Brain rhythms provide the timing for recruitment of brain activity required for linking together neuronal ensembles engaged in specific tasks. The γ-oscillations (30 to 120 Hz) orchestrate neuronal circuits underlying cognitive processes and working memory. These oscillations are reduced in numerous neurological and psychiatric disorders, including early cognitive decline in Alzheimers disease (AD). Here, we report on a potent brain-permeable small molecule, DDL-920 that increases γ-oscillations and improves cognition/memory in a mouse model of AD, thus showing promise as a class of therapeutics for AD. We employed anatomical, in vitro and in vivo electrophysiological, and behavioral methods to examine the effects of our lead therapeutic candidate small molecule. As a novel in central nervous system pharmacotherapy, our lead molecule acts as a potent, efficacious, and selective negative allosteric modulator of the γ-aminobutyric acid type A receptors most likely assembled from α1β2δ subunits. These receptors, identified through anatomical and pharmacological means, underlie the tonic inhibition of parvalbumin (PV) expressing interneurons (PV+INs) critically involved in the generation of γ-oscillations. When orally administered twice daily for 2 wk, DDL-920 restored the cognitive/memory impairments of 3- to 4-mo-old AD model mice as measured by their performance in the Barnes maze. Our approach is unique as it is meant to enhance cognitive performance and working memory in a state-dependent manner by engaging and amplifying the brains endogenous γ-oscillations through enhancing the function of PV+INs.

Visual working memory (WM) is a central cognitive ability but is capacity-limited due to competition between remembered items. Understanding whether inter-item competition depends on the similarity of the features being remembered has important implications for determining if competition occurs in sensory or post-sensory stages of processing. Experiment 1 compared the precision of WM across homogeneous displays, where items belonged to the same feature type (e.g., colorful circles), and heterogeneous displays (e.g., colorful circles and oriented bars). Performance was better for heterogeneous displays, suggesting a feature-specific component of interference. However, Experiment 2 used a retro-cueing task to isolate encoding from online maintenance and revealed that inter-item competition during storage was not feature-specific. The data support recent models of WM in which inter-item interference - and hence capacity limits in WM - occurs in higher-order structures that receive convergent input from a diverse array of feature-specific representations.

While the majority of studies have concluded that sleep deprivation causes detrimental effects on various cognitive processes, some studies reported conflicting results. We examined the effects of a 108-h total sleep deprivation (TSD) on working memory in the northern fur seal, an animal with unusual sleep phenomenology and long-range annual migrations. The performance of fur seals was evaluated in a two-choice visual delayed matching to sample (DMTS) task, which is commonly used to evaluate working memory. In baseline conditions, the performance of fur seals in a DMTS task based on the percentage of errors was somewhat comparable with that in nonhuman primates at similar delays. We have determined that a 108-h TSD did not affect fur seals' performance in a visual DMTS task as measured by overall percentage of errors and response latencies. On the contrary, all fur seals improved task performance over the study, including the baseline, TSD and recovery conditions. In addition, TSD did not change the direction and strength of the pattern of behavioral lateralization in fur seals. We conclude that a 108-h TSD did not interfere with working memory in a DMTS test in northern fur seals.

Working memory, the process through which information is transiently maintained and manipulated over a brief period, is essential for most cognitive functions1-4. However, the mechanisms underlying the generation and evolution of working-memory neuronal representations at the population level over long timescales remain unclear. Here, to identify these mechanisms, we trained head-fixed mice to perform an olfactory delayed-association task in which the mice made decisions depending on the sequential identity of two odours separated by a 5 s delay. Optogenetic inhibition of secondary motor neurons during the late-delay and choice epochs strongly impaired the task performance of the mice. Mesoscopic calcium imaging of large neuronal populations of the secondary motor cortex (M2), retrosplenial cortex (RSA) and primary motor cortex (M1) showed that many late-delay-epoch-selective neurons emerged in M2 as the mice learned the task. Working-memory late-delay decoding accuracy substantially improved in the M2, but not in the M1 or RSA, as the mice became experts. During the early expert phase, working-memory representations during the late-delay epoch drifted across days, while the stimulus and choice representations stabilized. In contrast to single-plane layer 2/3 (L2/3) imaging, simultaneous volumetric calcium imaging of up to 73,307 M2 neurons, which included superficial L5 neurons, also revealed stabilization of late-delay working-memory representations with continued practice. Thus, delay- and choice-related activities that are essential for working-memory performance drift during learning and stabilize only after several days of expert performance.

COVID-19 forced students to rely on online learning using multimedia tools, and multimedia learning continues to impact education beyond the pandemic. In this study, we combined behavioral, eye-tracking, and neuroimaging paradigms to identify multimedia learning processes and outcomes. College students viewed four video lectures including slides with either an onscreen human instructor, an animated instructor, or no onscreen instructor. Brain activity was recorded via fMRI, visual attention was recorded via eye-tracking, and learning outcome was assessed via post-tests. Onscreen presence of instructor, compared with no instructor presence, resulted in superior post-test performance, less visual attention on the slide, more synchronized eye movements during learning, and higher neural synchronization in cortical networks associated with socio-emotional processing and working memory. Individual variation in cognitive and socio-emotional abilities and intersubject neural synchronization revealed different levels of cognitive and socio-emotional processing in different learning conditions. The instructor-present condition evoked increased synchronization, likely reflecting extra processing demands in attentional control, working memory engagement, and socio-emotional processing. Although human instructors and animated instructors led to comparable learning outcomes, the effects were due to the dynamic interplay of information processing vs. attentional distraction. These findings reflect a benefit-cost trade-off where multimedia learning outcome is enhanced only when the cognitive benefits motivated by the social presence of onscreen instructor outweigh the cognitive costs brought about by concurrent attentional distraction unrelated to learning.

Parietal alpha activity shows a specific pattern of phasic changes during working memory. It decreases during the encoding and recall phases but increases during the maintenance phase. This study tested whether online rTMS delivered to the parietal cortex during the maintenance phase of a working memory task would increase alpha activity and hence improve working memory. Then, 46 healthy volunteers were randomly assigned to two groups to receive 3-day parietal 10 Hz online rTMS (either real or sham, 3600 pulses in total) that were time-locked to the maintenance phase of a spatial span task (180 trials in total). Behavioral performance on another spatial span task and EEG signals during a change detection task were recorded on the day before the first rTMS (pretest) and the day after the last rTMS (posttest). We found that rTMS improved performance on both online and offline spatial span tasks. For the offline change detection task, rTMS enhanced alpha activity within the maintenance phase and improved interference control of working memory at both behavioral (K score) and neural (contralateral delay activity) levels. These results suggested that rTMS with alpha frequency time-locked to the maintenance phase is a promising way to boost working memory.

The hippocampus plays an essential role in long-term episodic memory by supporting the recollection of contextual details, whereas surrounding regions such as the perirhinal cortex support familiarity-based recognition discriminations. Working memory - the ability to maintain information over very brief periods of time - is traditionally thought to rely heavily on frontoparietal attention networks, but recent work has shown that it can also rely on the hippocampus. However, the conditions in which the hippocampus becomes involved in working memory tasks are unclear and whether it contributes to recollection or familiarity-based responses in working memory is only beginning to be explored. In the current paper, we first review and contrast the existing amnesia literature examining recollection and familiarity in episodic and working memory. The results indicate that recollection and familiarity contribute to both episodic and working memory. However, in contrast to episodic memory, in working memory the hippocampus is particularly critical for familiarity-based rather than recollection-based discrimination. Moreover, the results indicate that the role of the hippocampus in working memory can be obscured due to criterion-induced process-masking because it primarily supports intermediate-confidence recognition decisions. We then report results from a new working memory study examining the ability of amnesics to detect global and local changes in novel complex objects (i.e., fribbles), which indicates that the hippocampus plays an especially critical role in working memory when the task requires the detection of global rather than discrete changes. We conclude by considering the results in light of neurocomputational models and proposing a general framework for understanding the relationship between episodic and working memory.

