BACKGROUND: Hyperphosphatemia is associated with increased morbidity and mortality in patients with end-stage kidney disease (ESKD). Whereas clinical and observational studies have demonstrated the effectiveness of sucroferric oxyhydroxide (SO) in controlling serum phosphorus (sP) in ESKD, data on the real-world impact of switching to SO in patients on peritoneal dialysis (PD) are limited. In this retrospective database analysis, we examine the impact of SO on sP management over a 1-year period among PD patients prescribed SO as part of routine clinical care. METHODS: We analyzed de-identified data from adults on PD in Fresenius Kidney Care clinics who were prescribed SO monotherapy between May 2018 and December 2019 as part of routine clinical management. Changes from baseline in sP levels, phosphate binder (PB) pill burden, and laboratory parameters were evaluated during the four consecutive 91-day intervals of SO treatment. RESULTS: The mean age of the 402 patients who completed 1 year of SO was 55.2 years at baseline, and they had been on PD for an average of 19.9 months. SO was initiated with no baseline PB recorded in 36.1% of patients, whereas the remaining 257 patients were switched to SO from sevelamer (39.7%), calcium acetate (30.4%), lanthanum (1.2%), ferric citrate (14.0%), or more than one PB (14.8%). Mean sP at baseline was 6.26 mg/dL. After being prescribed SO, the percentage of patients achieving sP ≤ 5.5 mg/dL increased from 32.1% (baseline) to 46.5-54.0% during the 1-year follow-up, whereas the mean number of PB pills taken per day decreased from 7.7 at baseline (among patients on a baseline PB) to 4.6 to 5.4. Serum phosphorus and PB pill burden decreased regardless of changes in residual kidney function over the 12-month period. Similar results were observed for the full cohort (976 patients who either completed or discontinued SO during the 1-year follow-up). CONCLUSIONS: Patients on PD who were prescribed SO as part of routine care for phosphorus management experienced significant reductions in SP and PB pills per day and improvements in sP target achievement, suggesting the effectiveness of SO on SP management with a concurrent reduction in pill burden.

Development of high yielding cowpea varieties coupled with good taste and rich in essential minerals can promote consumption and thus nutrition and profitability. The sweet taste of cowpea grain is determined by its sugar content, which comprises mainly sucrose and galacto-oligosaccharides (GOS) including raffinose and stachyose. However, GOS are indigestible and their fermentation in the colon can produce excess intestinal gas, causing undesirable bloating and flatulence. In this study, we aimed to examine variation in grain sugar and mineral concentrations, then map quantitative trait loci (QTLs) and estimate genomic-prediction (GP) accuracies for possible application in breeding. Grain samples were collected from a multi-parent advanced generation intercross (MAGIC) population grown in California during 2016-2017. Grain sugars were assayed using high-performance liquid chromatography. Grain minerals were determined by inductively coupled plasma-optical emission spectrometry and combustion. Considerable variation was observed for sucrose (0.6-6.9%) and stachyose (2.3-8.4%). Major QTLs for sucrose (QSuc.vu-1.1), stachyose (QSta.vu-7.1), copper (QCu.vu-1.1) and manganese (QMn.vu-5.1) were identified. Allelic effects of major sugar QTLs were validated using the MAGIC grain samples grown in West Africa in 2017. GP accuracies for minerals were moderate (0.4-0.58). These findings help guide future breeding efforts to develop mineral-rich cowpea varieties with desirable sugar content.

(1) Background: Clinical results on the effects of excess sugar consumption on insulin sensitivity are conflicting, possibly due to differences in sugar type and the insulin sensitivity index (ISI) assessed. Therefore, we compared the effects of consuming four different sugars on insulin sensitivity indices derived from oral glucose tolerance tests (OGTT). (2) Methods: Young adults consumed fructose-, glucose-, high-fructose corn syrup (HFCS)-, sucrose-, or aspartame-sweetened beverages (SB) for 2 weeks. Participants underwent OGTT before and at the end of the intervention. Fasting glucose and insulin, Homeostatic Model Assessment-Insulin Resistance (HOMA-IR), glucose and insulin area under the curve, Surrogate Hepatic Insulin Resistance Index, Matsuda ISI, Predicted M ISI, and Stumvoll Index were assessed. Outcomes were analyzed to determine: (1) effects of the five SB; (2) effects of the proportions of fructose and glucose in all SB. (3) Results: Fructose-SB and the fructose component in mixed sugars negatively affected outcomes that assess hepatic insulin sensitivity, while glucose did not. The effects of glucose-SB and the glucose component in mixed sugar on muscle insulin sensitivity were more negative than those of fructose. (4) Conclusion: the effects of consuming sugar-SB on insulin sensitivity varied depending on type of sugar and ISI index because outcomes assessing hepatic insulin sensitivity were negatively affected by fructose, and outcomes assessing muscle insulin sensitivity were more negatively affected by glucose.

Summary: The sweetness of apple fruit is a key factor in the improvement of apple varieties, with fructose being the sweetest of the soluble sugars, playing a crucial role in determining the overall sweetness of the apple. Therefore, uncovering the key genes controlling fructose accumulation and deciphering the regulatory mechanisms of fructose are vitally important for the improvement of apple varieties.In this study, through BSA‐seq and transcriptome analysis of the 'Changfu 2' × 'Golden Delicious' F1 hybrid population, MdNAC5 was identified as a key regulatory gene for fructose content.MdNAC5 was shown to significantly influence fructose accumulation in both apples and tomatoes. Furthermore, we conducted a detailed identification of sugar transporters and metabolic enzymes in apples, discovering that MdNAC5 can enhance fructose accumulation in vacuoles and the conversion of sucrose to fructose by binding to and activating the promoters of the vacuolar sugar transporter MdTST2 and the neutral invertase MdNINV6. Additionally, MdNAC5 regulated the MdEIN3.4‐MdSWEET15a module, strengthening the unloading of sucrose in the phloem of the fruit.Our results reveal a new mechanism by which MdNAC5 regulates fructose accumulation in apples and provide theoretical foundations for improving apple sweetness through genetic modification. The glycosylation of an alcohol group from a sugar nucleotide substrate involves proton release, so the reaction is favored thermodynamically at high pH. Here, we explored expansion of the alkaline pH range of sucrose synthase (SuSy; EC 2.4.1.13) to facilitate enzymatic glycosylation from uridine 5'-diphosphate (UDP)-glucose. The apparent equilibrium constant of the SuSy reaction (UDP-glucose + fructose ↔ sucrose + UDP) at 30 °C increases by ∼4 orders of magnitude as the pH is raised from 5.5 to 9.0. However, the SuSy in solution loses ≥80 % of its maximum productivity at pH ∼7 when alkaline reaction conditions (pH 9.0) are used. We therefore immobilized the SuSy on nanocellulose-based biocomposite carriers (∼48 U/g carrier; ≥ 50 % effectiveness) and reveal in the carrier-bound enzyme a substantial broadening of the pH-productivity profile to high pH, with up to 80 % of maximum capacity retained at pH 9.5. Using reaction by the immobilized SuSy with automated pH control at pH ∼9.0, we demonstrate near-complete conversion (≥ 96 %) of UDP-glucose and fructose (each 100 mM) into sucrose, as expected from the equilibrium constant (K eq = ∼7 × 102) under these conditions. Adeno-associated virus (AAV) vectors are widely used to deliver therapeutic transgenes due to their superior safety, relatively low immunogenicity, and ability to target diverse tissues. AAV gene therapy products are typically formulated as frozen liquid and stored below − 60 °C, and therefore are subjected to multiple freeze/thaw cycles during manufacturing and administration. Recent studies have shown that genome DNA leakage could be induced by freeze/thaw stress. DNA leakage from AAV capsids has been reported to potentially impact product stability, induce immune responses, and compromise product efficacy. Thus, further characterization to improve the understanding of genome DNA leakage is necessary for mitigating the risks associated with genome DNA leakage during AAV product development. In this work, we developed an optimized size-exclusion chromatography (SEC) method for quantifying the leakage of genome DNA across multiple different AAV serotypes and demonstrated satisfactory assay performance in sensitivity, precision, and linearity. Furthermore, we showed that this method could also be applied to quantifying additional quality attributes of AAV, including the percentage of full capsids and quantification of AAV dimers. By using this optimized SEC method, we demonstrated that significantly increased free DNA was observed with increasing freeze/thaw cycles or at a temperature approaching the onset temperature for genome DNA ejection, which was effectively mitigated by the addition of 1.5% w/v sucrose in the AAV formulation. Thus, this optimized SEC method can serve as an invaluable tool for AAV formulation, product, and process development in ensuring the quality and stability of AAV gene therapy products. Aim Congenital sucrase-isomaltase deficiency (CSID) is an