Abstract The severity of autism spectrum disorder (ASD) shows wide variations, though the reason remains unclear. Vitamin D (VitD) deficiency is considered a risk factor for ASD and its supplementation was reported to reduce symptom severity. Since VitD, either synthesized in the skin or absorbed from the food, is transported to the liver by the vitamin D binding protein (DBP), we have analyzed DBP genetic polymorphisms [rs7041 (A/C), rs4588 (G/T), and rs3755967 (C/T)] affecting DBP function [Case = 411; Control = 397], levels of plasma 25(OH)D and DBP [Case = 25; Control = 26], and DBP mRNA expression [Case = 74; Control = 44] in a group of Indo-Caucasoid ASD probands and neurotypical subjects. ASD probands with rs7041’CC’, rs4588 ‘TT’, and rs3755967 ‘TT’ genotypes exhibited higher scores for a few traits. Scores for Imitation and Listening response were also higher in the presence of the “A-T” haplotype (rs7041–rs4588). Plasma 25(OH)D and DBP levels as well as DBP mRNA expressions were significantly lower in the ASD probands as compared to the neurotypical subjects. We infer that DBP deficiency, in the presence of risk genetic variants, could be one of the reasons for the reported 25(OH)D deficiency of the ASD probands.

Latar Belakang: Sekitar 50% orang di seluruh dunia dan 95% wanita usia subur di Indonesia mengalami kekurangan vitamin D. Kadar 25(OH)D tubuh dapat dipengaruhi oleh asupan mikronutrien. Kadar 25(OH)D serum dan asupan mikronutrien seperti asupan vitamin D, magnesium, dan zink berkaitan dengan kejadian obesitas. Tujuan: Penelitian ini dilaksaakan untuk mengetahui perbedaan kadar 25(OH)D serum, asupan vitamin D, magnesium, dan zink pada mahasiswi dengan status gizi obesitas dan normal di Fakultas Kedokteran Universitas Andalas. Metode: Penelitian ini adalah penelitian observasional dengan case control design, dilaksanakan di Fakultas Kedokteran dan Laboratorium Biomedik Universitas Andalas pada bulan November 2021-Juli 2022. Sampel penelitian berjumlah 64 orang mahasiswi dengan status gizi obesitas dan normal. Teknik pengambilan sampel yaitu systematic random sampling. Data penelitian didapatkan melalui pengambilan darah dan pengisian kuesioner SQ-FFQ. Analisis bivariat menggunakan uji t-test independent. Hasil: Hasil penelitian ini menampilkan rerata kadar 25(OH)D serum, asupan vitamin D, magnesium, dan zink pada kelompok obesitas adalah 10,98 ± 2,12 ng/mL; 5,07 ± 1,47 mcg; 166,04 ± 65,27 mcg; dan 6,88 ± 2,73 mcg, sedangkan rerata pada kelompok normal adalah 13,94 ± 4,57 ng/mL; 5,92 ± 1,77 mcg; 205,88 ± 92,84 mcg; dan 7,62 ± 2,67 mcg. Rerata kadar 25(OH)D serum, asupan vitamin D, magnesium, dan zink lebih rendah pada kelompok obesitas (p-value = 0,001; 0,040; 0,044; 0,280). Kesimpulan: Kesimpulan penelitian adalah terdapat perbedaan yang bermakna kadar 25(OH)D serum, asupan vitamin D, magnesium pada mahasiswi dengan status gizi obesitas dan normal. Tidak terdapat perbedaan yang bermakna asupan zink pada mahasiswi dengan status gizi obesitas dan normal.

Objective: The aim of our study was to compare the total 25(OH)D fraction, the bioavailable vitamin fraction, and the free vitamin D fraction in spring and fall in a group of healthy individuals.Methods: In our study, we collected blood samples from healthy participants at the end of both summer and winter, and measured serum levels of albumin, DBP, and 25(OH)D. Utilizing these data, we calculated the percentage of free and bioavailable vitamin D. Our cohort comprised 87 participants, with a male-to-female ratio of 14:73, aged 35.95 ± 12.55 years, ranging from 19 to 70 years. We employed the chemiluminescence method to determine the vitamin 25(OH)D levels, the ELISA method was utilized to determine DBP levels, the albumin BCP Assay was performed using the ADVIA biochemical analyzer (Siemens) and an online calculator was used to determine the free and bioavailable 25(OH)D levels.Results: Our findings indicate significantly lower 25(OH)D levels in winter (44.13 ± 17.82 nmol/L) compared to summer (74.97 ± 22.75 nmol/L; p < 0.001). For vitamin D binding protein there was no significant difference from summer (236.2 ± 164.39 mg/L) to winter (239.86 ± 141.9 mg/L; p = 0.77), albumin levels were significantly higher in summer (49.37 ± 4.15 g/L vs. 47.97 ± 3.91 g/L, p = 0.01), but the magnitude of the change may not be large enough to be solely responsible for the stability of vitamin D levels throughout the year. In the winter season a significantly lower calculated bioavailable 25(OH)D vitamin (7.45 ± 5.66 nmol/L against 13.11 ± 8.27 nmol/L; p < 0.001) was observed, and the free fraction also showed a significant decrease (17.3 ± 12.9 pmol/L versus 29.7 ± 19.1 pmol/L; p < 0.0001). We observed a moderately positive correlation between 25(OH)D and bioavailable percentage in winter (r = 0.680; p < 0.001), in contrast with a lower positive association in summer (r = 0.343; p < 0.001).Conclusion: Our data suggest a positive correlation between total and bioavailable 25(OH)D levels. In addition to the statistically significant variation in 25(OH)D between the two observation periods, there was an additional variation in the free vitamin D percentage. The summertime synthesis of vitamin D in the skin could contribute directly to the free fraction of vitamin D. Standardizing the measurement of free 25(OH)D and clinical studies is necessary to establish reference values before these methods can be implemented in clinical practice.

