Polyethylene glycol (PEG 6000) was used to establish osmotic stress conditions during growth of the type strain Azospirillum brasilense Sp7 and its spontaneous variants Sp7.4 and Sp7.8, because it causes a stable decrease in the water potential and thus makes it possible to simulate the effect of drought on the bacterial population. While PEG suppressed the motility of azospirilla, it had no effect on the ability of strains Sp7 and Sp7.8 to form biofilms, as well as on the metabolic activity of the biofilms formed in the absence of stress. PEG 6000-caused osmotic stress promoted biofilm formation in Sp7.8. While the biofilms of the Sp7.4 variant were those most sensitive to the negative effect of the water stress, the growth variables of the planktonic culture of this variant under stress conditions exceeded the values for both Sp7 and Sp7.8. In biofilms, strains Sp7, Sp7.4, and Sp7.8 produced polysaccharides and the plant hormone IAA; desiccation-resistant cell forms emerged. The variants Sp7.4 and Sp7.8, similarly to Sp7, formed biofilms during plant root colonization and affected the morphology of the root system of wheat seedlings. Our results show that spontaneous variants of strain Sp7 may be of interest for further research directed at selection of promising Azospirillum strains to enhance the drought resistance of plants. The plant growth-promoting bacterium Azospirillum brasilense contains several genes encoding proteins involved in the biosynthesis and degradation of the second messenger cyclic-di-GMP, which may control key bacterial functions, such as biofilm formation and motility. Here, we analysed the function and expression of the cdgD gene, encoding a multidomain protein that includes GGDEF-EAL domains and CHASE and PAS domains. An insertional cdgD gene mutant was constructed, and analysis of biofilm and extracellular polymeric substance production, as well as the motility phenotype indicated that cdgD encoded a functional diguanylate protein. These results were correlated with a reduced overall cellular concentration of cyclic-di-GMP in the mutant over 48 h compared with that observed in the wild-type strain, which was recovered in the complemented strain. In addition, cdgD gene expression was measured in cells growing under planktonic or biofilm conditions, and differential expression was observed when KNO3 or NH4Cl was added to the minimal medium as a nitrogen source. The transcriptional fusion of the cdgD promoter with the gene encoding the autofluorescent mCherry protein indicated that the cdgD gene was expressed both under abiotic conditions and in association with wheat roots. Reduced colonization of wheat roots was observed for the mutant compared with the wild-type strain grown in the same soil conditions. The Azospirillum-plant association begins with the motility of the bacterium towards the plant rhizosphere followed by the adsorption and adherence of these bacteria to plant roots. Due to the primary localization of both epiphytic and endophytic plant growth-promoting rhizobacteria on the surface of the plant root system, biofilm formation is an adaptive trait for these microorganisms. Under conditions of nitrogen limitation in liquid media, nitrogen-fixing Azospirillum brasilense strains switch mainly to the biofilm mode of growth. Overall ultrastructural similarities of the cells within A. brasilense biofilms were revealed, and their resistance to desiccation and oxidative stress was characterized. In strains Sp7, Cd, and Sp245, several types of single and undivided cells were revealed, as well as cystlike cells with pronounced morphological diversity. Resistance to desiccation and to oxidative stress was higher in the biofilm populations of these strains than in planktonic cultures. Dormant forms remained viable in dry biofilms of strains Sp245, Sp7, and Cd formed in a nitrogen-free medium after storage for 120 days. Viability of the biofilms of the same strains formed in the presence of nitrogen was retained for 120, 90, and 60 days, respectively. The minimal inhibitory concentration of H2O2 for biofilms was 1.0% for strains Sp7 and Cd and 0.1% for strain Sp245. Both the dormant forms and biofilms of strain Sp245 were more sensitive to H2O2 than those of strains Sp7 and Cd. Peroxidase activity was not previously reported in Azospirillum biofilms. Abstract—: The structure and functions of the biopolymers of the matrix of Azospirillum brasilense biofilms formed at the liquid-solid interface were studied. Calcofluor-binding polysaccharides (CBPS) and the complexes containing lipopolysaccharides (LPS) and proteinaceous structures were shown to fix mature biofilms on solid surfaces and to perform a framework function. The matrix of mature biofilms contained a carbohydrate hapten with a specific affinity to wheat germ agglutinin (WGA), which is important for associative