autosomal recessive disease with a mutation in the sucrase-isomaltase (SI) gene and disaccharide maldigestion. With limited data, the estimated incidence of this disease is 57.59/10⁶ births and its heterozygosity rate is 1/132. We aimed to evaluate cases who underwent whole exome sequencing (WES) analysis in the medical genetics unit with regards to the frequency and clinic of CSID. Materials and Methods The patients' files who underwent WES between 2018-2023 were evaluated retrospectively. The demographic characteristics, complaints, physical examination, and laboratory findings of those patients with SI gene mutations were recorded. Cases with mutations were recontacted and symptom questioning was performed. Results Mutations were detected in 25 (8.3%) of 300 patients who underwent WES analysis. One case had a compound heterozygous mutation, while 24 cases were heterozygous. The mean age was 22.4±17.6 years (1.8-52 years) and 16 (64%) were females. Nine of the cases (36%) were symptomatic. Conclusion Data on the frequency of CSID are insufficient and variable in the literature. The SI heterozygosity rate in our study was higher than some studies. There were similar rates when compared to the study conducted on symptomatic pediatric patients in our country. Although the data of our study includes a heterogeneous group to evaluate the frequency of CSID, it consists of a group with suspected genetic diseases rather than healthy individuals. Sucrose functions as a signaling molecule in several metabolic pathways as well as in various developmental processes. However, the molecular mechanisms by which sucrose regulates these processes remain largely unknown. In the present study, we demonstrate that sucrose promotes flowering by mediating the stability of a regulatory protein that represses flowering in rice. Exogenous application of sucrose promoted flowering by inducing florigen gene expression. Reduction of sucrose levels in the phloem through genetic modifications, such as the overexpression of the vacuolar invertase OsVIN2 or the mutation of OsSUT2, a sucrose transporter, delayed flowering. Analysis of relative transcript levels of floral regulatory genes showed that sucrose activated Ehd1 upstream of the florigen, with no significant effect on the expression of other upstream genes. Examination of protein stability after sucrose treatment of major floral repressors revealed that the Ghd7 protein was specifically degraded. The Ghd7 protein interacted with the E3 ligase IPA INTERACTING PROTEIN1 (IPI1), and sucrose‐induced K48‐linked polyubiquitination of Ghd7 via IPI1, leading to protein degradation. Mutants defective in IPI1 delayed flowering, confirming its role in modulating proteins involved in flowering. Numerous studies have evaluated snap bean pod yield and quality at different pod maturities, but only a few have evaluated yield and pod quality across different seasons within the same year or for pods harvested at the same relative maturity successively from the same plants. Hence, the aim of this study was to determine if snap bean yield or pod sugar accumulation differed for pods at the same relative stage of maturity from plants harvested multiple times in spring and fall planting seasons in north central Oklahoma. Nine snap bean cultivars (seven green-podded and two yellow-podded) were field-grown in Spring and Fall 2022 and 2023. Pods were harvested weekly for 3 weeks from the same plants and at the same relative pod development stage. Pod yield and sugar concentration were measured. The total pod yield pooled across seasons and years of 'Sybaris' was lower (approximately 4100 kg-ha21) than that of six of the other cultivars, which yielded between 7000 to 8600 kg·ha21. In spring, pod yield increased from 900 to 1300 kg·ha21 in the first harvest to 3500 to 3700 kg·ha21 by the last harvest. In Fall 2022, the yield decreased from 1900 kg·ha21 in the first harvest to 690 kg·ha21 by the last harvest, whereas in Fall 2023 the pod yield remained steady at 2600 to 3000 kg·ha21 at each weekly harvest. The Fall 2022 decrease in yield was likely caused by low temperatures during pod development and a freeze that shortened the harvest season. From the first to the last harvest, the total pod sugar concentration decreased from 145.5 to 107.2 mgg21, glucose decreased from 48.4 to 33.0 mg·g21, and fructose decreased from 90.7 to 52.0 mg-g21. However, the sucrose concentration increased from 9.2 to 21.9 mg·g21. This decrease in the total and monomeric sugar concentrations, coupled with a disproportionally smaller increase in sucrose, is typical for overly mature snap bean pods, which are considered less desirable because of lower sweetness. Snap bean pods harvested later in the growing season in a successive harvest system at the same relative stage of maturity as earlier harvested pods had sugar concentrations resembling those of overly mature pods, indicating that snap bean plant age may influence the pod sugar concentration. The aim of the current research is to dry apples using a solar dryer with a desiccant cycle in two modes: direct radiation -convection and indirect radiation -convection. Two types of drying methods, direct and indirect with desiccant and without desiccant, moisture content, and drying in open air were used. Pre - prepared apple slices (flavored with cinnamon) were placed on a specific -sized mesh, the initial weight of the samples was measured, and they were placed inside the solar dryer with a desiccant cycle in two modes: direct radiation -convection and indirect radiation -convection. During the drying process, the evaporation rate, texture, wrinkling, water reabsorption, vitamin C retention, color changes, and sensory characteristics at different treatment levels (30% sucrose and without sucrose, 0.5%, 1%, and 2% cinnamon) were examined. Additionally, several samples were dried as controls without cinnamon flavoring using the solar dryer. Data analysis was conducted using a completely randomized factorial design and the statistical software SPSS version 26. The results of the study showed that in both direct and indirect methods, with and without desiccant, the moisture removal rate in samples without sucrose was higher compared to samples with sucrose and cinnamon. Comparing control samples in both methods with and without desiccant moisture showed that the evaporation rate of samples with desiccant moisture was significantly higher. Furthermore, in samples with sucrose, the evaporation rate was lower in the desiccant method, and in direct sunlight drying, the drying rate was significantly lower compared to solar dryer drying. The results of evaporation rates in the indirect drying method also indicated that similar to the direct method, the evaporation rate of samples with cinnamon treatment and without sucrose was higher than that of samples with sucrose and controls. Tissue analysis results also indicated that the highest firmness was observed in the direct method without desiccant for samples without sucrose, with no significant difference in means. Cinnamon also increased the firmness in samples without desiccant moisture in the indirect drying method, and samples with sucrose had less wrinkling, with an increase in cinnamon concentration leading to more wrinkling, with the highest increase observed in the direct method without desiccant at 2% cinnamon and in the direct method with desiccant at 1% cinnamon concentration. The water reabsorption rate in both direct and indirect drying methods showed that samples with sucrose had better water reabsorption conditions, and the vitamin C degradation rate indicated that the use of sucrose improved vitamin C retention. Color change analysis also showed that samples with sucrose had more favorable conditions in both methods. Therefore, the results related to sensory evaluation characteristics indicated that samples with sucrose and different cinnamon concentrations received the highest sensory evaluation scores. The content of nonstructural carbohydrates (glucose, fructose, sucrose, starch) and activities of sucrose-utilizing enzymes (vacuolar, apoplastic, and cytoplasmic invertases, as well as sucrose synthase) were examined in halophyte plants Triglochin maritima L. (sea arrowgrass), Tripolium vulgare Ness. (sea aster), and Glaux maritima L. (sea milkwort) inhabiting the littoral zone of the White Sea coast. Studies of plants growing in tidal cycle conditions revealed species-specific differences in sugar content and the activity of sucrose-metabolizing enzymes. T. maritima featured high concentrations of monosaccharides in leaves and elevated content of sucrose and storage starch in underground organs. T. vulgare was characterized by low monosaccharide content and the activated synthesis of sucrose that is deposited in stems and plays the main role in maintaining the plant osmotic balance. G. maritima occupies a special place: unlike other species, it accumulates abundant starch reserves in aboveground and underground organs. This species is a crinohalophyte. Its protection against ion toxicity is mediated by the release of excess toxic ions via salt-removing glands, while mineral nutrition is assisted by the mycorrhiza. Summary: The quality of processed products can be adversely affected by uncontrollable batches of mangoes, which exhibit heterogeneous characteristics. This study aimed to establish predictive models for sugar and organic acid contents (dependent variables) in processed products using the initial compositions of fresh mangoes. Three mango cultivars (cv. 