Abstract Fieldwork, including work done at sea, is a key component of many geoscientists' careers. Recent studies have highlighted the pervasive harassment faced by women and LGBTQ+ people during fieldwork. However, transgender and gender diverse (TGD) scientists face obstacles which have not yet been thoroughly examined. We fill this gap by sharing our experiences as TGD people. We have experienced sexual harassment, misconduct, privacy issues, and legal and medical struggles as we conduct seagoing work. In this work, we provide recommendations for individuals, cruise leaders, and institutions for making seagoing work safer for our communities.

Abstract Twin girls born at 30 weeks' gestation with spinal muscular atrophy (SMA) received onsasemnogene‐abeparvovec (OA) at 3.5 weeks of life. They had no treatment‐related adverse events, normal acquisition of motor milestones, and normal neurological examination at 19 months. Genotyping revealed 0 copies of SMN1 and a single, hybrid SMN2 gene containing the positive genetic modifier c.835‐44A>G. This was associated with full‐length SMN2 blood mRNA expression levels similar to a 2 copy SMA infant. The observed favorable outcomes are likely due to the genetic modifier combined with early drug administration enabled by prematurity.

Abstract Understanding how biotic interactions shape ecosystems and impact their functioning, resilience and biodiversity has been a sustained research priority in ecology. Yet, traditional assessments of ecological complexity typically focus on species-species interactions that mediate a particular function (e.g., pollination), overlooking both the synergistic effect that multiple functions might develop as well as the resulting species-function participation patterns that emerge in ecosystems that harbor multiple ecological functions. Here we propose a mathematical framework that integrates various types of biotic interactions observed between different species. Its application to recently collected data of an islet ecosystem—reporting 1537 interactions between 691 plants, animals and fungi across six different functions (pollination, herbivory, seed dispersal, decomposition, nutrient uptake, and fungal pathogenicity)—unveils a non-random, nested structure in the way plant species participate across different functions. The framework further allows us to identify a ranking of species and functions, where woody shrubs and fungal decomposition emerge as keystone actors whose removal have a larger-than-random effect on secondary extinctions. The dual insight—from species and functional perspectives—offered by the framework opens the door to a richer quantification of ecosystem complexity and to better calibrate the influence of multifunctionality on ecosystem functioning and biodiversity.

Abstract Although many studies predict extensive future biodiversity loss and redistribution in the terrestrial realm, future changes in marine biodiversity remain relatively unexplored. In this work, we model global shifts in one of the most important marine functional groups—ecosystem-structuring macrophytes—and predict substantial end-of-century change. By modelling the future distribution of 207 brown macroalgae and seagrass species at high temporal and spatial resolution under different climate-change projections, we estimate that by 2100, local macrophyte diversity will decline by 3–4% on average, with 17 to 22% of localities losing at least 10% of their macrophyte species. The current range of macrophytes will be eroded by 5–6%, and highly suitable macrophyte habitat will be substantially reduced globally (78–96%). Global macrophyte habitat will shift among marine regions, with a high potential for expansion in polar regions.

Abstract Objective Cenobamate is an antiseizure medication (ASM) associated with high rates of seizure freedom and acceptable tolerability in patients with focal seizures. To achieve the optimal cenobamate dose for maximal potential effectiveness while avoiding or minimizing drug‐related adverse events (AEs), the administration of cenobamate with other ASMs must be managed through concomitant ASM load reduction. A panel of Spanish epilepsy experts aimed to provide a Spanish consensus on how to adjust the dose of concomitant ASMs in patients with drug‐resistant epilepsy (DRE) in order to improve the effectiveness and tolerability of adjunctive cenobamate. Methods A three‐stage modified Delphi consensus process was undertaken, including six Spanish epileptologists with extensive experience using cenobamate. Based on current literature and their own expert opinion, the expert panel reached a consensus on when and how to adjust the dosage of concomitant ASMs during cenobamate titration. Results The expert panel agreed that tailored titration and close follow‐up are required to achieve the best efficacy and tolerability when initiating cenobamate in patients receiving concomitant ASMs. When concomitant clobazam, phenytoin, phenobarbital, and sodium channel blockers are taken at high dosages, or when the patient is receiving two or more sodium channel blockers, dosages should be proactively lowered during the cenobamate titration period. Other concomitant ASMs should be reduced only if the patient reports a moderate/severe AE at any stage of the titration period. Significance Cenobamate is an effective ASM with a dose‐dependent effect. To maximize effectiveness while maintaining the best tolerability profile, co‐medication management is needed. The recommendations included herein provide practical guidance for proactive and reactive management of co‐medication in cenobamate‐treated patients with DRE and a high drug load. Plain Language Summary Patients with epilepsy may continue to have seizures even after treatment with several different antiseizure medications (ASMs). Cenobamate is an ASM that can reduce seizures in these patients. In this study, six Spanish experts in epilepsy discussed the best way to use cenobamate in drug‐resistant epilepsy. They provide practical guidance on when and how the dose of other ASMs might be adjusted to reduce side effects and optimize the use of cenobamate.

Summary: The blood enzyme glutamate-oxaloacetate transaminase (GOT) has been postulated as an effective therapeutic to protect the brain during stroke. To demonstrate its potential clinical utility, a new human recombinant form of GOT (rGOT) was produced for medical use.We tested the pharmacokinetics and evaluated the protective efficacy of rGOT in rodent and non-human primate models that reflected clinical stroke conditions.We found that continuous intravenous administration of rGOT within the first 8 h after ischemic onset significantly reduced the infarct size in both severe (30%) and mild lesions (48%). Cerebrospinal fluid and proteomics analysis, in combination with positron emission tomography imaging, indicated that rGOT can reach the brain and induce cytoprotective autophagy and induce local protection by alleviating neuronal apoptosis.Our results suggest that rGOT can be safely used immediately in patients suspected of having a stroke. This study requires further validation in clinical stroke populations.

