Severe burns are one of the most devastating injuries, in which sustained inflammation and ischemia often delay the healing process. Pro-angiogenic growth factors such as vascular endothelial growth factor (VEGF) have been widely studied for promoting wound healing. However, the short half-life and instability of VEGF limit its clinical applications. In this study, we develop a photo-crosslinked hydrogel wound dressing from methacrylate hyaluronic acid (MeHA) bonded with a pro-angiogenic prominin-1-binding peptide (PR1P). The materials were extruded in wound bed and in situ formed a wound dressing via exposure to short-time ultraviolet radiation. The study shows that the PR1P-bonded hydrogel significantly improves VEGF recruitment, tubular formation, and cell migration in vitro. Swelling, Scanning Electron Microscope, and mechanical tests indicate the peptide does not affect the overall mechanical and physical properties of the hydrogels. For in vivo studies, the PR1P-bonded hydrogel dressing enhances neovascularization and accelerates wound closure in both deep second-degree burn and full-thickness excisional wound models. The Western blot assay shows such benefits can be related to the activation of the VEGF–Akt signaling pathway. These results suggest this photo-crosslinked hydrogel dressing efficiently promotes VEGF recruitment and angiogenesis in skin regeneration, indicating its potential for clinical applications in wound healing. Panicle-type structure is an important factor affecting rice yield, and an excellent panicle type has become a key indicator in rice breeding. In this study, the indica rice variety R766, which has an open panicle type, was obtained through natural mutation and hybrid selection. Through analyzing differences in panicle structure, panicle yield, and chemical composition between open panicle rice R766 and conventional panicle rice R2928, we found that the angles of the middle and lower branches in R766 were 186.99% and 135.93% greater than those in R2928, respectively. By comparing the grain-filling characteristics of different panicle positions in the two rice varieties, we found that the grain filling at the middle and lower panicle positions of R2928 was significantly lower, accompanied by an increase in the percentage of empty spikelets. However, in R766, the grain-filling rates in the middle and lower panicle positions were consistent with those in the upper panicle position, with significantly higher rates of grain filling and grain plumpness in the middle and lower panicle positions than in R2928. The empty grain rate at the lower panicle position of R766 was 15.25% lower than that of R2928, and the grain filling was 24.75% higher than that of R2928. Additionally, the variation in the 1000-grain weight of grains at different panicle positions in R766 was relatively small, with decreases of 1.55% and 0.38% in the middle and lower panicle positions, respectively, compared with the upper position, whereas R2928 showed decreases of 5.99% and 7.12% in the 1000-grain weight of grains at the middle and lower panicle positions, respectively, compared with the upper position. The cellulose content in the stems of R766 was 7.51% higher than that of R2928, with no significant difference in the cellulose content in the panicle axis and primary branches compared to R2928. The lignin content of the panicle axis in R766 was 8.03% higher than that in R2928, whereas there was no significant difference between the lignin content of the stems and primary branches. This preliminary study revealed the open panicle characteristics of R766 and the reasons for its high basal grain setting rate. This study provides a reference for promoting this open panicle-type indica rice variety to improve yield and disease resistance in environments with high humidity and low sunlight levels. CO2 huff-n-puff is an important method for the development of shale oil reservoirs. In this study, HPMI and NMR technology was used to characterize the pore distribution of the cores. The CO2 huff-n-puff experiment experiments were conducted to study the effects of injection pressure, soaking time, and heterogeneity on the CO2 huff-n-puff. The results showed that the Jimsar core pores are predominantly nanopores. Mesopores with a pore radius between 2 nm and 50 nm accounted for more than 70%. CO2 huff-n-puff has been shown to effectively enhance shale oil recovery. When the injection pressure was greater than the miscible pressure, higher injection pressures were able to improve the recovery of macropores, particularly in cores with higher permeability. Appropriately extending the soaking time enhanced the diffusion of CO2 in the mesopores, and the recovery increased to above 10%. Determining the optimal soaking time is crucial to achieve maximum CO2 huff-n-puff recovery. Artificial fractures can enhance the recovery of mesopores around them, resulting in