'Kent', cv. 'Keo Romeat', and cv. 'Keo Chen') were classified as low‐density and high‐density groups. Each group of mangoes at the green‐mature, mid‐ripe, and ripe stages was processed into pasteurised purees, dried slices, and mango chips. Prediction models were established using a mix of simple linear regression (SLR) based on the initial content and Analysis of Variance (ANOVA) to identify the impact of qualitative variables (ripening stage, cultivar‐density, and processing technique). In processed mangoes, 13% sucrose content was estimated to accumulate with the three qualitative variables, whereas glucose and fructose contents decreased from their initial levels by 10% and 7%, respectively. Processing techniques can predict the ratio of sugars/acids (S/A) in processed products, regardless of the ripening stage or cultivar‐density. Similar to S/A, citric acid and malic acid contents in mango products were significantly increased by processing techniques. The initial content and processes were insufficient to predict the final contents of the same parameters in processed mangoes; therefore, some models need to include the effects of ripening stage and cultivar‐density to improve the prediction. These relevant explanatory variables contributed significantly to the development of the models, resulting the accuracy of predictive models with normalised root mean square errors (NRMSEs) lower than 10%, except for malic acid (14.04%). In conclusion, it is feasible to estimate the sugar and acidity levels in processed mangoes, offering promising possibilities for ensuring consistent quality of mango‐based products. The hippocampus (HC) is recognized for its pivotal role in memory‐related plasticity and facilitating adaptive behavioral responses to reward shifts. However, the nature of its involvement in the response to reward downshifts remains to be determined. To bridge this knowledge gap, we explored the HC's function through a series of experiments in various tasks involving reward downshifts and using several neural manipulations in rats. In Experiment 1, complete excitotoxic lesions of the HC impaired choice performance in a modified T‐maze after reducing the quantity of sugar pellet rewards. In Experiment 2, chemogenetic inhibition of the dorsal HC (dHC) disrupted anticipatory behavior following a food‐pellet reward reduction. Experiments 3–5 impaired HC function by using peripheral lipopolysaccharide (LPS) administration. This treatment, which induces peripheral inflammation affecting HC function, significantly increased cytokine levels in the dHC (Experiment 3) and impaired anticipatory choice behavior (Experiment 4). None of these dorsal hippocampal manipulations affected consummatory responses in animals experiencing sucrose downshifts. Accordingly, we found no evidence of increased neural activation in either the dorsal or ventral HC, as measured by c‐Fos expression, after a sucrose downshift task involving consummatory suppression (Experiment 6). The results highlight the HC's pivotal role in adaptively modulating anticipatory behavior in response to a variety of situations involving frustrative nonreward, while having no effect on adjustments on consummatory behavior. The data supporting this conclusion were obtained under heterogeneous experimental conditions derived from a multi‐laboratory collaboration, ensuring the robustness and high reproducibility of our findings. Spatial orientation, memory update, choice of reward signals of different values, and anticipatory versus consummatory adjustments to reward downshift are discussed as potential mechanisms that could account for the specific effects observed from HC manipulations. A previous study found that mass transfer occurring during the osmotic dehydration (OD) process. This phenomena is possible causing modification in physicochemical properties; therefore, understanding the OD effect on quality attributes is important. This research aimed to study the impact of sucrose concentration and immersion time on the kinetics of mass transfer and the physicochemical properties of avocado (Persea americana var. Miki) during the OD process. The samples were immersed in a sucrose solution (45, 55 and 65 ° Brix) for up to 300 min, then the mass transfer parameter and physicochemical properties were evaluated. The results showed that the highest rate of water removal (WR) was -0.037 g.g-1.h-1 using the 55 ° Brix solution, which also led to an increase in the soluble solid gain (SSG) value to 0.0126 g.g-1.h-1, while the highest WR/SSG ratio was obtained after 180 min of immersion using 65 ° Brix (2.451). A 2nd-order polynomial model accurately describes avocado mass transfer kinetics and their relationship to physicochemical properties. The model showed the best adjustment for WR, SSG, and weight loss (WL), as pointed out by R2 = 0.9671, 0.8968 and 0.9573, respectively. WR has a negative effect on firmness (R2 = 0.9278), vitamin C (R2 = 0.9349), and total fat (R2 = 0.8954). The uptake of sucrose correlated with decrease of vitamin C (R2 = 0.9391), whereas WL also affected the loss of total fat (R2 = 0.8288). The OD process modified the chroma of the avocado by reducing lightness, increasing greenness and yellowness, and yet protecting the flesh from the browning effect. Overall, this study provides insights into the relationship between mass transfer and physicochemical properties, and it is useful to predict the final condition of the osmo-avocado before subsequent processes. Purpose: The objective of this study is to investigate the impact of ferroptosis on depression and elucidate the molecular mechanism underlying melatonin's inhibitory effect on ferroptosis in the treatment of depression. Methods: In this study, a depression-like behavior model was induced in mice using LPS, and the effect of melatonin on depression-like behavior was evaluated through behavioral experiments (such as forced swimming test (FST) and sucrose preference test (SPT)). Additionally, molecular biological techniques (including real-time fluorescence quantitative PCR, Western blotting, immunoprecipitation) were employed to detect the expression levels and interactions of METTL3, SIRT6 and ferroptosis-related genes in mouse brain tissue. Furthermore, both in vitro and in vivo experiments were conducted to verify the regulatory effect of melatonin on Nrf2/HO-1 pathway and explore its potential molecular mechanism for regulating ferroptosis. Results: Melatonin was found to significantly ameliorate depression-like behavior in mice, as evidenced by reduced immobility time in the forced swimming test and increased sucrose intake in the sucrose preference test. Subsequent investigations revealed that melatonin modulated SIRT6 stability through METTL3-mediated ubiquitination of SIRT6, leading to its degradation. As a deacetylase, SIRT6 plays a pivotal role in cellular metabolism regulation and antioxidative stress response. This study elucidated potential signaling pathways involving Nrf2/HO-1 through which SIRT6 may exert its effects. Hypothalamic inflammation underlies diet-induced obesity and diabetes in rodent models. While diet normalization largely allows for recovery from metabolic impairment, it remains unknown whether long-term hypothalamic inflammation induced by obesogenic diets is a reversible process. In this study, we aimed at determining sex specificity of hypothalamic neuroinflammation and gliosis in mice fed a fat- and sugar-rich diet, and their reversibility upon diet normalization. Mice were fed a 60%-fat diet complemented by a 20% sucrose drink (HFHSD) for 3 days or 24 weeks, followed by a third group that had their diet normalized for the last 8 weeks of the study (reverse diet group, RevD). We determined the expression of pro- and anti-inflammatory cytokines, and of the inflammatory cell markers IBA1, CD68, GFAP and EMR1 in the hypothalamus, and analyzed morphology of microglia (IBA-1+ cells) and astrocytes (GFAP+ cells) in the arcuate nucleus. After 3 days of HFHSD feeding, male mice showed over-expression of IL-13, IL-18, IFN-γ, CD68 and EMR1 and reduced expression of IL-10, while females showed increased IL-6 and IBA1 and reduced IL-13, compared to controls. After 24 weeks of HFHSD exposure, male mice showed a general depression in the expression of cytokines, with prominent reduction of TNF-α, IL-6 and IL-13, but increased TGF-β, while female mice showed over-expression of IFN-γ and IL-18. Furthermore, both female and male mice showed some degree of gliosis after HFHSD feeding for 24 weeks. In mice of both sexes, diet normalization after prolonged HFHSD feeding resulted in partial neuroinflammation recovery in the hypothalamus, but gliosis was only recovered in females. In bee diets, nectar is the primary source of carbohydrates. During scarcity of bee flora, such as in winter, beekeepers typically provide supplemental sugar syrup as a nectar substitute. We examined four types of sugars (sucrose, fructose, glucose, and a mixture of fructose and glucose) on bee longevity, bee appetitive, and their impact on pollen digestion. The results showed that sucrose and glucose had extended the longevity in cages. Bees preferred sucrose at a 50% concentration since they consumed more than the other sugar types in cages and in the multiple-choice test experiment in apiary. Bees given sucrose solution consumed the most pollen. In contrast, the cages provided the fructose solution consumed the least amount of pollen, increasing body fat percentage, which can be considered evidence of a