Entrepreneurial ecosystems (EEs) are a growing focus for policymakers and academics seeking to foster innovation and economic growth. While existing literature has analysed various aspects of EEs, limited research has examined how public policies can contribute to their development. Through a single-case study, this study addresses this gap by analysing the role of a regional government in overcoming the challenges of building an EE. We explore the creation and development of the Biscay Startup Bay (BSB) EE in Bilbao City (Bizkaia, Basque Country, Spain). We describe the unique framework that they crafted and implemented, highlighting the key attributes that generate value for all stakeholders. This single-case study of the BSB EE in Spain offers valuable insights for policymakers seeking to design and implement successful Open Innovation-driven approaches to creating and developing regional EEs.

BackgroundPediatric kidney transplantations are rarely performed, and there is limited knowledge about the diversity in current clinical practices across Europe. This study aims to explore the utility of clinical snapshot studies in identifying these disparities, establishing a foundation for future snapshot studies and standardization efforts.MethodsA pilot clinical snapshot study was conducted, with invitations extended to all 109 pediatric kidney transplant centres in Europe. Each participating centre provided pre-, peri-, and postoperative data concerning their most recent thirty transplantations. The primary outcomes encompassed the evaluation of disparities in donor-recipient selection, surgical techniques, post-operative drainage procedures, and immunosuppressive therapy protocols. Secondary outcomes involved the analysis of rejection rates, incidence of infections, and graft survival.ResultsThe study involved 439 patients from fifteen centres (14%) in twelve countries, with varying transplant volumes (range 1–29 transplantations per year) and follow-up periods. Significant differences were found among centres in terms of donor types, cold and warm ischemia time, pre-emptive transplant rates, and kidney transplant drainage methods. The rate of living donors varied between 3% and 90% and the median duration of cold ischemia ranged was 770 min after deceased donation and 147 min after living donation. Basiliximab was the dominant induction therapy, yet steroid withdrawal varied widely. Infection, rejection, and graft survival rates also varied significantly between centres.ConclusionThis study revealed substantial variation in clinical practices among European centres performing pediatric kidney transplantations. These findings could serve as a stimulus for international dialogue and collaboration.

Over millennia, mammals have coexisted with humans across the forest-savannah mosaics characterising part of West Africa. While some savannahs have long been used for rice cultivation in Guinea-Bissau, savannas and forests are now being rapidly converted into cashew monocultures. To understand how species cope with such land-use changes, we investigated how mammals make use of forest patches, cashew orchards and rice paddies over two contrasting periods: early and late rainy season. Using camera-trapping, we examined mammal diversity and trophic guild activity across seven landscapes, each with one sampling site per habitat type in Northern Guinea-Bissau. Based on 937 records from 21 species, species richness and activity were similar across habitat types, but generally higher in the late-rainy season. In this season, species composition differed between the cashew orchards and rice paddies. At the guild-level, carnivore activity was lower in the rice paddies than in forests, while herbivore activity was lower in the cashew orchards in comparison to that in forests in the early-rainy season. The relatively similar assemblage-wide diversity across habitat types seems to be enabled by the high habitat heterogeneity, but likely offset by previous extinctions from the area. Yet, guild-level habitat use highlighted the fundamental role of forests to sustain functionally healthy mammal assemblages. Maintaining habitat heterogeneity, including patches of native forests, seems to be key in optimizing the long-term conservation of mammal diversity in rural West Africa.

Background and objectives: Fibrosis contributes to 45% of deaths in industrialized nations and is characterized by an abnormal accumulation of extracellular matrix (ECM). There are no specific anti-fibrotic treatments for liver fibrosis, and previous unsuccessful attempts at drug development have focused on preventing ECM deposition. Because liver fibrosis is largely acknowledged to be reversible, regulating fibrosis resolution could offer novel therapeutical options. However, little is known about the mechanisms controlling ECM remodeling during resolution. Changes in proteolytic activity are essential for ECM homeostasis and macrophages are an important source of proteases. Herein, in this study we evaluate the role of macrophage-derived cathepsin D (CtsD) during liver fibrosis. Methods: CtsD expression and associated pathways were characterized in single-cell RNA sequencing and transcriptomic datasets in human cirrhosis. Liver fibrosis progression, reversion and functional characterization were assessed in novel myeloid-CtsD and hepatocyte-CtsD knock-out mice. Results: Analysis of single-cell RNA sequencing datasets demonstrated CtsD was expressed in macrophages and hepatocytes in human cirrhosis. Liver fibrosis progression, reversion and functional characterization were assessed in novel myeloid-CtsD (CtsDΔMyel) and hepatocyte-CtsD knock-out mice. CtsD deletion in macrophages, but not in hepatocytes, resulted in enhanced liver fibrosis. Both inflammatory and matrisome proteomic signatures were enriched in fibrotic CtsDΔMyel livers. Besides, CtsDΔMyel liver macrophages displayed functional, phenotypical and secretomic changes, which resulted in a degradomic phenotypical shift, responsible for the defective proteolytic processing of collagen I in vitro and impaired collagen remodeling during fibrosis resolution in vivo. Finally, CtsD-expressing mononuclear phagocytes of cirrhotic human livers were enriched in lysosomal and ECM degradative signaling pathways. Conclusions: Our work describes for the first-time CtsD-driven lysosomal activity as a central hub for restorative macrophage function during fibrosis resolution and opens new avenues to explore their degradome landscape to inform drug development.

