We systematically compare the temporal and spectral properties of 53 supernova (SN)-associated and 15 kilonova (KN)-associated gamma-ray bursts (GRBs). We find that the spectral parameters of both types of GRBs are identically and lognormally distributed, consistent with those normal GRBs. The bolometric luminosities of SN/GRBs and KN/GRBs have a triple form, with the corresponding break luminosities of SN/GRBs are roughly two orders of magnitude larger than those of KN/GRBs. We build the power-law relations between the spectral lag and the luminosity of prompt γ-rays with indices of −1.43 ± 0.33 for SN/GRBs and −2.17 ± 0.57 for KN/GRBs in the laboratory frame, which are respectively coincident with the rest-frame values. We verify that both SN/GRBs and KN/GRBs comply with their own Amati relations that match those of long and short GRBs, respectively. Analysing X-ray afterglows with good plateau segments, we build the power-law relations between the X-ray luminosity and the plateau time with an index of −1.12 ± 0.17 for KN/GRBs and −1.08 ± 0.22 for SN/GRBs, which can be well explained by the relativistic shock driven by an energy injection. The plots of luminosity-lag, Amati relation, and luminosity-time show heavy overlap between the two types of GRBs, implying that they might share the same radiation mechanism despite originating from different progenitors or central engines. Aims: Probiotics and plant extracts have been used to prevent the development of type 2 diabetes mellitus (T2DM). The study aimed to explore the effect of the interaction between potential probiotics and bitter gourd extract (BGE) or mulberry leaf extract (MLE) on T2DM. Methods and Results: Potential probiotics were tested for their gastrointestinal tract viability and growth situation combined with BGE and MLE in vitro. The diabetes model was constructed in C57BL/6 mice, and the potential effect and mechanism of regulating blood glucose were verified. Hematoxylin‐eosin staining (HE), gas chromatography (GC), ELISA, and RT‐PCR were also used for analysis. The results showed that Lactobacillus casei K11 had outstanding gastrointestinal tract viability and growth situation with plant extracts. Administration of L. casei K11 combined with BGE and MLE significantly reduced blood glucose levels and ameliorated insulin resistance in diabetic mice than the administration of Lactobacillus paracasei J5 combined with BGE and MLE. Moreover, in L. casei K11 combined with BGE and MLE groups, lipid metabolism, oxidative stress, and proinflammatory cytokine levels were regulated. Furthermore, the results indicated that L. casei K11 combined with BGE and MLE improved free fatty acid receptor 2 (FFAR2) upregulation, glucagon‐like peptide‐1 (GLP‐1) secretion, and short‐chain fatty acid (SCFA) levels. Conclusions: These findings showed that L. casei K11 combined with BGE and MLE modified the SCFA–FFAR2–GLP‐1 pathway to improve T2DM. We systematically analyse two GRB samples with radio-loud and radio-quiet afterglows, respectively. It is interestingly found that the radio-selected GRB samples exhibit a clear dichotomy in terms of their distributions of intrinsic durations (T int), isotropic energies in γ-rays (E γ, iso), the circum-burst medium density (n), the spectral radio peak luminosity (L ν, p ) and flux densities (F host) of host galaxies. On average, the values of T int, E γ, iso, n, L ν, p , and F host of radio-quiet GRBs are relatively smaller than those of radio-loud ones. However, the redshifts and host flux densities of both samples are similarly distributed. In addition, a positive power-law correlation of |$L_{\nu ,p}\propto E_{\gamma ,\rm iso}^{0.41\pm 0.04}$| is found for the radio-loud sample, especially in accord with the supernova-associated GRBs, which is marginally consistent with that of the radio-quiet GRB sample. A negative correlation between T int and z is confirmed to similarly hold for both radio-loud and radio-quiet GRBs. The dividing line between short and long GRBs in the rest frame is at T int ≃1 s. Gamma-ray bursts (GRBs) are the most intense explosions in the Universe. GRBs with extended emission (GRBs EE) constitute a small subclass of GRBs. GRBs EE are divided into EE-I GRBs and EE-II GRBs, according to the Amati empirical relationship rather than duration. We test here if these two types of GRB have different origins based on their luminosity function (and formation rate). Therefore, we use Lynden-Bell's c − method to investigate the luminosity function and formation rate of GRBs with EE without any assumption. We calculate the formation rate of two types of GRBs. For EE-I GRBs, the fitting function can be written as ρ(z) ∝ (1 + z)−0.34 ± 0.04 for z < 2.39 and ρ(z) ∝ (1 + z)−2.34 ± 0.24 for z > 2.39. The formation rate of EE-II can describe as ρ(z) ∝ (1 + z)−1.05 ± 1.10 for z < 0.43 and ρ(z) ∝ (1 + z)−8.44 ± 1.10 for z > 0.43. The local formation rate is |$\rho (0) = 0.03\, {\rm Gpc}^{-3} \, {\rm yr}^{-1}$| for some EE-I GRBs and |$\rho (0) = 0.32 \, {\rm Gpc}^{-3} \, {\rm yr}^{-1}$| for EE-II GRBs. Based on these results, we provide new evidence that the origins of EE-I GRBs are different from EE-II GRBs from the perspective of event rate. The EE-I GRB could be produced from the death of the massive star, but EE-II GRB may come from other processes that are unrelated to the star formation rate. Aims: The aims of the study were to (i) improve the evaluation criteria of detoxifying Jatropha curcas L. cake (JCC), (ii) isolate and characterize a JCC tolerant strain, (iii) explore its JCC detoxifying potential. Methods and Results: The zebrafish was employed as a survival model to screen the strains capable of detoxifying JCC. A strain identified as Mucor circinelloides SCYA25, which is highly capable of degrading all toxic components, was isolated from soil. Different solid‐state fermentation parameters were optimized by response surface methodology. The optimal values for inoculation amount, moisture content, temperature, and time were found to be 18% (1·8 × 106 spores g−1 cake), 66%, 26, and 36 days, respectively, to achieve maximum detoxification of the JCC (92%). Under optimal fermentation conditions, the protein content of JCC was increased, while the concentrations of ether extract, crude fiber, toxins, and anti‐nutritional substances were all degraded considerably (P < 0·05). Scanning electron microscopy and Fourier transform infrared spectrometer analysis revealed that the fermentation process could disrupt the surface structure and improve the ratio of α‐helix to β‐folding in the JCC protein, which may improve the digestibility when the detoxified JCC is used as a feedstuff. Conclusions: Our results indicate that M. circinelloides SCYA25 is able to detoxify JCC and improve its nutritional profile, which is