Abstract

Deciphering ion channel activity and signaling interactions within cells is one of the key tasks of neuroscience. Currently, measuring this electrophysiological activity is done using patch-clamp1-4 or voltage-sensitive imaging5-8. Unfortunately, these techniques are unable to balance between single-channel sensitivity and highthroughput detection. Here we introduce a label-free electrochemical-modulated interferometric scattering microscope (EM-iSCAT) to measure ion channel activity on living cells at both whole-cell and single-channel levels. We visualize the cellular responses dynamics to osmotic stimulation, and record open-close trajectories of single N-methyl-D-aspartate receptors channels with a frame rate of 1.5 kHz. Furthermore, we localize and distinguish different kinds of ion channels (Na+, K+, Ca2+) on cell membrane and monitor spatio-temporal heterogeneous responses between different cells in a network. The high-throughput and single-channel sensitive nature of EMiSCAT microscopy allows monitoring simultaneously the activity of individual channels, their localization, and clustering in the cellular community. Our imaging concept opens the possibility to study any kind of ion channels, and more broadly, cell communication mediated by ion channels.

2. Decrypting material performance by wide-field femtosecond interferometric imaging of energy carrier evolution

Abstract

Energy carrier evolution is crucial for material performance. Ultrafast microscopy has been widely applied to visualize the spatiotemporal evolution of energy carriers. However, direct imaging of small amounts of energy carriers on nanoscale remains difficult due to extremely weak transient signals. Here we present a method for ultrasensitive and high-throughput imaging of energy carrier evolution in space and time. This method combines femtosecond pump-probe techniques with interferometric scattering microscopy (iSCAT), named Femto-iSCAT. The interferometric principle and unique spatially-modulated contrast enhancement increase the transient image contrast by >2 orders of magnitude and enable the exploration of new science. We address three important and challenging problems: transport of different energy carriers at various interfaces, heterogeneous hot electron distribution and relaxation in single plasmonic resonators, and distinct structure-dependent edge state dynamics of carriers and excitons in optoelectronic semiconductors. Femto-iSCAT holds great potential as a universal tool for ultrasensitive imaging of energy carrier evolution in space and time.

26. Hyperbranched Chain Hybridization for Artificial Aggregates: Prolonging Intracellular Retention for Image‐Guided Cellular Fate Modulation.

Abstract

Designing dynamic assemblies in living cells is crucial for creating organelle‐like structures, yet precisely controlling their morphological transitions in response to specific signals is a significant challenge. In this study, a DNA framework is combined with hybridization chain reaction (HCR) to achieve specific assembly of hyperbranched aggregates in cancer cells. HCR, distinguished for its signal amplification and linear extension capabilities, enables the morphological transition of precursors to be specifically triggered by trace amounts of endogenous microRNA‐21 (miR‐21). The spatial constraints of the framework and the diversity of hairpin orientations significantly accelerate the assembly kinetics of hyperbranched networks, and the resulting micrometer‐scale aggregates possess enhanced intracellular retention capabilities. Introducing Ce6 molecules as a proof of concept, the regulatory function of aggregates can be activated under light irradiation and remains effective over a long period. The probe we constructed demonstrates good stability and biocompatibility, offers easy functionalization, and works inside cells long‐term, making it an ideal candidate material for the construction of organelle‐like structures. 37. Smart Molecular Imaging and Theranostic Probes by Enzymatic Molecular In Situ Self-Assembly

Abstract

Enzymatic molecular in situ self-assembly (E-MISA) that enables the synthesis of high-order nanostructures from synthetic small molecules inside a living subject has emerged as a promising strategy for molecular imaging and theranostics. This strategy leverages the catalytic activity of an enzyme to trigger probe substrate conversion and assembly in situ, permitting prolonging retention and congregating many molecules of probes in the targeted cells or tissues. Enhanced imaging signals or therapeutic functions can be achieved by responding to a specific enzyme. This E-MISA strategy has been successfully applied for the development of enzyme-activated smart molecular imaging or theranostic probes for in vivo applications. In this Perspective, we discuss the general principle of controlling in situ self-assembly of synthetic small molecules by an enzyme and then discuss the applications for the construction of “smart” imaging and theranostic probes against cancers and bacteria. Finally, we discuss the current challenges and perspectives in utilizing the E-MISA strategy for disease diagnoses and therapies, particularly for clinical translation.