digestive disorder. The identification and characterization of the sucrose phosphate synthase (SPS) genes family have been carried out in several plant species; nevertheless, studies on the analysis of SPS genes in cassava are limited. We report the study of four SPS genes; (sucrose phosphate synthase 3 F, sucrose phosphate synthase 1F, sucrose phosphate synthase family protein and sucrose phosphate synthase 2 F) identified from transcriptome analysis (NCBI Bio project ID: PRJNA998707) of the total RNA isolated from storage roots of TME-419 and H-165 cassava varieties. The sequence analysis revealed that the expression of SPS genes was mainly between the third and sixth months of storage root development in the two cassava varieties. However, TME-419 variety recorded 3.4-fold higher in expression of SPS than H-165 variety. Variations observed in the level of expression of the SPS genes and the reliability of the selected differentially expressed genes was validated via reverse transcription-polymerase chain reaction. The chromosome position of the SPS genes were located on Chr 18, Chr 15, Chr 14 and Chr 16 for sucrose phosphate synthase family protein, Sucrose phosphate synthase 3 F, Sucrose phosphate synthase 1F and Sucrose phosphate synthase 2 F respectively. The correlation analysis showed that the expression of SPS genes is positively correlated with the level of sucrose and starch contents in the cassava storage roots. This implies that high expression of SPS genes in cassava storage roots has the ability to expedite the rate of starch accumulation, which is important for the cassava industry, food security, and breeding of elite cassava and related starch-producing crops. The rubber tree Hevea brasiliensis (Willd. Ex Adr. De Juss.) Müell Arg.] is an important source of latex for the production natural rubber. Natural rubber is an important biopolymer used in various industries, but aspects related to hormonal regulation in biosynthesis are still unknown, which would allow optimizing its production. We review the molecular and physiological mechanisms of increases latex regeneration and flow by the stimulation of rubber trees with exogenous applications of ethylene and jasmonate. We found that the increase in latex regeneration by ethylene is due to the increase in gene level expression and enzymatic activity of key photosynthesis and glycolysis enzymes for the generation of precursors in the first phase of rubber biosynthesis. Latex flow is supported by up-regulated genes in sucrose metabolism such as invertases, induction of sucrose transporters (SUT), and aquaporins (PIP) to maintain flow and turgor pressure in laticifers. Meanwhile, the increase in latex yield mediated by jasmonate may be due to the induction of laticifer differentiation in the long term and in the short term be mediated by the induction of small rubber particles (SRPP) as non-enzymatic cofactors in the production of latex. This information contributes to the knowledge of latex biosynthesis, which allows for a greater support for the exogenous application of jasmonates and ethylene to regulate its production. Sucrose is a commonly utilized nutritive sweetener in food and beverages due to its abundance in nature and low production costs. However, excessive intake of sucrose increases the risk of metabolic disorders, including diabetes and obesity. Therefore, there is a growing demand for the development of nonnutritive sweeteners with almost no calories. d‐Allulose is an ultra‐low‐calorie, rare six‐carbon monosaccharide with high sweetness, making it an ideal alternative to sucrose. In this study, we developed a cell factory for d‐allulose production from sucrose using Escherichia coli JM109 (DE3) as a chassis host. The genes cscA, cscB, cscK, alsE, and a6PP were co‐expressed for the construction of the synthesis pathway. Then, the introduction of ptsG‐F and knockout of ptsG, fruA, ptsI, and ptsH to reprogram sugar transport pathways resulted in an improvement in substrate utilization. Next, the carbon fluxes of the Embden‐Meyerhof‐Parnas and the pentose phosphate pathways were regulated by the inactivation of pfkA and zwf, achieving an increase in d‐allulose titer and yield of 154.2% and 161.1%, respectively. ABSTRACT Fruit taste quality is greatly influenced by the content of soluble sugars, which are predominantly stored in the vacuolar lumen. However, the accumulation and regulation mechanisms of sugars in most fruits remain unclear. Recently, we established the citrus fruit vacuole proteome and discovered the major transporters localized in the vacuole membrane. Here, we demonstrated that the expression of tonoplast sugar transporter 2 (CsTST2) is closely associated with sugar accumulation during sweet orange (Citrus sinensis) ripening. It was further demonstrated that CsTST2 had the function of transporting hexose and sucrose into the vacuole. Overexpression of CsTST2 resulted in an elevation of sugar content in citrus juice sac, calli, and tomato fruit, whereas the downregulation of its expression led to the reduction in sugar levels. CsTST2 was identified as interacting with CsCIPK23, which binds to the upstream calcium signal sensor protein CsCBL1. The phosphorylation of the three serine residues (Ser277, Ser337, and Ser354) in the loop region of CsTST2 by CsCIPK23 is crucial for maintaining the sugar transport activity of CsTST2. Additionally, the expression of CsCIPK23 is positively correlated with sugar content. Genetic evidence further confirmed that calcium and CsCIPK23‐mediated increase in sugar accumulation depends on CsTST2 and its phosphorylation level. These findings not only unveil the functional mechanism of CsTST2 in sugar accumulation, but also explore a vital calcium signal regulation module of CsCBL1/CIPK23 for citrus sweetness quality. A series of sulfonated carbon acid catalysts with strong acidity was prepared by simultaneous carbonization and sulfonation of biomass sucrose in the presence of the organic sulfonating agent sulfosalicylic acid under hydrothermal conditions at temperatures ranging from 150 to 200 °C. It was found from FTIR and XPS spectra that the surface of carbon was efficiently functionalized with –SO3H groups. Research on the mechanism of the sulfonation process indicated that the intermediate 5-hydroxymethyl furfural (5-HMF), which was easily hydrolyzed from sucrose, was prone to carbonization and functionalized with –SO3H groups simultaneously. Compared with 5-HMF and fructose used as the initial carbon precursor, the slow hydrolysis of sucrose to intermediate 5-HMF to suppress its rapid carbonization is favorable for the efficient grafting of –SO3H groups when sucrose is used as the initial carbon precursor. The prepared sulfonated carbons were evaluated as acid catalysts in a typical ester hydrolysis reaction, namely, hydrolysis of ethyl acetate. The sulfonic acid groups were identified to be the active sites and quantified by a cation-exchange process. The activity of the sulfonated carbon was primarily correlated with the total number of active sites. The absorption of the essential element nitrogen by plants affects various aspects of plant physiological activity, including gene expression, metabolite content and growth. However, the molecular mechanism underlying the potato tuberization response to nitrogen remains unclear. Potato plants were subjected to pot experiments under nitrogen deficiency, normal nitrogen levels and nitrogen sufficiency. A comprehensive analysis of the physiological responses, transcriptomic profiles, and metabolic pathways of potato stolons subjected to nitrogen stress was conducted. Transcriptomic analysis revealed 2756 differentially expressed genes (DEGs) associated with nitrogen stress. Metabolomic analysis identified a total of 600 differentially accumulated metabolites (DAMs). Further correlation analysis of the major DEGs and DAMs revealed that 9 key DEGs were associated with alpha-linolenic acid metabolism, 16 key DEGs with starch and sucrose metabolism, 7 key DEGs with nitrogen metabolism, and 16 key DEGs with ABC transporters. Nitrogen deficiency significantly increased the sucrose, GDP-glucose and L-glutamic acid levels and promoted stolon growth by increasing the expression of AMY (alpha-amylase), BE (1,4-alpha-glucan branching enzyme), SS (starch synthase), SPS (sucrose‒phosphate synthase) and AGPS (glucose‒1-phosphate adenylyltransferase). However, high nitrogen levels had the opposite effect. In addition, high nitrogen levels upregulated EG (endoglucanase), SUS (sucrose synthase) and GDH (glutamate dehydrogenase) and led to significant accumulation of 9-Hydroperoxy-10,12,15-octadecatrienoate (9(S)-HpOTrE), (13 S)-Hydroperoxyoctadeca-9,11,15-trienoate (13 (S)-HpOTrE) and L-glutamine, ultimately affecting the balance between plant growth and defense. Overall, our comprehensive study revealed the co-expressed genes and potential pathways related to potato tuber formation under different nitrogen conditions. The conformational ensemble and function of intrinsically disordered proteins (IDPs) are sensitive to their solution environment. The inherent malleability of disordered proteins, combined with the exposure of their residues, accounts for this sensitivity. One context in which IDPs play important roles that are concomitant with massive changes to the intracellular environment is during desiccation (extreme drying). The ability of organisms to survive desiccation has long been linked to the accumulation of high levels of cosolutes such as trehalose or sucrose as well