beneficial to the safe utilization of JCC as a protein feedstuff. Significance and Impact of the Study: The newly identified M. circinelloides SCYA25 detoxified JCC in a safe manner to provide a potential alternative to soybean meal for the feed industry. Abstract: In this study, a glutathione S‐transferase gene (gst) from sensitive Physarum polycephalum was selected for its ability to detect nanosized TiO2 (nTiO2) exposure under dark conditions. The concentration of nTiO2 (25, 40 and 60 nm) for subsequent assays was first determined (5–18 mg ml−1) and total GST enzyme activity of P. polycephalum was confirmed to be increased 6–44 fold in groups treated with nTiO2. Second, an RNA‐seq study was performed to identify candidate gst genes before isolation of an optimum gst gene of P. polycephalum (Ppgst), which encoded 223 amino acids. Third, the transcriptional level of the Ppgst gene was further confirmed to be positively correlated with nTiO2 exposure within the concentration range of (5–15 mg ml−1) by qPCR. In conclusion, these results indicated that the transcriptional level of Ppgst can reflect nTiO2 exposure, suggesting that it may be employed as a new biomarker for nTiO2 pollution under dark conditions. Aims Drug susceptibility testing ( DST) of clinical isolates of Mycobacterium tuberculosis is critical in treating tuberculosis. We demonstrate the possibility of using a microbial sensor to perform DST of M. tuberculosis and shorten the time required for DST. Methods and Results The sensor is made of an oxygen electrode with M. tuberculosis cells attached to its surface. This sensor monitors the residual oxygen consumption of M. tuberculosis cells after treatment with anti- TB drugs with glycerine as a carbon source. In principle, after drug pretreatment for 4-5 days, the response differences between the sensors made of drug-sensitive isolates are distinguishable from the sensors made of drug-resistant isolates. The susceptibility of the M. tuberculosis H37Ra strain, its mutants and 35 clinical isolates to six common anti- TB drugs: rifampicin, isoniazid, streptomycin, ethambutol, levofloxacin and para-aminosalicylic acid were tested using the proposed method. The results agreed well with the gold standard method ( LJ) and were determined in significantly less time. The whole procedure takes approximately 11 days and therefore has the potential to inform clinical decisions. Conclusions To our knowledge, this is the first study that demonstrates the possible application of a dissolved oxygen electrode-based microbial sensor in M. tuberculosis drug resistance testing. This study used the microbial sensor to perform DST of M. tuberculosis and shorten the time required for DST. Significance and Impact of the Study The overall detection result of the microbial sensor agreed well with that of the conventional LJ proportion method and takes less time than the existing phenotypic methods. SDSS J010448.46+153501.8 has previously been classified as an sdM9.5 subdwarf. However, its very blue J - K colour (-0.15 ± 0.17) suggests a much lower metallicity compared to normal sdM9.5 subdwarfs. Here, we re-classify this object as a usdL1.5 subdwarf based on a new optical and near-infrared spectrum obtained with X-shooter on the Very Large Telescope. Spectral fitting with BT-Settl models leads to Teff = 2450 ± 150 K, [Fe/H] = -2.4 ± 0.2 and log g = 5.5 ± 0.25. We estimate a mass for SDSS J010448.46+153501.8 of 0.086 ± 0.0015M☉ which is just below the hydrogen-burning minimum mass at [Fe/H] = -2.4 (~0.088 M☉) according to evolutionary models. Our analysis thus shows SDSS J010448.46+153501.8 to be the most metal-poor and highest mass substellar object known to-date.We found that SDSS J010448.46+153501.8 is joined by another five known L subdwarfs (2MASS J05325346+8246465, 2MASS J06164006−6407194, SDSS J125637.16−022452.2, ULAS J151913.03−000030.0 and 2MASS J16262034+3925190) in a 'halo brown dwarf transition zone' in the Teff-[Fe/H] plane, which represents a narrow mass range in which unsteady nuclear fusion occurs. The treatment of choice for leiomyoma, the most common benign esophageal tumor, is thoracoscopic enucleation. One of the most difficult aspects of thoracoscopic enucleation is the precise localization of small tumors (=1.5 cm) and tumors without external protrusion. No simple, feasible solutions to this problem are available. We developed a novel methylene blue staining technique to localize small esophageal leiomyomas and evaluated the feasibility of our technique. Between January 2013 and July 2014, eight patients with small esophageal leiomyomas (=1.5 cm) underwent thoracoscopic enucleation in Tongji Hospital. Preoperative endoscopic ultrasonography was performed in all patients. The leiomyomas were located in the middle (n = 5) and lower (n = 3) thirds of the esophagus. We preoperatively injected 0.5-1.0mLmethylene blue in the submucosa adjacent to the tumors under standard gastroscope guidance. The entire staining process took about 10 minutes. Staining was successful in all patients. The unstained tumor was exposed after the blue-stained mediastinal pleura, and overlying muscle were incised longitudinally. All procedures were successfully completed without conversion to open surgery. No abnormalities were detected in the esophageal mucosa. The median operating time was 60minutes (range, 40-90 minutes). Postoperative histopathology confirmed leiomyoma in all patients. The median postoperative hospital stay was 6 days (range, 5-7 days). No major complications, such as esophageal leakage or esophageal diverticulum, occurred. Endoscopic methylene blue staining is safe and feasible for localizing small esophageal leiomyomas during thoracoscopic enucleation. This method will enable precise and easy enucleation. Bacillus subtilis ZK0, which was isolated from cotton, produces a type of lipopeptide antibiotic iturin A that inhibits the growth of pathogenic fungi on agricultural crops. However, the low level of iturin A production by B. subtilis ZK0 does not support its large-scale application. In this study, B. subtilis ZK0 was subjected to genetic manipulation to improve iturin A production. By the independent or simultaneous overexpression of two regulatory genes ( comA and sigA), iturin A production by B. subtilis ZK0 was significantly increased. When both genes were simultaneously overexpressed under optimal conditions, iturin A production increased up to 215 mg l−1 (an approximate 43-fold increase compared with B. subtilis ZK0). Moreover, overexpression of both genes was unexpectedly found to inhibit biofilm formation by B. subtilis ZK0, indicating that the phenomenon of 'stuck fermentation' would be avoided during B. subtilis ZK0 fermentation. In conclusion, a genetic manipulation method that improves