Identifying the collection sites for the wild seeds of a highly valued species is crucial for the conservation and management of its wild population and habitat. This study is the first to document the distribution and the characteristics of spiny lobster puerulus collection sites in Palawan, Philippines. The data were obtained from various key informants, site visits, and historical imagery from Google Earth Pro and Sentinel 2A of Copernicus between January 2021-December 2023. The data were loaded into the QGIS v3.28 software to produce a map of each locality projecting the distribution and characteristics of spiny lobster puerulus collection sites. The results showed that the collection began in the municipality of Sofronio Española in 2019 and eventually spread southward and northward along the east coast of Palawan. The municipalities on the east coast of Palawan such as Sofronio Española, Narra, and Roxas had the highest number of collection sites, whereas fewer on the west coast such as in Rizal. Most of the collection sites were located generally along the intertidal zones or embayment with sandy-muddy to rubble substrates adjacent to mangrove forests, or within seagrass or seaweed, and coral reef ecosystems. To conserve and manage the spiny lobster puerulus fishery, the revision of the provincial ordinance on spiny lobster puerulus collection in Palawan should include the proper zoning of collection sites, imposing catch limits, and annual catch monitoring. A Caribbean spiny lobster, Panulirus argus, was submitted for necropsy after a number of species-specific fatalities in a public aquarium. The hemolymph was opaque and did not clot and the hepatopancreas had multiple foci of necrosis centered on gram-negative bacteria. Pure cultures of Vibrio harveyi were isolated, identified initially by matrix laser desorption ionization-time of flight mass spectrometry, and confirmed by multilocus sequencing of the gyrB, recA, rpoA, and pyrH genes. As Caribbean spiny lobsters continue to be used for consumption and displayed in public aquariums, chronicling potential pathogens is warranted to inform differential diagnoses and to develop management strategies to reduce incidence of infectious disease in captive populations. Abstract Palinurid rock (or spiny) lobsters constitute valuable seafood fisheries around the world. Post‐harvest mortality can have a substantial economic impact on the value of these fisheries and damage the brand identity of an otherwise fungible commodity. In this study, sudden, large‐scale mortalities in the Australian Rock Lobster industry were investigated through comprehensive haemolymph biochemistry panels, measurements of immune status and nutritional condition. A cohort of lobsters (n = 25) was sampled over a 4‐week period at a commercial processing facility, with the final sample occurring during a mortality event. A comparison of 22 biochemical parameters, including electrolytes, minerals, ions, metabolites and enzymes, showed a notable disruption in osmoregulation, protein content and enzymes during weeks 3 and 4 of the study. Haemocyte counts showed a decline in circulating haemocytes by the time of the mortality event along with a shift in the proportion of hyalinocytes to granulocytes favouring the former. The condition index was variable over the sampling schedule, possibly linked to observed incidences of cannibalism, with no evidence of an overall low condition that would explain mortalities. The lobsters sampled in this study did not show any consistent or progressive changes in biochemistry that may indicate a pathogen or disease as a causative agent. Rather, analysis of the water quality within the facility suggested that mortality was due to sub‐optimal water quality characterized by a combination of two factors, a substantial decrease in the pH and potential nitrogen toxicity, that caused a physiological response comprised of an ion imbalance, altered enzyme activity and, ultimately, mortality. These results offer a comprehensive characterization of the impact of unexplained morbidity on the biochemistry and physiology of lobsters, illustrate the harm of sub‐optimal aquaculture practices and highlight the need for a better understanding of aquaculture practices and their effects on economically important seafood industries.

Este trabajo pretende actualizar la lista de especies de langostas espinosas para el Archipiélago de Abrolhos. Los datos se obtuvieron a través de censos visuales de buceo (SCUBA) durante el día (8 am a 5 pm) y la noche (7 pm a 11 pm), a profundidades de 2 a 12 metros. Este estudio presenta el primer registro de Panulirus laevicauda en el archipiélago de Abrolhos, confirma la presencia de Panulirus echinatus y refuerza la ocurrencia de Panulirus argus. Todas las especies de langostas del género Panulirus registradas en la costa brasileña están presentes en el Parque Nacional Marinho dos Abrolhos, lo que refuerza la importancia de esta área marina protegida.

