Abstract

During the past decade, state-of-the-art planet-finder instruments like SPHERE@VLT, coupling coronagraphic devices and extreme adaptive optics systems, unveiled, thanks to large surveys, around 20 planetary mass companions at semi-major axis greater than 10 astronomical units. Direct imaging being the only detection technique to be able to probe this outer region of planetary systems, the SPHERE infrared survey for exoplanets (SHINE) was designed and conducted from 2015 to 2021 to study the demographics of such young gas giant planets around 400 young nearby solar-type stars. In this paper, we present the observing strategy, the data quality, and the point sources analysis of the full SHINE statistical sample as well as snapSHINE. Both surveys used the SPHERE@VLT instrument with the IRDIS dual band imager in conjunction with the integral field spectrograph IFS and the angular differential imaging observing technique. All SHINE data (650 datasets), corresponding to 400 stars, including the targets of the F150 survey, are processed in a uniform manner with an advanced post-processing algorithm called PACO ASDI. An emphasis is put on the classification and identification of the most promising candidate companions. Compared to the previous early analysis SHINE F150, the use of advanced post-processing techniques significantly improved by one or 2 magnitudes (x3-x6) the contrast detection limits, which will allow us to put even tighter constraints on the radial distribution of young gas giants. This increased sensitivity directly places SHINE as the largest and deepest direct imaging survey ever conducted. We detected and classified more than 3500 physical sources. One additional substellar companion has been confirmed during the second phase of the survey (HIP 74865 B), and several new promising candidate companions are awaiting second epoch confirmations., Comment: Accepted for publication in A&A. 19 pages, 19 figures. Abstract

[Full abstract in the paper] We aim to better constrain the atmospheric properties of the directly imaged exoplanet 51~Eri~b by using a retrieval approach on higher signal-to-noise data than previously reported. In this context, we also compare the results of using the atmospheric retrieval code \texttt{petitRADTRANS} vs a self-consistent model to fit atmospheric parameters. We present a higher signal-to-noise $YH$ spectrum of the planet and revised $K1K2$ photometry (M$_{K1} = 15.11 \pm 0.04$ mag, M$_{K2} = 17.11 \pm 0.38$ mag). The best-fit parameters obtained using an atmospheric retrieval differ from previous results using self-consistent models. In general, we find that our solutions tend towards cloud-free atmospheres (e.g. log $\tau_{\rm clouds} = -5.20 \pm 1.44$). For our ``nominal'' model with new data, we find a lower metallicity ([Fe/H] $= 0.26\pm$0.30 dex) and C/O ratio ($0.38\pm0.09$), and a slightly higher effective temperature (T$_{\rm{eff}} = 807\pm$45 K) than previous studies. The surface gravity (log $g = 4.05\pm0.37$) is in agreement with the reported values in the literature within uncertainties. We estimate the mass of the planet to be between 2 and 4 M$_{\rm{Jup}}$. When comparing with self-consistent models, we encounter a known correlation between the presence of clouds and the shape of the $P-T$ profiles. Abstract

Objectives: To compare the efficacy of venetoclax‐azacitidine (VEN–AZA) with AZA in the real‐life for patients with first relapsed or refractory acute myeloid leukaemia (R/R AML). Methods: We retrospectively analysed R/R AML patients treated with VEN–AZA at the Institut Paoli Calmettes between September 2020 and February 2022. We compared them to a historical cohort of patients treated with AZA between 2010 and 2021. Results: Thirty‐five patients treated with VEN–AZA were compared with 140 patients treated with AZA. There were more favourable cytogenetics (25.7% vs. 8.6%; p = 0.01) and less FLT3‐ITD mutated AML (8.8% vs. 25.5%; p =.049) in the VEN–AZA group. The overall 30‐day mortality rate was 7.4% and the overall 90‐day mortality was 20%, with no difference between the groups. The complete remission rate was 48.6% in the VEN–AZA group versus 15% (p <.0001). The composite complete response rate was 65.7% in the VEN–AZA group versus 23.6% (p <.0001). OS was 12.8 months in the VEN–AZA group versus 7.3 months (p = 0.059). Patients with primary refractory AML, poor‐risk cytogenetics, prior hematopoietic stem‐cell transplantation (HSCT) and FLT3‐ITD mutated AML had lower response and survival rates. Conclusion: VEN–AZA was associated with a better response rate and a longer survival than AZA monotherapy in AML patients who relapsed after or were refractory to intensive chemotherapy. Abstract