as the enrichment of IDPs, such as late embryogenesis abundant (LEA) proteins or cytoplasmic abundant heat- soluble (CAHS) proteins. Despite knowing that IDPs play important roles and are co- enriched alongside endogenous, species- specific cosolutes during desiccation, little is known mechanistically about how IDP- cosolute interactions influence desiccation tolerance. Here, we test the notion that the protective function of desiccation- related IDPs is enhanced through conformational changes induced by endogenous cosolutes. We find that desiccation- related IDPs derived from four different organisms spanning two LEA protein families and the CAHS protein family synergize best with endogenous cosolutes during drying to promote desiccation protection. Yet the structural parameters of protective IDPs do not correlate with synergy for either CAHS or LEA proteins. We further demonstrate that for CAHS, but not LEA proteins, synergy is related to self- assembly and the formation of a gel. Our results suggest that functional synergy between IDPs and endogenous cosolutes is a convergent desiccation protection strategy seen among different IDP families and organisms, yet the mechanisms underlying this synergy differ between IDP families. The profound contamination of reservoirs triggered by industrial dyes and the growing phenomenon of antibiotic-resistant bacteria present substantial risks to both the environment and human health. Conventional approaches to water treatment and microbiological disinfection can frequently become ineffective, expensive, and detrimental to the environment, and this issue underscores the need for novel materials that possess refined photocatalytic and antibacterial characteristics that thrive efficiently in a setting of visible light. An innovative nanostructured nanocomposite, Mo-CeO 2 @C, was synthesized using a green chemistry method coupled with Mo-CeO 2 nanoparticles and pristine CeO 2. The impact of Ce, Mo, and carbon-based nanocomposites has been validated by leveraging XRD, FTIR, UV–vis spectra, and FE-SEM/EDX. The sunlight-mediated photodegradation performance of carbon-based nanocomposite exhibited a superior photodegradation efficiency of 99.5 %, higher than others. The photocatalytic activity of M.B. dye concentration, catalyst dosage, scavenger test, and recyclability were also conducted, which confirm the optimal catalyst loading of 20 mg, dye concentration 40 ppm, and pH 8 with reusability up to sixth cycles. The carbon-coated doped nanocomposite displays superior antibacterial efficacy against gram-positive S. aureus , gram-negative E. coli , and P. vulgaris bacteria in comparison to CeO 2 and Mo-doped CeO 2. This may be attributed to the combined effects of the generation of reactive oxygen species (R.O.S.) and the intrinsic characteristics of the dopant and carbon matrix. The results of our research emphasize the potential of carbon-based nanoparticles as versatile agents in the realms of environmental rehabilitation and biomedical research. ABSTRACT Blueberry flower buds cultivated in greenhouses develop during both autumn and spring, with floral induction being a critical process for flowering, influenced by environmental factors. This study aimed to clarify the regulatory mechanisms governing floral induction in greenhouse blueberries, focusing on the similarities and differences in flower bud differentiation between the spring and autumn seasons. Understanding these mechanisms is pivotal for enhancing blueberry production. In this study, we analysed the phenotypic characteristics associated with flower bud differentiation and observed that short photoperiods markedly affect the induction process. Transcriptomic analyses revealed distinct major metabolic pathways activated in autumn compared to spring. Seasonal variations in carbohydrate metabolism were also noted, with sucrose hydrolysis being prominent in autumn and sucrose synthesis prevailing in spring. The interplay between circadian rhythms and photosynthesis appeared to facilitate the allocation of sugars for bud development. Subsequent investigations underscored the sensitivity of VcCO3 to variations in photoperiod. Predominantly localised in the nucleus, VcCO3 facilitated floral induction in response to short photoperiods by activating the expression of downstream genes, including VcFT, VcLFY, VcAP3, and VcSOC1. Eremosparton songoricum (Litv.) Vass. is a desert legume exhibiting extreme drought tolerance and the ability to withstand various harsh environments, making it a good candidate for investigating stress tolerance mechanisms and exploring valuable stress-resistant genes. However, the absence of a genetic transformation system for E. songoricum poses significant limitations for functionally validating these stress-resistant genes in situ. In this study, we developed an Agrobacterium-mediated transient transformation system for E. songoricum utilizing the β-glucuronidase (GUS) gene as a reporter. We investigated three types of explants (seedlings, assimilated branches and callus) and the effects of different Agrobacterium strains, seedling ages, OD600 values, acetosyringone (AS) concentrations, sucrose concentrations and infection times on the transformation efficiency. The results reveal that the optimal transformation system was infecting one-month-old regenerating assimilated branches with the Agrobacterium strain C58C1. The infection solution comprised 1/2 MS medium with 3% sucrose and 200 μM AS at an OD600 of 0.8, infection for 3 h and then followed by 2 days of dark cultivation, which achieving a maximum transformation rate of 97%. The maximum transformation rates of the seedlings and calluses were 57.17% and 39.51%, respectively. Moreover, we successfully utilized the assimilated branch transient transformation system to confirm the role of the previously reported transcription factor EsDREB2B in E. songoricum. The overexpression of EsDREB2B enhanced drought tolerance by increasing the plant's reactive oxygen species (ROS) scavenging capacity in situ. This study established the first transient transformation system for a desert legume woody plant, E. songoricum. This efficient system can be readily applied to investigate gene functions in E. songoricum. It will expedite the exploration of genetic resources and stress tolerance mechanisms in this species, offering valuable insights and serving as a reference for the transformation of other desert plants and woody legumes. Background: 2-O-α-D-glucosyl glycerol (2-αGG) is a valuable ingredient in cosmetics, health-care and food fields. Sucrose phosphorylase (SPase) is a favorable choice for biosynthesis of 2-αGG, while its glucosyl-acceptor affinity and thermodynamic feature remain largely unknown, limiting 2-αGG manufacturing. Results: Here, three SPases were obtained from lactobacilli and bifidobacteria, and the one encoded by Lb. reuteri SDMCC050455 (LrSP) had the best transglucosylation ability, with 2-αGG accounting for 86.01% in the total product. However, the LrSP exhibited an initial forward reaction rate of 11.83/s and reached equilibrium of 56.90% at 110 h, indicating low glycerol affinity and conversion rate. To improve catalytic efficiency, the LrSP was overexpressed in Lb. paracasei BL-SP, of which the intracellular SPase activity increased by 6.67-fold compared with Lb. reuteri SDMCC050455. After chemically permeabilized with Triton X-100, the whole-cell biocatalysis of Lb. paracasei BL-SP was prepared and showed the highest activity, with the initial forward reaction rate improved to 50.17/s and conversion rate risen to 80.79% within 17 h. Using the whole-cell biocatalyst, the final yield of 2-αGG was 203.21 g/L from 1 M sucrose and 1 M glycerol. Conclusion: The food grade strain Lb. paracasei was used for the first time as cell factory to recombinantly express the LrSP and construct a whole-cell biocatalyst for the production of 2-αGG. After condition optimization and cell permeabilization, the whole-cell biocatalyst exhibited 23.89% higher equilibrium conversion and 9.10-fold of productivity compared with the pure enzyme catalytic system. [Display omitted] Aqueous solutions of long-chain water-soluble sucrose ester surfactants exhibit non-trivial response to temperature variations, revealing a peak in viscosity around 40–50 °C. While previous investigations have explored the structures within sucrose stearate systems at various constant temperatures, a comprehensive understanding of the entire temperature dependence and the underlying molecular factors, contributing to this phenomenon is currently missing. Temperature dependent properties and supramolecular structures formed in aqueous solutions of commercial sucrose palmitate were examined using SAXS/WAXS, DSC, optical microscopy, rheological measurements, NMR, and cryo-TEM. The underlying mechanism governing this unusual behavior is revealed and is shown to relate to the mono- to di-esters ratio in the solutions. Solutions primarily containing sucrose monoesters (monoesters molecules ≳ 98% of all surfactant molecules) exhibit behavior typical of nonionic surfactants, with minimal changes with temperature. In contrast, the coexistence of mono- and di-esters results in the formation of discrete monodisperse diester particles and a network of partially fused diester particles at low temperature. As the temperature approaches the diesters' melting point, wormlike mixed micelles form, causing a viscosity peak. The height of this peak increases significantly with the diester concentration. Further temperature