core recovery of up to 60%. However, artificial fractures exacerbate reservoir heterogeneity and reduce the CO2 huff-n-puff recovery of matrix. Increasing the cycles of CO2 huff-n-puff can effectively reduce the impact of heterogeneity on the recovery of matrix. In conclusion, expanding the area of the fracturing transformation zone and selecting the appropriate injection pressure and soaking time for the multiple cycles of CO2 huff-n-puff can effectively improve the recovery of shale oil reservoirs. Sulfide is a toxic and hazardous substance in the agricultural environment, which can cause damage to humans and livestock when exposed to large amounts of air. In this study, we performed one-factor optimization of the culture conditions and culture fractions of the Cellulosimicrobium sp. strain L1 and combined it with a biological trickling filter cell for the degradation of hydrogen sulfide for 24 consecutive days. The degradation effect of strain L1 and the biological trickling filter (BTF) on hydrogen sulfide was investigated, and the changes in intermediate products in the degradation process were briefly analyzed. The results showed that strain L1 had the highest conversion efficiency when incubated with 3 g/L sucrose as the carbon source and 1 g/L NH4Cl as the nitrogen source at a temperature of 35 °C, an initial pH of 5, and a NaCl concentration of 1%. The concentration of thiosulfate increased and then decreased during the degradation process, and the concentration of sulfate increased continuously. When strain L1 was applied to the biological trickling filter, it could degrade 359.53 mg/m3 of H2S. This study provides a deeper understanding of sulfide degradation in biological trickling filters and helps promote the development of desulfurization technology and the treatment of malodorous gasses produced by the accumulation of large quantities of livestock manure. Most older individuals prefer to age in place during their later years; however, achieving this aspiration presents significant challenges. Naturally Occurring Retirement Communities (NORCs) represent a potential option for promoting healthy aging, both from the perspective of meeting seniors' real needs and cost-effectiveness. This article aims to analyze the distribution of NORCs in Australia and compares census data from 2011 to 2021 to understand the overall distribution patterns and changes across the nation, by providing a localized analysis of the hotspot distribution of NORCs in eight Greater Capital Cities. The study employs methods of geovisualization, Global Moran's I, and Getis-Ord Gi* analysis to examine the spatial correlations and clustering effects of NORCs. The results indicate that NORCs are rapidly growing in Australia, with their distribution primarily influenced by sea change and urbanization. Understanding the trends in NORC distribution can assist the government in developing effective and localized policies and interventions to help older Australians to better age in place. Chicken egg white (EW) proteins possess various useful techno-functionalities, including foaming, gelling or coagulating, and emulsifying. The gelling property is one of the most important functionalities of EW proteins, affecting their versatile applications in the food and pharmaceutical industries. However, it is challenging to develop high-quality gelled foods and innovative nutraceutical supplements using native EW and its proteins. This review describes the gelling properties of EW proteins. It discusses the development and action mechanism of the physical, chemical, and biological methods and exogenous substances used in the modification of EW gels. Two main applications of EW gels, i.e., gelling agents in foods and gel-type carriers for nutraceutical delivery, are systematically summarized and discussed. In addition, the research and technological gaps between modified EW gels and their applications are highlighted. By reviewing the new modification strategies and application trends of EW gels, this paper provides insights into the development of EW gel-derived products with new and functional features. Myeloid-derived suppressor cells (MDSCs) play an essential role in suppressing the antitumor activity of T lymphocytes in solid tumors, thus representing an attractive therapeutic target to enhance the efficacy of immunotherapy. However, the differences in protein expression between MDSCs and their physiological counterparts, particularly polymorphonuclear neutrophils (PMNs), remain inadequately characterized, making the specific identification and targeting of MDSCs difficult. PMNs and PMN-MDSCs share markers such as CD11b+CD14−CD15+/CD66b+, and some MDSC-enriched markers are emerging, such as LOX-1 and CD84. More proteomics studies are needed to identify the signature and markers for MDSCs. Recently, we reported the induced differentiation of isogenic PMNs or MDSCs (referred to as