Introduction: ACE I/D variant and short-term endurance workout performance, glycemic, anthropometrical, and biochemical responses are unknown. Thus, this study investigated how ACE I/D gene polymorphism affects physiological performance after four weeks of endurance training. Materials and methods: Seventeen ACE I/D genotype-screened volunteers trained three times a week for four weeks at 45 minutes per session, equivalent to their 60% VO2max in the first two weeks, 70% in the third, and 75% in the fourth and final week. The oral glucose tolerance test examined anthropometric, cardiovascular, lipid, and blood glucose before and after four weeks of endurance training. The dominant model groups (II/ID) (n = 10) and recessive model groups (DD/ID) (n = 14) were compared using the paired samples t-test. Results: The training significantly reduced total cholesterol in the recessive model group (-0.4 ± 0.6 mmol/l, p = 0.035). Both dominant and recessive groups showed increased maximal oxygen consumption (Dominant: 0.2 ± 0.3 l/min, p = 0.036; Recessive: 0.2 ± 0.2 l/min, p = 0.007) and peak power output (Dominant: 23.2 ± 18.6 l/min, p = 0.003; Recessive: 24.0 ± 14.6 l/min, p = 0.000). After training, the dominant model group had lower blood glucose than the recessive model group, however the difference was minor. When asked to remember a color, do people remember a point estimate (e.g., a particular shade of red), a point estimate plus an uncertainty estimate, or are memory representations rich probabilistic distributions over feature space? We asked participants to report the color of a circle held in working memory. Rather than collecting a single report per trial, we had participants place multiple bets to create trialwise uncertainty distributions. Bet dispersion correlated with performance, indicating that internal uncertainty guided bet placement. While the first bet was on average the most precisely placed, the later bets systematically shifted the distribution closer to the target, resulting in asymmetrical distributions about the first bet. This resulted in memory performance improvements when averaging across bets, and overall suggests that memory representations contain more information than can be conveyed by a single response. The later bets contained target information even when the first response would generally be classified as a guess or report of an incorrect item, suggesting that such failures are not all-or-none. This paradigm provides multiple pieces of evidence that memory representations are rich and probabilistic. Crucially, standard discrete response paradigms underestimate the amount of information in memory representations.

Spatial reconstruction, a method for evaluating how individuals remember the placement of objects, has traditionally been evaluated through the aggregate estimation of placement errors. However, this approach may obscure the nature of task errors. Specifically, recent data has suggested the importance of examining the precision of responses, as well as absolute performance on item-context bindings. In contrast to traditional analysis approaches based on the distance between the target and the reconstructed item, in this study we further explored three types of errors (swap error, global error, and local distance) that may all contribute to the distance, with particular emphasis on swap errors and local distance due to their associations with item-context bindings and memory precision, respectively. We examined these errors in children aged 3-18 years, making comparisons between children with typical development (TD) and children with Down syndrome (DS), a population with known memory challenges. As expected, older children outperformed younger children in terms of overall memory accuracy. Of importance is that we measured uneven maturational trajectories of memory abilities across the various error types. Specifically, both remembered locations (irrespective of object identity) and swap errors (object-location binding errors) align with the overall memory accuracy. Memory precision, as measured by local distance in simpler set size 2 trials, mirrored overall memory accuracy. However, for more complex set size 3 trials, local distance remained stable before age 8 and showed age-related change thereafter. The group with DS showed reduced precision compared to a TD matched group, and measures of precision, and to a lesser extent binding errors, correlated with standard neuropsychological outcomes. Overall, our study contributed to a fine-grained understanding of developing spatial memory ability in a large sample of typical developing children and a memory impaired population. These findings contribute to a growing body of research examining precision as a key factor in memory performance.

Aged rhesus monkeys, like aged humans, show declines in cognitive function. We present cognitive test data from a large sample of male and female rhesus monkeys, 34 young (aged 3.5-13.6 years) and 71 aged (aged 19.9-32.5 years at the start of cognitive testing). Monkeys were tested on spatiotemporal working memory (delayed response), visual recognition memory (delayed nonmatching to sample), and stimulus-reward association learning (object discrimination), tasks with an extensive evidence base in nonhuman primate neuropsychology. On average, aged monkeys performed worse than young on all 3 tasks. Acquisition of delayed response and delayed nonmatching to sample was more variable in aged monkeys than in young. Performance scores on delayed nonmatching to sample and object discrimination were associated with each other, but neither was associated with performance on delayed response. Sex and chronological age were not reliable predictors of individual differences in cognitive outcome among the aged monkeys. These data establish population norms for multiple cognitive tests in young and aged rhesus monkeys in the largest sample reported to date. They also illustrate independence of cognitive aging in task domains dependent on the prefrontal cortex and medial temporal lobe.

Ensemble perception is a process by which we summarize complex scenes. Despite the importance of ensemble perception to everyday cognition, there are few computational models that provide a formal account of this process. Here we develop and test a model in which ensemble representations reflect the global sum of activation signals across all individual items. We leverage this set of minimal assumptions to formally connect a model of memory for individual items to ensembles. We compare our ensemble model against a set of alternative models in five experiments. Our approach uses performance on a visual memory task for individual items to generate zero-free-parameter predictions of interindividual and intraindividual differences in performance on an ensemble continuous-report task. Our top-down modelling approach formally unifies models of memory for individual items and ensembles and opens a venue for building and comparing models of distinct memory processes and representations.

Objectives: Estimating the risk of diabetes mellitus (DM) is important for the proper management of patients with spinal cord injury (SCI). We investigated the short-term and long-term risks of DM among patients with SCI, according to the presence or severity of post-SCI disability and the level of injury. Methods: We conducted a retrospective cohort study using the Korean National Health Insurance Service (2010–2018) database. After matching by age and sex, 6129 SCI patients and 22,979 controls were included. The primary outcome was incident DM, and risk was evaluated for both the short term (within 1 year after SCI) and the long term (after 1 year of SCI diagnosis). Results: The risk of DM was higher among patients with cervical- and thoracic-level SCI accompanied by disability compared to the controls during follow-up (4.6 ± 2.6 years). The short-term risk of DM was higher among patients with SCI (odds ratio [OR] 2.51, 95% confidence interval [CI] 1.91–3.27) than among the controls and it was even higher among patients with severe disability (OR 5.38, 95% CI 2.91–9.27). According to the level of injury, patients with cervical SCI had the highest short-term risk of DM (with disability, OR 4.93, 95% CI 3.07–7.63). There was no significant increase of DM risk in the long term, after 1 year of SCI diagnosis. Conclusions: Patients with SCI accompanied by severe disability and cervical-level injury had higher risks of pronounced DM in the short term. The findings of this study emphasize the need for active surveillance of DM among patients with high-level SCI and disability, especially in the short term, in addition to continuous monitoring and proper management of DM in the long term. Simple Summary: Superovulation is a hormonal treatment used to produce multiple oocytes in cows simultaneously, which is essential for embryo transfer. Its success depends on various factors such as hormone levels, protocols, timing of insemination, weather, and nutrition. However, the impact of feed changes, specifically the energy levels in the concentrate, on embryo production is not well understood. This study explores how changes in concentrate intake, with unlimited access to hay, affect embryo production in indigenous Korean (Hanwoo) cows. We compared embryo production and physiological changes between cows with altered concentrate intake and those with consistent intake. The results offer new insights for managing superovulation in relation to feed concentrate. Superovulation is a technique used to increase the number of oocytes released for fertilization. This study investigated the effects of short-term differences in concentrate feed intake on in vivo embryo production through superovulation in indigenous Korean (Hanwoo) cows. The cows were given fresh water and hay ad libitum and randomly divided into three groups (control (CON, n = 9): 2.0 kg/day (unchanged diet); low concentrate (LC, n = 10): 0 kg/day; and high concentrate (HC, n = 8): 4.0 kg/day) according to the amount of formula they were fed. This feeding treatment began seven days before the start of the hormonal treatment for superovulation. From the results, the LC group had the greatest weight change and the lowest body condition score at harvest, followed by the CON and HC groups (p < 0.05). The LC group had the highest number of harvesting embryos, followed by the HC and CON groups (p < 0.05). Estradiol, progesterone, glucose, total cholesterol, high-density lipoprotein, low-density lipoprotein, total protein, and blood urea nitrogen concentrations did not differ between the groups, except for a temporary increase in the HC group on day 0. These findings suggest that more embryos may be harvested when short-term changes in concentrate intake are made during superovulatory responses in Hanwoo cows. [ABSTRACT FROM AUTHOR]

