Objectives: to analyze the use of spirituality/religiosity by oncology nurse residents in caring for patients with cancer., Methods: a census, descriptive, sectional study, with 46 nurse residents from three public hospitals in Rio de Janeiro. Data collection took place between August 2020 and January 2021, using a sociodemographic questionnaire, including a question about the use of spirituality/ religiosity to deal with work situations. Descriptive analysis was carried out using SPSS software version 22.0., Results: participants stated that they use religiosity/spirituality in work situations related to patients or themselves. In relation to patients, death was the most mentioned situation among professionals, and for themselves, everyday situations and emotional vulnerability were the most mentioned., Final Considerations: spirituality and religiosity are dimensions that guide oncology nurse residents' attitudes.

This study investigated the potential of selected compounds as inhibitors of SARS-CoV-2 M pro through pharmacokinetic and toxicological analyses, molecular docking, and molecular dynamics simulations. In silico molecular docking simulations revealed promising ligands with favorable binding affinities for M pro , ranging from -6.2 to -9.5 kcal/mol. Moreover, molecular dynamics simulations demonstrated the stability of protein-ligand complexes over 200 ns, maintaining protein secondary structures. MM-PBSA analysis revealed favorable interactions between ligands and M pro , with negative binding energy values. Hydrogen bond formation capacity during molecular dynamics was confirmed, indicating consistent interactions with M pro catalytic residues. The mango weevil, Sternochetus mangiferae (Fabricius) (Curculionidae), pest present in Brazil and is restricted to some municipalities in the Rio de Janeiro State. This curculionid attacks the mango crop exclusively and puts mango production globally at risk, especially those destined for export. Using ecological modeling tools, this study is the first to map the potential risk of S. mangiferae in Brazil. We aimed to identify the potential distribution of this pest in Brazilian states, drawing up thematic maps of regions that present suitable and unsuitable climatic conditions for the establishment of the pest using the MaxEnt ecological niche model. The average annual temperature, the annual precipitation, the average daytime temperature range, and the annual temperature range were the variables that contributed most to the selected model. The MaxEnt model predicted highly suitable areas for S. mangiferae throughout the Brazilian coast, especially on the northeast coast. The region responsible for more than 50% of mango production in Brazil, the São Francisco Valley, was classified by the model with suitability for the pest; it can impacts exportations due to the imposition of phytosanitary barriers. This information can be used in strategies to prevent the introduction and establishment of this pest in new areas and monitor programs in areas with recent occurrence. Objective: To investigate the association between body mass index (BMI), obesity, clinical outcomes, and mortality following coronary artery bypass grafting (CABG) in Brazil using a large sample with one year of follow-up from the Brazilian Registry of Cardiovascular Surgeries in Adults (or BYPASS) Registry database., Methods: A multicenter cohort-study enrolled 2,589 patients submitted to isolated CABG and divided them into normal weight (BMI 20.0-24.9 kg/m2), overweight (BMI 25.0-29.9 kg/m2), and obesity (BMI > 30.0 kg/m2) groups. Inpatient postoperative outcomes included the most frequently described complications and events. Collected post-discharge outcomes included rehospitalization and mortality rates within 30 days, six months, and one year of follow-up., Results: Sternal wound infections (SWI) rate was higher in obese compared to normal-weight patients (relative risk [RR]=5.89, 95% confidence interval [CI]=2.37-17.82; P=0.001). Rehospitalization rates in six months after discharge were higher in obesity and overweight groups than in normal weight group (χ=6.03, P=0.049); obese patients presented a 2.2-fold increase in the risk for rehospitalization within six months compared to normal-weight patients (RR=2.16, 95% CI=1.17-4.09; P=0.045). Postoperative complications and mortality rates did not differ among groups during time periods., Conclusion: Obesity increased the risk for SWI, leading to higher rehospitalization rates and need for surgical interventions within six months following CABG. Age, female sex, and diabetes were associated with a higher risk of mortality. The obesity paradox remains controversial since BMI may not be sufficient to assess postoperative risk in light of more complex and dynamic evaluations of body composition and physical fitness.

The carambola fruit fly Bactrocera carambolae Drew and Hancock (Diptera: Tephritidae) is an invasive fruit fly reported in North Brazil that threatens Brazilian fruit culture. Assessing the potential risk of establishing this pest is necessary to reduce the threat of B. carambolae dispersion to other countries and Brazilian regions and to avoid damage to the fruit trade. In this study, the CLIMEX model was used to understand the response of B. carambolae to climate change and to determine its potential global distribution with and without irrigation practices. Based on ecophysiological parameters, the model simulates factors limiting species distribution concerning the climate. To assess the seasonal variation in the density of B. carambolae, monitoring data in Uiramutã municipality, Roraima, from 2013 to 2019 was used. According to the CLIMEX forecast, large parts of America, Africa, and Asia, mainly in areas closest to the equator, are highly suitable for the survival of B. carambolae. Brazil is a good part of its territory with high suitability for B. carambolae, especially the North, South, and Southeast regions and the entire coastal area. The periods of the highest climatic suitability in the five Brazilian regions were January-May and October-December. The potential distribution area expands under irrigation and is highly suitable for most areas without cold stress. Staphylococcus aureus is a microorganism with high morbidity and mortality due to antibiotic-resistant strains, making the search for new therapeutic options urgent. In this context, computational drug design can facilitate the drug discovery process, optimizing time and resources. In this work, computational methods involving ligand- and structure-based virtual screening were employed to identify potential antibacterial agents against the S. aureus MRSA and VRSA strains. To achieve this goal, tetrahydroxybenzofuran, a promising antibacterial agent according to in vitro tests described in the literature, was adopted as the pivotal molecule and derivative molecules were considered to generate a pharmacophore model, which was used to perform virtual screening on the Pharmit platform. Through this result, twenty-four molecules were selected from the MolPort ® database. Using the Tanimoto Index on the BindingDB web server, it was possible to select eighteen molecules with greater structural similarity in relation to commercial antibiotics (methicillin and oxacillin). Predictions of toxicological and pharmacokinetic properties (ADME/Tox) using the eighteen most similar molecules, showed that only three exhibited desired properties (LB255, LB320 and LB415). In the molecular docking study, the promising molecules LB255, LB320 and LB415 showed significant values in both molecular targets. LB320 presented better binding affinity to MRSA (-8.18 kcal/mol) and VRSA (-8.01 kcal/mol) targets. Through PASS web server, the three molecules, specially LB320, showed potential for antibacterial activity. Synthetic accessibility (SA) analysis performed on AMBIT and SwissADME web servers showed that LB255 and LB415 can be considered difficult to synthesize and LB320 is considered easy. In conclusion, the results suggest that these ligands, particularly LB320, may bind strongly to the studied targets and may have appropriate ADME/Tox properties in experimental studies.