Casitas, low-lying artificial shelters that mimic large crevices, are used in some fisheries for Caribbean spiny lobsters (Panulirus argus). These lobsters are highly gregarious and express communal defense of the shelter. Scaled-down casitas have been shown to increase survival, persistence, and foraging ranges of juveniles. Therefore, the use of casitas has been suggested to help enhance local populations of juvenile P. argus in Caribbean seagrass habitats, poor in natural crevice shelters, in marine protected areas. Following the emergence of Panulirus argus virus 1 (PaV1), which is lethal to juveniles of P. argus, concern was raised about the potential increase in PaV1 transmission with the use of casitas. It was then discovered that lobsters tend to avoid shelters harboring diseased conspecifics, a behavior which, alone or in conjunction with predatory culling of diseased lobsters, has been proposed as a mechanism reducing the spread of PaV1. However, this behavior may depend on the ecological context (i.e., availability of alternative shelter and immediacy of predation risk). We conducted an experiment in a lobster nursery area to examine the effect of the use of casitas on the dynamics of the PaV1 disease. We deployed 10 scaled-down casitas per site on five 1-ha sites over a reef lagoon (casita sites) and left five additional sites with no casitas (control sites). All sites were sampled 10 times every 3–4 months. Within each site, all lobsters found were counted, measured, and examined for clinical signs of the PaV1 disease. Mean density and size of lobsters significantly increased on casita sites relative to control sites, but overall prevalence levels remained similar. There was no relationship between lobster density and disease prevalence. Dispersion parameters (m and k of the negative binomial distribution) revealed that lobsters tended to avoid sharing natural crevices, but not casitas, with diseased conspecifics. These results confirm that casitas provide much needed shelter in seagrass habitats and that their large refuge area may allow distancing between healthy and diseased lobsters. On eight additional sampling times over two years, we culled all diseased lobsters observed on casita sites. During this period, disease prevalence did not decrease but rather increased and varied with site, suggesting that other factors (e.g., environmental) may be influencing the disease dynamics. Using scaled-down casitas in shelter-poor habitats may help efforts to enhance juvenile lobsters for conservation purposes, but monitoring PaV1 prevalence at least once a year during the first few years would be advisable.

The attractive price for spiny lobster (Panulirus spp.) puerulus has recently encouraged many fishers to engage in its collection as an alternative livelihood at the height of the COVID-19 pandemic. This booming fishery prompted the investigation of the settlement traps, luring/shading materials, trap installation and harvesting methods, and problems encountered in the spiny lobster puerulus fishery in Palawan, Philippines. Reviewed videos posted on social media, personal observation, and data gathered from key informants (KIs) revealed the use of various modified and newly developed settlement traps. The most common were the concrete cylindrical and disc-shaped traps used by 63.33% of the KIs. Some KIs (40%) provided the settlement traps with luring/shading materials like Sargassum thalli. Settlement traps were mostly installed using the stake-hanging method. The harvesting season starts in March and ends in August where the daily collection coincides with the moon phase. Fishers collected settled puerulus through diving or pulling the traps out of the water. However, some issues like unstable prices and the absence of fishing regulations threaten the sustainability of pueruli collection. Hence, the establishment of collection zones, issuance of fishing permits, price regulation, and long-term monitoring to maintain this promising fishery industry is suggested in crafting ordinances. The Japanese spiny lobster Panulirus japonicus lives on rocky shores and is mainly distributed along the Pacific coast around Japan. Due to the high demand for it, the development of aquaculture systems and increasing its resource volume requires further expansive production. However, a major factor preventing the establishment of aquaculture technology for this lobster is the difficulty with rearing processes from larval to juvenile production. A recent study shed light on the molecular mechanisms underlying larval development from the perspective of physiological functions of endocrine factors such as molting hormones. However, physiological studies of P. japonicus are still lacking. In decapod crustaceans, the X-organ/sinus gland complex is a well-known endocrine system that secretes the crustacean hyperglycemic hormone (CHH)-superfamily peptides that regulate growth, molting, sexual maturation, reproduction, and change in body color. In this study, we identified two CHHs from the sinus glands of P. japonicus using reversed-phase high-performance liquid chromatography in order to elucidate their physiological function for the first time.

