This research examines food delivery couriers’ preferred employment status and factors explaining their opinions. Previous studies have used qualitative research methods and are unable to explain couriers’ general views on employment status. In this research, a survey of 1,539 Wolt couriers was carried out in Finland with logistic regression, cross-tabulation, and content analysis as analysis methods. The results show that 56% of the couriers wanted to work as self-employed and 25% as employed. The opinion was most strongly explained by valuing work-related freedom and flexibility, which were associated with the right to refuse delivery tasks offered and to choose the amount of work, working hours and delivery vehicle. The preference for self-employment was also increased by the duration of courier work, one’s own choice to work as a courier, and age. Freedom and flexibility are dependent on the sufficient availability of delivery tasks, posing challenges when the demand is low.

Abstract Amongst the potential contribution of protein or peptide-display systems to study epitopes with relevant immunological features, the RAD display system stands out as a highly stable scaffold protein that allows the presentation of constrained target peptides. Here, we employed the RAD display system to present peptides derived from the SARS-CoV-2 Spike (S) protein as a tool to detect specific serum antibodies and to generate polyclonal antibodies capable of inhibiting SARS-CoV-2 infectivity in vitro. 44 linear S-derived peptides were genetically fused with the RAD scaffold (RAD-SCoV-epitopes) and screened for antigenicity with sera collected from COVID-19-infected patients. In a second step, selected RAD-SCoV-epitopes were used to immunize mice and generate antibodies. Phenotypic screening showed that some of these antibodies were able to recognize replicating viral particles in VERO CCL-81 and most notably seven of the RAD-SCoV-epitopes were able to induce antibodies that inhibited viral infection. Our findings highlight the RAD display system as an useful platform for the immunological characterization of peptides and a potentially valuable strategy for the design of antigens for peptide-based vaccines, for epitope-specific antibody mapping, and for the development of antibodies for diagnostic and therapeutic purposes.

Abstract Age-associated B cells (ABC) accumulate with age and in individuals with different immunological disorders, including cancer patients treated with immune checkpoint blockade and those with inborn errors of immunity. Here, we investigate whether ABCs from different conditions are similar and how they impact the longitudinal level of the COVID-19 vaccine response. Single-cell RNA sequencing indicates that ABCs with distinct aetiologies have common transcriptional profiles and can be categorised according to their expression of immune genes, such as the autoimmune regulator (AIRE). Furthermore, higher baseline ABC frequency correlates with decreased levels of antigen-specific memory B cells and reduced neutralising capacity against SARS-CoV-2. ABCs express high levels of the inhibitory FcγRIIB receptor and are distinctive in their ability to bind immune complexes, which could contribute to diminish vaccine responses either directly, or indirectly via enhanced clearance of immune complexed-antigen. Expansion of ABCs may, therefore, serve as a biomarker identifying individuals at risk of suboptimal responses to vaccination.

Abstract COVID-19 has stimulated the rapid development of new antibody and small molecule therapeutics to inhibit SARS-CoV-2 infection. Here we describe a third antiviral modality that combines the drug-like advantages of both. Bicycles are entropically constrained peptides stabilized by a central chemical scaffold into a bi-cyclic structure. Rapid screening of diverse bacteriophage libraries against SARS-CoV-2 Spike yielded unique Bicycle binders across the entire protein. Exploiting Bicycles’ inherent chemical combinability, we converted early micromolar hits into nanomolar viral inhibitors through simple multimerization. We also show how combining Bicycles against different epitopes into a single biparatopic agent allows Spike from diverse variants of concern (VoC) to be targeted (Alpha, Beta, Delta and Omicron). Finally, we demonstrate in both male hACE2-transgenic mice and Syrian golden hamsters that both multimerized and biparatopic Bicycles reduce viraemia and prevent host inflammation. These results introduce Bicycles as a potential antiviral modality to tackle new and rapidly evolving viruses.