Abstract Audiovisual news is a critical cultural phenomenon that has been influencing audience worldviews for more than a hundred years. To understand historical trends in multimodal audiovisual news, we need to explore them longitudinally using large sets of data. Despite promising developments in film history, computational video analysis, and other relevant fields, current research streams have limitations related to the scope of data used, the systematism of analysis, and the modalities and elements to be studied in audiovisual material and its metadata. Simultaneously, each disciplinary approach contributes significant input to research reducing these limitations. We therefore advocate for combining the strengths of several disciplines. Here we propose a multidisciplinary framework for systematically studying large collections of historical audiovisual news to gain a coherent picture of their temporal dynamics, cultural diversity, and potential societal effects across several quantitative and qualitative dimensions of analysis. By using newsreels as an example of such complex historically formed data, we combine the context crucial to qualitative approaches with the systematicity and ability to cover large amounts of data from quantitative methods. The framework template for historical newsreels is exemplified by a case study of the “News of the Day” newsreel series produced in the Soviet Union during 1944–1992. The proposed framework enables a more nuanced analysis of longitudinal collections of audiovisual news, expanding our understanding of the dynamics of global knowledge cultures.

Abstract Background The emergence of new SARS-CoV-2 variants and the waning of immunity raise concerns about vaccine effectiveness and protection against COVID-19. While antibody response has been shown to correlate with the risk of infection with the original variant and earlier variants of concern, the effectiveness of antibody-mediated protection against Omicron and the factors associated with protection remain uncertain. Methods We evaluated antibody responses to SARS-CoV-2 spike (S) and nucleocapsid (N) antigens from Wuhan and variants of concern by Luminex and their role in preventing breakthrough infections 1 year after a third dose of mRNA vaccination, in a cohort of health care workers followed since the pandemic onset in Spain (N = 393). Data were analyzed in relation to COVID-19 history, demographic factors, comorbidities, vaccine doses, brand, and adverse events. Results Higher levels of anti-S IgG and IgA to Wuhan, Delta, and Omicron were associated with protection against vaccine breakthroughs (IgG against Omicron S antigen HR, 0.06, 95%CI, 0.26–0.01). Previous SARS-CoV-2 infection was positively associated with antibody levels and protection against breakthroughs, and a longer time since last infection was associated with lower protection. In addition, priming with BNT162b2 followed by mRNA-1273 booster was associated with higher antibody responses than homologous mRNA-1273 vaccination. Conclusions Data show that IgG and IgA induced by vaccines against the original strain or by hybrid immunization are valid correlates of protection against Omicron BA.1 despite immune escape and support the benefits of heterologous vaccination regimens to enhance antibodies and the prioritization of booster vaccination in individuals without recent infections.

Newsreels, short documentary news films, were an influential channel of mass communication and propaganda in the Soviet Union. They served as an important means of visualizing the world for audiences in the way the Soviet authorities wanted it to be depicted. Studies in Soviet visual culture have recognized both continuities of repeating patterns and changes in the post-World War II period. This understanding is based primarily on temporally limited source selections, while a more systematic study of the developments in Soviet visual culture over a longer period is pending. In this article, we reveal long-term continuities, subtle changes, and sudden shifts in the official visual discourse in the Soviet newsreel series ‘Novosti dnia’ (News of the Day) 1945 to 1992. We study visual patterns in approximately 1,700 digitized newsreel issues, each about ten minutes long, using multidimensional vector embeddings. These embeddings, produced from the central frames of 205,678 shots, help visually evaluate the footage and assess visual similarities based on ResNet50 feature vectors. For this, we use the _Collection Space Navigator_ tool. The article demonstrates how multidimensional vector embeddings can be used to study the internal time of the films, and the external time of the years running by.

Genetic differentiation between populations inhabiting ecologically different habitats might appear because of limited dispersal and gene flow, which may lead to patterns of phenotypic divergence and local adaptation. In this study, we use dispersal, genotypic (24 microsatellite loci) and phenotypic (body size and clutch size) data to analyse patterns of genetic structuring and phenotypic divergence in a blue tit (Cyanistes caeruleus) population inhabiting a continuous and heterogeneous woodland along a valley. The two slopes of the valley differ in their forest formations and environmental conditions. Findings showed that most blue tits reproduced within their natal slope. Accordingly, microsatellite analyses revealed that populations of blue tits established in the two slopes show subtle genetic differentiation. The two genetic populations diverged in clutch size, exceeding the level of differentiation expected based on genetic drift, hence suggesting divergent selection (or other processes promoting divergence) on this life-history trait. Our findings reveal that restricted dispersal and spatial heterogeneity may lead to genetic differentiation among bird populations at a surprisingly small scale. In this respect, it is worth highlighting that such differentiation occurs for an organism with high dispersal capacity and within a continuous woodland. Moreover, we show that small-scale ecological differences, together with limited gene flow, can result in selection favouring different phenotypes even within the same continuum population.

Abstract At the end of 2020, an outbreak of HPAI H5N8 was registered in captive African houbara bustards (Chlamydotis undulata) in the United Arab Emirates. In order to better understand the pathobiology of this viral infection in bustards, a comprehensive pathological characterization was performed. A total of six birds were selected for necropsy, histopathology, immunohistochemistry, RNAscope in situ hybridization and RT-qPCR and nanopore sequencing on formalin-fixed and paraffin-embedded (FFPE) tissue blocks. Gross lesions included mottled and/or hemorrhagic pancreas, spleen and liver and fibrinous deposits on air sacs and intestine. Necrotizing pancreatitis, splenitis and concurrent vasculitis, hepatitis and fibrino-heterophilic peritonitis were identified, microscopically. Viral antigens (nucleoprotein) and RNAs (matrix gene) were both detected within necro-inflammatory foci, parenchymal cells, stromal cells and endothelial cells of affected organs, including the myenteric plexus. Molecular analysis of FFPE blocks successfully detected HPAI H5N8, further confirming its involvement in the lesions observed. In conclusion, HPAI H5N8 in African houbara bustards results in hyperacute/acute forms exhibiting marked pantropism, endotheliotropism and neurotropism. In addition, our findings support the use of FFPE tissues for molecular studies of poorly characterized pathogens in exotic and endangered species, when availability of samples is limited.

