Sulfur (S) fertilization and rhizobium inoculation are important agronomic practices for improving soybean growth and yield. However, their combined effects on yield components and the resultant impacts on yield need further investigation. Our study aimed to verify the combined effects of S fertilizer and rhizobium inoculant on the yield components and seed yield of soybeans, as well as photosynthesis dynamics during the V5–R6 growth stages. A randomized block design incorporating two factors was employed for field experiments on soybean, involving the application of five rates of sulfur (0, 5.0, 9.8, 14.8, and 19.6 kg S ha−1) with rhizobium inoculation or without. A reduction of 50% in mineral fertilizer according to farmer practice (FP) was applied alongside different rates of sulfur for field experiments on soybeans. The findings indicated that the application of sulfur fertilization significantly enhanced soybean yield by increasing the number of grains per plant (NG) and the number of pods per plant (NP). Maximum grain yield was reached under treatment with an S rate of 19.6 kg S ha−1, which increased NG by 39% more than an S rate of 0 kg S ha−1. Correlation analysis indicated that higher photosynthesis was closely associated with increased yield components. This study demonstrated that applying S fertilizers could improve soybean production by combining the appropriate sulfur concentration and rhizobia inoculation. [ABSTRACT FROM AUTHOR]

D-glucaric acid is an important bio-based building block of polymers and is a high value-added chemical that can be used in a variety of applications. In the present study, the Udh target genes from Pseudomonas putida and Pseudomonas syringae were used together to construct the expression vector pETDuet-2 × Udh. The transformants of BL21 (DE3) with vector pETDuet-2 × Udh were applied to produce glucaric acid from glucuronic acid. After optimizing the induction conditions, the highest Udh expression was achieved when 0.4 mmol·L−1 isopropyl-β-d–thiogalactoside (IPTG) was added to the cell cultures at an OD600 value of 0.6 followed by culturing at 26 °C for 6 h. The production of glucaric acid substantially reached 5.24 ± 0.015 g·L−1 in fed-batch cultures in a 30 L tank. Simple Summary: In order to increase the growth performance and make up for the lack of high-quality roughage, farmers are accustomed to adopting high-concentration diets during the fattening period of ruminants. The long-term consumption of high-concentration diets induces a series of nutritional metabolic diseases. In production practice, the major mutton sheep fattening region (e.g., Jiangsu, China) generally uses diets with a concentrate ratio of up to 80% to 90% during the fattening period, and reports of cases of nutritional metabolic diseases are also rare. This seems to indicate that sheep and goats are more adaptable to high-concentration diets than cattle. Our work shows that high-concentrate diets can significantly promote the digestion of nutrients; the liver enhances the adaptability of goats to high-concentration diets by regulating the expression of genes involved in nutrient metabolism and toxin clearance. Therefore, the liver plays a vital role in the adaptation of ruminants to high-concentration diets. The results here can lay the foundation for the rational application of high-concentration diets in production. The objective of this study was to determine the effect of high-concentrate diets on the blood parameters and liver transcriptome of goats. Eighteen goats were allocated into three dietary treatments: the high level of concentrate (HC) group, the medium level of concentrate (MC) group, and the low level of concentrate (LC) group. The blood parameters and pathological damage of the gastrointestinal tract and liver tissues were measured. In hepatic portal vein blood, HC showed higher LPS, VFAs, and LA; in jugular vein blood, no significant differences in LPS, VFAs, and LA were recorded among groups (p > 0.05). Compared to the LC and MC groups, the HC group showed significantly increased interleukin (IL)-1β, IL-10, TNF-α, and diamine oxidase in jugular vein blood (p < 0.05). Liver transcriptome analysis discovered a total of 1269 differentially expressed genes (DEGs) among the three groups and most of them came from the HC vs. LC group. There were 333 DEGs up-regulated and 608 down-regulated in the HC group compared to the LC group. The gene ontology enrichment analysis showed that these DEGs were mainly focused on the regulation of triacylglycerol catabolism, lipoprotein particle