iturin A production and inhibits biofilm formation in B. subtilis ZK0 is reported for the first time and this method has the potential to be widely applied in B. subtilis ZK0 commercial fermentation. Our newly engineered strains could be applied to commercial fermentation by enhancing yields of iturin A and reducing the rate of 'stuck fermentation'. The effectiveness of neoadjuvant chemotherapy (nCT) or chemoradiotherapy (nCRT) for esophageal cancer patients is well established. However, neoadjuvant therapy may induce severe adverse effects that could increase postoperative morbidity. The current study evaluated the impacts of nCT and nCRT on postoperative cardiopulmonary complications in patient with esophageal cancer. We conducted a prospective study in esophageal cancer patients who received nCT (n = 126) or nCRT (n = 141) prior to surgery. Surgery was performed in all these patients following nCT or nCRT treatment. More patients occurred pneumonia in the nCRT-treated group compared with the nCT group (P < 0.01). The E-velocity (early diastolic filling velocity) decreased significantly (P = 0.026), while the N-terminal pro-brain natriuretic peptide (NT-proBNP) significantly increased (P < 0.01) in patients of the nCRT group compared with patients from the nCT group. Furthermore, a multivariate analysis revealed that nCRT was correlated with the incidence of pneumonia and NT-proBNP level significantly. The nCRT caused more cardiopulmonary toxicity than nCT. The strategies are needed to prevent the postoperative cardiopulmonary complications especially in patients with nCRT treatment. [ABSTRACT FROM AUTHOR]

By incorporating high-density markers into breeding value prediction models, the whole genomic prediction (WGP) method can effectively accelerate genetic improvement in livestock breeding. However, the performance of WGP varies across species and populations and is affected by the underlying genetic architecture. In particular, very little is known about the performance of WGP for many chicken breeds. Here we estimate the genetic parameters and evaluate the performance of WGP for 18 growth and carcass traits in a Chinese quality chicken population. In total, 435 chickens were systematically phenotyped and genotyped using a 600K genotyping array. Two variance component estimation scenarios, 3 breeding value prediction methods, and 2 validation procedures were compared. The results showed that the heritability of these 18 traits was medium to high (ranging from 0.28 to 0.60) and that deviations existed between the heritability estimated from pedigrees and markers. Compared with conventional breeding methods, WGP could potentially increase the selection accuracy by 20% or more depending on the prediction model used, the trait under consideration, and the genetic connectedness between the training and validation individuals. Feeding efficiency is a multifactorial and economically important trait in pigs. Genetic improvement of feeding efficiency will greatly benefit the pig industry. In the past decades, the hog market weight has increased worldwide. However, whether the genetic architecture of feeding efficiency is same or not at early and late fattening periods is unclear. To map genomic regions for feed efficiency and feeding behavior traits at early (n ≥ 384) and late (n ≥ 334) growth stages in pigs, we performed genomewide association studies for feed to gain ratio (FCR), residual feed intake (RFI), daily feed intake, daily visit times, daily feeding time (DFT), feed intake per second (FIPS), and feed intake per visit during 3 periods (2 stages and overall) in a White Duroc × Erhualian F2 intercross population. Six chromosomal regions showed significant association with these traits, of which 4 loci were reported for the first time. Our results confirmed the QTL of FCR around 34 Mb on SSC7 and RFI around 134 Mb on SSC12. Of note, 2 regions were associated with more than 1 trait. One was around 36 Mb on SSC7, and there were 47 and 67 SNP associated with FCR from 120 to 210 and from 120 to 240 d, respectively. The top SNP is located in a 2.88-Mb linkage disequilibrium (LD) block that harbors 44 genes. We propose the high mobility group AT-hook 1 gene as a plausible candidate gene in this region. The other was evidenced around 53 Mb on SSC12, which had multiple association signals for DFT and FIPS. The top SNP is located in a 211-kb LD block that harbors only 1 annotated gene, WSCD1, which encodes a protein with sulfotransferase activity and involves the glucose metabolism and, therefore, appears to be a plausible candidate gene. Except the region on SSC12 associated with DFT at both stages, the rest of the regions associated with the traits at only 1 stage, so the genetic architectures of the 2 stages are not same. Aims: This study gives a systematic profile of ammonium overconsumption in anammox consortia beyond the stoichiometry ratio, with aim to reveal another ammonium removal process by oxidants OH· without nitrite under anaerobic conditions. Methods and Results: Percoll centrifugation, flow cytometry, cloning and chemical analysis demonstrated that this ammonium removal was related to low Planctomycetes and high biomass decay. Armatimonadetes, Acidobacteria, Verrucomicrobia and Nitrosomonas initially existed in the consortia with the overconsumption of ammonium, and their levels then declined with decreasing overconsumption. The inevitable release of oxidants, such as OH, during bacterial inhibition or decay was found to be a potential source of electron acceptors capable of oxidizing ammonium. Conclusions: Anaerobic ammonium removal without nitrite could occur in anammox consortia. However, it was unsteady and was not sustained over a long period. Under suitable conditions, anammox bacteria competed with others and became the dominant micro-organism responsible for the simultaneous removal of ammonium and nitrite. Significance and Impacts of the Study: This investigation will have importance to better understand the anammox process operation. Studies in mammalian species showed that Krüppel-like factor 2 (KLF2) regulates adipogenesis. However, its role in birds is unclear. The objective of the current study was to explore the expression and function of KLF2 in chicken adipogenesis. Results showed that chicken KLF2 (Gallus gallus KLF2 [gKLF2]) was greatly expressed in abdominal adipose tissue, and its transcripts fluctuated during adipose tissue development. In addition, gKLF2 transcripts in abdominal adipose tissue of lean broilers were greater at 1 wk of age but lower at 3, 5, and 8 wk of age than those in fat broilers (P < 0.05). The gKLF2 was more greatly expressed in preadipocytes than in mature adipocytes (P < 0.05), and its expression level decreased during the preadipocyte differentiation