Abstract

The temperature of the upper 300 m of the North Atlantic increased by about 0.57°C between 1984 and 1999. but this underlying trend was overlain with substantial geographic and interannual variability. Nor thward shifts occurred in the distribution of many commercial and non-commercial fish species in the NE Atlantic during the 1990s. New records were established for a number of Mediterranean and NW African species on the south coast of Portugal. Red mullet (Mullus surmuletus) and bass (Dicentrarchus labrax) extended their ranges northward to western Norway and catches of the former increased throughout the 1990s in the North Sea. Abundance or relative abundance o f warm-water commercial species of gadoids and flatfish generally increased during the 1990s, but like the warming trend the changes in distribution and abundance were by no means uniform and there was considerable interannual variability. There were also examples o f southward shifts lor some species, which can be related to local hydrographic conditions, such as upwelling. Information on distribution and abundance of Greenland cod (Gadus morhua L.) and Norwegian springspawning herring (Clupea harengus) during a previous warming period in the late 1920s and 1930s is also presented and compared with changes in the 1990s. Abstract

Background: Tuberculosis diagnosis might be delayed or missed in children with severe pneumonia because this diagnosis is usually only considered in cases of prolonged symptoms or antibiotic failure. Systematic tuberculosis detection at hospital admission could increase case detection and reduce mortality. Methods: We did a stepped-wedge cluster-randomised trial in 16 hospitals from six countries (Cambodia, Cameroon, Côte d'Ivoire, Mozambique, Uganda, and Zambia) with high incidence of tuberculosis. Children younger than 5 years with WHO-defined severe pneumonia received either the standard of care (control group) or standard of care plus Xpert MTB/RIF Ultra (Xpert Ultra; Cepheid, Sunnyvale, CA, USA) on nasopharyngeal aspirate and stool samples (intervention group). Clusters (hospitals) were progressively switched from control to intervention at 5-week intervals, using a computer-generated random sequence, stratified on incidence rate of tuberculosis at country level, and masked to teams until 5 weeks before switch. We assessed the effect of the intervention on primary (12-week all-cause mortality) and secondary (including tuberculosis diagnosis) outcomes, using generalised linear mixed models. The primary analysis was by intention to treat. We described outcomes in children with severe acute malnutrition in a post hoc analysis. This study is registered with ClinicalTrials.gov (NCT03831906) and the Pan African Clinical Trial Registry (PACTR202101615120643). Findings: From March 21, 2019, to March 30, 2021, we enrolled 1401 children in the control group and 1169 children in the intervention group. In the intervention group, 1140 (97·5%) children had nasopharyngeal aspirates and 942 (80·6%) had their stool collected; 24 (2·1%) had positive Xpert Ultra. At 12 weeks, 110 (7·9%) children in the control group and 91 (7·8%) children in the intervention group had died (adjusted odds ratio [OR] 0·986, 95% CI 0·597–1·630, p=0·957), and 74 (5·3%) children in the control group

Abstract

Monte Carlo (MC) simulation can be used in teaching the physics of ionizing radiation with applications in medical physics. However, the free MC codes available are not user-friendly for new and inexperienced users, limiting their use as an educational tool. This study focuses on the development of a user-friendly graphical interface for MC simulations with a focus on learning the interaction of ionizing radiation applied to Medical Physics. The interface is an executable file developed in Python 3.9. The interface serves as a frontend for the simulations performed by the PENELOPE v. 2014 code with the penEasy v. 2015 extension. Two types of simulations are available: (i) Particle tracking and (ii) Dose deposition. The results obtained in case (i) are presented graphically as three figures that show the trajectory of electrons, photons and positrons, respectively. For case (ii), the result is the dose as a function of the depth of a box-shaped volume. In summary, this interface, with simple geometry and fast simulations, in an interactive environment, offers a wide range of possibilities for understanding the interaction of radiation with matter, which is the basis of medical physics. 17. Risk Factors in Serrated Pathway Lesions: N-Glycosylation Profile as a Potential Biomarker of Progression to Malignancy.

Abstract

Introduction: The serrated pathway contributes to interval colorectal cancers, highlighting the need for new biomarkers to assess lesion progression risk. The β1,6-GlcNAc branched N -glycans expression in CRC cells was associated with an invasive phenotype and with immune evasion. Therefore, this study aims to identify potential risk factors for progression of serrated lesions (SLs) to malignancy, analyzing the N -glycosylation profile of epithelial/infiltrating immune cells., Methods: A retrospective cohort study was performed with data from 53 colonoscopies (48 patients). Sixty-three serrated pathway lesions (SPLs) were characterized based on N -glycosylation profile (lectin histochemistry/flow cytometry) and MGAT5 expression. Statistical analysis was performed to search for associations between the glycoprofile and clinical variables from each patient., Results: Increased β1,6-GlcNAc branched N -glycans expression in epithelial cells is found associated with age ( p = 0.007 in SPL), smoking ( p = 0.038 in SL), increased BMI ( p = 0.036 in sessile serrated lesions [SSL]), and polyp dimensions ≥10 mm ( p = 0.001 in SL), while increased expression of these structures on immune cells is associated with synchronous CA number (CD4 + T cells: p = 0.016; CD8 + T cells: p = 0.044 in SL) and female gender ( p = 0.026 in SL). Moreover, a lower high-mannose N -glycans expression in immune cells is associated with smoking ( p = 0.010 in SPL) and synchronous CA presence ( p = 0.010 in SPL). Higher expression of these glycans is associated with female ( p = 0.016 in SL) and male ( p = 0.044 in SL) gender, left colon location ( p = 0.028), dysplasia ( p = 0.028), and adenocarcinoma ( p = 0.010)., Conclusions: We identified an association between an abnormal glycoprofile and several clinical risk factors, proposing the N- glycosylation profile as a potential biomarker of tumor progression in the serrated pathway. Abstract

Advancing from gene discovery in autism spectrum disorders (ASDs) to the identification of biologically relevant mechanisms remains a central challenge. Here, we perform parallel in vivo functional analysis of 10 ASD genes at the behavioral, structural, and circuit levels in zebrafish mutants, revealing both unique and overlapping effects of gene loss of function. Whole-brain mapping identifies the forebrain and cerebellum as the most significant contributors to brain size differences, while regions involved in sensory-motor control, particularly dopaminergic regions, are associated with altered baseline brain activity.
