This review summarizes the fundamental concepts, recent advancements, and emerging trends in the field of stimuli-responsive hydrogels. While numerous reviews exist on this topic, the field continues to evolve dynamically, and certain research directions are often overlooked. To address this, we classify stimuli-responsive hydrogels based on their response mechanisms and provide an in-depth discussion of key properties and mechanisms, including swelling kinetics, mechanical properties, and biocompatibility/biodegradability. We then explore hydrogel design, synthesis, and structural engineering, followed by an overview of applications that are relatively well established from a scientific perspective, including biomedical uses (biosensing, drug delivery, wound healing, and tissue engineering), environmental applications (heavy metal and phosphate removal from the environment and polluted water), and soft robotics and actuation. Additionally, we highlight emerging and unconventional applications such as local micro-thermometers and cell mechanotransduction. This review concludes with a discussion of current challenges and future prospects in the field, aiming to inspire further innovations and advancements in stimuli-responsive hydrogel research and applications to bring them closer to the societal needs.

A novel approach to selectively modify narrow subareas of metallic nanostructures adjacent to plasmonic hotspots, where strong electromagnetic field amplification occurs upon localized surface plasmon (LSP) excitation, is reported. In contrast to surface plasmon-triggered polymerization, it relies on plasmonically enhanced multiphoton crosslinking (MPC) of polymer chains carrying photoactive moieties. When they are contacted with metallic nanostructures and irradiated with a femtosecond near-infrared beam resonantly coupled with LSPs, the enhanced field intensity locally exceeds the threshold and initiates MPC only at plasmonic hotspots. This concept is demonstrated by using gold nanoparticle arrays coated with two specifically designed polymers. Local MPC of a poly( N , N -dimethylacrylamide)-based copolymer with an anthraquinone crosslinker is shown via atomic force microscopy. Additionally, MPC is tested with a thermoresponsive poly( N -isopropylacrylamide)-based terpolymer. The reversible thermally induced collapse and swelling of the MPC-formed hydrogel at specific nanoparticle locations are confirmed by polarization-resolved localized surface plasmon resonance (LSPR) spectroscopy. The centrosome is the main microtubule organizing center in stem cells, and its mother centriole, anchored to the cell membrane, serves as the basal body of the primary cilium. Prolonged anchorage of centrosomes and primary cilia to the apical segment of the membrane of apical neural progenitor cells is considered vital for interkinetic nuclear translocation and repetitive cycling in the ventricular zone. In contrast, the basolateral anchorage of primary cilia has been regarded as the first step in delamination and conversion of apical to basal neural progenitor cells or neurons. Using electron microscopy analysis of serial sections, we show that centrosomes, in a fraction of cells, anchor to the basolateral cell membrane immediately after cell division and before development of cilia. In other cells, centrosomes situate freely in the cytoplasm, increasing their probability of subsequent apical anchorage. In mice, anchored centrosomes in the cells shortly after mitosis predominate during the entire cerebral neurogenesis, whereas in macaque monkeys, cytoplasmic centrosomes are more numerous. Species-specific differences in the ratio of anchored and free cytoplasmic centrosomes appear to be related to prolonged neurogenesis in the ventricular zone that is essential for lateral expansion of the cerebral cortex in primates., (© The Author(s) 2024. It was also seen trafficking between excitatory neurons within synapses on spines, where it was exposed to the extracellular space, and thus accessible to cerebrospinal fluid (CSF)/blood. Alzheimer's disease cortical tau pathology initiates in the layer II cell clusters of entorhinal cortex, but it is not known why these specific neurons are so vulnerable. Aging macaques exhibit the same qualitative pattern of tau pathology as humans, including initial pathology in layer II entorhinal cortex clusters, and thus can inform etiological factors driving selective vulnerability. Macaque data have already shown that susceptible neurons in dorsolateral prefrontal cortex express a "signature of flexibility" near glutamate synapses on spines, where cAMP-PKA magnification of calcium signaling opens nearby potassium and hyperpolarization-activated cyclic nucleotide-gated channels to dynamically alter synapse strength. This process is regulated by PDE4A/D, mGluR3, and calbindin, to prevent toxic calcium actions; regulatory actions that are lost with age/inflammation, leading to tau phosphorylation. The current study examined whether a similar "signature of flexibility" expresses in layer II entorhinal cortex, investigating the localization of PDE4D, mGluR3, and HCN1 channels. Results showed a similar pattern to dorsolateral prefrontal cortex, with PDE4D and mGluR3 positioned to regulate internal calcium release near glutamate synapses, and HCN1 channels concentrated on spines. As layer II entorhinal cortex stellate cells do not express calbindin, even when young, they may be particularly vulnerable to magnified calcium actions and ensuing tau pathology., (© The Author(s) 2023. Mitochondrial malfunction and morphologic disorganization have been observed in brain cells as part of complex pathological changes. However, it is unclear what may be the role of mitochondria in the initiation of pathologic processes or if mitochondrial disorders are consequences of earlier events. We analyzed the morphologic reorganization of organelles in an embryonic mouse brain during acute anoxia using an immunohistochemical identification of the disordered mitochondria, followed by electron microscopic three-dimensional (3D) reconstruction. We found swelling of the mitochondrial matrix after 3 h anoxia and probable dissociation of mitochondrial stomatin-like protein 2 (SLP2)-containing complexes after 4.5 h anoxia in the neocortex, hippocampus, and lateral ganglionic eminence. Surprisingly, deformation of the Golgi apparatus (GA) was detected already after 1 h of anoxia, when the mitochondria and other organelles still had a normal ultrastructure. The disordered GA showed concentrical swirling of the cisternae and formed spherical onion-like structures with the trans-cisterna in the center of the sphere. Such disturbance of the Golgi architecture likely interferes with its function for post-translational protein modification and secretory trafficking. Thus, the GA in embryonic mouse brain cells may be more vulnerable to anoxic conditions than the other organelles, including mitochondria.