Background: Few studies have correlated maternal and neonatal Vit D (25(OH)D) levels at birth in Greece. We investigated this potential association, taking into account the administration or not of low doses (400–800 IU) of prenatal Vit D supplements. Our study contributes evidence not only to the small amount of existing literature regarding the above correlation, but also to the topic of maternal and neonatal vitamin D deficiency (VDD) during pregnancy in Mediterranean countries, such as Greece. Methods: A cross-sectional study was conducted on 248 neonates and their mothers from September 2019 to January 2022. Blood samples of 25(OH)D were studied at the time of delivery. Frequency counts and percentages were registered, and logistic regression was used to investigate the independent factors associated with maternal Vit D status. The Chi-square test and the Pearson coefficient were used to demonstrate a possible association between maternal and neonatal 25(OH)D levels. Results: Our findings show a high prevalence of VDD in Greek women and their newborns at birth. This was observed not only in women who did not receive Vit D supplements, but also in all the study groups, especially in the autumn and winter months. We observed that mothers who received low doses (400–800 IU) of prenatal Vit D supplements increased both their own 25(OH)D concentrations and those of their newborns; however, the latter did not seem to be completely covered by the prenatal administration of Vit D because, although their 25(OH)D concentrations increased, they never reached sufficient 25(OH)D levels, unlike their mothers who reached sufficient concentrations. Conclusions: Overall, this study highlights the strong association between maternal and neonatal 25(OH)D concentrations at the end of gestation. However, neonates tended to show even lower 25(OH)D concentrations relative to maternal 25(OH)D concentrations. The same phenomenon was observed irrespective of the administration of Vit D supplements during pregnancy. Moreover, this is what was observed concerning the administration of formulations with 400–800 IU of Vit D, which the doctors in our clinic used in the present study. In any case, more clinical studies related to the administration of higher doses of Vit D supplementation to pregnant women would lead to more reliable conclusions. Without a doubt, the measurement of maternal vitamin D status during pregnancy provides opportunities for preventive and therapeutic interventions in the mother–infant pair.

Background: Neonatal bone mass may potentially be influenced by existing maternal vitamin D (25(OH)D) levels. Few studies evaluated maternal vitamin D deficiency (VDD) with neonatal anthropometrics such as weight, height and head circumference (HC), especially in Greece, which is a Mediterranean country with plenty of sunshine and consequently benefits the synthesis of 25(OH)D. We investigated this potential association in Greece, taking into account the administration or not of prenatal vitamin D supplements. The purpose of our study is to ascertain if there is a possible association between maternal VDD and neonatal specific anthropometric characteristics (weight, height and HC) at birth. If this is confirmed by future clinical studies, it would be of interest to develop a prenatal pregnancy selection program that would detect VDD early or during pregnancy in order to improve fetal–neonatal development in a Mediterranean country like ours. Methods: We performed a cross-sectional study on 248 early early term infants (after 37 + 0 to 38 + 6 weeks of gestation) but also on full-term infants (after 39 to 40 weeks of gestation) and their Greek mothers from September 2019 to January 2022. Blood samples of 25(OH)D were taken from the mother at the beginning of labor and cord blood was taken from the newborn. Pregnant women were divided into two groups: those who received or did not receive a normal dose of calcium (500 mg/day) and vitamin D supplements (400–800 IU/day) as instructed by their treating physicians. Results: Our findings revealed a positive association between maternal VDD and low neonate birth weight (LBW) in women receiving vitamin D during pregnancy and no association between maternal VDD and neonatal height or head circumference (HC) at birth. Conclusions: Overall, this study highlighted the association between maternal VDD at the end of gestation and LBW neonates born to mothers who received vitamin D supplementation. We did not find any correlation in two of the three somatometric characteristics studied, height and HC. In any case, more clinical studies are needed to further corroborate any potential association of maternal VDD with other neonatal somatometric characteristics.

There is limited data on the vitamin D status of UK-based professional academy footballers. Therefore, the objective of this study was to report total 25(OH)D, free 25(OH)D and free 1,25(OH) 2 D at the end of the winter (March) and summer periods (October) in a cohort ( n = 27) of professional academy footballers in northern England. Blood samples were collected to measure total 25(OH)D, parathyroid hormone, vitamin D binding protein, albumin and calcium. Free 25(OH)D and 1, 25(OH) 2 D were calculated. Dietary vitamin D intake and retrospective summer sunlight exposure were also collected. At the end of winter, 2/27 (7.4%) players were vitamin D deficient (25(OH)D 30 nmol/l < 50 nmol/l). By the end of summer, none were deficient but 3/14 (21.4%) were still insufficient. Median total 25(OH)D(82.2 nmol/l [IQR: 50.3–90.2] vs. 54.2 nmol/l [IQR: 36.8–71.9]; P = .02), free 25(OH)D (25.8 pmol/l [IQR: 15.1–33.1] vs. 13.2 pmol/l [IQR: 9.0–14.9]; P = .005) and free 1, 25(OH)2D (389 fmol/l [IQR: 209–594] vs. 212 fmol/l [IQR: 108–278]; P = .034) were significantly higher at the end of summer than the end of winter. At the end of winter, free 25(OH)D was lower (P = .003) in those vitamin D insufficient (8.8 pmol/l [IQR: 5.5–11.8]) vs. sufficient (13.7 pmol/l [IQR: 12.0 17.0]). There was a high prevalence of vitamin D insufficiency at the end of the winter. Free 25(OH)D was also lower at the winter timepoint and in players that were insufficient vs. sufficient.

Objective: To investigate the correlation between homocysteine (Hcy), 25-hydroxyvitamin D [25-(OH)-D], high mobility group protein B1 (HMGB1) and oxidative stress indexes and inflammatory cytokines in serum and aqueous humor of patients with age-related cataract (ARC). Methods: 135 ARC patients admitted to The Sixth People's Hospital of Chengdu from January 2021 to January 2023 were selected as ARC group and 60 patients with ocular trauma in the same period were selected as control group. The levels of Hcy, 25(OH)-D, HMGB1, oxidative stress indexes [superoxide dismutase (SOD), malondialdehyde (MDA), catalase (CAT), glutathione peroxidase (GSH-PX)] and inflammatory cytokines [tumor necrosis factor-a (TNF-a), interleukin (IL) -1B, IL-6, IL-8] in serum and aqueous humor were detected. The correlation between serum and aqueous humor Hcy, 25(OH)-D, HMGB1 and oxidative stress and inflammatory cytokine levels in ARC patients were analyzed by Pearson correlation analysis. Results: The levels of Hcy, HMGB1, MDA, TNF-a, IL-1B, IL-6 and IL-8 in serum and aqueous humor in ARC group were higher than those in control group, while the levels of 25-(OH)-D, SOD, CAT and GSH-PX were lower than those in control group(P<0.05). Pearson correlation analysis showed that, Hey and HMGB1 in serum and aqueous humor of ARC patients were negatively correlated with SOD, CAT and GSH-PX, and positively correlated with MDA, TNF-α, IL-1B, IL-6 and IL-8 (P<0.05). 25-(OH)-D was positively correlated with SOD, CAT and GSH-PX, and negatively correlated with MDA, TNF-α, IL-1B, IL-6 and IL-8 (P<0.05). Conclusions: The levels of Hey and HMGB1 in serum and aqueous humor of ARC patients increase, and the level of 25(OH)-D decrease, which is significantly correlate with oxidative stress and inflammatory cytokines. Hcy, HMGB1, 25-(OH)-D may be involve in the development of ARC through oxidative stress and inflammatory response. [ABSTRACT FROM AUTHOR]