A network of marine reserves can enhance yield in depleted fisheries by protecting populations, particularly large, old spawners that supply larvae for interspersed fishing grounds. The ability of marine reserves to enhance sustainable fisheries is much less evident. We report empirical evidence of a marine reserve network improving yield regionally for a sustainable spiny lobster fishery, apparently through the spillover of adult lobsters and behavioral adaptation by the fishing fleet. Results of a Before-After, Control-Impact analysis found catch, effort, and Catch-Per-Unit Effort increased after the establishment of marine reserves in the northern region of the fishery where fishers responded by fishing intensively at reserve borders, but declined in the southern region where they vacated once productive fishing grounds. The adaptation of the northern region of the fishery may have been aided by a history of collaboration between fishers, scientists, and managers, highlighting the value of collaborative research and education programs for preparing fisheries to operate productively within a seascape that includes a large marine reserve network., (© 2024. The hydroxyapatite (HAp; Ca 10 (PO 4 ) 6 (OH) 2 )) has good biocompatibility, bioactivity, and osteoconductivity as a bone implant because the main inorganic mineral of human bone is HAp. The use of scaffold HAp from biogenic resources that contain high calcium and polymer as a pore forming agent to support bone growth is a longstanding area of interest. In this study, porous scaffolds based on HAp were synthesized from sand lobster (SL; Panulirus homarus) shells as a source of calcium using the porogen leaching method with polyethylene oxide (PEO) and chitosan (Chs) as polymeric porogen. The present study aims to synthesize HAp derived from SL shells and evaluate the effect variations of PEO on the physicochemical properties of the scaffold and cytotoxicity in cell viability assay. Briefly, the SL shell powder was calcinated with temperature variations of 600°C, 800°C, and 1000°C for 6 h. Based on the characterization, it was shown that 1000°C was the optimum calcination temperature for SL shells to synthesize HAp using the precipitation method. The characterization results of HAp using energy dispersive x-ray (EDX) revealed that the molar ratio of Ca/P was 1.67. The Fourier transform infrared (FTIR) and x-ray diffractometer (XRD) spectral patterns indicated that HAp had been successfully synthesized with minor β-tricalcium phosphate (β-TCP), a calcium phosphate with high biocompatibility. Porous scaffolds were synthesized by varying the concentration of PEO at 0, 5, 10, and 15 wt %. Physicochemical analysis revealed that a higher concentration of PEO affected decreased crystallinity and compressive strength, but on the other hand, the porosity and pore sizes increased. Based on the physicochemical analysis, the synthesized porous scaffold showed that HAp/PEO/Chs 15 wt % had the most potential as a scaffold for biomedical applications. MTT Assay, after 24 h incubation, revealed that the scaffold was safe for use at low concentrations on the MC3T3E1 osteoblast cells, with a percentage of cell viability of 83.23 ± 3.18% at 23.4375 μg/mL. Although the cell viability decreased at higher concentrations, the HAp/PEO/Chs 15 wt % scaffold was cytocompatible with the cells. Lobster seacage aquaculture has a significant social-economic contribution to many coastal communities throughout the central provinces of Vietnam. One of the issues of lobster aquaculture is that the industry entirely relies on the wild juvenile lobster (lobster seed) captured using traps made of coral stones, which affects the coastal ecology and environment. The present study was carried out to compare the catch efficiency of traps made of round timber poles (wooden trap) with conventional dead corals (coral stones) during the juvenile lobster fishing season in 2019 in the coastal area of Nha Trang city, Vietnam. Results showed that experimental wooden traps maintained the conventional catch rate level, with a significant proportion of Panulirus ornatus caught at larger carapace length than those caught by the coral