Background: Chimeric antigen receptor T cell (CAR-T) therapy stands as a precise and targeted approach in the treatment of malignancies. In this study, we investigated the feasibility of targeting Cadherin 17 (CDH17) with CDH17 CAR-T cells as a therapeutic modality for small cell lung cancer (SCLC). Methods: CDH17 expression levels were assessed in human SCLC tumor tissues and cell lines using qPCR and Western blot. Subsequently, we established CDH17 CAR-T cells and assessed their cytotoxicity by co-culturing them with various SCLC cell lines at different effector-to-target (E:T) ratios, complemented by ELISA assays. To ascertain the specificity of CDH17 CAR-T cells, we conducted experiments on SCLC cells with and without CDH17 expression (shRNAs). Furthermore, we employed an SCLC xenograft model to evaluate the in vivo efficacy of CDH17 CAR-T cells. Results: Our results revealed a significant upregulation of CDH17 in both SCLC tissues and cell lines. CDH17 CAR-T cells exhibited robust cytotoxic activity against SCLC cells in vitro, while demonstrating no cytotoxicity towards CDH17-deficient SCLC cells and HEK293 cells that lack CDH17 expression. Importantly, the production of IFN-γ and TNF-α by CDH17 CAR-T cells correlated with their cytotoxic potency. Additionally, treatment with CDH17 CAR-T cells significantly decelerated the growth rate of SCLC-derived xenograft tumors in vivo. Remarkably, no significant difference in body weight was observed between the control group and the group treated with CDH17 CAR-T cells. To explore the relationship between physical exercise and smoking behavior among Chinese residents aged 16 years and older. Analysis based on 29,466 validated cases in the 2018 China Family Panel Studies (CFPS 2018). The chi-square test and Mann–Whitney U test were used for comparative analysis between groups. Logistic regression analysis was used to explore the relationship between physical exercise and smoking behavior. Gender and birth cohort differences in the relationship between physical exercise and smoking behavior were explored based on stratified regression analysis using gender and birth cohort as stratified variables, respectively. Robustness testing based on multiple linear regression analysis using a replacement data approach. There were 8735 cases of smokers among the respondents. After controlling for relevant confounders, there was a significant negative association between physical exercise and smoking behavior among residents [OR 0.718, 95% CI (0.673, 0.765), P < 0.01]. Physical exercise was more significantly associated with smoking behavior among male residents [OR 0.694, 95% CI (0.649, 0.743), P < 0.01], while it was not significantly associated with smoking behavior among female residents [OR 0.901, 95% CI (0.743, 1.093), P > 0.05]. Physical exercise was more significantly associated with smoking behavior in the pre-1948 (OR 0.748), 1959–1968 (OR 0.748), 1969–1978 (OR 0.812), 1989–1998 (OR 0.576) and post-1999 (OR 0.411) birth cohorts, and the association decreased over time and with social change. The results of the robustness test showed that frequency of exercise was significantly and negatively associated with smoking behavior among residents [OR 0.961, 95% CI (0.951, 0.970), P < 0.01]. Physical exercise is negatively associated with smoking behavior among Chinese residents aged 16 years and older, especially among male residents. The understanding of the mechanical behavior of granite containing fissures is of great significance for geothermal energy exploitation and high-level nuclear waste disposal repository. To consider the grain interlocking and rotational resistance behavior in granite specimens, the grain-based model 3D (GBM3D) was constructed to investigate the fracture behavior of granite specimens containing a single fissure. The numerical result indicates that this GBM3D can be used to investigate the mechanical behavior of granite constituted by different minerals, and it can provide more information to understand the mechanical behavior of specimens under loading process. The inter-granular cracks initiate in the first place, but the initiation of intra-granular cracks mainly concentrates after the peak strength. The tensile cracks between different minerals are more than that between the same minerals, and the intra-granular cracks in muscovite initiate firstly, and the number of them is more than that in other minerals. Segmental overgrowth of the skeletal muscles with bone involvement in body extremities, predominantly affecting the upper limb, is an extremely rare condition with only 40–50 affected children described clinically. The molecular pathogenesis of this disorder remains largely unclear except for the identification of a somatic PIK3CA mutation in each of the six patients genetically tested, all restricted to upper limbs in the literature. This study aimed to further characterize the molecular defects for patients affected with segmental overgrowth of the skeletal muscles by analyzing a 9-gene panel selected from the PI3K/AKT/mTOR pathway and genes associated with other related conditions. Nineteen unrelated patients were chosen for this study, comprising ten upper limb (nine unilateral and one bilateral) and nine lower limb (eight unilateral and one bilateral) cases with variable bone involvement. In each case, an activating PIK3CA mutation (p.E110del, p.N345K, p.E542K, p.E545K, p.H1047R, or p.H1047L) was identified in the affected muscle tissue with variant allele frequencies ranging from 13.88 to 30.43%, while no mutation was detected in the paired peripheral blood sample, indicating somatic mosaicism. All detected mutations were limited to PIK3CA and were previously reported in other overgrowth syndromes currently categorized under the PIK3CA-Related Overgrowth Spectrum (PROS). Our study provides strong molecular evidence that isolated segmental overgrowth of the skeletal muscle with bone involvement is a subtype of PROS. Our findings expand the PROS clinical presentations with a newly molecularly classified condition and can provide guidance in clinical and molecular diagnosis and treatment for patients with this condition. In this work, we determine up/down-quark mass m q = u / d in the isoscalar scalar channel from both the Shifman–Vainshtein–Zakharov (SVZ) and the Monte-Carlo-based QCD sum rules. The relevant spectral function, including the contributions from the f 0 (500) , f 0 (980) and f 0 (1370) resonances, is determined from a sophisticated U(3) chiral study. Via the traditional SVZ QCD sum rules, we give the prediction to the average light-quark mass m q (2 GeV) = 1 2 (m u (2 GeV) + m d (2 GeV)) = (3. 46 - 0.22 + 0.16 ± 0.33) MeV . Meanwhile, by considering the uncertainties of the input QCD parameters and the spectral functions of the isoscalar scalar channel, we obtain m q (2 GeV) = (3.44 ± 0.14 ± 0.32) MeV from the Monte-Carlo-based QCD sum rules. Granite is a viable host for deep nuclear waste disposal because its low permeability and high strength enable the stable and safe operation of the repository. We examined the evolution of the permeability and triaxial mechanical behaviour of granite after high-temperature treatment. First, the effect of the high temperature on the physical behaviour and permeability evolution of granite was analysed in detail. The mass, P-wave velocity, and thermal conductivity of granite decrease, but the volume increases with increasing temperature. The permeability of intact granite increases by four orders of magnitude as the cycled temperature increases from 25 to 800 °C. Subsequently, the effect of high temperature on the triaxial deformation and acoustic emission (AE) behaviour of granite was investigated. Under uniaxial compression at T ≤ 300 °C, the stress decreases before the peak strength is reached, corresponding to a significant AE event, which