remodeling, and cholesterol transport. The Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed that the liver of the HC group enhanced the metabolism of nutrients such as VFAs through the activation of AMPK and other signaling pathways and enhanced the clearance and detoxification of LPS by activating the toll-like receptor signaling pathway. Although crop rotations have been widely shown as an effective approach for improving yield or soil quality in the long term, the relationship between crop rotations and quality traits of crop or biochar-based fertilization is still unclear. To address this, we conducted a long-term field experiment in the Heilongjiang province of China to investigate the effects of crop rotation and biochar-based fertilizer application on the crop yield, soil parameters, crop quality, and agronomic traits in 2014–2020. The effects of rotations on crop production and soil properties were evaluated with the average yield variability during the 7 years of this study. Our results showed that the diversified corn and soybean rotations had a significant positive effect on average crop yield compared with their monocultures. The corn yield was enhanced by 0.6 Mg ha−1 (5.4%) in the corn–soybean–corn (CSC) crop sequence compared with monoculture corn. Similarly, soybean yield was enhanced by 0.21 Mg ha−1 (9.7%) in the soybean–corn–corn (SCC) crop sequence compared with monoculture soybean. However, a negative effect of crop rotations was detected on the protein content of soybean compared with the monoculture soybean, while a positive effect was detected on oil content. Additionally, no differences were detected in crop yield between biochar-based fertilization and mineral fertilization treatments, but a significant positive effect of biochar-based fertilization was observed for any crop on both protein and oil content. A significant effect of crop rotation was found on the percentage of total soil N (TN), available soil N (AN), and available soil K (AK) content. The SSS crop sequence treatment illustrated the highest TN values at 0.18%. The CCC crop sequence treatment increased AN and AK content by 9.1% and 7.8%, respectively, compared with SSS (p < 0.05). We conclude that crop rotations increase crop yield and biochar-based fertilizer application, improving crop quality traits in the long term. Thus, the addition of biochar-based fertilizer could efficiently enhance the yield and quality of crop in the rotation cropping system. Simple Summary: The specific enzymes secreted by microorganisms in the gastrointestinal tract (GIT) of ruminants, such as phytase, can catalyze the decomposition of calcium compounds (e.g., phytic acid) and release bound calcium for the absorption of animals. Therefore, we speculate that gastrointestinal microbes could be a factor affecting digestion and absorption of dietary calcium. However, little related research has been reported. In the present study, we found that the true digestibility of calcium (TDC) in goats is related to gastrointestinal bacteria. Some gastro-intestinal bacteria, such as ruminal Prevotella, were beneficial for true host digestibility of dietary calcium. The current study was performed to examine the relationship between the true digestibility of calcium (TDC) in the diet and bacterial community structure in the gastrointestinal tract (GIT) of goats. Twenty-six Nubian healthy female goats were selected as experimental animals, and their TDC was determined using metabolic experiments. Eight goats were grouped into the high digestibility of Calcium (HC) phenotype, and another eight were grouped into the low digestibility of Calcium (LC) phenotype. Their bacterial 16S rRNA gene amplicons from the rumen, abomasum, jejunum, cecum, and colon contents were sequenced using next-generation high-throughput sequencing technology. In the rumen, 239 genera belonging to 23 phyla, 319 genera belonging to 30 phyla in the abomasum, 248 genera belonging to 36 phyla in the jejunum, 248 genera belonging to 25 phyla in the colon and 246 genera belonging to 23 phyla in the cecum were detected. In addition, there was a significant correlation between the TDC and the relative abundance of Candidatus_Saccharimonas, Christensenellaceae_R-7_group, Mogibacterium, Prevotella_1, Prevotella_UCG_004, Ruminococcus_2, Saccharibacteria in the rumen, Eubacterium_coprostanoligens_group, Lachnospiraceae_ND3007_group, Lachnospiraceae_NK3A20_group, p-1088-a5_gut_group, and Planctomycetes in the abomasum, Butyrivibrio in the cecum, and Fibrobacter in the cecum were observed.
