Abstract Background Previous studies have established a positive link between nurse managers’ caring leadership and nurses’ work engagement, but the processes and conditions through which this leadership style influences positive work behaviors remain largely unexplored. To address this gap and contribute to the existing body of knowledge, we developed a chain-mediated effects model to elucidate the impact of caring leadership on nurses’ work engagement and the underlying mechanisms. In this model, we identified professional mission and affective organizational commitment as the mediating variables, offering a novel perspective on the relationship between caring leadership and work engagement. Methods A robust multi-center and large-sample cross-sectional survey was conducted, involving 2502 first-line nurses from six general tertiary hospitals across the eastern, central, and western regions of China. The data collection instruments included a comprehensive questionnaire covering demographic information, the caring leadership scale, the Chinese calling scale, the affective organizational commitment scale, and the Utrecht work engagement scale. Data were meticulously screened and analyzed, employing descriptive analysis to summarize the demographic information, correlation analysis to test the relationship among the variables, stepwise regression analysis to explore the mediating role of calling and affective organization commitment, and the bootstrap method to test the chain mediating effect. This rigorous methodology not only ensures the reliability and validity of research findings but also instills confidence in the robustness of this research. Abstract This retrospective study aimed to determine the incidence and risk factors for osteomyelitis recurrence and present our experiences in treating traumatic osteomyelitis in the lower leg and foot. We retrospectively reviewed data from 174 patients with distally based sural flaps for treating traumatic osteomyelitis with soft tissue defects in the lower leg and foot from November 2003 to February 2021. Possible risk factors for osteomyelitis recurrence were compared between the osteomyelitis control and recurrence groups. A total of 162 (93.1%) flaps survived uneventfully, while 12 (6.9%) flaps developed partial necrosis. Five patients had a bone defect with an average length of 5 cm. The free vascularized bone grafts were performed in two patients, and bone transportations were performed in three patients. All patients were followed up with an average period of 72.8 months. There were 152 patients (87.4%) in control group and 22 patients (12.6%) in recurrence group. Objective The clinical management of patients with chronic limb‐threatening ischemia (CLTI) faces great challenges. Enhancing wound healing and limb preservation rates in this cohort is a critical objective. This study investigates the effectiveness of combining tibial cortex transverse transport (TTT) and endovascular therapy (EVT) for the treatment of patients with severe CLTI. We aim to evaluate the therapeutic results of this combined approach on the specified patient group. Methods We conducted a retrospective study to compare EVT with the combination of TTT and EVT in patients (Rutherford category 5 and above) with CLTI at Guangxi Medical University's First Affiliated Hospital from June 2017 to June 2023. This cohort was subjected to a follow‐up period ranging from a minimum of 6 months to a maximum of 12 months. The primary outcome measures included amputation‐free survival (AFS) (avoidance of above‐ankle amputation or death from any cause), overall mortality, limb salvage rates, wound healing efficiency, and the technical efficacy of the applied treatments. A variety of statistical analyses including chi‐square tests, Fisher's exact tests, and Pearson's and Spearman's correlation analyses. Results In this study, 131 patients with CLTI were included: 76 in the control group receiving only EVT treatment and 55 in the TTT + EVT group. The two groups were matched on demographic and clinical characteristics. In the TTT + EVT group, after more than 6 months of follow‐up, 85.5% of patients achieved AFS, and wound healing was observed in 54.5% (30 of 55 patients). After more than 12 months of follow‐up, 81.9% achieved AFS, with wound healing in 32 patients. Furthermore, after more than 24 months, 74.2% of patients remained amputation‐free, with wound healing in all surviving patients. In the control group, after more than 6 months of follow‐up, 72.4% of patients achieved AFS, and wound healing was observed in 51.3% (39 of 96 patients). After more than 12 months, 48.9% achieved AFS, with wound healing in 21 patients. Conclusion We found that combining therapy of TTT and EVT is safe and can be successfully administered in patients with CLTI and it enhances wound healing and AFS.

Abstract Studying crystal anisotropy is of great importance for understanding the thermal–mechanical coupling behavior of crystalline rocks in deep underground engineering. In this study, a microscopic parameter calibration method incorporating the size effect is proposed. Subsequently, a thermal–mechanical coupling model accounting for the quartz crystal anisotropy is established to investigate the thermal–mechanical coupling behavior of monomineral quartzite. The results show that thermal-induced microcracks are exclusively distributed along crystal boundaries, and initiate preferentially from crystal boundaries with a larger average linear thermal expansion coefficient, eventually leading to the formation of a crack network. With the increase in temperature, the peak strength of monomineral quartzite increases slightly at first and then decreases rapidly, and the transition threshold temperature is 200 °C. Both elastic modulus and Poisson’s ratio show a monotonic pattern, with abrupt changes occurring at 200 and 300 °C, respectively. The monomineral quartzite exhibits a significant compaction stage under uniaxial compression, and the ductile strengthening critical temperature for monomineral quartzite are between 400 and 500 °C. The quartz crystal anisotropy leads to an anisotropic distribution of inclination angles for tensile microcracks under high temperatures while having no obvious effect on the shear microcracks. In addition, the average size of fragments generated under uniaxial compression is influenced by thermal cracking, demonstrating an initial decrease followed by an increase, and the distribution of fragment sizes is solely correlated with the temperature, which is more concentrated with the increase in temperature.

The types，preparation methods，thermal and mechanical properties of rare earth hafnate materials and their corrosion behaviors exposed to low melting point silicate（CMAS） and high temperature water vapor are summarized. The previous investigations indicate that the rare earth hafnates have the characteristics of low thermal conductivity，excellent high-temperature phase stability and good resistance to CMAS corrosion，which shows a favorable application prospect in the field of thermal/environmental barrier coatings（T/EBCs）. However，in order to overcome the limitations of a single-phase rare earth hafnate exposed to water vapor corrosion and CMAS，it is still necessary to carry out systematic studies on multi-rare earth components/high-entropy rare earth hafnate in the future，and further clarify the mechanism of influence of lattice distortion caused by components on physical and chemical properties of materials. Moreover，the coupled control method of thermal，mechanical and chemical properties of hafnate with integrated functions such as thermal protection，water vapor corrosion resistance and CMAS resistance and their corresponding material preparation processes should be explored.

Rapid tissue reconstruction in acute and chronic injuries are challengeable, the inefficient repair mainly due to the difficulty in simultaneous promoting the regeneration of peripheral nerves and vascular, which are closely related. Main clinical medication strategy of tissue repair depends on different cytokines to achieve nerves, blood vessels or granulation tissue regeneration, respectively. However, their effect is still limited to single aspect with biorisk exists upon long-time use. Herein, for the first time, we have demonstrated that NA isolated from Malania oleifera has potential to simultaneously promote both neurogenesis and angiogenesis in vitro and in vivo. First, NA was identified by NMR and FTIR structural characterization analysis. In a model of oxidative stress in neural cells induced by hydrogen peroxide, the cells viability of RSC96 and PC12 were protected from oxidative stress injury by NA. Similarly, based on the rat wound healing model, effective blood vessel formation and wound healing can be observed in tissue staining under NA treatment. In addition, according to the identification of nerve and vascular related markers in the wound tissue, the mechanism of NA promoting nerve regeneration lies in the upregulation of the secretion NGF, NF-200 and S100 protein, and NA treatment was also able to up-regulate VEGF and CD31 to directly promote angiogenesis during wound healing. This study provides an important candidate drug molecules for acute or chronic wound healing and nerve vascular synchronous regeneration.