Abstract Bioactive glass (BAG) is a bone substitute that can be used in orthopaedic surgery. Following implantation, the BAG is expected to be replaced by bone via bone growth and gradual degradation of the BAG. However, the hydroxyapatite mineral forming on BAG resembles bone mineral, not providing sufficient contrast to distinguish the two in X-ray images. In this study, we co-registered coded-excitation scanning acoustic microscopy (CESAM), scanning white light interferometry (SWLI), and scanning electron microscopy with elemental analysis (Energy Dispersive X-ray Spectroscopy) (SEM–EDX) to investigate the bone growth and BAG reactions on a micron scale in a rabbit bone ex vivo. The acoustic impedance map recorded by the CESAM provides high elasticity-associated contrast to study materials and their combinations, while simultaneously producing a topography map of the sample. The acoustic impedance map correlated with the elemental analysis from SEM–EDX. SWLI also produces a topography map, but with higher resolution than CESAM. The two topography maps (CESAM and SWLI) were in good agreement. Furthermore, using information from both maps simultaneously produced by the CESAM (acoustic impedance and topography) allowed determining regions-of-interest related to bone formation around the BAG with greater ease than from either map alone. CESAM is therefore a promising tool for evaluating the degradation of bone substitutes and the bone healing process ex vivo.

Reactivity-controlled compression ignition (RCCI) is a promising new combustion technology for marine applications. It has offered the potential to achieve low NOx emissions and high thermal efficiency, which are both important considerations for marine engines. However, the performance of RCCI engines is sensitive to a number of factors, including the start of injection. This study used computational fluid dynamics (CFD) to investigate the effects of start of ignition (SOI) on the performance of a marine RCCI engine. The CFD model was validated against experimental data, and the results showed that the SOI has a significant impact on the combustion process. In particular, the SOI affected the distribution of fuel and air in the combustion chamber, which in turn affected the rate of heat release and the formation of pollutants. Ten different SOIs were implemented on a validated closed-loop CFD model from 96 to 42 CAD bTDC (crank angle degree before top dead center) at six-degree intervals. A chemical kinetic mechanism of 54 species and 269 reactions tuned and used for simulation of in-cylinder combustion. The results show that in early injection, high-reactivity fuel was distributed close to the liner. This distribution was around the center of late injection angles. A homogeneity study was carried out to investigate the local equivalence ratio. It showed a more homogenous mixture in early injection until 66 CAD bTDC, after which point, earlier injection timing had no effect on homogeneity. Maximum indicated mean effective pressure (IMEP) was achieved at SOI 48 CAD bTDC, and minimum amounts of THC (total hydrocarbons) and NOx were observed with middle injection timing angles around 66 CAD bTDC.

Several northern European countries have announced ambitious plans to become carbon neutral already before the year 2050. Recent research has, however, highlighted how potential bottlenecks in raw material and resource availability could significantly delay or hinder wind and solar photovoltaic (PV) expansion and continued biomass usage in parts of Europe. To address this issue, this paper assesses how exposed the national energy and climate plans (NECPs) of Finland, Estonia, Germany, Sweden, and Denmark are to resource limitations and techno-economic risks by reviewing and analysing 2030 NECP targets compared to statistical energy use data in these countries. The results indicate that the NECPs of Denmark and Germany are particularly exposed to risks related to global raw material availability, as Denmark plans to rapidly grow the share of wind and solar PV in electricity generation to 81% and 13% by 2030, respectively, followed by Germany, which outlines a 39% and 16% share of wind and solar PV in its national climate strategy. The NECPs of Finland and Germany are also shown to be vulnerable to limitations in biomass availability, as there is a significant disparity between the projected biomass usage and legally binding European Union (EU) targets for land use, land use change, and forestry (LULUCF) sector emissions in 2030 in these countries.

Here, the authors establish a protease selection system based on bacterial alpha-2-macroglobulin protease inhibitors and evolve staphylococcal proteases for increased activity and altered specificity.

A TGFβ/BMP receptor library with Halo- and SNAP-tags is generated and with the application of labeled growth factor ligands, a ligand surface binding assay is developed, allowing assessment of preferential ligand-receptor binding in cells.