iPMNs and iMDSCs, respectively) from the human promyelocytic cell line HL60. Here, we profiled the global proteomics and membrane proteomics of these cells with quantitative mass spectrometry, which identified a 41-protein signature ("cluster 6") that was upregulated in iMDSCs compared with HL60 and iPMN. We further integrated our cell line-based proteomics data with a published proteomics dataset of normal human primary monocytes and monocyte-derived MDSCs induced by cancer-associated fibroblasts. The analysis identified a 38-protein signature that exhibits an upregulated expression pattern in MDSCs compared with normal monocytes or PMNs. These signatures may provide a hypothesis-generating platform to identify protein biomarkers that phenotypically distinguish MDSCs from their healthy counterparts, as well as potential therapeutic targets that impair MDSCs without harming normal myeloid cells. New and efficient sensors of nerve agents are urgently demanded to prevent them from causing mass casualties in war or terrorist attacks. So, in this work, a novel hierarchical nanoheterostructure was synthesized via the direct growth of α-Fe2O3 nanorods onto multiwall carbon nanotube (MWCNT) backbones. Then, the composites were functionalized with hexafluoroisopropanol (HFIP) and successfully applied to detect dimethyl methylphosphonate (DMMP)-sarin simulant gas. The observations show that the HFIP-α-Fe2O3@MWCNT hybrids exhibit outstanding DMMP-sensing performance, including low operating temperature (220 °C), high response (6.0 to 0.1 ppm DMMP), short response/recovery time (8.7 s/11.9 s), as well as low detection limit (63.92 ppb). The analysis of the sensing mechanism demonstrates that the perfect sensing performance is mainly due to the synergistic effect of the chemical interaction of DMMP with the heterostructure and the physical adsorption of DMMP by hydrogen bonds with HFIP that are grafted on the α-Fe2O3@MWCNTs composite. The huge specific surface area of HFIP-α-Fe2O3@MWCNTs composite is also one of the reasons for this enhanced performance. This work not only offers a promising and effective method for synthesizing sensitive materials for high-performance gas sensors but also provides insight into the sensing mechanism of DMMP. Malignant pleural mesothelioma (MPM) is a rare and aggressive cancer affecting the pleural lining of the lungs. Celastrol (Cela), a pentacyclic triterpenoid, has demonstrated promising therapeutic potential as an antioxidant, anti-inflammatory, neuroprotective agent, and anti-cancer agent. In this study, we developed inhaled surface-modified Cela-loaded poly(lactic-co-glycolic) acid (PLGA) microparticles (Cela MPs) for the treatment of MPM using a double emulsion solvent evaporation method. The optimized Cela MPs exhibited high entrapment efficiency (72.8 ± 6.1%) and possessed a wrinkled surface with a mean geometric diameter of ~2 µm and an aerodynamic diameter of 4.5 ± 0.1 µm, suggesting them to be suitable for pulmonary delivery. A subsequent release study showed an initial burst release up to 59.9 ± 2.9%, followed by sustained release. The therapeutic efficacy of Cela MPs was evaluated against four mesothelioma cell lines, where Cela MP exhibited significant reduction in IC50 values, and blank MPs produced no toxicity to normal cells. Additionally, a 3D-spheroid study was performed where a single dose of Cela MP at 1.0 µM significantly inhibited spheroid growth. Cela MP was also able to retain the antioxidant activity of Cela only while mechanistic studies revealed triggered autophagy and an induction of apoptosis. Therefore, these studies highlight the anti-mesothelioma activity of Cela and demonstrate that Cela MPs are a promising inhalable medicine for MPM treatment. [ABSTRACT FROM AUTHOR]

Non-small cell lung cancer (NSCLC) is a global concern as one of the leading causes of cancer deaths. The treatment options for NSCLC are limited to systemic chemotherapy, administered either orally or intravenously, with no local chemotherapies to target NSCLC. In this study, we have prepared nanoemulsions of tyrosine kinase inhibitor (TKI), erlotinib, using the single step, continuous manufacturing, and easily scalable hot melt extrusion (HME) technique without additional size reduction step. The formulated nanoemulsions were optimized and evaluated for their physiochemical properties, in vitro aerosol deposition behavior, and therapeutic activity against NSCLC cell lines both in vitro and ex vivo. The optimized nanoemulsion showed suitable aerosolization characteristics for deep lung deposition. The in vitro anti-cancer activity was tested against the NSCLC A549 cell line which exhibited 2.8-fold lower IC50 for erlotinib-loaded nanoemulsion, as compared to erlotinib-free solution. Furthermore, ex vivo studies using a 3D spheroid model also revealed higher efficacy of erlotinib-loaded nanoemulsion against NSCLC. Hence, inhalable nanoemulsion can be considered as a potential therapeutic approach for the local lung delivery of erlotinib to NSCLC. [ABSTRACT FROM AUTHOR]

