Abstract

Beyond conventional productivity metrics, human interaction and collaboration dynamics merit careful consideration in our increasingly digital workspace. This research proposes a conjectural neuro-adaptive room that enhances group interactions by adjusting the physical environment to desired internal states. Drawing inspiration from previous work on collective consciousness, the system leverages computer vision and machine learning models to analyze physiological and behavioral cues, such as facial expressions and speech analysis, to infer the overall internal state of occupants. Environmental conditions of the room, such as visual projections, lighting and sound, are actively adjusted to create an optimal setting for inducing the desired state, including focus or collaboration. 2. Dynamical Mean-Field Theory of Self-Attention Neural Networks

Abstract

Transformer-based models have demonstrated exceptional performance across diverse domains, becoming the state-of-the-art solution for addressing sequential machine learning problems. Even though we have a general understanding of the fundamental components in the transformer architecture, little is known about how they operate or what are their expected dynamics. Recently, there has been an increasing interest in exploring the relationship between attention mechanisms and Hopfield networks, promising to shed light on the statistical physics of transformer networks. However, to date, the dynamical regimes of transformer-like models have not been studied in depth. In this paper, we address this gap by using methods for the study of asymmetric Hopfield networks in nonequilibrium regimes --namely path integral methods over generating functionals, yielding dynamics governed by concurrent mean-field variables. Assuming 1-bit tokens and weights, we derive analytical approximations for the behavior of large self-attention neural networks coupled to a softmax output, which become exact in the large limit size. Our findings reveal nontrivial dynamical phenomena, including nonequilibrium phase transitions associated with chaotic bifurcations, even for very simple configurations with a few encoded features and a very short context window. Finally, we discuss the potential of our analytic approach to improve our understanding of the inner workings of transformer models, potentially reducing computational training costs and enhancing model interpretability.

Abstract

Pressure evolution of the crystal structure and magnetism of the honeycomb $\alpha$-RuBr$_3$ is studied using high-pressure x-ray diffraction, magnetometry, and density-functional band-structure calculations. Hydrostatic compression transforms antiferromagnetic $\alpha$-RuBr$_3$ ($R\bar 3$) into paramagnetic $\alpha'$-RuBr$_3$ ($P\bar 1$) where short Ru-Ru bonds cause magnetism collapse above 1.3 GPa at 0 K and 2.5 GPa at 295 K. Below this critical pressure, the N\'eel temperature of $\alpha$-RuBr$_3$ increases with the slope of 1.8 K/GPa. Pressure tunes $\alpha$-RuBr$_3$ away from the Kitaev limit, whereas increased third-neighbor in-plane coupling and interlayer coupling lead to a further stabilization of the collinear zigzag state. Both $\alpha$- and $\alpha'$-RuBr$_3$ are metastable at ambient pressure, but their transformation into the thermodynamically stable $\beta$-polymorph is kinetically hindered at room temperature., Comment: 8 pages + Supplemental Material: published version

Abstract

Cannabichromene (CBC) is a nonpsychoactive phytocannabinoid well-known for its wide-ranging health advantages. However, there is limited knowledge regarding its human metabolism following CBC consumption. This research aimed to explore the metabolic pathways of CBC by various human liver cytochrome P450 (CYP) enzymes and support the outcomes using in vivo data from mice. The results unveiled two principal CBC metabolites generated by CYPs: 8-hydroxy-CBC and 6,7-epoxy-CBC, along with a minor quantity of 1″-hydroxy-CBC. Notably, among the examined CYPs, CYP2C9 demonstrated the highest efficiency in producing these metabolites. Moreover, through a molecular dynamics simulation spanning 1 μs, it was observed that CBC attains stability at the active site of CYP2J2 by forming hydrogen bonds with I487 and N379, facilitated by water molecules, which specifically promotes the hydroxy metabolites formation. Additionally, the presence of cytochrome P450 reductase (CPR) amplified CBCs binding affinity to CYPs, particularly with CYP2C8 and CYP3A4. Furthermore, the metabolites derived from CBC reduced cytokine levels, such as IL6 and NO, by approximately 50% in microglia cells. This investigation offers valuable insights into the biotransformation of CBC, underscoring the physiological importance and the potential significance of these metabolites.