stone traps. Many digenean trematodes require three hosts to complete their life cycle. For Cymatocarpus solearis (Brachycoeliidae), the first intermediate host is unknown; the Caribbean spiny lobster Panulirus argus is a second intermediate host, and the loggerhead turtle Caretta caretta, a lobster predator, is the definitive host. Trophically-transmitted parasites may alter the behavior or general condition of intermediate hosts in ways that increase the hosts' rates of consumption by definitive hosts. Here, we examined the effects of infection by C. solearis on P. argus by comparing several physiological and behavioral variables among uninfected lobsters (0 cysts) and lobsters with light (1-10 cysts), moderate (11-30 cysts), and heavy (>30 cysts) infections. Physiological variables were hepatosomatic index, growth rate, hemocyte count, concentration in hemolymph of cholesterol, protein, albumin, glucose, dopamine (DA) and serotonin (5-HT). Behavioral variables included seven components of the escape response (delay to escape, duration of swimming bout, distance traveled in a swimming bout, swim velocity, acceleration, force exerted, and work performed while swimming). There was no relationship between lobster size or sex and number of cysts. Significant differences among the four lobster groups occurred only in concentration of glucose (lower in heavily infected lobsters) and 5-HT (higher in heavily and moderately infected lobsters) in plasma. Panulirus argus virus 1 (PaV1) is the first and only naturally occurring pathogenic virus described in the Caribbean spiny lobster, Panulirus argus . PaV1 infection in decapod species that commonly co-occur with P. argus , including the spotted spiny lobster Panulirus guttatus , has not been previously described. In 2016, 14 Caribbean and 5 spotted spiny lobsters were collected near Summerland Key, Florida, to supplement the resident population of the Audubon Aquarium of the Americas in New Orleans, Louisiana. After 5 months in quarantine, Caribbean and spotted spiny lobsters began to exhibit clinical signs of lethargy and dying in the molt. Initial histologic evaluation revealed intranuclear inclusion bodies in circulating hemocytes in the spongy connective tissue of the epidermis, suggesting a viral infection. Samples of hepatopancreas and hemolymph from deceased Caribbean and spotted spiny lobsters tested negative for white spot syndrome virus and positive for PaV1 using real-time quantitative polymerase chain reaction (qPCR). Intranuclear, eosinophilic to amphophilic, Cowdry type A inclusion bodies observed primarily within fixed phagocytes and circulating hemocytes in the hepatopancreas of freshly euthanized Caribbean spiny lobsters were consistent with PaV1 infection. Transmission electron microscopy revealed that hemocytes associated with hepatopancreatic tubules contained viral inclusions with location, size, and morphology consistent with previously described PaV1 infection. These findings highlight the significance of using molecular diagnostics in conjunction with histopathology and electron microscopy in the investigation and diagnosis of PaV1 in spiny lobsters. Further study is required to investigate the relationship of PaV1-associated mortality events and microscopic lesions in the spotted spiny lobster.

Crustaceans exhibit discontinuous growth as they shed hard shells periodically. Fundamentally, the growth of crustaceans is typically assessed through two key components, length increase after molting (LI) and time intervals between consecutive molts (TI). In this article, we propose a unified likelihood approach that combines a generalized additive model and a Cox proportional hazard model to estimate the parameters of LI and TI separately in crustaceans. This approach captures the observed discontinuity in individuals, providing a comprehensive understanding of crustacean growth patterns. Our study focuses on 75 ornate rock lobsters ( Panulirus ornatus ) off the Torres Strait in northeastern Australia. Through a simulation study, we demonstrate the effectiveness of the proposed models in characterizing the discontinuity with a continuous growth curve at the population level.