(1) Background: Sheath blight is one of the most economically significant rice diseases worldwide. A study was conducted in order to find the relationship and impact of the amount of nitrogen (N) and phosphorus (P) application on the hybrid rice population microclimate and the severity of sheath blight. (2) Methods: Four N and four P application levels were used to determine their impact on plant type, temperature, and humidity variation in different positions of population and the severity of sheath blight in the later stage. (3) Results: We found that N and P application levels could affect the plant type and change the population temperature and humidity by increasing the leaf length and leaf angle. (4) Conclusions: N application had a more significant (p < 0.05) impact on the plant type. High N application caused decreased temperature (hybrid rice population), while increased humidity (especially the population base layer at grain filling stage) resulted in severe sheath blight. High P application had similar impacts; however, P application increased material and nitrogen transport in plants and reduced the severity of sheath blight. [ABSTRACT FROM AUTHOR]

Compound droughts and heatwaves (CDHWs) are likely to cause more severe natural disasters than a single extreme event, and they have been exacerbated by rapid global warming. Based on high-resolution grid data, this study combines the daily-scale ERA5-Land dataset and the monthly-scale SPEI dataset with multiple indicators to analyze CDHWs. We calculated and analyzed the temporal and spatial modal distribution of CDHWs in Central Asia from 1981 to 2018, and in this paper, we discuss the sequence relationship between drought events, heatwave events, and CDHWs. The results show that the number of CDHWs in the study region have increased over time and expanded in terms of area, especially in eastern and southwestern Central Asia. The tsum (total frequency of CDHWs) was 0.5 times higher than the total heatwave frequency and it increased at a rate of 0.17/yr. The maximum duration of tmax (maximum duration of CDHWs in days) was 17 days. Furthermore, the occurrence rate of tmax was 96.67%, and the AH (CDHWs' accumulated heat) had a rate of 97.78%, which, upon examination of the spatial trend pattern, accounted for the largest increase in terms of area. We also found that the TAH (CDHWs' average temperature anomalies, SPEI < −0.5) shows obvious seasonality, with the increases in winter and spring being significantly greater than the increases in summer and autumn. The intensity of the CDHWs was stronger than that of a single extreme event, the temperature anomaly was higher than the average of 0.4–0.8 °C, and there was a north–south spatial pattern across the study region. In eastern and northwestern Central Asia, the AH and heatwaves (SPEI < −0.5) increased by 15–30 times per year on average. During the transition from the base period to the reference period, CDHWs increased by 25%, and the number of dry days prior to the CDHWs decreased by 7.35 days. As an alternative to ageing at home in the community, naturally occurring retirement communities (NORCs) have great potential to facilitate ageing in place; however, they have not attracted much research attention. This study conducts an overview of NORCs, aiming to examine the previous research in a comprehensive manner in order to explore how NORCs impact ageing in place, with the goal of guiding future research. The research presented here employs the content analysis method to review prior NORC-related studies and categorise research themes and findings following top-down coding principles. A total of 49 articles were selected from the Scopus and Web of Science databases, and the results show that the "social environment", which was the most discussed topic (n = 24), provides the necessary mental support and physical motivation for older adults to live actively in NORCs, and that NORCs play a positive role in preserving public resources and promoting individual health. The limitations of this study include the fact that there is little public information on NORC programs and the subjective classification of themes, among others. This study acts as a foundation for future research on NORCs, which serve as a perfect model for healthy ageing in place. [ABSTRACT FROM AUTHOR]