Regular weed surveys provide information on changes in weed communities. The composition of weed flora in spring cereal fields was investigated in southern and central Finland during 2020–2022. The survey was conducted in 16 regions, 226 farms and 549 fields. The occurrence of weeds was assessed in the second half of July from 90 organically cropped and 459 conventionally cropped fields. The weed flora was dominated by broad-leaved species, representing 81% of the total number of 138 identified species. The average species number per field was 21 in organic fields and 12 in conventional fields. The most frequent weed species in organic fields were Chenopodium album 99%, Erysimum cheiranthoides 84%, Viola arvensis 83% and Stellaria media 82%. In conventionally cropped fields, the most frequent species were Viola arvensis 83%, Stellaria media 65%, Galeopsis spp. 59% and Galium spurium 59%. The two most common grass species in both production systems were Poa annua and Elymus repens. The frequency and density of Poa annua had increased substantially in conventional cropping since the previous survey in 2007–2009. The average density of weeds was 384 plants m-2 in organic fields and 147 plants m-2 in sprayed conventional fields. The average air-dry biomass of weeds was 678 kg ha-1 and 151 kg ha-1, respectively. Elymus repens produced almost a quarter of the total weed biomass in both production systems. Changes in weed flora were minor in terms of frequency and density of the most common broad-leaved weed species. Increased infestation of Poa annua in conventional cropping calls for special attention. The dominance of Elymus repens decreased in both production systems since the previous survey.

The increase of popularity of reactivity-controlled compression ignition (RCCI) is attributed to its capability of achieving ultra-low nitrogen oxides (NOx) and soot emissions with high brake thermal efficiency (BTE). The complex and nonlinear nature of the RCCI combustion makes it challenging for model-based control design. In this work, a model-based control system is developed to control the combustion phasing and the indicated mean effective pressure (IMEP) of RCCI combustion through the adjustments of total fuel energy and blend ratio (BR) in fuel injection. A physics-based nonlinear control-oriented model (COM) is developed to predict the main combustion performance indicators of an RCCI marine engine. The model is validated against a detailed thermo-kinetic multizone model. A novel linear parameter-varying (LPV) model coupled with a model predictive controller (MPC) is utilized to control the aforementioned parameters of the developed COM. The developed system is able to control combustion phasing and IMEP with a tracking error that is within a 5% error margin for nominal and transient engine operating conditions. The developed control system promotes the adoption of RCCI combustion in commercial marine engines.

Introduction: Oral tongue squamous cell carcinoma (OTSCC) is the most common cancer of the oral cavity. Contradictory results have been observed on the involvement of herpes simplex virus 1 (HSV-1) in oral squamous cell carcinomas. Here, we aimed to study the predominance of HSV-1 or HSV-2 in oral HSV infections and to investigate the presence of HSV-1 in OTSCC and its effect on carcinoma cell viability and invasion.Methods: The distribution of HSV types one and two in diagnostic samples taken from suspected oral HSV infections was determined from the Helsinki University Hospital Laboratory database. We then analysed 67 OTSCC samples for HSV-1 infection using immunohistochemical staining. We further tested the effects of HSV-1 using six concentrations (0.00001–1.0 multiplicity of infection [MOI]) on viability and two concentrations (0.001 and 0.1 MOI) on invasion of highly invasive metastatic HSC-3 and less invasive primary SCC-25 OTSCC cell lines using MTT and Myogel-coated Transwell invasion assays.Results: Altogether 321 oropharyngeal samples were diagnosed positive for HSV during the study period. HSV-1 was the predominant (97.8%) HSV type compared with HSV-2 (detected in 2.2% of samples). HSV-1 was also detected in 24% of the OTSCC samples and had no association with patient survival or recurrence. OTSCC cells were viable even after 6 days with low viral load (0.00001, 0.0001, 0.001 MOI) of HSV-1. In both cell lines, 0.001 MOI did not affect cell invasion. However, 0.1 MOI significantly reduced cell invasion in HSC-3 cells.Discussion: HSV-1 infection is predominant compared with HSV-2 in the oral cavity. HSV-1 is detected in OTSCC samples without clinical significance, and OTSCC cell survival or invasion was not affected at low doses of HSV-1.