The present work compares microplastics (MPs) contamination in two charismatic crustaceans: European spiny lobster Palinurus elephas and langoustine Nephrops norvegicus. Samples (P. elephas n = 14; N. norvegicus n = 15) were collected between 76 and 592 m depth, from four sites in west Sardinia, Italy. An extraction protocol was applied on stomachs and intestines, separately, and over 500 particles were further characterized through μFT-IR. We document 100 % occurrence in specimens from both species, with P. elephas being significantly more contaminated (9.1 ± 1.75 vs. 3.2 ± 0.45 MPs individual -1 ), ingesting larger MPs with different polymeric composition. The scavenging-based feeding strategy of both species could explain such exposure to MPs, mostly derived by single-use plastic. The ornate spiny rock lobster, Panulirus ornatus, is an attractive candidate for aquaculture. The larval stages of spiny lobsters, known as phyllosoma, are complex with many developmental stages. Very little is known about the inorganic element composition of phyllosoma. In this study, a novel method using synchrotron X-ray fluorescence microscopy (XFM) was applied to investigate the distributions of metals potassium (K), calcium (Ca), copper (Cu), zinc (Zn), the metalloid arsenic (As), and nonmetal bromine (Br) within individual phyllosoma at stages 3, 4, and 8 of their development. For the first time, 1 µm resolution synchrotron XFM images of whole phyllosoma as well as closer examinations of their eyes, mouths, setae, and tails were obtained. Elements accumulated in certain locations within phyllosoma, providing insight into their likely biological role for these organisms. Marine crustaceans produce broadband sounds that are useful for passive acoustic monitoring to support conservation and management efforts. However, the propagation characteristics and detection ranges of their signals are poorly known, limiting our leveraging of these sounds. Here, we used a four-hydrophone linear array to measure source levels (SLs) and sound propagation from Caribbean spiny lobsters (Panulirus argus) of a wide range of sizes within a natural, shallow water habitat (3.3 m depth). Source level in peak-peak (SLpp) varied with body size; larger individuals produced SLpp up to 166 dB re 1 μPa. However, transmission losses (TL) were similar across all sizes, with a global fitted TL of 12.1 dB. Correspondingly, calculated detection ranges varied with body size, ranging between 14 and 364 m for small and large individuals (respectively). This increased up to 1612 m for large spiny lobsters when considering lower ambient noise levels. Despite the potential ease of tank studies, our results highlight the importance of empirical in situ sound propagation studies for marine crustaceans. Given the important ecological and economic role of spiny lobsters, these data are a key step to supporting remote monitoring of this species for fisheries management and efforts to acoustically quantify coral reefs' health.

European marine waters are infested with plastic, with an average density of 1 plastic item in every four square-meters. Research relevant to MPs-NPs ingestion by wild decapods in European waters is limited, none of which regards the European spiny lobster. Totally 4102 plastic particles were extracted from the spiny lobster stomach tissues of both sexes. Out of the 63 samples analysed only three (4.8%) of them were found with no plastic particles. The range of number of MPs in stomachs was from 20 to 273 MPs individual -1 . The 98.3% were fragments. In total 3833 plastic particles were extracted from the gill tissues of both sexes. MPs were found in all samples (n = 50),99.2% of the detected particles were fragments. The MPs detected in gills ranged from 11 to 339 MPs individual -1 . The DLS method was used in order to evaluate the NPs presence. Nanoplastics were detected in 22.6% of stomachs and in the 48.1% of gills. A total of 43 polymer types were identified in both tissues. Also, our study assessed the accumulation of heavy metals at the edible tail muscle. Certain elements were detected above the EU's Maximum Residue Level, including arsenic. Stage I phyllosoma larvae of the spiny lobster Panulirus ornatus hatch in tropical oceanic waters with limited and variable food resources. To better understand how these larvae cope with food deprivation, this study examined specific (mU mg−1 protein) and total (mU larva−1) activities of major digestive enzymes (i.e., α-amylase, non-specific esterase, trypsin-like protease) during a series of ex situ experiments. Specifically, temporal changes in enzyme profiles were examined in three experiments simulating scenarios in which phyllosoma hatch and begin development in an environment where (1) prey is either continuously present or absent; (2) prey is initially present, but subsequently absent; (3) prey is initially absent, but subsequently present. Results indicated that the accessibility of suitable prey provides an overarching influence on digestive enzyme activities and substrate utilisation in Stage I phyllosoma of P. ornatus , with enzymatic responses to both intermittent prey availability and food deprivation being influenced by their nutritional history. In the absence of prey, larval digestive