- Abstract
Experiencing contact with the world helps to develop a connection, a sense of belonging, care and ecological awareness, generating a desire to experience more sustainable existences (Mortari, 2020). The immersive and experiential encounter with the environment can foster a deep connection with the world in which children are asked to take responsibility towards places (Sobel, 2002): so, it is possible to develop respectful, empathic and sustainable attitudes (Guerra, 2020; 2021) which allow each one to feel part of the planet, and not outsiders of something to be exploited. Ecological education (Mortari, 2018) enhances a sense of deep connection with the world (Chawla, Derr, 2012) through occasions of encounter, knowledge and a desire to care for places, which progressively acquire significance (Judson, 2019). Caring is a practice acquired through experiential learning: in this sense, it appears crucial to promote immersive experiences outdoors where caring is embodied in practices (Mortari, 2020). In light of these considerations, a project by a research group of the Department of Human Sciences for Education of the University of Milano-Bicocca, within the National Biodiversity Future Center, proposes exploratory experiences of the Vivaio Bicocca, an outdoor place in the neighbourhood, neglected for years and recently redeveloped through interventions and cultural initiatives. Among the actions envisaged there are initial proposals and tools for observation, collection, cataloguing and documentation, discussed with a group of researchers from the Department of Biotechnology and Biosciences. These tools, useful for keeping track of questions and hypotheses with narratives, sketches and photographs, making discoveries visible and supporting reflective processes, have been tested with children aged 3-11 and their teachers/educators through a material-reflective experience (Guerra, 2019) that aims to observe, describe, look for relationships and questioning, through an

- Abstract
A literature review of more than 70 articles on Biodiversity Education reveal certain trends that highlight critical issues that educational research may handle. For example, most studies have a strong didactic connotation and education seems to be meant as outreach and dissemination activities. Even if the audience reached by the studies considered is broad (from kids to adults), Biodiversity Education (Saito, 2013) is mostly conceived "one-way" awareness-raising and information initiatives, and less often experiential long-term involvement. The above seems to be at odds with some of the evidence from studies of Ecological education (Mortari, 2020) which shows that immersive and experiential encounter with the environment can foster a deep connection with the world in which individuals are asked to take responsibility towards places. Towards exploration it is possible to develop respectful, empathic and sustainable attitudes which allow each one to feel part of the planet, and not outsiders of something to be exploited: a core attitude of understanding the importance of biodiversity too. In order to fill this gap, a research group of the Department of Human Sciences for Education, in collaboration with the Department of Biotechnology and Biosciences of the University of Milano-Bicocca, designed and implemented a toolkit to promote outdoor explorations aimed at knowing and valuing biodiversity. The poster will present the kit and the first results of the educational activities that have been promoted: a material-reflective experience (Guerra, 2019) that aims to observe, describe, look for relationships and questioning, through an immersive occasion that triggers the desire to learn more.

- Abstract
Alla luce dell'analisi e della letteratura sul tema vengono poi delineate possibili azioni volte a favorire l'avvicinamento ed il reclutamento di professionisti sottorappresentati in questo ambito professionale., Taking as its starting point the feminisation of education and care work, this article examines the phenomenon of the male educators and teacher’s shortage in ECEC, workplaces where gender segregation is extremely pronounced. The article analyses the professional trajectories, narratives and turning points of ECEC professionals. Based on an unpublished part of a larger research study conducted between 2016 and 2022, the paper identifies possible actions to facilitate the outreach and recruitment of underrepresented professionals in the sector.

- Abstract
This paper reflects the application in education of an autocorrect for Italian designed to make it easier to adopt gender-fair language consistently in administrative documents. Sexism in Italian produces important effects in educational contexts as confirmed, for example, by research analyzing school texts. The literature highlights how certain gendered expressions influence our cognition, and how masculine terms evoke masculine images with the effect of excluding, depowering and make invisible women, non-binary and trans people. The analysis of sexism in Italian is thorough and several national and international bodies issued gender-fair language guidelines, that constitute a vast know-that on the subject. However, there is a lack of operational tools to facilitate their implementation, bridging the gap between the know-that and the know-how. This contribution aims to explore, from an interdisciplinary perspective, the pedagogical and training potentialities arising from the use of a gender-fair autocorrect in education. In particular, we argue that the benefit of such a tool is twofold: on the one hand, it produces fair texts and, on the other, it helps its users to develop the ability to recognize and replace sexist expressions.