interaction and plant root colonization. The carbohydrate components, predominating over the proteinaceous matrix components, mediate preferable biofilm attachment to hydrophilic surfaces, which is especially evident in the case of A. brasilense Sp245 mutants with impaired flagella formation. Azospirillum brasilense is an important plant-growth promoting bacterium (PGPB) that requires several critical steps for root colonization, including biofilm and exopolysaccharide (EPS) synthesis and cell motility. In several bacteria these mechanisms are mediated by quorum sensing (QS) systems that regulate the expression of specific genes mediated by the autoinducers N-acyl-homoserine lactones (AHLs). We investigated QS mechanisms in strains Ab-V5 and Ab-V6 of A. brasilense, which are broadly used in commercial inoculants in Brazil. Neither of these strains carries a luxI gene, but there are several luxR solos that might perceive AHL molecules. By adding external AHLs we verified that biofilm and EPS production and cell motility (swimming and swarming) were regulated via QS in Ab-V5, but not in Ab-V6. Differences were observed not only between strains, but also in the specificity of LuxR-type receptors to AHL molecules. However, Ab-V6 was outstanding in indole acetic acid (IAA) synthesis and this molecule might mimic AHL signals. We also applied the quorum quenching (QQ) strategy, obtaining transconjugants of Ab-V5 and Ab-V6 carrying a plasmid with acyl-homoserine lactonase. When maize ( Zea mays L.) was inoculated with the wild-type and transconjugant strains, plant growth was decreased with the transconjugant of Ab-V5-confirming the importance of an AHL-mediated QS system-but did not affect plant growth promotion by Ab-V6. Bacteria Azospirillum brasilense Sp245 with mixed flagellation are able to form biofilms on various surfaces. A nonflagellated mutant of this strain with inactivated chromosomal copy of the flhB gene ( flhB1) was shown to exhibit specific traits at the later stages of biofilm formation on a hydrophilic (glass) surface. Mature biofilms of the flhB1::Omegon-Km mutant Sp245.1063 were considerably thinner than those of the parent strain Sp245. The biofilms of the mutant were more susceptible to the forces of hydrodynamic shear. A. brasilense Sp245 cells in biofilms were not found to possess lateral flagella. Cells with polar flagella were, however, revealed by atomic force microscopy of mature native biofilms of strain Sp245. The thickness and antigenic properties of biofilms produced by Azospirillum brasilense Sp245 and its mutants deficient in the synthesis of lipopolysaccharides (Lps) and calcofluor-binding polysaccharides (CBPS) at the interface between water and hydrophilic or hydrophobic solid surfaces were compared. The mutants deficient in acidic LpsI synthesis produce thicker biofilms on hydrophilic surfaces. Biofilms produced on hydrophobic surfaces by bacteria that are unable to synthesize CBPS are less pronounced. Defects in CBPS production in Azospirillum mutants with impaired flagellar motility can cause adverse effects on the cell ability to attach to hydrophobic and hydrophilic surfaces. The loss of the neutral LpsII antigen by the mutants capable of producing CBPS does not affect their behavior on hydrophobic surfaces, which is probably due to the compensatory increase in the total polysaccharide production. Bacteria Azospirillum brasilense may swim and swarm owing to the rotation of a constitutive polar flagellum (Fla) and inducible lateral flagella (Laf). They also construct sessile biofilms on various interfaces. As compared to the wild-type strain Sp245, the previously characterized Fla− Laf−flhB1 mutant Sp245.1063 accumulated less biomass in mature biofilms, which also were susceptible to the forces of hydrodynamic shear. In this study, we compared biofilms formed by strain Sp245 and its previously constructed derivatives on the interfaces between a minimal (malate-salt medium, or MSM) or rich (LB) liquid growth medium and a hydrophilic (glass) or hydrophobic (polystyrene) solid surface under static or dynamic conditions. In all experimental settings, the alterations in Sp245.1063's mature biofilm traits were partially (in MSM) or completely (in LB) rescued in the complemented mutant Sp245.1063 (pRK415-150177), which received the pRK415-borne coding sequence for the putative FlhB1 protein of the flagellar type III secretion system. Although Laf were not found in the biofilms of azospirilla, Fla was present on the biofilm cells of the complemented mutant Sp245.1063 (pRK415-150177) and other studied strains, which had normal flagellation on liquid and solid nutritional media. Accordingly, mature biofilms of these strains contained more biomass and were significantly more resistant to shaking at 140 rpm, as compared to the biofilms of the flagella-free mutant bacteria.