in vitro (P < 0.05). The functional analysis showed that gKLF2 overexpression inhibited chicken preadipocyte differentiation (P < 0.05), accompanied by the reduced expression of CCAAT/enhancer binding protein α (C/EBPα) and peroxisome proliferator-activated receptor γ (PPARγ) and the elevated expression of GATA binding protein 2 (GATA2). Additionally, the luciferase reporter assays showed that gKLF2 overexpression suppressed the promoter activities of chicken C/EBPα and PPARγ (P < 0.05). Polymerase chain reaction denaturing gradient gel electrophoresis (PCR-DGGE), and a culture-dependent technique were used to study the diversity of the intestinal bacterial community in adult Dastarcus helophoroides (Fairmaire) (Coleoptera: Bothrideridae). Universal bacterial primers targeting 200 bp regions of the 16S rDNA gene were used in the PCR-DGGE assay, and 14 bright bands were obtained. The intestinal bacteria detected by PCR-DGGE were classified to Enterococcus (Lactobacillales: Enterococcaceae), Bacillus (Bacillales: Bacillaceae), Cellvibrio (Pseudomonadales: Pseudomonadaceae), Caulobacter (Caulobacterales: Caulobacteraceae), and uncultured bacteria, whereas those isolated by the culture-dependent technique belonged to Staphylococcus (Bacillales: Staphylococcaceae), Pectobacterium Enterobacteriales: Enterobacteriaceae), and Enterobacter (Enterobacteriales: Enterobacteriaceae). These intestinal bacteria represented the groups Lactobacillales (Enterococcus), Pseudomonadales (Cellvibrio), Caulobacterales (Caulobacter), Bacilli (Bacillus and Staphylococcus), and Gammaproteobacteria (Pectobacterium and Enterobacter). Our results demonstrated that PCR-DGGE analysis and the culture-dependent technique were useful in determining the intestinal bacteria of D. helophoroides and the two methods should be integrated to characterize the microbial community and diversity. In this work, we restudy the dependence of luminosity function and event rates for different gamma-ray burst samples on the criteria of sample selection and threshold effect. To compare with many previous studies, we have chosen two samples including 88 and 118 long bursts with known redshift and peak flux over 2.6 ph cm−2 s−1, from which 79 bursts are picked out to constitute our complete sample. It is found that the evolution of luminosity with redshift can be expressed by L ∝(1 + z) k with a diverse k relied more on the sample selection. Interestingly, the cumulative distributions of either non-evolving luminosities or redshifts are found to be also determined by the sample selection rather than the instrumental sensitivity. Nevertheless, the non-evolving luminosities of our samples are similarly distributed with a comparable break luminosity of L 0 ∼ 1051 erg s−1. Importantly, we verify with a K-S test that three cases of event rates for the two burst samples evolve with redshift similarly except a small discrepancy due to sampling differences at low-redshift of z < 1, in which all event rates show an excess of Gaussian profile instead of monotonous decline no matter whether the sample is complete. Most importantly, it is found that the burst rates violate the star formation rate at low redshift, while both of them are good in agreement with each other in the higher-redshift regions as many authors discovered previously. The ALMA interferometer has played a key role in revealing a new component of the Sun-like star forming process: the molecular streamers, i.e. structures up to thousands of au long funnelling material non-axisymmetrically to discs. In the context of the FAUST ALMA LP, the archetypical VLA1623-2417 protostellar cluster has been imaged at 1.3 mm in the SO(56–45), SO(66–55), and SiO(5–4) line emission at the spatial resolution of 50 au. We detect extended SO emission, peaking towards the A and B protostars. Emission blue-shifted down to 6.6 km s−1 reveals for the first time a long (∼ 2000 au) accelerating streamer plausibly feeding the VLA1623 B protostar. Using SO, we derive for the first time an estimate of the excitation temperature of an accreting streamer: 33 ± 9 K. The SO column density is ∼ 1014 cm−2, and the SO/H2 abundance ratio is ∼ 10−8. The total mass of the streamer is 3 × 10−3 M ⊙, while its accretion rate is 3–5 × 10−7 M ⊙ yr−1. This is close to the mass accretion rate of VLA1623 B, in the 0.6–3 × 10−7 M ⊙ yr−1 range, showing the importance of the streamer in contributing to the mass of protostellar discs. The highest blue- and red-shifted SO velocities behave as the SiO(5–4) emission, the latter species detected for the first time in VLA1623-2417: the emission is compact (100–200 au), and associated only with the B protostar. Using spectroscopy from the Sloan Digital Sky Survey Data Release Seven, we systematically determine the electron density ne and electron temperature Te of active galaxies and star-forming galaxies, while mainly focusing on the narrow-line regions (NLRs). Herein, active galaxies refer to composites, low-ionization narrow emission-line regions (LINERs) and Seyfert galaxies, following the Baldwin-Phillips-Terlevich diagram classifications afforded by the SDSS data. The plasma diagnostics of ne and Te are determined through the I[S ii] λ6716/λ6731 and I[O III] λ5007/λ4363 ratios, respectively. By simultaneously determining ne from [S ii] and Te from [O III] in our [O III] λ4363 emission sample of 15 019 galaxies, we find two clear sequences: TLINER ≳ Tcomposite > TSeyfert > Tstar-forming and nLINER ≳ nSeyfert > ncomposite > nstar-forming. The typical range of nee in the NLRs of active galactic nuclei (AGNs) is 102 - 3 cm-3. The temperatures in the NLRs range from 1.0 to 2.0 x 104 K for Seyferts, and the ranges are even higher and wider for LINERs and composites. The transitions of ne and Te from the NLRs to the discs are revealed. We also present a comparative study, including stellar mass (M*), specific star formation rate (SFR/M*) and plasma diagnostic results. We propose that YL ≳ YSY > YC > YSF, where Y is the characteristic present-day star-formation time-scale. One remarkable feature of the Seyferts shown on an M*-SFR/M* diagram, which we call the evolutionary pattern of AGNs with high ionization potential, is that the strong [O III] λ4363 Seyferts distribute uniformly with the weak Seyferts, definitely a reverse of the situation for star-forming galaxies. It is a natural and well-known consensus that strong [O III] λ4363 emissions in star-forming galaxies imply young stellar populations and thus low stellar masses. Motivation: In many situations, genome-wide association studies are performed in populations presenting stratification. Mixed models including a kinship matrix accounting for genetic relatedness among individuals have been shown to correct for population and/or family structure. Here