[INLINE:1] Plaques of amyloid-β (Aβ) and neurofibrillary tangles are the main pathological characteristics of Alzheimer's disease (AD). However, some older adult people with AD pathological hallmarks can retain cognitive function. Unraveling the factors that lead to this cognitive resilience to AD offers promising prospects for identifying new therapeutic targets. Our hypothesis focuses on the contribution of resilience to changes in excitatory synapses at the structural and molecular levels, which may underlie healthy cognitive performance in aged AD animals. Utilizing the Morris Water Maze test, we selected resilient (asymptomatic) and cognitively impaired aged Tg2576 mice. While the enzyme-linked immunosorbent assay showed similar levels of Aβ42 in both experimental groups, western blot analysis revealed differences in tau pathology in the pre-synaptic supernatant fraction. To further investigate the density of synapses in the hippocampus of 16–18 month-old Tg2576 mice, we employed stereological and electron microscopic methods. Our findings indicated a decrease in the density of excitatory synapses in the stratum radiatum of the hippocampal CA1 in cognitively impaired Tg2576 mice compared with age-matched resilient Tg2576 and non-transgenic controls. Intriguingly, through quantitative immunoelectron microscopy in the hippocampus of impaired and resilient Tg2576 transgenic AD mice, we uncovered differences in the subcellular localization of glutamate receptors. Specifically, the density of GluA1, GluA2/3, and mGlu5 in spines and dendritic shafts of CA1 pyramidal cells in impaired Tg2576 mice was significantly reduced compared with age-matched resilient Tg2576 and non-transgenic controls. Notably, the density of GluA2/3 in resilient Tg2576 mice was significantly increased in spines but not in dendritic shafts compared with impaired Tg2576 and non-transgenic mice. These subcellular findings strongly support the hypothesis that dendritic spine plasticity and synaptic machinery in the hippocampus play crucial roles in the mechanisms of cognitive resilience in Tg2576 mice.

Introduction: We conducted a comprehensive comparative analysis of the Okazaki, Umehara, and American College of Rheumatology/European League Against Rheumatism (ACR/EULAR) classification criteria for diagnosing immunoglobulin G4-related disease (IgG4-RD). Materials and Methods: A retrospective study was conducted in a single tertiary hospital, using expert clinical judgment as the gold standard. We compared the diagnostic accuracy of the Okazaki, Umehara, and ACR/EULAR criteria in a cohort of 41 patients with suspected IgG4-RD. We assessed sensitivity, specificity, and positive and negative predictive values for each criterion, and conducted a separate analysis based on four IgG4-RD subtypes. Results: A total of 30 patients were confirmed to have IgG4-RD and 11 were identified as mimickers. The Umehara criteria demonstrated the highest sensitivity (83.33%), followed by the ACR/EULAR 2019 (66.67%) and Okazaki (60.0%) criteria. All three criteria exhibited 100% specificity, with overall diagnostic accuracy ranging from 70% to 88%. The areas under the curve (AUC) were 0.917 (Umehara), 0.800 (Okazaki), and 0.833 (ACR/EULAR 2019), indicating significant diagnostic effectiveness (p < 0.000). Subtype analysis revealed that the Umehara and ACR/EULAR 2019 criteria were more effective in diagnosing pancreato-hepato-biliary involvement (subtype 1), while the Okazaki and ACR/EULAR 2019 criteria were more effective in diagnosing retroperitoneal fibrosis and/or aortitis (subtype 2). Conclusions: Our study provides valuable insights into the diagnostic performance of the Okazaki, Umehara, and ACR/EULAR criteria for a cohort of patients with suspected IgG4-RD. The Umehara criterion demonstrated the highest sensitivity, suggesting its potential utility for screening purposes, while all three criteria showed consistent specificity.

Wild food plants (WFPs) are crucial for the subsistence of many human populations. While there are at least 7000 edible plant species in the world, only approximately 420 are considered food crops. WFPs are often studied from the phytochemical and pharmacological point of view, because they include available food components with nutraceutical value. The present study aims to highlight the nutritional value of WFPs traditionally used in the Catalan linguistic area, providing detailed insights and discussing the significance of these properties. Information about the nutritional properties of 93 taxa, coming from ethnobotanical prospection, has been collected through an extensive bibliographic research. The results reveal that WFPs are rich in nutrients, especially micronutrients. Furthermore, in selected species, those for which nutritional information and a cultivated homologue are available, the nutrient content in wild taxa exceeds than of phylogenetically related crop plants with similar use. Traditional wild plant preparation forms for food and the nutritional value of a menu constituted by wild food plants are presented. This research represents a preliminary step toward selecting certain taxa that could be developed into new small- or large-scale crops or sustainably harvested in the wild, contributing to food security.