- Abstract
Ognuna delle ricerche evidenzia alcuni nuclei etici, quali ad esempio la legittimità, i contenuti e i significati condivisi o meno in riferimento all’umorismo e al suo utilizzo nella rappresentazione di chi è (consideratə) ‘altrə’; così come il processo riflessivo di chi fa ricerca; l’esposizione pubblica e la diffusione di prodotti che, nel contribuire alla diffusione di contenuti rilevanti, possono essere importanti testimonianze di esperienze biografiche, in alcuni casi sensibili; il riconoscimento dell’autorialità e della proprietà intellettuale e la problematica dell’archiviazione dei prodotti., Starting from an analysis of different research experiences with people belonging to minoritised or socially vulnerable groups, I intend to explore some ethical issues that have been identified in educational research with young people and adults, particularly in the use of creative methods, from the data collection stage to its dissemination. Humorous writing and its translation into comic strips, photo-voice and finally photo elicitation are the creative methods I will consider. Each research considered highlights ethical cores. First, the legitimacy and shared or unshared meanings of humouring and using it when writing about 'the other'. Second, the reflexive process of doing research. Third, the public dissemination of the results, which, while contributing relevant content, can also be important accounts of biographical experiences that are in some cases sensitive. Fourth, recognition of authorship and ownership, and issues of archiving output.

- Abstract
The aim of this presentation is to address the male shortage in early education and teaching professions, through an interdisciplinary frame. Critical studies on masculinities and literature on role models in ECEC and primary schools are here in dialogue with pedagogical perspectives on hidden curriculum studies and inclusive education theory. My proposal describes a theoretical framework and research project in its early stages aimed at investigate whether a more diverse teaching staff may increase inclusion in education, understood as the never-ending commitment to developing better ways of responding to diversity. Although international literature is quite attentive to minority teachers, the Italian scenario does not look so rich of theory or empirical study. While reframing the male shortage definition in an intersectional framework, including gender identities, but also considering ethnicity, (dis)abilty, religious or cultural backgrounds, I intend to investigate if a diverse teaching staff may challenge the mainstream curricula and pedagogical practices, and increases the level of inclusiveness of schools in a diverse society.

- Abstract
The contribution aims to explore whether and how it is possible to indulge children's on-gender curiosity in a binary and heteronormative society. From a theoretical perspective, the contribution draws on those theoretical approaches that, in questioning gender role models, propose the plurality of adult figures as a means of deconstructing stereotypical gender roles. From a methodological point of view, this is an experimental qualitative research, mainly carried out with a reflexive approach inspired to autoethnography. Autoethnographic events are considered ex-post in order to highlight both some rising problems for a non binary gender education, and related countermeasures, and the difficulty of making synthesis between my professional expertise on the subject, the existing literature and the sense of displacement when not discomfort in proposing a non-binary educational approach to gender in a society expressly so.

- Abstract

Within the European project Story_S, supporting successful educational paths for Roma youth through peer to peer education, a participatory anti-discrimination campaign was built with young people of different backgrounds, in Milan and Rome. An intersectional approach is proposed to reflect together with young students on the topic of discrimination and to build, through the photovoice, a photographic campaign addressed to their peers. The photovoice process fostered group dynamics, reflection, and a photographic work, during two Awareness Raising Workshops of 3 days each. Discrimination is recognized and experienced by the youth on grounds of gender and sexuality, race and ethnicity, in definitions of normality, poverty, online, offline and in the media. Metaphors of coexistence or of the beauty of diversity were represented through photos; also experiences of discrimination regarding housing conditions and public services were captured. Strategies to combat discrimination have been proposed and discussed by participants. Through the online and offline dissemination of the campaign, counter narratives were promoted and reached wider audiences, in an attempt to support anti-discrimination cultures and empowerment in schools and youth environments. By discussing both the process and the results of the photovoice, we contribute to the understanding of young people’s perspective on discrimination and on participation to social change.