BACKGROUND: The burden of disease due to lead exposure continues to increase. Lead interferes with 25(OH)D hydroxylation and calcium transport, increasing osteoclastic activity and bone resorption. Pyridinoline crosslinks (PYD), as an indicator of bone damage, can be seen earlier compared to imaging changes. Therefore, it is necessary to determine the correlation between serum 25(OH)D levels and the urinary PYD/creatinine ratio in workers exposed to lead, since up to now, there are only limited studies related to it. METHODS: This cross-sectional study involved 104 workers exposed to lead, selected from parents whose children had blood lead levels above 10 µg/dL. Questionnaires and physical examination were performed to obtain characteristic data from subjects. Data regarding blood lead levels, serum 25(OH)D levels, urinary PYD levels, and urinary creatinine levels were also obtained from various laboratory methods. RESULTS: Most subjects (86.5%) had inadequate serum 25(OH)D. Median blood lead levels was 6.3 (1.2-35.5) µg/dL, chronic lead exposure index was 35.3 (1.2-535.8) years µg/dL, serum 25(OH)D levels was 22 (8-52) ng/mL, and urinary PYD/creatinine ratio was 5.3 (3.6-28.1)×10-6. There was a significant negative correlation between serum 25(OH)D levels and urinary PYD/creatinine ratio in workers exposed to lead. There was also a significant positive correlation between chronic lead exposure index and the urinary PYD/creatinine ratio. CONCLUSION: Since urinary PYD/creatinine ratio is correlated with serum chronic lead exposure index and serum 25(OH)D levels, it suggests that pyridinoline might be a potential biomarker to detect bone metabolism disorder due to the chronic lead exposure. Vitamin D adequacy is also an important factor in preventing bone metabolism disorder amidst chronic lead exposure. KEYWORDS: 25(OH)D, bone resorption, pyridinoline, lead, worker

Abstract Background Mounting evidence suggests that vitamin D deficiency is associated with a higher risk of many chronic non-skeletal, age-associated diseases as well as mortality. Aim To determine, in older patients aged ≥ 80, the prevalence of vitamin D deficiency and its association with comorbidity, laboratory tests, length of stay and mortality within one year from blood withdrawal on admission to acute geriatrics ward. Methods We retrospectively surveyed electronic hospital health records of 830 older patients. The recorded data included patient demographics (e.g., age, sex, stay duration, readmissions number, death within one year from blood withdrawal on admission), medical diagnoses, laboratory results, including 25-hydroxyvitamin D [25(OH)D], and medications. We compared the characteristics of the patients who survived to those who died within one year. Results On admission, in 53.6% patients, vitamin D levels were lower than 50 nmol/L, and in 32%, the levels were ≤ 35 nmol/L. Persons who died were likely to be older, of male sex, were likely to be admitted for pneumonia or CHF, were likely to have lower level of albumin or hemoglobin, lower level of vitamin D or higher vitamin B12 and higher level of creatinine, were also likely to have had a lengthier hospitalization stay, a greater number of hospitalizations in the last year, a higher number of comorbidities, to have consumption of ≥5 drugs or likely to being treated with insulin, diuretics, antipsychotics, anticoagulants or benzodiazepines. Vitamin D is commonly used to prevent and treat osteoporosis, with studies indicating its potential to reduce fractures, falls, and mortality. However, meta-analyses present inconsistent findings regarding its efficacy, particularly reflecting significant variability in data and outcomes related to various dosing regimens. In this meta-analysis, we assessed the impact of high-dose intermittent oral administration of vitamin D3 on serum 25(OH)D levels, fractures, falls, and mortality among elderly individuals. We included 14 randomized controlled trials and employed Review Manager 5.4 for statistical analysis. Our findings indicate that intermittent monthly administration of vitamin D3 (over 800 IU per day) significantly raised serum 25(OH)D levels at all timepoints after six months, maintaining levels above 75 nmol/L throughout the year. This regimen showed no increase in all-cause mortality, with a risk ratio (95% CI) of 0.95 (0.87-1.04). Likewise, it did not significantly reduce the risks of falls and fractures, with risk ratios of 1.02 (0.98-1.05) and 0.95 (0.87-1.04) respectively. Although one-year intermittent administration significantly increased the concentration of 25(OH)D in serum, further research is needed to determine if this method would increase the incidence of falls. Therefore, it is not recommended at this stage due to the lack of demonstrated safety in additional relevant RCTs. This study had been registered on PROSPERO (CRD42022363229).

The mechanism of hepatic encephalopathy is complex and has not been conclusively established. Recent studies support lower serum 25-Hydroxy Vitamin D [25(OH) D] levels in patients with hepatic encephalopathy. This study aimed to evaluate the association between serum 25(OH) D and hepatic encephalopathy in patients with decompensated cirrhosis of liver. A total of 70 cirrhosis patients (35 cases of hepatic encephalopathy and 35 patients without encephalopathy as control, mean age 53.07 ± 12.99 years, 67 % male) were recruited for this study. Assessment of the severity of cirrhosis was done by using a model for end-stage liver disease(MELD) and Child Turcotte Pugh (CTP) scores, and assessment of the severity of hepatic encephalopathy was done according to West Haven criteria. Serum 25 (OH) D level was measured by Chemiluminescent Microparticle Immuno Assay(CMIA). The mean serum 25(OH) D level among hepatic encephalopathy patients was significantly lower in comparison to the control group without encephalopathy (18.76 ± 8.84 nmol/L vs 31.19 ± 13.9 nmol/L, P <0.0001). 91.4 % of hepatic encephalopathy patients had moderate to severe 25(OH)D deficiency as compared to 51.4 % in the control group. There was a significant correlation observed between the severity of the 25 (OH) D deficiency and the severity of liver disease (r = − 0.35, P = 0.002). No statistically significant difference in serum 25(OH) D levels was found among patients with different hepatic encephalopathy grades (P = 0.416). A significant association was found between a low serum 25(OH) D leveland hepatic encephalopathy. It requires further large-scale multicenter studies to establish it as a risk factor and predictor of hepatic encephalopathy. [ABSTRACT FROM AUTHOR]