Is there a link between levels of terrorism and the presence of organized crime groups? Which factors does terrorism affect that could impact the presence and expansion of organized crime groups? This study aims to empirically analyze this link. Dealing with potential endogeneity bias using matching methods, we provide quantitative evidence showing that terrorist attacks increase the future levels of organized crime group activities. Using Structural Equation Modeling techniques (SEM), we also show that the main mechanisms through which this relation seems to occur are through the effects of terrorism on state capacity and state legitimacy. Thus, organized crime groups seem to take advantage of the turbulent situation created by terrorist attacks in order to expand their activities. The dataset presented here is an achievement of the H2020 European project "Integrated Pest Management of the Invasive Japanese Beetle, Popillia japonica (IPM-Popillia)". This project addresses the challenge of a new risk to plant health in Europe, the invasion of the Japanese beetle, Popillia japonica (Newman, 1838) (Coleoptera, Rutelidae) and provides an environmentally friendly IPM Toolbox to control the expanding pest populations across Europe. This study aims to present the records of terrestrial arthropod diversity with a special focus on four groups belonging to Carabids and Staphylinid beetles (Coleoptera), Opiliones and Anisolabididae (Dermaptera), collected with the potential to be used as biocontrol agents against P. japonica in future Integrated Pest Management programmes. A thorough sampling programme was conducted in maize and pasture fields in two Islands of the Azores (São Miguel and São Jorge) in the summer of 2022.We provided an inventory of the arthropods recorded in two Azorean agroecosystems (maize and pasture fields) from São Miguel and São Jorge Islands. A total of ten maize and ten pasture fields were sampled and a total of 360 pitfall traps were installed, 216 in São Miguel and 144 in São Jorge, for seven consecutive days in August and September of 2022.We collected 18559 specimens belonging to the phylum Arthropoda, four classes, twelve orders, twenty-six families and forty morphospecies (two identified at the family level as carabid and Staphylinid larvae and 38 identified at the species level). We identified 38 taxa at the species level (n = 18281). Of the 38 identified taxa, 18 species were predators, 15 were plant feeders and five were omnivores. The 18 predators belong to the following families: 10 species were Carabidae, two Staphylinidae, one Anisolabididae, one Chrysopidae, one Leiobunidae, one Nabidae, one Phalangiidae and one Scathophagidae. Concerning the origin of the predators, we recorded five native species: two Carabidae, one Leiobunidae, one Scathophagidae and one Nabidae. The other 13 predator species were introduced or indeterminate.

Abstract The natural world is under unprecedented and accelerating pressure. Much work on understanding resilience to local and global environmental change has, so far, focussed on ecosystems. However, understanding a system’s behaviour requires knowledge of its component parts and their interactions. Here we call for increased efforts to understand ‘biological resilience’, or the processes that enable components across biological levels, from genes to communities, to resist or recover from perturbations. Although ecologists and evolutionary biologists have the tool-boxes to examine form and function, efforts to integrate this knowledge across biological levels and take advantage of big data (e.g. ecological and genomic) are only just beginning. We argue that combining eco-evolutionary knowledge with ecosystem-level concepts of resilience will provide the mechanistic basis necessary to improve management of human, natural and agricultural ecosystems, and outline some of the challenges in achieving an understanding of biological resilience.

Abstract Progressive hepatic damage and fibrosis are major features of chronic liver diseases of different etiology, yet the underlying molecular mechanisms remain to be fully defined. N-RAS, a member of the RAS family of small guanine nucleotide-binding proteins also encompassing the highly homologous H-RAS and K-RAS isoforms, was previously reported to modulate cell death and renal fibrosis; however, its role in liver damage and fibrogenesis remains unknown. Here, we approached this question by using N-RAS deficient (N-RAS−/−) mice and two experimental models of liver injury and fibrosis, namely carbon tetrachloride (CCl4) intoxication and bile duct ligation (BDL). In wild-type (N-RAS+/+) mice both hepatotoxic procedures augmented N-RAS expression in the liver. Compared to N-RAS+/+ counterparts, N-RAS−/− mice subjected to either CCl4 or BDL showed exacerbated liver injury and fibrosis, which was associated with enhanced hepatic stellate cell (HSC) activation and leukocyte infiltration in the damaged liver. At the molecular level, after CCl4 or BDL, N-RAS−/− livers exhibited augmented expression of necroptotic death markers along with JNK1/2 hyperactivation. In line with this, N-RAS ablation in a human hepatocytic cell line resulted in enhanced activation of JNK and necroptosis mediators in response to cell death stimuli. Of note, loss of hepatic N-RAS expression was characteristic of chronic liver disease patients with fibrosis. Collectively, our study unveils a novel role for N-RAS as a negative controller of the progression of liver injury and fibrogenesis, by critically downregulating signaling pathways leading to hepatocyte necroptosis. Furthermore, it suggests that N-RAS may be of potential clinical value as prognostic biomarker of progressive fibrotic liver damage, or as a novel therapeutic target for the treatment of chronic liver disease.

Biological and cultural diversity are inextricably linked and rapidly eroding worldwide. As a response, many conservation efforts foster synergies between cultural and biological diversity agendas through biocultural approaches. However, such approaches do not always address biocultural conflicts, where certain cultural practices can lead to biodiversity loss and, in turn, threaten the continuance of such practices. In this study, we examined a biocultural conflict in the Dimi ceremony, the most important rite of passage of the Daasanach agro-pastoralists of north Kenya, in which skins from threatened carnivore species are used extensively as traditional ornaments. We quantified the current use of skins in Dimi as well as changes in the cultural ceremony that exacerbate its impacts on wildlife. We collected field-based data on the context of the use of skins through structured interviews, focus-group discussions, participant observation and counts of skins in two Dimi ceremonies. We counted a total of 121 skins of four carnivore species being used in a single ceremony. We also found that Dimi has become environmentally unsustainable, threatening distant cheetah Acinonyx jubatus and leopard Panthera pardus populations and local species with spotted skins (African civet Civettictis civetta, common genet Genetta genetta and serval Leptailurus serval). The young Daasanach are deeply concerned about the lack of availability of skins in their area, as well as the prohibitive prices, and they are calling for alternatives to the use of skins in Dimi. Overall, our study shows that acknowledging biocultural conflicts and opening space for dialogue with local communities are essential for the maintenance of both biological and cultural diversity.