Objective: Multirooted teeth respond less favorably to non-surgical periodontal treatment and long-term tooth prognosis is influenced by the degree of furcation involvement (FI). Therapeutic strategy for multirooted teeth is essentially based on accurate diagnosis of the FI. The aim of this systematic review is to evaluate the accuracy of the different furcation assessment methods and to determine if radiographic help is needed to determine early stage of FI. Methods: Electronic databases were searched up to March 2021. Comparative studies describing the reliability of different clinical and/or radiological furcation assessment methods were identified. Results: A total of 22 studies comparing at least 2 furcation assessment methods, among which 15 retrospective studies, 5 prospective studies, 1 randomized controlled trial and 1 case series, were included in this review. The reliability of cone beam CT (CBCT), intraoral radiographs (IOs), orthopantomograms (OPGs) and MRI to identify FI was evaluated. Using OFS as a reference for FI detection and diagnosis, agreement ranged from 43.3 to 63% for OPG, 38.7 to 83.1% for IO and 82.4 to 84% for CBCT. The validity of the measurements was mainly influenced by the location of the furcation entrance. For radiological diagnosis, CBCT displayed the closest agreement with OFS while the accuracy of IO and OPG showed modest agreement and were influenced by the examiner’s experience. Conclusion: Altogether, it appears that the use of IO, OPG or CBCT allows detection of FI but could not be considered as gold-standard techniques.

Abstract

Background The INNODIA consortium has established a pan-European infrastructure using validated centres to prospectively evaluate clinical data from individuals with newly diagnosed type 1 diabetes combined with centralised collection of clinical samples to determine rates of decline in beta-cell function and identify novel biomarkers, which could be used for future stratification of phase 2 clinical trials. Methods In this context, we have developed a Master Protocol, based on the “backbone” of the INNODIA natural history study, which we believe could improve the delivery of phase 2 studies exploring the use of single or combinations of Investigational Medicinal Products (IMPs), designed to prevent or reverse declines in beta-cell function in individuals with newly diagnosed type 1 diabetes. Although many IMPs have demonstrated potential efficacy in phase 2 studies, few subsequent phase 3 studies have confirmed these benefits. Currently, phase 2 drug development for this indication is limited by poor evaluation of drug dosage and lack of mechanistic data to understand variable responses to the IMPs. Identification of biomarkers which might permit more robust stratification of participants at baseline has been slow. Discussion The Master Protocol provides (1) standardised assessment of efficacy and safety, (2) comparable collection of mechanistic data, (3) the opportunity to include adaptive designs and the use of shared control groups in the evaluation of combination therapies, and (4) benefits of greater understanding of endpoint variation to ensure more robust sample size calculations and future baseline stratification using existing and novel biomarkers.

Abstract

Current practice is to report and manage likely pathogenic/pathogenic variants in a given cancer susceptibility gene as though having equivalent penetrance, despite increasing evidence of intervariant variability in risk associations. Using existing variant interpretation approaches, largely based on full-penetrance models, variants in which reduced penetrance is suspected may be classified inconsistently and/or as variants of uncertain significance. We aimed to develop a national consensus approach for such variants within the Cancer Variant Interpretation Group UK (CanVIG-UK) multidisciplinary network.

Abstract

This paper describes the elastic tire–pavement tractive rolling contact, by means of a semi-analytical method, in order to analyse top-down cracking. The model includes the actual tire profile and assumes the tyre to be elastic, linear and homogeneous, as modelled by an equivalent Young's modulus. The pavement is modelled as a half-space layer composed of asphalt concrete acting as a homogeneous elastic material. Results display the vertical contact stress and shear contact stress in both braking and turning cases; they also highlight the stick/slip zones within the contact area. When applied, these stresses allow computing the tensile strain responses at the pavement surface. Analysis shows that the maximum tensile strains are able to cause top-down cracking. Such strains are higher than the level considered in current pavement design models. A top-down cracking analysis is ultimately conducted according to the French method of bottom-up pavement design. Abstract

Understanding the complex workings of auditory mecha-no-electrical transduction (MET) in the auditory hair cells is essential for comprehending the mechanisms underlying hearing. We present a novel approach integrating single-cell proteomics, affinity-purification mass spectrometry (AP-MS), and AI modeling to identify new components of the lower auditory MET complex, crucial for auditory function. Specifically, we seek to identify new components of the lower tip-link complex via AP-MS with specialized antibodies against the known tip-link component protocadherin 15 (Pcdh15). Protein-protein interactions between potential new and known MET components will be validated using Alphafold2. Simultaneously, we construct an exhaustive proteomic profile of inner hair cells (IHCs) and outer hair cells (OHCs) isolated from the murine cochlea, employing a suction pipette technique for single-cell isolation. Our sample preparation method yields promising results, with over 500 protein groups per single OHC. While proteomic analyses of vestibular hair bundles exist, none focus specifically on IHCs and OHCs. Ultimately, the aim is to conduct a comparative analysis between OHCs and the more difficult-to-iso-late IHCs. Furthermore, we intend to correlate this dataset with established molecular structures in auditory mechanisms and have them serve as complimentary validation for AP-MS experiments with known components of the hearing machinery. These findings will offer critical insights into cochlear mechanosensation, advancing our understanding of auditory biology and providing a framework for innovative therapeutic strategies to address hearing impairments. Abstract