is due to the development of multiple splitting tensile fractures along the loading direction. At T ≥ 450 °C, AE event is observed once a minor stress is applied, which results from failure is controlled by thermally induced cracks. However, under triaxial compression, the temperature has little effect on the AE characteristics. The granite fails along the shear fracture plane, which becomes wider with increasing confining pressure. At T ≥ 600 °C, it is easier to form intragranular cracks and the stress quickly decreases after the peak strength is reached. The shear plane is smoother under high confining pressure. Third, the effect of high temperature on the peak strength and crack damage threshold of granite was further analysed. Generally, under uniaxial compression, the peak strength and crack damage threshold first remain relatively constant at T ≤ 300 °C, begin to decrease at T = 450 °C, and decrease more rapidly at T = 600 °C. The confining pressure notably reduces the effect of the temperature on the peak strength and crack damage threshold. Finally, the effect and mechanism of high temperature on the triaxial strength parameters of granite were further discussed. At T ≤ 300 °C, thermally induced cracks are not notable and the temperature has little effect on the strength. At 450 °C≤ T ≤ 600 °C, thermally induced cracks are more notable and the temperature has a significant effect on the strength behaviour. Understanding thermo-mechanical properties of granites with different grain sizes is of great significance for the site selection of high-level radioactive waste disposal. In this paper, a cluster model was used to investigate the uniaxial compressive behaviors of granite specimens with different grain sizes. It turns out the results of simulation match experimental results. Grain size has a significant effect on the mechanical behaviors of granite. Macro-cracks can be easily observed in the coarse granite specimens after high-temperature treatment. However, the number of inter-cracks in the granite specimens decreased with increasing the grain sizes. When T ≥ 600 °C, main cracks were not observed in the compressed specimens, and the ultimate failure modes were mostly dominated by thermally induced cracks. The isolated grains can be observed in the coarse granite specimens. The reduction in the uniaxial compressive strength of the coarse-grained granite specimens was lower than that for the fine-grained granite specimens. [ABSTRACT FROM AUTHOR]

A variety of researches on boron oxide clusters have indicated the key role of boronyl (BO) group in the structures and bonding. Based upon global structural searches and electronic structure calculations at the B3LYP and single-point coupled cluster single double (triple) (CCSD(T)) levels, we present the possibility of construction of lithium-doped boron oxide B8O2Li+/0 clusters (1-2). Different from the structures of pure B6+/0/− and B6(BO)20/−, the B8O2Li+/0 which can be formulated as B6(BO)2Li+/0 are not the double-chain structures, they are the crown-like structure, and the Li is like a diamond that links the crown. Detailed AdNDP analyses indicate the π aromaticity of B8O2Li+ (1). The results obtained in this work reveal that the metal could influence the structures and properties of boron oxides significantly. Janus polymerization is featured as a combination of cationic and anionic growing ends in one living polymer chain. In the copolymerization of THF and CL catalyzed by lutetium triflates and initiated by propylene oxide, three stages are identified by kinetic study including (1) fast cationic polymerization with slow anionic one, (2) fast anionic polymerization with dormant cationic one, and (3) reactivation of cationic polymerization with coupling of anionic and cationic chain ends. In this work, density functional theory (DFT) calculation is employed to investigate the reaction details of ionic polymerization and dormancy. A "tripedal crow" configuration is proposed to illustrate the unique high-coordinated ligand exchange configuration in anionic polymerization in different stages. Administration of CHK1-targeted anticancer therapies is associated with an increased cumulative risk of cardiac complications, which is further amplified when combined with gemcitabine. However, the underlying mechanisms remain elusive. In this study, we generated hiPSC-CMs and murine models to elucidate the mechanisms underlying CHK1 inhibition combined with gemcitabine-induced cardiotoxicity and identify potential targets for cardioprotection. Mice were intraperitoneally injected with 25 mg/kg CHK1 inhibitor AZD7762 and 20 mg/kg gemcitabine for 3 weeks. hiPSC-CMs and NMCMs were incubated with 0.5 uM AZD7762 and 0.1 uM gemcitabine for 24 h. Both pharmacological inhibition or genetic deletion of CHK1 and administration of gemcitabine induced mtROS overproduction and pyroptosis in cardiomyocytes by disrupting mitochondrial respiration, ultimately causing heart atrophy and cardiac dysfunction in mice. These toxic effects were further exacerbated with combination administration. Using mitochondria-targeting sequence-directed vectors to overexpress CHK1 in cardiomyocyte (CM) mitochondria, we identified the localization of CHK1 in CM mitochondria and its crucial role in maintaining mitochondrial redox homeostasis for the first time. Mitochondrial CHK1 function loss mediated the cardiotoxicity induced by AZD7762 and CHK1-knockout. Mechanistically, mitochondrial CHK1 directly phosphorylates SIRT3 and promotes its expression within mitochondria. On the contrary, both AZD7762 or CHK1-knockout and gemcitabine decreased mitochondrial SIRT3 abundance, thus resulting in respiration dysfunction. Further hiPSC-CMs and mice experiments demonstrated that SIRT3 overexpression maintained mitochondrial function while alleviating CM pyroptosis, and thereby improving mice cardiac function. In summary, our results suggest that targeting SIRT3 could represent a novel therapeutic approach for clinical prevention and treatment of cardiotoxicity induced by CHK1 inhibition and gemcitabine. In tunnel engineering, due to the effect of excavation disturbance, the surrounding rock mass can produce an excavation damage zone with different damage extents. Therefore, knowledge of rock deformation and damage behavior is especially significant for the design of deep tunnel support. However to date, a few experiments and numerical simulations have been conducted to investigate the deformation and mechanical failure behavior of damaged rocks. Therefore, in this research, multi-stage triaxial compression test was used to investigate the mechanical behavior of mudstone specimens with different damage extents by experiment and two-dimensional particle flow code. First, a group of micro-parameters was calibrated by single-stage triaxial compression experiments of mudstone, and the numerical results agree very well with the experimental results. Then, multi-stage triaxial compression experiment and discrete element modeling of mudstone specimens were carried out. The more axial strain the specimens sustained, the less strength they had (because the degree of damage increased). A damage variable was defined by the ratio of the area of micro-cracks to the total area of the specimen. As the post-stress reducing ratio increases, the damage variable increases rapidly until the post-stress reducing ratio reaches 0.4; then, it remains constant. Abstract Granite, an excellent medium for deep geological disposal projects, geothermal systems and geological carbon storage, may be affected by its grain size and pre-existing flaws. Thus, three types of granite specimens with different grain sizes containing two non-coplanar fissures under uniaxial compression were investigated experimentally and numerically. Notably, the ligament angle had more effect on the peak strength