enzyme activities (mU larva−1) initially remained static from hatch but eventually declined with extended food deprivation. When prey became available, enzyme activities increased, with delayed access to prey having minimal impact on this enzymatic response. Furthermore, phyllosoma were able to adjust α-amylase and non-specific esterase activities (within 8 h) in response to the disappearance of prey, demonstrating adaptive changes to endure periods of food deprivation that were based on their nutritional history. Effectiveness of restocking programs in Marine Fully Protected Areas' (FPAs) can be highly affected by the movement behavior of the species to protect. We analyzed the data of 744 Palinurus elephas specimens tagged, relocated inside 12 FPAs, established in the seas surrounding Sardinia (central-western Mediterranean), and then recaptured. The overall aims were to characterize the individuals' movements after the relocation and to discuss strengths and weaknesses of the current FPAs' design. Almost 80% of tagged individuals travelled progressively increasing distances, as a function of the release distance, up to a maximum value of 11,500 m. Then, for larger release distances, individuals' mobility appeared reduced and more chaotic. A similar trend was found analyzing the distances travelled with respect to the FPAs with the highest number of recaptured individuals and to the size. Sound is an important cue for arthropods. In insects, sound features and sound-producing apparatus are tightly correlated to enhance signal emission in larger individuals. In contrast, acoustic scaling in marine arthropods is poorly described even if they possess similar sound-producing apparatus. Here, the acoustic scaling of the European spiny lobster is analyzed by recording sounds in situ at 1 m from a wide range of body sizes. The dimensions of associated sound-producing apparatus increased with body size, indicating sound features would also be influenced by spiny lobster size. Indeed, temporal sound features changed with body size, suggesting differences in calling songs could be used for spiny lobster acoustic communication. Source levels (peak-peak) ranged from 131 to 164 dB re 1μPa for smaller and larger lobsters, respectively, which could be explained by more efficient resonating structures in larger animals. In addition, dominant frequencies were highly constrained by ambient noise levels, masking the low-frequency content of low intensity sounds from smaller spiny lobsters. Although the ecological function of spiny lobster sounds is not clear yet, these results suggest larger body sizes benefit because louder calls increase the broadcast area and potential interactions with conspecifics, as shown in the insect bioacoustic literature.

Panulirus argus virus 1 (PaV1) (Family Mininucleoviridae) causes chronic and systemic infection in wild juvenile spiny lobsters Panulirus argus (Latreille, 1804), ending in death by starvation and metabolic wasting. In marine decapods, the antennal gland is involved in osmoregulation and excretion. In this compact organ, fluid is filtered from the hemolymph, and ions are reabsorbed to produce a hypotonic urine. Although PaV1 is released with the urine in infected individuals, little is known regarding the metabolic effect of PaV1 in the antennal gland. The objective of this study was to perform a comparative evaluation of the metabolic profile of the antennal gland of clinically PaV1-infected lobsters versus those with no clinical signs of infection, using proton nuclear magnetic resonance analysis. Overall, 48 compounds were identified, and the most represented metabolites were those involved in carbohydrate, amino acid, energy, and nucleotide metabolism. Most of the metabolites that were down-regulated in the infected group were essential and non-essential amino acids. Some metabolites involved in the urea cycle and carbohydrate metabolism were also altered. This study represents a first approach to the metabolic evaluation of the antennal gland. We broadly discuss alterations in the content of several proteinogenic and non-proteinogenic amino acids and other key metabolites involved in energetic and nucleotide metabolism.

Ammonia is a common pollutant in the aquatic ecosystem and closed aquaculture systems. It may pose a threat to the lobster growth, reproduction and survival. However, there is lack of research of the mechanisms on the toxic effects ammonia at molecular levels. In this work, proteomics and metabolomics were integrated to analyze the proteome and metabolome responses in the ornate spiny lobster Panulirus ornatus treated with ammonia (20 mg L -1 ) for 48 h. A total of 199 proteins and 176 metabolites were significantly altered in P. ornatus following ammonia treatment. The responsive proteins and metabolites were predominantly involved in immune response, phase I and phase II biotransformation, carbohydrate metabolism, amino acid metabolism, and lipid metabolism. Furthermore, an increase in urea levels was observed, and amino acid metabolism was induced, indicating that the urea cycle was utilized to biotransform ammonia so as to reduce endogenous ammonia content. Ammonia exposure also affected the antioxidant system and induced cellular apoptosis. Overall, our