The primary endpoint of our randomized trial was recently reported (Belliveau et al, EAU2024) and demonstrated that intensified salvage radiotherapy guided by PSMA-PET (PSMAiSRT) after radical prostatectomy (RP) resulted in improved failure free survival (FFS). The aim of the present study was to evaluate which subgroups most benefited from PSMAiSRT, evaluate treatment free survival (TFS) outcomes, and determine whether intensification led to heightened toxicity or impact on quality of life (QoL). Between May 2018 and February 2021, there were 128 subjects with biochemical recurrence (BCR) after RP planned for salvage radiotherapy (SRT) were enrolled in the post-RP stratum of a phase II multicenter registry-based randomized controlled trial (RCT-NCT03525288). Patients were randomized 1:1 to standard of care SRT or 18F-DCFPyL PSMA-PET/CT with intensified SRT guided by newly identified disease sites. Secondary endpoints and subgroup analysis were analyzed using cox regression (Hazard Ratio (HR)) and Kaplan-Meier (survival curves and log-rank). Updated toxicity events (CTCAE version 5.0) grade ≥ 2 and minimal important decline (MID) quality of life (QoL) events based on EPIC-CP in 87 patients (n = 49 PSMAiSRT, n = 38 control) were analyzed. As recently published (Petit et al. IJROBP 2023), study arms were balanced with median PSA 0.3 ng/mL (range = 0.1-3) and intensified SRT was delivered in 33 of 64 imaged subjects (n = 16 addition of pelvic SRT, n = 2 addition of metastases-directed RT (MDRT), n = 19 lymph node boost, n = 15 prostate bed boost). The use of adjuvant hormone therapy (HT) was equally high in both arms (86% control vs. 84% PSMAiSRT). At median follow up of 37 months, PSMAiSRT most improved FFS outcomes in patients with PSA ≥ median 0.3 ng/mL (log rank P = 0.01, HR = 0.17, 95% CI = 0.04-0.79) and without adjuvant HT (log rank P = 0.01, HR = 0.009, 95% CI = 0.00-16.20). In secondary endpoint analysis, there was improved TFS with a higher rate of next-line HT events (+/- MDRT) in the control arm (PSMAiSRT 4 events vs. control 12 events, log rank P = 0.04, HR = 0.32, 95% CI = 0.11-1.02). There were no statistically significant differences in toxicity events at median follow up (grade ≥ 2 PSMAiSRT 35 events vs control 39 events, log rank P = 0.716, grade ≥ 3 PSMAiSRT 6 events vs. control 3 events, log rank P = 0.25). Two grade ≥ 3 events were found attributable to intensified RT. No significant differences were found in the rate of MIDs for all QoL domains at 12 and 24 months. (P = NS). The results of this randomized trial demonstrate no significant differences in toxicities or impact in QoL with intensified SRT after RP informed by PSMA-PET/CT, with improved FFS leading to improved TFS. In subgroup analysis, there was a pronounced advantage favoring PSMAiSRT in patients with PSA ≥ median 0.3 ng/mL and without adjuvant HT. Abstract

This thesis analyses the assumptions present in security proofs of post-quantum cryptosystems using Koblitz and Menezes' critique to guide the focus points. The classical analysis focusses in particular on isogeny-based and multivariate polynomial cryptosystems. Firstly, we present attacks against hardness assumptions on derivative computational problems arising in protocols for isogeny-based undeniable signature schemes. Secondly, we provide upper bounds on the solving degree of over-determined systems of n+l multivariate polynomials in n variables as an alternative to the degree of regularity, which gives a heuristic upper bound. We finish with an analysis of the resources of a quantum adversary, providing an improved method for quantum gate approximation that gives shorter sequences by a factor of 9/7.

At the heart of this sits the Brunauer–Emmett–Teller (BET) theory, which has been a remarkably successful contribution to the field of materials science. The BET method was developed in the 1930s for open surfaces but is now the most widely used metric for the estimation of surface areas of microand mesoporous materials. Despite its widespread use, the calculation of BET surface areas causes a spread in reported areas, resulting in reproducibility problems in both academia and industry. To prove this, for this analysis, 18 already-measured raw adsorption isotherms were provided to sixty-one labs, who were asked to calculate the corresponding BET areas. This roundrobin exercise resulted in a wide range of values. Here, the reproducibility of BET area determination from identical isotherms is demonstrated to be a largely ignored issue, raising critical concerns over the reliability of reported BET areas. To solve this major issue, a new computational approach to accurately and systematically determine the BET area of nanoporous materials is developed. Abstract