Background Mature rapeseed pods typically shatter when harvested, resulting in approximately 8–12% yield loss. Adverse weather conditions and mechanized harvesting can diminish pod yield by up to 50%, primarily owing to delays in harvesting and mechanical collisions. The pod shatter resistance index (PSRI) assesses pod damage. Recent research focused on comparing pod shatter resistance among varieties, evaluating methods, and studying gene knockout mechanisms. However, there remains a pressing need to broaden the scope of research. In particular, it is essential to recognize that pod shatter, a complex trait, influenced by genetics, environment, agronomic practices, and harvest techniques. Future studies should integrate these factors to develop comprehensive strategies to mitigate pod shatter, enhancing rapeseed yields and agricultural mechanization. This review explores factors affecting pod shatter resistance and strategies to improve it. Methodology Scoping literature review that adhered to the methodological framework for systematic reviews was performed using search engines such as Google Scholar, Web of Science, and the Chinese National Knowledge Infrastructure. This review aimed to identify pertinent articles, which were subsequently subjected to thorough screening and evaluation. The protocol for this literature review involved the following key steps: definition of research questions, development of a search strategy, development of data extraction strategy, synthesis of the extracted data, and organization and analysis of the extracted data. Results The review presents strategies for enhancing rapeseed yield during mechanized harvesting, focusing on four key areas: (i) selecting and breeding shatter-resistant varieties using DNA markers to establish a robust germplasm resource; (ii) optimizing cultivation technologies and agronomic measures to elicit favorable interactions between compact plant-type genotypes and the environment, thereby facilitating nutrient-related regulatory mechanisms of rapeseed pods to improve pod dry weight and resistance; (iii) innovating combine header design and structure to better suit rapeseed harvesting; and (iv) providing training for operators to enhance their harvesting skills. These comprehensive measures aim to minimize yield loss, increase production efficiency. Conclusion To effectively reduce yield loss during mechanized harvesting of rapeseed, it is crucial to enhance resistance to pod shattering by addressing both internal physiological factors and external environmental conditions. This requires a holistic approach that includes genetic improvements, optimization of ecological conditions, careful cultivation management, and precise harvesting timing, along with ongoing research into traits related to machine harvesting to boost production efficiency and sustainability.

Abstract Dendritic cell (DC)-targeted vaccination is a new mode of antigen delivery that relies on the use of monoclonal antibodies (mAb) to target antigen to specific DC subsets. The neonatal Fc receptor (FcRn) is a non-classical Fc receptor that binds to immunoglobulin G (IgG) in acidified endosomes and controls its intracellular transport and recycling. FcRn is known to participate in the antigen presentation of immune complexes, however its contribution to DC-targeted vaccination has not previously been examined. Here we have investigated the role of FcRn in antigen presentation using antigen conjugated to IgG mAb which target specific DC receptors, including DEC205 and Clec9A expressed by the conventional DC 1 (cDC1) subset. We show that FcRn is expressed at high levels by cDC1, both at steady-state and following activation and plays a significant role in MHC I cross-presentation and MHC II presentation of antigens that are targeted to cDC1 via mAb specific for DEC205. This effect of FcRn is intrinsic to cDC1 and FcRn impacts the efficacy of anti-DEC205-mediated vaccination against B cell lymphoma. In contrast, FcRn does not impact presentation of antigens targeted to Clec9A and does not regulate presentation of cell-associated antigen. These data highlight a new and unique role of FcRn in controlling the immunogenicity of anti-DEC205-based vaccination, with consequences for exploiting this pathway to improve DC-targeted vaccine outcomes.

Lactylation is a novel post-translational modification (PTM) involving proteins that is induced by lactate accumulation. Histone lysine lactylation alters chromatin spatial configuration, influencing gene transcription and regulating the expression of associated genes. This modification plays a crucial role as an epigenetic regulatory factor in the progression of various diseases. Glycolytic reprogramming is one of the most extensively studied forms of metabolic reprogramming, recognized as a key hallmark of cancer cells. It is characterized by an increase in glycolysis and the inhibition of the tricarboxylic acid (TCA) cycle, accompanied by significant lactate production and accumulation. The two processes are closely linked by lactate, which interacts in various physiological and pathological processes. On the one hand, lactylation levels generally correlate positively with the extent of glycolytic reprogramming, being directly influenced by the lactate concentration produced during glycolytic reprogramming. On the other hand, lactylation can also regulate glycolytic pathways by affecting the transcription and structural functions of essential glycolytic enzymes. This review comprehensively outlines the mechanisms of lactylation and glycolytic reprogramming and their interactions in tumor progression, immunity, and inflammation, with the aim of elucidating the relationship between glycolytic reprogramming and lactylation.

Cement production consumes large amounts of depleted resources and energy, and causes environmental pollution. In order to reduce cement consumption and realize the closed-loop recycling of solid waste, cold bonded solid waste wrap-shell lightweight aggregates (SWSLAs) with a sextuplet blended system composed of dredged sediments, mineral powder, steel slag, phosphogypsum, fly ash, and a minimal amount of cement were prepared. Through the single-factor experiment, the influences of the types and dosages of solid wastes in the nuclear phase of SWSLAs (SWSLAs-NP) and shell phase of SWSLAs (SWSLAs-SP) on the strength and water resistance were explored, and the optimum ratio was obtained. On this basis, the influence of the types and amounts of activators incorporated in SWSLAs on the physical properties of aggregates was studied, and the primary microstructure of the aggregate was characterized. Our results showed that the optimal ratio of solid waste in the SWSLAs-NP was: 40 wt% of dredged sediments, 24 wt% of mineral powder, 18 wt% of steel slag, and 18 wt% of phosphogypsum. The optimal weight ratio of cement and solid waste in the SWSLAs-SP was 5:95. The resulting aggregate had a bearing capacity of single grain of 73.83 N and a water-resistance coefficient of 0.67. After the above aggregates were treated with Na2SO4 (activator) at 2%, the bearing capacity and water-resistance coefficient of the aggregates increased by 47.00% and 11.94%, respectively. The performances were even better than that of the aggregate composed of100w cement in the shell phase. Microstructural observations demonstrated that the structure of the raw material was reconstructed under the action of Na2SO4, and the raw materials reacted with each other, and thus the densification of the aggregate and its core-shell interface structure was enhanced, which improved the comprehensive performance of the aggregate. This study represents a step toward efficient utilization of industrial solid wastes.

Objective(s)This study aimed to identify the key elements and develop a formation mechanism model of quality geriatric care behavior for nursing assistants.MethodsThis qualitative research employed the strategy of grounded theory proposed by Strauss and Corbin. Furthermore, the data was collected by participatory observation and semi-structured interviews. A total of 12 nursing managers, 63 nursing assistants, and 36 older people from 9 nursing homes in 6 cities were interviewed, whereas for the observatory survey, participants were recruited from 2 nursing homes.ResultsThe comparative and analysis process revealed 5 key elements of quality geriatric care behavior, including holistic care, personalized care, respect, positive interaction, and empowerment. Based on the Capability-Opportunity-Motivation-Behavior (COM-B) model, key elements and the 3 stages of quality geriatric care behavior (negative behavior cognition stage, practice exchange run-in stage, and positive behavior reinforcement stage), the theoretical framework of the formation mechanism was established.ConclusionThe results indicated that nursing assistants’ capabilities, motivation, and organizational and environmental support are vital for quality care behaviors. The theoretical framework established in this study provides theoretical support and practical reference to policymakers, institutional administrators, and healthcare professionals for improving nursing assistant’s care behaviors.