Abstract To the human eye, different images appear more or less complex, but capturing this intuition in a single aesthetic measure is considered hard. Here, we propose a computationally simple, transparent method for modeling aesthetic complexity as a multidimensional algorithmic phenomenon, which enables the systematic analysis of large image datasets. The approach captures visual family resemblance via a multitude of image transformations and subsequent compressions, yielding explainable embeddings. It aligns well with human judgments of visual complexity, and performs well in authorship and style recognition tasks. Showcasing the functionality, we apply the method to 125,000 artworks, recovering trends and revealing new insights regarding historical art, artistic careers over centuries, and emerging aesthetics in a contemporary NFT art market. Our approach, here applied to images but applicable more broadly, provides a new perspective to quantitative aesthetics, connoisseurship, multidimensional meaning spaces, and the study of cultural complexity.

IntroductionAtherosclerotic cardiovascular disease (ASCVD) is one of the main causes of morbidity and mortality in developed countries and entails high resources use and costs for health systems. The risk of suffering future cardiovascular (CV) events and the consequent resources use is higher in those patients who have already had a previous cardiovascular event. The objective of the study was to determine the average annual cost of patients with a new or recurrent atherosclerotic CV event during the 2 years after the event.MethodologyRetrospective observational study of electronic medical records of patients from the BIG-PAC® database (7 integrated health areas of 7 Autonomous Communities; n = 1.8 million). Patients with a new or recurrent episode of ASCVD (angina, acute myocardial infarction, transient ischemic attack, stroke, or peripheral arterial disease) between 1-Jan-2017 and 31-Dec-2018 were included. The resources use within two years of the diagnosis was estimated in order to estimate the average cost of patient follow-up.ResultsA total of 26,976 patients with an ASCVD episode were identified during the recruitment period; Out of them, 6,798 had a recurrent event during the follow-up period and 2,414 died. The average costs per patient were €11,171 during the first year and €9,944 during the second year.DiscussionPatients with ASCVD represent a significant economic burden for the health system and for society. Despite the perception that drug costs in the follow-up of chronic patients imply a high percentage of the costs, these accounted for only one tenth of the total amount. Implementing preventive programs and increasing the control of cardiovascular risk factors may have a significant social and health impact by helping to reduce mortality and costs for the Spanish National Health System. The costs derived from pharmacological treatments were obtained from the NHS pricing nomenclator database (https://www.sanidad.gob.es/profesionales/nomenclator.do).

El objetivo del presente artículo es analizar las aportaciones de las herramientas de ayuda automática (HAA) al aprendizaje consciente de la escritura académica (EA) en entornos universitarios, considerando tres factores de diversidad: las diferencias entre aprender EA en L1 o L2, la experiencia previa con feedback correctivo y las particularidades de las distintas HAA. Específicamente, comparamos las características de dos HAA (arText y Estilector), las ponemos a prueba al revisar un corpus de artículos académicos y exploramos cómo percibe su utilidad un grupo de 24 informantes. El marco teórico son los estudios acerca de la EA y sobre el uso de HAA. Nuestra metodología es mixta y entre los instrumentos de investigación contamos con una plantilla para analizar cualitativamente HAA y un cuestionario para cuantificar la opinión de los participantes. Los resultados muestran la utilidad de ambas HAA, sus rasgos específicos y la constatación de que aún son escasamente conocidas.

This article deals with recent Spanish and Portuguese political television series. Within this sub-genre, it is pertinent to consider the symbolic construction of politics, as well as the differences caused by each series’ geographical adscription. Six Spanish productions have been selected—Isabel (Isabella the Catholic), Carlos Rey Emperador (Charles the Emperor King), La Embajada (The Embassy), Crematorio (Crematorium), Vamos Juan/Venga Juan (Come on, Juan/Let’s go Juan), and El Partido (The Party)—along with three Portuguese productions—A Rainha e a Bastarda (The Queen and the Bastard), Teorias da Conspiração (Conspiracy Theories), and Os Boys (The Boys). The narrative of these audio-visual stories has been examined utilising qualitative content analysis, looking at the plotlines and characters involved. The type of characterisation of politics has been identified by means of the deconstruction of the main characters. The conclusion is that the evaluation is eminently negative, although differential frameworks are present, depending, in particular, on the fiction’s genre, either historical drama, drama-thriller, or comedy.

Abstract Traditional agroecological knowledge (i.e. TAeK) is gaining recognition for its potential contribution to climate change adaptation in food systems, ecosystems restoration and food insecurity. Despite the existing literature on Traditional Ecological Knowledge and its nexus with food security, how gender critically influences the distribution of such knowledge within agri-food systems has not yet been systematically analysed. In this regard, this systematic review attempts to answer four questions: 1) How does the literature on gender and TAeK in agri-food systems evolved temporally, geographically and in different agroecosystems? 2) How are gender and intersectionality mainly approached by such literature? 3) How do the articles address gendered dimensions in TAeK within the agri-food system activities? 4) What are the main drivers of change that influence TAeK and adaptive responses? The results show the gendered nature of TAeK in relation to food production, processing, and conservation activities, and how these activities are linked to tasks and activities, gender-specific knowledge, and spaces where gender discrimination is reproduced. The review also identifies elements that delimit and/or take part of the development of TAeK, such as gendered access to resources, gendered institutions, and the identification of the main drivers of change and impacts of TAeK erosion and biodiversity loss. These results are discussed in terms of power relations that interact with sociocultural norms and practices according to the specific geographical context and agroecosystem.