than the grain size. With an increasing ligament angle, the angle between the crack and the principal stress did not show an obvious trend for β ≤ 60°, whereas the angle between the crack and the principal stress increased when β ≥ 60°. This trend is similar to that for the peak strength. For the same ligament angle, the angle between the crack and the principal stress increased as the grain size decreased. Based on the micro-structure of the crack surface in the ligament (β = 60°), the hypothesis that a shear crack is more difficult to initiate in coarse granite specimens than in fine granite specimens was presented and verified by experiments and numerical simulations. Introduction: Impaired mitochondrial function is a key factor attributing to the lung ischemia reperfusion injury (LIRI). Methylene blue (MB) has been reported to attenuate brain and renal ischemia–reperfusion injury. We hypothesized that MB also could have a protective effect against LIRI by preventing mitochondrial oxidative damage.Methods: Isolated rat lungs were assigned to the following four groups (n = 6): a sham group: perfusion for 105 min without ischemia; I/R group: shutoff of perfusion and ventilation for 45 min followed by reperfusion for 60 min; and I/R + MB group and I/R + glutathione (GSH) group: 2 mg/kg MB or 4 μM glutathione were intraperitoneally administered for 2 h, and followed by 45 min of ischemia and 60 min of reperfusion.Results: MB lessened pulmonary dysfunction and severe histological injury induced by ischemia–reperfusion injury. MB reduced the production of reactive oxygen species and malondialdehyde and enhanced the activity of superoxide dismutase. MB also suppressed the opening of the mitochondrial permeability transition pore and partly preserved mitochondrial membrane potential. Moreover, MB inhibited the release of cytochrome c from the mitochondria into the cytosol and decreased apoptosis. Additionally, MB downregulated the mRNA expression levels of pro-inflammatory cytokines (TNF-α, IL-1β, IL-6, and IL-18).Conclusion: MB protects the isolated rat lungs against ischemia–reperfusion injury by attenuating mitochondrial damage. In this paper, a cluster model in particle flow code was used to simulate granite specimens after heat treatment under uniaxial compression. The results demonstrated that micro-cracks are randomly distributed in the specimen when the temperature is below 300 ∘C, and have partial coalescence when the temperature is up to 450 ∘C, then form macro-cracks when the temperature is above 600 ∘C. There is more inter-granular cracking than intra-granular cracking, and their ratio increases with increasing temperature. The micro-cracks are almost constant when the temperature decreases from 900 ∘C to room temperature, except for quartz α-β phase transition temperature (573 ∘C). The fracture evolution process is obviously affected by these cracks, especially at 600-900 ∘C. Elevated temperature leads to easily developed displacement between the grains, and the capacity to store strain energy becomes weaker, corresponding to the plasticity of granite after heat treatment. The deformation failure characteristics of marble subjected to triaxial cyclic loading are significant when evaluating the stability and safety of deep excavation damage zones. To date, however, there have been notably few triaxial experimental studies on marble under triaxial cyclic loading. Therefore, in this research, a series of triaxial cyclic tests was conducted to analyze the mechanical damage characteristics of a marble. The post-peak deformation of the marble changed gradually from strain softening to strain hardening as the confining pressure increased from 0 to 10 MPa. Under uniaxial compression, marble specimens showed brittle failure characteristics with a number axial splitting tensile cracks; in the range of σ = 2.5-7.5 MPa, the marble specimens assumed single shear fracture characteristics with larger fracture angles of about 65°. However, at σ = 10 MPa, the marble specimens showed no obvious shear fracture surfaces. The triaxial cyclic experimental results indicate that in the range of the tested confining pressures, the triaxial strengths of the marble specimens under cyclic loading were approximately equal to those under monotonic loading. With the increase in cycle number, the elastic strains of the marble specimens all increased at first and later decreased, achieving maximum values, but the plastic strains of the marble specimens increased nonlinearly. To evaluate quantitatively the damage extent of the marble under triaxial cyclic loading, a damage variable is defined according to the irreversible deformation for each cycle. The evolutions of the elastic modulus for the marble were characterized by four stages: material strengthening, material degradation, material failure and structure slippage. Based on the experimental results of the marble specimens under complex cyclic loading, the cohesion of the marble decreased linearly, but the internal friction angles did not depend on the damage extent. To describe the peak strength characteristics of the marble specimens under complex cyclic loadings with various deformation positions, a revised strength criterion for damaged rocks is offered. A thermally triggered shape memory polymer composite was prepared by blending short carbon fiber (SCF) into a blend of poly(styrene- b-butadiene- b-styrene) triblock copolymer (SBS)/linear low density polyethylene (LLDPE) prior to curing. These composites have excellent processability compared with other thermosets. The dynamic mechanical analysis (DMA) and differential scanning calorimetry (DSC) were investigated to assess the thermomechanical properties of the SCF/SBS/LLDPE composite. Scanning electron microscope (SEM) imaging of the samples was performed to show the distribution of the SCF in the composite. The study specifically focused on the effect of SCF on the shape memory behavior of the SCF/SBS/LLDPE composite. The results indicated that the large amount of SCF significantly improved the mechanical property of the polymer composites while not damaging the shape memory performance. The SCF/SBS/LLDPE composites exhibited excellent shape memory behavior when the SCF content was less than 15.0 wt%. To understand the fracture mechanism in all kinds of rock engineering, it is important to investigate the fracture evolution behavior of pre-fissured rock. In this research, we conducted uniaxial compression experiments to evaluate the influence of ligament angle on the strength, deformability, and fracture coalescence behavior of rectangular prismatic specimens (80 × 160 × 30 mm) of brittle sandstone containing two non-coplanar fissures. The experimental results show that the peak strength of sandstone containing two non-coplanar fissures depends on the ligament angle, but the elastic modulus is not closely related to the ligament angle. With the increase of ligament angle, the peak strength decreased at a ligament angle of 60°, before increasing up to our maximum ligament angle of 120°. Crack initiation, propagation, and coalescence were all observed and characterized from the inner and outer tips of pre-existing non-coplanar fissures using photographic monitoring. Based on the results, the sequence of crack evolution in sandstone containing two non-coplanar fissures was analyzed in detail. In order to fully understand the crack evolution mechanism of brittle sandstone, numerical simulations using PFC were performed for specimens containing two non-coplanar fissures under uniaxial compression. The results are in good agreement with the experimental results. By analyzing the stress field, the crack evolution mechanism in brittle sandstone containing two non-coplanar fissures under uniaxial compression is revealed. Four water-soluble dia(m)mine platinum complexes with 