we extend this methodology to generalized linear mixed models which properly model data under various distributions. In addition we perform association with ancestral haplotypes inferred using a hidden Markov model.Results: The method was shown to properly account for stratification under various simulated scenari presenting population and/or family structure. Use of ancestral haplotypes resulted in higher power than SNPs on simulated datasets. Application to real data demonstrates the usefulness of the developed model. Esophageal cancer (EC) is a common malignant cancer threatening people's health. There are no universally accepted parameters for its early diagnosis. The aim of this study was to observe the expression of human leukocyte antigen class I (HLA-I) on peripheral blood mononuclear cells (PBMCs) of EC patients and in individuals of high-incidence area of EC so as to evaluate the feasibility of using this parameter as a potential non-invasive biomarker for the early diagnosis of EC. The present study enrolled 58 pathological confirmed EC patients, 46 patients with benign esophageal disease, and 65 healthy volunteers. Expression levels of HLA-I protein and mRNA on PBMCs were determined by flow cytometry and quantitative reverse transcriptase polymerase chain reaction, respectively. Then, 181 volunteers from Lijiadian, a village with high morbidity of EC, and 153 age- and gender-matched health volunteers were involved in this study to observe HLA-I expressions in individuals of high-incidence area of EC. Compared with benign esophageal disease and health volunteers, the expressions of HLA-I protein and mRNA on PBMCs of EC patients are significantly decreased, especially in patients with stage III and IV EC, but was not influenced by patient's age and gender. Furthermore, individuals of high-incidence area of EC also show downregulated HLA-I protein, but not mRNA, expression on PBMCs. Altogether, HLA-I expression on PBMCs of EC patients and individuals from high-incidence area of EC is downregulated, and this parameter might be used as a potential predictor of EC. ABSTRACT Recent observations show that supergiant fast X-ray transients (SFXTs) spend most of their lifetime at an intermediate level luminosity of ∼1033-1034 erg−1, and, when a blackbody model for the spectra is adopted, the resulting radii of the emission region are always only a few hundred metres, supporting the idea that during the intermediate state SFXTs are accreting matter from the companion star. From these observational phenomena we derive possible constraints on the magnetic field strengths of the neutron stars in four SFXTs with known spin periods. While IGR J11215−5952, IGR J16465−4507 and IGR J18483−0311 may have magnetic fields (up to a few 1011-1012 G) similar to those of normal X-ray pulsars, the magnetic field of AX J1841.0−0536 is considerably low (≲1010 G). The high-mass companion stars in SFXTs imply that the neutron stars are relatively young objects, with age less than ∼107 yr. Analysis of the spin evolution shows that neutron stars like AX J1841.0−0536 should be born with relatively long spin periods (≲1 s). Considering the fact that among the four SFXTs only AX J1841.0−0536 is a 'proper' one, and the other three are either 'intermediate' SFXTs or have peculiar characteristics, we suggest that the neutron stars in some of the SFXTs may have similar characteristics as several young central compact objects in supernova remnants called 'anti-magnetars'. These features, combining with accretion from clumpy winds, could make them distinct from standard supergiant X-ray binaries - the low fields and relatively long spin periods guarantee accretion at a very low level, resulting in a large dynamic range (104-105) of X-ray luminosity. We identify 806 ultracool dwarfs (of which 34 are newly discovered L dwarfs) from their Sloan Digital Sky Survey (SDSS) riz photometry and obtain proper motions through cross-matching with the United Kingdom Infrared Telescope Infrared Deep Sky Survey (UKIDSS) and Two-Micron All-Sky Survey (2MASS). Proper-motion and distance constraints show that nine of our ultracool dwarfs are members of widely separated binary systems: SDSS 0101 (K5V+M9.5V), SDSS 0207 (M1.5V+L3V), SDSS 0832 (K3III+L3.5V), SDSS 0858 (M4V+L0V), SDSS 0953 (M4V+M9.5V), SDSS 0956 (M2V+M9V), SDSS 1304 (M4.5V+L0V), SDSS 1631 (M5.5V+M8V) and SDSS 1638 (M4V+L0V). One of these (SDSS 0832) is shown to be a companion to the bright K3 giant η Cancri. Such primaries can provide age and metallicity constraints for any companion objects, yielding excellent benchmark objects. η Cancri AB is the first wide ultracool dwarf + giant binary system identified. We present new observations and analysis that constrain the metallicity of η Cancri A to be near-solar, and use recent evolutionary models to constrain the age of the giant to be 2.2–6.1 Gyr. If η Cancri B is a single object, we estimate its physical attributes to be mass = 63–82 = 1800 ± 150 K, . Its colours are non-typical when compared with other ultracool dwarfs, and we also assess the possibility that η Cancri B is itself an unresolved binary, showing that the combined light of an L4 + T4 system could provide a reasonable explanation for its colours. The replication activity of 2009 pandemic H1N1 influenza virus in human lung cells was evaluated in this study. Twenty-two surgically removed human lung tissue samples were infected ex vivo with pandemic H1N1 influenza virus (A/California/04/2009), seasonal human H1N1 influenza virus (A/Shantou/92/09), or a highly pathogenic H5N1 influenza virus (A/Vietnam/1194/04). Examination of nucleoprotein expression and viral RNA replication in the infected human lung tissue samples showed that whereas the replication of pandemic H1N1 influenza virus varied between tissue samples, overall this virus replicated more efficiently than seasonal H1N1 influenza virus but less efficiently than H5N1 influenza virus. Double-immunostaining for viral antigens and cellular markers indicated that pandemic H1N1 influenza virus replicates in type 2 alveolar epithelial cells. [ABSTRACT FROM AUTHOR]