The inactivation of von Hippel–Lindau (VHL) is critical for clear cell renal cell carcinoma (ccRCC) and VHL syndrome. VHL loss leads to the stabilization of hypoxia-inducible factor α (HIFα) and other substrate proteins, which, together, drive various tumor-promoting pathways. There is inadequate molecular characterization of VHL restoration in VHL-defective ccRCC cells. The identities of HIF-independent VHL substrates remain elusive. We reinstalled VHL expression in 786-O and performed transcriptome, proteome and ubiquitome profiling to assess the molecular impact. The transcriptome and proteome analysis revealed that VHL restoration caused the downregulation of hypoxia signaling, glycolysis, E2F targets, and mTORC1 signaling, and the upregulation of fatty acid metabolism. Proteome and ubiquitome co-analysis, together with the ccRCC CPTAC data, enlisted 57 proteins that were ubiquitinated and downregulated by VHL restoration and upregulated in human ccRCC. Among them, we confirmed the reduction of TGFBI (ubiquitinated at K676) and NFKB2 (ubiquitinated at K72 and K741) by VHL re-expression in 786-O. Immunoprecipitation assay showed the physical interaction between VHL and NFKB2. K72 of NFKB2 affected NFKB2 stability in a VHL-dependent manner. Taken together, our study generates a comprehensive molecular catalog of a VHL-restored 786-O model and provides a list of putative VHL-dependent ubiquitination substrates, including TGFBI and NFKB2, for future investigation. Extreme weather and global warming are drastic agricultural issues causing water scarcity. Under these conditions, it is vital to select drought-tolerant hybrid super rice breeds to enhance the rice yield in dry areas. In the current study, a dry-shed experiment was carried out in 2019–2020 to analyze the root growth, gene expression, and enzyme activity of hybrid rice Deyou4727, its parent (Dexiang074B * Chenghui727), taking IR64 as a universal control. Results showed that Deyou4727 and its maintainer line Dexiang074B produced more roots (30–50 cm soil) and higher enzymatic activity than IR64 during the drought stress period, indicating a good drought-tolerant capacity for Deyou4727. The distribution and morphology of the root system in Deyou4727 were inherited by the maintainer line Dexiang074B, which was confirmed by the high upregulation of DRO1 and OsPIN1b gene expression and the sharing of the same ORF region in both the hybrid rice varieties. However, the DRO1 gene ORF study showed that, in the case of the control IR64, 'A' was missing at the 4th exon of the DRO1 gene, which may explain the shallow and weak root architecture system of IR64. Caffeoyl shikimate esterase (CSE) hydrolyzes caffeoyl shikimate into caffeate and shikimate in the phenylpropanoid pathway. In this study, we performed a systematic analysis of the CSE gene family and investigated the possible roles of CSE and CSE-like genes in Populus. We conducted a genome-wide analysis of the CSE gene family, including functional and phylogenetic analyses of CSE and CSE-like genes, using the poplar (Populus trichocarpa) genome. Eighteen CSE and CSE-like genes were identified in the Populus genome, and five phylogenetic groups were identified from phylogenetic analysis. CSEs in Group Ia, which were proposed as bona fide CSEs, have probably been lost in most monocots except Oryza sativa. Primary functional classification showed that PoptrCSE1 and PoptrCSE2 had putative function in lignin biosynthesis. In addition, PoptrCSE2, along with PoptrCSE12, might also respond to stress with a function in cell wall biosynthesis. Enzymatic assay of PoptoCSE1 (Populus tomentosa), -2 and -12 showed that PoptoCSE1 and -2 maintained CSE activity. PoptoCSE1 and 2 had similar biochemical properties, tissue expression patterns and subcellular localization. Most of the PoptrCSE-like genes are homologs of AtMAGL (monoacylglycerol lipase) genes in Arabidopsis and may function as MAG lipase in poplar. The widespread use of antibiotics in large-scale livestock production has led to serious antibiotic resistance. Proteus mirabilis is an important pathogenic bacterium on large-scale farms. Chromosomally localized mobilizable genetic elements (genomic islands) and mobile genetic elements (Tn7-like transposons) play an important role in the acquisition and transmission of resistance genes by P. mirabilis. To study the prevalence and resistance characteristics of antibiotic-resistant genomic islands in P. mirabilis of animal origin in China, we performed whole genome sequencing of P. mirabilis isolated from large-scale pig and chicken farms. Three new variants of PmGRI1 (HN31, YN8, and YN9), and a hybrid structure (HN2p) formed by the multidrug-resistant Tn7-like-HN2p transposon and a genomic island PmGRI1-HN2p, were identified from P. mirabilis. All variants underwent homologous recombination mediated by insertion sequence IS26. A genomic rearrangement in the chromosome between the Tn7-like-HN2p transposon and PmGRI1-HN2p occurred in HN2p. The heterozygous structure contained various antimicrobial resistance genes, including three copies of fluoroquinolone resistance gene qnrA1 and 16S rRNA methylase gene rmtB, which are rarely found in P. mirabilis. Our results highlight the structural genetic diversity of genomic islands by characterizing the novel variants of PmGRI1 and enrich the