombination dynamics in OPV devices are demonstrated. A ferroelectric blend interfacial layer composed of a solution-processable ferroelectric poly­mer and a wide bandgap semiconductor is introduced as a tunable electron selective layer in inverted OPV devices with non-Ohmic contact electrodes. The direct rendering of dipole alignment within the ferroelectric blend layer is found to increase the carrier selectivity of the charge collecting interfaces up to two orders of magnitude. Transient photovoltaic analyses reveal that the increase of carrier selectivity significantly reduces the diffusion and recombination among minority carriers in the vicinity of the electrodes, giving rise to the 85% increased charge carrier lifetime. Furthermore, the carrier-selective charge extraction leads to the constitution of the internal potential within the devices, even with energetically identical cathodes and anodes. With these carrier-selectivity-controlled interlayers, the devices based on various photoactive materials commonly display significant increments in the device performances, especially with the high fill factor of up to 0.76 under optimized conditions. Mitochondrial metabolism is highly responsive to nutrient availability and ongoing activity in neuronal circuits. The molecular mechanisms by which brain cells respond to an increase in cellular energy expenditure are largely unknown. Mild mitochondrial uncoupling enhances cellular energy expenditure in mitochondria and can be induced with 2,4-dinitrophenol ( DNP), a proton ionophore previously used for weight loss. We found that DNP treatment reduces mitochondrial membrane potential, increases intracellular Ca2+ levels and reduces oxidative stress in cerebral cortical neurons. Gene expression profiling of the cerebral cortex of DNP-treated mice revealed reprogramming of signaling cascades that included suppression of the mammalian target of rapamycin ( mTOR) and insulin - PI3K - MAPK pathways, and up-regulation of tuberous sclerosis complex 2, a negative regulator of mTOR. Genes encoding proteins involved in autophagy processes were up-regulated in response to DNP. CREB ( cAMP-response element-binding protein) signaling, Arc and brain-derived neurotrophic factor, which play important roles in synaptic plasticity and adaptive cellular stress responses, were up-regulated in response to DNP, and DNP-treated mice exhibited improved performance in a test of learning and memory. Immunoblot analysis verified that key DNP-induced changes in gene expression resulted in corresponding changes at the protein level. Our findings suggest that mild mitochondrial uncoupling triggers an integrated signaling response in brain cells characterized by reprogramming of mTOR and insulin signaling, and up-regulation of pathways involved in adaptive stress responses, molecular waste disposal, and synaptic plasticity. Aging is associated with increased vulnerability to inflammatory challenge. However, the effects of altered inflammatory response on the metabolic status of tissues or organs are not well documented. In this study, we present evidence demonstrating that lipopolysaccharide ( LPS)-induced upregulation of the inflammasome/ IL-1β pathway is accompanied with an increased inflammatory response and abnormal lipid accumulation in livers of aged rats. To monitor the effects of aging on LPS-induced inflammation, we administered LPS (2 mg kg−1) to young (6-month old) and aged (24-month old) rats and found abnormal lipid metabolism in only aged rats with increased lipid accumulation in the liver. This lipid accumulation in the liver was due to the dysregulation of PPARα and SREBP1c. We also observed severe liver inflammation in aged rats as indicated by increased ALT levels in serum and increased Kupffer cells in the liver. Importantly, among many inflammation-associated factors, the aged rat liver showed chronically increased IL-1β production. Increased levels of IL-1β were caused by the upregulation of caspase-1 activity and inflammasome activation. In vitro studies with HepG2 cells demonstrated that treatment with IL-1β significantly induced lipid accumulation in hepatocytes through the regulation of PPARα and SREBP1c. In summary, we demonstrated that LPS-induced liver inflammation and lipid accumulation were associated with a chronically overactive inflammasome/ IL-1β pathway in aged rat livers. Based on the present findings, we propose a mechanism of aging-associated progression of steatohepatitis induced by endotoxin, delineating a pathogenic role of the inflammasome/ IL-1β pathway involved in lipid accumulation in the liver. A series of four polymers containing benzo[1,2-b:4,5-b′]dithiophene (BDT) and 5,6-difluoro-4,7-diiodobenzo[c][1,2,5]thiadiazole (2FBT), PBDT2FBT, PBDT2FBT-O, PBDT2FBT-T, and PBDT2FBT-T-O, are synthesized with their four different side chains, alkyl-, alkoxy-, alkylthienyl-, and alkoxythienyl. Experimental results and theoretical calculations show that the molecular tuning of the side chains simultaneously influences the solubilities, energy levels, light absorption, surface tension, and intermolecular packing of the resulting polymers by altering their molecular coplanarity and electron affinity. The polymer solar cell (PSC) based on a blend of PBDT2FBT-T/[6,6]-phenyl-C71-butyric acid methyl ester (PC71BM) exhibits the best photovoltaic performance of the four PBDT2FBT derivatives, with a high open-circuit voltage of 0.98 V and a power conversion efficiency of 6.37%, without any processing additives, post-treatments, or optical spacers. Furthermore, PBDT2FBT-T-O, which has a novel side chain alkoxythienyl, showed promising properties with the most red-shifted absorption and strong intermolecular packing property in solid state. This study provides insight into molecular design and fabrication strategies via structural tuning of the side chains of conjugated polymers for achieving highly efficient PSCs. Background The electrophysiological inhibitory mechanism of cognitive control for alcohol remains largely unknown. The purpose of the study was to compare electroencephalogram (EEG) power spectra and cross-frequency phase-amplitude coupling ( CFPAC) at rest and during a simple subtraction task after acute alcohol ingestion. Methods Twenty-one healthy subjects participated in this study. Two experiments were performed 1 week apart, and the order of the experiments was randomly assigned to each subject. During the experiments, each subject was provided with orange juice containing alcohol or orange juice only. We recorded EEG activity and analyzed power spectra and CFPAC data. Results The results showed prominent theta-phase gamma-amplitude coupling at the frontal and parietal electrodes at rest. This effect was significantly reduced after alcohol ingestion. Conclusions Our findings suggest that theta-phase gamma-amplitude coupling is deficiently synchronized at rest after alcohol ingestion. Therefore, cross-frequency coupling could be a useful tool for studying the effects of alcohol on the brain and investigating alcohol addiction. [ABSTRACT FROM AUTHOR]

Background and Purpose: Whole-brain irradiation (WBI) therapy produces learning and memory deficits in patients with brain tumours. Although the pathological cascade of cognitive deficits remains unknown, it may involve reduced neurogenesis within the hippocampus. Baicalein is a flavonoid derived from the roots of Huangqin, Scutellaria baicalensis Georgi, and has been shown to have antioxidant effects. Here, we have investigated the protective effects of baicalein on irradiation-induced impairments in hippocampal neurogenesis and cognitive function.Experimental Approach: Radioprotective effects of baicalein were evaluated in C17.2 neural progenitor cells and 6-week-old male C57BL/6 mice during hippocampal neurogenesis. Mice were given a single dose of 5 Gy WBI. Changes in hippocampal neurogenesis, oxidative stress and BDNF-pCREB signalling were evaluated. Morris water maze and passive avoidance test were used to assess learning and memory.Key Results: Baicalein protected neural progenitor cells against irradiation-induced necrotic cell death. Pretreatment with baicalein attenuated the irradiation-induced impairment of hippocampal neurogenesis by modulating oxidative stress and elevating BDNF-pCREB signalling. Furthermore, baicalein prevented the spatial learning and memory retention deficits follwing WBI.Conclusions and Implications: Our findings suggest that baicalein can be viewed as a potential therapeutic agent that protects against the impaired neurogenesis induced by WBI, and its neurocognitive consequences. Hypoxic microenvironment plays an important role in determining stem cell fates. However, it is controversial to which direction between self-renewal and differentiation the hypoxia drives the stem cells. Here, we investigated whether a short exposure to hypoxia (termed 'hypoxic-priming') efficiently directed and promoted mouse embryonic stem cells (mESCs) to differentiate into vascular-lineage. During spontaneous differentiation of embryoid bodies (EBs), hypoxic region was observed inside EB spheroids even under normoxic conditions. Indeed, hypoxia-primed EBs more efficiently differentiated into cells of vascular-lineage, than normoxic EBs did. We found that hypoxia suppressed Oct4 expression via direct binding of HIF-1 to reverse hypoxia-responsive elements (rHREs) in the Oct4 promoter. Furthermore, vascular endothelial growth factor (VEGF) was highly upregulated in