The scale mealybug, Diaspis echinocacti (Bouché, 1833) (Hemiptera: Diaspididae), is one of the main pests of the cactus pear in Brazil. The objective was to study biological aspects of D. echinocacti at the constant temperatures of 25, 28, 30, 33 and 35 °C with relative humidity of 60 ± 10% and photoperiod of 12 hours in the laboratory on the cactus pear cultivar, "Orelha de Elefante Mexicana", Opuntia stricta [Haw.] Haw. The development period (22 to 35 days) and survival in the egg (92 to 100%) and nymph (21.8 to 100%) stages and of the egg-adult cycle (20 to 100%), longevity (34.1 to 59.6 days) and fecundity (33 to 112 eggs) of D. echinocacti females with the different temperature and absence of males at the highest temperatures (> 30°C), indicated that the range between 25 °C and 30°C is the most favorable for this scale mealybug. This information may help to improve integrated management programs for D. echinocacti, in areas subject to seasonal temperature changes in the Brazilian regions where cactus pear is cultivated.

The MutS homolog 6 (MSH6) is a nuclear DNA mismatch repair (MMR) gene that encodes the MSH6 protein. MSH6 interacts with MSH2 to form the MutSα heterodimer. MutSα corrects DNA mismatches and unpaired nucleotides arising during DNA replication, deamination of 5-methylcytosine, and recombination between non-identical DNA sequences. In addition to correcting DNA biosynthetic errors, MutSα also recognizes chemically damaged DNA bases. Here, we show that inactivation of MSH6 affects the basal susceptibility of Arabidopsis thaliana to Pseudomonas syringae pv tomato DC3000. The msh6 T-DNA insertional mutant exhibited a reduced susceptibility to the bacterial invasion. This heightened basal resistance of msh6 mutants appears to be dependent on an increased stomatal closure, an accumulation of H 2 O 2 and double-strand breaks (DSBs) and a constitutive expression of pathogenesis-related (NPR1 and PR1) and DNA damage response (RAD51D and SOG1) genes. Complementation of this mutant with the MSH6 wild type allele under the control of its own promoter resulted in reversal of the basal bacterial resistance phenotype and the stomatal closure back to wild type levels. When an epidemic started in the Chinese city of Wuhan in December 2019, coronavirus was identified as the cause. Infection by the virus occurs through the interaction of viral S protein with the hosts' angiotensin-converting enzyme 2. By leveraging resources such as the DrugBank database and bioinformatics techniques, ligands with potential activity against the SARS-CoV-2 spike protein were designed and identified in this investigation. The FTMap server and the Molegro software were used to determine the active site of the Spike-ACE2 protein's crystal structure. Virtual screening was performed using a pharmacophore model obtained from antiparasitic drugs, obtaining 2000 molecules from molport ® . The ADME/Tox profiles were used to identify the most promising compounds with desirable drug characteristics. The binding affinity investigation was then conducted with selected candidates. A molecular docking study showed five structures with better binding affinity than hydroxychloroquine. Ligand_003 showed a binding affinity of -8.645 kcal·mol -1 , which was considered an optimal value for the study. The values presented by ligand_033, ligand_013, ligand_044, and ligand_080 meet the profile of novel drugs. To choose compounds with favorable potential for synthesis, synthetic accessibility studies and similarity analyses were carried out. Molecular dynamics and theoretical IC 50 values (ranging from 0.459 to 2.371 µM) demonstrate that these candidates are promising for further tests. Chemical descriptors showed that the candidates had strong molecule stability. Theoretical analyses here show that these molecules have potential as SARS-CoV-2 antivirals and therefore warrant further investigation.