increase leads to fluidization of surfactant tails and formation of branched micelles, while excess diester molecules phase separate into distinct droplets. Binge eating disorder is the most prevalent eating disorder, affecting both sexes but more commonly found in women. Given the frequent co-occurrence of psychiatric disorders, this study aimed to establish a standardized experimental intermittent protocol to investigate overeating associated with depression. A 10-day protocol induced uncontrolled eating behavior in C57BL/6J female mice. The first experiment included the following groups: naive group (chow ad libitum), control group (chow and sucrose solution ad libitum), and fasting groups (16 and 20 h) exposed to an intermittent sucrose solution (10 %) and chow regimen. Subsequently, the feeding test, open field test, elevated plus maze test, tail suspension test, and light/dark conflict test were conducted. Furthermore, monoamine oxidase (MAO) A and B activities in brain structures and plasma corticosterone levels were assessed. Food overconsumption and depressive-like behavior were observed in both sucrose fasting groups, while risk-taking behaviors were specifically observed in the 20-hour fasting sucrose group. While both fasting sucrose groups caused reduced hippocampal MAO-A activity, only the F20 sucrose group inhibited MAO-B in the cortex and hypothalamus. Moreover, both fasting sucrose groups exhibited elevated corticosterone levels. In a separate design (Experiment 2), groups with 16 and 20 h of fasting alone (without sucrose) did not show the same behavioral results as the intermittent fasting sucrose groups, thus avoiding fasting bias. Based on these results, the 20-hour sucrose fasting group was chosen as the ideal protocol for mimicking overeating behavior associated with depression to investigate future therapeutic approaches for this comorbidity. Summary How carbon (sucrose) and nitrogen (amino acid) accumulation is coordinatively controlled in cereal grains remains largely enigmatic. We found that overexpression of the strigolactone (SL) biosynthesis gene CAROTENOID CLEAVAGE DIOXYGENASE 8 (CCD8) resulted in greater ear diameter and enhanced sucrose and amino acid accumulation in maize kernels. Loss of ZmCCD8 function reduced kernel growth with lower sugar and amino acid concentrations. Transcriptomic analysis showed down‐regulation of the transcription factors ZmMYB42 and ZmMYB63 in ZmCCD8 overexpression alleles and up‐regulation in zmccd8 null alleles. Importantly, ZmMYB42 and ZmMYB63 were negatively regulated by the SL signalling component UNBRANCHED 3, and repressed expression of the sucrose transporters ZmSWEET10 and ZmSWEET13c and the lysine/histidine transporter ZmLHT14. Consequently, null alleles of ZmMYB42 or ZmMYB63 promoted accumulation of soluble sugars and free amino acids in maize kernels, whereas ZmLHT14 overexpression enhanced amino acid accumulation in kernels. Moreover, overexpression of the SL receptor DWARF 14B resulted in more sucrose and amino acid accumulation in kernels, down‐regulation of ZmMYB42 and ZmMYB63 expression, and up‐regulation of ZmSWEETs and ZmLHT14 transcription. Together, we uncover a distinct SL signalling pathway that regulates sucrose and amino acid accumulation in kernels. Significant association of two SNPs in the 5′ upstream region of ZmCCD8 with ear and cob diameter implicates its potential in breeding toward higher yield and nitrogen efficiency. SUMMARY Cotton seed development and fiber elongation are the inseparable and overlapped development processes requiring the continuous supply of sucrose as the direct carbon source. However, little is known about the molecular mechanism of how sucrose is transported from the source tissues (leaves) into growing cotton seeds. Here, we identify the function of a sucrose transporter gene, Sugars Will Eventually be Exported Transporter 10, GhSWEET10 in cotton seed development. GhSWEET10 encodes a functional sucrose transporter, predominantly expressing in the funiculus, inner seedcoat, and endosperm during fiber elongation. GhSWEET10 RNAi plants (GhSWEET10i) accumulated less sucrose and glucose in growing seeds and that led to shorter fibers and smaller seeds, whereas GhSWEET10 overexpressed plants (GhSWEET10OE) had bigger seeds and longer fibers with more sugar accumulation during fiber elongation. GhSWEET10 gene is transcriptionally controlled by the transcription factor GhDOFD45. GhDOFD45 knockout plants (GhDOFD45‐KO) possessed the phenotypes of smaller seeds and shorter fibers like those of GhSWEET10i plants. Furthermore, GhSWEET10 mainly exports the sucrose from the funiculus into developing seeds according to the mimic‐analysis of sucrose transporting. Collectively, all these findings show that GhDOFD45 positively regulates GhSWEET10 expression to mainly transport sucrose from leaves into developing cotton seeds. Our findings also imply that the sucrose transport into enlarging seeds benefits fiber development, and thus GhSWEET10 can be selected as a target of breeding novel cotton varieties with larger and more vigorous seeds. Introduction: Carbohydrate compounds serve multifaceted roles, from energy sources to stress protectants, found across diverse organisms including bacteria, fungi, and plants. Despite this broad importance, the molecular genetic framework underlying carbohydrate biosynthesis pathways, such as starch, sucrose, and glycolysis/gluconeogenesis in Salvia guaranitica , remains largely unexplored. Methods: In this study, the Illumina-HiSeq 2500 platform was used to sequence the transcripts of S. guaranitica leaves, generating approximately 8.2 Gb of raw data. After filtering and removing adapter sequences, 38 million reads comprising 210 million high-quality nucleotide bases were obtained. De novo assembly resulted in 75,100 unigenes, which were annotated to establish a comprehensive database for investigating starch, sucrose, and glycolysis biosynthesis. Functional analyses of glucose-6-phosphate isomerase (SgGPI), trehalose-6-phosphate synthase/phosphatase (SgT6PS), and sucrose synthase (SgSUS) were performed using transgenic Arabidopsis thaliana. Results: Among the unigenes, 410 were identified as putatively involved in these metabolic pathways, including 175 related to glycolysis/gluconeogenesis and 235 to starch and sucrose biosynthesis. Overexpression of SgGPI , SgT6PS , and SgSUS in transgenic A. thaliana enhanced leaf area, accelerated flower formation, and promoted overall growth compared to wild-type plants. Discussion: These findings lay a foundation for understanding the roles of starch, sucrose, and glycolysis biosynthesis genes in S. guaranitica , offering insights into future metabolic engineering strategies for enhancing the production of valuable carbohydrate compounds in S. guaranitica or other plants. This study was conducted in order to explore the pathogens that cause stem rot of fresh onions during postharvest storage, identify the incidence of stem rot, investigate the influence of pathogen infection on the active components of onion, and provide a theoretical basis for disease control during the postharvest storage of fresh onions. The pathogens were isolated and purified from the junction between the rotten and healthy tissues of onion stem rot that occurred naturally during storage at room temperature by tissue separation; then, the pathogens were identified by morphological and molecular biological techniques, the biological characteristics of the pathogens were analyzed, and finally, the influence of pathogen infection on the active ingredients of onion was studied. The results suggested that the main pathogens causing stem rot of fresh onions during postharvest storage were Talaromyces pinophilus, Trichoderma simmonsii, and Talaromyces minioluteus. The optimum colony growth conditions for T. pinophilus were as follows: a temperature of 30 °C, a pH of 7, light for 24 h, maltose as a carbon source, and peptone as a nitrogen source; the lethal temperature was 65 °C for 15 min. For T. simmonsii, the lethal temperature was 60 °C for 15 min, and the optimum sporulation conditions were a temperature of 25 °C, a pH of 5–7, light for 24 h, a carbon source of sucrose, and a nitrogen source of yeast powder. For T. minioluteus, the lethal condition was 65 °C for 15 min; the optimum colony growth conditions were a temperature of 25 °C, a pH of 8–9, 24 h of darkness, a carbon source of maltose, and a nitrogen source of peptone. The relative content of sulfur compounds, as the active components of onions, was much lower in the infected onions than in the healthy onions due to infection by the pathogens T. pinophilus, T.simmonsii, and T.minioluteus. Background: The differing floret opening times between subsp. indica and subsp. japonica in rice limit the potential for increased hybrid seed production. Objectives: To elucidate the physiological basis underlying the differences in floret opening time between indica and japonica rice. Materials: A comparative analysis involved nine indica and ten japonica rice varieties. Methods: Using paraffin sectioning, transcriptome sequencing, RT-PCR, and endogenous substance quantification, we investigated the structural variations in floral organs, the differences in the initiation timing of floret opening regulatory pathways, and endogenous regulators. Results: The results indicated insignificant differences in lemma thickness, lodicule thickness, lodicule area, and the coupling-lodicule length between indica and japonica rice. However, japonica rice