Abstract

Bayesian theories of biological and brain function speculate that Markov blankets (a conditional independence separating a system from external states) play a key role for facilitating inference-like behaviour in living systems. Although it has been suggested that Markov blankets are commonplace in sparsely connected, nonequilibrium complex systems, this has not been studied in detail. Here, we show in two different examples (a pair of coupled Lorenz systems and a nonequilibrium Ising model) that sparse connectivity does not guarantee Markov blankets in the steady-state density of nonequilibrium systems. Conversely, in the nonequilibrium Ising model explored, the more distant from equilibrium the system appears to be correlated with the distance from displaying a Markov blanket. These result suggests that further assumptions might be needed in order to assume the presence of Markov blankets in the kind of nonequilibrium processes describing the activity of living systems.

Abstract

Chronic myelomonocytic leukemia (CMML) is a rare blood cancer of older adults (3 in every 1,000,000 persons) characterized by poor survival and lacking effective mutation-specific therapy. Mutations in the ubiquitin ligase Cbl occur frequently in CMML and share biological and molecular features with a clonal disease occurring in children, juvenile myelomonocytic leukemia (JMML). Here we analyzed the clinical presentations, molecular features and immunophenotype of CMML patients with CBL mutations enrolled in a prospective Phase II clinical trial stratified according to molecular markers. Clinically, CBL mutations were associated with increased bone marrow blasts at diagnosis, leukocytosis and splenomegaly, similar to patients harboring NRAS or KRAS mutations. Interestingly, 64% of patients presented with more than one CBL variant implying a complex subclonal architecture, often with co-occurrence of TET2 mutations. We found CBL mutations in CMML frequently clustered in the RING domain in contrast to JMML, where mutations frequently involve the linker helix region (P<0.0001). According to our comparative alignment of available X-ray structures, mutations in the linker helix region such as Y371E give rise to conformational differences that could be exploited by targeted therapy approaches. Furthermore, we noted an increased percentage of CMML CD34+ stem and progenitor cells expressing CD116 and CD131 in all CBL mutant cases and increased CD116 receptor density compared to healthy controls, similar to CMML overall. In summary, our data demonstrate that CBL mutations are associated with distinct molecular and clinical features in CMML and are potentially targetable with CD116-directed immunotherapy. Abstract

In contrast to the many reports of successful real-world cases of personalized bacteriophage therapy (BT), randomized controlled trials of non-personalized bacteriophage products have not produced the expected results. Here we present the outcomes of a retrospective observational analysis of the first 100 consecutive cases of personalized BT of difficult-to-treat infections facilitated by a Belgian consortium in 35 hospitals, 29 cities and 12 countries during the period from 1 January 2008 to 30 April 2022. We assessed how often personalized BT produced a positive clinical outcome (general efficacy) and performed a regression analysis to identify functional relationships. The most common indications were lower respiratory tract, skin and soft tissue, and bone infections, and involved combinations of 26 bacteriophages and 6 defined bacteriophage cocktails, individually selected and sometimes pre-adapted to target the causative bacterial pathogens. Clinical improvement and eradication of the targeted bacteria were reported for 77.2% and 61.3% of infections, respectively. In our dataset of 100 cases, eradication was 70% less probable when no concomitant antibiotics were used (odds ratio = 0.3; 95% confidence interval = 0.127–0.749). In vivo selection of bacteriophage resistance and in vitro bacteriophage–antibiotic synergy were documented in 43.8% (7/16 patients) and 90% (9/10) of evaluated patients, respectively. We observed a combination of antibiotic re-sensitization and reduced virulence in bacteriophage-resistant bacterial isolates that emerged during BT. Bacteriophage immune neutralization was observed in 38.5% (5/13) of screened patients. Fifteen adverse events were reported, including seven non-serious adverse drug reactions suspected to be linked to BT. While our analysis is limited by the uncontrolled nature of these data, it indicates that BT can be effective in combination with antibiotics and can inform the design of future controlled clinical trials.