The common bed bug, Cimex lectularius L., is a difficult urban pest to control. A simulated field study was conducted to compare the efficacy of steam application and an insecticide mixture spray (0.05% acetamiprid and 0.06% bifenthrin mixture) against C. lectularius. Three types of furniture (desk chair, upholstered armchair, and wooden table) were treated in the laboratory. The efficacy of the treatments was evaluated by visual inspection and placement of interceptor traps under the legs of the furniture. One hundred mixed stages of an insecticide-resistant population of C. lectularius were released onto each furniture item. After a 10-day acclimation period, each furniture item received steam treatment, insecticide spray, or no treatment. The second application of treatment was conducted 14 d later. Bed bug counts from interceptors and visual inspections were recorded at 13 d and 28 d after the initial treatment. At 28 d, the mean (± SE) live bed bug count in the steam, spray, and control group was 1 ± 0, 2 ± 1, and 83 ± 10, respectively. Both treatment methods were highly effective in controlling bed bugs on furniture. The mean bed bug count from interceptors in the steam, spray, and control groups were 0.3 ± 0.2, 11 ± 7, and 47 ± 9, respectively. There was no significant difference in the efficacy between steam and spray treatments based on either visual inspection or bed bug counts from interceptors. Skin Cancer (SC) is among the most common type of cancers worldwide. The search for SC therapeutics using molecular modeling strategies as well as considering natural plant-derived products seems to be a promising strategy. The phytochemical Rocaglamide A (Roc-A) and its derivatives rise as an interesting set of reference compounds due to their in vitro cytotoxic activity with SC cell lines. In view of this, we performed a hierarchical virtual screening study considering Roc-A and its derivatives, with the aim to find new chemical entities with potential activity against SC. For this, we selected 15 molecules (Roc-A and 14 derivatives) and initially used them in docking studies to predict their interactions with Checkpoint kinase 1 (Chk1) as a target for SC. This allowed us to compile and use them as a training set to build robust pharmacophore models, validated by Pearson's correlation ( p ) values and hierarchical cluster analysis (HCA), subsequentially submitted to prospective virtual screening using the Molport ® database. Outputted compounds were then selected considering their similarities to Roc-A, followed by analyses of predicted toxicity and pharmacokinetic properties as well as of consensus molecular docking using three software. 10 promising compounds were selected and analyzed in terms of their properties and structural features and, also, considering their previous reports in literature. This study aimed to identify potential inhibitors and investigate the mechanism of action on SARS-CoV-2 ACE2 receptors using a molecular modeling study and theoretical determination of biological activity. Hydroxychloroquine was used as a pivot structure and antimalarial analogues of 1,2,4,5 tetraoxanes were used for the construction and evaluation of pharmacophoric models. The pharmacophore-based virtual screening was performed on the Molport ® database (~7.9 million compounds) and obtained 313 structures. Additionally, a pharmacokinetic study was developed, obtaining 174 structures with 99% confidence for human intestinal absorption and penetration into the blood-brain barrier (BBB); posteriorly, a study of toxicological properties was realized. Toxicological predictions showed that the selected molecules do not present a risk of hepatotoxicity, carcinogenicity, mutagenicity, and skin irritation. Only 54 structures were selected for molecular docking studies, and five structures showed binding affinity (ΔG) values satisfactory for ACE2 receptors (PDB 6M0J), in which the molecule MolPort-007-913-111 had the best ΔG value of -8.540 Kcal/mol, followed by MolPort-002-693-933 with ΔG = -8.440 Kcal/mol. Theoretical determination of biological activity was realized for 54 structures, and five molecules showed potential protease inhibitors. Additionally, we investigated the Mpro receptor (6M0K) for the five structures via molecular docking, and we confirmed the possible interaction with the target. In parallel, we selected the TopsHits 9 with antiviral potential that evaluated synthetic accessibility for future synthesis studies and in vivo and in vitro tests.

Plasmodium the causative agent of malaria is a member of the phylum Apicomplexa, where all invasive forms have a substrate-dependent motility called gliding, key to malaria transmission. Gliding allows parasite host-cell recognition, binding, cell entry and trespassing the cytoplasm. In this process Plasmodium releases molecules from micronemes and the cell surface that are deposited on trails left behind on the substratum as the parasite progresses. Previously we identified the heat shock protein 70-1 (HSP 70-1) on the surface and micronemes of P. berghei ookinetes, the parasite form that invades the mosquito midgut. Soybean (Glycine max) is the world's most cultivated legume; currently, most of its varieties are Bt. Spodoptera spp. (Lepidoptera: Noctuidae) are important pests of soybean. An artificial neural network (ANN) is an artificial intelligence tool that can be used in the study of spatiotemporal dynamics of pest populations. Thus, this work aims to determine ANN to identify population regulation factors of Spodoptera spp. and predict its density in Bt soybean. For two years, the density of Spodoptera spp. caterpillars, predators, and parasitoids, climate data, and plant age was evaluated in commercial soybean fields. The selected ANN was the one with the weather data from 25 days before the pest's density evaluation. ANN forecasting and pest densities in soybean fields presented a correlation of 0.863. It was found that higher densities of the pest occurred in dry seasons, with less wind, higher atmospheric pressure and with increasing plant age. Pest density increased with the increase in temperature until this curve reached its maximum value. ANN forecasting and pest densities in soybean fields in different years, seasons, and stages of plant development were similar. Globally, people use sugarcane (Saccharum officinarum) to produce sugar and ethanol. Rainfed or irrigated sugarcane agricultural systems are available. Among the pests affecting this crop, the weevil Sphenophorus levis, Vaurie 1978 (Coleoptera: Curculionidae), is increasingly becoming a significant threat in southern South America. Sphenophorus levis populations are controlled using chemical or biological measures. Control decisions hinge upon the economic injury level (EIL). The EIL delineates the pest density that results in financial losses for producers. This study aims to determine the EIL for S. levis, considering the factors favoring this insect pest and chemical and biological control methods in rainfed and irrigated systems. The intensity of S. levis attacks was monitored in commercial sugarcane plantations over four years