The application of high‐frequency radar as an instrument for assimilating tsunami‐induced current fields is garnering increasing interest. The performance of surface current velocity measurements depends on the azimuthal differences between the crossing radar beams at the measurement points. This study aimed to incorporate the measurement error distributions of the east‐west and north‐south velocity components into tsunami data assimilation based on an optimal interpolation method, assuming Gaussian noise with the time‐invariant and a uniform standard deviation (STD = 5 cm/s) of radial velocity measurements. Through the empirical orthogonal function (EOF) analysis of radar‐derived surface currents in the Kii Channel, Japan, the velocities reconstructed using higher modes (EOFs 16–274) were associated with measurement errors, portraying nonuniform distribution depending on the crossing beam angle of two radar beams. Based on independent fifteen‐time assimilation experiments for two different tsunami scenarios, for a uniform water depth of 500 m, we observed a significant improvement of up to 29% and 0.9% in the assimilation performance (on average) over the along‐coast stations for scenarios with 1‐ and 5‐m maximum initial sea surface heights, respectively. The measurement errors dependent on the crossing beam angle reduced the error‐induced tsunamis, resulting in stable assimilations, with lower STDs in the fifteen‐time assimilation performances. When the STD of Gaussian noise varies with time, it is important to consider the temporal change in the radial velocity measurement errors and/or noise‐filtering techniques, to maintain a certain level of noise intensity. Plain Language Summary: High‐frequency radar is valuable for monitoring tsunamis as it measures tsunami‐induced surface current velocities, not the tsunami height, which is crucial for tsunami evacuation. In this study, we estimated a reasonable tsunami wave height using a data assimilation method incorporating high‐frequency radar measurements with tsunami numerical simulation. To minimize the adverse effects of high‐frequency radar measurement errors and obtain accurate tsunami wave heights, it is essential to estimate the errors appropriately, especially considering their dependency on the radar beam angle. We estimated the beam angle‐dependent errors through a principal component analysis and integrated them into the assimilation process. By doing so, we successfully reproduced the tsunami wave heights, demonstrating an improvement over estimations made under the assumption of constant errors and mitigating the negative impact of measurement errors. Background: Clinical amyopathic dermatomyositis is characterized by cutaneous symptoms but lacks muscle symptoms. Anti-melanoma differentiation-associated gene 5 antibodies are frequently found in Japanese patients with clinical amyopathic dermatomyositis. Patients with rapidly progressive interstitial lung disease with positive anti-melanoma differentiation-associated gene 5 antibodies have poor prognoses, and majority of them are treated with combination immunosuppressive therapy; however, the best treatment is yet to be determined. Case presentation: A 52-year-old Asian male patient presented with a chief complaint of dyspnea on exertion. He had a typical skin rash and rapidly progressive interstitial pneumonia. Additionally, anti-melanoma differentiation-associated gene 5 antibodies were detected; therefore, he was diagnosed with dermatomyositis-associated interstitial pneumonia. Respiratory failure worsened despite administering steroid pulse therapy, tacrolimus, and cyclophosphamide. Consequently, plasma exchange was performed on day 13 of admission. After a slight improvement, the patient's respiratory failure worsened. Thus, cyclophosphamide was replaced by tofacitinib on day 28. Although respiratory failure improved and the progression of interstitial pneumonia seemed under control, βD-glucan level increased and Aspergillus antigen was detected on day 49. Micafungin and voriconazole were administered, but the patient succumbed to worsening respiratory failure on day 61. The pathological autopsy revealed multiple nodular lesions with cavity formation in both lungs and the presence of Aspergillus with severe neutrophilic infiltration and necrosis, which supported the diagnosis of invasive pulmonary aspergillosis. Conclusion: The patient with anti-melanoma differentiation-associated gene 5 antibody-related rapidly progressive interstitial lung disease, whose disease was difficult to control after the administration of triple immunosuppressive therapy (steroids, tacrolimus, and cyclophosphamide), showed good response with tofacitinib. Unfortunately, the patient died of invasive pulmonary aspergillosis owing to severe immunosuppression; thus, the signs of complications should be promptly detected. [ABSTRACT FROM AUTHOR]

After exocytosis, release sites are cleared of vesicular residues to replenish with transmitter-filled vesicles. Endocytic and scaffold proteins are thought to underlie this site-clearance mechanism. However, the physiological significance of this mechanism at diverse mammalian central synapses remains unknown. Here, we tested this in a physiologically optimized condition using action potential evoked EPSCs at fast calyx synapse and relatively slow hippocampal CA1 synapse, in post-hearing mice brain slices at 37°C and in 1.3 mM [Ca 2+ ]. Pharmacological block of endocytosis enhanced synaptic depression at the calyx synapse, whereas it attenuated synaptic facilitation at the hippocampal synapse. Block of scaffold protein activity likewise enhanced synaptic depression at the calyx but had no effect at the hippocampal synapse. At the fast calyx synapse, block of endocytosis or scaffold protein activity significantly enhanced synaptic depression as early as 10 ms after the stimulation onset. Unlike previous reports, neither endocytic blockers nor scaffold protein inhibitors prolonged the recovery from short-term depression. We conclude that the release-site clearance by endocytosis can be a universal phenomenon supporting vesicle replenishment at both fast and slow synapses, whereas the presynaptic scaffold mechanism likely plays a specialized role in vesicle replenishment predominantly at fast synapses., Competing Interests: SM, TT No competing interests declared, (© 2023, Mahapatra and Takahashi.)

Thymic adenocarcinomas are rare. We herein report for the first time a case of thymic adenocarcinoma with positivity for thyroid transcription factor-1 (TTF-1) and a BRAF V600E mutation. A 50-year-old woman had persistent suffocation and chest pain. Computed tomography revealed a 42×28-mm tumor in the anterior mediastinum. The patient underwent tumor resection using video-assisted thoracoscopic surgery. Histopathological findings showed thymic papillary adenocarcinoma, Masaoka stage II. Immunohistochemically, the tumor was positive for TTF-1. A sequencing analysis revealed a BRAF V600E mutation. The patient underwent postoperative radiotherapy and was in good health without recurrence at five months after resection.