Objective: To investigate the relationship between serum hemoglobin A (HbA1c), 25 hydroxyvitamin D [25(OH)D], matrix metalloproteinase-9 (MMP-9) and postoperative dry eye in patients with type 2 diabetes mellitus (T2DM) complicated with cataract. Methods: 162 patients with T2DM complicated with cataract who underwent phacoemulsification combined with intraocular lens implantation in our hospital from January 2019 to December 2023 were selected, patients were divided into dry eye group (53 cases) and non-dry eye group (109 cases) according to whether dry eye occurred after surgery. The levels of serum HbA1c 25(OH)D and MMP-9 were detected. The factors of postoperative dry eye in patients with T2DM complicated with cataract surgery were analyzed by multivariate Logistic regression, the predictive value of serum HbA1c 25 (OH)D and MMP-9 levels for postoperative dry eye in patients with T2DM complicated with cataract were analyzed by receiver operating characteristic (ROC) curve. Results: Compared with non-dry eye group, the levels of serum HbA1c and MMP-9 in dry eye group increased, and the level of 25(OH)D decreased (P < 0.05) Multivariate Logistic regression analysis showed that prolonged course of T2DM ( OR = 1.245 . 95%CI: 1.007~1.539), elevated HbAte (OR = 3.132, 95% CI: 1.745-5.624), and elevated MMP-9 (OR = 1.206 95%CI: 1.091~1.333) were independent risk factors for postoperative dry eye in patients with T2DM complicated with cataract surgery, Elevated 25 (OH) D ( OR = 0.854, 95% * CI 0.773~0.945) was an independent protective factor (P < 0.05) ROC curve analysis showed that, the area under the curve of combined prediction of serum HbA1c, 25(OH)D and MMP-9 levels for postoperative dry eye in T2DM patients with cataract was 0.921 (95%CI: 0.868~0.957), which was greater than 0.793 (95%CI: 0.723~0.853), 0.779 (95%CI: 0.707~0.840) and 0.783 (95%CI: 0.711~0.844) predicted by serum HbA1 25(OH)D and MMP-9 levels alone. Conclusion: Elevated preoperative serum HbAte and MMP-9 levels and the decrease of 25 (OH)D levels in patients with T2DM complicated with cataract are closely related to postoperative dry eye, and the combination of serum HbA1c 25 (OH)D and MMP-9 has a value in predicting postoperative dry eye in patients with T2DM complicated with cataract surgery. Simple Summary: Advances in the treatment of acute lymphoblastic leukemia (ALL) have led to a marked improvement in the survival rate of patients. Nevertheless, these patients may develop adverse effects during and after treatment, such as bone abnormalities and vitamin D deficiency. Bone remodeling allows for bone volume and structure to be maintained, which is controlled by the receptor activator of the nuclear factor-kB (RANK)/RANK ligand (RANKL)/osteoprotegerin (OPG) system-determining pathway in the balance between bone formation and resorption. Some reports have explored the role of corticosteroids in modulating the RANKL and OPG levels and RANKL/OPG ratio in pediatric patients. Nevertheless, studies evaluating the role of RANKL and OPG in the bone health of pediatric ALL patients during treatment are limited. During remission, we observed an increase in the RANKL/OPG ratio, increased RANKL levels, and decreased OPG levels in ALL patients. These changes may predispose such patients to the development of bone health disorders in their adult lives. Background: The receptor activator of the nuclear factor-kB (RANK)/RANK ligand (RANKL)/osteoprotegerin (OPG) pathway is a determining pathway in the balance between bone formation and resorption, and disruptions in this complex can affect bone metabolism. Methods: This study analyzes the changes in RANKL, OPG, and 25(OH)D levels; the RANKL/OPG ratio; and other bone turnover markers (BTMs) from diagnosis to complete remission in children with acute lymphoblastic leukemia (ALL). This is a prospective observational cohort study, carried out at the Instituto Mexicano del Seguro Social, Mexico City, including 33 patients (4–17 years) with newly diagnosed B-cell ALL. The patients were treated with the HP09 chemotherapy protocol. Children who had previously been treated with corticosteroids were excluded. A peripheral blood sample at diagnosis and remission was collected to determine the 25(OH)D and BTM concentrations. Results: Increased RANKL (p = 0.001) and osteocalcin (p < 0.001) levels and RANKL/OPG ratio (<0.001) and a decreased OPG level (p = 0.005) were observed at remission, predominantly in the high-risk (HR) relapse and vitamin D deficiency groups. A negative association between RANKL and OPG (r = −0.454, p = 0.008) was observed. Conclusions: we suggest that the RANKL/OPG ratio could serve as a bone remodeling marker in ALL patients. Objective To investigate the changes in serum 25-hydroxyvitamin D ［25（OH）D］ and sex hormone-binding globulin （SHBG） levels in pregnant women with obese polycystic ovary syndrome （PCOS) and their predictive value for adverse pregnancy. Methods Totally 125 pregnant women with obese PCOS （PCOS group) and 52 healthy pregnant women in the same period （control group) were selected, and the pregnant women with obese PCOS were divided into the adverse pregnancy group （48) and good pregnancy group （77) according to the pregnancy outcome. Serum 25（OH）D and SHBG levels were measured by enzyme-linked immunosorbent assay. Factors of adverse pregnancy in the obese pregnant women with PCOS were analyzed by multifactorial Logistic regression, and the predictive value of serum 25（OH）D and SHBG levels for adverse pregnancy in pregnant women with PCOS was analyzed by receiver operating characteristic （ROC） curve. Results Compared with the control group, serum 25（OH）D and SHBG levels were reduced in the PCOS group （both P<0. 05). The incidence of adverse pregnancy in 125 pregnant women with obese PCOS was 38. 40% （48/ 125）. Serum 25（OH）D and SHBG levels were lower in the adverse pregnancy group than those in the good pregnancy group （both P<0. 05). Elevated luteinizing hormone/follicle-stimulating hormone, elevated testosterone, elevated low-density lipoprotein cholesterol, and elevated homeostasis model assessment-insulin resistance were independent risk factors for adverse pregnancy in pregnant women with obese PCOS, and elevated 25（OH）D and elevated SHBG were independent protective factors （all P<0. 05). The area under the curve of serum 25（OH）D combined with SHBG in predicting obese PCOS was 0. 882, which was greater than that （0. 783 and 0. 775） of serum 25（OH）D and SHBG levels alone （both P<0. 05）. Conclusion Serum 25（OH）D and SHBG levels in pregnant women with obese PCOS decrease, which are closely associated with adverse pregnancy, and the combined detection of serum 25（OH）D and SHBG is of high value in predicting adverse pregnancy in pregnant women with obese PCOS. Resumo Introdução: Alterações fisiológicas do envelhecimento contribuem para osteoporose e fraturas relacionadas à baixa concentração sérica de 25(OH)D. Objetivo: Investigar a associação entre concentração sérica de 25(OH)D, ingestão de alimentos fonte de vitamina D e cálcio, condições de saúde e presença de osteoporose e fraturas em idosos. Método: Estudo transversal com 274 idosos do Piauí. Realizado em duas etapas: questionário sobre ingestão de alimentos fonte de vitamina D, cálcio e condições de saúde (incluindo a variável desfecho, presença de osteoporose e fraturas) e coleta de sangue para exame bioquímico de concentração sérica de 25(OH)D. Realizou-se análise descritiva, inferencial e regressão logística. Resultados: A maioria dos idosos apresentava comorbidades (86,4%). A prevalência dos desfechos foi de 23,7% e 14,6%, respectivamente. Na análise múltipla, ingerir bebidas alcoólicas (OR=6,58; IC95% 1,88–23) e reposição com cálcio (OR=2,87; IC95% 1,25–6,61) mostraram-se associados às fraturas, enquanto sexo feminino (OR=5,69; IC95% 1,12–28,9) e reposição com cálcio (OR=5,62; IC95% 1,62–19,5) mostraram-se associados à osteoporose. As maiores medianas de 25(OH)D foram encontradas naqueles que consumiam queijo e iogurte regularmente (p=0,021). Conclusão: A análise das concentrações séricas de 25(OH)D apontou insuficiência, indicando a necessidade de intervenções para a mudança do estilo de vida.

