Senescence is an important cellular program occurring in development, tissue repair, cancer, and aging. Increased senescence is also associated with disease states, including obesity and Type 2 diabetes (T2D). Characterizing and quantifying senescent cells at a single cell level has been challenging and particularly difficult in large primary cells, such as human adipocytes. In this study, we present a novel approach that utilizes reflected light for accurate senescence-associated beta-galactosidase (SABG) staining measurements, which can be integrated with immunofluorescence and is compatible with primary mature adipocytes from both human and mouse, as well as with differentiated 3T3-L1 cells. This technique provides a more comprehensive classification of a cell's senescent state by incorporating multiple criteria, including robust sample-specific pH controls. By leveraging the precision of confocal microscopy to detect X-gal crystals using reflected light, we achieved superior sensitivity over traditional brightfield techniques. This strategy allows for the capture of all X-gal precipitates in SABG-stained samples, revealing diverse X-gal staining patterns and improved detection sensitivity. Additionally, we demonstrate that reflected light outperforms western blot analysis for the detection and quantification of senescence in mature human adipocytes, as it offers a more accurate representation of SABG activity. The number and area of forest fires in northern Europe have been dramatically reduced during the past century, and several fire-favoured species are now threatened. To promote the recovery of these species, prescribed burning is often used as a conservation measure, and to optimise the use of these conservation burns, knowledge is needed on suitable fire frequency, size and placement in the landscape. The aim of this study was to analyse the effect of recent fire history (12 yrs) on beetles sampled using smoke attraction traps at 21 forest sites in a 10,000 km 2 region. We analysed the odds of finding a fire-favoured beetle species or individual among the beetles in each trap using a new spatiotemporal connectivity measure and compared the results to non-fire-favoured and saproxylic species. For fire-favoured beetles, both the number of species and individuals significantly increased with connectivity to previous fires, while the other two groups did not. The spatiotemporal connectivity that best explained the patterns suggests that fire-favoured beetles mainly respond to fires within a 2 km range up to 2-3 years after the fire. Hence, to preserve fire-favoured insects, prescribed fires must be close in space and time to other fires-whether prescribed or natural.

Mathematical language (i.e., content-specific language used in mathematics) and emergent literacy skills predict children's broad numeracy development. However, little work has examined whether these domains predict development of individual numeracy skills (e.g., cardinality, number order). Thus, the aim of the current study was to examine longitudinal relations among mathematical language, emergent literacy skills, and specific early numeracy skills. Participants included 114 preschool children aged 3.12 to 5.26 years (M = 4.17 years, SD = 0.59). Specifically, this study examined whether mathematical language and three emergent literacy skills (print knowledge, phonological awareness, and general vocabulary) in the fall of preschool predicted 12 individual early numeracy skills in the spring, controlling for age, sex, rapid automatized naming, parent education, and autoregressors. Results indicated that mathematical language predicted development of most of the early numeracy skills (e.g., set comparison, numeral comparison, numeral identification), but findings for emergent literacy skills were not robust. Among the three emergent literacy skills, only print knowledge was a significant predictor of development in some specific numeracy skills, including verbal counting, number order, and story problems. Results highlight the important role of mathematical language in children's numeracy development and provide the foundation for future work in designing interventions to improve early numeracy skills., (Copyright © 2024 Elsevier Inc. A limited body of work has examined the nature and scope of young children's science-related activities outside of the school context, and thus there is little understanding or consensus regarding what comprises the home science environment (HSE; e.g., interactions, activities, resources) and how specific factors of the HSE relate to children's science performance. The two primary goals of this study were to (a) examine the factor structure of a parent-report measure of home science interactions and (b) evaluate how these factors relate to the science core knowledge of young children from families with low incomes. A total of 125 families with children aged 3 to 5 years (52 girls) participated in the study. Children were assessed on a measure of science core knowledge, and parents completed a brief questionnaire on their home science interactions that included questions pertaining to both home science disciplinary core idea (DCI) engagement and home science and engineering practice (SEP) engagement. Findings revealed that although separating home science interactions into distinct DCI and SEP factors represented the data well, the best overall representation of home science interactions was a one-factor model that included only home DCI engagement items. In addition, home DCI engagement was significantly predictive of children's science core knowledge over and above a large group of covariates, including children's age, race/ethnicity, sex, and performance on math, executive function, and vocabulary tasks as well as their parents' education., (Published by Elsevier Inc.)

While loss-of-function variants in the WAS gene are associated with Wiskott-Aldrich syndrome and lead to microthrombocytopenia, gain-of-function variants of WAS are associated with X-linked neutropenia (XLN) and the absence of microthrombocytopenia. Only a few XLN families have been reported so far, and their platelet phenotype was not described in detail. To date, no renal involvement was described in XLN. In the present study, we report exome sequencing of individuals from 3 generations of a family with a dominant disease combining neutropenia, macrothrombocytopenia, and renal failure. We identified a heterozygous missense gain-of-function variant in the WAS gene (c.881T>C, p.I294T) that segregates with the disease and is already known to cause XLN. There was no pathogenic variant in MYH9, TUBB1, or ACTN1. The development of B cells relies on an intricate network of transcription factors critical for developmental progression and lineage commitment. In the B cell developmental trajectory, a temporal switch from predominant Foxo3 to Foxo1 expression occurs at the CLP stage. Utilizing VAV-iCre mediated conditional deletion, we found that the loss of FOXO3 impaired B cell development from LMPP down to B cell precursors, while the loss of FOXO1 impaired B cell commitment and resulted in a complete developmental block at the CD25 negative proB cell stage. Strikingly, the combined loss of FOXO1 and FOXO3 resulted in the failure to restrict the myeloid potential of CLPs and the complete loss of the B cell lineage. This is underpinned by the failure to enforce the early B-lineage gene regulatory circuitry upon a predominantly pre-established open chromatin landscape.