in João Pinheiro, Minas Gerais, Brazil. Sampling occurred in a 50 × 50 × 30-cm-deep trench dug in the soil surrounding the sugarcane clump. The total number of stumps in the clump, including those attacked by S. levis, was tallied. The EILs for this pest were 5.93% and 4.85% of targeted stumps for chemical control in rainfed and irrigated crops, respectively. Biological control in sugarcane plots resulted in an EIL of 4.15% and 3.40% for stumps attacked in rainfed and irrigated crops, respectively. Damage from Sinoxylon anale Lesne, a woodboring beetle not previously known to be established in Brazil, was observed in young jabuticaba trees (Plinia cauliflora, Myrtaceae) in a nursery in the municipality of Laranjal Paulista, state of São Paulo. We immediately advised MAPA ("Ministério da Agricultura, Pecuária e Abastecimento") and collected samples from the nursery and from different hosts in nearby areas, to identify the specimens and investigate the dynamics of the infestation in the jabuticaba trees. Sinoxylon anale was also collected in ethanol-baited and ultraviolet-light traps and in dry branches of the native species pau-jacaré (Piptadenia gonoacantha, Fabaceae) and inga (Inga vera, Fabaceae), and the exotic pau-d'água (Dracaena fragrans, Asparagaceae) in the municipality of Piracicaba, state of São Paulo. These collections established that S. anale larvae and adults develop in dead branches of four new host plants. Taxonomic studies using morphological parameters and DNA barcoding confirmed the identification of S. anale. An illustrated key to the three Sinoxylon species now recorded in Brazil is provided, and the COI gene sequences have been made available in a public database. Sinoxylon anale probably attacked the young jabuticaba trees after they were killed by larvae of long-horned beetles (Cerambycidae). Postprandial hyperglycemia, typical manifestation of Type 2 Diabetes Mellitus (T2DM), is associated with notable global morbidity and mortality. Preventing the advancement of this condition by delaying the rate of glucose absorption through inhibition of α-amylase and α-glucosidase enzymatic activities is of utmost importance. Finding a safe antidiabetic drug is essential since those that are currently on the market have drawbacks like unpleasant side effects. The current study utilized computer-aided drug design (CADD), as a quick and affordable method to find a substitute drug template that can be used to control postprandial hyperglycemia by modulating the activity of α-amylase and α-glucosidase enzymes. The Natural Products Activity and Species database (NPASS) (30,926 compounds) was screened in silico, with a focus on evaluating drug-likeness, toxicity profiles and ability to bind on a target protein. Two molecules NPC204580 (Chrotacumine C) and NPC137813 (1-O-(2-Methoxy-4-Acetylphenyl)-6-O-(E-Cinnamoyl)-Beta-D-Glucopyranoside) were identified as potential dual inhibitors for α-amylase and α-glucosidase with free binding energies of -14.46 kcal/mol and -12.58 kcal/mol for α-amylase, and -8.42 kcal/mol and -8.76 kcal/mol for α-glucosidase, respectively. The molecules showed ionic, H-bonding and hydrophobic interactions with critical amino acid residues of both enzymes. Moreover, 100 ns molecular dynamic simulations showed that both molecules are stable on the receptors' active sites based on root mean square deviation (RMSD), root mean square fluctuation (RMSF), and the Generalized Born surface area (GBSA) energy calculated. The two compounds are thus promising therapeutic agents for T2DM that merit further investigation due to their excellent binding energies, encouraging pharmacokinetics, toxicity profiles, and stability as demonstrated in simulated studies. According to fruit growers and theMinnesota Department of Agriculture, population size and feeding damage has been an increasing concern since 2010. Based on trapcatch data, populations have recently exceeded 4,000 beetles/trap/week during July-August near raspberry fields, and can increase by an order of magnitude within 7-10 days. The primary goals of this study were to assess the spatial distribution of P. japonica adults in raspberry, and to develop and validate a practical fixed-precision sequential sampling plan for grower use. Taylor's Power Law (TPL) regression was used to characterize the beetle's spatial pattern in research plots and commercial fields, either with or without insecticide applications. We then used Green's plan to develop an enumerative sequential sampling plan to estimate P. japonica density in primocane raspberry. Beetle population data were collected at two locations in southern Minnesota, including the Rosemount Research andOutreach Center, and a commercial field near Forest Lake. The TPL results, via slope comparisons, indicated no significant differences in P. japonica spatial pattern between insecticide treated plots versus untreated plots, or among 4 different insecticides (P>0.05). Utilizing all spatial pattern data, we characterized the distribution of P. japonica beetles to be highly aggregated in raspberry, with TPL slopes ranging from b = 1.38 to 1.55; all slopes were found to be >1.0. Although the slopeswere not significantly different, we accounted for variability in spatial pattern by using 33 independent data sets, and the Resampling for Validation of Sampling Plans (RVSP) model to validate a sampling plan with a final average precision level of 0.25 (SEM/mean), recommended for integrated pest management (IPM) purposes. The final sampling plan required an average sample number of only 15, 1-m-row samples, while providing high relative net precision (RNP), and thus a cost-effective, efficient sample plan for growers. Although the slopeswere not significantly different, we accounted for variability in spatial pattern by using 33 independent data sets, and the Resampling for Validation of Sampling Plans (RVSP) model to validate a sampling plan with a final average precision level of 0.25 (SEM/mean), recommended for integrated pest management (IPM) purposes. Background: While most research has focused on the association between intracytoplasmic sperm injection (ICSI) and neurodevelopmental disorders in children, relatively little attention has been given to its metabolic effects. Previous studies have reported that low serum lipid levels are associated with mental health problems. Our objective was to analyze the impact of ICSI on metabolic alterations compared to their in vitro fertilization (IVF) counterparts in male offspring, as well as its interaction with paternal overweight/obesity. Methods: We recruited families between January 2006 and December 2017 at the Center for Reproductive Medicine, Shandong University, China. Prospective data of offspring were obtained for body mass index (BMI), blood pressure, glucose, and lipid profile in their 0–11 years old. Linear mixed models were utilized to compute the mean difference and 95% confidence intervals (CI). Results: A total of 14,196 offspring visits were identified. In offspring aged 4–11 years, ICSI-conceived offspring exhibited significantly lower fasting glucose z-scores, total cholesterol