research base of multidrug resistance genomic islands. Dehydrins (DHN) belong to the late embryogenesis abundant II family and have been found to enhance plant tolerance to abiotic stress. In the present study, we reported four DHNs in Larix kaempferi (LkDHN) which were identified from the published transcriptome. Alignment analysis showed that these four LkDHNs shared close relationships and belonged to SK3-type DHNs. The electrophoretic mobility shift assay indicated that these four LkDHNs all possess sequence-independent binding capacity for double-strands DNAs. The subcellular localizations of the four LkDHNs were in both the nucleus and cytoplasm, indicating that these LkDHNs enter the nucleus to exert the ability to bind DNA. The preparation of tobacco protoplasts with different concentrations of mannitol showed that LkDHNs enhanced the tolerance of plant cells under osmotic stress. The overexpression of LkDHNs in yeasts enhanced their tolerance to osmotic stress and helped the yeasts to survive severe stress. In addition, LkDHNs in the nucleus of salt treated tobacco increased. All of these results indicated that the four LkDHNs help plants survive from heavy stress by participating in DNA protection. These four LKDHNs played similar roles in the response to osmotic stress and assisted in the adaptation of L. kaempferi to the arid and cold winter of northern China. [ABSTRACT FROM AUTHOR]

Caffeoyl shikimate esterase (CSE) has been reported to be involved in lignin biosynthesis; however, studies of CSE in gymnosperms are lacking. In this study, CSE was successfully cloned from Larix kaempferi (LkCSE) based on Larix laricina transcriptome screening. LkCSE was likely to have catalytic activity based on homologous sequence alignment and phylogenetic analyses of CSEs from different species. In vitro assays with the recombinant enzyme validated the catalytic activity of LkCSE, indicating its function in converting caffeoyl shikimate into caffeate and shikimate. Additionally, the optimum reaction pH and temperature of LkCSE were determined to be 6.0 and 30 °C, respectively. The values of Km and Vmax of CSE for caffeoyl shikimate were 98.11 μM and 14.44 nM min−1, respectively. Moreover, LkCSE was observed to have tissue expression specificity and was abundantly expressed in stems and leaves, especially stems, which was 50 times higher than the expression levels of roots. Lastly, translational fusion assays using LkCSE fused with green fluorescent proteins (GFP) in tobacco leaves indicated that LkCSE was localized in the plasma membrane and endoplasmic reticulum (ER). These results revealed that CSE clearly functions in gymnosperms and it is possible for LkCSE to interact with other ER-resident proteins and regulate mass flux in the monolignol biosynthesis pathway. Incorporating oilseed-rape straw in soil is one of the effective methods for enhancing the use efficiency of agricultural resources in the rape-rice rotation system. However, the impacts of oilseed-rape straw incorporation on root growth and dynamic changes in soil are still unclear. In order to provide a deeper understanding of the oilseed rape straw return on rice growth and productivity, the experiment was conducted in the field and in a specially-designed pots system from 2016 to 2017 by means of two straw returning methods and four straw returning amounts. In the early stage of rice growth (0–36 days after rice transplanting) the straw returning treatments decreased 1.0–8.6 mg/plant in bleeding density and 0.10–6.11, 0.06–0.31, and 0.52–0.84 μmol/(g h) in the activity of glutamine synthetase (GS), glutamic-oxalacetic transaminase (GOT), and glutamic-pyruvic transaminase (GPT), respectively. Oilseed rape straw returned by mulching induced negative impacts on new germinating roots in 0–10 cm of soil, while the negative impacts were observed on roots in 10–30 cm of soil for straw returned by plowing. In the later stage of rice growth (56–75 days after rice transplanting), oilseed rape straw returning produced some positive impacts on rice roots, which enhanced the yield of rice. Conclusively, our findings suggested that dynamic root growth and the activity of root enzymes are two major factors behind the slow reviving of rice after transplanting in the straw returning field. Drought is one of the major factors limiting rice yield worldwide. A total of 46 hybrid rice varieties were chosen to investigate their root distribution and their response to drought. A field experiment was carried out in a dry shed building to evaluate the drought tolerance capacity of hybrid rice varieties on the basis of CTIRDE (complex tolerance index of rice under drought environment) values. Next, the experiment was conducted in a specially designed pot system and seed bags to analyze the root distribution and activity of antioxidant enzymes in different rice varieties. Moreover, the DEEPER ROOTING 1 (DRO1) gene