The COVID-19 pandemic has caused fluctuations in electricity demand, altering people's lifestyles and electricity usage patterns, thereby affecting the accuracy of demand predictions. However, existing studies on electricity forecasting have not adequately considered the incorporation of COVID-19-related features and the analysis of electricity usage characteristics across different regions of the UK. Therefore, this paper, based on data of the UK's national electricity demand, conducts an analysis around the scenario of a large-scale health emergency in society. We explore the changing patterns and regional characteristics of electricity consumption during the COVID-19 pandemic, comparing the forecast results before and during the pandemic to illustrate its impact on the UK's electricity demand. By introducing COVID-19-related features into the models, we compare the forecast results before and after their inclusion. The results indicate that the COVID-19 pandemic has had a certain impact on the electricity prediction in the UK, leading to a 22.8% decrease in prediction accuracy. However, the models' correlation improved with the inclusion of COVID-19-related features, resulting in a 13.2% enhancement in prediction accuracy compared to the previous models. Additionally, the study summarizes other factors influencing electricity demand, such as power imports/exports and clean energy usage, as considerations for electricity distribution planning. This contributes to improving the accuracy of predicting the UK's electricity demand during COVID-19 pandemic, enabling the government to adjust power dispatching plans reasonably based on relevant factors, achieving rational distribution and efficient scheduling of power resources. Cenchrus fungigraminus is a new species and is largely used as forage and mushroom substrate. However, it can usually not be planted on farmland on account of local agricultural land policy. Interplanting Cenchrus fungigraminus with other crops annually (short-term) is an innovative strategy to promote the sustainable development of the grass industry in southern China. To further investigate this, C. fungigraminus mono-planting (MC), C. fungigraminus–potato interplanting (CIP) and C. fungigraminus–broad bean interplanting (CIB) were performed. Compared to MC, soil microbial biomass carbon (SMBC), soil organic matter (SOM), ammoniacal nitrogen (AMN), pH and soil amino sugars had a positive effect on the rhizosphere soil of CIP and CIB, as well as enhancing soil nitrogenase, nitrite reductase, and peroxidase activities (p < 0.05). Moreover, CIP improved the root vitality (2.08 times) and crude protein (1.11 times). In addition, CIB enhanced the crude fiber of C. fungigraminus seedlings. These two interplanting models also improved the microbial composition and diversity (Actinobacteria, Firmicutes, and Bacteroidota, etc.) in the rhizosphere soil of C. fungigraminus seedlings. Among all the samples, 189 and 59 genes were involved in methane cycling and nitrogen cycling, respectively, which improved the presence of the serine cycle, ribulose monophosphate, assimilatory nitrate reduction, methane absorption, and glutamate synthesis and inhibited denitrification. Through correlation analysis and the Mantel test, the putative functional genes, encoding functions in both nitrogen and methane cycling, were shown to have a significant positive effect on pH, moisture, AMN, SOM, SMBC, and soil peroxidase activity, while not displaying a significant effect on soil nitrogenase activity and total amino sugar (p < 0.05). The short-term influence of the interplanting model was shown to improve land use efficiency and economic profitability per unit land area, and the models could provide sustainable agricultural production for rural revitalization. Background: Disorders of consciousness (DoC) represent a spectrum of neurological conditions that pose significant treatment challenges. Percutaneous short-term spinal cord stimulation (SCS) has emerged as a promising experimental diagnostic treatment to assess and potentially improve consciousness levels. However, the effectiveness of this intervention is frequently compromised by the shift of electrodes, particularly in the cervical region, which can negatively affect therapeutic outcomes. Methods: This retrospective study aimed to study if electrodes shift in percutaneous short-term SCS in patients with DoC would affect the outcome. We analyzed the relationship between electrode shift length and patient outcome, as well as the correlation with various anatomical parameters, including the actual length of the cervical spine, linear length, spinal canal transverse diameter, spinal canal diameter, and C2 cone height, in a cohort of patients undergoing the procedure. Results: Our findings revealed that in patients with better outcome, there are significant less patient with electrode shift (p = 0.019). Further, a linear correlation was found between the length of electrode shift and patients' outcome (Rho = 0.583, p = 0.002), with longer shift lengths associated with poorer outcomes. Contrary to our expectations, there was no significant association between the measured anatomical parameters and the extent of electrode shift. However, a trend was found between the actual length of the cervical spine and the shift of the electrode (p = 0.098). Notably, the shorter spinal canal transverse diameter was found to be significantly associated with better outcome in patients with DoC receiving percutaneous short-term SCS (p = 0.033). Conclusion: These results highlight the clinical importance of electrode stability in the cervical region during SCS treatment for patients with DoC. Ensuring secure placement of electrodes may play a crucial role in enhancing patients' outcome and minimize postoperative complications. Given the lack of association with expected anatomical parameters, future research should investigate other factors that could impact electrode stability to optimize this therapeutic intervention. Primary gaseous air pollutants have been associated with death from multiple causes, however, it remains unknown if they play a role in premature mortality among individuals living with HIV/AIDS. Data on HIV/AIDS patients were collected from the Hubei Provincial Center for Disease Control and Prevention, with a total of 1,467 AIDS-related deaths (ARD) between 2013 and 2020. Daily mean sulfur dioxide (SO2), nitrogen dioxide (NO2), and carbon monoxide (CO) were generated by artificial intelligence algorithms combined with big data. We employed a time-stratified case-crossover approach and conditional logistical regression models to investigate the acute effects of primary gaseous air pollutants on ARD. Per interquartile range increase in the concentrations of SO2 was significantly linked with ARD, with a corresponding odds ratio (OR) of 1.17 [95% confidence intervals (CIs): 1.01, 1.35] at lag 4 day. Furthermore, our findings indicated that males exhibited a heightened vulnerability to the adverse effects of SO2 and NO2, for example, the ORs were 1.24 (95% CIs: 1.05, 1.47) and 1.16 (95% CIs: 1.01, 1.34), respectively. Moreover, individuals aged over 65 years were more susceptible to SO2 and CO. Additionally, we identified the warm season as a sensitive period for mortality associated with SO2 and NO2. Background and Objectives: Investment has become a paramount concern for various individuals, particularly investors, in today's financial landscape. Cryptocurrencies, encompassing various types, hold a unique position among investors, with Bitcoin being the most prominent. Additionally, Bitcoin serves as the foundation for some other cryptocurrencies. Given the critical nature of investment decisions, diverse methods have been employed, ranging from traditional statistical approaches to machine learning and deep learning techniques. However, among these methods, the Generative Adversarial Network (GAN) model has not been utilized in the cryptocurrency market. This article aims to explore the applicability of the GAN model for predicting short-term Bitcoin prices. Methods: In this article, we employ the GAN model to predict short-term Bitcoin prices. Moreover, Data for this study has been collected from a diverse set of sources, including technical data, fundamental data, technical indicators, as well as additional data such as the number of tweets and Google Trends. In this research, we also evaluate the model's accuracy using the RMSE, MAE and MAPE metrics. Results: The results obtained from the experiments indicate that the GAN model can be effectively utilized in the cryptocurrency market for short-term price prediction. Conclusion: In conclusion, the results of this study suggest that the GAN model exhibits promise in predicting short-term prices in the cryptocurrency market, affirming its potential utility within this domain. These insights can provide investors and analysts with enhanced knowledge for making more informed investment decisions, while also paving the way for comparative analyses against alternative models operating in this dynamic field. OBJECTIVE: To identify the risk of acute respiratory distress syndrome (ARDS) and in-hospital mortality using long short-term memory (LSTM) framework in a mechanically ventilated (MV) non-COVID-19 cohort and a COVID-19 cohort. METHODS: We included MV ICU patients between 2017 and 2018 and reviewed patient records for ARDS and death. Using active learning, we enriched this cohort with MV patients from 2016 to 2019 (MV non-COVID-19, n=3905). We collected a second validation cohort of hospitalised patients with COVID-19 in 2020 (COVID+, n=5672). We trained an LSTM model using 132 structured features on the MV non-COVID-19 training cohort and validated on the MV non-COVID-19 validation and COVID-19 cohorts. RESULTS: Applying LSTM (model score 0.9) on the MV non-COVID-19 validation cohort had a sensitivity of 86% and specificity of 57%. The model identified the risk of ARDS 10 hours before ARDS and 9.4 days before death. The sensitivity (70%) and specificity (84%) of the model on the COVID-19 cohort are lower than MV non-COVID-19 cohort. For the COVID-19 + cohort and MV COVID-19 + patients, the model identified the risk of in-hospital mortality 2.4 days and 1.54 days before death, respectively. DISCUSSION: Our LSTM algorithm accurately and timely identified the risk of ARDS or death in MV non-COVID-19 and COVID+ patients. By alerting the risk of ARDS or death, we can improve the implementation of evidence-based ARDS management and facilitate goals-of-care discussions in high-risk patients. CONCLUSION: Using the LSTM algorithm in hospitalised patients identifies the risk of ARDS or death.