z-scores, and low-density lipoprotein cholesterol (LDL-C) z-scores compared with their IVF counterparts (fasting glucose z-score: adjusted mean difference: − 0.13, 95% CI: − 0.23 to − 0.03; total cholesterol z-score: adjusted mean difference: − 0.13, 95% CI: − 0.23 to − 0.02; LDL-C z-score: adjusted mean difference: − 0.12, 95% CI: − 0.22 to − 0.01). Paternal overweight/obesity significantly influenced the relationship between ICSI and metabolic changes in offspring. In offspring born from fathers with overweight/obesity, ICSI-conceived offspring displayed significantly lower fasting glucose and total cholesterol z-scores than their IVF controls (fasting glucose z-score: adjusted mean difference: − 0.20, 95% CI: − 0.32 to − 0.08; total cholesterol z-score: adjusted mean difference: − 0.15, 95% CI: − 0.27 to − 0.02). In offspring born to fathers with normal weight, ICSI-conceived offspring showed significantly lower systolic blood pressure z-scores compared to those conceived via the IVF procedures (adjusted mean difference: − 0.21, 95% CI: − 0.37 to − 0.05). Conclusions: The findings of this study suggested that ICSI was associated with altered glucose and lipid profiles compared to their IVF controls, characterized by lower fasting glucose z-scores, total cholesterol z-scores, and LDL-C z-scores. The soybean pod borer Leguminivora glycinivorella (Matsumura) is one of the most important soybean pests and often causes serious damage to Glycine max (L.) Merr. a leading source of dietary protein and oil in animal feed. However, the potential distribution patterns of this economically important pest and its driving factors require further investigation. Here, we used the optimized MaxEnt model to predict the potential distribution of this pest with multiple variables associated with climate, land use, and host plant, at its recorded range and a globe scale. Based on 4 variable combinations, the results show that the current suitable habitats of L. glycinivorella are primarily distributed in most of China, the Korean Peninsula, and Japan. Whereas no suitable area is present in other continents. In future projections, the suitable region shows a slight northward expansion compared with the result predicted with current climatic conditions, and the suitable areas of almost all future projections were stable in size. Among the 9 bioclimatic factors, BIO03 (isothermality) consistently highly contributes to the predictions, indicating that temperature may be a key factor influencing the habitat distribution of L. glycinivorella. Comparative analyses of projections further show that non-climatic factors are informative in the modeling as routinely used bioclimate variables. The spatio-temporal distribution patterns of suitable habitats and the regulatory factors predicted in this study could provide important guidance for L. glycinivorella management. CRISPR/Cas9-mediated gene editing requires high efficiency to be routinely implemented, especially in species which are laborious and slow to transform. This requirement intensifies further when targeting multiple genes simultaneously, which is required for genetic screening or more complex genome engineering. Species in the Citrus genus fall into this category. Here we describe a series of experiments with the collective aim of improving multiplex gene editing in the Carrizo citrange cultivar using tRNA-based sgRNA arrays. We evaluate a range of promoters for their efficacy in such experiments and achieve significant improvements by optimizing the expression of both the Cas9 endonuclease and the sgRNA array. In the case of the former we find the UBQ10 or RPS5a promoters from Arabidopsis driving the zCas9i endonuclease variant useful for achieving high levels of editing. The choice of promoter expressing the sgRNA array also had a large impact on gene editing efficiency across multiple targets. In this respect Pol III promoters perform especially well, but we also demonstrate that the UBQ10 and ES8Z promoters from Arabidopsis are robust alternatives. Ultimately, this study provides a quantitative insight into CRISPR/Cas9 vector design that has practical application in the simultaneous editing of multiple genes in Citrus, and potentially other eudicot plant species. Thyrinteina arnobia (Lepidoptera: Geometridae) is a native American species. Despite its historical importance as an insect pest in Eucalyptus plantations, more information is needed regarding the population diversity, demography, and climatic variables associated with its distribution in different regions of Brazil. We used a phylogeographic approach to infer the genetic diversity, genetic structure, and demographic parameters of T. arnobia. We also conducted an ecological niche modeling (ENM) to predict suitable areas for T. arnobia occurrence in Brazil and other countries worldwide. Although T. arnobia populations have low genetic diversity in Brazil, we identified mitochondrial haplogroups predominating in different Brazilian regions and high ФST and ФCT values in AMOVA, suggesting a low frequency of insect movement among these regions. These results indicate that outbreaks of T. arnobia in Eucalyptus areas in different regions of Brazil are associated with local or regional populations, with no significant contribution from long-distance dispersal from different regions or biomes, suggesting that pest management strategies would be implemented on a regional scale. In Brazil, the demographic and spatial expansion signals of T. arnobia seem to be associated with the history of geographical expansion of Eucalyptus plantations, a new sustainable host for this species. ENM indicated that isothermality and annual rainfall are critical climatic factors for the occurrence of T. arnobia in tropical and subtropical areas in the Americas. Vepris Comm. ex A. Juss. is a genus of 96 species extending from Africa to India that are distinct in their unarmed stems and their digitately (1-)3(-5) foliolate leaflets, and whose many secondary compounds earn them uses in traditional medicine. Mziray (1992) subsumed six related genera into Vepris, with Vepris amaniensis (Engl.) Mziray becoming somewhat of a dustpan for ambiguous specimens (Cheek & Luke, 2023). This study, using material from the Kew herbarium, sought to pull out novel species from those previously incorrectly filed as Vepris amaniensis, and here describes the new species Vepris usambarensis sp. nov. This species is morphologically distinct from Vepris amaniensis with its canaliculate to winged petioles, 0.5–2.3 cm long inflorescences, 1–3 foliolate leaflets, and hairs on inflorescences and stem apices. Phytochemical analysis attributed seven compounds to Vepris usambarensis: tecleanthine (1), evoxanthine (2), 6-methoxytecleanthine (3), tecleanone (4), 1-(3,4-methylenedioxyphenyl)-1,2,3-propanetriol (5), lupeol (6), and arborinine (7). This is a unique mixture of compounds for a species of Vepris, though all are known to occur in the genus, with the exception of 1-(3,4-methylenedioxyphenyl)-1,2,3-propanetriol (5) which was characterized from a species in the Asteraceae. An attempt at constructing a phylogeny for Vepris using the ITS and trnL-F regions was made, but these two regions could not be used to differentiate at species level and it is suggested that 353 sequencing is used for further research. Originally more than one new species was hypothesized to be within the study group; however, separating an additional species was unsupported by the data produced. Further phylogenetic analysis is recommended to fully elucidate species relationships and identify any cryptic species that may be present within Vepris usambarensis. [ABSTRACT FROM AUTHOR]