Aims: To isolate and characterize microbes in the soils containing high contents of phenolics and to dissolve the allelopathic inhibition of plants through microbial degradation. Methods and Results: Four microbes were isolated from plant soils using a screening medium containing p-coumaric acid as sole carbon source. The isolates were identified by biochemical analysis and sequences of their 16S or 18S rDNA, and designated as Pseudomonas putida 4CD1 from rice ( Oryza sativa) soil, Ps. putida 4CD3 from pine ( Pinus massoniana) soil, Pseudomonas nitroreducens 4CD2 and Rhodotorula glutinis 4CD4 from bamboo ( Bambusa chungii) soil. All isolates degraded 1 g l−1 of p-coumaric acid by 70–93% in inorganic and by 99% in Luria-Bertani solutions within 48 h. They also effectively degraded ferulic acid, p-hydroxybenzoic acid and p-hydroxybenzaldehyde. The microbes can degrade p-coumaric acid and reverse its inhibition on seed germination and seedling growth in culture solutions and soils. Low pHs inhibited the growth and phenolic degradation of the three bacteria. High temperature inhibited the R. glutinis. Co2+ completely inhibited the three bacteria, but not the R. glutinis. Cu2+, Al3+, Zn2+, Fe3+, Mn2+, Mg2+ and Ca2+ had varying degrees of inhibition for each of the bacteria. Conclusions: Phenolics in plant culture solutions and soils can be decomposed through application of soil microbes in laboratory or controlled conditions. However, modification of growth conditions is more important for acidic and ions-contaminated media. Significance and Impact of the Study: The four microbes were first isolated and characterized from the soils of bamboo, rice or pine. To identify QTL for feed consumption and feeding behavior traits in pigs, ADFI, feed conversion ratio (FCR), number of visits to the feeder per day (NVD), and average feeding rate (AFR) were recorded in 577 F2 animals from a White Duroc x Chinese Erhualian resource population during the fattening period of 120 to 240 d. A whole genome scan was performed with 183 microsatellites covering the pig genome across the entire resource population. A total of 8 QTL were identified on 5 pig chromosomes, including 3 genomewide significant QTL for FCR on SSC2, 7, and 9, 1 significant QTL for ADFI on SSC3, and 1 for NVD on SSC7. These QTL were identified for the first time, except for the QTL for FCR on SSC2. Four of the 5 significant QTL were adjacent to the known QTL for growth, carcass, and fat deposition traits, supporting the existence of gene(s) with pleiotropic effects on these traits. White Duroc alleles were generally associated with greater phenotypic values, except for those on SSC7 and 9. Comparison of QTL for feed consumption and feeding behaviors indicated that distinct chromosomes had effects on the 2 types of traits. Erosive pustular dermatosis ( EPD) of the leg is an unusual form of amicrobial pustulosis, which typically affects elderly patients with chronic venous insufficiency. We report a patient who presented with painful erythema, crusted erosions and pustules on her right lower leg. Histology showed a dermal infiltration made up predominantly of neutrophils. Bacteria were isolated from the lesions. The condition was initially diagnosed as cellulitis, but antibiotic treatment was unsuccessful. Background The comparative efficacy and tolerability of conventional and biologic treatments for moderate-to-severe plaque psoriasis are unknown. Objectives To perform a systematic review and meta-analysis of randomized controlled trials (RCTs) reporting efficacy of systemic treatments approved for moderate-to-severe psoriasis by means of the Psoriasis Area and Severity Index (PASI). Methods We identified relevant articles by systematic electronic searches (Cochrane Library, Medline, Embase, Scopus). Efficacy was defined as proportion of participants with ≥ 75% decrease in PASI (PASI-75) at primary efficacy measurement (week 8–16). PASI-75 response rates of double-blind placebo-controlled trials were summarized as risk differences (RDs) and pooled using random effects models. Tolerability was assessed from rates of withdrawals and adverse events. Results Twenty-four RCTs totalling 9384 patients were analysed qualitatively. Sixteen double-blind placebo-controlled trials were eligible for meta-analysis. Infliximab was significantly superior to all other interventions [RD 77%, 95% confidence interval (CI) 72–81%]. Adalimumab (RD 64%, 95% CI 61–68%) was superior to ciclosporin (RD 33%, 95% CI 13–52%), efalizumab (RD 24%, 95% CI 19–30%), etanercept 50 mg twice weekly (RD 44%, 95% CI 40–48%) and etanercept 25 mg twice weekly (RD 30%, 95% CI 25–35%). Efalizumab was significantly less efficacious than fumaric acid esters (RD 48%, 95% CI 32–64%). Rates of withdrawals due to adverse events were highest for methotrexate and fumaric acid esters. Conclusions The efficacy of systemic agents approved for moderate-to-severe psoriasis differs considerably both within and between biologics and nonbiologics. Infliximab is most efficacious, followed by adalimumab. Patients receiving infliximab have an excess chance of 77% over placebo to achieve PASI-75 response. The widespread use of the set of multiple-trait derivative-free REML programs for prediction of breeding values and estimation of variance components has led to significant improvement in traits of economic importance. The initial version of this software package, however, was generally limited to pedigree-based relationships. With continued advances in genomic research and the increased availability of genotyping, relationships based on molecular markers are obtainable and desirable. The addition of a new program to the set of multiple-trait derivative-free REML programs is described that allows users the flexibility to calculate relationships using standard pedigree files or an arbitrary relationship matrix based on genetic marker information. We also discuss three factors that affect the detection power in the context of this crossbreed pedigree: being able to detect and correct genotyping errors, increasing marker density for chromosomes with a sparse coverage, and adding individuals to the mapping population as soon as they become available. Kocevski, Ryde & Liang have proposed a semi-empirical function (the KRL function) of gamma-ray burst (GRB) pulses, which could well describe those pulses comprising a fast rise and an exponential decay (FRED) phase. Meanwhile, a theoretical model that could give rise to this kind of pulse, based on the Doppler effect of the expanding fireball surface, has been put forward in detail by Qin and co-workers. To provide a physical explanation to the parameters of the KRL function, we try to fit light curves of the Doppler model with the KRL function so that parameters in both models can be directly related. We pay attention only to single GRB pulses whose profiles are that of FRED and hence employ the sample presented by Kocevski, Ryde & Liang (the KRL sample) to study this issue. We find from our analysis that, for light curves, which arise from exponential rise and exponential decay local pulses, of the Doppler model, the ratio of the rise index r to the decay index d, derived when fitted by the KRL function, increases quickly first. It then remains nearly invariant with the relative width (relative to the time-scale of the initial fireball radius Rc/ c) of local pulses when the width exceeds 2 (the relative width is dimensionless). The rise