3-oxo-cyclobutane- 1,1-dicarboxylate as the leaving group have been synthesized. These compounds were evaluated for their in vitro anticancer activity against three human A549, SK-OV-3, and HT-29 cancer cell lines. All the tested compounds showed potent activity in lung carcinoma (A549), ovarian carcinoma (SK-OV-3), and colon cancer cell (HT-29) lines. Complex 2(IC value = 1.57 µM for A549 and IC value = 0.88 µM for HT-29 cells) and Complex 4 (IC value = 15.3 µM for SK-OV-3 cells) displayed the most potent anticancer activity of the series. Also, preliminary acute toxicity of the platinum complexes derivatives has shown the same level as carboplatin and lower toxicity than cisplatin in vivo. A new xanthone, 2,8-dihydroxy-9-oxo-9H-xanthene-6-carboxylic acid (1), together with six known compounds, 2,8-dihydroxy-1-methoxycarbonyl-9-oxo-9H-xanthene-6-carboxylic acid (2), 8-hydroxy-3-methyl-9-oxo-9Hxanthene-1-carboxylic acid methyl ether (3), 1,3,6-trihydroxy-8-methylxanthone (4), 1,6-dihydroxy-3-methoxy-8-methylxanthone (5), decarboxycitrinone (6), and formoic acid A (7), were isolated from an endophytic fungus Arthrinium arundinis of Anoectochilus roxburghii. Symbiotic microbiomes of Sphagnum have been confirmed to play a fundamental role in carbon and nitrogen cycles, however, little is known about microbiomes associated with other bryophytes in subtropical peatland ecosystems. To explore the differences in community structure, metabolic potential and interaction relationship of bacterial microbiomes associated with different bryophytes species, the gametophytes of three bryophyte species (Sphagnum palustre, Aulacomnium androgynum, and Polytrichum commune) and their underlying peat sediments were collected from the subtropical Dajiuhu Peatland and subjected to Illumina high-throughout sequencing of 16S rRNA gene. Results showed that bacterial diversity was lowest in S. palustre, the dominant moss species, among the three moss species investigated in Dajiuhu Peatland. Bacterial communities from bryophytes clearly separated with those from sediments as indicated by both phylogenetic and taxonomical approaches. Linear discriminant analysis effect size (LEfSe) identified 30 and 36 indicator taxa in mosses and peat sediments. Bacteroidetes, Verrucomicrobia and Thermoleophilia significantly enriched in S. palustre, A. androgynum and P. commune, Chloroflexi, Proteobacteria and Acidobacteria subgroup 6 was indicator taxa for corresponding underlying sediments, respectively. Despite of these differences in compositions, bacterial functional structures were similar among all bryophytes, such as abundant aerobic heterotrophs, rare nitrifiers and denitrifiers. This phenomenon was also observed among the underlying sediments. Network analysis indicated that Proteobacteria and Acidobacteria located in the center of network and exerted strong interactions to other taxa. The sub-network of bacterial communities in sediments was more connected and microbial groups were more competitive than those in bryophytes subnetwork. Our results offer new insight into the community structure, ecological function and interaction pattern of bacterial microbiomes in the Dajiuhu Peatland across different habitats. Purpose: Chronic granulomatous disease (CGD) is an inherited disorder, with phagocytes failing to produce antimicrobial superoxide due to deficient NADPH oxidase activity. Mutations in the gene encoding CYBB are responsible for the majority of the CGD cases. To date, there have been no reports on large samples of children with CGD in China. Therefore, in this study, we described the clinical and molecular features of 38 suspected CGD patients from 36 unrelated Chinese families. Methods: Clinical diagnosis was performed using dihydrorhodamine assays detected by flow cytometry. Molecular analysis was used to identify underlying CGD-causative genes. Results: The mean age of onset in our 38 patients was 3.4 months, while the mean age at diagnosis was 31.7 months. Apart from recurrent pneumonia and abscesses, tuberculosis (TB) and Bacille Calmette-Guerin (BCG) infections were notable features in our cohort. Overall, 17 cases died and patient 1 did not participate in the follow-up period . In total, we identified 29 different CYBB gene mutations in 31 patients. We found NCF1 and CYBA mutations in 3 and 2 patients, respectively. In addition, we identified 31 carriers and prenatally diagnosed 4 CGD and 4 healthy fetuses. Conclusions: The results of our study demonstrate that children with BCG infections or recurrent TB infections should have immune function screening tests performed. Moreover, newborns with family histories of primary immunodeficiency diseases should avoid of BCG vaccination. The most stable mono-layer boron sheets were predicted to have both the isolated hexagonal hole and the twin-hexagonal hole. Previous investigations indicate that planar B 18H nq ( n = 3–6, q =  n − 4) are the building blocks of boron sheets with isolated hexagonal holes. Extensive DFT investigations performed in this work show that D2h B 26H 8, D2h B 26H 82+, and C2 B 26H 6, may serve as the building blocks of boron sheets with twin-hexagonal holes. These bicyclic clusters possess planar or quasi-planar geometries at B3LYP/6-311+G(d,p) level, with 16, 14, and 14 delocalized π electrons, respectively. Detailed analyses indicate that they are overall aromatic in nature, with the formation of islands of both σ and π aromaticity. They are analogous to D2h C 16H 14 and D2h C 16H 142+ in π bonding patterns, respectively, but fundamentally different from the latter in σ-bonding. Remarkably, all of them appear to be energetically the lowest-lying isomers obtained, which are promising targets for future gas phase syntheses. These hydroboron clusters, together with B 18H nq clusters, establish the molecular basis for modeling the short-range structures, nucleation, and growth processes of monolayer boron sheets. Effects of pulsed electric fields (PEF) on quality characteristics and microbial inactivation of soymilk were studied with different PEF parameters. PEF did not affect significantly the values of pH, 'a' (an indicator of redness ranging from '−a' to '+a', −a = green, +a = red) and electric conductivity. The values of 'L' (white if 'L' = 100, black if 'L' = 0) increased slightly, whereas values of viscosity and 'b' (an indicator of yellowness ranging from '−b' to '+b', −b = blue, +b = yellow) decreased slightly as PEF time increased from 0 to 547 μs. Cysteine, tyrosine, phenylalanine, and serine reduced with the increase of PEF time. The relative activities of soybean lipoxygenase (SLOX) decreased with PEF time increasing from 0 to 1,036 μs. When PEF time and strength increased, the inactivation of Escherichia coli and Staphylococus aureus increased significantly ( p < 0.05), achieving a maximum of 5.20 and 3.51 log cycles reduction at PEF time 547 μs and pulsed electric strength 40 kV/cm, respectively. The inactivation of E. coli, S. aureus, and SLOX as a function PEF time followed Weibull distribution. This study demonstrated that PEF could inactivate efficiently E. coli, S. aureus, and SLOX without affecting the quality characteristics of soymilk. The Vitreoscilla hemoglobin ( VHb) gene was expressed in yeast-like conidia (YLCs) of Tremella fuciformis ( T. fuciformis) to increase cell density in submerged fermentation by enhancing oxygen uptake. With the intention of doing this, an integrated expression vector containing the VHb gene and the hygromycin B phosphotransferase ( hph) gene derived from Escherichia coli ( E. coli) as the selectable marker was constructed, and then transformed into protoplasts of YLCs from T. fuciformis with restriction enzyme-mediated DNA integration (REMI). Hygromycin-resistant transformants had been generated during the transformation. Molecular evidences including PCR assay, Southern