Natural products received much attention as an environmentally beneficial solution for pest management. Therefore, the extracts of invasive silverleaf nightshade (Solanum elaeagnifolium Cav.) weeds using their berries parts (seeds, peels and mucilage) supported by bioassay-guided fractionation were tested against both the greater wax moth (Galleria mellonella) and Erwinia carotovora pv. carotovora causes of the blackleg of potatoes. The seeds and peels of S. elaeagnifolium were successively extracted by maceration using dichloromethane (DCM), ethyl acetate (EtOAc), and ethanol (EtOH), respectively. While, its mucilage was extracted using EtOAc. The successive EtOH extract of the plant seeds had promising inhibition efficacy and the best minimal inhibition concentration (MIC) of 50 µg/ml against E. Carotovora amongst other extracts (DCM and EtOAc of the plant berries parts). Depending on dose response activity, EtOH extract had G. mellonella larval mortality and pupal duration rates (LC50; 198.30 and LC95; 1294.73 µg/ml), respectively. Additionally, this EtOH extract of seeds was fractionated using preparative TLC to three characteristic bands. The insecticidal and bacterial activities of these isolated bands (SEA, SEB, and SEC) were evaluated at a dose of 100 µg/ml, causing mortality by 48.48, 62.63 and 92.93% (G. mellonella larvae) and inhibition by 15.22, 0.00 and 31.66 mm (E. carotovora), respectively. Moreover, the separated major three bands were tentatively identified using LC-ESI-MS analysis revealing the presence of two phenolic acids; chlorogenic acid (SEA) and dicaffeoyl quinic acid (SEB) in addition to one steroidal saponin (SEC) annotated as borassoside E or yamoscin. Finally, the plant seeds' successive EtOH extract as well as its active constituents, exhibited potential broad-spectrum activity and the ability to participate in future pest management initiatives. Methyl jasmonate (MeJA) is a phytohormone involved in plant defense against stress. However, its application as pretreatment in soybean seeds is limited. Here, we investigated whether seed pretreatment with MeJA mitigated the negative effects of water restriction (WR) and mechanical wounding (MW) in soybean seedlings at the V1 vegetative stage. Seeds of Glycine max (Monsoy 6410 variety) were pretreated with water or 12.5 µM MeJA for 14 h. The obtained seedlings were transferred to pots containing substrate (soil and sand) kept in a greenhouse and subjected to different growth conditions: control (no stress), WR (40% water retention), and MW. The experiment was conducted in a 2 × 3 factorial scheme (2 seed pretreatments × 3 growth conditions). The variables analyzed were ethylene levels, hydrogen peroxide, lipid peroxidation, antioxidant system enzymes, sugars, amino acids, proteins, proline, and growth (root and shoot length). WR negatively affected seedling growth, regardless of seed pretreatment, but proline levels increased with MeJA application. In seedlings subjected to MW, MeJA increased ethylene release, which was related to reduced damage. It suggests that pretreatment of soybean seeds with MeJA is a promising tool to mitigate the deleterious effects of biotic and abiotic stresses during seedling establishment, inducing distinct tolerance strategies. Increased consumption of palm oil results in more palm oil by-products. To meet environmental sustainability goals, deodorizing distillate palm oil (DDPO) also calls attention to diversifying its technological or commercial application. Because of this, the present work, to present new economic-scientific alternatives for this by-product, aimed to evaluate the larvicide effect of DDPO as to investigate the synergistic effect of the combination of these free fatty acids (FFA's) present in the composition of DDPO, against larvae of the 3rd instar of Ae. aegypti. The DDPO larvicide assay showed a high mortality rate, with an LC50 of 6.18 μg.mL− 1 at 24 h and 8.73 μg.mL− 1 at 48 h of treatment. In addition, the results of LC50, among saturated fatty foods and between the combination of free fatty acids (myristic, lauric, stearic, and oleic) with palmitic acid were met positively by second-degree polynomial regression analysis. Finally, the study of molecular docking, corroborated the potential inhibitors of the binding of juvenile hormones. Together, the results suggest that DDPO can be a potential natural larvicide agent, making it an alternative to the excessive use of synthetic insecticides, thus minimizing its impacts on the environment, and promoting new technologies for the use of this palm oil by-product. Cassava (Manihot esculenta) is among the most important staple crops globally, with an imperative role in supporting the Sustainable Development Goal of 'Zero hunger'. In sub-Saharan Africa, it is cultivated mainly by millions of subsistence farmers who depend directly on it for their socio-economic welfare. However, its yield in some regions has been threatened by several diseases, especially the cassava brown streak disease (CBSD). Changes in climatic conditions enhance the risk of the disease spreading to other planting regions. Here, we characterise the current and future distribution of cassava, CBSD and whitefly Bemisia tabaci species complex in Africa, using an ensemble of four species distribution models (SDMs): boosted regression trees, maximum entropy, generalised additive model, and multivariate adaptive regression splines, together with 28 environmental covariates. We collected 1,422 and 1,169 occurrence records for cassava and Bemisia tabaci species complex from the Global Biodiversity Information Facility and 750 CBSD occurrence records from published literature and systematic surveys in East Africa. Our results identified isothermality as having the highest contribution to the current distribution of cassava, while elevation was the top predictor of the current distribution of Bemisia tabaci species complex. Cassava harvested area and precipitation of the driest month contributed the most to explain the current distribution of CBSD outbreaks. The geographic distributions of these target species are also expected to shift under climate projection scenarios for two mid-century periods (2041–2060 and 2061–2080). Our results indicate that major cassava producers, like Cameron, Ivory Coast, Ghana, and Nigeria, are at greater risk of invasion of CBSD. These results highlight the need for firmer agricultural management and climate-change mitigation actions in Africa to combat new outbreaks and to contain the spread of CBSD. [ABSTRACT FROM AUTHOR]