Abstract Huge advances in understanding patterns of human communication, and the underlying social networks where it takes place, have been made recently using massive automatically recorded data sets from digital communication, such as emails and phone calls. However, it is not clear to what extent these results on human behaviour are artefacts of contemporary communication technology and culture and if the fundamental patterns in communication have changed over history. This paper presents an analysis of historical epistolary metadata with the aim of comparing the underlying historical communication patterns with those of contemporary communication. Our work uses a new epistolary dataset containing metadata on over 150,000 letters sent between the 16th and 19th centuries. The analyses indicate striking resemblances between contemporary and epistolary communication network patterns, including dyadic interactions and ego-level behaviour. Certain aspects of the letter datasets are insufficient to corroborate other similarities or differences for these communication networks. Despite these drawbacks, our work helps confirm that several features of human communication are not artefacts of contemporary mediums or culture, but are likely elements of human behaviour.

Intra-abdominal lymphatic malformations (LMs) are relatively rare congenital anomalies and can be divided into intra- and retroperitoneal subgroups. This study aims to evaluate the outcomes after surgical resection of intraperitoneal LMs in children. The records of all patients ≤16 years of age with intraperitoneal LMs managed at our tertiary center between 2007–2022 were reviewed. Patients with retroperitoneal LMs were excluded. Our preferred approach is surgical exploration for large (>5 cm), symptomatic lesions if they are potentially resectable on imaging. A total of 12 patients (10 males) were diagnosed with intraperitoneal LMs located in the small bowel mesentery or omentum at a median age of 6.6 (range, 0.6−14.4) years. Ten (83%) patients presented with acute symptoms, including abdominal pain (n = 8; 67%) and vomiting (n = 3; 25%). Two (17%) intraperitoneal LMs were found on imaging performed for other reasons. Surgical exploration was performed in 11/12 (92%) cases. Intestinal volvulus around a mesenteric macrocyst had occurred in 4 (36%) patients. Complete macroscopic resection was achieved in 10/11 (91%) patients. Most lesions were localized to a short intestinal segment. One mixed type LM with an extensive mesenteric involvement was biopsied as the lesion was deemed to be unresectable in preoperative imaging and the diagnosis was initially unclear. There was one recurrence (8%) during postoperative follow-up on imaging (median 3.6 (range, 1.5−6.9) years) but the patient has remained asymptomatic and not undergone interventions. 6/8 (75%) of screened intraperitoneal LMs were positive for a somatic PIK3CA mutation. Most large, pedunculated macrocystic LMs in the small bowel mesentery or omental location were amenable to macroscopic resection. Intraperitoneal LMs tend to present with acute symptoms including a risk for intestinal volvulus based on anatomical location. Our results suggest low recurrence rates or need for further interventions over follow-up. III. • What is currently known about this topic? In this work, a model predictive controller is developed for a multiple injection combustion model. A 1D engine model with three distinct injections is used to generate data for identifying the state-space representation of the engine model. This state-space model is then used to design a controller for controlling the start of injection and injected fuel mass of the post injection. These parameters are used as inputs for the engine model to control the maximum cylinder pressure and indicated mean effective pressure.