exhibited larger lodicule-lemma gaps and more vascular bundles compared to indica rice. Within the 9:00 a.m. to 10:00 a.m. interval, the expression of OsAOS1 in α-linolenic acid metabolism and OsISA3 in starch and sucrose metabolism notably increased in indica rice, with no significant change in japonica rice. Additionally, the endogenous JA and α-amylase surged more significantly in indica rice than in japonica rice. The increase in soluble carbohydrate in indica rice is greater than in japonica rice, but the difference is not significant. Conclusions: These findings suggest that in the process of the floret opening, the α-linolenic acid metabolism and starch and sucrose metabolism are initiated earlier in indica rice, accompanied by a more pronounced elevation in endogenous JA and α-amylase. A field experiment was carried out at Vasantdada Sugar Research Institute (VSI), Manjari, Pune, India during 2019-20, 2020-21 and 2021-22 crop seasons (36 months) to evaluate the performance of different harvesting periods of advanced varieties on yield and quality of sugarcane (Saccharum officinarum L.) under agro-climatic condition of peninsular zone of Maharashtra. Three harvesting dates i.e. H1-10 month, H2-12 month and H3-14 month and 10 different varieties including five early maturing sugarcane varieties V1-MS10001, V2-CoM 9057, V3-Co 09004 with two zonal check namely V4-CoC 671 and V5-VSI434 and five midlate maturing varieties V6-VSI08005, V7-CoVSI18121, V8-VSI12003 with two zonal check namely V9-CoM 0265 and V10-Co 86032 were evaluated in the split plot design with three replications. The varieties differed in stalk diameter and weight, and millable stalk number. Improvements in internodes number, stalk diameter and weight were increased with harvest age. Varieties produced better juice quality in canes harvested at 14 months. On the basis of pooled results obtained from three plant cane, it can be concluded that, Maximum cane yield (140.15 t ha-1) and CCS yield (19.33 t ha-1) can be secured by harvesting the sugarcane at 14 months after planting. With respect to varieties CoM 0265 recorded significantly higher cane yield (156.36 t ha-1) as well as CCS yield (18.81 t ha-1) which was followed by varieties VSI 08005 and CoVSI 18121. Sucrose and glycerol have gained attention as additives for hydrogels, owing to their capacity to exert considerable influence over the physicochemical, mechanical, and biological characteristics of these materials. Herein, these effects on agarose hydrogels (AHs) are explored. A series of AHs are synthesized using sucrose (30% and 300% w/v) and glycerol as additives. The storage modulus (10.0–13.7 kPa) and hydrophilicity of the hydrogels (contact angle < 50°) do not vary significantly with sucrose or glycerol addition. However, sucrose enhances the hydration capacity of the hydrogels by up to 170%, whereas glycerol reduces it. Interestingly, sucrose and glycerol individually do not have bacteriostatic effects against Staphylococcus epidermidis, but their combination significantly (p ≤ 0.001) inhibits the growth of both S. epidermidis and Pseudomonas aeruginosa by 63% and 29%, respectively, in comparison to native agarose. Cytotoxicity testing on NIH/3T3 murine fibroblasts reveals that sucrose increases cell viability up to 98%, while glycerol reduces it below 60%. Overall, these hydrogels hold promise for antibacterial biomedical applications as wound dressing materials and surface coatings for medical devices and can also be used to formulate bioinks for 3D bioprinting. Background and Objectives: Blood establishments strive to ensure the safety and comfort of blood donors while minimizing adverse events. This review aims to assess the efficacy and effectiveness of eating and/or drinking interventions before, during and/or after blood donation in reducing vasovagal reactions (VVRs). Materials and Methods: We analysed randomized and non‐randomized controlled trials comparing eating and/or drinking interventions to no intervention, placebo or usual practice on (pre‐)syncopal VVRs and related symptoms. The GRADE (Grading of Recommendations, Assessment, Development and Evaluation) approach was used to assess the risk of bias and overall certainty of the evidence. Results: Pre‐donation water ingestion likely results in reduced on‐site VVRs, compared to no water (2 fewer per 100 donors, moderate‐certainty evidence). A pre‐donation isotonic drink likely results in reduced VVRs, compared to usual practice (2 fewer per 100 donors, moderate‐certainty evidence). Pre‐donation salt‐loaded sweetened lemon water may result in fewer off‐site VVRs, compared to sweetened lemon water only (1 fewer per 100 donors, low‐certainty evidence). Pre‐donation water and a gel cap containing sucrose with 250 mg caffeine may result in fewer blood donor reaction ratings, compared to pre‐donation water only (low‐certainty evidence). Conclusions: Pre‐donation plain water ingestion or isotonic drink probably results in a large reduction in on‐site and off‐site VVRs. Pre‐donation water ingestion with caffeine consumption or salt supplementation may result in a VVR reduction, compared to water ingestion only. Corn starch‐based direct expanded products incorporated with 2% and 10% (w/w) sugar (fructose, glucose, sucrose, and xylose) were produced using a 20 mm co‐rotating twin‐screw extruder. The pasting and thermal properties of raw corn starch‐sugar mixes were analyzed before extrusion processing. The independent variables for extrusion processing included two sugar inclusion levels (2% and 10% w/w) and two screw speeds (150 and 250 rpm). The extrudates were characterized by their initial expansion ratio (IER), expansion ratio (ER), and shrinkage. ER values were high for fructose at 2% and 150 rpm and 10% glucose and sucrose extrudates at 250 rpm. The extrudates with 2% sucrose inclusion shrunk significantly higher than the control. Fourier transform infrared (FTIR) spectroscopy of the extruded blends did not indicate the presence of any new covalent bond formed between starch and sugar post‐extrusion. The interactions between sugar concentration and screw speed significantly influenced extrudate expansion characteristics. Due to their thermal and plasticizing properties, sugar inclusion (glucose, fructose, sucrose, and xylose) enhanced the extrudate expansion by altering their melt viscosity. Practical Application: The findings of this study can improve the expansion characteristics of high‐fiber‐based extruded snacks. Ingredients high in fiber generally hinder the starch transformation during extrusion and negatively impact the expansion properties. The presence of sugar at low concentrations can improve melt properties during extrusion processing and, in turn, significantly improve the textural properties of snacks. [ABSTRACT FROM AUTHOR]

Sweetened condensed milk (SCM) is a product widely used by both consumers and other food production branches. However, it contains a lot of sucrose. This study aimed to examine the effects provided by sugar substitutes—trehalose, isomaltulose, and polydextrose—upon the SCM sensory profile and valuable quality indicators, such as water activity (Aw), viscosity, acidity, crystal size, and Maillard reaction potential (browning index, color change, loss of free AA). The study was performed by making model systems of SCM (MSCM) using the method of reconstitution of powdered ingredients. All the presented carbohydrate compositions in MSCM provided Aw value typical of intermediate-moisture food, which contributes to the long-term shelf life of the product. However, only 2 MSCM showed stability of Aw within a 14-d storage period: a composition of isomaltulose and trehalose (each at 22.55%), and a composition of trehalose (28.19%), sucrose (5.64%), polydextrose (5.64%), and isomaltulose (5.64%). Trehalose and polydextrose in MSCM with monocarbohydrate added fraction demonstrated their high structure-forming ability, expressed in high values of dynamic viscosity (>30 Pa·s). The MSCM containing a predominant amount of trehalose in the carbohydrate compositions (≥50%) showed lower average crystal size (<16 μm) compared with MSCM with di- and tetra-carbohydrate added fractions with predominant amounts of isomaltulose or polydextrose. Isomaltulose and polydextrose added to MSCM led to pronounced browning, whereas trehalose and sucrose reduced this effect in MSCM with di- and tetra-carbohydrate added fractions. Polydextrose added at 22.55% to carbohydrate fraction of MSCM caused bitterness, whereas the addition at 5.64% did not affect the taste. Based on the results of all the research conducted, the optimal carbohydrate compositions to produce SCM with fewer calories, lower sucrose content, and stable, adequate values of processing and sensory properties were trehalose (22.55%) with isomaltulose (22.55%), and trehalose (28.19%) with sucrose (5.64%), polydextrose (5.64%), and isomaltulose (5.64%). Objectives: Binge eating disorder (BED) is the most prevalent eating disorder associated with multiple adverse health effects, especially mental health issues, including substance use disorders and mood and anxiety disorders. Given these high comorbidities, the objective of our study was to examine whether bingeing behavior would lead to altered perception of reinforcing properties of EtOH and changes in well-being. Methods: We used a sucrose bingeing model based on an intermittent access paradigm with a two-bottle choice, without fasting, in male and female mice. We examined the effect of 2-week sucrose paradigm on ethanol-reinforcing