Cholinesterases (ChEs) are pivotal in the pathophysiology of various neuromuscular diseases, including Parkinson's disease, Alzheimer's disease, myasthenia gravis, and vascular dementia. The involvement of ChEs in open-angle glaucoma establishes them as promising drug targets. This study employed hierarchical virtual screening (HVS) to identify lead compounds against cholinesterase drug targets. A four-step, knowledge-based, and time-efficient HVS protocol was implemented, resulting in the identification of 41 novel scaffolds capable of evolving into diverse functional ChE inhibitors. This includes inhibitors with selectivity for acetylcholinesterase (AChE) or butyrylcholinesterase (BChE), dual-target, and dual-binding-site inhibitors. The proposed HVS scheme integrates structure-based pharmacophores, docking methodologies, and physicochemical-descriptor-based filters. Among the identified scaffolds, 13 potential ChE inhibitors were selected based on their non-covalent interactions with key binding site residues of both AChE and BChE. Furthermore, an assessment of the physicochemical and pharmacokinetic profiles of these compounds was conducted. The selected potential inhibitors are recommended for further evaluation through in vitro and in vivo assay studies. Aim: To develop an alternative drug molecule design to fludarabine which is commonly used in chronic lymphocytic leukemia. Methods: The molecular properties and biological activities of the drug molecules were determined using Molinspiration software. We investigated the biological activity and drug properties of fludarabine by changing the positions of bioisosteres on the molecular structure. Results: In our studies of derivatives of the fludarabine drug molecule, we obtained data by adding different structures to the Y part without changing the X structure (F) of fludarabine. We have used the abbreviation 'M' to refer to the molecules in these experiments. We predict that the M6 derivative of fludarabine will have higher ion channel modulator, kinase and protease activity compared to fludarabine. We predict that the M15 derivative of fludarabine will have higher G- protein coupled receptors, ion channel modulator, kinase, and protease and enzyme inhibition activity compared to fludarabine. In our experiments with fludarabine derivatives, we have experimented by binding different molecules to both the X and Y structures of fludarabine at the same time. We have used the abbreviation 'C' to refer to the molecules in these experiments. In these experiments, we did not achieve higher biological activity than fludarabine. Conclusions: The results suggest that this newly designed M15 derivative of fludarabine molecule may be a better antileukemic drug molecules in the future and may be useful for further drug molecule development research in medical biochemistry, chemistry and pharmacology. One invasive species that decreases citrus fruit yield is the Asian citrus psyllid, Diaphorina citri Kuwayama. The psyllids are the carriers of Candidatus Liberibacter species, the pathogen that causes citrus greening disease (commonly called huanglongbing, or HLB), which is the most devastating citrus disease in the world. Originally from Southeast Asia, the Asian citrus psyllid, a highly invasive species, is spreading rapidly throughout the world. Regarding geographical distribution and population growth, climate change will aggravate its impacts on many citrus agroecosystems. However, limited information on the mechanisms underpinning the pest's invasion impedes the formulation of biosecurity risk measures. We highlight the potential suitable areas for D. citri, combining their climatic requirements, temperature and humidity, in the CLIMEX model. The model's performance was high, and it can be utilized for biosecurity risk assessment, as only 2.82% of the 496 known occurrence spots were located in areas deemed unsuitable by the model's fit. The potential habitat suitability for D. citri covered all continents except Antarctica, and the suitability ranges from unsuitable (EI < 0) to highly suitable areas (EI ≥ 30). Our model predicted suitable areas outside its known native and invaded regions, especially in Africa, South America, and Oceania. Most of these regions will remain suitable for D. citri in the future. During the oestrous cycle, the bovine endometrium undergoes morphological and functional changes, which are regulated by alterations in the levels of oestrogen and progesterone and consequent changes in gene expression. To clarify these changes before and after oestrus, RNA-seq was used to profile the transcriptome of oestrus-synchronized beef heifers. Endometrial samples were collected from 29 animals, which were slaughtered in six groups beginning 12 h after the withdrawal of intravaginal progesterone releasing devices until seven days post-oestrus onset (luteal phase). The groups represented proestrus, early oestrus, metoestrus and early dioestrus (luteal phase). Changes in gene expression were estimated relative to gene expression at oestrus. Ingenuity Pathway Analysis (IPA) was used to identify canonical pathways and functional processes of biological importance. A total of 5,845 differentially expressed genes (DEGs) were identified. The lowest number of DEGs was observed at the 12 h post-oestrus time point, whereas the greatest number was observed at Day 7 post-oestrus onset (luteal phase). A total of 2,748 DEGs at this time point did not overlap with any other time points. Prior to oestrus, Neurological disease and Organismal injury and abnormalities appeared among the top IPA diseases and functions categories, with upregulation of genes involved in neurogenesis. Lipid metabolism was upregulated before oestrus and downregulated at 48h post-oestrus, at which point an upregulation of immune-related pathways was observed. The use of insecticides on a large scale and applied at the wrong time can increase the cost of potato production. Good agricultural pest management practices can reduce the number of insecticide applications and promote the sustainability of potato production in modern agriculture. Thus, the objective was to compare the costs and revenues of a pest management system that uses monitoring (IPM-integrated pest management) with a conventional system (calendar-based insecticide application) in the potato crop. Two experiments were carried out: experiment 1, carried out in the summer with the cultivars Ágata, Atlantic, and FLs 2027, 1867, 2215, and 2221, and experiment 2, carried out in autumn with the cultivar Ágata. The densities of sucking insects, defoliators, and natural enemies were evaluated and compared. In both experiments, treatments were based on pest monitoring (insecticide applications when reaching economic thresholds [ETs]) and a conventional system (calendar-based insecticide application). In experiment 2, a control group was also carried out without the use of insecticides. In experiment 1, with the use of pest monitoring, the number of sprays was reduced by 66.67%, pest control costs were decreased by 60.86%, and there was an increase of 8.97% in net income from potato production. Whilst in experiment 2, the number