Soil salinization poses a significant threat to agricultural productivity, necessitating innovative agronomic strategies to mitigate its impact. This study focuses on improving salt stress resistance in tomato plants through the application of silicon (Si) in roots. A greenhouse experiment was carried out under normal conditions (control, and 1 and 4 mM Si) and under salinity stress (salt control, and 1 and 4 mM Si). Various parameters were analyzed in leaves and roots. Under normal conditions, tomato plants grown in non-saline conditions exhibited some toxicity when exposed to Na2SiO3. As for the experiments under salt stress conditions, Si mitigated oxidative damage, preserving root cell membrane integrity. The concentration of malondialdehyde was reduced by 69.5%, that of proline was reduced by 56.4% and there was a 57.6% decrease in catalase activity for tomato plants treated with 1 mM Si under salt stress. Furthermore, Fe uptake and distribution, under salt conditions, increased from 91 to 123 mg kg−1, the same concentration as that obtained for the normal control. In all cases, the lower dose produced better results under normal conditions than the 4 mM dose. In summary, this research provides a potential application of Si in non-fertigated crop systems through a radicular pathway.

Las sociedades Cooperativas de Energía están cambiando la realidad del sector energético. Esto es un tema incipiente de investigación que da lugar a una emergente e invisible estructura cientifica. Conocer a esta estructura puede originar unas oportunidades y un apoyo de referencia. A partir de este objetivo y mediante un análisis de red (UCINET-6), se construyeron tres redes: las personas autoras, los centros de afiliación y los países donde radican los trabajos académicos sobre las cooperativas de energía. Se analizaron 47 aportaciones, 24 fuentes, 113 personas autoras pertenecientes a 79 centros de investigación ubicados en 20 países. Los resultados indicaron que los personas autoras suelen cooperar en la producción científica, aunque la colaboración es local, pues ella se origina entre personas pertenecientes a mismos centros y países. Se halló una estructura de red fragmenta y centralizada donde 10 personas científicas son responsables de una producción específica.

Introdução: A faixa etária infantil constitui um grupo suscetível à deficiência do ferro, devido ao alto consumo deste mineral em função da exponencial velocidade de crescimento. Associada a susceptibilidade, alguns hábitos negativos da alimentação na infância podem aumentar esse desarranjo, como por exemplo, consumo escasso de alimentos fontes de ferro. Objetivo: Analisar as frequências de internação por anemia ferropriva entre crianças nordestinas e sua distribuição segundo variáveis demográficas e territoriais entre 2014 e 2023. Material e métodos: Trata-se de um estudo ecológico e descritivo. Foram utilizados dados do Ministério da Saúde - Sistema de Informações Hospitalares do SUS (SIH/SUS). Informações de crianças (0-14 anos) internados devido anemia por deficiência de ferro entre 2014 e 2023. Essa variável principal foi categorizada segundo sexo, faixa de idade, cor/raça, unidade da federação e região geográfica de moradia. Todos os dados foram coletados utilizando o TABNET. Resultados: Analisou-se 3062 internações por anemia ferropriva no Nordeste. Dentre os estados, Bahia (694) e Pernambuco (662) foram os que tiveram mais casos. As faixas etárias com mais internações foram a de menor de 1 ano, com 1009 casos (32,95%), e a de entre 1 a 4 anos, com 1004 casos (32,79%). Em relação à raça, a mais acometida foi a parda, com 1787 internações (58,36%), seguida da categoria sem informação (997). O sexo com mais internações foi o masculino com 1613 casos (52,68%). Discussão: A anemia ferropriva é a responsável pela maior parte das anemias encontradas e está associada a mais de 60% dos casos de anemia no mundo. Esta patologia afeta o crescimento e o desenvolvimento da criança, que são pilares da saúde infantil, como: o desenvolvimento de habilidades de linguagem, comportamentais e capacidades motoras. Ainda, propicia a cáries dentárias, alterações no sistema imunológico, paladar e apetite. Logo, é possível perceber o motivo do predomínio de casos de anemia ferropriva ser em menores de 1 ano, por ser uma parcela que está propensa a ter um consumo maior que a ingestão de ferro, associado com a raça parda que é uma das raças com menos acesso à saúde, sendo passíveis a falta de informação adequada quanto a alimentação infantil, elencado com o sexo masculino que desde a infancia é negligenciado quanto aos cuidados na saúde. Conclusão: O estudo evidenciou que a frequência de internações por anemia ferropriva é maior na faixa etária de risco, na raça parda e nos meninos. Mesmo com as medidas de prevenção, notou-se um número expressivo de casos que precisaram de internação durante o período analisado. Logo, deve-se reforçar tanto para as equipes de saúde, quanto para a população, o uso profilático de sulfato ferroso na primeira infância.

IntroductionRice heavily relies on nitrogen fertilizers, posing environmental, resource, and geopolitical challenges. This study explores sustainable alternatives like animal manure and remote sensing for resource-efficient rice cultivation. It aims to assess the long-term impact of organic fertilization and remote sensing monitoring on agronomic traits, yield, and nutrition.MethodsA six-year experiment in rice fields evaluated fertilization strategies, including pig slurry (PS) and chicken manure (CM) with mineral fertilizers (MIN), MIN-only, and zero-fertilization. Traits, yield, spectral responses, and nutrient content were measured. Sentinel-2 remote sensing tracked crop development.ResultsCost-effective organic fertilizers (PS and CM) caused a 13% and 15% yield reduction but still doubled zero-fertilization yield. PS reduced nitrogen leaching. Heavy metals in rice grains were present at safe amounts. Organic-fertilized crops showed nitrogen deficiency at the late vegetative stages, affecting yield. Sentinel-2 detected nutrient deficiencies through NDVI.DiscussionOrganic fertilizers, especially PS, reduce nitrogen loss, benefiting the environment. However, they come with yield trade-offs and nutrient management challenges that can be managed and balanced with reduced additional mineral applications. Sentinel-2 remote sensing helps manage nutrient deficiencies. In summary, this research favors cost-effective organic fertilizers with improved nutrient management for sustainable rice production.

