Healthy aging is a vital process that encompasses developing and maintaining functional ability to ensure wellbeing in older age. Social factors, including social relationships, support, engagement, and socioeconomic status, significantly influence successful aging outcomes. The UN Decade of Healthy Ageing, from 2021 to 2030, aims to transform societal perceptions of aging, create age-friendly environments, and provide integrated healthcare services to enhance the lives of older individuals globally. Demographic shifts towards aging populations pose challenges such as labour shortages and increased healthcare costs but also present opportunities to harness the potential of older societies through work incentives and community engagement. Social determinants, including economic stability, education, and cultural influences, play crucial roles in shaping healthy aging outcomes. Intergenerational relationships, social participation, and volunteerism are key components in promoting well-being in older adults. Psychological and physical well-being are intertwined, with social connections playing a significant role in enhancing overall health outcomes. Policy implications, technological innovations, and social integration strategies are essential for promoting healthy aging and ensuring the quality of life for older individuals. Chronic gastrointestinal disorders such as GERD, Gastritis, hiatus hernia, ZES etc require prolong treatment with pantoprazole and domperidone. Owing to its serious cardiac and psychological effects domperidone was banned in USA by USFDA for human use. Osteoporosis-related fracture, hypomagnesemia and adverse cardiac events are reported with longterm PPI usage. Neverthless, the combination of pantoprazole and domperidone is still widely used in rest of the world for long term management of chronic hyperacidic conditions in patients, that could result in serious adverse effects. Thus the study was conducted to evaluate long-term adverse effects associated with combined treatment of Domperidone and Pantoprazole in rodents at regular (Std) and higher (DD) therapeutic doses for 2 months. Our results demonstrated no mortality and no apparent morphological changes in rodents from both group however the complete blood count test revealed a dose dependent decline in blood Hb, lymphocytes, neutrophils and electrolytes levels in both groups. Moreover, Liver function test revealed significant alterations in BUN, ALP, ALT, Bilirubin and total cholesterol levels. ECG studies revealed tachycardia and a sharp change in P and R amplitude in both groups suggesting cardiac abnormalities. Liver and kidney histopathology revealed congested central vein and glomerulus, respectively. Thus, domperidone and pantoprazole combination may cause alteration in vital organ functions and hence the usage of this combination should be limited for reasonable cause by clinicians and regular therapeutic drug monitoring should be advised for patients on long term use. Erdheim-Chester disease (ECD) is an extremely rare non-Langerhans cell disorder that is believed to include both inflammatory and neoplastic characteristics. It is caused due to genetic mutations in proto-oncogenes like BRAF and MEK, while immunological pathways have an essential role in the onset and progression of the disease. Despite its rarity, ECD poses significant diagnostic and therapeutic challenges due to its heterogeneous clinical presentation and limited understanding of its underlying pathophysiology. Multiple organs can be affected, with the most frequent being long bones, central nervous system and retro-orbital abnormalities, pericardial and myocardial infiltration, interstitial lung disease, retroperitoneal fibrosis, and large blood vessel aberrations. Here, in this review, we comprehensively underline the current knowledge of ECD, including its epidemiology, clinical manifestations, genetics, pathophysiology, diagnostic modalities, and treatment options.