Abstract Skin pigmentation is negatively associated with circulating vitamin D (VD) concentration. Therefore, genetic factors involved in skin pigmentation could influence the risk of vitamin D deficiency (VDD). We evaluated the impact genetic variants related to skin pigmentation on VD in Mexican population. This cross-sectional analysis included 848 individuals from the Health Worker Cohort Study (ratio males to females ~ 1:3). Eight genetic variants: rs16891982 (SLC45A2), rs12203592 (IRF4), rs1042602 and rs1126809 (TYR), rs1800404 (OCA2), rs12913832 (HERC2), rs1426654 (SLC24A5), and rs2240751 (MFSD12); involved in skin pigmentation were genotyped. Skin pigmentation was assessed by self-report. Linear and logistic regression were used to assess the association between the variants of interest and VD and VDD, as appropriate. In our study, eight genetic variants were associated with skin pigmentation. A genetic risk score built with the variants rs1426654 and rs224075 was associated with lower VD levels (β = − 1.38, 95% CI − 2.59, − 0.17, p = 0.025). Nevertheless, when examining gene–gene interactions, we observed that rs2240751 × rs12203592 were associated with VD levels (P interaction = 0.021). Whereas rs2240751 × rs12913832 (P interaction = 0.0001) were associated with VDD. Our results suggest that skin pigmentation-related gene variants are associated with lower VD levels in Mexican population. These results underscore the importance of considering genetic interactions when assessing the impact of genetic polymorphisms on VD levels.

There is limited data on the vitamin D status of UK-based professional academy footballers. Therefore, the objective of this study was to report total 25(OH)D, free 25(OH)D and free 1, 25(OH)2D at the end of the winter (March) and summer periods (October) in a cohort (n = 27) of professional academy footballers in northern England. Blood samples were collected to measure total 25(OH)D, parathyroid hormone, vitamin D binding protein, albumin and calcium. Free 25(OH)D and 1, 25(OH)2D were calculated. Dietary vitamin D intake and retrospective summer sunlight exposure were also collected. At the end of winter, 2/27 (7.4%) players were vitamin D deficient (25(OH)D < 30 nmol/l) and 11/27 (40.7%) were insufficient (25(OH)D > 30 nmol/l < 50 nmol/l). By the end of summer, none were deficient but 3/14 (21.4%) were still insufficient. Median total 25(OH)D (82.2 nmol/l [IQR: 50.3-90.2] vs. 54.2 nmol/l [IQR: 36.8-71.9]; P = .02), free 25(OH)D (25.8 pmol/l [IQR: 15.1-33.1] vs. 13.2 pmol/l [IQR: 9.0-14.9]; P = .005) and free 1, 25(OH)2D (389 fmol/l [IQR: 209-594] vs. 212 fmol/l [IQR: 108-278]; P = .034) were significantly higher at the end of summer than the end of winter. At the end of winter, free 25(OH)D was lower (P = .003) in those vitamin D insufficient (8.8 pmol/l [IQR: 5.5-11.8]) vs. sufficient (13.7 pmol/l [IQR: 12.0-17.0]). There was a high prevalence of vitamin D insufficiency at the end of the winter. Free 25(OH)D was also lower at the winter timepoint and in players that were insufficient vs. sufficient. BackgroundVitamin D deficiency is common in patients with systemic lupus erythematosus (SLE) and may affect their disease activity and severity.ObjectiveThis study aims to assess the vitamin D status in patients with initial-onset SLE during childhood and its association with the clinical and laboratory markers of disease activity.MethodThis is a retrospective study that includes 168 patients with initial-onset SLE during childhood and 109 healthy children as controls. Clinical and laboratory data were recorded. According to recent literature, there is a limited amount of data about the correlation of vitamin 25(OH)D, potassium (K), oxidative stress parameters, and other biomarkers with dyslipidemia, which is an established risk factor for cardiovascular diseases (CVDs). This study aims to investigate the correlation of lipid profile and atheromatic index TC/HDL with several biomarkers and oxidative stress parameters. A total of 102 volunteers, 67 with atheromatic index TC/HDL > 3.5 (Group A) and 35 with TC/HDL < 3.5 (Group B), aged from 26 to 78 years, participated in this study. Serum levels of triglycerides (TG), total cholesterol (TC), low- and high-density lipoproteins (LDL and HDL), vitamin 25(OH)D [25(OH)D], potassium (K), sodium (Na), lactose dehydrogenase (LDH), liver enzymes including serum glutamic oxaloacetic and glutamic pyruvic transaminases (SGOT and SGPT), gamma-glutamyl transferase (γ-GT), and alkaline phosphatase (ALP) were analyzed using standard photometric methods. Oxidative stress parameters such as reactive oxygen species (ROS) were detected with fluorometric methods, whereas total oxidative (TOS) and antioxidative status (TAS) were measured with spectrophotometric methods. According to the results, negative correlations of HDL (r = −0.593) and 25(OH)D (r = −0.340) and K (r = −0.220) were found, and positive expected correlations of LDL (r = 0.731), TC (r = 0.663), and TG (r = 0.584) with atheromatic index in the total studied sample were found. In conclusion, patients with a dyslipidemic profile should frequently check not only their lipid profile but also other biomarkers such as 25(OH)D, potassium, and oxidative stress markers to predict dyslipidemia and avoid subsequent disorders.