- Abstract
Reliable predictions of the aero- and hydrodynamic loads of fixed-bottom and floating offshore wind turbines are paramount for assessing fatigue life and designing load and power control systems. However, significant uncertainty affecting aerodynamic predictions still exists. This study presents cross-comparative analyses of the predictions of aerodynamic loads and power of fixed-foundation and floating wind turbine rotors with and without yaw errors using time- and frequency-domain Navier-Stokes Computational Fluid Dynamics, and the Blade Element Momentum theory. The considered test case is the National Renewable Energy Laboratory 5 MW reference turbine, assumed to be mounted in the floating case on a semi-submersible platform and undergoing pitching motion about the tower base. Although the overall qualitative agreement of the low- and high-fidelity predictions is found to be fair in all cases, for the considered regimes the agreement between the two methods is better for the pitching rotor in aligned wind than for the yawed flows regardless of the tower motion.

Frailty affects the physical, cognitive, and social domains exposing older adults to an increased risk of cardiovascular disease and death. The mechanisms linking frailty and cardiovascular outcomes are mostly unknown. Here, we studied the association of abundance (flow cytometry) and gene expression profile (RNAseq) of stem/progenitor cells (HSPCs) and molecular markers of inflammaging (ELISA) with the cardiorespiratory phenotype and prospective adverse events of individuals classified according to levels of frailty. Two cohorts of older adults were enrolled in the study. In a cohort of pre-frail 35 individuals (average age: 75 years), a physical frailty score above the median identified subjects with initial alterations in cardiorespiratory function. RNA sequencing revealed S100A8/A9 upregulation in HSPCs from the bone marrow (>10-fold) and peripheral blood (>200-fold) of individuals with greater physical frailty. Moreover higher frailty was associated with increased alarmins S100A8/A9 and inflammatory cytokines in peripheral blood. We then studied a cohort of 104 more frail individuals (average age: 81 years) with multidomain health deficits. Reduced levels of circulating HSPCs and increased S100A8/A9 concentrations were independently associated with the frailty index. Remarkably, low HSPCs and high S100A8/A9 simultaneously predicted major adverse cardiovascular events at 1-year follow-up after adjustment for age and frailty index. In conclusion, inflammaging characterized by alarmin and pro-inflammatory cytokines in pre-frail individuals is mirrored by the pauperization of HSPCs in frail older people with comorbidities. S100A8/A9 is upregulated within HSPCs, identifying a phenotype that associates with poor cardiovascular outcomes.