Abstract Post-translational modifications (PTMs) of the non-histone protein high-mobility group protein B1 (HMGB1) are involved in modulating inflammation and immune responses. Recent studies have implicated that the RNA-binding protein (RBP) Musashi-2 (MSI2) regulates multiple critical biological metabolic and immunoregulatory functions. However, the precise role of MSI2 in regulating PTMs and tumor immunity in colorectal cancer (CRC) remains unclear. Here, we present data indicating that MSI2 potentiates CRC immunopathology in colitis-associated colon cancer (CAC) mouse models, cell lines and clinical specimens, specifically via HMGB1-mediated dendritic cell (DC) maturation and migration, further contributes to the infiltration of CD4+ and CD8+ T cells and inflammatory responses. Under stress conditions, MSI2 can exacerbate the production, nucleocytoplasmic transport and extracellular release of damage-associated molecular patterns (DAMPs)-HMGB1 in CRC cells. Mechanistically, MSI2 mainly enhances the disulfide HMGB1 production and protein translation via direct binding to nucleotides 1403–1409 in the HMGB1 3′ UTR, and interacts with the cytoplasmic acetyltransferase P300 to upregulate its expression, further promoting the acetylation of K29 residue in HMGB1, thus leading to K29-HMGB1 nucleocytoplasmic translocation and extracellular release. Furthermore, blocking HMGB1 activity with glycyrrhizic acid (Gly) attenuates MSI2-mediated immunopathology and immune infiltration in CRC in vitro and in vivo. Collectively, this study suggests that MSI2 may improve the prognosis of CRC patients by reprogramming the tumor immune microenvironment (TIME) through HMGB1-mediated PTMs, which might be a novel therapeutic option for CRC immunotherapy.

Abstract Background Musashi-2 (MSI2) is a critical RNA-binding protein (RBP) whose ectopic expression drives the pathogenesis of various cancers. Accumulating evidence suggests that inducing ferroptosis of tumor cells can inhibit their malignant biological behavior as a promising therapeutic approach. However, it is unclear whether MSI2 regulates cell death in colorectal cancer (CRC), especially the underlying mechanisms and biological effects in CRC ferroptosis remain elusive. Methods Experimental methods including qRT‒PCR, immunofluorescence, flow cytometry, western blot, co-immunoprecipitation, CCK-8, colony formation assay, in vitro cell transwell migration and invasion assays, in vivo xenograft tumor experiments, liver and lung CRC metastasis models, CAC mice models, transmission electron microscopy, immunohistochemistry, histopathology, 4D label-free proteomics sequencing, bioinformatic and database analysis were used in this study. Results Here, we investigated that MSI2 was upregulated in CRC and positively correlated with ferroptosis inhibitor molecules. MSI2 deficiency suppressed CRC malignancy by inhibiting cell proliferation, viability, migration and invasion in vitro and in vivo; and MSI2 deficiency triggered CRC ferroptosis by changing the intracellular redox state (ROS levels and lipid peroxidation), erastin induced cell mortality and viability, iron homeostasis (intracellular total irons and ferrous irons), reduced glutathione (GSH) levels and mitochondrial injury. Mechanistically, through 4D-lable free proteomics analysis on SW620 stable cell lines, we demonstrated that MSI2 directly interacted with p-ERK and MSI2 knockdown downregulated the p-ERK/p38/MAPK axis signaling pathway, which further repressed MAPKAPK2 and HPSB1 phosphorylation, leading to decreased expression of PCNA and Ki67 and increased expression of ACSL4 in cancer cells. Furthermore, HSPB1 could rescue the phenotypes of MSI2 deficiency on CRC ferroptosis in vitro and in vivo. Conclusions This study indicates that MSI2 deficiency suppresses the growth and survival of CRC cells and promotes ferroptosis by inactivating the MAPK signaling pathway to inhibit HSPB1 phosphorylation, which leads to downregulation of PCNA and Ki67 and upregulation of ACSL4 in cancer cells and subsequently induces redox imbalance, iron accumulation and mitochondrial shrinkage, ultimately triggering ferroptosis. Therefore, targeted inhibition of MSI2/MAPK/HSPB1 axis to promote ferroptosis might be a potential treatment strategy for CRC.

In order to achieve metallurgical bonding in the form of solid solution at the Cu/W interface and avoid the formation of intermetallic compounds, the novel high entropy alloys (HEAs) were designed on the basis of the mature high entropy alloy criteria. The CuCrCoFeNixTi high entropy alloys interlayers were applied to weld the CuW and CuCr bimetals by sintering–infiltration technology. Scanning electron microscope, energy dispersive spectrometry, and X-ray diffraction were used to explore the interfacial microstructure evolutions and strengthening mechanism of CuW/CuCr joints with applied HEA interlayers. The interfacial characterization results show that HEAs were diffused and dissolved into bimetallic materials, and a diffusion solution layer of 2–3 μm thickness was formed at the Cu/W phase interface, and there is no new phase generated at the CuW/CuCr interface. When CuCrCoFeNi1.5Ti interlayer was infiltrated into the CuW/CuCr interface, the electrical conductivity of CuCr side is 71.6%IACS, and the interfacial tensile strength reaches 484.5 MPa. Compared with the CuW/CuCr integral material without interlayer, the interfacial bonding strength is increased by 43.1%. And the SEM fracture morphology presents a larger amount of cleavage fractures of W particles. It indicates the appropriate solid solution layer on edge of W skeletons formed at the Cu/W phases interface. The Cu/W phase interface is strengthened, and it can effectively transfer and disperse the external load. Tungsten phase with higher elastic modulus endures a large amount of load, resulting in enhancing the CuW/CuCr interfacial bonding strength. When CuCrCoFeNi2Ti high entropy alloy interlayer was applied, the W skeleton near the CuW/CuCr interface was eroded, the imperfect W skeleton cannot withstand the tensile load effectively, resulting in decrease in the CuW/CuCr interfacial bonding strength. In the interfacial fracture, appears some fragmentations of W particles, and fewer W particles occur at the cleavage fracture.

Abstract The inherent unpredictability of the maritime environment leads to low rates of survival during accidents. Life jackets serve as a crucial safety measure in underwater environments. Nonetheless, most conventional life jackets lack the capability to monitor the wearer's underwater body movements, impeding their effectiveness in rescue operations. Here, we present an intelligent self‐powered life jacket system (SPLJ) composed of a wireless body area sensing network, a set of deep learning analytics, and a human condition detection platform. Six coaxial core‐shell structure triboelectric fiber sensors with high sensitivity, stretchability, and flexibility are integrated into this system. Additionally, a portable integrated circuit module is incorporated into the SPLJ to facilitate real‐time monitoring of the wearer's movement. Moreover, by leveraging the deep‐learning‐assisted data analytics and establishing a robust correlation between the wearer's movements and condition, we have developed a comprehensive system for monitoring drowning individuals, achieving an outstanding recognition accuracy of 100%. This groundbreaking work introduces a fresh approach to underwater intelligent survival devices, offering promising prospects for advancing underwater smart wearable devices in rescue operations and the development of ocean industry.

Elucidating the temporal process of immune remodeling under immunosuppressive treatment after liver transplantation (LT) is critical for precise clinical management strategies. Here, we performed a single-cell multi-omics analysis of peripheral blood mononuclear cells (PBMCs) collected from LT patients (with and without acute cellular rejection [ACR]) at 13 time points. Validation was performed in two independent cohorts with additional LT patients and healthy controls. Our study revealed a four-phase recovery process after LT and delineated changes in immune cell composition, expression programs, and interactions along this process. The intensity of the immune response differs between the ACR and non-ACR patients. Notably, the newly identified inflamed NK cells, CD14+RNASE2+ monocytes, and FOS-expressing monocytes emerged as predictive indicators of ACR. This study illuminates the longitudinal evolution of the immune cell landscape under tacrolimus-based immunosuppressive treatment during LT recovery, providing a four-phase framework that aids the clinical management of LT patients.

