Abstract  The DNA polymerase gene (dnapol) of thePieris rapaegranulovirus (PiraGV) was completely sequenced and located between 73.1 and 76 m.u. on the PiraGV genome. Its open reading frame (ORF) has 3135 nucleotides (35% G-C content) encoding 1045 amino acids with a predicted molecular mass of 122.16 kDa. Homology analysis indicated that PiraGVdnapolhad 28–66% amino acid identity to that of other known baculoviruses. Comparative sequence analyses demonstrated that the PiraGVdnapolgene contains conserved 3′–5′ exonuclease motifs and DNA binding functional domains of the DNA polymerase enzyme found in all known baculovirusdnapols. Northern blot results showed that in infectedPieris rapaelarvae the PiraGVdnapolgene was transcribed as a predominant 3.7 kb mRNA. 5′ and 3′ RACE indicated that the PiraGVdnapoltranscript was initiated from the thymine residue located at −378 nt upstream from the ATG start codon and terminated at the polyadenylation signal AATAAA. Phylogenetic analysis ofdnapolsequences suggests that the PiraGVdnapolis more closely related to that ofCydia pomonellaGV andCryptophlebia leucotretaGV than to those of other baculoviruses. A platinum (Pt) on pure ceria (CeO2) supported by carbon black (CB) anode was synthesized using a combined process of precipitation and coimpregnation methods. The electrochemical activity of methanol oxidation reaction on synthesized Pt-CeO2/CB anodes was investigated by cyclic voltammetry and chronoamperometry experimentation. To improve the anode property on Pt-CeO2/CB, the influence of particle morphology and particle size on anode properties was examined. The morphology and particle size of the pure CeO2 particles could be controlled by changing the preparation conditions. The anode properties (i.e., peak current density and onset potential for methanol oxidation) were improved by using nanosize CeO2 particles. This indicates that a larger surface area and higher activity on the surface of CeO2 improve the anode properties. The influence of particle morphology of CeO2 on anode properties was not very large. The onset potential for methanol oxidation reaction on Pt-CeO2/CB, which consisted of CeO2 with a high surface area, was shifted to a lower potential compared with that on the anodes, which consisted of CeO2 with a low surface area. The onset potential on Pt-CeO2/CB at 60 °C became similar to that on the commercially available Pt-Ru/carbon anode. We suggest that the rate-determining steps of the methanol oxidation reaction on Pt-CeO2/CB and commercially available Pt-Ru/carbon anodes are different, which accounts for the difference in performance. In the reaction mechanism on Pt-CeO2/CB, we conclude that the released oxygen species from the surface of CeO2 particles contribute to oxidation of adsorbed CO species on the Pt surface. This suggests that the anode performance of the Pt-CeO2/CB anode would lead to improvements in the operation of direct methanol fuel cells at 80 °C by the enhancement of diffusion of oxygen species created from the surface of nanosized CeO2 particles. Therefore, we conclude that fabrication of nanosized CeO2 with a high surface area is a key factor for development of a high-quality Pt-CeO2/CB anode in direct methanol fuel cells. We constructed a peptide consisting of a staphylococcal AgrD1 pheromone fused to the channel-forming domain of colicin Ia and named it pheromonicin. This fusion peptide had bactericidal effects against methicillin-sensitive and methicillin-resistant Staphylococcus aureus (MSSA and MRSA, respectively), but not against Staphylococcus epidermidis or Streptococcus pneumoniae. Growth rates, vital staining and colony forming unit (CFU) counts showed that pheromonicin did not merely suppress growth but killed S. aureus cells. The specificity of pheromonicin was shown by the absence of bactericidal effects against an accessory gene regulator (agr) locus knockout of S. aureus, and a dose-dependent inhibition of the bactericidal effects of pheromonicin by competition with corresponding free AgrD pheromone. In vivo, all pheromonicin-treated mice survived administration of MRSA that was lethal to controls. No toxicity was detectable in human liver or renal cells in culture, or in livers, kidneys or spleens of pheromonicin-treated mice.