blotting, and Western blot analysis indicated the VHb gene had been integrated into the genome of transgenic T. fuciformis strains and was expressed successfully. Shake-flask fermentation and bioreactor cultivation results showed that the expression of VHb in this fungus could enhance growth of YLCs. The final cell density was higher in the culture of VHb-expressing strain than that of the wild-type strain. Purpose: Ultrasomics is a radiomics technique that extracts high-throughput quantitative data from ultrasound imaging. The aim of this study was to differentiate malignant from benign focal liver lesions (FLLs) using two-dimensional shear wave elastography (2D-SWE)-based ultrasomics. Methods: A total of 175 FLLs in 169 patients were prospectively analyzed. The study population was divided into a training cohort (n = 122) and a validation cohort (n = 53). The maxima, minima, mean, and standard deviation of 2D-SWE measurements were expressed in kilopascals (Emax, Emin, Emean, and ESD). The ultrasonics technique was used to extract the features from the 2D-SWE images. Support vector machine was used to establish two prediction models: the ultrasomics score (ultrasomics features only) and the combined score (SWE measurements and ultrasomics features). The diagnostic performance of the models in differentiating FLLs was analyzed. Results: A total of 1044 features were extracted and 15 features were selected. The AUC for the combined score, ultrasomics score, Emax, Emean, Emin and ESD were 0.94, 0.91, 0.92, 0.89, 0.67, and 0.89, respectively. The combined score had the best diagnostic performance. The sensitivity, specificity, PPV, NPV, +LR, LR of the combined score were 92.59%, 87.50%, 94.59%, 82.50%, 7.35%, and 0.09%, respectively. The decision curve analysis results showed that when the threshold probability was > 29%, the combined score showed improved benefits for patients compared to using the ultrasomics score and 2D-SWE measurements. In this study, a new adaptive morphological filter is developed based on the mathematical morphology algorithm and characteristics of the subtle differences in the waveform morphology in seismic data. The algorithm improves the traditional morphological dilation and corrosion operations. In this study, we propose a multiscale adaptive operator based on the principle of morphological structural "probes" and present the corresponding mathematical proof. Simulation experiments and actual seismic data processing results show that compared with traditional morphological filters, the constructed OCCO-based multistructure adaptive morphological filter can suppress noise to the greatest extent. Motifs of planar metalloborophenes, cage-like metalloborospherenes, and metal-centered double-ring tubular boron species have been reported. Based on extensive first-principles theory calculations, we present herein the possibility of doping the quasi-planar C2v B56 (A-1) with an alkaline-earth metal to produce the penta-ring tubular Ca©B56 (B-1) which is the most stable isomer of the system obtained and can be viewed as the embryo of metal-doped (4,0) boron α-nanotube Ca©BNT(4,0) (C-1). Ca©BNT(4,0) (C-1) can be constructed by rolling up the most stable boron α-sheet and is predicted to be metallic in nature. Detailed bonding analyses show that the highly stable planar C2v B56 (A-1) is the boron analog of circumbiphenyl (C38H16) in π-bonding, while the 3D aromatic C4v Ca©B56 (B-1) possesses a perfect delocalized π system over the σ-skeleton on the tube surface. The IR and Raman spectra of C4v Ca©B56 (B-1) and photoelectron spectrum of its monoanion C4v Ca©B56− are computationally simulated to facilitate their spectroscopic characterizations. Although genome-wide association studies (GWASs) have identified over 100 colorectal cancer (CRC) risk loci, an understanding of causal genes or risk variants and their biological functions in these loci remain unclear. Recently, genomic loci 10q26.12 with lead SNP rs1665650 was identified as an essential CRC risk loci of Asian populations. However, the functional mechanism of this region has not been fully clarified. Here, we applied an RNA interfering-based on-chip approach to screen for the genes essential for cell proliferation in the CRC risk loci 10q26.12. Notably, HSPA12A had the most significant effect among the identified genes and functioned as a crucial oncogene facilitating cell proliferation. Moreover, we conducted an integrative fine-mapping analysis to identify putative casual variants and further explored their association with CRC risk in a large-scale Chinese population consisting of 4054 cases and 4054 controls and also independently validated in 5208 cases and 20,832 controls from the UK biobank cohort. We identified a risk SNP rs7093835 in the intron of HSPA12A that was significantly associated with an increased risk of CRC (OR 1.23, 95% CI 1.08–1.41, P = 1.92 × 10–3). Mechanistically, the risk variant could facilitate an enhancer–promoter interaction mediated by the transcriptional factor (TF) GRHL1 and ultimately upregulate HSPA12A expression, which provides functional evidence to support our population findings. Collectively, our study reveals the important role of HSPA12A in CRC development and illustrates a novel enhancer–promoter interaction module between HSPA12A and its regulatory elements rs7093835, providing new insights into the etiology of CRC. Highlights: Ultrathin P and Fe co-doped NiSe2 nanosheets supported on modified Ni foam are synthesized, which shows desirable bifunctional electrocatalytic hydrogen evolution reaction (HER)/hydrazine oxidation reaction (HzOR) performance in hydrazine-assisted water electrolysis and Zn-Hz battery. Herein, we demonstrate the development of ultrathin P/Fe co-doped NiSe2 nanosheets supported on modified Ni foam (P/Fe-NiSe2) synthesized through a facile electrodeposition process and subsequent heat treatment. Based on electrochemical measurements, characterizations, and density functional theory calculations, a favorable "2 + 2" reaction mechanism with a two-step HER process and a two-step HzOR step was fully proved and the specific effect of P doping on HzOR kinetics was investigated. P/Fe-NiSe2 thus yields an impressive electrocatalytic performance, delivering a high current density of 100 mA cm−2 with potentials of − 168 and 200 mV for HER and HzOR, respectively. The identification of additional Echinococcus granulosus sensu lato (s.l.) complex species/genotypes in recent years raises the possibility that there might be more variation among this species in China than is currently understood. The aim of this study was to explore intra- and inter-species variation and population structure of Echinococcus species isolated from sheep in three areas of Western China. Of the isolates, 317, 322, and 326 were successfully amplified and sequenced for cox1, nad1, and nad5 genes, respectively. BLAST analysis revealed that the majority of the isolates were E. granulosus s.s., and using the cox1, nad1, and nad5 genes, respectively, 17, 14, and 11 isolates corresponded to Elodea canadensis (genotype G6/G7). In the three study areas, G1 genotypes were the most prevalent. There were 233 mutation sites along with 129 parsimony informative sites. A transition/transversion ratio of 7.5, 8, and 3.25, respectively, for cox1, nad1, and nad5 genes was obtained. Every mitochondrial gene had intraspecific variations, which were represented in a star-like network with a major haplotype with observable mutations from other distant and minor haplotypes. The Tajima's D value was significantly negative in all populations, indicating a substantial divergence from neutrality and supporting the demographic expansion of E. granulosus s.s. in the study areas. The phylogeny inferred by the maximum likelihood (ML) method using nucleotide sequences of cox1–nad1–nad5 further