results provide comprehensive insights into the molecular mechanisms underlying the response of P. ornatus to ammonia stress. The anaesthetic isoeugenol has been used as metabolic suppressant for commercial transport of live lobsters in order to decrease energy expenditure and improve survival. Given the central role of mitochondria in metabolism and structural similarities of isoeugenol to the mitochondrial electron carrier coenzyme Q, we explored the influence on mitochondrial function of isoeugenol. Mitochondrial function was measured using high-resolution respirometry and saponin-permeabilised heart fibres from the Australasian red spiny lobster, Jasus edwardsii. Relative to vehicle (polysorbate), isoeugenol inhibited respiration supported by complex I (CI) and cytochrome c oxidase (CCO). While complex II (CII), which also reduces coenzyme Q, was largely unaffected by isoeugenol, respiration supported by CII when uncoupled was depressed. Titration of isoeugenol indicates that respiration through CI has a half-maximal inhibitory concentration (IC50) of 2.4±0.1 µmol l-1, and a full-maximal inhibitory concentration (IC100-) of approximately 6.3 µmol l-1. These concentrations are consistent with those used for transport and euthanasia of J. edwardsii and indicate that CI is a possible target of isoeugenol, like many other anaesthetics with quinone-like structures., Competing Interests: Competing interests The authors declare no competing or financial interests., (© 2022. Paralytic shellfish toxins (PST) are found in the hepatopancreas of Southern Rock Lobster Jasus edwardsii from the east coast of Tasmania in association with blooms of the toxic dinoflagellate Alexandrium catenella . Tasmania's rock lobster fishery is one of the state's most important wild capture fisheries, supporting a significant commercial industry (AUD 97M) and recreational fishing sector. A comprehensive 8 years of field data collected across multiple sites has allowed continued improvements to the risk management program protecting public health and market access for the Tasmanian lobster fishery. High variability was seen in toxin levels between individuals, sites, months, and years. The highest risk sites were those on the central east coast, with July to January identified as the most at-risk months. Relatively high uptake rates were observed (exponential rate of 2% per day), similar to filter-feeding mussels, and meant that lobster accumulated toxins quickly. Similarly, lobsters were relatively fast detoxifiers, losing up to 3% PST per day, following bloom demise. Mussel sentinel lines were effective in indicating a risk of elevated PST in lobster hepatopancreas, with annual baseline monitoring costing approximately 0.06% of the industry value. In addition, it was determined that if the mean hepatopancreas PST levels in five individual lobsters from a site were <0.22 mg STX equiv. kg -1 , there is a 97.5% probability that any lobster from that site would be below the bivalve maximum level of 0.8 mg STX equiv. kg -1 . The combination of using a sentinel species to identify risk areas and sampling five individual lobsters at a particular site, provides a cost-effective strategy for managing PST risk in the Tasmanian commercial lobster fishery.

An ultimate benefit of marine protected areas (MPAs) is to reverse trophic cascades caused by human-driven collapse of critical ecological interactions. Here we demonstrate that, despite a small scale (0.28 km 2 ) and not being fully protected, an MPA with strict fishing management and habitat enhancement by artificial reefs (ARs) in southwest Japan can lead to well-established macroalgal communities on widespread sea urchin barrens through cascading effects of predator recovery. Areas with low urchin densities occurred in and around daytime lobster (Panulirus japonicus) shelters primarily formed by quarry-rock ARs inside the MPA. We confirmed in the laboratory that lobsters preyed on two dominant sea urchins (Echinometra sp. A and Heliocidaris crassispina), with size- and species-dependent predation. The area with few urchins extended farther (˜65 m) from an AR with numerous lobsters than from a natural shelter (patch reef) with far fewer lobsters. Causation of this pattern was confirmed by a tethering experiment showing that predation on urchins was similarly high at and near lobster shelters but decreased at ˜100 m from the AR to a similar level as at an unprotected site. Time-lapse photography revealed that predation on tethered urchins was due mostly to the largest size class of lobsters (>100 mm carapace length), which comprised only 7% of the population, highlighting the importance of large-sized lobsters in controlling urchin abundance in localized areas adjacent to urchin-dominated barrens. Despite an ongoing once-a-year fishing event permitted within the MPA, lobster populations were persistent, demonstrating that the cascading effect of the lobsters on urchins and ultimately macroalgae was robust to temporary reductions in predator population size. Erect macroalgal cover was not simply accounted for by snapshot urchin density or biomass, suggesting a hysteresis effect of the phase shifts between macroalgal dominance and urchin barren states., (© 2021 by the Ecological Society of America.)
