Inflammation is the critical component of neuropathic pain; therefore, this study aimed to assess the potential anti-inflammatory effects of Cannabis sativa L. extracts in a vincristine-induced model of neuropathic pain. The effects of different doses (5.0–40.0 mg/kg) of two Cannabis sativa L. extracts (B and D) on COX-1, COX-2, TNF-α, and NF-κB mRNA and protein levels were examined in the rat hippocampus, cerebral cortex, and blood lymphocytes. There were statistically significant differences in COX-1, COX-2, and TNF-α mRNA and protein expression in the hippocampus and cerebral cortex, with significant differences in COX-2 and TNF-α in the lymphocytes. Extract D dose-dependently increased COX-1 mRNA and protein in the hippocampus and cortex. In contrast, Extract B dose-dependently increased COX-1 mRNA and decreased COX-2 mRNA (in a dose of 7.5 mg/kg) and TNF-α protein levels in the cortex. Cannabis sativa L. extracts significantly influenced the expression of inflammatory genes and proteins, with effects varying based on dose and tissue type. The increased expression of COX-1, COX-2, and TNF-α (in comparison to groups receiving NaCl, vincristine, and gabapentin) in the rat hippocampus and COX-1 in the cerebral cortex suggests that Cannabis may have a pro-inflammatory effect. Due to species specificity, the results of our research based on rats require confirmation in humans. However, Cannabis sativa should be recommended with caution for treating pain with an inflammatory component.

In a power grid with multiple two-state loads, the total power can vary over a significant range. This results in the inability to supply this system from a low-power source. The solution is an algorithm that shapes energy demand depending on its availability. For this purpose, a new load distribution method is proposed based on introducing a buffer between the temperature controller output and the heater and filtering the load using a master Proportional–Integral (PI) controller. The aim of the work was to evaluate the quality of the developed algorithm for limiting power peaks in the power grid. The research was conducted on a model of the Creep Test Laboratory with 389 heaters simulated in MATLAB Simulink R2023b. The algorithm was tested with various settings of the master controller parameters. By experimentally adjusting these parameters, a ten-fold reduction in peak power was achieved. The standard deviation for the L1 phase was reduced from 7.6 kW to 0.6 kW. Similar results were obtained for phases L2 and L3. The tested control system tracked changes in the average power value by changing the number of loads switched on and by frequency-modulating the signal when the change was less than the power of a single load. It was demonstrated that the controlled delayed switching of electrical loads can modify the shape of the total electric power without affecting their operation. The proposed solution features a low computational complexity, which allows its implementation in various systems.

Hypercholesterolemia is a condition characterized by total cholesterol levels exceeding 200 mg/dl. A breakthrough in its treatment was the introduction of the first representative of the status - lovastatin. The mechanism of their action relies on inhibition of the activity of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase, the enzyme responsible for converting HMG-CoA to mevalonic acid, the main substrate in the synthesis of endogenous cholesterol. The progress of experimental and clinical studies on action statins indicates a number of other biological properties of this group of compounds, and thus their pharmacological potential. The possibility of their use in the pharmacotherapy of cardiovascular diseases, hyperandrogenism in women with polycystic ovary syndrome (PCOS), osteoporosis, type 2 diabetes, neurodegenerative diseases, or cancer has been so far highlighted. The aim of this paper was to systematize the knowledge of statin drugs in terms of the progress of studies on their biological activity indicating the possibility of their potential use in the treatment of diseases other than those related to the cardiovascular system, along with verification of their safety profile and demonstrated adverse effects (DN) as a result of their intake. The literature review was conducted by a screening of the following scientific databases - PubMed and Google Scholar (the analysis lasted from March 2022 to February 2023). The keywords used for searching were disease-related and DN-related. Statins have shown potential to alleviate symptoms of androgen excess in women suffering from polycystic ovary syndrome (PCOS). Studies relating to their effect on the treatment of Alzheimer's disease and dementia indicate a reduction in progressive neurodegenerative changes, and thus suggest their potential use in the prevention and treatment of comorbid cardiovascular disease. Moreover, statins tend to reduce morbidity, increase survival, and alleviate the progression of cancer. The results of studies of the effects of these drugs on the possibility or development of Parkinson's disease, osteoporosis and the etiology of muscle symptoms appear to be inconsistent in the relationship of their benefits to DN. Unfortunately, it seems that they have also led to disturbances in carbohydrate metabolism and increased the risk of type II diabetes mellitus (NODM). Studies on the effect of the nocebo effect during statin therapy on the frequency with which patients experience DN have confirmed the presence of this effect. Analysis of the collected studies shows considerable potential for expanding the current therapeutic indications of the drugs in question. However, further randomized controlled trials are required to determine the precise effective dose, duration of therapy for a specific statin.