Background: In the Mediterranean basin, three Leishmania species have been identified: L. infantum, L. major and L. tropica, causing zoonotic visceral leishmaniasis (VL), zoonotic cutaneous leishmaniasis (CL) and anthroponotic CL, respectively. Despite animal models and genomic/transcriptomic studies provided important insights, the pathogenic determinants modulating the development of VL and CL are still poorly understood. This work aimed to identify host transcriptional signatures shared by cells infected with L. infantum, L. major, and L. tropica, as well as specific transcriptional signatures elicited by parasites causing VL (i.e., L. infantum) and parasites involved in CL (i.e., L. major, L. tropica)., Methodology/principal Findings: U937 cells differentiated into macrophage-like cells were infected with L. infantum, L. major and L. tropica for 24h and 48h, and total RNA was extracted. RNA sequencing, performed on an Illumina NovaSeq 6000 platform, was used to evaluate the transcriptional signatures of infected cells with respect to non-infected cells at both time points. The EdgeR package was used to identify differentially expressed genes (fold change > 2 and FDR-adjusted p-values < 0.05). Then, functional enrichment analysis was employed to identify the enriched ontology terms in which these genes are involved. At 24h post-infection, a common signature of 463 dysregulated genes shared among all infection conditions was recognized, while at 48h post-infection the common signature was reduced to 120 genes. Aside from a common transcriptional response, we evidenced different upregulated functional pathways characterizing L. infantum-infected cells, such as VEGFA-VEGFR2 and NFE2L2-related pathways, indicating vascular remodeling and reduction of oxidative stress as potentially important factors for visceralization., Conclusions: The identification of pathways elicited by parasites causing VL or CL could lead to new therapeutic strategies for leishmaniasis, combining the canonical anti-leishmania compounds with host-directed therapy., Competing Interests: The authors have declared that no competing interests exist., (Copyright: © 2024 Diotallevi et al. - Abstract
The Ketogenic Diet (KD) improves memory and longevity in aged C57BL/6 mice. We tested 7 months KD vs. control diet (CD) in the mouse Alzheimer's Disease (AD) model APP/PS1. KD significantly rescued Long-Term-Potentiation (LTP) to wild-type levels, not by changing Amyloid-β (Aβ) levels. KD's 'main actor' is thought to be Beta-Hydroxy-butyrate (BHB) whose levels rose significantly in KD vs. CD mice, and BHB itself significantly rescued LTP in APP/PS1 hippocampi. KD's 6 most significant pathways induced in brains by RNAseq all related to Synaptic Plasticity. KD induced significant increases in synaptic plasticity enzymes p-ERK and p-CREB in both sexes, and of brain-derived neurotrophic factor (BDNF) in APP/PS1 females. We suggest KD rescues LTP through BHB's enhancement of synaptic plasticity. LTP falls in Mild-Cognitive Impairment (MCI) of human AD. KD and BHB, because they are an approved diet and supplement respectively, may be most therapeutically and translationally relevant to the MCI phase of Alzheimer's Disease., (© 2024. The Author(s).)

- Abstract
The histone deacetylase sirtuin 6 (SIRT6) has been endowed with anti-cancer capabilities in many tumor types. Here, we investigate the impact of SIRT6-overexpression (SIRT6-OE) in Delta16HER2 mice, which are a bona fide model of HER2-positive breast cancer. After an initial delay in the tumor onset, SIRT6-OE induces a more aggressive phenotype of Delta16HER2 tumors promoting the formation of higher number of tumor foci and metastases than controls. This phenotype of SIRT6-OE tumors is associated with cancer stem cell (CSC)-like features and tumor dormancy, and low senescence and oxidative DNA damage. Accordingly, a sub-set of HER2-positive breast cancer patients with concurrent SIRT6-OE has a significant poorer relapse-free survival (RFS) probability than patients with low expression of SIRT6. ChIP-seq, RNA-seq and RT-PCR experiments indicate that SIRT6-OE represses the expression of the T-box transcription factor 3 (Tbx3) by deacetylation of H3K9ac. Accordingly, loss-of-function mutations of TBX3 or low TBX3 expression levels are predictive of poor prognosis in HER2-positive breast cancer patients. Our work indicates that high levels of SIRT6 are indicative of poor prognosis and high risk of metastasis in HER2-positive breast cancer and suggests further investigation of TBX3 as a downstream target of SIRT6 and co-marker of poor-prognosis. Our results point to a breast cancer subtype-specific effect of SIRT6 and warrant future studies dissecting the mechanisms of SIRT6 regulation in different breast cancer subtypes., (© 2023. - Abstract
Acute promyelocytic leukemia (APL) is an aggressive subtype of acute myeloid leukemia (AML) in which the PML/RARα fusion protein exerts oncogenic activities by recruiting repressive complexes to the promoter of specific target genes. Other epigenetic perturbations, as alterations of histone H3 lysine 9 trimethylation (H3K9me3), have been frequently found in AMLs and are associated with leukemogenesis and leukemia progression. Here, we characterized the epigenomic effects of maltonis, a novel maltol-derived molecule, in APL cells. We demonstrate that maltonis treatments induce a profound remodulation of the histone code, reducing global H3K9me3 signal and modulating other histone post-translational modifications. Transcriptomic and epigenomic analyses revealed that maltonis exposure induces changes of genes expression associated with a genomic redistribution of histone H3 lysine 4 trimethylation (H3K4me3) and lysine 27 acetylation (H3K27ac). Upregulation of interferon alpha and gamma response and downregulation of c-MYC target genes, in function of c-MYC reduced expression (monitored in all the hematopoietic neoplasms tested), represent the most significant modulated pathways. These data demonstrate the ability of maltonis to epigenetically reprogram the gene expression profile of APL cells, inducing an intriguing antiviral-like response, concomitantly with the downregulation of c-MYC-related pathways, thus making it an attractive candidate for antileukemic therapy., (© 2022. - Abstract
Objectives: The aim of the present randomized clinical trial (RCT) with a parallel arm design was to evaluate the clinical and microbiological efficacy of repeated ICG-aPDT as an adjunct to full-mouth subgingival debridement in the treatment of periodontitis., Materials and Methods: Twenty-four periodontitis patients were treated with full-mouth ultrasonic subgingival debridement (FMUD). Initial sites with probing depth (PD) > 4 mm were randomly assigned to receive the test (ICG-aPDT with an 810 nm diode laser) or the control treatment (off-mode aPDT) one and four weeks after FMUD. Clinical parameters were registered after 3 and 6 months. The presence of the main periodontal pathogens in subgingival samples was assessed with real-time PCR., Results: Both treatment modalities resulted in significant clinical improvements at 3 and 6 months. The only significant differences in favour of the test group were found at 6 months for a higher PD reduction in initial deep pockets (PD ≥ 6 mm) and a higher percentage of closed pockets (PD ≤ 4 mm/no bleeding on probing). Limited microbiological changes were observed in both groups after treatment with no inter-group difference, except for a more significant reduction in Aggregatibacter actinomycetemcomitans and Parvimonas micra levels in the test group at 3 months., Conclusion: The combination of repeated ICG-aPDT and FMUD provided no benefits except for selective clinical and microbiological improvements compared to FMUD alone., Clinical Relevance: Based on the obtained results, only limited adjunctive effects could be found for the combined use of ICG-aPDT and FMUD. Further, well-designed RCT with larger sample sizes are required to confirm these findings., Trial Registration: ClinicalTrials.gov NCT04671394., (© 2023. The Author(s).)