[Purpose/Significance] The construction of a reading promotion standard system is the key to achieving the standardization and professional development of reading promotion. The construction of standardized reading promotion is based on reading theory, and aims to make reading promotion a standardized service with scientific theoretical guidance, constrained by the values and ethics of libraries, and with clear service purposes and target groups. From the perspective of high-quality development, the construction process of the standardized reading promotion system also reflects the development level of the "people-centered" library of the new era, which is of great significance. [Method/Process] Aiming to conduct a systematic and comprehensive investigation of the existing reading promotion standards in China and to promote the research and practice of reading promotion standards and evaluation in China, this report comprehensively collects the relevant standards in the library field issued in China before 2024, and screens out the standards related to reading promotion. By counting the drafting units, responsible agencies and types, and analyzing the content of the standards, this report reviews the construction of the reading promotion standard system in China. [Results/Conclusions] Standardized reading promotion work in China began to take shape during the "13th Five-Year Plan" period, and is now gradually gaining momentum and entering a new stage of rapid and high-quality development. The major achievements include: first, a standardized reading promotion system has been established for the first time, with an increasing number of relevant standards and the gradual introduction of specific local standards; second, public libraries have played a leading role in the standardized construction of reading promotion, demonstrating continuous professional expertise in the field; third, local standards have shown great effectiveness, fully absorbing and transforming many innovative experiences in reading promotion work; and fourth, many important and practical issues in reading promotion are reflected and addressed in the standard texts, with an increasing emphasis on demand and effectiveness orientation. Accordingly, this report puts forward a number of recommendations for the future construction of the reading promotion standard system, including: strengthening the launch of national and industry standard construction projects for reading promotion, continuously developing specialized standards for reading promotion, strengthening the integrated application of standards related to reading promotion, and promoting the participation of various disciplines in standard construction.

Abstract Background With China's rapidly aging population, meeting the diverse care needs of senior citizens is becoming more challenging. Although voluntary social services have numerous advantages and are popular among older adults, there is little information on the need for assistance from volunteer-based social services, particularly those with a medical background, and influencing factors among urban home-based older adults. This study aimed to assess the need for assistance from voluntary services and related factors among urban home-based older adults in China. Methods A cross-sectional study was conducted in 2022 on communities in four cities in China. The 27-item Home-Based Older Adults Assistance Need Scale was used to measure the assistance needs of 498 participants aged 60 and above. Multiple linear regression models were conducted to identify salient variables associated with the need for assistance from voluntary services. Results The mean score of the need for assistance from voluntary services was 88.60 ± 24.37. The mean scores of the items examining four dimensions, namely, health maintenance, visiting communication, social intercourse, and daily life, were 3.64 ± 1.08, 3.49 ± 1.04, 3.33 ± 1.08, and 2.78 ± 1.08, respectively. The level of depression, willingness to assist older adults, attaching importance to health preservation, ability to self-comfort, desire to accept assistance from others, and the presence of more children or none at all were all positively correlated with the perceived need for assistance from voluntary services. In contrast, social care obtained from visiting medical institutions was negatively correlated. These seven factors explained 28.5% of the total variance. Conclusions Urban home-based older adults in China were found to have significant requirements for assistance from volunteer services, and several complex factors were associated with more significant assistance needs. These findings may encourage the extremely limited numbers of social volunteers, particularly those with a medical background, to identify priorities in providing assistance services to the large numbers of urban home-based older adults and thus improve service delivery.

Background: Tibial Cortex Transverse Transport (TTT) has been demonstrated to be an effective treatment for unilateral diabetic foot ulcers (UDFUs). However, this retrospective study was designed to compare the efficacy and safety of unilateral TTT on bilateral diabetic foot ulcers (BDFUs). Methods: This retrospective study included a review of patients with TTT treated from January 2017 to August 2019, Propensity Score Matching (PSM) was performed to compare patients with BDFUs to those with UDFUs. Ulcer healing, recurrence, and major amputation rates were evaluated at 1-year follow-up. Changes in foot vessels were assessed in the BDFUs group using computed tomography angiography (CTA). Results: A total of 140 patients with DFUs (106 UDFUs and 34 BDFUs) were included in the study. UDFUs and BDFUs were matched in a 1:1 ratio (34 in each group) using PSM. No significant difference was observed at 1-year-follow-up [91.2% (31/34) vs. 76.5% (26/34), OR 0.315 (95% CI 0.08 to 1.31), P ​= ​0.10] and 6-month-follow-up [70.6% (24/34) vs. 50.0% (17/34), OR 0.85 (95% CI 0.15 to 1.13), P ​= ​0.08] in two groups. Significant differences in rates of major amputation and recurrence between the groups (P ​> ​0.05) were not observed. The BDFUs group appeared more angiogenesis of the foot by CTA after 8 weeks of operation. Conclusion: Results of this study suggest that severe BDFUs can be effectively treated by unilateral TTT. TTT is easy to operate and effective, which may be a good alternative for treating severe BDFUs. The translational potential of this article: In previous retrospective clinical studies, TTT has demonstrated promising clinical outcomes in the management of diabetic foot ulcers. In this current study, we aim to investigate the potential use of TTT in treating distant tissue defects by evaluating the limited availability and safety of TTT for the management of bilateral diabetic foot. While additional basic and clinical research is necessary to fully elucidate the underlying mechanisms, our study offers insight into the potential therapeutic use of TTT for this condition.

Rapeseed siliques easily shatter after ripening, resulting in a significant amount of grain loss, which delimits the development of rapeseed machine harvest. However, the effect of nitrogen (N) and density interaction on the characteristics of rape siliques and shattering resistance index is still vague. During the 2021–2022 and 2022–2023 growing seasons, we selected the Jiayou No. 5 rapeseed variety and set three N application levels (N1: 90 kg ha−1, N2: 180 kg ha−1, N3: 270 kg ha−1) and two density treatments (M1: 150,000 plants ha−1, M2: 300,000 plants ha−1) to research the effects of N and density interaction on morphological indexes, physiological indexes, shatter resistance index and yield of direct-seeding rapeseed siliques. The silique shatter resistance index, silique’s length, weight, moisture content, silique shell’s weight, thickness, lignin content, cellulose content and phenylalaninase (PAL) activity all increased first and then decreased with the increase in the N application rate; the N2 treatment increased by 18.38% and 26.92%, respectively, compared to the N1 and N3 treatments; 3.65%, 2.48%; 6.70%, 3.58%; 20.46%, 18.33%; 5.97%, 5.96%; 8.82%, 9.60%; 9.12%, 19.90%; 43.85%, 69%; 2.10%, 11.04%. Compared with the M1 treatment, the silique shatter resistance index, silique’s length, weight, moisture content, silique shell’s weight, thickness, lignin content, cellulose content and PAL activity were lower under M2 treatment. Correlation analysis demonstrated that the silique’s length, water content, silique shell’s weight, thickness, lignin content, cellulose content and PAL activity were significantly positively correlated with the silique shatter resistance index. Therefore, this study shows that N2M1 treatment can carry off synergy between silique shatter resistance and yield.