Oscillatory activity is commonly observed during the maintenance of information in short-term memory, but its role remains unclear. Non-oscillatory models of short-term memory storage are able to encode stimulus identity through their spatial patterns of activity, but are typically limited to either an all-or-none representation of stimulus amplitude or exhibit a biologically implausible exact-tuning condition. Here we demonstrate a simple mechanism by which oscillatory input enables a circuit to generate persistent or sequential activity that encodes information not only in the spatial pattern of activity, but also in the amplitude of activity. This is accomplished through a phase-locking phenomenon that permits many different amplitudes of persistent activity to be stored without requiring exact tuning of model parameters. Altogether, this work proposes a class of models for the storage of information in working memory, a potential role for brain oscillations, and a dynamical mechanism for maintaining multi-stable neural representations.

BackgroundWorking memory involves the temporary storage and manipulation of information and is frequently an area of challenge for individuals with Down syndrome (DS). Despite the potential benefits of intervention, laboratory assessments of working memory that could capture intervention effects have not undergone rigorous evaluation for use with young children with DS. It is critical to evaluate assessments of working memory in young children with DS to ensure the reliable and accurate measurement of performance.AimThis study evaluated an adapted laboratory measure of working memory for young children with DS 2-8 years old.MethodA self-ordered pointing task, the Garage Game, was administered to 78 children with DS (mean = 5.17 years; SD = 1.49). Adaptations were made to the task to minimize potential DS phenotype-related language and motor confounds.ResultsResults indicate that the measure is feasible, scalable, and developmentally sensitive, with minimal floor and practice effects for this population within this chronological and developmental age range.ConclusionThese findings demonstrate that the Garage Game is promising for use in studies of early working memory and treatment trials that aim to support the development of this critical dimension of executive functioning for children with DS.

Adverse experiences in early life can shape neuronal structures and synaptic function in multiple brain regions, leading to deficits of distinct cognitive functions later in life. Focusing on the pyramidal cells of the prelimbic cortex (PrL), a main subregion of the medial prefrontal cortex, the impact of early-life adversity (ELA) was investigated in a well-established animal model generated by changing the rearing environment during postnatal days 2 to 9 (P2-P9), a sensitive developmental period. ELA has enduring detrimental impacts on the dendritic spines of PrL pyramidal cells, which is most apparent in a spatially circumscribed region. Specifically, ELA affects both thin and mushroom-type spines, and ELA-provoked loss of spines is observed on selective dendritic segments of PrL pyramidal cells in layers II-III and V-VI. Reduced postsynaptic puncta represented by postsynaptic density protein-95 (PSD-95), but not synaptophysin-labelled presynaptic puncta, in ELA mice supports the selective loss of spines in the PrL. Correlation analysis indicates that loss of spines and postsynaptic puncta in the PrL contributes to the poor spatial working memory of ELA mice, and thin spines may play a major role in working memory performance. To further understand whether loss of spines affects glutamatergic transmission, AMPA- and NMDA-receptor-mediated synaptic currents (EPSCs) were recorded in a group of Thy1-expressing PrL pyramidal cells. ELA mice exhibited a depressed glutamatergic transmission, which is accompanied with a decreased expression of GluR1 and NR1 subunits in the PrL. Finally, upregulating the activation of Thy1-expressing PrL pyramidal cells via excitatory DREADDs can efficiently improve the working memory performance of ELA mice in a T-maze-based task, indicating the potential of a chemogenetic approach in restoring ELA-provoked memory deficits.

Multiple types of memory guide attention: Both long-term memory (LTM) and working memory (WM) effectively guide visual search. Furthermore, both types of memories can capture attention automatically, even when detrimental to performance. It is less clear, however, how LTM and WM cooperate or compete to guide attention in the same task. In a series of behavioral experiments, we show that LTM and WM reliably cooperate to guide attention: Visual search is faster when both memories cue attention to the same spatial location (relative to when only one memory can guide attention). LTM and WM competed to guide attention in more limited circumstances: Competition only occurred when these memories were in different dimensions - particularly when participants searched for a shape and held an accessory color in mind. Finally, we found no evidence for asymmetry in either cooperation or competition: There was no evidence that WM helped (or hindered) LTM-guided search more than the other way around. This lack of asymmetry was found despite differences in LTM-guided and WM-guided search overall, and differences in how two LTMs and two WMs compete or cooperate with each other to guide attention. This work suggests that, even if only one memory is currently task-relevant, WM and LTM can cooperate to guide attention; they can also compete when distracting features are salient enough. This work elucidates interactions between WM and LTM during attentional guidance, adding to the literature on costs and benefits to attention from multiple active memories.