The role of vitamin D in predicting clinical outcome for COVID-19 patients is unknown. Here, we aimed to determine the prognostic value of plasma 25(OH)D level in COVID-19 patients.Patients and Methods: A total of 158 patients infected with novel COVID-19 Omicron variants in Shanghai were recruited in this study and were categorized into three groups by the tertile levels of plasma 25(OH)D. Plasma 25(OH)D level was determined along with routine blood tests related to liver and renal functions in newly diagnosed COVID-19 patients at admission. The nucleic acid negative conversion time of throat swab samples was evaluated as the primary clinical outcome. The prognostic value of clinical characteristics and plasma 25(OH)D level was assessed using the Kaplan–Meier plot and Cox proportional hazards regression tests.Results: Higher level of plasma 25(OH)D level in COVID-19 patients was independently associated with shorter nucleic acid negative conversion time from COVID-19 infection (multivariate adjusted HR: 0.54, 95%CI: 0.35– 0.82, P=0.004, tertile 2 vs 1; multivariate adjusted HR: 0.60, 95%CI: 0.39– 0.90, P=0.014, tertile 3 vs 1).Conclusion: Plasma 25(OH)D level may serve as an independent prognostic factor in COVID-19 patient. Obejective: To investigate the clinical efficacy of relationship between serum 25-hydroxyvitamin D [25 (OH)D], neutrophil gelatinase-associated lipocalin (NGAL), Hepcidin （Hepcidin）and roxadustat capsules in the treatment of renal anemia in maintenance hemodialysis (MHD) patients. Methods: 137 MHD patients with renal anemia who were treated with roxadustat capsules in the Affiliated Hospital of Xuzhou Medical University from October 2020 to October 2022 were selected, and patients were divided into effective group (86 cases) and ineffective group (51 cases) according to the effect of 12 weeks of treatment. The serum levels of 25(OH) D, NGAL and Hepcidin were detected before treatment, the factors affecting the efficacy of roxadustat capsules were analyzed by multivariate Logistic regression model, the value of 25(OH)D, NGAL and Hepcidin in predicting the efficacy of roxadustat capsules were analyzed by receiver operating characteristic (ROC) curve. Results: The level of serum 25 (OH)D in ineffective group before treatment was lower than that in effective group (P<0.05), and the levels of NGAL and Hepcidin were higher than those in effective group (P<0.05). High level of NGAL and high level of Hepcidin were risk factors for ineffective treatment of renal anemia in MHD patients with roxadustat capsules (P<0.05), high level of 25 (OH)D, combined with hemofiltration or hemoperfusion, and urea clearance index (Kt/v) >1.2 were protective factors (P<0.05). The area under the curve (AUC) of 25 (OH)D, NGAL, and Hepcidin combined to predict the efficacy of rosastat capsules was 0.853, which was higher than the prediction of individual indicators. Conclusion: The serum 25 (OH) D level decreased and NGAL and Hepcidin level increased in patients with MHD renal anemia before treatment with roxadustat capsules, the above level changes were related to the treatment effect of roxadustat capsules, the combination of 25 (OH)D, NGAL and Hepcidin could improve the predictive value of the therapeutic effect of roxadustat capsules. Objective: To investigate the value of electrocardiogram QT interval dispersion (QTd) combined with 25-hydroxyvitamin D [25-(OH) D], growth differentiation factor-15 (GDF-15) and pentraxin 3 (PTX3) in the evaluation of coronary artery lesions (CAL) in children with Kawasaki disease. Methods: 180 children with Kawasaki disease admitted to our hospital from June 2018 to June 2023 were selected, and patients were divided into CAL group (36 cases) and non-CAL group (144 cases) according to whether CAL occurred. All children received electrocardiogram examination to obtain QTd and corrected QT dispersion (QTcd), and serum 25-(OH)D, GDF-15 and PTX3 levels were detected. The factors affecting the occurrence of CAL in children with Kawasaki disease were analyzed by multivariate Logistic regression analysis, and a Log P model of multi-index combined application was constructed. The predictive value of QTd, 25-(OH)D, GDF-15 and PTX3 for CAL in children with Kawasaki disease were analyzed by receiver operating characteristic (ROC) curve. Results: The levels of QTd, QTcd, GDF-15 and PTX3 in CAL group were higher than those in non-CAL group (P<0.05), and the level of 25-(OH)D was lower than that in non-CAL group(P<0.05). Treatment non-response, high QTd, high QTcd, high GDF-15, and high PTX3 were risk factors for CAL in children with Kawasaki disease (P<0.05), and high 25-(OH)D was a protective factor (P<0.05). The Log P models of multi-index combined application were constructed as follows: A: electrocardiogram parameter model: Ln (P/1-P)=0.512 ×QTd+0.596 QTcd; B: serum 3 index model: Ln (P/1-P)=0.712×GDF-15+0.626×PTX3-0.609×25- (OH)D; C: 5-factor joint application model (above A+B model): Ln (P/1-P)=0.512×QTd+0.596 QTcd+0.712×GDF-15+0.626×PTX3-0.609×25-(OH)D. ROC analysis showed that, models A, B and C had certain predictive efficacy for CAL in children with Kawasaki disease, and their ROC-AUC (0.95CI) were 0.709 (0.490~0.921), 0.787 (0.579~0.966) and 0.835 (0.699~0.955) respectively. C combined application model has the highest prediction efficiency. Conclusion: The QTd increase, QTcd increased, GDF-15 increase, PTX3 increase, and 25-(OH)D decrease in children with Kawasaki disease with CAL, the combined detection of the above indicators has a high predictive efficiency for the occurrence of CAL in children with Kawasaki disease. This study investigated the association between serum 25(OH)D 3 levels and cardiovascular risk-related indicators. 4 727 participants aged 20 and above from the National Health and Nutrition Examination Survey 2015-2018 database were enrolled. Body mass index, hypersensitive C-reactive protein, high density lipoprotein cholesterol, systolic blood pressure, waist-height ratio, and total cholesterol were selected as the research indicators. Weighted multiple linear regression models, subgroup analyses, smooth curve fitting, and saturation threshold effect analyses were employed to explore the relationship between serum 25(OH)D 3 and these indicators. The results showed that after full adjustment for covariates, every 1 nmol/L increase in serum 25(OH)D 3 , the changes in β (95% CI ) values for body mass index(BMI), hypersensitive C-reactive protein(hs-CRP), systolic blood pressure(SBP), waist-height ratio(WHtR), high density lipoprotein cholesterol(HDL-C), and total cholesterol(TC) were -0.05 (-0.06, -0.04) kg/m 2 , -0.01 (-0.02, -0.01) mg/L, -0.02 (-0.04, -0.01) mmHg, -0.000 7 (-0.000 8, -0.000 6), 0.10 (0.08, 0.11) mg/dl, and 0.08 (0.04, 0.12) mg/dl, respectively. Female participants were more sensitive to changes in serum 25(OH)D 3 , while participants aged 60 and above were relatively less sensitive. The relationship between serum 25(OH)D 3 and these indicators partially exhibited nonlinear patterns across different gender and age subgroups. The saturation threshold effect analysis revealed 8 meaningful inflection points. In summary, vitamin D has a close association with cardiovascular risk-related indicators.