hypoxia-primed EBs, which differentiated towards endothelial cells in the absence of exogenous VEGF. Interestingly, this differentiation was abolished by the HIF-1 or VEGF blocking. In vivo transplantation of hypoxia-primed EBs into mice ischemic limb elicited enhanced vessel differentiation. Collectively, our findings identify that hypoxia enhanced ESC differentiation by HIF-1-mediated inverse regulation of Oct4 and VEGF, which is a novel pathway to promote vascular-lineage differentiation. The cause of elevated level of amyloid β-peptide (Aβ42) in common late-onset sporadic [Alzheimer's disease (AD)] has not been established. Here, we show that the membrane lipid peroxidation product 4-hydroxynonenal (HNE) is associated with amyloid and neurodegenerative pathologies in AD and that it enhances γ-secretase activity and Aβ42 production in neurons. The γ-secretase substrate receptor, nicastrin, was found to be modified by HNE in cultured neurons and in brain specimens from patients with AD, in which HNE-nicastrin levels were found to be correlated with increased γ-secretase activity and Aβ plaque burden. Furthermore, HNE modification of nicastrin enhanced its binding to the γ-secretase substrate, amyloid precursor protein (APP) C99. In addition, the stimulation of γ-secretase activity and Aβ42 production by HNE were blocked by an HNE-scavenging histidine analog in a 3xTgAD mouse model of AD. These findings suggest a specific molecular mechanism by which oxidative stress increases Aβ42 production in AD and identify HNE as a novel therapeutic target upstream of the γ-secretase cleavage of APP. We used two-wave panel data based on a sample of 137 employees from a small Korean manufacturing company to examine first the discriminant validity of, and then the interaction between perceived organizational support (POS) and perceptions of organizational politics (POP). We focused on the relationships between POS, POP, and three employee work-related outcomes, affective organizational commitment, intention to stay, and individually oriented organizational citizenship behaviour (OCBI). Confirmatory factor analysis revealed that POS and POP were distinct constructs. Moreover, time-lagged LISREL estimates showed that POS and POP were differentially related to the three outcomes in a theoretically predictable way. POP did not moderate the relationship between POS and either affective commitment or intention to stay. POS, however, significantly moderated the POP-OCBI relationship. Adult neural stem cells in the dentate gyrus of the hippocampus are negatively and positively regulated by a broad range of environmental stimuli that include aging, stress, social interaction, physical activity, and dietary modulation. Interestingly, dietary regulation has a distinct outcome, such that reduced dietary intake enhances neurogenesis, whereas excess calorie intake by a high-fat diet has a negative effect. As a type of metabolic stress, dietary restriction (DR) is also known to extend life span and increase resistance to age-related neurodegenerative diseases. However, the potential application of DR as a 'neurogenic enhancer' in humans remains problematic because of the severity of restriction and the protracted duration of the treatment required. Therefore, the authors consider that an understanding of the neurogenic mechanisms of DR would provide a basis for the identification of the pharmacological and nutraceutical interventions that mimic the beneficial effects of DR without limiting caloric intake. Research on the relationship between perceived organizational support (POS) and affective organizational commitment (AC) has primarily adopted a social exchange perspective. In this study we considered complementary socio-emotional explanations of the POS–AC relationship. We focused on the mediating role of organization-based self-esteem (OBSE) and tested competing models of the POS–OBSE-AC relationship separately on data from two Korean banks that experienced different levels of downsizing following the 1997 Korean financial crisis. We further extended the analysis by examining the extent to which the relationship between POS and OBSE and AC, respectively, was affected by employees' perceived sense of job insecurity in the two banks. The results showed that OBSE was a significant mediator of the POS–AC relationship in both organizations. POS, however, also retained a strong independent direct effect on commitment. Moreover, as expected, perceived job insecurity tended to attenuate the POS–OBSE relationship, but augmented the direct relationship between perceived organizational support and affective commitment. Lysophosphatidylcholine induces expression of adhesion molecules; however, the underlying molecular mechanisms