Working memory (WM) and timing are generally considered distinct cognitive functions, but similar neural signatures have been implicated in both. To explore the hypothesis that WM and timing may rely on shared neural mechanisms, we used psychophysical tasks that contained either task-irrelevant timing or WM components. In both cases, the task-irrelevant component influenced performance. We then developed recurrent neural network (RNN) simulations that revealed that cue-specific neural sequences, which multiplexed WM and time, emerged as the dominant regime that captured the behavioural findings. During training, RNN dynamics transitioned from low-dimensional ramps to high-dimensional neural sequences, and depending on task requirements, steady-state or ramping activity was also observed. Analysis of RNN structure revealed that neural sequences relied primarily on inhibitory connections, and could survive the deletion of all excitatory-to-excitatory connections. Our results indicate that in some instances WM is encoded in time-varying neural activity because of the importance of predicting when WM will be used.

Decades of evidence across taxa have established the importance of dopamine (DA) signaling in the pFC for successful working memory performance. Genetic and hormonal factors can shape individual differences in prefrontal DA tone. The catechol-o-methyltransferase (COMT) gene regulates basal prefrontal DA, and the sex hormone 17β-estradiol potentiates DA release. E. Jacobs and M. DEsposito [Estrogen shapes dopamine-dependent cognitive processes: Implications for womens health. Journal of Neuroscience, 31, 5286-5293, 2011] investigated the moderating role of estradiol on cognition using the COMT gene and COMT enzymatic activity as a proxy for pFC DA tone. They found that increases in 17β-estradiol within women at two time points during the menstrual cycle influenced working memory performance in a COMT-dependent manner. Here, we aimed to replicate and extend the behavioral findings of Jacobs and DEsposito by employing an intensive repeated-measures design across a full menstrual cycle. Our results replicated the original investigation. Within-person increases in estradiol were associated with improved performance on 2-back lure trials for participants with low basal levels of DA (Val/Val carriers). The association was in the opposite direction for participants with higher basal levels of DA (Met/Met carriers). Our findings support the role of estrogen in DA-related cognitive functions and further highlight the need to consider gonadal hormones in cognitive science research.

Working memory (WM) is the ability to maintain and manipulate information no longer accessible in the environment. The brain maintains WM representations over delay periods in noisy population-level activation patterns, resulting in variability in WM representations across items and trials. It is established that participants can introspect aspects of the quality of WM representations, and that they can accurately compare which of several WM representations of stimulus features like orientation or color is better on each trial. However, whether this ability to evaluate and compare the quality of multiple WM representations extends to spatial WM tasks remains unknown. Here, we employed a memory-guided saccade task to test recall errors for remembered spatial locations when participants were allowed to choose the most precise representation to report. Participants remembered either one or two spatial locations over a delay and reported one items location with a saccade. On trials with two spatial locations, participants reported either the spatial location of a randomly cued item, or the location of the stimulus they remembered best. We found a significant improvement in recall error and increase in response time (RT) when participants reported their best-remembered item compared with trials in which they were randomly cued. These results demonstrate that participants can accurately introspect the relative quality of neural WM representations for spatial position, consistent with previous observations for other stimulus features, and support a model of WM coding involving noisy representations across items and trials.

There is consistent debate over whether capacity in working memory (WM) is subject to an item limit, or whether an unlimited number of items can be held in this online memory system. The item limit hypothesis clearly predicts guessing responses when capacity is exceeded, and proponents of this view have highlighted evidence for guessing in visual working memory tasks. Nevertheless, various models that deny item limits can explain the same empirical patterns by asserting extremely low fidelity representations that cannot be distinguished from guesses. To address this ambiguity, we employed a task for which guess responses elicited a qualitatively distinct pattern from low fidelity memories. Inspired by work from Rouder et al. (2014), we employed an orientation WM task that required subjects to recall the precise orientation of each of six memoranda presented 1 s earlier. The orientation stimuli were created by rotating the position of a "clock hand" inside a circular region that was demarcated by four colored quadrants. Critically, when observers guess with these stimuli, the distribution of responses is biased towards the center of these quadrants, creating a "banded" pattern that cannot be explained by a low precision memory. We confirmed the presence of this guessing pattern using formal model comparisons, and we show that the prevalence of this pattern matches observers' own reports of when they thought they were guessing. Thus, these findings provide further evidence for guessing behaviors predicted by item limit models of WM capacity.

When viewing scenes, observers differ in how long they linger at each fixation location and how far they move their eyes between fixations. What factors drive these differences in eye-movement behaviors? Previous work suggests individual differences in working memory capacity may influence fixation durations and saccade amplitudes. In the present study, participants (N = 98) performed two scene-viewing tasks, aesthetic judgment and memorization, while viewing 100 photographs of real-world scenes. Working memory capacity, working memory processing ability, and fluid intelligence were assessed with an operation span task, a memory updating task, and Ravens Advanced Progressive Matrices, respectively. Across participants, we found significant effects of task on both fixation durations and saccade amplitudes. At the level of each individual participant, we also found a significant relationship between memory updating task performance and participants fixation duration distributions. However, we found no effect of fluid intelligence and no effect of working memory capacity on fixation duration or saccade amplitude distributions, inconsistent with previous findings. These results suggest that the ability to flexibly maintain and update working memory is strongly related to fixation duration behavior.

Distraction disrupts Working Memory (WM) performance, but how the brain filters distraction is not known. One possibility is that neural activity associated with distractions is suppressed relative to a baseline/passive task (biased competition). Alternatively, distraction may be denied access to WM, with no suppression. Furthermore, behavioural work indicates separate mechanisms for ignoring distractions which occur (1) while we put information into WM (Encoding Distraction, ED) and (2) while we maintain already encoded information during the WM delay period (Delay Distraction, DD). Here we used fMRI in humans to measure category-sensitive cortical activity and probe the extent to which ED/DD mechanisms involve enhancement/suppression during a WM task. We observed significant enhancement of task-relevant activity, relative to a passive view task, which did not differ according to whether or when distractors appeared. For both ED and DD we found no evidence of suppression, but instead a robust increase in stimulus specific activity in response to additional stimuli presented during the passive view task, which was not seen for the WM task, when those additional stimuli were to be ignored. The results indicate that ED/DD resistance does not necessarily involve suppression of distractor-related activity. Rather, a rise in distractor-associated activity is prevented when distractors are presented, supporting models of input gating, and providing a potential mechanism by which input-gating might be achieved.

[C. Koch, S. Ullman, Hum. Neurobiol.4, 219-227 (1985)] proposed a 2D topographical salience map that took feature-map outputs as its input and represented the importance saliency of the feature inputs at each location as a real number. The computation on the map, winner-take-all, was used to predict action priority. We propose that the same or a similar map is used to compute centroid judgments, the center of a cloud of diverse items. [P. Sun, V. Chu, G. Sperling, Atten. Percept. Psychophys.83, 934-955 (2021)] demonstrated that following a 250-msec exposure of a 24-dot array of 3 intermixed colors, subjects could accurately report the centroid of each dot color, thereby indicating that these subjects had at least three salience maps. Here, we use a postcue, partial-report paradigm to determine how many more salience maps subjects might have. In 11 experiments, subjects viewed 0.3-s flashes of 28 to 32 item arrays composed of M, M = 3,...,8, different features followed by a cue to mouse-click the centroid of items of just the post-cued feature. Ideal detector response analyses show that subjects utilized at least 12 to 17 stimulus items. By determining whether a subjects performance in (M-1)-feature experiments could/could-not predict performance in M-feature experiments, we conclude that one subject has at least 7 and the other two have at least five salience maps. A computational model shows that the primary performance-limiting factors are channel capacity for representing so many concurrently presented groups of items and working-memory capacity for so many computed centroids.