and decay times of pulses are found to be related to the Lorentz factor by a power law, where the power-law index associated with the rise time is less than that of the decay time, and both are close to −2. In addition, the mean asymmetry shows a slight trend of decreasing with Lorentz factors. In plots of decay indices versus asymmetry, there is a descending phase and after this phase there is a rising portion. We find that these long GRBs of the KRL sample are mainly associated with those light curves arising from comoving pulses with the relative width being larger than 0.1. We show in our analysis that the effect of the comoving pulse shape on the KRL function parameters of the resulting pulses is considerable and can be distinguished by the decay index d when the relative comoving pulse width is less than 2 (when the relative width is larger than 2, it would be difficult to discern the difference in the resulting pulse shapes). SUMMARY Infection by maedi-visna virus, a lentivirus of sheep, leads to chronic inflammatory reactions of various tissues. In this report we have analysed the role of specific cytokines in the disease process. A significant increase in expression of interleukin-6, interleukin-10, granulocyte macrophage-colony stimulating factor (GM-CSF) and transforming growth factor-β1 mRNA was observed in alveolar macrophages isolated from the lungs of naturally infected animals when compared with lungs of seronegative controls. Levels of GM-CSF mRNA expression in alveolar macrophages correlated with the presence of lung lesions, but there was no correlation of interleukin-10, interleukin-6, tumour necrosis factor-α and transforming growth factor-β1 mRNA levels in alveolar macrophages from animals with pulmonary lesions. In vitro investigation showed that GM-CSF in the range 0·1–10ng/ml induced a significant increase in viral p25 production after 7 days in acutely infected blood monocyte-derived macrophages. The production of p25 peaked between 7 and 14 days exposure to 10ng/ml of GM-CSF. Quantitative polymerase chain reaction showed that the level of viral DNA in monocyte-derived macrophages was dose-dependent following GM-CSF treatment in the range 0·1–100ng/ml after 7 days. Viral mRNA expression was also enhanced. These findings indicate a role for GM-CSF in the pathogenesis of lymphoid interstitial pneumonia in infected animals. [ABSTRACT FROM AUTHOR]

SUMMARY Schistosome antigen-driven cytokine responses and antischistosome antibody levels of residents of a Schistosoma japonicum endemic island in Poyang Lake, Jiangxi Province were studied before and 45 days after treatment with praziquantel. IL-4, IL-5, IL-10 and INF-γ were all detected in the supernatants of whole-blood cultures after stimulation with schistosome soluble egg antigen (SEA) and soluble worm antigen preparation (SWAP). The percentages of subjects producing detectable amounts of each cytokine assayed were higher in the group who were negative by stool examination at the start of the study than in those who were initially stool positive. After praziquantel treatment the percentages of subjects producing both type I and type II cytokines increased. This suggests that the production of both types of cytokine was down-regulated in the presence of live, egg-laying S. japonicum adult worms but that this was reversible by treatment. In contrast, the antibody studies showed higher levels of SWAP and SEA-specific antibodies (IgE, total IgG, IgG4, IgM) in subjects who were originally stool-positive than in those who were stool-negative. After treatment specific IgE responses were elevated, but total IgG and IgG4 anti-SEA and IgM anti-SWAP antibody levels all fell significantly. [ABSTRACT FROM AUTHOR]

The lifetimes of three short-lived levels belonging to the 4f[sup 9]6p configuration and of two long-lived levels of the 4f[sup 9]5d configuration of Dy iiihave been measured for the first time using time-resolved laser-induced fluorescence techniques. Motor slowing is a universal feature of human aging, and parkinsonian signs are commonly expressed in human senescence. In the present study, age-associated declines in motor functions in 31 female rhesus monkeys were quantified by activity monitors and an automated test panel, and the incidence of parkinsonian signs was scored using a movement dysfunction assessment scale. Activity levels in middle-aged monkeys (12-17 years old) were less than half that of young animals (5-8 years old) and were further depressed in aged monkeys (21-27 years old). Movement dysfunction scores increased significantly with increasing age. Two or more parkinsonian signs were exhibited by 20% of the middle-aged monkeys and 36% of the aged monkeys. Slowing performance times on fine-motor hand tasks correlated significantly with increasing age. Motor learning was seen in all age groups, but improved faster in the young monkeys. The data suggest that aging rhesus monkeys provide an appropriate model to analyze the biological processes leading to motor slowing and the expression of parkinsonian signs in human senescence. The role of neutrophils in experimental cerebral malaria (ECM) is not well understood. In this study we used a MoAb, RB6-8C5, to deplete the peripheral neutrophils of ECM-susceptible CBA/NSlc mice 24 h before Plasmodium berghei ANKA (PbA) infection. We found that early neutrophil depletion prevented the development of ECM and dramatically decreased the sequestration of monocytes and microhaemorrhage in the brain. The depletion of neutrophils also down-regulated tumour necrosis factor-alpha, interferon-gamma and IL-2 mRNAs and abrogated IL-12p40 mRNA expression in the brain as examined by competitive reverse transcriptase-polymerase chain reaction. Although depletion of neutrophils decreased the expression of Th1 cytokines in both spleen and brain, our results did not show the shift of a Th1 to a Th2 immune response since there was no obvious augmentation of expression of Th2 cytokine mRNAs (IL-4 and IL-10). Five thousand actinomycetes were isolated from soil samples collected from rainforests in Singapore and the generic identities of these isolates were determined by using a procedure that combined morphological, chemotaxonomic and 16S rDNA sequence-based phylogenetic analyses. Actinomycetes belonging to a total of 36 genera were identified. The most abundant isolates are members of Streptomyces, Micromonospora, Actinoplanes, Actinomadura, Nonomuria, Nocardia and Streptosporangium. By phylogenetic analysis of 16S rDNA sequences of our isolates together with those of known actinomycete species, we also evaluated the species diversity of several genera including Streptomyces, Micromonospora, Nonomuria, and Actinomadura. We found that: first, the tropical isolates are present in most clades represented