confirmed their identity. The nodes assigned to the G1, G3, and G6 clades as well as the reference sequences utilized had maximal posterior probability values (1.00). In conclusion, our study confirms the existence of a significant major haplotype of E. granulosus s.s. where G1 is the predominant genotype causing of CE in both livestock and humans in China. To investigate the role of fissure angle and heat treatment temperature on the mechanical properties and deformation failure behavior, uniaxial compression tests were carried out on granite specimens containing a single fissure. Using stress–strain curves, the peak strength, peak strain, and elastic modulus of the one-fissured granite specimens were analyzed in detail. The mechanical parameters are closely related to the fissure angle and the high temperature. As the fissure angle increases from 0° to 90°, the peak strength and elastic modulus first decrease and then increase, while the peak strain increases slowly. However, the peak strength and elastic modulus first increase and then decrease, while the peak strain first decreases and then increases with increasing treatment temperature. During the experiments, the crack evolution process and acoustic emission (AE) counts were obtained using real-time photography and the AE monitoring technique. In the granite specimens containing a pre-existing fissure, large AE counts are clearly observed before the peak strength, which indicates crack initiation and propagation. The accumulated AE count first increases slowly, but is followed by a sharp increase, with increasing deformation. The AE events in the one-fissured specimen also depend on the heat treatment temperature. As the temperature increases, the rate of increase of the accumulated AE count curve is reduced. Finally, using a digital image correlation method, the full fields of surface deformation were obtained for the entire testing process. In addition, the local strain around the pre-existing fissure was measured using strain gauges. Bathyarchaeota are believed to have roles in the carbon cycle in marine systems. However, the ecological knowledge of Bathyarchaeota is limited in peatland ecosystems. Here, we investigated the vertical distribution of Bathyarchaeota community structure using quantitative PCR and high-throughput sequencing technology of ribosomal 16S rRNA gene integrated with detailed chemical profiling in the Dajiuhu Peatland, central China. Eight archaeal phyla were observed in peat samples, which mainly composed of Bathyarchaeota with a mean relative abundance about 88%, followed by Thaumarchaeota (9%). Bathyarchaeota were further split into 17 subgroups, and some subgroups showed habitat specificity to peat horizons with distinct lithological and physicochemical properties, for example, Bathy-6 and Bathy-15 had preference for the acrotelm, Bathy-5b, Bathy-16, and Bathy-19 were enriched in the catotelm, Bathy-5a, Bathy-8, and Bathy-11 were specific for the clay horizon. This spatial distribution pattern of archaeal communities along peat profile was mainly influenced by water content as indicated by RDA ordination and permutational MANOVA, whereas organic matter content exclusively affected Bathyarchaeota distribution along the peat profile significantly. The abundance of archaeal 16S rRNA genes ranged from 105 to 107 copies per gram dry sediment, and the highest archaeal biomass was observed in the periodically oxic mesotelm horizon with more dynamic archaeal interaction relationship as indicated by the network analysis. Bathyarchaeota dominated the archaeal interaction network with 82% nodes, 96% edges, and 71% keystone species. Our results provide an overview of the archaeal population, community structure, and relationship with environmental factors that affect the vertical distribution of archaeal communities and emphasize the ecology of bathyarchaeotal lineages in terrestrial peatland ecosystems. Introduction: Inflammation is a defensive response of the organism to irritation which is manifested by redness, swelling, heat, pain and dysfunction. The inflammatory response underlies the role of various diseases. Ferroptosis, a unique modality of cell death, driven by iron-dependent lipid peroxidation, is regulated by multifarious cellular metabolic pathways, including redox homeostasis, iron processing and metabolism of lipids, as well as various signaling pathways associated with diseases. A growing body of evidence suggests that ferroptosis is involved in inflammatory response, and targeting ferroptosis has great prospects in preventing and treating inflammatory diseases. Materials and methods: Relevant literatures on ferroptosis, inflammation, inflammatory factors and inflammatory diseases published from January 1, 2010 to now were searched in PubMed database. Conclusion: In this review, we summarize the regulatory mechanisms associated with ferroptosis, discuss the interaction between ferroptosis and inflammation, the role of mitochondria in inflammatory ferroptosis, and the role of targeting ferroptosis in inflammatory diseases. As more and more studies have confirmed the relationship between ferroptosis and inflammation in a wide range of organ damage and degeneration, drug induction and inhibition of ferroptosis has great potential in the treatment of immune and inflammatory diseases. In the "treat all" era, the high rate of late HIV diagnosis (LHD) worldwide remains an impediment to ending the HIV epidemic. In this study, we analyzed LHD in newly diagnosed people living with HIV (PLWH) and its impact on HIV transmission in Northeast China. Sociodemographic information, baseline clinical data, and plasma samples obtained from all newly diagnosed PLWH in Shenyang, the largest city in Northeast China, between 2016 and 2019 were evaluated. Multivariate logistic regression analysis was performed to identify risk factors associated with LHD. A molecular network based on the HIV pol gene was constructed to assess the risk of HIV transmission with LHD. A total of 2882 PLWH, including 882 (30.6%) patients with LHD and 1390 (48.2%) patients with non-LHD, were enrolled. The risk factors for LHD were older age (≥ 30 years: p <.01) and diagnosis in the general population through physical examination (p <.0001). Moreover, the molecular network analysis revealed that the clustering rate (p <.0001), the fraction of individuals with ≥ 4 links (p =.0847), and the fraction of individuals linked to recent HIV infection (p <.0001) for LHD were significantly or marginally significantly lower than those recorded for non-LHD. Our study indicates the major risk factors associated with LHD in Shenyang and their limited contribution to HIV transmission, revealing that the peak of HIV transmission of LHD at diagnosis may have been missed. To understand the high temperature effects on the mechanical and failure behaviors of rock, uniaxial compression tests were carried out on granite specimens containing three pre-existing holes using a rock testing system. Based on the experimental results, the influences of testing temperature on the physical and mechanical parameters of granite were analyzed in detail. An obvious color change of tested granite occurs from gray at room temperature to reddish after 450 °C and to red-brown after 900 °C high temperature treatment. The granite volume increases, mass decreases and density decreases with increasing testing temperature. As the temperature increases, the peak strength first increases and then decreases, while the elastic modulus decreases. However, the peak strain changes slightly before 450 °C, increases dramatically up 450 °C. As the bridge angle increases, the mechanical parameters of granite specimens first decrease and then increase. And then, the crack initiation, propagation and coalescence behavior of granite specimens after high temperature exposure