The Internet-of-things ecosystem has been a driving force in the creation of smart communities where a variety of physical phenomena can be monitored continuously, e.g., air quality, traffic conditions on roads, energy consumption in buildings, etc. In this paper, we address how IoT can be quickly and effectively deployed for short-term and sporadic events (e.g., fire spread in a wildland area and flood propagation), where monitoring the evolving event is critical. In particular, we propose QuIC-IoT, a model-driven planning platform that aims to temporarily deploy a custom IoT infrastructure for monitoring short-term events, where phenomena-spread is driven by models that are physics-based. Our driving usecase event is a quasi-planned prescribed fire or RxFire - this is a wildfire resilience technique where intentional small fires are ignited apriori by forestry personnel to destroy fuel and help contain the spread of actual wildfires. Anomalies that may occur during these quasi-planned events must be rapidly captured by the IoT deployment, e.g., escaped RxFires can escalate to catastrophic wildfires under unpredictable conditions of wind, vegetation, etc. QuIC-IoT incorporates domain expert-developed models to guide IoT deployment; the event area is partitioned into subregions and a criticality metric that quantifies the likelihood of anomalies at each location is computed. QuIC-IoT allows us to mix fixed and quasi-mobile IoT devices to flexibly deploy IoT in challenging terrain and as the phenomena (RxBurn) evolves. We evaluate QuIC-IoT in two real-world forest settings (large and small) in Blodgett Forest, CA, USA, with concrete burn plans developed by wildfire experts. Our experimental results reveal that QuIC-IoT enables over 3X improvement in cost-effectiveness and performance (timely detection of anomalies) as compared to baseline IoT deployment algorithms.

Flexible behavior requires gating mechanisms that encode only task-relevant information in working memory. Extant literature supports a theoretical division of labor whereby lateral frontoparietal interactions underlie information maintenance and the striatum enacts the gate. Here, we reveal neocortical gating mechanisms in intracranial EEG patients by identifying rapid, within-trial changes in regional and inter-regional activities that predict subsequent behavioral outputs. Results first demonstrate information accumulation mechanisms that extend prior fMRI (i.e., regional high-frequency activity) and EEG evidence (inter-regional theta synchrony) of distributed neocortical networks in working memory. Second, results demonstrate that rapid changes in theta synchrony, reflected in changing patterns of default mode network connectivity, support filtering. Graph theoretical analyses further linked filtering in task-relevant information and filtering out irrelevant information to dorsal and ventral attention networks, respectively. Results establish a rapid neocortical theta network mechanism for flexible information encoding, a role previously attributed to the striatum.

Human learning and decision-making are supported by multiple systems operating in parallel. Recent studies isolating the contributions of reinforcement learning (RL) and working memory (WM) have revealed a trade-off between the two. An interactive WM/RL computational model predicts that although high WM load slows behavioral acquisition, it also induces larger prediction errors in the RL system that enhance robustness and retention of learned behaviors. Here, we tested this account by parametrically manipulating WM load during RL in conjunction with EEG in both male and female participants and administered two surprise memory tests. We further leveraged single-trial decoding of EEG signatures of RL and WM to determine whether their interaction predicted robust retention. Consistent with the model, behavioral learning was slower for associations acquired under higher load but showed parametrically improved future retention. This paradoxical result was mirrored by EEG indices of RL, which were strengthened under higher WM loads and predictive of more robust future behavioral retention of learned stimulus-response contingencies. We further tested whether stress alters the ability to shift between the two systems strategically to maximize immediate learning versus retention of information and found that induced stress had only a limited effect on this trade-off. The present results offer a deeper understanding of the cooperative interaction between WM and RL and show that relying on WM can benefit the rapid acquisition of choice behavior during learning but impairs retention.SIGNIFICANCE STATEMENT Successful learning is achieved by the joint contribution of the dopaminergic RL system and WM. The cooperative WM/RL model was productive in improving our understanding of the interplay between the two systems during learning, demonstrating that reliance on RL computations is modulated by WM load. However, the role of WM/RL systems in the retention of learned stimulus-response associations remained unestablished. Our results show that increased neural signatures of learning, indicative of greater RL computation, under high WM load also predicted better stimulus-response retention. This result supports a trade-off between the two systems, where degraded WM increases RL processing, which improves retention. Notably, we show that this cooperative interplay remains largely unaffected by acute stress.

We argue that critical areas of memory research rely on problematic measurement practices and provide concrete suggestions to improve the situation. In particular, we highlight the prevalence of memory studies that use tasks (like the "old/new" task: "have you seen this item before? yes/no") where quantifying performance is deeply dependent on counterfactual reasoning that depends on the (unknowable) distribution of underlying memory signals. As a result of this difficulty, different literatures in memory research (e.g., visual working memory, eyewitness identification, picture memory, etc.) have settled on a variety of fundamentally different metrics to get performance measures from such tasks (e.g., A', corrected hit rate, percent correct, d', diagnosticity ratios, K values, etc.), even though these metrics make different, contradictory assumptions about the distribution of latent memory signals, and even though all of their assumptions are frequently incorrect. We suggest that in order for the psychology and neuroscience of memory to become a more cumulative, theory-driven science, more attention must be given to measurement issues. We make a concrete suggestion: The default memory task for those simply interested in performance should change from old/new ("did you see this item'?") to two-alternative forced-choice ("which of these two items did you see?"). In situations where old/new variants are preferred (e.g., eyewitness identification; theoretical investigations of the nature of memory signals), receiver operating characteristic (ROC) analysis should be performed rather than a binary old/new task.

The ability to use past experience to effectively guide decision-making declines in older adulthood. Such declines have been theorized to emerge from either impairments of striatal reinforcement learning systems (RL) or impairments of recurrent networks in prefrontal and parietal cortex that support working memory (WM). Distinguishing between these hypotheses has been challenging because either RL or WM could be used to facilitate successful decision-making in typical laboratory tasks. Here we investigated the neurocomputational correlates of age-related decision-making deficits using an RL-WM task to disentangle these mechanisms, a computational model to quantify them, and magnetic resonance spectroscopy to link them to their molecular bases. Our results reveal that task performance is worse in older age, in a manner best explained by working memory deficits, as might be expected if cortical recurrent networks were unable to sustain persistent activity across multiple trials. Consistent with this, we show that older adults had lower levels of prefrontal glutamate, the excitatory neurotransmitter thought to support persistent activity, compared to younger adults. Individuals with the lowest prefrontal glutamate levels displayed the greatest impairments in working memory after controlling for other anatomical and metabolic factors. Together, our results suggest that lower levels of prefrontal glutamate may contribute to failures of working memory systems and impaired decision-making in older adulthood.

Rainfall plays a crucial role in shaping the weather in Indonesia, influenced by factors such as latitude, elevation, wind patterns, land and water distribution, as well as topography. Rising temperatures contribute to the increased intensity of extreme rainfall, amplifying the potential risk of disasters. Therefore, it is necessary to conduct analyses to predict weather based on historical time series data. This study aims to identify short-term rainfall patterns and trends using the SARIMAX method. The initial stage involves data processing and splitting the data into training data (2005-2014) and test data (2015-2019). Time series decomposition is then performed to identify patterns, followed by period identification and stationarity testing using the ADF test. The SARIMAX model is selected based on the lowest AIC value, followed by parameter estimation and diagnostic tests. Rainfall predictions are evaluated using model performance evaluation methods, while inferential statistics are used to describe population attributes through confidence intervals. The forecasting results show that using the SARIMAX (0, 0, 1)(0, 1, 1, 12)12 model for short-term rainfall prediction achieved the best performance with the smallest MSE, MAE, and MAPE values on the test sample with a 95% confidence level. Repeating rainfall patterns with a mid-year decline and significant variation in magnitude were identified from the sample data for August 2020-2027, with the lowest rainfall at 39.05 mm and the highest at 397.03 mm in December. The implications of this study support efforts to mitigate natural disasters due to unexpected weather changes by integrating this model into early warning systems and water resource planning.