properties using a conditioned place preference test (CPP). Well-being, anxiety- and depressive-like behavioral tests were performed to assess emotional state following 2 and 8-week sucrose bingeing paradigm. Results: Mice with intermittent access to sucrose developed a binge-like behavior assessed by higher sucrose intake and escalation rate during the 1st hour of access, in comparison with mice with a continuous sucrose access. We show for the first time that sucrose bingeing in mice modifies positive reinforcing effect of EtOH in a CPP paradigm without marked alteration of emotional state. Interestingly, prolonging sucrose access for 8 weeks revealed an exacerbated bingeing behavior in female mice, and some signs of emotional state alterations in female with continuous access. Discussion: In sum, our findings broaden the understanding of behavioral alterations associated with bingeing, highlighting the need to investigate addictive-like properties of palatable food both in male and female mice. The present study evaluated the potential of a saccharide mixture (galactose, glucose, fructose, and sucrose in a 2:1:1:1 ratio) as a chemoattractant (attraction) or/and feeding stimulant (ingestion) for the purple mud crab Scylla tranquebarica. In Experiment I, the S. tranquebarica appetitive response toward the 3%, 5%, or 7% (w/v) saccharide mixture solutions was examined and evaluated by a scoring method. The 5% and 7% treatments yielded the highest mean score (2.05 out of 3.00), with 20% of the S. tranquebarica exhibiting the highest response order (probing movement) in the latter treatment. In Experiment II, the S. tranquebarica feeding response toward the pelleted feeds containing the saccharide mixture at 3%, 5%, or 7% (SAC3, SAC5, or SAC7) was examined and evaluated using a scoring method. No significant difference (P > 0.05) was found among the mean scores of SAC3, SAC5, and SAC7. However, the mean score of the SAC7 diet was significantly higher (P < 0.05) than that of the control feed (without saccharide mixture inclusion), with 25% of the S. tranquebarica ingesting the feed. Summary: Grain chalkiness is an undesirable trait that negatively regulates grain yield and quality in rice. However, the regulatory mechanism underlying chalkiness is complex and remains unclear. We identified a positive regulator of white‐belly rate (WBR). The WBR7 gene encodes sucrose synthase 3 (SUS3). A weak functional allele of WBR7 is beneficial in increasing grain yield and quality. During the domestication of indica rice, a functional G/A variation in the coding region of WBR7 resulted in an E541K amino acid substitution in the GT‐4 glycosyltransferase domain, leading to a significant decrease in decomposition activity of WBR7A (allele in cultivar Jin23B) compared with WBR7G (allele in cultivar Beilu130). The NIL(J23B) and knockout line NIL(BL130)KO exhibited lower WBR7 decomposition activity than that of NIL(BL130) and NIL(J23B)COM, resulting in less sucrose decomposition and metabolism in the conducting organs. This caused more sucrose transportation to the endosperm, enhancing the synthesis of storage components in the endosperm and leading to decreased WBR. More sucrose was also transported to the anthers, providing sufficient substrate and energy supply for pollen maturation and germination, ultimately leading to an increase rate of seed setting and increased grain yield. Our findings elucidate a mechanism for enhancing rice yield and quality by modulating sucrose metabolism and allocation, and provides a valuable allele for improved rice quality. Cherry tomatoes are nutritious food, often processed into long shelf-life preserved foods. However, there is increasing concern about the dangers of high sucrose in traditional preserved fruit. In this paper, the effect of enzyme-assisted osmosis of seven kinds of functional sugars, including psicose, tagatose, isomaltose, trehalose, galactooligosaccharides, isomaltooligosaccharides, and resistant dextrin on cherry tomato was studied. The dry matter content after enzyme treatment and sugar soaking treatment was evaluated. Sensory quality, physicochemical quality, antioxidant capacity, sugar composition, and flavor substances were also measured. The addition of enzymes and functional sugars made cherry tomatoes redder and softer, and increased the soluble solid content (1.45- to 1.67-fold), lycopene content (2.14- to 2.18-fold), total polyphenol content (1.28- to 1.41-fold), and DPPH radical scavenging rate (1.81- to 1.95-fold). Furthermore, the sugar composition and flavor substances were improved. Thus, it is a good choice of adding these functional sugars to produce cherry tomato products with probiotic potential, and this new method can provide a reference for the processing of other fruit products. Objective: Increased intake of sweets or sugar craving may occur in response to chronic stress representing a risk factor for development of eating disorders and obesity. However, no safe treatment of stress‐induced sugar craving is available. In this study we analysed effects of two Lactobacillus strains on food and sucrose intake in mice before and during their exposure to a chronic mild stress (CMS). Research Methods & Procedures: C57Bl6 mice were gavaged daily for 27 days with a mix of L. salivarius (LS) LS7892 and L. gasseri (LG) LG6410 strains or with 0.9% NaCl as a control. Following 10 days of gavage, mice were individually placed into the Modular Phenotypic cages, and after 7 days of acclimation were exposed to a CMS model for 10 days. Food, water and 2% sucrose intakes as well as meal pattern were monitored. Anxiety and depressive‐like behaviour were analysed by standard tests. Results: Exposure of mice to CMS was accompanied by increased size of sucrose intake in the control group likely reflecting the stress‐induced sugar craving. A consistent, about 20% lower total sucrose intake, was observed in the Lactobacilli‐treated group during stress which was mainly due to a reduced number of intakes. Lactobacilli treatment also modified the meal pattern before and during the CMS, showing a decrease of meal number and an increase of meal size with a tendency of reduced total daily food intake. Mild anti‐depressive behavioural effects of the Lactobacilli mix were also present. Conclusion: Supplementation of mice with LS LS7892 and LG LG6410 decreases sugar consumption suggesting a potential utility of these strains against stress‐induced sugar craving. Sourdough fermentation is rarely used for gluten-free flatbread (GFFB), a product that is challenging to produce, especially when using high-fiber ingredients that bring nutritional benefits but lead to physical deterioration. The aim of this study was therefore to evaluate the fermentation performance of carob flour (CSPF), proso millet flour (PMF), and proso millet bran (PMB) individually and in combination with Limosilactobacillus fermentum and Kluyveromyces marxianus (LF + KM) and to compare the performance of LF + KM with a commercial starter (LIVENDO® LV1). A mixture design (n = 13) was used to evaluate the fermentation performance of LF + KM (total titratable acidity (TTA); lactobacilli and yeast growth; acetic and lactic acid, fructose, glucose, and saccharose content) at 35 °C for 16 h. The comparison of LF + KM with LV1 fermentation was based on the acidity rate, fermentation quotient, TTA, and finally by determining the physical properties (texture, shape, color) of a rice–corn GFFB in which 10% of flour was supplemented with the sourdoughs. PMB promoted the growth of lactobacilli and the production of organic acids, especially in combination of CSPF and PMF. The optimum flour ratio was 2.4:1:1.2 (PMB:PMF:CSPF). LF + KM shortened the sourdough fermentation time by 2.5 times compared to LV1. The use of LF + KM sourdough reduced the hardness (32%) and chewiness (28%) of the GFFB, while the volume (35%) was increased compared to LV1 sourdough. Background/Objectives: Sweet taste preferences are currently targeted to aid with reducing free sugar intakes, but associations between sweet taste liking, sweet food intakes, and sugar intakes are not well established. Methods: UK consumers (n = 179) who were consuming >5% of total energy intakes from free sugars provided several laboratory measures of sweet taste liking, laboratory test meal measures of sweet food choice and sugar intakes, and 3-day food diary measures of free-living free sugar and total sugar intakes. Liking measures included liking for a 1 M sucrose solution, and pleasantness, desire to eat, and sweet taste intensity ratings for seven foods of a range of sweet taste intensities in a taste test. Results: Wide individual differences in sweet taste liking, in liking for a high sweet taste intensity, and in the relationships between sweet taste intensity and sweet taste liking were found. The majority of participants confirmed high liking and increasing liking for increasingly sweet tastes, but differing patterns of responses were also found. Higher liking for sweet foods was associated with increased sweet food selection and consumption at the test meal, and to some degree with free sugar and total sugar consumption in this restricted scenario. However, we found no associations between sweet taste liking, regardless of measure, and either free-living free sugar or total sugar intakes. Conclusions: These findings cast doubt on assumptions that sweet taste preferences are high for all and that these high sweet taste preferences drive high free sugar intakes. [ABSTRACT FROM AUTHOR]