was investigated using an acoustic emission (AE) and photography monitoring technique. The cracking process shows that the propagation of crack from the surface of holes leads to the coalescence between adjacent holes. A large AE count and a stress drop are observed during the crack initiation and propagation. The failure modes can be generally classified into three categories: splitting mode, shear mode and mixed mode and they are closely related to heat treatment temperature and bridge angle. A magnetically separable photocatalyst, the composite compound CoFeO-AgO, was fabricated successfully by a simple precipitation method and used for photodegradation of organic pollutants under visible light (>420 nm) irradiation. Its magnetic separation performance was evaluated. Results showed that the CoFeO-AgO with 60-wt% content of AgO had the best photocatalytic performance, stability, and magnetic separation performance. Methyl orange, methylene blue, rhodamine B, and phenol can be completely photodegraded by the CoFeO-AgO photocatalyst in a short period. After five cycles, CoFeO-AgO kept its performance stability. As prepared, CoFeO-AgO (60%) has a coercivity of 2500 Oe and a saturation magnetization of 22.45 emu g; it can be completely separated magnetically in 20 s with magnetic recovery mass rate of 85% under an external magnetic field. This superb photocatalytic performance and separation recovery confirms that the CoFeO-AgO photocatalyst is a promising candidate for future use in photo-oxidative degradation of organic contaminants. Because of its novel physical properties, two-dimensional materials have attracted great attention. From first-principle calculations and vibration frequencies analysis, we predict a new family of two-dimensional materials based on the idea of octet stability: honeycomb lattices of pnictogens (N, P, As, Sb, Bi). The buckled structures of materials come from the sp3 hybridization. These materials have indirect band gap ranging from 0.43 eV to 3.7 eV. From the analysis of projected density of states, we argue that the s and p orbitals together are sufficient to describe the electronic structure under tight-binding model, and the tight-binding parameters are obtained by fitting the band structures to first-principle results. Surprisingly large on-site spin-orbit coupling is found for all the pnictogen lattices except nitrogen. Investigation on the electronic structures of both zigzag and armchair nanoribbons reveals the possible existence of spin-polarized ferromagnetic edge states in some cases, which are rare in one-dimensional systems. These edge states and magnetism may exist under the condition of high vacuum and low temperature. Parasitic infection is one of the many challenges facing livestock production globally. Cysticercosis tenuicollis is a common parasitic disease in domestic and wild ruminants (intermediate host) caused by the larval stage of Taenia hydatigena that primarily infects dogs (definitive host). Although genetic studies on this parasite exist, only a few describe the genetic variation of this parasite in Mongolia. Our aim was thus, to identify the mitochondrial differences in ovine isolates of Cysticercus tenuicollis entering China from Mongolia and comparison with existing Chinese isolates from sheep and goats based on the recently described PCR–RFLP method and mitochondrial genes of NADH dehydrogenase subunit 4 (nad4) and the NADH dehydrogenase subunit 5 (nad5). Sixty-nine isolates were collected during routine veterinary meat inspections from sheep that originated from Mongolia, at the modern slaughterhouses in Erenhot City, Inner Mongolia. Additional 114 cysticerci were also retrieved from sheep and goats from northern (Inner Mongolia Autonomous Region, Ningxia Hui Autonomous Region, and Gansu Province), western (Tibet Autonomous Region), and southern (Jiangxi Province and Guangxi Province) China. The PCR–RFLP approach of the nad5 showed nine mitochondrial subclusters A1, A2, A3, A5, A8, A9, A10, A11, and B of T. hydatigena isolates from sheep and goats from Mongolia and China. Meanwhile, haplogroup A1 RFLP profile was more widespread than other variants. These data supplements existing information on the molecular epidemiology of T. hydatigena in China and Mongolia and demonstrate the occurrence of similar genetic population structures in both countries. Purpose: The prognostication for the injured recurrent laryngeal nerve (RLN) with incomplete loss of signal (LOS) and its function outcome have not been well unified. A warning criterion was proposed to predict RLN injury during monitored thyroidectomy. Methods: A retrospective review of prospectively collected data from consecutive 357 patients with 560 nerves at risk was conducted. Vocal cords mobility with laryngoscope was performed preoperatively, on the second day, and once a month postoperatively until complete recovery. Different cutoff values of the percentage reduction in sum of the amplitude of left and right channel at the end of the surgery, for postoperative vocal cord paralysis (VCP) prediction were compared. Results: Percentage reduction in sum of the amplitude of left and right channel at the end of operation ranged from 30.2 to 63.6% in 27 nerves with incomplete LOS (absolute amplitude value of final R2 > 100 μV with reduction > 50% of R1). Seven (1.25%) nerves experienced transient postoperative VCP, in which one nerve with postoperative VCP showed no amplitude reduction. The positive predictive value of VCP for the sum amplitude reduction exceeding 30, 40, 50, and 60% was 22.2, 40, 85.7, and 100%, respectively. Accuracy was 96.1, 98.2, 99.6, 99.4%, respectively. Conclusion: Percentage reduction in sum of the amplitude of left and right channel is a meaningful method to improve the accuracy of VCP prediction. As an effective tool for data analysis, formal concept analysis (FCA) is widely used in software engineering and machine learning. The construction of concept lattice is a key step of the FCA. How to effectively to update the concept lattice is still an open, interesting and important issue. To resolve this problem, an incremental algorithm for concept lattice on image structure similarity (SsimAddExten) was presented. The proposed method mapped each knowledge class on the conceptlattice into a graphic, when a new object was added or deleted in a knowledge class, the boundary profile of graphic will be changed, the graphic edge structure similarity was introduced as the calculation index of the change degree before and after the knowledge, and the concept lattice will be updated on the basis of the index. We performed experiments to test SsimAddExtent, whose computational efficiency obtains obvious advantages over mainstream methods on almost all test points, especially on the data set with a large number of attributes. But, its complexity is not reduced compared with mainstream methods. Both theoretical analysis and performance test show SsimAddExtent algorithm is better choice when we apply the FCA to large scale data or non-sparse data. [ABSTRACT FROM AUTHOR]