The increasing demands for reducing fuel consumption and emissions in contemporary technology solutions lead to the use of more sensors, actuators, and control applications. With this increasing engine complexity, the feedback design is complex due to the coupling between inputs and combustion parameters. To be able to design the controller systematically, model predictive control (MPC) comes to the scope because of its advantages in the design of multi-input multi-output (MIMO) systems, especially with its constraints handling ability and performance in simultaneously optimizing the engine fuel efficiency and emission reduction. Multi-injection is one of the promising techniques for achieving better engine performance. In this work, post-injection control is implemented utilizing MPC MIMO strategy with the target of exploring the possibility of reducing emissions and improving engine efficiency by controlling post-injection duration and injection timing. The workflow of the MPC controller design from control-oriented model (COM) establishing to MPC problem formation and solution methodology is discussed in this work. Moreover, one contribution from this work is the different implementation angle when compared with the state-of-the-art approaches, where the MPC controller is implemented purely by Matlab Simulink to enable the rapid control prototyping design. The simulation result demonstrated the ability of the controller’s tracking performance and showed a preliminary step towards the nonlinear combustion model-based multi-injection MPC design. The systematic model-based controller framework developed in this work can be applied to other control applications and enables a fast path from design to test.

Abstract With a limited coding capacity of 4.7 kb, adeno-associated virus (AAV) genome has evolved over-lapping genes to maximise the usage of its genome. An example is the recently found ORF in the cap gene, encoding membrane-associated accessory protein (MAAP), located in the same genomic region as the VP1/2 unique domain, but in a different reading frame. This 13 KDa protein, unique to the dependovirus genus, is not homologous to any known protein. Our studies confirm that MAAP translation initiates from the first CTG codon found in the VP1 ORF2. We have further observed MAAP localised in the plasma membrane, in the membranous structures in close proximity to the nucleus and to the nuclear envelope by co-transfecting with plasmids encoding the wild-type AAV (wt-AAV) genome and adenovirus (Ad) helper genes. While keeping VP1/2 protein sequence identical, both inactivation and truncation of MAAP translation affected the emergence and intracellular distribution of the AAV capsid proteins. We have demonstrated that MAAP facilitates AAV replication and has a role in controlling Ad infection. Additionally, we were able to improve virus production and capsid integrity through a C-terminal truncation of MAAP while other modifications led to increased packaging of contaminating, non-viral DNA. Our results show that MAAP plays a significant role in AAV infection, with profound implications for the production of therapeutic AAV vectors.

Although the RDF query language SPARQL has a reputation for being opaque and difficult for traditional humanists to learn, it holds great potential for opening up vast amounts of Linked Open Data to researchers willing to take on its challenges. This is especially true in the field of premodern manuscripts studies as more and more datasets relating to the study of manuscript culture are made available online. This paper explores the results of a two-year long process of collaborative learning and knowledge transfer between the computer scientists and humanities researchers from the Mapping Manuscript Migrations (MMM) project to learn and apply SPARQL to the MMM dataset. The process developed into a wider investigation of the use of SPARQL to analyse the data, refine research questions, and assess the research potential of the MMM aggregated dataset and its Knowledge Graph. Through an examination of a series of six SPARQL query case studies, this paper will demonstrate how the process of learning and applying SPARQL to query the MMM dataset returned three important and unexpected results: 1) a better understanding of a complex and imperfect dataset in a Linked Open Data environment, 2) a better understanding of how manuscript description and associated data involving the people and institutions involved in the production, reception, and trade of premodern manuscripts needs to be presented to better facilitate computational research, and 3) an awareness of need to further develop data literacy skills among researchers in order to take full advantage of the wealth of unexplored data now available to them in the Semantic Web.

Hyperornithinemia with gyrate atrophy of the choroid and retina (HOGA) is a severe recessive inherited disease, causing muscular degeneration and retinochoroidal atrophy that progresses to blindness. HOGA arises from mutations in the ornithine aminotransferase (OAT) gene, and nearly one-third of the known patients worldwide are homozygous for the Finnish founder mutation OAT c.1205 T > C p.(Leu402Pro). We have corrected this loss-of-function OAT mutation in patient-derived induced pluripotent stem cells (iPSCs) using CRISPR/Cas9. The correction restored OAT expression in stem cells and normalized the elevated ornithine levels in cell lysates and cell media. These results show an efficient recovery of OAT function in iPSC, encouraging the possibility of autologous cell therapy for the HOGA disease.