Background Iron deficiency is known to impair muscle function and reduce athletic performance, while vitamin D has been reported to induce iron deficiency. However, the mechanism underlying exercise-induced changes in iron metabolism and the involvement of vitamins in this mechanism are unclear. The present study examined changes in biological iron metabolism induced by continuous training and the effects of vitamin D on these changes. Methods Diet, physical characteristics, and blood test data were collected from 23 female high school students in a dance club on the last day of each of a 2-month continuous training period and a 2-week complete rest periods. Results Serum hepcidin-25 levels were significantly lower during the training period than the rest period (p = 0.013), as were the red blood cell count, hemoglobin, and hematocrit (all p < 0.001). Serum erythropoietin was significantly higher (p = 0.001) during the training period. Significant positive correlations were observed between 25(OH)D levels and serum iron, serum ferritin, and transferrin saturation during the training period. Multiple regression analysis with serum 25(OH)D level as the dependent variable and serum ferritin and iron levels as independent variables during the training period revealed a significant association with serum ferritin. Conclusion Continuous training may promote hemolysis and erythropoiesis, contributing to the suppression of hepcidin expression. The relationship between serum 25(OH)D and iron in vivo may be closely related to metabolic changes induced by the exercise load.

Abstract Background We explored the potential novel anticancer mechanisms of 25‐hydroxyvitamin D (25(OH)D), a vitamin D metabolite with antitumour effects in breast cancer. It is stable in serum and is used to assess vitamin D levels in clinical practice. Transfer RNA‐derived small RNAs are small noncoding RNAs that generate various distinct biological functions, but more research is needed on their role in breast cancer. Methods Small RNA microarrays were used to explore the novel regulatory mechanism of 25(OH)D. High‐throughput RNA‐sequencing technology was used to detect transcriptome changes after 25(OH)D treatment and tRF‐1‐Ser knockdown. RNA pull‐down and high‐performance liquid chromatography–mass spectrometry/mass spectrometry were used to explore the proteins bound to tRF‐1‐Ser. In vitro and in vivo functional experiments were conducted to assess the influence of 25(OH)D and tRF‐1‐Ser on breast cancer. Semi‐quantitative PCR was performed to detect alternative splicing events. Western blot assay and qPCR were used to assess protein and mRNA expression. Results The expression of tRF‐1‐Ser is negatively regulated by 25(OH)D. In our breast cancer (BRCA) clinical samples, we found that the expression of tRF‐1‐Ser was higher in cancer tissues than in paired normal tissues, and was significantly associated with tumour invasion. Moreover, tRF‐1‐Ser inhibits the function of MBNL1 by hindering its nuclear translocation. Functional experiments and transcriptome data revealed that the downregulation of tRF‐1‐Ser plays a vital role in the anticancer effect of 25(OH)D. Conclusions In brief, our research revealed a novel anticancer mechanism of 25(OH)D, unveiled the vital function of tRF‐1‐Ser in BRCA progression, and suggested that tRF‐1‐Ser could emerge as a new therapeutic target for BRCA.