Poverty in Indonesia is a problem that needs special attention by the government. Poverty is due to the high level of inequality, unequal distribution of income, and the number of poor people, which continue to increase. Therefore, it can affect society’s economy. This research aims to identify incongruities between endogenous and exogenous variables that influence poverty levels in Indonesia. By applying a descriptive quantitative analysis approach for 2013-2022 in the form of secondary data from the Indonesian Central Statistics Agency, the research model uses dynamic panel data regression analysis based on the Generalized Method Of Moment method (GMM). The method was developed by Arellano-Bond, of which the two best models are First-Difference GMM and System GMM, which create an impartial, consistent, and efficient model for determining short-term ang long-term effects. The research results show that HDI has a significant negative impact on short-term and long-term relationships with poverty levels, resulting in low human resources. Exports have a significant negative impact on poverty levels in the short and long term. This means that if exports increase, poverty levels can be reduced. Imports have a significant positive impact on short-term and long-term with poverty levels. In import activities, the higher the price increases, the more people's purchasing power decreases. The results are declared to have a significant effect because the p-value is significance of level 5% or 0.05. It is expected that the research will become reference material for macroeconomic development and further research regarding poverty alleviation.

One critical approach for promoting the efficiency of memory is to adopt selective encoding strategies to prioritize more valuable information. Past neuroimaging studies have shown that value-directed modulation of verbal memory depends heavily on the engagement of left-lateralized semantic processing regions, particularly in the ventrolateral prefrontal cortex (VLPFC). In the present study, we used high-definition transcranial direct current stimulation (HD-tDCS) to seek evidence for a causal role of left VLPFC in supporting the memory advantage for high-value items. Three groups of healthy young adult participants were presented with lists of words to remember, with each word accompanied by an arbitrarily assigned point value. During the first session, all participants received sham stimulation as they encoded five lists of 30 words each. Two of these lists were immediately tested with free recall, with feedback given to allow participants to develop metacognitive insight and strategies to maximize their point total. The second session had the exact same structure as the first, but the groups differed in whether they received continued sham stimulation (N = 22) or anodal stimulation of the left VLPFC (N = 21) or right VLPFC (N = 20). Those lists not tested with immediate recall were tested with recognition judgments after a one-day delay. Since no brain stimulation was applied during this Day 2 test, any performance differences can be attributed to the effects of stimulation on Day 1 encoding processes. Anodal stimulation of left VLPFC significantly boosted participants' memory encoding selectivity. In comparison, no such effect was seen in participants who received right VLPFC or sham stimulation. Estimates of recollection- and familiarity-based responding revealed that left VLPFC stimulation specifically amplified the effects of item value on recollection. These results demonstrate a causal role for left VLPFC in the implementation of selective value-directed encoding strategies, putatively by boosting deep semantic processing of high-value words. Our findings also provide further evidence on the hemispheric lateralization of value-directed verbal memory encoding.

Background and objectiveIndividuals with intellectual disability (ID) experience protracted cognitive development compared with typical youth. Sensitive measurement of cognitive change in this population is a critical need for clinical trials and other intervention studies, but well-validated outcome measures are scarce. This study's aim was to evaluate the sensitivity of the NIH Toolbox Cognition Battery (NIHTB-CB) to detect developmental changes in groups with ID-fragile X syndrome (FXS), Down syndrome (DS), and other ID (OID)-and to provide further support for its use as an outcome measure for treatment trials.MethodsWe administered the NIHTB-CB and a reference standard cross-validation measure (Stanford-Binet Intelligence Scales, Fifth Edition [SB5]) to 256 individuals with FXS, DS, and OID (ages 6-27 years). After 2 years of development, we retested 197 individuals. Group developmental changes in each cognitive domain of the NIHTB-CB and SB5 were assessed using latent change score models, and 2-year growth was evaluated at 3 age points (10, 16, and 22 years).ResultsOverall, effect sizes of growth measured by the NIHTB-CB tests were comparable with or exceeded those of the SB5. The NIHTB-CB showed significant gains in almost all domains in OID at younger ages (10 years), with continued gains at 16 years and stability in early adulthood (22 years). The FXS group showed delayed gains in attention and inhibitory control compared with OID. The DS group had delayed gains in receptive vocabulary compared with OID. Unlike the other groups, DS had significant growth in early adulthood in 2 domains (working memory and attention/inhibitory control). Notably, each group's pattern of NIHTB-CB growth across development corresponded to their respective pattern of SB5 growth.DiscussionThe NIHTB-CB is sensitive to developmental changes in individuals with ID. Comparison with levels and timing of growth on the cross-validation measure shows that the NIHTB-CB has potential to identify meaningful trajectories across cognitive domains and ID etiologies. Sensitivity to change within the context of treatment studies and delineation of clinically meaningful changes in NIHTB-CB scores, linked to daily functioning, must be established in future research to evaluate the battery more completely as a key outcome measure.

The neuroanatomical changes that underpin cognitive development are of major interest in neuroscience. Of the many aspects of neuroanatomy to consider, tertiary sulci are particularly attractive as they emerge last in gestation, show a protracted development after birth, and are either human- or hominoid-specific. Thus, they are ideal targets for exploring morphological-cognitive relationships with cognitive skills that also show protracted development such as working memory (WM). Yet, the relationship between sulcal morphology and WM is unknown-either in development or more generally. To fill this gap, we adopted a data-driven approach with cross-validation to examine the relationship between sulcal depth in lateral prefrontal cortex (LPFC) and verbal WM in 60 children and adolescents between ages 6 and 18. These analyses identified 9 left, and no right, LPFC sulci (of which 7 were tertiary) whose depth predicted verbal WM performance above and beyond the effect of age. Most of these sulci are located within and around contours of previously proposed functional parcellations of LPFC. This sulcal depth model outperformed models with age or cortical thickness. Together, these findings build empirical support for a classic theory that tertiary sulci serve as landmarks in association cortices that contribute to late-maturing human cognitive abilities.

ABSTRACTPeople can support abstract reasoning by using mental models with spatial simulations. Such models are employed when people represent elements in terms of ordered dimensions (e.g. who is oldest, Tom, Dick, or Harry). We test and find that the process of forming and using such mental models can influence the liking of its elements (e.g. Tom, Dick, or Harry). The presumed internal structure of such models (linear-transitive array of elements), generates variations in processing ease (fluency) when using the model in working memory (see the Symbolic Distance Effect, SDE). Specifically, processing of pairs where elements have larger distances along the order should be easier compared to pairs with smaller distances. Elements from easier pairs should be liked more than elements from difficult pairs (fluency being hedonically positive). Experiment 1 shows that unfamiliar ideographs are liked more when at wider distances and therefore easier to process. Experiment 2 replicates this effect with non-words. Experiment 3 rules out a non-spatial explanation of the effect while Experiments 4 offers a high-powered replication. Experiment 5 shows that the spatial effect spontaneously emerges after learning, even without a task that explicitly focuses on fluency. Experiment 6 employed a shorter array, but yielded no significant results.