Abstract Background Single nucleotide polymorphisms (SNPs) and insertions/deletions (InDels) are the most abundant genetic variations and widely distribute across the genomes in plant. Development of SNP and InDel markers is a valuable tool for genetics and genomic research in radish (Raphanus sativus L.). Results In this study, a total of 366,679 single nucleotide polymorphisms (SNPs) and 97,973 insertion-deletion (InDel) markers were identified based on genome resequencing between ‘YZH’ and ‘XHT’. In all, 53,343 SNPs and 4,257 InDels were detected in two cultivars by transcriptome sequencing. Among the InDel variations, 85 genomic and 15 transcriptomic InDels were newly developed and validated PCR. The 100 polymorphic InDels markers generated 207 alleles among 200 Chinese radish germplasm, with an average 2.07 of the number of alleles (Na) and with an average 0.33 of the polymorphism information content (PIC). Population structure and phylogenetic relationship revealed that the radish cultivars from northern China were clustered together and the southwest China cultivars were clustered together. RNA-Seq analysis revealed that 11,003 differentially expressed genes (DEGs) were identified between the two cultivars, of which 5,020 were upregulated and 5,983 were downregulated. In total, 145 flowering time-related DGEs were detected, most of which were involved in flowering time integrator, circadian clock/photoperiod autonomous, and vernalization pathways. In flowering time-related DGEs region, 150 transcriptomic SNPs and 9 InDels were obtained. Conclusions The large amount of SNPs and InDels identified in this study will provide a valuable marker resource for radish genetic and genomic studies. The SNPs and InDels within flowering time-related DGEs provide fundamental insight into for dissecting molecular mechanism of bolting and flowering in radish.

Abstract Background With the growing challenge of an aging population, addressing the needs of elderly individuals who face living difficulties and lack family support becomes increasingly difficult. Volunteer services are crucial in this context, yet their effectiveness is hindered by unclear service scopes and uncertain volunteer inclinations. Aim To explore the role and specific preference of volunteers with nursing backgrounds in support of older adults living in the urban community. Design, Setting and Participants A descriptive qualitative study was conducted between September and October 2022. Twenty‐three participants (hospital nurses [10], community nurses [4], nursing teachers [4] and nursing students [5]) were selected. Data analysis followed conventional content analysis. Results Nine major themes were identified based on interview data. Four themes described the service scope of nursing volunteers: (1) environment domain, (2) physiological domain, (3) psychosocial domain and (4) health‐related behaviours domain. Another five themes highlighted the service inclination of these volunteers: (1) service frequency, (2) service duration per person/time, (3) service coverage, (4) service place and (5) service object. Conclusion This study clarifies the service focus and scope of necessary support for volunteers, exploring the potential service capabilities of scarce volunteers to the greatest extent. Meanwhile, the results of this study also provide a foundation for stakeholders to fully exploit the synergy. The important findings of this study will help the government and relevant authorities better understand the service attributes of nursing volunteers, allowing them to develop detailed training plans and provide nursing volunteers with targeted support and assistance to meet the health expectations of urban community‐living older adults in need. The level of informatization literacy of college teachers is related to the comprehensive development of design professionals. In this paper, from the perspective of design practice teaching, we utilize the OBE model to construct a “one-core, six-double” design professional talent cultivation model. We also point out the prospects for applying the OBE model in design practice teaching. The Delphi method is used to construct the teachers’ information literacy evaluation index system. To avoid the influence of the correlation between the indexes on the evaluation results, the AHP-gray correlation combination algorithm is used to calculate the weights of the evaluation indexes. Finally, the complete evaluation index system was used to perform empirical analysis. The results show that the F-statistic of teachers’ informatization awareness and attitude (P1) is 2.349, and the P-value is 0.168, which is greater than 0.1, indicating that there is no significant difference between the scores of teachers’ informatization awareness and attitude in universities in Guangzhou and Shanghai. This study provides a foundation and reference for improving the computer literacy of design practice teaching faculty in applied colleges and universities. It aids in the advancement of design practice teaching.

With the rapid development of the information age, the traditional data center network management can no longer meet the rapid expansion of network data traffic needs. Therefore, the research uses the biological ant colony foraging behavior to find the optimal path of network traffic scheduling, and introduces pheromone and heuristic functions to improve the convergence and stability of the algorithm. In order to find the light load path more accurately, the strategy redefines the heuristic function according to the number of large streams on the link and the real-time load. At the same time, in order to reduce the delay, the strategy defines the optimal path determination rule according to the path delay and real-time load. The experiments show that under the link load balancing strategy based on ant colony algorithm, the link utilization ratio is 4.6% higher than that of ECMP, while the traffic delay is reduced, and the delay deviation fluctuates within ±2 ms. The proposed network data transmission scheduling strategy can better solve the problems in traffic scheduling, and effectively improve network throughput and traffic transmission quality.

In this paper, the focal depth of earthquakes with magnitude 2.5 or above in Jiaokou reservoir area in 2019 was determined using the CAP method, the sPL depth phase method and the double difference positioning method. The inversion is carried out by CAP method to calculate the optimal focal mechanism solution and focal depth. The clear sPL seismic phase is identified at the near station about 50 km away from the epicenter, the theoretical waveform of various focal depths is drawn using the frequency wave velocity (F-K) method, and the focal depth is determined through fitting with the actual waveform. The seismic event pair is established, and its relative position and depth are determined using the residual between the observed value and the theoretical value of travel time difference. The results of various methods are basically the same. In contrast, the double difference positioning method is more suitable for focal depth determination of earthquakes in Jiaokou reservoir area.

Jiaokou reservoir earthquakes in Ningbo from 2009 to 2021 were relocated using the double difference positioning method. The focal mechanism solutions of large earthquakes were calculated by HASH method. Based on the location distribution, depth distribution and overall focal mechanism solution characteristics of the earthquakes after precise positioning, the seismogenic structure and seismogenic mechanism were discussed by combining with the regional geological tectonic background, geological conditions in the reservoir area and the change of reservoir water level. The results show that the seismogenic mechanism of the Jiaokou reservoir earthquake sequence in 2009—2012 is similar to that in 2018—2021, and the regional fault Changxing—Fenghua may be the seismogenic fault. The reservoir water infiltrates through the fracture fracture zone, the pore water pressure diffuses, the reservoir water softens and weakens the rock for a long time, causing the fault plane and some weak parts in the deep part to become unstable areas, and finally triggering the earthquake under the loading and unloading stress field caused by the change of reservoir water.

ABSTRACT The 2022 World Health Organization guidelines recommend use of two core anti-tuberculosis (TB) drugs, bedaquiline (BDQ) and clofazimine (CFZ), for treatment of drug-resistant (DR)-TB. However, several mutated Mycobacterium tuberculosis (MTB) genes, conferring BDQ and CFZ resistance, have been reported that predominantly arose from sporadic mutations that have not been comprehensively characterized. Herein, MTB clinical isolates collected from drug-susceptible (DS)-, multidrug-resistant (MDR)-, and extensively drug-resistant (XDR)-TB patients were cultured in vitro with BDQ or CFZ to generate progeny strains with resistance to these drugs. Progeny strains exposed to CFZ exhibited increased CFZ minimum inhibitory concentrations (MICs) that exceeded MIC increases of BDQ-exposed progeny strains. Notably, mmpR and pepQ mutations accounted for 83% and 17% of BDQ-induced spontaneous gene mutations, respectively, and 86% and 14% of CFZ-induced spontaneous gene mutations, respectively. Analyses of predicted mutation-induced changes in amino acid sequences and structures of MmpR and PepQ mutants revealed several point mutations affected sequence conversation and functionality as an underlying mechanism for observed acquired BDQ/CFZ resistance. Moreover, our results revealed differences in patterns of BDQ- and CFZ-induced acquired spontaneous mutations that may enhance our understanding of MTB BDQ/CFZ-resistance mechanisms. IMPORTANCE This study of MTB drug resistance mechanisms revealed patterns of spontaneous MTB mutations associated with acquired BDQ and CFZ resistance that arose after clinical MTB isolates were cultured in vitro with BDQ or CFZ. Results of protein sequence and structural analyses provided insights into potential mechanisms underlying associations between MTB gene mutations and DR phenotypes. Taken together, these results revealed differences in acquired BDQ and CFZ resistance mechanisms as a new perspective that may enhance our understanding of BDQ/CFZ resistance mechanisms and facilitate the development of new methods for detecting MTB drug resistance genes.