was sequenced to elucidate its role in the root distribution of typical rice varieties. On the basis of CTIRDE values, the 46 hybrid rice varieties were classified as tolerant (CTIRDE ≥ 0.75), semi-tolerant (0.75 > CTIRDE > 0.65), or sensitive (CTIRDE ≤ 0.65) to drought stress. The tolerant varieties (Chuanguyou208 and Deyou4727) displayed a significantly larger length, had higher number and weight of roots in the 30–50 cm soil layer, and exhibited a significantly higher activity of Superoxide dismutase (SOD) and Peroxidase (POD) enzymes in roots during the drought stress period. The DRO1 gene sequencing results revealed that the tolerant and sensitive varieties exhibited a single-nucleotide polymorphism (SNP) in the 3-exon region, and the tolerant varieties (Chuanguyou208 and Deyou4727) exhibited a larger root growth angle with the horizontal axis, hence developing a deeper root system as compared with the other two group varieties. A significant correlation was found not only between the DRO1 gene and root distribution but also between DRO1 and the activity of SOD and POD enzymes. Conclusively, as a key feature, a deep root system enabled tolerant rice varieties (Chuanguyou208 and Deyou4727) to avoid drought stress by absorbing more water stored in deep soil layers. The root distribution, activity of POD and SOD enzymes in roots, and DRO1 gene can be used to screen tolerant rice varieties that can survive better under drought stress during the seedling stage of rice growth. Light is one of the most important environmental factors that affect many aspects of plant growth, including chlorophyll (Chl) synthesis and flowering time. Here, we identified a rice mutant, yellow leaf and early flowering (ye1), and characterized the gene YE1 by using a map-based cloning method. YE1 encodes a heme oxygenase, which is localized to the chloroplasts. YE1 is expressed in various green tissues, especially in leaves, with a diurnal-rhythmic expression pattern, and its transcripts is also induced by light during leaf-greening. The mutant displays decreased Chl contents with less and disorderly thylakoid lamellar layers in chloroplasts, which reduced the photosynthesis rate. The early flowering phenotype of ye1 was not photoperiod-sensitive. Furthermore, the expression levels of Chl biosynthetic genes were downregulated in ye1 seedlings during de-etiolation responses to light. We also found that rhythmic expression patterns of genes involved in photoperiodic flowering were altered in the mutant. Based on these results, we infer that YE1 plays an important role in light-dependent Chl biogenesis as well as photoperiodic flowering pathway in rice. [ABSTRACT FROM AUTHOR]

Larix olgensis is an economically important tree species native to northeastern China. The use of somatic embryogenesis (SE) is efficient and enables the rapid production of varieties with desirable qualities. Here, isobaric labeling via tandem mass tags was used to conduct a large-scale quantitative proteomic analysis of proteins in three critically important stages of SE in L. olgensis : the primary embryogenic callus, the single embryo, and the cotyledon embryo. We identified 6269 proteins, including 176 shared differentially expressed proteins across the three groups. Many of these proteins are involved in glycolipid metabolism, hormone response/signal transduction, cell synthesis and differentiation, and water transport; proteins involved in stress resistance and secondary metabolism, as well as transcription factors, play key regulatory roles in SE. The results of this study provide new insights into the key pathways and proteins involved in SE in Larix . Our findings have implications for the expression of totipotency, the preparation of synthetic seeds, and genetic transformation.

The performance of a self-flocculating microalga Chlorococcum sp. GD on the flocculation, growth, and lipid accumulation in wastewater with different ammonia nitrogen concentrations was investigated. It was revealed that relative high ammonia nitrogen concentration (20-50 mg·L -1 ) was beneficial to the flocculation of Chlorococcum sp. GD, and the highest flocculating efficiency was up to 84.4%. It was also found that the highest flocculating efficiency occurred in the middle of the culture (4-5 days) regardless of initial ammonia concentration in wastewater. It was speculated that high flocculating efficiency was likely related to the production of extracellular proteins. 20 mg·L -1 of ammonia was found to be a preferred concentration for both biomass production and lipid accumulation. 92.8% COD, 98.8% ammonia, and 69.4% phosphorus were removed when Chlorococcum sp. GD was cultivated in wastewater with 20 mg·L -1 ammonia. The novelty and significance of the investigation was the integration of flocculation, biomass production, wastewater treatment, and lipid accumulation, simultaneously, which made Chlorococcum sp.