IntroductionManaging postsurgical complications is crucial in optimizing the outcomes of bariatric surgery, for which preoperative nutritional assessment is essential. In this study, we aimed to evaluate and validate the efficacy of vitamin D levels as an immunonutritional biomarker for bariatric surgery prognosis.MethodsThis matched retrospective cohort study included adult patients who underwent bariatric surgery at a tertiary medical center in China between July 2021 and June 2022. Patients with insufficient and sufficient 25(OH)D (< 30 ng/mL) were matched in a 1:1 ratio. Follow-up records of readmission at 3 months, 6 months, and 1 year were obtained to identify prognostic indicators.ResultsA matched cohort of 452 patients with a mean age of 37.14 ± 9.25 years and involving 69.47% females was enrolled. Among them, 94.25 and 5.75% underwent sleeve gastrectomy and gastric bypass, respectively. Overall, 25 patients (5.54%) were readmitted during the 1-year follow-up. The prognostic nutritional index and controlling nutritional status scores calculated from inflammatory factors did not efficiently detect malnourishment. A low 25(OH)D level (3.58 [95% CI, 1.16–11.03]) and surgery season in summer or autumn (2.68 [95% CI, 1.05–6.83]) increased the risk of 1-year readmission in both the training and validation cohorts. The area under the receiver operating characteristic curve was 0.747 (95% CI, 0.640–0.855), with a positive clinical benefit in the decision curve analyses. The relationship between 25(OH)D and 6-month readmission was U-shaped.ConclusionSerum 25(OH)D levels have prognostic significance in bariatric surgery readmission. Hence, preferable 25(OH)D levels are recommended for patients undergoing bariatric surgery.

Background. Hand eczema (HE) commonly occurs in healthcare services, especially during COVID-19 pandemic era. Vitamin D shows various effects on skin inflammation. This study aims to analyze the correlation between HE severity and vitamin D levels as measured by serum 25(OH)D in healthcare workers. Method. This is an analytic descriptive study with a cross-sectional design conducted in Indonesia between September-October 2022. HE severity was assessed using Hand Eczema Severity Index (HECSI). Results. Among 44 healthcare workers with HE, 29 people had mild HE, 11 had moderate HE, and 4 had severe HE. The mean serum 25(OH)D level in subjects with mild, moderate, and severe HE was 17.85 ng/mL, 16.45 ng/mL, and 17.87 ng/mL, respectively, which belonged to vitamin D deficiency category. There was no statistically significant correlation between serum 25(OH)D levels and HE severity (r = -0.056; p = 0.359). Conclusions. There was no significant difference in serum 25(OH)D levels between subjects with mild, moderate, and severe HE. Serum 25(OH)D level was not inversely correlated with HE severity.

BackgroundVitamin D deficiency is a widespread issue globally, resulting in increased use of vitamin D supplements. However, it is unclear whether intermittent (weekly or monthly) vitamin D supplementation is as effective as daily supplementation in improving circulating 25-hydroxyvitamin D [25(OH)D] levels.MethodsThree databases including Medline, EMBASE, and the Cochrane Library were systematically searched up to 10 November 2020. The risk of bias was evaluated according to Cochrane Collaboration’s tool for rating methodological quality assessment. Direct and indirect comparisons between interventions and controls were performed by a Bayesian network meta-analysis (NMA), where the mean difference (MD) and its 95% confidence interval (CI) were used to indicate the efficacy.ResultsThis NMA analysis included 116 RCTs with a total of 11,376 participants. Generally, we observed that 25(OH)D concentrations were significantly elevated regardless of vitamin D supplementation frequency. Although the findings of SUCRA indicated that daily vitamin D supplementation had a higher rank value than intermittent supplementation when the supplement dosage was similar, no statistically significant pooled mean differences of 25(OH)D concentration were noted between the daily supplementation group and intermittent supplementation group. Additionally, weekly supplementation with a total of 600,000 IU vitamin D supplementation during 3 months had the best efficacy in elevating 25(OH)D concentration (pooled MD = 63 nmol/L, 95%CI: 49–77). To achieve optimal 25(OH)D concentration (>75 nmol/L), we recommend 60,000 IU vitamin D supplementation monthly (~2,000 IU/day).ConclusionThe efficacy of intermittent vitamin D supplementation was similar to daily supplementation. BackgroundT helper 17 (Th17) cells and regulatory T cells (Treg) are known to play a crucial role in the pathogenesis of systemic lupus erythematosus (SLE). This study aimed to detect seasonal and species differences in serum 25-hydroxy vitamin D (25(OH)D) concentrations during summer and winter months in captive pachyderms in Europe. Both elephant species had low 25(OH)D while African elephants did not show a seasonal variation. Asian elephants had significantly higher 25(OH)D compared to their African counterparts but also did not show a seasonal difference. Both rhinoceros species investigated had higher 25(OH)D compared to both elephant species; the Indian rhinoceros had high circulating levels year-round, while the black rhinoceroses showed significantly lower 25(OH)D in winter. Malayan tapirs have very low 25(OH)D, comparable to horses. The higher 25(OH)D of elephants and rhinoceroses could indicate that elephants and rhinoceroses are capable of producing vitamin D. This might indicate that the Indian rhinoceroses are capable of producing enough endogenous vitamin D year-round at latitudes around 52° N, while both elephant species and the black rhinoceros are not. This study also showed that it is likely that both elephant species and rhinoceros species are capable of absorbing cholecalciferol from the digestive tract, according to the existing literature, while tapirs may not.

Background: Deficiency in vitamin D is widely prevalent around the world. Oral vitamin D supplementation is suggested for older adults to sustain appropriate 25-hydroxyvitamin D (25(OH)D) levels throughout the year. At present, cholecalciferol (vitamin D3) and ergocalciferol (vitamin D2) are the most commonly used compounds. Supplementation with calcifediol (25OHD3) rather than vitamin D itself should also be considered for the treatment. We performed a systematic review of the literature with a meta-analysis to assess the effects of cholecalciferol (vitamin D3) compared to calcifediol (25OHD3) on increasing serum 25(OH)D levels., Methods: A search of online databases was performed electronically for all relevant observational published population-based studies until November 2023, without geographical restrictions. We included studies that directly compared the effects of cholecalciferol and calcifediol on increasing concentrations of serum 25(OH)D. Only papers in English or French languages were considered. Records were screened and data were retrieved through a standardized extraction process., Results: Seventeen studies including 1575 participants were reviewed. Twelve intervention trials showed that, in spite of the dosage or the frequency of administration, calcifediol supplementation was more efficacious in raising serum 25(OH)D concentrations compared with cholecalciferol. Two studies showed that calcifediol and cholecalciferol were identically potent. According to three studies, cholecalciferol was more effective than calcifediol in raising 25(OH)D concentrations. A meta-analysis including randomized controlled trials (RCTs) and non-randomized trials revealed that calcifediol supplementation had a better impact on elevating serum 25(OH)D concentrations compared with the effect of cholecalciferol., Conclusion: This meta-analysis suggests that calcifediol is more effective in increasing serum 25(OH)D concentrations compared to cholecalciferol.