This paper examined the effects of different doses of electron beam irradiation on the sensory evaluation, microbial content and changes in volatile components of Zhaotong soybean paste, aiming to investigate the feasibility of irradiation sterilization for Zhaotong soybean paste. According to the results, when the electron beam irradiation dose was 3.68 kGy, the comprehensive sensory evaluation of Zhaotong soybean paste was good, and the total number of bacteria, fungi and coliform contents met the health standards. A total of 41 volatile components were identified in Zhaotong soybean paste before irradiation and 100 volatile components were detected after irradiation. The volatile components after irradiation mainly consisted of 32 esters, 4 aldehydes, 26 alkenoids, 15 alcohols, 11 ketones, 1 ether, 2 phenols and 9 other substances. Among them, the irradiation dose had greater effects on alcohols, esters, aldehydes and other types of volatile substances. The types and contents of esters increased significantly after irradiation, while the contents of aldehydes and alkylenes decreased. The volatile components of Zhaotong soybean paste before irradiation were mainly isobutanol, linalool, 4,7-dimethyl undecane and methyl linoleate, accounting for 36.90%, 20.29%, 11.70% and 4.94% of the total content, respectively. On the other hand, the volatile components in irradiated Zhaotong paste were dominantly linalool, methyl linoleate, ethyl benzoate and Ligustrazine. The relative contents of isobutanol decreased significantly after irradiation while linalool remarkably increased. Based on the results, it could be concluded that 3.68 kGy was a reliable dose for the irradiation sterilization of Zhaotong soybean paste, and the results would provide reference information for the promotion of Zhaotong soybean paste to the highland local specialties.

A Co32Ni21Cr8Al0.5Y alloy (by mass percentage) has been additively manufactured by selective laser melting (SLM). The as-deposited alloy has a γ matrix dispersing Al-rich β phase which is coarsened with time at high temperatures. The alloy grows a double-layered oxide scale consisting of outer (Co,Ni)(Cr,Al)2O4 and inner Al2O3 during 1000 h isothermal oxidation in air at 900 and 1000 ℃. The Al2O3 growth is dominated after a short term, particularly at the higher temperature. The growth rate of inner Al2O3 layer during cyclic oxidation with respect to isothermal oxidation at 1000 ℃ appears to be promoted, leading to extensive incorporation of Y-rich particles into the alumina-dominated scale. The CoNiCrAlY alloy mainly forms a major Al2O3 scale after 40 h during hot corrosion at 800 and 900 ℃ under Na2SO4+NaCl mixture. The dependence of the microstructure of the oxide scales formed on the different corrosion conditions is fully discussed.

Field experiments were conducted to study the effects of different proportions of controlled-release nitrogen fertilizer mixed with quick-acting nitrogen fertilizer on the yield and nitrogen utilization efficiency of direct-seeding rapeseed. Using a conventional nitrogen application rate of 180 kg ha−1 as a control, a total of 5 types of available nitrogen fertilizers and different proportions of controlled-release nitrogen fertilizers were mixed for fertilizer treatment. The proportion of available nitrogen fertilizer used was 135 kg ha−1, and the addition ratios of the five types of controlled-release nitrogen fertilizers were 0%, 30%, 50%, 70%, and 100%, respectively (i.e., the proportion of controlled-release nitrogen to the total nitrogen application amount). These ratios were represented as N135R0, N135R1, N135R2, N135R3, and N135R4, respectively. The results showed that there was no significant difference in the number of pods per plant, the number of seeds per pod, or the grain yield under the treatment of controlled-release nitrogen fertilizer mixed with quick-acting nitrogen fertilizer for proportions of 30–50% (N135R1~R3) when compared with the control, and a stable yield was achieved. Mixing controlled-release nitrogen fertilizer under reduced nitrogen application can significantly improve the apparent utilization rate of rapeseed nitrogen fertilizer, but it first increases and then decreases with the increase of the controlled-release nitrogen mixing ratio, reaching its highest under the N135R2 treatment. The agronomic utilization efficiency and partial productivity of nitrogen fertilizer first increased and then decreased with the increased proportion of controlled-release nitrogen, and both reached their highest utilization with the N135R2 treatment. The mixed treatment of controlled-release nitrogen did not affect soil urease activity, but significantly increased soil sucrase activity. The mixed treatment of controlled-release nitrogen also increased soil microbial biomass nitrogen and carbon content. Especially in the flowering stage, the soil microbial biomass nitrogen and carbon content was significantly higher under a controlled-release nitrogen mixing ratio of 30–50%. At the same time, it had a similar effect on soil inorganic nitrogen content. Therefore, a controlled-release nitrogen mixing treatment provided sufficient nitrogen for the key growth period of rapeseed. Under the condition of reducing the amount of nitrogen fertilizer by 25% based on the amount of nitrogen fertilizer applied to conventional rapeseed, the application of controlled-release urea mixed with common nitrogen fertilizer mixed at a ratio of 30–50% can be an effective way to maintain grain yield levels and improve nitrogen utilization efficiency.

Abstract Focussing on the insulating material of power transformer—cellulose insulation paper, this paper first introduces the basic characteristics and engineering application background of insulation paper. Then, the research progress of improving the mechanical properties, thermal stability and electrical properties of insulation paper by nano modification in recent years is compared, and the promotion mechanism is analysed. In addition, in practical engineering application, the oil–paper composite insulation system composed of insulation paper and insulating oil is used for the internal insulation of oil immersed transformer. Therefore, the influence of modified insulation paper on the performance of oil–paper composite insulation system is analysed. Finally, the newly reported new insulation paper is introduced, and the future development trend of modification of traditional insulation paper is analysed.

Abstract Background Glioblastoma (GBM) has a high incidence rate, invasive growth, and easy recurrence, and the current therapeutic effect is less than satisfying. Pyroptosis plays an important role in morbidity and progress of GBM. Meanwhile, the tumor microenvironment (TME) is involved in the progress and treatment tolerance of GBM. In the present study, we analyzed prognosis model, immunocyte infiltration characterization, and competing endogenous RNA (ceRNA) network of GBM on the basis of pyroptosis-related genes (PRGs). Methods The transcriptome and clinical data of 155 patients with GBM and 120 normal subjects were obtained from The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx). Lasso (Least absolute shrinkage and selection operator) Cox expression analysis was used in predicting prognostic markers, and its predictive ability was tested using a nomogram. A prognostic risk score formula was constructed, and CIBERSORT, ssGSEA algorithm, Tumor IMmune Estimation Resource (TIMER), and TISIDB database were used in evaluating the immunocyte infiltration characterization and tumor immune response of differential risk samples. A ceRNA network was constructed with Starbase, mirtarbase, and lncbase, and the mechanism of this regulatory axis was explored using Gene Set Enrichment Analysis (GSEA). Results Five PRGs (CASP3, NLRP2, TP63, GZMB, and CASP9) were identified as the independent prognostic biomarkers of GBM. Prognostic risk score formula analysis showed that the low-risk group had obvious survival advantage compared with the high-risk group, and significant differences in immunocyte infiltration and immune related function score were found. In addition, a ceRNA network of messenger RNA (CASP3, TP63)–microRNA (hsa-miR-519c-5p)–long noncoding RNA (GABPB1-AS1) was established. GSEA analysis showed that the regulatory axis played a considerable role in the extracellular matrix (ECM) and immune inflammatory response. Conclusions Pyroptosis and TME-related independent prognostic markers were screened in this study, and a prognosis risk score formula was established for the first time according to the prognosis PRGs. TME immunocyte infiltration characterization and immune response were assessed using ssGSEA, CIBERSORT algorithm, TIMER, and TISIDB database. Besides a ceRNA network was built up. This study not only laid foundations for further exploring pyroptosis and TME in improving prognosis of GBM, but also provided a new idea for more effective guidance on clinical immunotherapy to patients and developing new immunotherapeutic drugs.