Glutamate carboxypeptidase-II (GCPII) expression in brain is increased by inflammation, e.g. by COVID19 infection, where it reduces NAAG stimulation of metabotropic glutamate receptor type 3 (mGluR3). GCPII-mGluR3 signaling is increasingly linked to higher cognition, as genetic alterations that weaken mGluR3 or increase GCPII signaling are associated with impaired cognition in humans. Recent evidence from macaque dorsolateral prefrontal cortex (dlPFC) shows that mGluR3 are expressed on dendritic spines, where they regulate cAMP-PKA opening of potassium (K + ) channels to enhance neuronal firing during working memory. However, little is known about GCPII expression and function in the primate dlPFC, despite its relevance to inflammatory disorders. The present study used multiple label immunofluorescence and immunoelectron microscopy to localize GCPII in aging macaque dlPFC, and examined the effects of GCPII inhibition on dlPFC neuronal physiology and working memory function. GCPII was observed in astrocytes as expected, but also on neurons, including extensive expression in dendritic spines. Recordings in dlPFC from aged monkeys performing a working memory task found that iontophoresis of the GCPII inhibitors 2-MPPA or 2-PMPA markedly increased working memory-related neuronal firing and spatial tuning, enhancing neural representations. These beneficial effects were reversed by an mGluR2/3 antagonist, or by a cAMP-PKA activator, consistent with mGluR3 inhibition of cAMP-PKA-K + channel signaling. Systemic administration of the brain penetrant inhibitor, 2-MPPA, significantly improved working memory performance without apparent side effects, with largest effects in the oldest monkeys. We propose a novel non-toxic method of diagnostic biomarker extraction and concentration from biofluids. The method is based on the usage of (1) magnetic nanoparticles of a few nanometres in size bearing molecular traps for biomarkers on their surface and (2) additional larger (several tens of nanometres) magnetic nanoparticles for catching smaller magnetic nanoparticles in a strong magnetic field gradient with their consequent concentration into the detection area. It is shown that the interference of an external permanent gradient magnetic field with the magnetic field of large magnetic nanoparticles allows one to catch small magnetic nanoparticles from their trajectories in a fluid at a distance around ten radii of the large nanoparticles. Theoretical analysis and mathematical simulation show the validity of the proposed non-toxic method for fast and robust biomarker extraction and concentration for increasing the sensitivity of biomarker detection. The hippocampal-entorhinal system supports cognitive functions, has lifelong neurogenic capabilities in many species, and is selectively vulnerable to Alzheimer's disease. To investigate neurogenic potential and cellular diversity, we profiled single-nucleus transcriptomes in five hippocampal-entorhinal subregions in humans, macaques, and pigs. Integrated cross-species analysis revealed robust transcriptomic and histologic signatures of neurogenesis in the adult mouse, pig, and macaque but not humans. Doublecortin (DCX), a widely accepted marker of newly generated granule cells, was detected in diverse human neurons, but it did not define immature neuron populations. To explore species differences in cellular diversity and implications for disease, we characterized subregion-specific, transcriptomically defined cell types and transitional changes from the three-layered archicortex to the six-layered neocortex. Notably, METTL7B defined subregion-specific excitatory neurons and astrocytes in primates, associated with endoplasmic reticulum and lipid droplet proteins, including Alzheimer's disease-related proteins.