Introduction: Cannabis sativa L. (CSL) extract has pain-relieving potential due to its cannabinoid content, so the effects of two CSL extracts on alleviating neuropathic pain were investigated in vivo. Methods and groups: Male Wistar rats (n = 130) were divided into groups and received vincristine (0.1 mg/kg) and gabapentin (60 mg/kg) to induce and relieve neuropathic pain or CSL extracts (D and B). The mRNA and protein expression of the cannabinoid receptors type 1 and 2 (CB1R, CB2R) were evaluated in the cerebral cortex, hippocampus, and lymphocytes. Behavioural tests (Tail-Flick and von Frey) were performed on all animals. Results: VK-induced neuropathic pain was accompanied by decreased CB1R protein level and CB2R mRNA expression in the cortex. Gabapentin relieved pain and increased CB1R protein levels in the hippocampus compared to the vincristine group. Hippocampus CB1R protein expression increased with the administration of extract D (10 mg/kg, 40 mg/kg) and extract B (7.5 mg/kg, 10 mg/kg) compared to VK group. In the cerebral cortex CSL decreased CB1R protein expression (10 mg/kg, 20 mg/kg, 40 mg/kg of extract B) and mRNA level (5 mg/kg, 7.5 mg/kg of extract B; 20 mg/kg of extract D) compared to the VK-group.CB2R protein expression increased in the hippocampus after treatment with extract B (7.5 mg/kg) compared to the VK-group. In the cerebral cortex extract B (10 mg/kg, 20 mg/kg) increased CB2R protein expression compared to VK-group. Conclusion: Alterations in cannabinoid receptor expression do not fully account for the observed behavioural changes in rats. Therefore, additional signalling pathways may contribute to the initiation and transmission of neuropathic pain. The Cannabis extracts tested demonstrated antinociceptive effects comparable to gabapentin, highlighting the antinociceptive properties of Cannabis extracts for human use.

The aim of the study was to investigate the effect of baicalein or Scutellaria baicalensis root extract interaction with methyldopa in pregnant spontaneously hypertensive rats (SHR) at the pharmacodynamic, molecular, and biochemical levels. The rats, after confirming pregnancy, received baicalein (200 mg/kg/day, p.o.) and extract (1000 mg/kg/day, p.o.), in combination with methyldopa (400 mg/kg/day; p.o.), for 14 consecutive days, 1 h before blood pressure and heart rate measurements. In the heart and placenta from mothers after giving birth to their offspring, mRNA expression of factors related to inflammatory processes (TNF-α, Il-1β, IL-6) and vascular diseases (TGF-β, HIF-1α, VEGF, PlGF) was measured. Levels of markers of oxidative stress (superoxide dismutase and malondialdehyde) in the placenta and indicators of myocardial damage (troponin cTnC and cTnI, creatine kinase, myoglobin, and lactate dehydrogenase) in the heart were also assessed. Baicalein co-administered with methyldopa was associated with reduced blood pressure, especially during the first three days. The interactions were more pronounced for such factors as TGF-β, HIF-1α, VEGF, and PlGF than TNF-α, Il-1β, and IL-6. Combined application of baicalein and extract with methyldopa may be of value in the development of a new antihypertensive medication intended for patients suffering from preeclampsia or pregnancy-induced hypertension.

Recent research trends show an increasing interest in non-equilibrium plasmas operated at atmospheric pressure, which are often used to tackle several environmental and health issues. Nevertheless, due to the complexity of the applications, these trends also show the need for a comprehensive characterization of such plasmas for a deeper understanding of the observed effects. One of the diagnostic methods for experimental determination of key parameters which affect the reactivity of a plasma, i.e., electron temperature, electron density and heavy particle temperature, is laser scattering. In this work, an approach based on a simple and compact laser scattering setup is proposed, which allows an estimation of the above parameters without any additional changes in the acquisition settings. Thus, the experimental effort and possible sources of error can be reduced. The proposed setup is tested experimentally with a commercially available pulsed plasma system, and the results are compared to available data. From this comparison, it is found that the plasma parameters estimated with the proposed scattering setup are plausible.

The advantages of pulsed low-current high-voltage discharges operated at atmospheric pressure and the ease with which such discharges can be implemented industrially contributed to their popularity. However, the high reactivity of a pulsed plasma implies that thorough diagnostics are needed to fully understand the interactions inside these plasmas. Some of the key parameters determining plasma properties of low-current discharges are the electron number density and the temperature of heavy particles. Both parameters can be determined experimentally with spectroscopic techniques, for example by investigating the broadening of spectral lines due to the Stark effect and by fitting synthetic spectra to molecular transitions. To the authors’ knowledge, experimentally determined electron densities for pulsed low-current discharges operated in a power range between 300 W and 1000 W have not been performed in previous works. Thus, in this work, the electron number density and temperature of heavy particles of one of several commercially available plasma systems are determined by means of emission spectroscopy.

Non-equilibrium conditions in plasma are often achieved by pulsed power delivery, where the pulse shape and repetition rate determine the properties of the plasma constituents and thus its chemical reactivity. The evaluation of the latter is becoming increasingly important to understand the observed effects, especially when new application fields are targeted. The composition of the plasma and the occurring chemical reactions can be calculated using various models. Thereby, the temperature of the electrons, the electron number density, as well as the heavy particle temperature are usually required as the basis of such calculations. In this work, the influence of pulse amplitude and repetition rate on these plasma parameters is determined by laser scattering for a low-current, high-voltage discharge operated with nitrogen at atmospheric pressure. In particular, the characteristic parameters regarding the plasma free electrons in such discharges have not yet been experimentally determined to this extent. The results are validated by spectroscopic measurements, i.e., the electron density is estimated from the Stark broadening of the hydrogen beta line and the heavy particle temperature is estimated by fitting the spectrum of nitrogen molecular transitions. Depending on the operating frequency, a pure nitrogen discharge with an input power of about 650 W displays an electron density between 1.7×1021m−3 and 2.0×1021m−3 with electron temperatures in the range of 40,000 K and heavy particle temperatures of about 6000 K in the core of the discharge channel. Furthermore, a relatively slow electron recombination rate in the range of 20 µs is observed.