The linear Minimum Mean Square Error (MMSE) signal detection algorithm in the massive Multiple-Input Multiple-Output (MIMO) system is difficult to use in practical engineering due to the complex inversion of the high-dimensional matrix of the signal detection algorithm. In order to solve this problem, the article proposes a low-complexity hybrid iterative algorithm based on the idea of matrix block and combined with Neumann series expansion algorithm. Using the Neumann series second-order expansion of the inverse matrix of the weighted matrix in the MMSE algorithm as the initial value of the iterative inversion of the block matrix can effectively improve the convergence speed of the algorithm. The simulation results show that this method can have similar performance as the traditional MMSE algorithm with fewer iterations and lower complexity.

The oxidation of β-NiAl at high temperatures leads to the preferential formation of metastable alumina, such as θ-Al2O3, which exhibits a significantly faster growth rate compared to stable α-Al2O3. However, our recent research has shown that through the use of the surface-dispersing nanoparticles (NPs) of metal oxides with a hexagonal closed pack (hcp), such as α-Al2O3, the thermal growth of α-Al2O3 can be facilitated. The present study employed laser additive manufacturing (LAM) to develop an integrated α-Al2O3 NPs surface-seeded two-phase intermetallic alloy comprising brittle β-NiAl and tougher γ’-Ni3Al, which demonstrated better comprehensive mechanical properties. It was found that seeding the α-Al2O3 NPs promoted the early stage growth of α-Al2O3 on both β and γ’ phases during oxidation in air at 1000 °C. This led to a decrease in the oxidation rate but an enhancement in adhesion of the formed alumina scale in comparison to the naked β/γ’ two-phase alloy. The reasons for this result were interpreted.

Background: Teng-Long-Bu-Zhong-Tang (TLBZT) is a traditional Chinese herbal medicine used to treat colorectal cancer (CRC). In the present study, we observed the anti-cancer effects of TLBZT on human RKO CRC. Materials and Methods: Mice were subcutaneously transplanted with RKO cells, divided into control, 5-Fu-administered, TLBZT-administered, and TLBZT and 5-Fu combination-administered groups, and treated with 5-Fluorouracil (5-Fu) and/or TLBZT. Apoptosis was detected by TdT-mediated dUTP nick-end labeling assay. The activity of caspase-3,-8, and-9 was detected using specific commercial kits. Cell senescence was assessed using senescence β-galactosidase staining. Protein expression was evaluated by immunohistochemistry. Results: TLBZT inhibited RKO CRC tumor growth, enhanced the anti-cancer effects of 5-Fu, induced apoptosis, and activated caspase-3,-8, and-9. TLBZT induced cell senescence accompanied by the downregulation of cyclin E1 and cyclin-dependent kinase 2 expressions. TLBZT also inhibited angiogenesis and the expression of hypoxia-inducible factor 1 subunit alpha and vascular endothelial growth factor-A. Conclusions: TLBZT inhibited RKO CRC tumor growth and enhanced the anti-cancer effects of 5-Fu, and it could be associated with apoptosis and cell senescence induction, and angiogenesis inhibition.

Radish (Raphanus sativus L.) is an important root vegetable crop belonging to the Brassicaceae family. Anthocyanin rich radish varieties are popular among consumers because of their bright color and high nutritional value. However, the underlying molecular mechanism responsible for skin and flesh induce anthocyanin biosynthesis in transient overexpression, gene silencing and transcriptome sequencing were used to verify its function in radish anthocyanin accumulation, radish remains unclear. Here, we identified a long noncoding RNA LINC15957, overexpression of LINC15957 was significantly increased anthocyanin accumulation in radish leaves, and the expression levels of structural genes related to anthocyanin biosynthesis were also significantly increased. Anthocyanin accumulation and expression levels of anthocyanin biosynthesis genes were significantly reduced in silenced LINC15957 flesh when compared with control. By the transcriptome sequencing of the overexpressed LINC15957 plants and the control, 5,772 differentially expressed genes were identified. A total of 3,849 differentially expressed transcription factors were identified, of which MYB, bHLH, WD40, bZIP, ERF, WRKY and MATE were detected and differentially expressed in the overexpressed LINC15957 plants. KEGG enrichment analysis revealed the genes were significant enriched in tyrosine, L-Phenylalanine, tryptophan, phenylpropanol, and flavonoid biosynthesis. RT-qPCR analysis showed that 8 differentially expressed genes (DEGs) were differentially expressed in LINC15957-overexpressed plants. These results suggested that LINC15957 involved in regulate anthocyanin accumulation and provide abundant data to investigate the genes regulate anthocyanin biosynthesis in radish.

Polystyrene nanoplastics (PS-NPs) are typical accumulated nanoplastics in the marine environment and organisms, and have strong potential risks to marine ecological environment and human health. MiRNAs could respond to and participate in the response process of environmental stressors. However, the response of miRNAs to nanoplastics has not been fully explored. In this study, miRNA responses of digestive glands in mussels Mytilus galloprovincialis treated by 200 nm PS-NPs (20, 200, 2000 μg/L) for 7 days were characterized by BGISEQ-500 deep sequencing and bioinformatics analysis, along with histopathological quantification with planimetric parameters on hematoxylin and eosin (H&E) staining. Results showed that one novel miRNA (novel_mir63) and seven known miRNAs (miR-34_2, miR-34_5, miR-281_8, let-7–5p_6, miR-10, miR-124, miR-29b-3p) were significantly (adjusted P-value < 0.05) differentially expressed after PS-NPs treatments, and most of them were down-regulated expect for novel_mir63 and miR-34_2. Function analysis of target genes corresponding to these differentially expressed miRNAs indicated that PS-NPs disturbed the process related to metabolism, aging, cardiac function, neural excitation, and repairment. Among them, acetyl-CoA C-acetyltransferase and purine metabolism pathway played vital connection roles. Meanwhile, significantly morphology changes of digestive tubes obtained from H&E stained sections also implied severely disrupted metabolic capability in digestive glands, reflected by significantly increased mean diverticular radius (MDR) and mean luminal radius (MLR) values and the ratio of MLR to mean epithelial thickness (MET), and significantly decreased MET value and MET/MDR. Overall, these findings have revealed new characterization of miRNAs and their target genes in mussel M. galloprovincialis under PS-NPs stress, and provide important clues to further elucidate the toxicity mechanisms of PS-NPs.