Background: Bile duct injury after ablation of malignant liver tumors (MLTs) was not unusual and should be avoided. However, few studies have focused on evaluating the risk factors for intrahepatic bile duct injury.Aim: To evaluate the risk factors for intrahepatic bile duct injury after ablation of MLTs and to evaluate the minimum safe distance for ablating tumors abutting bile ducts.Methods: Sixty-five patients with intrahepatic bile duct injury after ablation of MLTs, and 65 controls were recruited. Risk factors for intrahepatic bile duct injury were analyzed. Tumor location was recorded as ≤5 mm (group A), 5-10 mm (group B), and >10 mm (group C) from the right/left main duct or segmental bile duct.Results: Ascites history (P < 0.001), TACE treatment history (P = 0.025), intrahepatic bile duct dilatation before ablation (P < 0.001), and tumor location (P = 0.000) were identified as significant risk factors for intrahepatic bile duct injury. Significant differences in the risk of intrahepatic bile duct injury were found between groups B and C (P = 0.000), but not between groups A and B (P = 0.751). Ascites history (P = 0.002) and tumor location (P < 0.001) were independent predictors with the OR (95 % confidence interval) of 39.31(3.95-391.69) and 16.56 (5.87-46.71), respectively.Conclusions: Bile duct injury after ablation of MLTs was the result of local treatment-related factors combined with the patients' general condition. Microwave-assisted mechanical rock-breaking can effectively solve the problems of low efficiency of hard rock-breaking and severe wear and tear of equipment. This study used granite as the research object and analysed electromagnetic–thermal characteristics and dynamic tensile characteristics of microwave-irradiated granite. The microwave irradiation problem was decomposed into the electromagnetic–thermal unidirectional coupling and thermal–mechanical unidirectional coupling problems for numerical solutions, and the weakening mechanism of microwave irradiation was discussed at the microscopic level. The results showed that the thermal stress generated by the rapid heating and selective heating characteristics of microwave irradiation is the dominant factor in microwave fracturing. The presence of thermal cracks affects the crack extension path, making the failure more complicated. Tensile failure is the primary mechanical mechanism that causes the macroscopic fracture of the specimen, and intergranular failure dominates the overall failure of the specimen. Microwave irradiation reduces the shear resistance of the specimen. Microscopically, the proportion of shear cracks gradually increases with the increasing of irradiation duration (TM). Strength and deformability characteristics of rock with pre-existing fissures are governed by cracking behavior. To further research the effects of pre-existing fissures on the mechanical properties and crack coalescence process, a series of uniaxial compression tests were carried out for rock-like material with two unparallel fissures. In the present study, cement, quartz sand, and water were used to fabricate a kind of brittle rock-like material cylindrical model specimen. The mechanical properties of rock-like material specimen used in this research were all in good agreement with the brittle rock materials. Two unparallel fissures (a horizontal fissure and an inclined fissure) were created by inserting steel during molding the model specimen. Then all the pre-fissured rock-like specimens were tested under uniaxial compression by a rock mechanics servo-controlled testing system. The peak strength and Young's modulus of pre-fissured specimen all first decreased and then increased when the fissure angle increased from $$0^{\circ }$$ to $$75^{\circ }$$ . In order to investigate the crack initiation, propagation and coalescence process, photographic monitoring was adopted to capture images during the entire deformation process. Moreover, acoustic emission (AE) monitoring technique was also used to obtain the AE evolution characteristic of pre-fissured specimen. The relationship between axial stress, AE events, and the crack coalescence process was set up: when a new crack was initiated or a crack coalescence occurred, the corresponding axial stress dropped in the axial stress-time curve and a big AE event could be observed simultaneously. Finally, the mechanism of crack propagation under microscopic observation was discussed. These experimental results are expected to increase the understanding of the strength failure behavior and the cracking mechanism of rock containing unparallel fissures. A new organic salt of dyclonine (DYC) with p-toluenesulfonic acid ( p-TSA) has been synthesized and characterized by elemental analysis, single-crystal X-ray diffraction, IR spectra, UV spectra, and hot stage microscopy. The structure determination shows that the asymmetric unit of the salt consisting of two entire ions of DYC and p-TSA, and that they are not symmetric. The salt is primarily stabilized by a strong N-H···O hydrogen bonding interaction between DYC and p-TSA. In addition, the luminescent properties and biological activities are discussed. Alloimmune inflammation damages the microvasculature of solid organ transplants during acute rejection. Although immunosuppressive drugs diminish the inflammatory response, they do not directly promote vascular repair. Repetitive microvascular injury with insufficient regeneration results in prolonged tissue hypoxia and fibrotic remodeling. While clinical studies show that a loss of the microvascular circulation precedes and may act as an initiating factor for the development of chronic rejection, preclinical studies demonstrate that improved microvascular perfusion during acute rejection delays and attenuates tissue fibrosis. Therefore, preservation of a functional microvasculature may represent an effective therapeutic strategy for preventing chronic rejection. Here, we review recent advances in our understanding of the role of the microvasculature in the long-term survival of transplanted solid organs. Microvascular ischemia and infections are associated with the development of chronic rejection following lung transplantation. The von Hippel-Lindau protein (VHL) controls protein levels of hypoxia-inducible factors (HIFs), regulates vascular repair, and improves tissue perfusion. Here, we studied the role of VHL in microvascular repair by orthotopically transplanting tracheas into mice with VHL haplodeficiency in Tie2 lineage cells. We showed that VHL haplodeficiency prolonged airway microvascular perfusion and promoted tissue blood flow through the production of the angiogenic factors, SDF-1 and angiopoietin 1. VHL-haplodeficient pulmonary endothelial cells exhibited increased angiogenic activity, resistance to serum deprivation-induced cell death, and enhanced microvascular repair. By contrast, in recipient mice with HIF-1α deficiency in Tie2 lineage cells, microvascular repair was significantly diminished and suggested that recipient-derived HIF-1α normally participates in the repair of alloimmune-mediated microvascular damage. To evaluate the translational impact of our findings, we compared VHL-haplodeficient mice with wild-type controls using a model of Aspergillus airway infection. In 83 % of the VHL-haplodeficient recipients, Aspergillus fumigatus was noninvasive in contrast to 75 % of wild-type mice in which the mold was deeply invasive. Our study demonstrated that stabilization of HIF-1α in angiogenic cells, through Tie2 cell VHL haplodeficiency, promoted airway microvascular regeneration and vascular normalization and thereby minimized tissue ischemia and hypoxia. By also mitigating the virulence of A. fumigatus, a common pathogen and itself a risk factor for the development of lung transplant rejection, the selective enhancement of HIF-1α expression has the prospect of offering several novel therapeutic effects to transplant recipients. TRAIL was a tumor-specific protein in development as a novel anticancer therapeutic agent. Generally, when expressed in recombinant Escherichia coli, TRAIL protein was prone to form inclusion bodies. In this study, coexpression of human TRAIL protein and protein isoaspartate methyltranferase (PIMT) from E. coli on plasmid pBV–TRAIL–PCM in E. coli C600 was investigated to overcome the difficulties in soluble expression. The results showed that this PIMT coexpression strategy exerted a positive effect on the TRAIL protein expression in recombinant E. coli, which led to a mean increase in the intracellular concentration of soluble and total protein of TRAIL by 1.57-fold and 1.33-fold, respectively.