- Abstract
Human amniotic fluid cells (hAFSCs) are a fascinating foetal cell-type that have important stem cell characteristics; however, they are a heterogeneous population that ranges from totally differentiated or progenitor cells to highly multipotent stem cells. There is no single approach to isolating the stem cell component, but the selection of a subpopulation of hAFSCs expressing c-Kit is widely employed, while a deep characterization of the two populations is still lacking. Here we performed single-cell and bulk RNAseq analysis to compare the gene expression profiles of adherent amniotic fluid cells and their subpopulation c-Kit
+ . Information deriving from this high throughput technology on the transcriptome was then confirmed for specific targets with protein expression experiments and functional analysis. In particular, transcriptome profiling identified changes in cellular distribution among the different clusters that correlated with significant differential expression in pathways related to stemness, proliferation, and cell cycle checkpoints. These differences were validated by RT-PCR, immunofluorescence, WB, and cell cycle assays. Interestingly, the two populations produced secretomes with different immune-modulating and pro-regenerative potentials. Indeed, the presence of TGFβ, HGF, IDO was higher in EVs deriving from c-Kit+ cells, unlike IL-6. These results suggest the existence of deep intra-population differences that can influence the stemness profile of hAFSCs. This study represents a proof-of-concept of the importance of selecting c-Kit positive fractions with higher potential in regenerative medicine applications.- Published