of sprays was reduced by 71.42%, pest control costs were decreased by 51.38%, and an increase of 12.75% in net revenue from potato production was observed. Thus, it is concluded that the application of insecticides when the pest population reaches the economic threshold (ET) reduces insecticide applications and costs and increases net income in the potato crop. The soybean bud borer, a soybean pest in Brazil, was initially identified as Crocidosema aporema (Walsingham 1914) (Lepidoptera: Tortricidae). Outbreaks of this species have recently increased, but identification of this pest remains uncertain, and the historical factors associated with its geographic distribution in Brazil are little known. Here, we conducted a species characterization and phylogeographic analysis based on molecular and morphological evidence. Ninety individuals of bud-borers Lepidoptera were collected in different regions of Brazil. We sequenced COI and COII mitochondrial genes and examined wing patterns and male genital morphology. DNA barcoding approach revealed that 10 individuals were Argyrotaenia sphaleropa (Meyrick 1909) (Lepidoptera: Tortricidae) and 80 were a species of the genus Crocidosema Zeller. The morphology of the adult genitalia and wings proved to be insufficient to confirm the identification of Brazilian individuals as C. aporema , a species originally described from a high-elevation site in Costa Rica. Furthermore, the genetic distance between putative C. aporema specimens from Brazil and Costa Rica (ranging from 5.2% to 6.4%) supports the hypothesis that the Brazilian specimens are not referable to C. aporema. Our analysis revealed a single genetic strain (i.e. species) with low genetic diversity on soybean crops. We found no indication that the genetic structure was related to geographic distance among populations or edaphoclimatic regions. The population expansion of the soybean bud borer coincides with the increase in the area of soybean production in Brazil, suggesting that expanded soybean farming has allowed a significant increase in the effective population size of this pest. Background: The Asian citrus psyllid (ACP) Diaphorina citri Kuwayama (Hemiptera: Liviidae) is a destructive, invasive species that poses a serious threat to the citrus industry wherever it occurs. The psyllid vectors the phloem-limited bacteria 'Candidatus Liberibacter americanus' and 'Ca. L. asiaticus', causal agents of the incurable citrus greening disease or huanglongbing (HLB). It is essential to understand which regions and areas are suitable for colonization by ACP to formulate appropriate policy and preventive measures. Considering its biology and ecology, we used a machine learning algorithm based on the MaxEnt (Maximum Entropy) principle, to predict the potential global distribution of ACP using bioclimatic variables and elevation., Results: The model predictions are consistent with the known distribution of ACP and also highlight the potential occurrence outside its current ecological range, that is, primarily in Africa, Asia and the Americas. Background: Present climate studies on invasive species imply that climate change will alter the habitat suitability of invasive pests, especially given the projected rise in average global temperatures by the end of 2100. However, globally, limited information exists on the habitat suitability of the mango stone weevil, Sternochetus mangiferae Fabricius, which impedes the development of early detection and preventive measures. Herein, we used the MaxEnt model to estimate the potential global geographical distribution of S. mangiferae., Results: Our results revealed that thermal conditions played a significant role in explaining the invasion risk of S. mangiferae. Habitat suitability was found in all continents, except Antarctica. Under the present condition, habitat suitability covered 5.67 × 10 7  km 2 . For ssp126, habitat suitability will decrease from the 2060s (5.58 × 10 7  km 2 ) and 2080s (5.57 × 10 7  km 2 ). Similarly, under ssp585, suitable areas will decrease from 5.62 × 10 7 to 5.51 × 10 7  km 2 for the 2060s and 2080s, respectively., Conclusion: Our study has estimated variability in the habitat suitability of S. mangiferae which establishes a foundation for determining global risk assessment and response plans for the pest. Background: Over the past two decades, bed bugs (Cimex spp.) have resurged as common urban pests around the world. The search for efficient and safe control measures has become a key interest among researchers, manufacturers, and pest control professionals. In this study, we evaluated and compared the efficacy of steam, diatomaceous earth (DE) dust, and a combination of both against tropical bed bugs (Cimex hemipterus (F.)) under laboratory and field conditions., Results: In the laboratory study, the mortality of bed bugs after 2 days of exposure to DE dust was 100%. When bed bugs stayed on the surface of an object or in cracks, a brief steam treatment (1 s) caused 100% mortality. However, when bed bugs were hidden under a fabric cover, steam application for 10 s only caused 89 ± 6% mortality. Bed bugs that survived steam treatment exhibited reduced feeding activity. In a 14-week long study, there was no significant difference in the reduction rate of bed bugs between steam treatment and DE dust treatment. A 37-week long control study showed that steam and steam plus DE dust treatments eliminated 97-100% of the infestations., Conclusion: Applying steam and DE dust are effective strategies for eliminating natural tropical bed bug infestations. CRISPR/Cas9 ribonucleoproteins enable DNA-free genome editing; thus, improving protoplast culture is essential for the efficient development of mutant plants. However, the use of protoplast cultures is limited because a universal method cannot be applied to diverse plants. Solanum lycopersicum 'Heinz 1706,' a model cultivar for tomato genome analysis, has not yet been studied for DNA-free genome editing. We optimized the protoplast culture method for the tomato model cultivar 'Heinz 1706' using combinations of plant growth regulators (PGRs) and basal media. Isolated protoplasts were cultured in R-Ini medium for cell division and micro-calli proliferation, and then the medium was changed to G-207.3 medium for callus formation. Among different concentrations of 1-naphthaleneacetic acid (NAA) and 6-benzylaminopurine (BAP), the combination of 0.05 mg/L NAA and 0.5 mg/L BAP in the R-Ini medium was observed to be highly efficient for micro-calli development. G-207.3 liquid medium was more efficient for mini-calli formation than the R-Ini liquid medium. For calli formation from the mini-calli, 0.1 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D), 0.05 mg/L NAA, and 0.5 mg/L BAP were used in G-207.3 solid medium. In addition, five single guide RNAs (sgRNAs) were designed to target SlPelo using polyethylene glycol-mediated transfection, thereby developing a tool for tomato yellow leaf curl virus (TYLCV) resistance breeding. sgRNAs and Cas9 complexes were delivered into protoplasts using PEG-mediated transfection, and sgRNA2 resulted in a high mutagenesis efficiency.