of this are not well elucidated. In this study, the intracellular signaling by which lysophosphatidylcholine upregulates vascular cell adhesion molecule-1 and P-selectin was delineated using YPEN-1 and HEK293T cells. The results showed that lysophosphatidylcholine dose-dependently induced expression of vascular cell adhesion molecule-1 and P-selectin, accompanied by the activation of transcription factor nuclear factor κB. However, the nuclear factor κB inhibitor caffeic acid phenethyl ester (CAPE) and the antioxidant N-acetylcysteine only partially blocked lysophosphatidylcholine-induced adhesion molecules. Subsequently, we found that the lysophosphatidylcholine receptor G protein-coupled receptor 4 (GPK4) was expressed in YPEN-1 cells and triggered the cAMP/protein kinase A/cAMP response element-binding protein pathway, resulting in upregulation of adhesion molecules. Further evidence showed that overexpression of human GPK4 enhanced lysophosphatidylcholine-induced expression of adhesion molecules in YPEN-1 cells, and enabled HEK293T cells to express adhesion molecules in response to lysophosphatidylcholine. In conclusion, the current study suggested two pathways by which lysophosphatidylcholine regulates the expression of adhesion molecules, the lysophosphatidylcholine/nuclear factor-κB/adhesion molecule and lysophosphatidylcholine/GPK4/cAMP/protein kinase A/cAMP response element-binding protein/adhesion molecule pathways, emphasizing the importance of the lysophosphatidylcholine receptor in regulating endothelial cell function. Clip-domain serine proteases (SPs) have been identified in invertebrates as crucial enzymes that are involved in diverse extracellular signalling pathways. Prophenoloxidase (proPO) activating factor-I (PPAF-I), a catalytically active clip-domain SP, cleaves proPO. To date, no crystal structures of a catalytically active clip-domain SP have been determined. Here, the results of crystallization and preliminary X-ray analysis of the SP domain of PPAF-I are reported. The crystal of the PPAF-I SP domain was obtained using the hanging-drop vapour-diffusion method in a precipitant solution containing 0.15 M lithium sulfate, 30% polyethylene glycol 4000 and 0.1 M Tris–HCl pH 8.0. The crystal diffracts X-rays to 1.7 Å resolution using a synchrotron-radiation source. The crystal belongs to space group P212121, with one molecule in the asymmetric unit and unit-cell parameters a = 38.3, b = 53.3, c = 116.6 Å, α = β = γ = 90°. Titanium oxide is the most widely used material for photocatalytic applications due to its low cost and environmental friendliness. One of the grand challenges to improve its energy conversion efficiency is to utilize more visible light while inhibiting the recombination of photogenerated electrons and holes. A one‐step hydrosolvothermal method is used to obtain colored ultrafine nanowires of rutile and nanoparticles of anatase with edge dislocations, which induce broadened visible solar absorption (400–900 nm) and improve photocatalytic efficiency up to 1.8 times that of rutile or anatase without defects. Enhanced photocatalytic activity of these structures is demonstrated by photodegrading methylene blue measurements under simulated solar light irradiation. Results show the existence of Ti3+, induced by edge dislocations, and subsequent electronic band structure–property relationships. This work highlights a strategy for generating sufficient desired defects in TiO2 nanostructures, leading to broadened visible solar absorption and improved photocatalytic efficiency under visible light irradiation. In this study, a comprehensive analytical model to quantify the total nongeminate recombination losses, originating from bimolecular as well as bulk and surface trap‐assisted recombination mechanisms in nonfullerene‐based bulk heterojunction organic solar cells is developed. This proposed model is successfully employed to obtain the different contributions to the recombination current of the investigated solar cells under different illumination intensities. Additionally, the model quantitatively describes the experimentally measured open‐circuit voltage versus light intensity dependence. Most importantly, it is possible to calculate the experimental results with the same fitting parameter values from the presented model. The validity of this model is also proven by a combination of other independent, steady‐state, and transient experimental techniques. This new powerful analytical tool will enable researchers in the photovoltaic community to take into account the synergetic contribution from all relevant types of nongeminate recombination losses in different optoelectronic systems and target their analysis of recombination dynamics at any operating voltage.