Aims: This study aimed to compare 12-month metabolic outcomes in patients with type 1 diabetes (T1D) treated with either MiniMed 780G (Guardian 4) or Control-IQ (Dexcom G6) automated insulin delivery (AID) systems and identify interaction with patient characteristics., Materials and Methods: We conducted a single-centre, retrospective study including all patients (aged ≥16) with T1D who were started on either MiniMed 780G or Control-IQ between January 2021 and October 2022 and continued for ≥12 months. We used propensity score matching to compare the average marginal effects between MiniMed 780G and Control-IQ regarding the primary outcome (time in range [TIR]) and secondary outcomes (time below range [TBR], glucose monitoring indicator [GMI] and coefficient of variation [CV]) after 12 months. We tested for interaction effects between baseline characteristics (age, sex, socio-professional background, body mass index, insulin daily dose, carbohydrate counting practice) and treatment effect., Results: We included 245 patients (58% women): 178 treated with Control-IQ and 67 with MiniMed 780G. The mean ± SD age and haemoglobin A1c were 39 ± 15 years and 8.7 ± 1.8% (72 ± 20 mmol/mol) respectively. In the propensity score-matched sample (n = 221), we observed significant differences in 12-month TIR (MiniMed 780G minus Control-IQ [95% CI]: 6.4 [3.4;9.5]), GMI (-0.42 [-0.59; -0.25]) and CV (-2.12 [-3.68; -0.55]), while TBR showed no significant difference (-0.04 [-0.47; +0.40]). The 12-month TIR difference was consistent across subgroups, including baseline carbohydrate counting characteristics., Conclusion: MiniMed 780G is associated with moderate metabolic superiority compared to Control-IQ, without interaction with patient characteristics. These results suggest that neither model is more appropriate for certain populations, particularly patients without carbohydrate counting practice., (© 2024 The Author(s). Abstract

Background: Recurrence of focal segmental glomerulosclerosis (FSGS) is common after kidney transplantation and is classically associated with a significant decrease in graft survival. A major risk factor is a prior history of FSGS recurrence on a previous graft. This analysis reports the impact of a prophylactic treatment of FSGS recurrence in very high-risk patients who experienced a recurrence on a previous graft., Methods: We performed a retrospective multicentre observational study in 25 French transplantation centres. The inclusion criteria were patients aged more than 18 years who had undergone kidney transplant between 31 December 2004 and 31 December 2020, and who had a history of FSGS recurrence on a previous graft., Results: We identified 66 patients: 40 received prophylactic treatment (PT+), including intravenous cyclosporine and/or rituximab and/or plasmapheresis, and 26 did not receive any prophylactic treatment (PT-). The time to progression to end-stage kidney disease was similar between groups. The PT+ group was younger at FSGS diagnosis and at the time of kidney retransplantation and lost their previous graft faster. The overall recurrence rate was 72.7% (76.9% in the PT- group and 70.0% in the PT+ group, P = .54). At least partial remission was achieved in 87.5% of patients. The 5-year graft survival was 67.7% [95% confidence interval (CI) 53.4%-78.4%]: 65.1% (95% CI 48.7%-77.4%) in patients with FSGS recurrence vs 77.3% (95% CI 43.8%-92.3%) in patients without recurrence (P = .48)., Conclusion: Our study suggests that prophylactic treatment should not be used routinely in patients receiving a second transplantation after recurrence of FSGS on a previous graft. The recurrence rate is high regardless of the use of prophylactic treatment. However, the 5-year graft survival remains satisfactory., (© The Author(s) 2024. Abstract

Auditory neuropathies affect the spiral ganglion neurons of the auditory nerve or their synapses with the sensory hair cells, distorting the sound information transmitted from the ear to the brain. Deciphering the underlying pathophysiological mechanisms remains challenging owing to the diversity of spiral ganglion neuron subtypes and associated central auditory circuits. An auditory neuropathy mechanism is unraveled by investigating the origin of auditory hyperexcitability in a mouse model for hereditary congenital deafness. Otogl encodes the large Otogelin-like protein, which is related to secreted epithelial mucins and is implicated in the mechanical stimulation of cochlear outer hair cells. Heterozygous Otogl +/- mutant mice display auditory hyperexcitability, highlighted by their susceptibility to audiogenic seizures induced by loud sounds. It is shown that Otogl is transiently expressed in a subpopulation of spiral ganglion neurons during cochlear development. Despite their apparently normal hearing, Otogl +/- mice display poor activation of the spiral ganglion neurons processing loud sounds and an elevation of the activation threshold of the middle the ear muscle reflex that attenuates loud sounds. The findings reveal how a neuropathy affecting spiral ganglion neurons specialized in loud sound processing and associated with the middle the ear muscle reflex can manifest itself as auditory hyperexcitability., (© 2025 The Author(s).