- Abstract
Background: Early-life stress due to poor parental care has been suggested to increase cancer risk, though, so far, no experimental evidence established a link between defective parental behavior and spontaneous tumorigenesis in progeny. Essential maternal behavior is regulated, in particular, by the oxytocin (OT) hormonal circuit, which in turn responds to stimuli from the offspring and impinges on the central nervous systems., Methods: By providing L-368,899 OT receptor (OTR) inhibitor to lactating mothers, we set up a model of defective maternal care in p53 knockout mice., Results: The progeny of these dams showed, later in life, higher cortisol levels, shortened life span and increased tumorigenic potential of bone marrow cells (BMC). Notably, these phenotypes were transmitted to the following generation., Conclusions: Therefore, the inhibition of OT function in mothers is a novel paradigm of early-life stress that is inherited across generations and increases cancer risk in tumor-prone mice., (© 2022 The Authors. - Abstract
To detect the epigenetic drift of time passing, we determined the genome-wide distributions of mono- and tri-methylated lysine 4 and acetylated and tri-methylated lysine 27 of histone H3 in the livers of healthy 3, 6 and 12 months old C57BL/6 mice. The comparison of different age profiles of histone H3 marks revealed global redistribution of histone H3 modifications with time, in particular in intergenic regions and near transcription start sites, as well as altered correlation between the profiles of different histone modifications. Moreover, feeding mice with caloric restriction diet, a treatment known to retard aging, reduced the extent of changes occurring during the first year of life in these genomic regions.

- Abstract
The history of the modern Italian gynecology is closely related to the figure of an enlightened clinician and passionate master, Luigi Mangiagalli (1850-1928), who, with good reason, can be considered one of the fathers of this discipline. In 1906, Mangiagalli founded the Obstetric-Gynecological Institute in Milan, directing this structure until 1925. The aim of this paper is to describe some aspects of Italian gynecology at the beginnings of the twentieth century through the analysis of 250 original medical records, dating back to the years 1906-1912, fragments of real life of women, midwives and doctors in a pioneering era of obstetrics and gynecology.

- Abstract
S100A8/A9 is upregulated within HSPCs, identifying a phenotype that associates with poor cardiovascular outcomes., (© 2022 The Authors. Background: Epigenetic remodeling is emerging as a critical process for both the onset and progression of Alzheimer's disease (AD), the most common form of neurodegenerative dementia. However, it is not clear to what extent the distribution of histone modifications is involved in AD., Methods: To investigate histone H3 modifications in AD, we compared the genome-wide distributions of H3K4me3 and H3K27me3 in entorhinal cortices from severe sporadic AD patients and from age-matched healthy individuals of both sexes., Results: AD samples were characterized by typical average levels and distributions of the H3K4me3 and H3K27me3 signals. However, AD patients showed a lower H3K4me3 and higher H3K27me3 signal, particularly in males. Interestingly, the genomic sites found differentially trimethylated at the H3K4 between healthy and AD samples involve promoter regions of genes belonging to AD-related pathways such as glutamate receptor signaling., Conclusions: The signatures of H3K4me3 and H3K27me3 identified in AD patients validate the role of epigenetic chromatin remodeling in neurodegenerative disease and shed light on the genomic adaptive mechanisms involved in AD.

Background: Women represent the majority of Alzheimer's disease patients and show typical symptoms. Genetic, hormonal, and behavioral mechanisms have been proposed to explain sex differences in dementia prevalence. However, whether sex differences exist in the epigenetic landscape of neuronal tissue during the progression of the disease is still unknown., Methods: To investigate the differences of histone H3 modifications involved in transcription, we determined the genome-wide profiles of H3K4me3, H3K27ac, and H3K27me3 in brain cortexes of an Alzheimer mouse model (PSAPP). Gastrocnemius muscles were also tested since they are known to be different in the two sexes and are affected during the disease progression., Results: Correlation analysis distinguished the samples based on sex for H3K4me3 and H3K27me3 but not for H3K27ac. The analysis of transcription starting sites (TSS) signal distribution, and analysis of bounding sites revealed that gastrocnemius is more influenced than brain by sex for the three histone modifications considered, exception made for H3K27me3 distribution on the X chromosome which showed sex-related differences in promoters belonging to behavior and cellular or neuronal spheres in mice cortexes., Conclusions: H3K4me3, H3K27ac, and H3K27me3 signals are slightly affected by sex in brain, with the exception of H3K27me3, while a higher number of differences can be found in gastrocnemius.