Background & Aims: : The accumulation of adipose tissue macrophages (ATMs) in obesity has been associated with hepatic injury. However, the contribution of ATMs to hepatic fibrosis in non-alcoholic fatty liver disease (NAFLD) remains to be elucidated. Herein, we investigate the relationship between ATMs and liver fibrosis in patients with patients with NAFLD and evaluate the impact of modulation of ATMs over hepatic fibrosis in an experimental non-alcoholic steatohepatitis (NASH) model. Methods: Adipose tissue and liver biopsies from 42 patients with NAFLD with different fibrosis stages were collected. ATMs were characterised by immunohistochemistry and flow cytometry and the correlation between ATMs and liver fibrosis stages was assessed. Selective modulation of the ATM phenotype was achieved by i.p. administration of dextran coupled with dexamethasone in diet-induced obesity and NASH murine models. Chronic administration effects were evaluated by histology and gene expression analysis in adipose tissue and liver samples. In vitro crosstalk between human ATMs and hepatic stellate cells (HSCs) and liver spheroids was performed. Results: Patients with NAFLD presented an increased accumulation of pro-inflammatory ATMs that correlated with hepatic fibrosis. Long-term modulation of ATMs significantly reduced pro-inflammatory phenotype and ameliorated adipose tissue inflammation. Moreover, ATMs modulation was associated with an improvement in steatosis and hepatic inflammation and significantly reduced fibrosis progression in an experimental NASH model. In vitro, the reduction of the pro-inflammatory phenotype of human ATMs with dextran–dexamethasone treatment reduced the secretion of inflammatory chemokines and directly attenuated the pro-fibrogenic response in HSCs and liver spheroids. Conclusions: Pro-inflammatory ATMs increase in parallel with fibrosis degree in patients with NAFLD and their modulation in an experimental NASH model improves liver fibrosis, uncovering the potential of ATMs as a therapeutic target to mitigate liver fibrosis in NAFLD. Impact and implications: We report that human adipose tissue pro-inflammatory macrophages correlate with hepatic fibrosis in non-alcoholic fatty liver disease (NAFLD). Furthermore, the modulation of adipose tissue macrophages (ATMs) by dextran-nanocarrier conjugated with dexamethasone shifts the pro-inflammatory phenotype of ATMs to an anti-inflammatory phenotype in an experimental murine model of non-alcoholic steatohepatitis. This shift ameliorates adipose tissue inflammation, hepatic inflammation, and fibrosis. Our results highlight the relevance of adipose tissue in NAFLD pathophysiology and unveil ATMs as a potential target for NAFLD.

Energy consumption in public education buildings depends on use and occupancy. To improve energy performance, energy audits are essential to identify specific solutions for each building. In this study, we conducted an energy audit of a historical building used as a public education centre. We collected gas and electricity bills and recorded indoor temperatures for over a year to determine heating and cooling set points and the schedules. Our analysis showed that 47.42% of the electricity bills were unused. To reduce energy demand and improve thermal comfort, we both developed and validated a Building Energy Modelling (BEM) approach using TRNSYS18 and weather data during year 2021. The BEM model allowed us to propose efficient measures to meet the Standard Passive requirements. Our results demonstrate the effectiveness of energy audits and BEM modelling in reducing energy consumption in public education buildings.

Purpose: The aim of this paper is to form a comprehensive definition to describe the term “job crafting”. Design/methodology/approach: Through a systematic review of the literature found in databases, such as Web of Science, Scopus, Emerald and ABI, articles containing the words “job crafting” in the title and abstract were identified. After following several criteria applied by previous similar analyses, the resulting final sample was 47 articles. Findings: In this research paper on the term “job crafting”, 76 different definitions of its meaning were identified. After a systematic analysis of these definitions, with the aim of extracting the common elements that characterise job crafting, a definition is proposed, which makes it possible to include any typology, based on three key elements: the employee’s proactivity, the impact on the environment and the context in which the activity is carried out. Research limitations/implications: The analysis is based on a review of extant literature, which previously has not considered the impact of such definitions of job crafting on hybrid (physical and remote) and virtual (metaverse) environments. Practical implications: The paper aims to provide a simple and all-inclusive definition of job crafting that serves as a basis for identifying attitudes and behavior that will help in talent attraction, selection and development processes. Social implications: This integrated definition will help individuals to develop the attitudes and behavior which will enable them to adapt their professional activities in a context where, according to the World Economic Forum (2020 study), 50% of the jobs we know today will disappear by 2030. Originality/value: This article attempts to provide a succinct and all-inclusive definition of the term job crafting; something which has not previously been achieved.

Summary: Conventional dendritic cells (cDCs) are found in most tissues and play a key role in initiation of immunity. cDCs require constant replenishment from progenitors called pre-cDCs that develop in the bone marrow (BM) and enter the blood circulation to seed all tissues. This process can be markedly accelerated in response to inflammation (emergency cDCpoiesis). Here, we identify two populations of BM pre-cDC marked by differential expression of CXCR4. We show that CXCR4lo cells constitute the migratory pool of BM pre-cDCs, which exits the BM and can be rapidly mobilized during challenge. We further show that exit of CXCR4lo pre-cDCs from BM at steady state is partially dependent on CCR2 and that CCR2 upregulation in response to type I IFN receptor signaling markedly increases efflux during infection with influenza A virus. Our results highlight a fine balance between retention and efflux chemokine cues that regulates steady-state and emergency cDCpoiesis.