[Purpose/Significance] Improving college students' digital literacy is of great significance to building a digital talent power. [Method/Process] By combing the relevant policies before and after the release of Action Plan for Enhancing Digital Literacy and Skills of the People, this paper analyzes the main contents of the Action Plan and believes that it has the following significance: it determines the concept of digital literacy with Chinese characteristics and international vision, pays attention to the frontier requirements such as creativity improvement and special group service, expands four digital scenes of life, work, learning and innovation, and deploys comprehensive operations according to key tasks and projects. [Results/Conclusions] The authors argue that for various fields of library, information science and archives, the Action Plan will have extensive influence theoretically and practically, and its impact on digital literacy education of college students, in particular, is perhaps the most direct and far-reaching according to the research pattern and tradition of information literacy. The authors argue that the layout and action should be started as soon as possible for the education around digital literacy of college students.

Introduction COVID-19 has posed a serious threat to people worldwide, especially the older adults, since its discovery. Tai Chi as a traditional Chinese exercisethat belongs to traditional Chinese medicine has proven its effectiveness against COVID-19. However, no high-quality evidence is found on the dose–response relationships between Tai Chi and clinical outcomes in patients with COVID-19. This study will evaluate and determine the clinical evidence of Tai Chi as a treatment in elderly patients with COVID-19.Methods and analysis The following electronic bibliographical databases including PubMed, EMBASE, Web of Science, Cochrane Library, China National Knowledge Infrastructure, VIP Database and Wanfang Database will be screened from their inception date to 30 June 2022. All eligible randomised controlled trials or controlled clinical trials related to Tai Chi for elderly patients with COVID-19 will be included. The primary outcomes are forced expiratory volume in 1 s (FEV1), forced vital capacity (FVC) and FEV1/FVC ratio (FEV1%). The secondary outcomes are the time of main symptoms disappearance, length of hospital stay, serum levels of interleukin (IL)-6, IL-1b and tumour necrosis factor-α, and adverse event rate. Two independent reviewers will select the studies, extract the data, and analyse them on EndNote V.X9.0 and Stata V.12.1. The robust error meta-regression model will be used to establish the dose–response relationships between Tai Chi and clinical outcomes. The heterogeneity and variability will be analysed by I2 and τ2 statistics. Risk of bias, subgroup analysis and sensitivity analysis will also be performed. The quality of evidence will be assessed by the Grading of Recommendations Assessment, Development and Evaluation, and the risk of bias will be evaluated by using the Physiotherapy Evidence Database Scale.Ethics and dissemination This study will review published data; thus, obtaining ethical approval and consent is unnecessary. The results will be disseminated through peer-reviewed publications.PROSPERO registration number CRD42022327694.

Abstract Optical imaging is a most useful and widespread technique for the investigation of the structure and function of the cellular genomes. However, an analysis of immensely convoluted and irregularly compacted DNA polymer is highly challenging even by modern super-resolution microscopy approaches. Here we propose fluorescence lifetime imaging (FLIM) for the advancement of studies of genomic structure including DNA compaction, replication as well as monitoring of gene expression. The proposed FLIM assay employs two independent mechanisms for DNA compaction sensing. One mechanism relies on the inverse quadratic relation between the fluorescence lifetimes of fluorescence probes incorporated into DNA and their local refractive index, variable due to DNA compaction density. Another mechanism is based on the Förster resonance energy transfer (FRET) process between the donor and the acceptor fluorophores, both incorporated into DNA. Both these proposed mechanisms were validated in cultured cells. The obtained data unravel a significant difference in compaction of the gene-rich and gene-poor pools of genomic DNA. We show that the gene-rich DNA is loosely compacted compared to the dense DNA domains devoid of active genes.

IntroductionCadmium (Cd) is one of the most dominant heavy metals in the Bohai Sea. Our previous study proved that Cd could induce gill mitochondrial toxicity in marine animals. Herein, we aimed to elucidate the toxicity mechanism of Cd on liver mitochondria, as liver is the main metabolic and detoxification organ and generally rich in mitochondria.MethodsThe mitochondrial responses induced by Cd (5 and 50 μg/L) were characterized by observing mitochondrial morphology, measuring mitochondrial membrane potential (MMP), and proteomic and metabolomic analysis in juvenile olive flounder Paralichthys olivaceus livers.ResultsAfter water-bonre exposure for 14 days, two Cd treatments decreased MMPs significantly and caused ultrastructural-damaged mitochondria in flounder livers. NMR-based metabolomics revealed that Cd exposure mainly altered the abundances of metabolites (ATP, AMP, phosphocholine, lactate and succinate) related to energy metabolism in flounder livers. iTRAQ-based mitochondrial proteomics indicated that 27 differentially expressed proteins (DEPs) were screened out from liver mitochondria after Cd treatments. These proteins were mainly associated with energy metabolism (oxidative phosphorylation (OXPHOS) and tricarboxylic acid (TCA) cycle) and apoptosis.DiscussionThese results indicated that Cd disrupted mitochondrial morphology, energy homeostasis and apoptosis in liver mitochondria in flounder P. olivaceus. This work revealed a comprehensive view on Cd-induced mitochondrial responses in the liver tissues of flounder using an integrated proteomic and metabolomic approach.

Modern era of agriculture is concerned with the environmental influence on crop growth and development. Shading is one of the crucial factors affecting crop growth considerably, which has been neglected over the years. Therefore, a two-year field experiment was aimed to investigate the effects of shading at flowering (S1) and pod development (S2) stages on nitrogen (N) dynamics, carbohydrates and yield of rapeseed. Two rapeseed genotypes (Chuannong and Zhongyouza) were selected to evaluate the effects of shading on 15N trace isotopes, enzymatic activities, dry matter, nitrogen and carbohydrate distribution and their relationship with yield. The results demonstrated that both shading treatments disturbed the nitrogen accumulation and transportation at the maturity stage. It was found that shading induced the downregulation of the N mobilizing enzymes (NR, NiR, GS, and GOGAT) in leaves and pods at both developmental stages. Shading at both growth stages resulted in reduced dry matter of both varieties but only S2 exhibited the decline in pod shell and seeds dry weight in both years. Besides this, carbohydrates distribution toward economic organs was declined by S2 treatment and its substantial impact was also experienced in seed weight and seeds number per pod which ultimately decreased the yield in both genotypes. We also revealed that yield is positively correlated with dry matter, nitrogen content and carbohydrates transportation. In contrast to Chuannong, the Zhongyouza genotype performed relatively better under shade stress. Overall, it was noticed that shading at pod developmental stage considerable affected the transportation of N and carbohydrates which led to reduced rapeseed yield as compared to shading at flowering stage. Our study provides basic theoretical support for the management techniques of rapeseed grown under low light regions and revealed the critical growth stage which can be negatively impacted by low light.

Four Ni-Al intermetallic compounds with the Al concentrations in range of 28 to 35 (in atomic percentage) have been fabricated by laser metal deposition (LMD) through mixing appropriate contents of pure Ni powder with pre-alloyed high-Al nickel aluminide powder. Ni-35Al and Ni-32Al are both composed of more than 45 vol.% β-NiAl matrix phase dispersing γ′-Ni3Al, while Ni-30Al and Ni-28Al consisted of worm-like, needle-like γ′-Ni3Al mainly and less than 37 vol.% β-NiAl. After 20 h oxidation in air at 1000 °C, the results of thermobalance, X-ray diffraction and scanning electron microcopy, indicated that Ni-35Al and Ni-32Al are more oxidation-resistant than the Ni-30Al and Ni-28Al. Because the first two can thermally grow an external Al2O3 scale but the second two form a duplex oxide scale consisting of outer NiAl2O4 layer and inner Al2O3 layer.