BackgroundStudies have investigated white matter microstructure in relation to late-life cognitive impairments, with fractional anisotropy (FA) and mean diffusivity (MD) measures thought to capture demyelination and axonal degradation. However, new post-processing methods allow isolation of free water (FW), which captures extracellular fluid contributions such as atrophy and neuroinflammation, from tissue components. FW also appears to be highly relevant to late-life cognitive impairment. Here, we evaluated whether executive functions are associated with FW, and FA and MD corrected for FW (FAFWcorr and MDFWcorr).MethodWe examined 489 non-demented men in the Vietnam Era Twin Study of Aging (VETSA) at mean age 68. Two latent factors capturing 'common executive function' and 'working-memory specific' processes were estimated based on 6 tasks. Analyses focused on 11 cortical white matter tracts across three metrics: FW, FAFWcorr, and MDFWcorr.ResultsBetter 'common executive function' was associated with lower FW across 9 of the 11 tracts. There were no significant associations with intracellular metrics after false discovery rate correction. Effects also appeared driven by individuals with MCI (13.7% of the sample). Working memory-specific tasks showed some associations with FAFWcorr, including the triangularis portion of the inferior frontal gyrus. There was no evidence that cognitive reserve (i.e., general cognitive ability assessed in early adulthood) moderated these associations between executive function and FW or FA.DiscussionExecutive function abilities in early old age are associated primarily with extracellular fluid (FW) as opposed to white matter (FAFWcorr or MDFWcorr). Moderation analyses suggested cognitive reserve does not play a strong role in these associations, at least in this sample of non-demented men.

ObjectiveThe combination of d-methylphenidate and guanfacine (an α-2A adrenergic agonist) may be an effective alternative to either agent as monotherapy in children with attention-deficit/hyperactivity disorder (ADHD). This study investigated the neural mechanisms underlying medication effects using cortical source analysis of electroencephalography (EEG) data.MethodA total of 172 children with ADHD (aged 7-14; 118 boys) completed an 8-week randomized, double-blind, comparative study with 3 treatment arms: d-methylphenidate, guanfacine, or their combination. EEG modulations of brain oscillations at baseline and end point were measured during a spatial working memory task from cortical sources localized within the anterior cingulate (midfrontal) and primary visual cortex (midoccipital), based on previously reported ADHD and control differences. Linear mixed models examined treatment effects on EEG and performance measures.ResultsCombined treatment decreased midoccipital EEG power across most frequency bands and task phases. Several midoccipital EEG measures also showed significantly greater changes with combined treatment than with monotherapies. D-methylphenidate significantly increased midoccipital theta during retrieval, while guanfacine produced only trend-level reductions in midoccipital alpha during maintenance and retrieval. Task accuracy improved with combined treatment, was unchanged with d-methylphenidate, and worsened with guanfacine. Treatment-related changes in midoccipital power correlated with improvement in ADHD severity.ConclusionThese findings show that combined treatment ameliorates midoccipital neural activity associated with treatment-related behavioral improvements and previously implicated in visuo-attentional deficits in ADHD. Both monotherapies had limited effects on EEG measures, with guanfacine further showing detrimental effects on performance. Background: Evidence suggests that the quality of the therapeutic relationship impacts the outcome of therapy, with the ending phase having the potential to elicit painful and difficult emotions. There is limited research on this phase of the therapeutic relationship. Aims: The study aimed to explore the lived experiences of counsellors when ending with clients in short‐term planned settings those relationships in which they have worked at relational depth. Method: Interpretative phenomenological analysis was used to evaluate the counsellors' lived experiences of ending therapeutic relationships. Online semi‐structured interviews took place with six trainee and qualified counsellors who volunteered as counsellors for a local charity. Findings: Three main themes were identified during the study—impact of ending, impact of short‐term work and professional issues. Findings indicated that working at relational depth in short‐term counselling could elicit unprocessed powerful emotions within the ending phase of therapy. Limitations: A small sample size limits the wider application of these findings. Implications: Further guidance and support are needed for trainee and qualified practitioners in the management and experience of ending counselling relationships within short‐term work, including the exploration of counsellors' self‐disclosure. Implications for practice include the need for supervisors and counsellors to be proactive in managing reflective practice within supervision, supervisors recognising and challenging supervisees' non‐disclosure and counsellors looking at further ways for self‐reflection on endings in counselling relationships. Conclusion: The ending phase for counsellors working at relational depth within a seven‐session model can elicit powerful emotions, positive experiences and challenges. [ABSTRACT FROM AUTHOR]

As reverse total shoulder arthroplasty (rTSA) has evolved, the use of augmented glenoid baseplates has increased. While there are benefits of metal augmentation, literature concerning the mid- to long-term follow-up of these implants is emerging. The purpose of this study is to report early to short-term radiographic follow-up of patients with glenoid deformity treated with a metal-augmented glenoid baseplate. Our hypothesis is that we will see a low incidence of osteolysis, loosening, complications, and revision of these implants regardless of diagnosis, augment type, or patient demographics. We performed a single-center retrospective review of all patients undergoing primary reverse shoulder arthroplasty with an augmented metallic baseplate for rotator cuff arthropathy and glenohumeral arthritis. Patients undergoing revision procedures or reverse arthroplasty for fractures were excluded. All patients had a minimum of 1-year radiographic follow-up. Preoperative imaging was reviewed to confirm the diagnosis and glenoid deformity classification. Postoperative radiographs were reviewed for evidence of complications, revisions, osteolysis, baseplate loosening, and failure. Clinical data were reviewed for complications. Primary rTSA with a metal-augmented baseplate (38 full wedge and 22 half wedge augments) was performed on 60 patients who met inclusion criteria (mean age 74 ± 7). The average radiographic follow-up of all patients was 17 months. In our cohort, there was one patient with aseptic loosening of a full wedge implant that was not revised. There were no other complications, revision surgeries, baseplate failures, or acromial stress fractures. Six patients developed scapular notching, and five patients were found to have asymptomatic glenoid osteolysis. Two patients with full wedge implants developed aseptic humeral loosening. Pearson chi-square post hoc analysis found no correlation between augment type, gender, or glenoid deformity classification and radiographic changes such as glenoid or humeral osteolysis, loosening, or scapular notching. We did find a statistically significant correlation (P =.018) between patients with body mass index > 35 and glenoid osteolysis. Primary rTSA for patients with rotator cuff arthropathy or glenohumeral arthritis with glenoid deformity corrected with metal-augmented glenoid baseplates results in a low complication rate and an acceptable outcome at a minimum 1-year follow-up. Compared to other similar cohorts, our patients did not experience any acromial stress fractures, and only one patient had loosening of the glenoid component that was not ultimately revised. Further studies with longer follow-up will be needed, but early outcomes are promising and support the increase in the use of these implants. [ABSTRACT FROM AUTHOR]

The aim of this study was to explore the feasibility and preliminary effectiveness of a short-term, manualized schema therapy group for 77 patients with CBTresistant mood and/or anxiety and/or personality disorders (PDs) in primary care. The primary focus was on the effects of this treatment on Early Maladaptive Schemas (EMS), schema modes, and psychological well-being. These aspects were assessed pre-and post-treatment treatment using the Young Schema Questionnaire (YSQ), the Schema Mode Inventory version 1.1 (SMI), and the Symptom Questionnaire-48 (SQ-48). The treatment consisted of 16 sessions, incorporating cognitive, behavioral, and experiential techniques. EMS significantly decreased from pre-treatment to post-treatment, as along with maladaptive schema modes. Adaptive modes increased, as did psychological wellbeing. There were no significant differences between the DSM-5 classifications regarding changes in the aforementioned measures, except for the maladaptive modes, where the value of the corrected within-subject effect indicated a significant interaction. Post hoc comparisons were therefore conducted which showed that patients with a mood disorder experienced more positive changes in maladaptive modes compared to patients with anxiety disorders and PDs (p < 0.001). There was no significant difference between those with PDs and those with Anxiety Disorders. Our findings provide preliminary evidence that short-term, manualized schema therapy might be an effective treatment for patients with CBT-resistant mood and/or anxiety and/or PDs in primary care.