by known species; and second, many tropical isolates form new clades distant from the known species, indicating the presence of unidentified taxa at both species and genus levels. Based on these results, we conclude that actinomycete diversity in the tropical rainforest is very great and should represent an excellent source for discovery of novel bioactive compounds. In this study we have evaluated the prevalence of antibodies against core region peptides (residues 1–28, 21–38 and 51–68), the envelope 1, the non-structural (NS) 4 and 5 proteins of hepatitis C virus (HCV) in sera from 65 chronically HCV-infected patients, 47 with mixed cryoglobulinaemia (MC+) and 18 without (MC-). The major binding sites were located within the core region. Regions 1–28 and 51–68 were recognized by a similar proportion of MC+ and MC- patients, while peptide 21–38 was less frequently detected by samples from MC+ patients (65.5% versus 100%; P = 0.011). The patterns of the reactions showed a minimum of three binding sites: one, located within region 51–68, was shared by both groups; a second determinant was identified at residues 1–21 for MC+ patients and at residues 28–38 for MC- patients; a third, not exactly localized, lay between residues 1 and 38. Recognition of NS5 peptides was not significantly different between MC+ and MC- patients, but while the former mostly reacted either with peptide 1 (residues 2294–2309) (five of 15 sera) or with peptide 2 (residues 2304–2319) (nine of 15 sera), the latter group showed a more scattered reaction. Antibodies to HCV peptides prevalently belonged to IgG1 subclass. However, whereas IgG1 antibodies against peptide 21–38 and peptide 1 of NS5 were more frequently found in MC- rather than in MC+ patients (100% versus 63.8%, P = 0.003, and 22.2% versus 4.2%, P = 0.025, respectively), IgG3 antibodies against region 1–28 were more frequent in MC+ patients (53.19% versus 16.6%, P = 0.0078). The antigenic structure of the envelope 2 (e2) protein of the hepatitis C virus (HCV) was characterized by the use of 70 synthetic peptides and 131 human sera from persons with antibodies to HCV. Among 34 overlapping peptides spanning the e2 protein of HCV, two major antigenic regions were located to residues 484-499 and residues 554-569. The sequence of the two major antigenic regions of the e2 protein are partly well conserved within the described types of HCV. Both regions contain two Cys residues in close proximity, and the region at residues 554-569 contains a putative N-glycosylation site, which are factors that previously have been suggested to affect the immune recognition of the e2 protein. Using substitution peptide analogues where each position within residues 484-499 and 554- 569 were sequentially substituted by Ala or Gly, the most essential residues for antibody binding were found to be the Pro-498, Ala-499, Ala-566, Pro-567, and Pro-568. All of these, except for the Pro-498 and Ala-566, are conserved among different HCV strains. Also, according to previous studies, position 496 often shows variations, which could be explained by position 496 being contained within the antigenic region at residues 484 499. Interestingly, none of the Cys residues at positions 486,494, 564 and 569 were found to be essential for antibody binding, indicating [hat these are not essential in maintaining the e2 antigenicity of the peptides. In a material of 114 confirmed anti-HCV positive sera, derived from patients during the acute or the chronic phase of HCV infection, the prevalence of antibodies to the two major linear antigenic regions of the e2 protein was found to be 55% among HCV RNA-positive sera, and 53% among HCV RNA-negative sera. In conclusion, we have identified and characterized two major linear antigenic regions outside the two hypervariable regions of the e2 protein. Since these regions are accessible to the B cells of the infected host, these two regions are likely to be surface exposed either on the precursor polyprotein or the native e2 protein. The presence of antibodies reactive to a recently cloned host-derived antigen GOR is highly correlated with the presence of antibodies to the hepatitis C virus (HCV). We explored the molecular basis for this observation, and address the following question: are antibodies reactive with GOR19-27 (QKAKSNPNR) a result of a cross-reactivity triggered by the antigenic region at residues 9-17 of HCV core (RKTKRNTNR)? We compared the relative antibody avidity between antibodies reactive to both regions, and determined the residues essential for antibody binding using substitution peptide analogues. Of 96 sera assayed, 60 were found positive for anti-HCV, and of these 55 were found to react with HCV core. Twenty-nine sera were found reactive to the GOR peptide, and these were all reactive to HCV core. In most cases the relative antibody avidity of antibodies reactive to GOR was higher for the HCV core peptide. In 21 of the GOR-reactive sera we were able to determine the essential residues for antibody binding. The essential residues in > 50% of all tested sera coincided with the well conserved residues Lys10, Lys12, Asn14, and Asn16. Also, reactivity to GOR was not related to any certain serotype of antibodies to HCV. Taken together, these findings explain at the molecular level the observed cross-reactivity between these two proteins, since sequence homology per se is not evidence for cross-reactivity. Aims: To isolate, identify and characterize phenolic acid‐degrading bacteria and reduce plant growth inhibition caused by phenolic acids. Methods and Results: A total of 11 bacterial isolates with high phthalic acid (PA)‐degrading ability were obtained using mineral salt medium (MSM) medium containing PA as sole carbon source. These isolates were identified as Arthrobacter globiformis, Pseudomonas putida and Pseudomonas hunanensis by sequence analyses of the 16S rRNA gene. Among them, five Pseudomonas strains could also effectively degrade ferulic acid (FA), p‐hydroxybenzoic acid (PHBA) and syringic acid (SA) in MSM solution. P. putida strain 7 and P. hunanensis strain 10 showed highly efficient degradation of PA, SA, FA and PHBA, and could reduce their inhibition of lily, watermelon, poplar and strawberry seedling growth in soils respectively. These two strains could promote plant growth in soil with phenolic acids. Conclusions: In this study, bacterial strains with highly efficient phenolic acid‐degrading abilities could not only effectively reduce the autotoxicity of phenolic acids on plants but also were able to promote plant growth in soil with phenolic acids. Significance and Impact of the Study: In this study, Pseudomonas can promote plant growth while degrading phenolic acids. Our results provide new choices for the biological removal of autotoxins.
